Summary This paper reports the inhibitory effects of calmidazolium (CDZ), a calmodulin inhibitor, on electrical uncoupling by CO2. Membrane potential and coupling ratio (V 2/V1) are measured in two neighboring cells ofXenopus embryos (16 to 64 cell stage) for periods as long as 5.5 hr. Upon exposure to 100% CO2, control cells consistently uncouple even if the CO2 treatments are repeated every 15 min for 2.5 hr. CDZ (5×10–8–1×10–7 m) strongly inhibits uncoupling. The inhibition starts after 30, 50 and 60 min of treatment with 1×10–7, 7×10–8 and 5×10–8 m CDZ, respectively, is concentration-dependent and partially reversible. In the absence of CO2, CDZ also improves electrical coupling. CDZ has no significant effect on membrane potential and nonjunctional membrane resistance. These data suggest that calmodulin or a calmodulin-like protein participates in the uncoupling mechanism.  相似文献

Summary The effects of several sulfamoyl benzoic acid derivatives on Na–K–Cl cotransport were investigated in winter flounder intestine. The relative efficacy (IC50 values) and order of potency of these derivatives were benzmetanide, 5×10–8 m> bumetanide 3×10–7 m>piretanide 3×10–6 m>furosemide 7×10–6 m> amino piretanide 1×10–5 3-amino-4-penoxy-5-sulfamoyl benzoic acid. Binding of [3H] bumetanide was studied in microsomal membranes from winter flounder intestine and compared to that in bovine kidney outer medulla. Binding was also studied in brush-border membranes from winter flounder intestine. The estimated values forK d and number of binding sites (n) were: bovine kidney,K d =1.6×10–7,n=10.5 pmol/mg protein; winter flounder intestine,K d 1.2×10–7,n=7.3 pmol/mg protein, and brush-border membranes from winter flounder,K d =5.3×10–7,n=20.4 pmol/mg protein. The estimatedK d for bumetamide binding to winter flounder brush-border membranes derived from association and dissociation kinetics was 6.8×10–7 m. The similarity in magnitudes of IC50 andK d for bumetanide suggests that the brush-border cotransporter is ordinarily rate-limiting for transmural salt absorption and that bumetanide specifically binds to the cotransporter. Measurement of bumetanide binding at various concentrations of Na, K and Cl showed that optimal binding required all three ions to be present at about 5mm concentrations. Higher Na and K concentrations did not diminish binding but higher Cl concentrations (up to 100mm Cl) inhibited bumetanide binding by as much as 50%. Still higher Cl concentrations (500 and 900mm) did not further inhibit bumetanide binding. Scatchard analysis of bumetanide binding at 5 and 100mm Cl concentrations showed that bothK d andn were lower at the higher Cl concentration (5mm Cl:K d =5.29×10–7 m,n=20.4 pmol/mg protein; 100mm Cl:K d =2.3×10–7 m,n=8.8 pmol/mg protein). These data suggest two possibilities: that bumetanide and Cl binding are not mutually exclusive (in contrast to pure competitive inhibition) and that they each bind to separate sites or that two distinct bumetanide binding sites exist, only one of which exhibits Cl inhibition of binding. This inhibition would then be consistent with a competitive interaction with Cl.  相似文献

