Seventy-seven out of eighty-one group D streptococcal strains isolated from humans and animals were found to produce bacteriocins that were active on other streptococcal strains of gorup A and D, but inactive on their own cells. On the bases of the spectra of indicator strains, and the sensitivities to heat, chloroform, and trypsin, seven types of bacteriocins were classified. Streptococcus faecalis var. liquefaciens strain 4532 or strain A (liq-A) was UV-irradiated, and mutants which lost bacteriocin- as well as the β-hemolysin-forming activities (Bact.Hem) were obtained. Cells of the type I bacteriocin producer (SMr.TCr.Bact-I+.Hem+) and nonproducer 2025 (PCr.Bact-I.Hem), both belonging to S. faecalis var. liquefaciens, were mixed and incubated in broth. Recombinants (PCr.-SMs.TCs.Bact-I+.Hem+) were obtained at a high frequency (5.8% preinoculum size of PCr.-Bact-I.Hem), and the character was stable for at least ten transfers. In the mixed culture, a marked decrease in the recipient 2025 cell number was observed. The occurrence of recombinants was not inhibited by deoxyribonuclease. A cell-free filtrate of Bact+.Hem+ cells mixed with Bact.Hem cells did pot cause a mutation of the latter combined characters. The transfer of a genetic marker is discussed as an event of the cell-to-cell contact.  相似文献

A pot study was conducted to determine the effects of arbuscular mycorrhizal (AM) fungi (Glomus mosseae and Paraglomus occultum) and salt (NaCl) stress on growth, photosynthesis, root morphology and ionic balance of citrus (Citrus tangerine Hort. ex Tanaka) seedlings. Eighty-five-day-old seedlings were exposed to 100 mM NaCl for 60 days to induce salt stress. Mycorrhizal colonization of citrus seedlings was not affected by salinity when associated with P. occultum, but significantly decreased when with G. mosseae. Compared with the non-mycorrhizal controls, mycorrhizal seedlings generally had greater plant height, stem diameter, shoot, root and total plant biomass, photosynthetic rate, transpiration rate and stomatal conductance under the 0 and 100 mM NaCl stresses. Root length, root projected area and root surface area were also higher in the mycorrhizal than in the non-mycorrhizal seedlings, but higher root volume in seedlings with G. mosseae. Leaf Na+ concentrations were significantly decreased, but leaf K+ and Mg2+ concentrations and the K+/Na+ ratio were increased when seedlings with both G. mosseae and P. occultum. Under the salt stress, Na+ concentrations were increased but K+ concentrations decreased in the mycorrhizal seedlings. Under the salt stress, Ca2+ concentrations were increased in the seedlings with P. occultum or without AM fungi (AMF), but decreased with G. mosseae. Ratios of both Ca2+/Na+ and Mg2+/Na+ were also increased in seedlings with G. mosseae under the non-salinity stress, while only the Mg2+/Na+ ratio was increased in seedlings with P. occultum under the salt stress. Our results suggested that salt tolerance of citrus seedlings could be enhanced by associated AMF with better plant growth, root morphology, photosynthesis and ionic balance.  相似文献

Two intergeneric hybrids involving wild species Erucastrum cardaminoides (2n=18, Ecd Ecd) and two crop brassica species, Brassica rapa (2n=20, AA) and B. nigra (2n=16, BB), were synthesized through in vitro sequential ovary culture. Morphological, molecular and cytological studies were conducted to establish their hybridity. Both hybrids, though morphologically distinct, were intermediate phenotypically between their respective parents. Cytological analysis of the E. cardaminoides × B. rapa hybrid (2n=19), revealed the occurrence of 17 I+1 II at diakinesis/metaphase in the majority (28%) of the pollen mother cells (PMCs), whereas in E. cardaminoides × B. nigra hybrid (2n=17), 13 I+2 II was the predominant (32%) meiotic configuration. A maximum of 5 II was recorded in both hybrids, indicating homoeologous pairing in the respective combined genomes. Chromosome doubling by colchicine application gave rise to two new amphiploids (AA EcdEcd and BB EcdEcd) having normal chromosome pairing and pollen fertility. The occasional occurrence of one quadrivalent in the amphiploids confirmed partial homoeology between the Ec and A/B genomes. The E. cardaminoides ×B. nigra hybrid and amphiploid appeared to be tolerant to alternaria blight under field conditions.Communicated by H.F. Linskens  相似文献

