Abstract

Studies that allow computing values of aqueous proton dissociation constants (pKa), gas phase proton affinities, and the free energy of solvation have been performed for six members of angiotensin-I-converting enzyme (ACE) inhibitor family (captopril, enalaprilat, imidaprilat, ramiprilat, perindoprilat, and spiraprilat). Density functional theory (DFT) calculations using PBE1PBE functional on optimized molecular geometries have been carried out to investigate the thermodynamics of gas-phase protonation. The conductor-like polarizable continuum model (CPCM) solvation method at various levels of theory was applied to calculate the free energy of solvation for the ACE inhibitors and their respective anions. The CPCM solvation calculations were performed on both gas-phase and solvent-phase optimized structures. The combination of gas-phase and solvation energies according to the thermodynamic cycle enabled us to compute accurate pKa values for the all studied molecules.  相似文献

【目的】甲烷氧化细菌(MOB)长期以来一直被用作石油和天然气勘探的重要油气指示菌,其仅能利用甲烷作为唯一碳源。根据甲烷氧化菌菌群特征结合地质剖面可以较好地预测深部油气藏,为石油勘探提供良好的数据支撑。由于传统平板培养法只能针对可培养甲烷氧化菌,方法具有一定局限性。【方法】本文采用分子生物学技术结合地球化学烃类指标研究了顺北典型油气藏上方土壤中甲烷氧化菌的分布。【结果】研究结果显示,油气田上方pmoA基因拷贝数与酸解烃含量具有一定的正相关性,且油气区比背景区高0.5–2个数量级。16SrRNA基因高通量测序和pmoA基因的克隆文库结果显示顺北油藏上方土壤中甲烷氧化菌主要以I型为主,水平剖面中甲烷氧化菌随着离油田距离增加存在I型向II型演变的现象,且Methylomonassp.在背景区与油气区的丰度有较大差异,具有良好的油气指示潜力。【结论】综上所述,长期微渗透过程中甲烷氧化菌(MOB)的菌群特征对预测深层油藏具有一定的指示作用,结合地质剖面和地表烃类可以有效预测有利油藏区域。  相似文献

 A two-step concerted mechanism for the conversion of methane to methanol catalyzed by soluble methane monooxygenase (sMMO) is discussed. We propose that the enzymatic reaction mechanism is essentially the same as that of the gas-phase methane-methanol conversion by the bare FeO+ complex. In the initial stage of our mechanism, the ferryl (Fe—O) "iron" active site of intermediate Q and substrate methane come into contact to form the initial Q (CH4) complex with an OFe—CH4 bond. The C—H bonds of methane are significantly weakened by the formation of a five-coordinate carbon species, through orbital interactions between a C 3v - or D 2d -distorted methane and the Fe—O active site. The important transition state for an H atom abstraction exhibits a four-centered structure. The generated intermediate involves an HO—Fe—CH3 moiety, and it is then converted into the final product complex including methanol as a ligand through a methyl migration that occurs via a three-centered transition state. The two-step concerted mechanism is consistent with recent experiments on regioselectivity of enzyme-catalyzed alkane hydroxylations. The molecular species of legumin from broad bean seeds exhibit wide heterogeneity [S. Utsumi and T. Mori, Biochim. Biophys. Acta, 621, 179 (1980)]. The subunit compositions of these molecular species were analyzed according to the following procedure: the molecular species were separated by polyacrylamide gel electrophoresis, and then their subunit compositions were analyzed by sodium dodecyl sulfate-polyacrylamide gel electrophoresis. The results indicated that five groups with nine kinds of submolecular species having different molecular weights and subunit compositions were present as legumin in the broad bean. According to these results, we have demonstrated the presence of another group consisting of two kinds of submolecular species, in addition to the four groups and seven kinds of submolecular species reported previously. Various possible molecular species of legumin composed of subunit groups classified according to size were presented.  相似文献