Summary Basal lateral membrane vesicles were isolated from rat intestinal epithelial cells. The sodium potassium triphosphatase (Na/K-ATPase) of these plasma membranes has been characterized by (1) the molecular weight of the phosphorylated intermediate, (2) the sensitivity of the phosphorylated intermediate to hydroxylamine, (3) its ouabain binding constants, and (4) its susceptibility to digestion by pronase. The phosphorylated intermediate was shown by SDS polyacrylamide gel electrophoresis to be a protein of 100,000 Daltons apparent mol wt. Its extensive hydrolysis in hydroxylamine demonstrated that it was an acyl phosphate. The isolated basal lateral membranes bound ouabain with a dissociation constant,K m (1.5×10–5 m), similar to the inhibitory constantK I (3×10–5 m), measured for ouabain inhibition of the Na/K-ATPase activity. The association rate constant measured for ouabain binding at 22°C was 1.3×103 m –1 sec–1 and is similar to the association rate constants reported for other tissues and species. The high dissociation rate constant, 3.6×10–2 sec–1, is consistent with the insensitivity of the rat to ouabain. Digestion of the intact cells by pronase yielded basal lateral membranes in which the Na/K-ATPase had been unaffected. The phosphorylated intermediate ran as a sharp band at 100,000 Daltons on electrophoresis, and the ouabain dissociation constant appeared to be unchanged. In these membranes, protein stains of polyacrylamide gels revealed digestion of the major high mol wt proteins including the major protein at 100,000 Daltons. This suggests that the Na/K-ATPase represent a minor component, less than 1%, of the basal lateral membrane protein. From these characteristics of the phosphorylated intermediate and the ouabain binding constants, we conclude that the Na/K-ATPase of the basal lateral membranes of rat intestinal epithelial cells is similar to that found in other tissues and species. Estimates of the number of pump sites and the turnover number predict rates of Na transport that are consistent with observed values.This paper is dedicated to the memory of Professor David H. Summary The transepithelial water permeability in frog urinary bladder is believed to be essentially dependent on the ADH-regulated apical water permeability. To get a better understanding of the transmural water movement, the diffusional water permeability (P d) of the basolateral membrane of urinary bladder was studied. Access to this post-luminal barrier was made possible by perforating the apical membrane with amphotericin B. The addition of this antibiotic increasedP d from 1.12±0.10×10–4 cm/sec (n=7) to 4.08±0.33×10–4 cm/sec (n=7). The effect of mercuric sulfhydryl reagents, which are commonly used to characterize water channels, was tested on amphotericin B-treated bladders. HgCl2 (10–3 m) decreasedP d by 52% andpara-chloromercuribenzoic acid (pCMB) (1.4×10–4 m) by 34%. The activation energy for the diffusional water transport was found to increase from 4.52±0.23 kcal/mol (n=3), in the control situation, to 9.99±0.91 kcal/mol (n=4) in the presence of 1.4×10–4 m pCMB. Our second approach was to measure the kinetics of water efflux, by stop-flow light scattering, on isolated epithelial cells from urinary bladders.pCMB (0.5 or 1.4×10–4 m) was found to inhibit water exit by 91±2%. These data strongly support the existence of proteins responsible for water transport across the basolateral membrane, which are permanently present.  相似文献

Summary The properties of an anion-selective channel observed in basolateral membranes of microdissected, collagenase-treated, cortical thick ascending limbs of Henle's loop from mouse kidney were investigated using patch-clamp single-channel recording techniques. In basal conditions, single Cl currents were detected in 8% of cell-attached and excised, inside-out, membrane patches whereas they were observed in 24% of cell-attached and 67% of inside-out membrane patches when tubular fragments were preincubated with Forskolin (10–5 m) or 8-bromo-cAMP (10–4 m) and isobutylmethylxanthine (10–5 m). The channel exhibited a linear current-voltage relationship with conductances of about 40 pS in both cell-attached and cell-free membrane configurations. AP Na + P Cl ratio of 0.05 was estimated in the presence of a 142/42mm NaCl concentration gradient applied to inside-out membrane patches. Anionic selectivity of the channel followed the sequence Cl>Br>No 3 F; gluconate was not a permeant species. The open-state probability of the channel increased with membrane depolarization in cell-attached, i.e.,in situ membrane patches. In excised, inside-out, membrane patches, the channel was predominantly open with the open-state probability close to 0.8 over the whole range of potentials tested (–60 to +60 mV). The channel activity was not a function of internal calcium concentration between 10–9 and 10–3 m. We suggest that this Cl channel, whose properties are distinct from those in other epithelia, could account for the well-documented conductance which mediates Cl exit in the basolateral step of NaCl absorption in thick ascending limb of Henle's loop.  相似文献

Summary The charge-pulse technique has been used previously for the study of quasistationary processes in membranes which required only a moderate time resolution. It is shown here that a time resolution of about 400 nsec may be achieved with this technique and that it may be applied to the kinetic analysis of carrier-mediated ion transport. By this method we have studied the transport of alkali ions through optically black monoolein membranes in the presence of the ion carrier valinomycin. All three relaxation processes that are predicted by theory have been resolved. From the relaxation times and the relaxation amplitudes the rate constants for the association (k R ) and the dissociation (k D ) of the ioncarrier complex, as well as the translocation rate constants of the complex (k MS ) and the free carrier (k S ) could be obtained. For 1m Rb+ at 25° C the values arek R =3×105 m –1 sec–1,k D =2×105 sec–1,k MS =3×105 sec–1,k S =4×104 sec–1. The activation energies of the single rate constants which have been estimated from experiments at two different temperatures range between 50 and 90 kJ/mol.  相似文献