A requirement for potassium for growth and forembryogenesis in suspension cultures of wild carrot (Daucus carota L.) was demonstrated. The concentration of K+ required for maximal growth (1 mM) was less than that required for maximal embryogenesis (20 mM). Neither Na+ nor NH4+ could replace K+. Ammonium ion enhanced embryogenesis when K+ was present at suboptimal levels greater than 1 mM. Nitrogen sources strongly influenced growth and embryogenesis, but the effects of nitrogen were separable from those of K+. Subline differences were noted. Subline CSC-29 produced nearly half the maximum embryo number in 1 mM K+ while CSC-31 produced no embryos at that K+ concentration. Growth of CSC-29 was slightly repressed by Na+, but no more than by similarconcentrations of K+. Growth of CSC-31 in 1 mM K+ was strongly repressed by Na+. Embryogenesis in CSC-29 was unaffected by Na+. In CSC-31, Na+ repressed embryogenesis at lower concentrations of K+.  相似文献

The strictly anaerobic homoacetogenic bacteria Acetobacterium woodii and Sporomusa sphaeroides differ with respect to their energy metabolism. Since growth as well as acetate and ATP formation of A. woodii is strictly dependent on Na+, but that of S. sphaeroides is not, the question arose whether these organisms also use different coupling ions for mechanical work, i.e. flagellar rotation. During growth on fructose in the presence of Na+ (50 mM), cells of A. woodii were vigorously motile, as judged by light microscopy. At low Na+ concentrations (0.3 mM), the growth rate decreased by only 15%, but the cells were completely non-motile. Addition of Na+ to such cultures restored motility instantaneously. Motility, as determined in swarm agar tubes, was strictly dependent on Na+; Li+, but not K+ partly substituted for Na+. Of the amilorides tested, phenamil proved to be a specific inhibitor of the flagellar motor of A. woodii. Growth and motility of S. sphaeroides was neither dependent on Na+ nor inhibited by amiloride derivatives. These results indicate that flagellar rotation is driven by ΔμNa + in A. woodii, but by ΔμH + in S. sphaeroides. Received: 30 May 1995 / Accepted: 31 August 1995  相似文献

The unicellular alga Chlamydomonas reinhardtii and the bacterium Pseudomonas protegens serve as a model to study the interactions between photosynthetic and heterotrophic microorganisms. P. protegens secretes the cyclic lipopeptide orfamide A that interferes with cytosolic Ca2+ homeostasis in C. reinhardtii resulting in deflagellation of the algal cells. Here, we studied the roles of additional secondary metabolites secreted by P. protegens using individual compounds and co-cultivation of algae with bacterial mutants. Rhizoxin S2, pyrrolnitrin, pyoluteorin, 2,4-diacetylphloroglucinol (DAPG) and orfamide A all induce changes in cell morphology and inhibit the growth of C. reinhardtii. Rhizoxin S2 exerts the strongest growth inhibition, and its action depends on the spatial structure of the environment (agar versus liquid culture). Algal motility is unaffected by rhizoxin S2 and is most potently inhibited by orfamide A (IC50 = 4.1 μM). Pyrrolnitrin and pyoluteorin both interfere with algal cytosolic Ca2+ homeostasis and motility whereas high concentrations of DAPG immobilize C. reinhardtii without deflagellation or disturbance of Ca2+ homeostasis. Co-cultivation with a regulatory mutant of bacterial secondary metabolism (ΔgacA) promotes algal growth under spatially structured conditions. Our results reveal how a single soil bacterium uses an arsenal of secreted antialgal compounds with complementary and partially overlapping activities.  相似文献