【目的】基于肠道微生物与宿主代谢的相互关系,研究不同配方的益生菌对小鼠肥胖的影响。【方法】50只C57BL/6J雄性小鼠随机平均分成10组,分别给予正常饲料、高脂饲料以及高脂饲料加8种不同配方的益生菌产品(50亿CFU/只),所有动物连续喂养9周,每周测量小鼠体重1次。最后一周测定空腹血糖、葡萄糖耐量试验(glucose tolerance test,GTT)、血脂相关指标,称取内脏重量,并留取小鼠盲肠内容物,提取小鼠肠道菌群总DNA,利用16S rDNA测序检测相关细菌含量。【结果】部分益生菌可引起小鼠体重增速加快,而部分益生菌可减缓小鼠肥胖和降低内脏脂肪重量,同时缓解高血脂症。丹尼斯克品牌益生菌配方组小鼠肠道中厚壁菌/拟杆菌比例(F/B)是正常饮食组的22.8倍,Akkermansia muciniphila(Akkermansia)细菌含量几乎为0;而菌拉丁品牌益生菌配方组小鼠F/B比例与正常饲料饮食组类似,Akkermansia含量为0.5%,为正常饮食对照组小鼠的一半左右。【结论】益生菌可影响小鼠体重和代谢,但不同配方的益生菌效果截然相反。特定的益生菌配方对肥胖和高血脂的改善可能是由于其选用的菌株本身的特性以及菌株之间的相互配比能够降低小鼠肠道中F/B比例以及升高Akkermansia的含量所带来的。此研究为进一步开发可改善代谢的益生菌产品提供了参考。  相似文献

Abstract

The semi-empirical thermodynamic model for the binding of metal ions to an emulsion (intralipid 20%) reported previously (Hall et al., 1991 ; Gaskin et al., 1993) is incorporated into a thermodynamic computer model. This permits the zeta potential and emulsion stability together with precipitate formation to be estimated for any intravenous nutrition regimen. A regimen frequently used in the intravenous nutrition of patients is considered in this modeling study. The effect of solution pH and calcium on the zeta potential of the emulsion is predicted. Calcium and magnesium are the only metal cations which are predicted to be of importance when considering stability of this emulsion.  相似文献

Question: How well can mortality probabilities of deciduous trees(Fagus sylvatica) and conifers (Abies alba) be predicted using permanent plot data that describe growth patterns, tree species, tree size and site conditions? Location: Fagus forests in the montane belt of the Jura folds (Switzerland). Method: Permanent plot data were used to develop and validate logistic regression models predicting survival probabilities of individual trees. Backward model selection led to a reduced model containing the growth‐related variable ‘relative basal area increment’ (growth‐dependent mortality) and variables not directly reflecting growth such as species, size and site (growth‐independent mortality). Results: The growth‐mortality relationship was the same for both species (growth‐dependent mortality). However, species, site and tree size also influenced mortality probabilities (growth‐independent mortality). The predicted survival probabilities of the final model were well calibrated, and the model showed an excellent discriminatory power (area under the receiver operating characteristic curve = 0.896). Conclusion: Mortality probabilities of Fagus sylvatica and Abies alba can be predicted with high discriminatory power using a well calibrated logistic regression model. Extending this case study to a larger number of tree species and sites could provide species‐ and site‐specific tree mortality models that allow for more realistic projections of forest succession.  相似文献

Capsule Woodland birds were significantly less likely to occur in gardens in years of high beechmast crop.

Aim To test the hypothesis that woodland species that feed on beechmast will have significantly lower occurrence rates at garden feeders in mast years.

Methods Weekly winter occurrence rates at garden feeders between 1970/71 and 1999/2000 for 40 species were analysed in relation to annual beechmast abundance, classified into low, medium and high years. A repeated-measures logistic regression model was used to assess whether beechmast abundance explained further significant variation additional to underlying seasonal and annual trends.

Results Seven species that commonly feed on beechmast showed significantly lower occurrence in gardens in years of highest beechmast abundance: Great Spotted Woodpecker Dendrocopos major, Woodpigeon Columba palumbus, Great Tit Parus major, Coal Tit Periparus ater, Nuthatch Sitta europaea, Jay Garrulus glandarius and Chaffinch Fringilla coelebs. Blackbird Turdus merula and Siskin Carduelis spinus, which showed similar significant patterns, are likely to take beechmast as elements of their diet. Pied Wagtail Motacilla alba was the only insectivorous species to show significant effects, but occurrence was lowest in years of intermediate beechmast abundance. For the latter species, this may have been due to confounding effects of temperature, but there were no such confounding effects of either temperature, or the number of bird feeders provided in gardens, for the other nine species.