Summary The apical plasma membrane of toad bladder epithelial cells has been enzymatically iodinated, using lactoperoxidase, H2O2 (generated by a glucose-glucose oxidase system) and NaI. The site of labeling was demonstrated by electron-microscopic autoradiography; the silver grains (125I) were found exclusively overlying the luminal plasma membranes of the epithelium. The iodination reaction reached completion in less than 5 min. The dependence of the degree of iodination on NaI concentrations (range=6.3×10–8 to 6.3×10–2 m) in the mucosal medium was determined. The results suggest that three classes of sites are iodinated within this concentration range. At concentrations of NaI of 6.3×10–6 m or less, iodination of the apical membrane had no significant effect on either the fine structure of the epithelium or on electrophysiological properties. The baseline short-circuit current (SCC) remained steady and the response to vasopressin was unimpaired. At concentrations of 6.3×10–5 m NaI and greater, the baseline SCC was depressed and the response to vasopressin was partially inhibited. The results indicate that125I may serve as a covalent marker (specific for tyrosine and histidine residues) of the apical plasma membrane of epithelia.  相似文献

Summary A number of published data suggest a variable stoichiometry between the rates of cellular potassium uptake and net sodium transport (J Na) across the urinary bladder of the toad. This problem was examined by simultaneously studying the intracellular chemical activity of potassium (a K) with open-tip K+-selective microelectrodes and micropipets, and monitoringJ Na by measuring the short-circuit current (SCC). When bathed in the short-circuited state with solutions containing ana K of 2.7mm, the mean ±sem values for intracellulara K were 43±0.6mm.Ouabain, at a concentration of 10–2 m, reduced intracellulara K by 56–67% and SCC by 96–100%. At 5×10–4 m, ouabain reversibly reduced intracellulara K by 40–55%, and SCC by 63–68%; the inhibition of SCC was only partly reversible during the period of observation.Removal of external potassium reduced intracellulara K by 69–80% and SCC by 51–76%. Restoration of external potassium entirely returned intracellulara K to its control value, but only partially reversed the inhibition of SCC during the period of study. Furthermore, recovery ofa K began 19–43 min before that of SCC; recovery ofa K was 90–97% complete before any increase in SCC could be measured. Although other interpretations are possible, the simplest interpretation of the data is that the processes responsible for potassium accumulation and transepithelial sodium transport are not identical. We propose the existence of a separate transfer mechanism at the basolateral cell membrane, responsible for accumulating intracellular potassium, and not directly coupled to active sodium transport.  相似文献

Summary The Ca2+-activated K+ channel in rat pancreatic islet cells has been studied using patch-clamp single-channel current recording in excised inside-out and outside-out membrane patches. In membrane patches exposed to quasi-physiological cation gradients (Na+ outside, K+ inside) large outward current steps were observed when the membrane was depolarized. The single-channel current voltage (I/V) relationship showed outward rectification and the null potential was more negative than –40 mV. In symmetrical K+-rich solutions the single-channelI/V relationship was linear, the null potential was 0 mV and the singlechannel conductance was about 250 pS. Membrane depolarization evoked channel opening also when the inside of the membrane was exposed to a Ca2+-free solution containing 2mm EGTA, but large positive membrane potentials (70 to 80 mV) were required in order to obtain open-state probabilities (P) above 0.1. Raising the free Ca2+ concentration in contact with the membrane inside ([Ca2+]i) to 1.5×10–7 m had little effect on the relationship between membrane potential andP. When [Ca2+]i was increased to 3×10–7 m and 6×10–7 m smaller potential changes were required to open the channels. Increasing [Ca2+]i further to 8×10–7 m again activated the channels, but the relationship between membrane potential andP was complex. Changing the membrane potential from –50 mV to +20 mV increasedP from near 0 to 0.6 but further polarization to +50 mV decreasedP to about 0.2. The pattern of voltage activation and inactivation was even more pronounced at [Ca2+]i=1 and 2 m. In this situation a membrane potential change from –70 to +20 mV increasedP from near 0 to about 0.7 but further polarization to +80 mV reducedP to less than 0.1. The high-conductance K+ channel in rat pancreatic islet cells is remarkably sensitive to changes in [Ca2+]i within the range 0.1 to 1 m which suggests a physiological role for this channel in regulating the membrane potential and Ca2+ influx through voltage-activated Ca2+ channels.  相似文献