Phylogenetic relationships of polyploid Aegilops species sharing the U-genome were investigated by analyzing heterochromatin banding patterns of their somatic metaphase chromosomes as revealed by C-banding and fluorescence in situ hybridization (FISH) with the heterochromatin-limited repetitive DNA probes pSc119, pAs1, as well as the distribution of NOR and 5S DNA loci revealed by pTa71 (18S-26S rDNA), and pTa794 (5S rDNA) probes. Seven tetraploid (Ae. triuncialis, Ae. peregrina, Ae. kotschyi, Ae. geniculata, Ae. biuncialis, Ae. columnaris, and 4x Ae. neglecta) and one hexaploid (6x Ae. neglecta) Aegilops species of the U-genome cluster were studied. The Ut and Ct chromosomes of 4x Ae. triuncialis (UtCt) were similar to the diploid donors Ae. umbellulata (U) and Ae. caudata (C). However, the size of the NOR locus on chromosome 5Ut was reduced. Karyotypic analyses confirmed that 4x Ae. peregrina (SpUp) was derived from a hybridization of the diploid species Ae. umbellulata with Ae. longissima, whereas Ae. umbellulata and Ae. sharonensis (or an immediate precursor) were the diploid progenitor species of Ae. kotschyi (SkUk). In both 4x species, the NORs on S-genome chromosomes were inactivated and were accompanied with a decrease or loss of rDNA sequences. Karyotypes of the tetraploid species, Ae. geniculata (UgMg) and Ae. biuncialis (UbMb) differed from each other and from the putative diploid progenitors Ae. umbellulata and Ae. comosa indicating that various types of chromosomal alterations occurred during speciation. Inactivation of major NORs on the M-genome chromosomes, redistribution of 5S rDNA sites, and loss of some minor 18S-26S rDNA loci were observed in Ae. geniculata and Ae. biuncialis. Significant differences in the total amount and distribution of heterochromatin, the number and location of 5S and 18S-26S rDNA loci observed between Ae. columnaris (UcXc)/4x Ae. neglecta (UnXn) and Ae. geniculata/Ae. biuncialis indicate that these species have different origins. Similarities in C-banding and FISH patterns of most Ae. columnaris and 4x Ae. neglecta chromosomes suggest that they were probably derived from a common ancestor, whereas distinct differences of three chromosome pairs may indicate that the divergence of these species was probably associated with chromosomal rearrangements and/or introgressive hybridization. Ae. umbellulata contributed the U genome, however, the source of their second genomes remains unknown. The formation of 6x Ae. neglecta (UnXnNn) was not associated with large modifications of the parental genomes.  相似文献

Molecular typing was applied and optimized for genetic characterization for three pathogenic variants of Xanthomonas axonopodis pv. citri (Xac) from Taiwan. These three novel variants of atypical symptom–producing X. axonopodis pv. citri were designated as Xac‐Af, Xac‐Ap and Xac‐Ar. Based on polymerase chain reaction (PCR) with primers specific to X. axonopodis pv. citri, leucine‐responsive regulatory protein (lrp) gene assay and DNA fingerprintings generated by repetitive‐sequence PCR (rep‐PCR) and amplified fragment length polymorphism (AFLP) were used to compare strains including the three types of atypical symptom–producing strains Xac‐Af, Xac‐Ap and Xac‐Ar, and additional reference strains from pathotypes Xac‐A, Xac‐A*, Xac‐Aw, X. axonopodis pv. auruantifolii and X. axonopodis pv. citrumelo. These three types of X. axonopodis pv. citri variants can be detected with six sets of primer specific for X. axonopodis pv. citri. Cluster analyses by lrp sequence assay, AFLP and combing the band patterns of rep‐PCR clearly grouped the atypical symptom–producing variants in types Xac‐ Af, Xac‐Ar and Xac‐Ap into the same cluster with typical symptom‐producing strains in pathotype Xac‐A. These three types of X. axonopodis pv. citri variants could be excluded from strains of Xac‐A* and Xac‐Aw in these genotypic analyses. Strains of Xac‐A* and Xac‐Aw were closely related to Xac‐A strains in our results. No Taiwan isolate was related to X. axonopodis pv. auruantifolii or X. axonopodis pv. citrumelo. The results further confirmed the atypical symptom–producing variants of X. axonopodis pv. citri in Taiwan belong to pathotype Xac‐A.  相似文献