Conclusion Use of artificial food sources by birds in gardens is influenced by resources in the surrounding countryside, suggesting that food provided in gardens may play a significant part in the population dynamics of these species, that population monitoring without consideration of the garden habitat may be deficient, and that volunteer-based garden bird recording may provide data that can be used as an indicator of changes in the wider countryside.  相似文献

In response to the increased concern over agriculture’s contribution to greenhouse gas (GHG) emissions, more detailed assessments of current methane emissions and their variation, within and across individual dairy farms and cattle, are of interest for research and policy development. This assessment will provide insights into possible changes needed to reduce GHG emissions, the nature and direction of these changes, ways to influence farmer behavior and areas to maximize the adoption of emerging mitigation technologies. The objectives of this study were to (1) quantify the variation in enteric fermentation methane emissions within and among seasonal calving dairy farms with the majority of nutritional requirements met through grazed pasture; (2) use this variation to assess the potential of new individual animal emission monitoring technologies and their impact on mitigation policy. We used a large database of cow performance records for milk production and survival from 2 398 herds in New Zealand, and simulation to account for unobserved variation in feed efficiency and methane emissions per unit of feed. Results showed an average of 120 ± 31.4 kg predicted methane (CH4) per cow per year after accounting for replacement costs, ranging 8.9–323 kg CH4/cow per year. Whereas milk production, survival and predicted live weight were reasonably effective at predicting both individual and herd average levels of per cow feed intake, substantial within animal variation in emissions per unit of feed reduced the ability of these variables to predict variation in per animal methane output. Animal-level measurement technologies predicting only feed intake but not emissions per unit of feed are unlikely to be effective for advancing national policy goals of reducing dairy farming enteric methane output. This is because farmers seek to profitably utilize all farm feed resources available, so improvements in feed efficiency will not result in the reduction in feed utilization required to reduce methane emissions. At a herd level, average per cow milk production and live weight could form the basis of assigning a farm-level point of obligation for methane emissions. In conclusion, a comprehensive national database infrastructure that was tightly linked to animal identification and movement systems, and captured live weight data from existing farm-level recording systems, would be required to make this effective. Additional policy and incentivization mechanisms would still be required to encourage farmer uptake of mitigation interventions, such as novel feed supplements or vaccines that reduce methane emissions per unit of feed.  相似文献

Acidic wetlands of the northern hemisphere are an important source of methane, a major greenhouse gas. The taxonomic identity of the aerobic methanotrophic bacteria, which colonize these environments and reduce the potential flux of methane to the atmosphere, has remained elusive for a long time. Both cultivation-independent molecular approaches and cultivation-based studies have been used to identify methanotrophs in this acidic habitat. It was shown that acidic peat is colonized mainly by methanotrophic representatives of the Alphaproteobacteria: Methylocystis spp., Methylocella spp. and Methylocapsa spp. Novel methanotrophic isolates from acidic wetlands display a number of unique characteristics and metabolic traits including unusual cell ultrastructure and fatty acid composition, ability to utilize some multicarbon compounds as growth substrates, and new regulatory mechanisms of methane oxidation. Several other methanotroph populations, which have been detected in acidic peat by molecular approaches but have so far eluded isolation, represent a challenge for further cultivation studies.  相似文献