Summary Cadmium ion (Cd++) significantly increased potential difference (PD) and short-circuit current (SCC) across isolated frog skin when added to the outside Ringer's solution at 10–4, 10–3 and 5×10–3 m concentration. Resistance was reduced by 10–4 m Cd++ but not significantly changed by the higher concentrations. When SCC was first stimulated by vasopressin, 10–4 and 10–3 m Cd++ produced additive stimulation which was reversible by washing with Cd++-free Ringer's. If SCC was first stimulated by Cd++, further stimulation by vasopressin was additive with 10–4 m Cd++ but completely inhibited by 10–3 m Cd++. Elevating the calcium ion (Ca++) concentration of the outer Ringer's from 10–3 m to 5×10–3 m or 10–2 m prior to Cd++ treatment did not reduce the magnitude of SCC stimulation by Cd++. Removal of Ca++ from the outside Ringer's with 2×10–3 m EDTA increased SCC as predicted. Subsequent addition of 5×10–3 m Cd++ drastically reduced SCC below control levels while equimolar concentrations of Cd++ and EDTA reduced SCC only to control levels. These results suggest that Cd++ interacts with the components of the apical plasma membranes of epithelial cells which are associated with the stimulation of SCC by vasopressin and Ca++ removal and may be a useful probe for elucidating these components.  相似文献

Summary Diffusion of inorganic mercury (Hg2+) through planar lipid bilayer membranes was studied as a function of chloride concentration and pH. Membranes were made from egg lecithin plus cholesterol in tetradecane. Tracer (203Hg) flux and conductance measurements were used to estimate the permeabilities to ionic and nonionic forms of Hg. At pH 7.0 and [Cl] ranging from 10–1000mm, only the dichloride complex of mercury (HgCl2) crosses the membrane at a significant rate. However, several other Hg complexes (HgOHCl, HgCl 3 and HgCl 4 2– ) contribute to diffusion through the aqueous unstirred layer adjacent to the membrane. The relation between the total mercury flux (J Hg), Hg concentrations, and permeabilities is: 1/J Hg=1/P ul[Hg t ]+1/P m [HgCl2], where [Hg t ] is the total concentration of all forms of Hg,P ul is the unstirred layer permeability, andP m is the membrane permeability to HgCl2. By fitting this equation to the data we find thatP m =1.3×10–2 cm sec–1. At Cl concentrations ranging from 1–100mm, diffusion of Hg t through the unstirred layer is rate limiting. At Cl concentrations ranging from 500–1000mm, the membrane permeability to HgCl2 becomes rate limiting because HgCl2 comprises only about 1% of the total Hg. Under all conditions, chemical reactions among Hg2+, Cl and/or OH near the membrane surface play an important role in the transport process. Other important metals, e.g., Zn2+, Cd2+, Ag+ and CH3Hg+, form neutral chloride complexes under physiological conditions. Thus, it is likely that chloride can facilitate the diffusion of a variety of metals through lipid bilayer and biological membranes.  相似文献