The biochemical rationale for the inhibition of citric acid fermentation by Aspergillus niger in the presence of Mn2+ ions has been investigated using high citric acid-yielding, Mn2+ ion-sensitive as well as Mn2+ ion-tolerant mutant strains of A. niger. In the presence of Mn2+ (1.5 mg/l), citric acid production by the Mn2+ ion-sensitive strain (KCU 520) was reduced by about 75% with no apparent effect on citric acid yield by the Mn2+ ion-tolerant mutant strain (GS-III) of A. niger. The significantly increased level of the Mn2+ ion-requiring NADP+-isocitrate dehydrogenase activity in KCU 520 cells and the lack of effect on the activity level of the enzyme in GS-III mutant cells by Mn2+ ions during fermentation seem to be responsible for the Mn2+ ion inhibition of citric acid production by the KCU 520 strain and the high citric acid yield by the mutant strain GS-III of A. niger even in the presence of Mn2+.  相似文献

The efficiency of different FinnishFrankia strains as symbionts onAlnus incana (L.) Moench was evaluated in inoculation experiments by measuring nitrogen fixation and biomass production. Since all available pure cultures ofFrankia are of the Sp type (sporangia not formed in nodules), but the dominant nodule endophyte ofA. incana in Finland is of the Sp+ type (sporangia formed in nodules), crushed nodules of thisFrankia type were included. The Sp pure cultures, whether originating fromA. incana orA. glutinosa, produced with one exception, similar biomass withA. incana. The highest biomass was produced with an American reference strain fromA. viridis crispa. Using Sp+ nodule homogenates fromA. incana as inoculum, the biomass production was only one third of that produced by Sp pure cultures from the same host. Hence, through selection of the endophyte it is possible to exert a considerable influence on the productivity ofAlnus incana.  相似文献

Summary We have initiated a cytogenetic analysis of chromosome region 89A of Drosophila melanogaster by isolating a set of radiation-induced mutations causing loss of function of P[(w)B]1-1, a transposon bearing the white locus inserted in 89A. Complementation tests and cytological examination of these chromosomes identified four new deficiencies (Df(3R)Po 2, Df(3R)Po 3, Df(3R)Po 4 and Df(3R)c(3)G 2 ). The new deficiencies and three previously identified deficiencies (Df(3R)sbd 26, Df(3R)sbd 45 and Df(3R)sbd 105) were tested for the ability to complement mutations in the enzyme loci Po and Aldox-1, the indirect flight muscle genes Tm2 and act88F, the morphological mutations jvl, sbd 2 and Sb, the vital loci srp, pnr and mor, and a newly described vital locus l(3)89Aa. We also used linkage analysis to determine the order and relative positions of P[(w)B]1-1 and an independent transposon insertion, P[w+]21, with respect to cv-c, Po, Aldox-1 and sbd 2. Cytological examination of the deficiencies and analysis of the transformed lines by in situ hybridization permits the correlation of genetically defined regions with specific polytene chromosome bands. A revised cytogenetic map of the 8817–8913 region is presented.  相似文献

Summary A mutational alteration either in adenylate cyclase (cya -) or in cyclic-35-AMP (cAMP) receptor protein (crp -) rendered Salmonella typhimurium incapable of producing flagella. The amount of mRNA specific for flagellin in these mutants was almost negligible when assayed in an in vitro protein synthesizing system. A secondary mutation, cfs, partially suppressing the cya - mutation, was identified among the revertants of cya -. A mutation in the same cistron as cfs resulted in a non-flagellate phenotype either by itself or in combination with cfs. The cistron, which was given the gene symbol flaT, was located between flaE and flaL. It was suggested that cAMP receptor protein together with cAMP modulates the gene flaT, which in turn acts as a positive effector on the synthesis of active mRNA specific for flagellin.  相似文献