Microalgae are a promising resource for the highly sustainable production of various biomaterials (food and feed), high‐value biochemicals, or biofuels. However, factors influencing the valued lipid production from oleaginous algae require a more detailed investigation. This study elucidates the variations in lipid metabolites between a marine diatom (Cylindrotheca closterium) and a freshwater green alga (Scenedesmus sp.) under nitrogen starvation at the molecular species level, with emphasis on triacylglycerols using liquid chromatography–electrospray ionization mass spectrometry techniques. A comprehensive analysis was carried out by comparing the changes in total lipids, growth kinetics, fatty acid compositions, and glycerolipid profiles at the molecular species level at different time points of nitrogen starvation. A total of 60 and 72 triacylglycerol molecular species, along with numerous other polar lipids, were identified in Scenedesmus sp. and C. closterium, respectively, providing the most abundant triacylglycerol profiles for these two species. During nitrogen starvation, more triacylglycerol of Scenedesmus sp. was synthesized via the “eukaryotic pathway” in the endoplasmic reticulum, whereas the increase in triacylglycerol in C. closterium was mainly a result of the “prokaryotic pathway” in the chloroplasts after 96 h of nitrogen starvation. The distinct responses of lipid synthesis to nitrogen starvation exhibited by the two species indicate different strategies of lipid accumulation, notably triacylglycerols, in green algae and diatoms. Scenedesmus sp. and Cylindrotheca closterium could serve as excellent candidates for the mass production of biofuels or polyunsaturated fatty acids for nutraceutical purposes.  相似文献

Abstract. A method is given for measuring two components of error (rate and direction) in predictions of compositional change through time. Observed compositional change between two times can be represented as a vector between two points in multidimensional species space. The point at the tail of this vector is the species composition at one particular time. A vector of predicted compositional change will diverge from the vector of observed change to some degree. The error in the predicted rate of change is measured by the difference between the lengths of the two vectors. The error in the predicted direction of change is measured by the angle between the vectors. The cosine of this angle is a non-standardized correlation coefficient (rn) between the predicted and observed species compositions. The quantity 1 - rn2 measures the error in direction of the predicted dynamics without being influenced by the overall rate of change. These measures in Euclidean space have useful counterparts in city-block space. The method is illustrated by comparing actual long-term changes in Midwestern old-growth forests with the changes predicted by a growth and yield model, TWIGS.  相似文献

【目的】以不同密度等级大柳塔长焰煤作为产气底物,前期驯化培养厌氧菌群进行生物模拟产气实验,研究不同密度等级煤中的灰分和无机矿物对生物产气的影响。【方法】利用小浮沉将大柳塔长焰煤分成不同密度等级的煤样,采用工业分析、XRD、XRF分析小浮沉处理得到煤样的理化性质,利用这些煤样进行生物产气模拟实验,以甲烷产量作为评价指标,分析不同密度等级煤样中灰分对产气的影响。最后,通过添加几种标准矿物方式比较了煤中无机矿物对生物产气的可能影响。【结果】不同密度等级煤样中灰分对产气量存在一般显著影响(P=0.035),且灰分与甲烷含量呈负相关关系,其灰分中的无机矿物如高岭土、菱铁矿、氧化亚铁镁等的积累对产气有抑制作用。不同矿物配比产气实验证实低含量的粘土矿物促进甲烷的生成,高含量的粘土矿物抑制产气。【结论】不同密度等级煤中的灰分对生物产气存在一般显著的影响,高灰分煤的产气量低,而低灰分煤的产气量高。  相似文献

【目的】通过体外静态模拟瘤胃发酵法研究溶菌酶对瘤胃发酵、甲烷生成及微生物菌群结构的影响。【方法】采用单因素多水平试验设计,溶菌酶添加水平分别为0(L-0,对照组)、0.1 mg/100 m L(L-0.1)、1 mg/100 m L(L-1)、10 mg/100 m L(L-10)和100 mg/100 m L(L-100),定时测定产气量和甲烷产量,培养24 h后,发酵液用于发酵参数和微生物菌群数量的q PCR测定,其中L-0、L-1和L-100三个组发酵液同时进行16S r RNA基因Illumina高通量测序。【结果】与对照组相比,低剂量溶菌酶添加(L-0.1组)不影响甲烷产量、氨氮浓度、干物质消失率、有机物消失率和总挥发性脂肪酸等瘤胃发酵参数(P0.05);随着剂量提高,L-1处理组甲烷产量、氨氮浓度显著降低(P0.05),丙酸浓度显著增加(P0.05),并且干物质消失率、有机物消失率和总挥发性脂肪酸不受影响(P0.05);而较高剂量组(L-10和L-100组)虽然甲烷产量显著降低,丙酸浓度显著增加(P0.05),但干物质消失率和有机物消失率也显著降低(P0.05)。q PCR结果显示高剂量组(L-100组)总菌、原虫、甲烷菌数量与对照组相比显著降低(P0.05),而L-0.1、L-1和L-10组总菌、真菌和原虫数量与对照组相比均无显著变化(P0.05)。高通量测序主成分分析(PCA)显示对照组与溶菌酶添加组间瘤胃细菌组成的明显区分,说明添加溶菌酶显著改变了瘤胃细菌菌群结构。溶菌酶通过增加月形单胞菌和琥珀酸弧菌等丙酸生成菌的相对丰度,使更多的氢被用于生成丙酸,导致甲烷产量降低;溶菌酶可抑制普雷沃氏菌和拟杆菌属等蛋白降解菌的生长,进而减少蛋白质过度降解,降低氨氮浓度。【结论】添加适宜浓度(1 mg/100 m L)的溶菌酶可通过调控瘤胃微生态改变瘤胃发酵模式,降低瘤胃甲烷和氨的生成,短期内并不影响饲料消化。  相似文献