Summary The effects of bathing solution HCO 3 /CO2 concentrations on baseline cell membrane voltages and resistances were measured inNecturus gallbladder epithelium with conventional intracellular microelectrode techniques. Gallbladders were bathed in either low HCO 3 /CO2 Ringer's solutions (2.4mm HCO 3 /air or 1mm HEPES/air) or a high HCO 3 /CO2 Ringer's (10mm HCO 3 /1% CO2). The principal finding of these studies was that the apical membrane fractional resistance (fR a) was higher in tissues bathed in the 10mm HCO 3 /CO2 Ringer's, averaging 0.87±0.06, whereasfR a averaged 0.63±0.07 and 0.48±0.08 in 2.4mm HCO 3 and 1mm HEPES, respectively. Intraepithelial cable analysis was employed to obtain estimates of the individual apical (R a) and basolateral membrane (R b) resistances in tissues bathed in 10mm HCO 3 /1% CO2 Ringer's. Compared to previous resistance measurements obtained in tissues bathed in a low HCO 3 /CO2 Ringer's, the higher value offR a was found to be due to both an increase inR a and a decrease inR b. The higher values offR a and lower values ofR b confirm the recent observations of others. To ascertain the pathways responsible for these effects, cell membrane voltages were measured during serosal solution K+ and Cl substitutions. The results of these studies suggest that an electrodiffusive Cl transport mechanism exists at the basolateral membrane of tissues bathed in a 10mm HCO 3 /1% CO2 Ringer's, which can explain in part the fall inR b. The above observations are discussed in terms of a stimulatory effect of solution [HCO 3 /PCO2 on transepithelial fluid transport, which results in adaptive changes in the conductive properties of the apical and basolateral membranes.  相似文献

Summary This paper describes the effect of trinitrocresolate anions (TNC) on the electrical conductance (G m ), and tracer-measured unidirectional Na and K fluxes (M Na andM K) across bilayers formed from sheep red cell lipids dissolved in decane. In the absence of TNC, typical low conductances were observed, while the cation fluxes were too low to measure by our techniques (<10–12 moles cm–2 sec–1). In the presence of TNC (10–2 m),G m increased and TNC was the main charge carrier in the system. The cationic fluxes were also much increased, but the membranes showed no significant selectivity between K and Na. Furthermore, the Na and K fluxes were at least two orders of magnitude larger than the ionic fluxes calculated fromG m . Thus, almost all of the K and Na transport across the membrane in the presence of TNC is electrically silent and is probably carried out as KTNC and NaTNC ion pairs.In the presence of valinomycin (10–6 m) and no TNC, both the ion fluxes andG m were 103 times larger in KCl than in NaCl, thus exhibiting the characteristic high selectivity of valinomycin for K over Na. In the presence of both valinomycin (10–6 m) and TNC (10–2 m), this selectivity disappeared in that bothG m andM Na in the NaCl system were similar to the respective values in the KCl system. Even under these conditions, most of the Na is still transported by a process which does not carry charge.BothG m andM x increased alike and monotonically with increasing temperature over the range 7 to 30°C. In the absence of TNC the enthalpies of activation were invariably higher in KCl than in NaCl. Addition of TNC produced equal enthalpies of activation for both Na and K containing systems suggesting a common, temperature-dependent, ratedetermining step in charge transfer and the electrically silent cation fluxes.  相似文献

Summary We have measured the intracellular potassium activity, [K+]i and the mechanisms of transcellular K+ transport in reabsorptive sweat duct (RSD) using intracellular ion-sensitive microelectrodes (ISMEs). The mean value of [K+]i in RSD is 79.8±4.1mm (n=39). Under conditions of microperfusion, the [K+]i is above equilibrium across both the basolateral membrane, BLM (5.5 times) and the apical membrane, APM (7.8 times). The Na+/K+ pump inhibitor ouabain reduced [K+]i towards passive distribution across the BLM. However, the [K+]i is insensitive to the Na+/K+/2 Cl cotransport inhibitor bumetanide in the bath. Cl substitution in the lumen had no effect on [K+]i. In contrast, Cl substitution in the bath (basolateral side) depolarized BLM from –26.0±2.6 mV to –4.7*±2.4 mV (n=3;* indicates significant difference) and decreased [K+]i from 76.0±15.2mm to 57.7* ±12.7mm (n=3). Removal of K+ in the bath decreased [K+]i from 76.3±15.0mm to 32.3*±7.6mm (n=4) while depolarizing the BLM from –32.5±4.1 mV to –28.3*±3.0 mV (n=4). Raising the [K+] in the bath by 10-fold increased [K+]i from 81.7±9.0mm to 95.0*±13.5mm and depolarized the BLM from –25.7±2.4 mV to –21.3*±2.9 mV (n=4). The K+ conductance inhibitor, Ba2+, in the bath also increased [K+]i from 85.8±6.7mm to 107.0*±11.5mm (n=4) and depolarized BLM from –25.8±2.2 mV to –17.0*±3.1 mV (n=4). Amiloride at 10–6 m increased [K+]i from 77.5±18.8mm to 98.8*±21.6mm (n=4) and hyperpolarized both the BLM (from –35.5±2.6 mV to –47.8*±4.3 mV) and the APM (from –27.5±1.4 mV to –46.0* ±3.5 mV,n=4). However, amiloride at 10–4 m decreased [K+]i from 64.5±0.9mm to 36.0*±9.9mm and hyperpolarized both the BLM (from –24.7±1.4 mV to –43.5*±4.2 mV) and APM (from –18.3±0.9 mV to –43.5*±4.2 mV,n=6). In contrast to the observations at the BLM, substitution of K+ or application of Ba2+ in the lumen had no effect on the [K+]i or the electrical properties of RSD, indicating the absence of a K+ conductance in the APM. Our results indicate that (i) [K+]i is above equilibrium due to the Na+/K+ pump; (ii) only the BLM has a K+ conductance; (iii) [K+]i is subject to modulation by transport status; (iv) K+ is probably not involved in carrier-mediated ion transport across the cell membranes; and (v) the RSD does not secrete K+ into the lumen.  相似文献