Summary Telve chloramphenicol resistant (CM r)-mutants were isolated from B. subtilis ATCC 6633 and were classified into the following six groups. Group I. No 50s ribosomal protein change was detectable. Ribosomes did not show alteration of the binding ability to CM or to erythromycin in vitro. Group II. A 50s protein, 50a, was altered. Ribosomes did not show alteration of the binding ability to CM or to erythromycin in vitro. The genes specifying the 50a protein was in the cysA-str region on B. subtilis chromosome. Group III. A 50s protein, 50b, was altered. Biological properties of the ribosomes were the same as Group I or II so fas as examined. The genes for 50b protein was in the cysA-str region. Group IV. A 50s protein, 50c, was altered. Ribosomes showed a definite decrease in ability to bind to CM in vitro. The binding of erythromycin to the ribosomes was not impaired. The chromosomal locus of the CM r (and for 50c protein) was in the cysA-str region. Group V. A 50s protein, 50e, was changed. The ability of the ribosomes to bind in vitro both to CM and to erythromycin was greatly reduced. The genetic locus of the CM r (and for 50e protein) was in the cysA-str region. Group VI. A 50s protein, 50f, was altered. Ribosomes showed a decrease in ability to bind in vitro both to CM and to erythromycin. The genes for 50f protein was in the cysA-str region.The results suggest that the ribosomal resistance to CM may be caused by an independent change of at least several 50s ribosomal protein species. The genetic data shown here and those reported previously show that at least two 30s and seven 50s ribosomal protein genes are situated in the cysA-str region on B. subtilis chromosome.  相似文献

以生长在我国荒漠和半荒漠地区的特有植物沙蒿为供体植物,沙米、虫实、草木樨状黄芪、狗尾草为受体植物,研究了沙蒿根、茎、叶、种子水浸提液对受体植物种子发芽、幼苗生长以及根对NH~+_4和K~+吸收的化感效应的差异,为沙区植被恢复中伴生杂草种类的选择提供理论依据。结果表明:沙蒿水浸提液对4种受体植物种子发芽和幼苗生长均具有显著的化感效应,沙米、虫实、草木樨状黄芪表现为抑制作用,狗尾草表现为促进作用。不同部位沙蒿浸提液的化感作用不同,根浸提液对沙米、虫实、狗尾草的化感效应最强,茎浸提液对草木樨状黄芪的抑制最强。不同植物种及植物的不同发育时期对化感作用的敏感性也不同,在4种受体植物中,沙米受抑制最强,其次为虫实、草木樨状黄芪,且随浸提液浓度增加而显著增强,而狗尾草具有一定的耐受力,表现为促进作用。沙蒿浸提液对受体植物根长的影响最强,其次是种子发芽、苗高和幼苗干质量。浸提液显著降低沙米、虫实、草木樨状黄芪根对NH~+_4和K~+的吸收,增加狗尾草根对NH~+_4和K~+的吸收。研究结果表明雨水淋溶是沙蒿向环境中释放化感物质的途径之一,化感作用在增强沙蒿生存竞争力,扩大种群过程中起着不容忽视的作用,可能是沙蒿单优势种群落形成的原因之一。  相似文献

Hafnia alvei 5-5, isolated from a soil-litter mixture underneath the canopy of the nickel-hyperaccumulating tree Sebertia acuminata (Sapotaceae) in New Caledonia, was found to be resistant to 30 mM Ni2+ or 2 mM Co2+. The 70-kb plasmid, pEJH 501, was transferred by conjugation to Escherichia coli, Serratia marcescens, and Klebsiella oxytoca. Transconjugant strains expressed inducible nickel resistance to between 5 and 17 mM Ni2+, and cobalt resistance to 2 mM Co2+. A 4.8-kb SalEcoRI fragment containing the nickel resistance determinant was subcloned, and the hybrid plasmid was found to confer a moderate level of resistance to nickel (7 mM Ni2+) even to E. coli. The expression of nickel resistance was inducible by exposure to nickel chloride at a concentration as low as 0.5 mM Ni2+. By random TnphoA′-1 insertion mutagenesis, the fragment was shown to have structural genes as well as regulatory regions for nickel resistance. Southern hybridization studies showed that the nickel-resistance determinant from pEJH501 of H. alvei 5-5 was homologous to that of pTOM9 from Alcaligenes xylosoxydans 31A. Succinic acid-producing Anaerobiospirillum succiniciproducens was anaerobically grown in a glycerol-fed continuous bioreactor in order to investigate the physiological responses of the cell to different pH values (5.9, 6.2, or 6.5) and various dilution rates, D. In these experiments, A. succiniciproducens showed a pH-dependent glycerol consumption behavior. When pH was maintained at 5.9 or 6.5, glycerol started to accumulate even at a very low D of 0.027 h−1. Succinic acid yield was not significantly affected by the pH of the culture or the Ds. However, more acetic acid formation was observed when the growth rate of A. succiniciproducens was fast on glycerol at pH 6.2 (at D ≥ 0.15 h−1). The highest obtainable succinic acid/acetic acid ratio was 40:1, which was 10 times higher than that obtained by batch cultures grown on glucose. The maximum obtainable productivity of succinic acid was 2.1 g L−1 h−1), which was 14 times higher than that obtained by batch culture.  相似文献