Background: There is a growing interest in understanding the gas exchange between the atmosphere and seasonally snow-covered regions, especially in light of projections that climate change will alter the timing and extent of seasonal snow cover. In snow-covered ecosystems, gas fluxes are due both to microbial activity in the snow-covered soils and to chemical and physical reactions with the various gases and/or dissolved constituents in the snowpack. Niwot Ridge, in the Colorado Rocky Mountains, has one of the most extensive sets of measurements of winter gas exchange globally.

Aims: Our goal was to examine the temporal patterns and environmental controls on Niwot Ridge of gas fluxes for gases with different sources and sinks.

Methods: Here, we review the concentrations and fluxes that have been measured for carbon dioxide, nitrous oxide, methane, nitrogen oxides, ozone, gaseous elemental mercury and volatile organic carbon compounds.

Results and Conclusions: We looked for similarities and differences among the gases, but in many cases, the origin, fate and controls of these fluxes still need to be determined. However, we believe that many of the biologically driven reactions are the result of exponential growth of a winter microbial community during the long period of stable environmental conditions under the seasonal snowpack.  相似文献

AMethanosarcina species, designated strain ChGul, was isolated from goat feces; this is the first fully described pure culture ofMethanosarcina obtained from feces. Antigenic fingerprinting suggests that isolate ChGul is a new immunotype. The mol% G + C content of DNA was 42.2%. Strain ChGul grew on methanol, methylamines, and acetate in a minimal salts medium. It grew on H2-CO2 only after adaptation. Growth occurred as a milky-white suspension and contained cells mostly in doublets and quadruplets of irregular cocci; many cells contained phase bright spots typical of gas vacuoles. The isolate did not grow on formate, or CO2 plus isopropanol, ethanol, or acetone as substrates and did not produce methane from formate. The optimum growth temperature was 35–37°C, and optimum pH was 6.2–6.8. ChGul is unusually sensitive to sulfide and has low tolerance for NaCl. Optimal levels of total sulfide and NaCl for growth were 0.5 mM and 20–40 mM, respectively. Since ChGul requires adaptation for growth on H2-CO2 and cannot use formate, it may be restricted to methylotropic or acetoclastic methanogenesis in the rumen, a function not observed in previously isolated rumen methanogens that use H2-CO2 and formate. Our work suggests that improper NaCl and sulfide concentrations, and cell lysis, may have made isolation of rumenMethanosarcina difficult in the past. It also underscores the need to evaluate feed compositions and media components for most probable number studies, with respect to NaCl and sulfide levels, to understand the role ofMethanosarcina in the rumen.  相似文献