Summary The effects of stepwise concentration changes of K+ and HCO 3 in the basolateral solution on the basolateral membrane potential (V bl) of proximal tubule cells of the doubly-perfusedNecturus kidney were examined using conventional microelectrodes. Apparent transference numbers were calculated from changes inV bl after alterations in external K+ concentration from 1.0 to 2.5mm (t K, 1.0–2.5), 2.5 to 10, and in external HCO 3 concentration (at constant pH) from 5 to 10mm (t HCO3, 5–10), 10 to 20, or 10 to 50.t K, 2.5–10 was 0.38±0.02 under control conditions but was sharply reduced to 0.08±0.03 (P>0.001) by 4mm Ba++. This concentration of Ba++ reducedV bl by 9±1 mV (at 2.5 external K+). Perfusion with SITS (5×10–4 m) for 1 hr hyperpolarizedV bl by 10±3 mV and increasedt K, 2.5–10 significantly to 0.52±0.01 (P<0.001). Ba++ application in the presence of SITS depolarizedV bl by 22±3 mV. In control conditionst HCO3, 10–50 was 0.63±0.05 and was increased to 0.89±0.07 (P<0.01) by Ba++ but was decreased to 0.14±0.02 (P<0.001) by SITS. In the absence of apical and basolateral chloride, the response ofV bl to bicarbonate was diminished but still present (t HCO3, 10–20 was 0.35±0.03). Intracellular pH, measured with liquid ion-exchange microelectrodes, increased from 7.42±0.19 to 7.57±0.17 (P<0.02) when basolateral bicarbonate was increased from 10 to 20mm at constant pH. These data show that the effects of bicarbonate onV bl are largely independent of effects on the K+ conductance and that there is a significant current-carrying bicarbonate pathway in the basolateral membrane. Hence, both K+ and HCO 3 gradients are important in the generation ofV bl, and their relative effects vary reciprocally.  相似文献