A strong biomass increase of two Anabaena species was observed in natural plankton community enclosed into nine large mesocosms (51 m3) and manipulated with mineral nutrients and an organic carbon source during a 3‐week period in the coastal Baltic Sea. The water column and settled material from the bottom of the mesocosms were sampled at 2‐day intervals. Planktonic populations of Anabaena lemmermannii Richter and A. cylindrica Lemmermann and sedimentation rates of akinetes to the bottom were quantified. Comparing mesocosms with artificially induced nitrogen and phosphorus limitation, we found that during the third week of the experiment, the population size of A. lemmermannii was clearly higher in nitrogen‐limited units (by a factor of 2.4), whereas the production rate of akinetes was higher in the phosphorus‐limited units (by a factor of 2.5). Input of freshly produced A. lemmermannii akinetes to the benthos was on average 15 × 106 and 6 × 106 cells· m?2·d?1 in the P? and N? limited mesocosms, respectively. Our estimates of specific akinete production rate of A. lemmermannii in P? and N? limited mesocosms revealed an even larger divergence (a factor of 5.5), being on average 2.4 and 0.4 akinetes·10?3 vegetative cells?1·d?1, respectively. The phosphorus addition effectively reduced akinete production of A. lemmermannii. Differences in the nutrient manipulation had no apparent effect on the biomass and akinete production of A. cylindrica. The akinete production pattern of A. cylindrica revealed a 1‐week delay compared with the vegetative population peak, whereas such a delay was not obvious in A. lemmermannii.  相似文献

Metronidazole (Mtz) is a critical component of combination therapies that are used against Helicobacter pylori, the major cause of peptic ulcer disease. Many H. pylori strains are Mtz resistant (MtzR), however, and here we show that MtzR results from loss of oxygen-insensitive NADPH nitroreductase activity. The underlying gene (called ‘rdxA’) was identified in several steps: transformation of Mtz-susceptible (MtzS) H. pylori with cosmids from a MtzR strain, subcloning, polymerase chain reaction (PCR) and DNA sequencing. We also found that (i) E. coli (normally MtzR) was rendered MtzS by a functional H. pylori rdxA gene; (ii) introduction of rdxA on a shuttle vector plasmid into formerly MtzRH. pylori rendered it MtzS; and (iii) replacement of rdxA in MtzSH. pylori with an rdxA::camR null insertion allele resulted in a MtzR phenotype. The 630 bp rdxA genes of five pairs of H. pylori isolates from infections that were mixed (MtzR/MtzS), but uniform in overall genotype, were sequenced. In each case, the paired rdxA genes differed from one another by one to three base substitutions. Typical rdxA genes from unrelated isolates differ by ≈ 5% in DNA sequence. Therefore, the near identity of rdxA genes from paired MtzR and MtzS isolates implicates de novo mutation, rather than horizontal gene transfer in the development of MtzR. Horizontal gene transfer could readily be demonstrated under laboratory conditions with mutant rdxA alleles. RdxA is a homologue of the classical nitroreductases (CNRs) of the enteric bacteria, but differs in cysteine content (6 vs. 1 or 2 in CNRs) and isoelectric point (pI = 7.99 vs. 5.4–5.6), which might account for its reduction of low redox drugs such as Mtz. We suggest that many rdxA (MtzR) mutations may have been selected by prior use of Mtz against other infections. H. pylori itself is an early risk factor for gastric cancer; the possibility that its carcinogenic effects are exacerbated by Mtz use, which is frequent in many societies, or the reduction of nitroaromatic compounds to toxic, mutagenic and carcinogenic products, may be of significant concern in public health.  相似文献

Summary A bacterial mutant is described which restricts bacteriophage amber suppressor psu +. Restriction, probably, operates at a translational level. The new strain provides the system of identification of bacteriophage amber mutants suppressed by psu + using suppressornegative (su -) bacterial strains.  相似文献