Synopsis Stunting in freshwater fish populations may be due to low availability of one or more prey components within their diet. If the limiting prey constitute a trophic level (i.e. zooplankton, benthos, or fish), we define the phenomenon as a trophic bottleneck. Growth of a non-stunted population of yellow perch, Perca flavescens, was simulated using an allocation model based on reported ontogenetic shifts in diet (planktivory to benthivory to piscivory). The model was then perturbed by limiting the available ration individually for each of the three feeding types. The resulting simulated growth curves all reflected lower growth rates than the unperturbed state and the shape of the curve differed between the limited food types. Only the reduced benthic ration produced a simulated growth that matched that observed for the stunted yellow perch of Lac Hertel (Québec). To test the hypothesis that benthic ration was limiting growth in Lac Hertel, all the fish species in the lake were sampled for diet composition and total length at age. We predicted that species feeding upon benthic invertebrates should exhibit the slow growth characteristic of stunting (based on the results of the allocation model). Of the seven fish species found in Lac Hertel, four were stunted: yellow perch, pumpkinseed, Lepomis gibbosus, rock bass, Ambloplites rupestris, and brown bullhead, Ictalurus nebulosus. Three species were non-stunted: northern pike, Esox lucius, golden shiner, Notemigonus crysoleucas, and white sucker, Catostomus commersoni. All stunted species fed on benthic invertebrates and all non-stunted species, except the white sucker, did not feed upon benthos. The prediction of the allocation model was thus supported.  相似文献

【目的】研究特基拉芽孢杆菌挥发物2-甲基丁酸对甘薯长喙壳菌的抑制作用,评价2-甲基丁酸对甘薯黑斑病的防治效果。【方法】采用I-分隔平皿和气相抑菌体系,研究不同剂量的2-甲基丁酸对甘薯长喙壳菌菌丝生长和孢子萌发的抑制作用;使用乳酸酚棉蓝染色观察2-甲基丁酸对甘薯长喙壳菌显微形态的影响;利用荧光探针钙荧光白和溴化丙锭检测2-甲基丁酸对甘薯长喙壳菌细胞壁结构与细胞膜通透性的影响;使用荧光探针2,7-二氯荧光素二乙酸酯检测甘薯长喙壳菌胞内活性氧含量变化;测定谷胱甘肽含量分析病原菌应对氧化损伤能力的改变;通过线粒体脱氢酶活力和丙酮酸含量的检测,分析2-甲基丁酸对甘薯长喙壳菌线粒体功能和能量代谢的影响;评价2-甲基丁酸在甘薯黑斑病防治中的使用效果。【结果】2-甲基丁酸显著抑制甘薯长喙壳菌菌丝生长和孢子萌发,降低其产孢能力,导致菌丝折叠弯曲并形成不连续的空腔。2-甲基丁酸使甘薯长喙壳菌的细胞壁结构改变,细胞膜通透性增加,胞内活性氧含量升高,谷胱甘肽含量显著降低,使病原菌应对氧化损伤的能力下降,线粒体脱氢酶活力和丙酮酸含量显著降低,诱发线粒体功能障碍,干扰细胞能量代谢,最终导致细胞死亡。此外,2-甲基丁酸对甘薯黑斑病也具有良好的防治作用。【结论】2-甲基丁酸对甘薯长喙壳菌具有显著的抑制作用,可作为安全高效的气相抑菌材料用于新型熏蒸制剂的研发。  相似文献

ABSTRACT

Background: According to modern coexistence theory, ecologically similar species can coexist if fitness differences between them are small, or niche differences between them are large. However, these predictions have not been tested extensively in real systems and are difficult to examine in traits-based studies.

Aims: The aim of our study was, by using the carnivorous pitcher plant genus Nepenthes as a model system, to examine (1) species growth ranks as a proxy for fitness; (2) modern coexistence theory at a geographical scale; (3) evidence for pitcher trait-mediated resource partitioning between sympatric Nepenthes species.

Methods: We used growth ranks, obtained from a survey of experienced Nepenthes nursery owners, as a proxy for fitness. Multivariate pitcher-, vegetative- or combined-trait distances, computed from morphometric-trait data from the literature were used as proxies for niche differences. Data on global Nepenthes species-pair sympatry was modelled against these fitness and niche differences.

Results: Niche and growth rate differences were positively and negatively correlated with sympatry, respectively, as expected from theory.

Conclusion: Our results agree with theory and suggest that fitness differences can be approximated from average species growth. Prey- and microhabitat-resource partitioning, operating through divergent pitcher and vegetative traits respectively, are likely mechanisms that stabilise the coexistence of sympatric Nepenthes species.  相似文献