Summary Near-instantaneous current-voltage relationships and shot-noise analysis of amiloride-induced current fluctuations were used to estimate apical membrane permeability to Na (P Na), intraepithelial Na activity (Na c ), single-channel Na currents (i) and the number of open (conducting) apical Na channels (N0), in the urinary bladder of the toad (Bufo marinus). To facilitate voltageclamping of the apical membrane, the serosal plasma membranes were depolarized by substitution of a high KCl (85mm) sucrose (50mm) medium for the conventional Na-Ringer's solution on the serosal side.Aldosterone (5×10–7 m, serosal side only) elicited proportionate increases in the Na-specific current (I Na and inP Na, with no significant change in the dependence ofP Na on mucosal Na (Na o ).P Na and the control ofP Na by aldosterone were substrate-dependent: In substrate-depleted bladders, pretreatment with aldosterone markedly augmented the response to pyruvate (7.5×10–3 m) which evoked coordinate and equivalent increases inI Na andP Na.The aldosterone-dependent increase inP Na was a result of an equivalent increase in the area density of conducting apical Na channels. The computed single-channel current did not change. We propose that, following aldosterone-induced protein synthesis, there is a reversible metabolically-dependent recruitment of preexisting Na channels from a reservoir of electrically undetectable channels. Summary The effect of the K-sparing diuretic amiloride was assessed electrophysiologically in the isolated cortical collecting tubule of the rabbit, a segment which absorbs Na and secretes K. Low concentrations of amiloride in the perfusate caused a rapid, reversible, decrease in the magnitude of the lumen negative transepithelial potential difference,V te, transepithelial conductanceG te, and equivalent short-circuit current,I sc, with an apparentK 1/2 of approximately 7×10–8 m. The effects of a maximum inhibitory concentration of amiloride (10–5 m) were identical to those observed upon Na removal from lumen and bath (Na removal from the bath alone has no effect). Removal of Na in the presence of 10–5 m amiloride had no affect onV te,G te, orI sc, and is consistent with the view that amiloride blocks the Na conductive pathways of the apical cell membrane. Further, in the absence of Na, the subsequent addition of amiloride had no influence. In tubules where active Na absorption was either spontaneously low, or abolished by removal of Na from lumen and bath, the elevation of K from 5 to 155 meq/liter in the perfusate caused a marked change of theV te in the negative direction and an increase in theG te. These effects could be attributed to a high K permeability of the apical cell membrane and not of the tight junctions. Amiloride (10–5 m) had no effect on these responses to K. It is concluded that amiloride selectively blocks the apical cell membrane Na channels but has no effect on the K conductive pathways(s). This selective nature of amiloride may indicate that Na and K are transported across the apical cell membrane via separate conductive pathways.  相似文献

Summary The effect of cholecystokinin (CCK) and internal Ca2+ on outward K+ current in isolated pig pancreatic acinar cells has been investigated using the patch-clamp method for whole-cell current recording under voltage-clamp conditions. CCK (2 × 10–10 M) applied to the bath evoked a marked increase in the outward K+ current associated with depolarizing voltage steps, and this effect was fully reversible and acutely dependent on the presence of external Ca2+. When strongly buffered Ca2+-EGTA solutions were used inside the cells CCK failed to evoke an effect. Increasing the internal Ca2+ concentration ([Ca2+] i ) from 5 × 10–10 M to 10–7 and 5 × 10–7 M mimicked the effect of CCK. It would appear therefore that CCK controls K+ conductance in the acinar cells via changes in the internal free ionized Ca2+ concentration.  相似文献

Summary Investigations into the properties of 6-PG dehydrogenase in cell free extracts of Escherichia coli revealed a pH optimum at pH 9.5 with a sharp decline on both sides of the optimum. The addition of 1.0×10-2 m MgCl2 produced maximal activity, whereas higher concentrations caused inhibition. The K m values were 2.5×10-4 m for 6-phosphogluconate and 2.5×10-5 m for NADP+ as substrate. The enzyme was extremely stable for at least 5 hours if stored at 4°C in Tris–NaCl–MgCl2 buffer at pH 7.5. 6-PG dehydrogenase activity was shown to be proportional to cell free extract concentration over the range 0–0.3 mg protein. Summary When present at nanomolar concentrations on one side of a lipid bilayer membrane,helianthus toxin (a protein of mol wt16,000) increases enormously membrane permeability to ions and nonelectrolytes by forming channels in the membrane. Membranes containing sphingomyelin are especially sensitive to toxin, but sphingomyelin isnot required for toxin action. Conductance is proportional to about the 4th power of toxin concentration. Single channel conductances are approximately 2×10–10 mho in 0.1m KCl. Toxin-treated membranes are more permeable to K+ and Na+ than to Cl and SO 4 = , but the degree of selectivity is pH dependent. Above pH 7 membranes are almost ideally selective for K+ with respect to SO 4 = , whereas below pH 4 they are poorly selective. The channels show classical molecular sieving for urea, glycerol, glucose, and sucrose — implying a channel radius >5 Å. In symmetrical salt solutions above pH 7, theI–V characteristic of the channel shows significant rectification: below pH 5 there is very little rectfication. Because of the effects of pH on ion selectivity and channel conductance, and also because of the rectification in symmetrical salt solutions and the effect of pH on this, we conclude that there are titratable negative charge groups in the channel modulating ion permeability and selectivity. Since pH changes on the side containing the toxin are effective whereas pH changes on the opposite side are not, we place these negative charges near the mouth of the channel facing the solution to which toxin was added.  相似文献
