 The migratory history of the brown trout, Salmo trutta, collected from Japanese rivers, was examined in terms of strontium (Sr) and calcium (Ca) uptake in the otolith, by means of wavelength dispersive X-ray spectrometry on an electron microprobe. Sea-run (anadromous) and freshwater-resident (nonanadromous) types of S. trutta were found to occur sympatrically. Otolith Sr concentration or Sr : Ca ratios of anadromous S. trutta fluctuated strongly along the life history transect in accordance with the migration (habitat) pattern from sea to freshwater. In contrast, the Sr concentration or the Sr : Ca ratios of nonanadromous fish remained at consistently low levels throughout the otolith. The higher ratios in anadromous S. trutta, in the otolith region from the core to 1500 μm, corresponded to the initial seagoing period, probably reflecting the ambient salinity or the seawater–freshwater gradient in Sr concentration. The findings clearly indicated that otolith Sr : Ca ratios reflected individual life histories, enabling the sea-run S. trutta to be distinguished from the freshwater-resident brown trout. The migratory history of Sakhalin taimen, Hucho perryi, was examined in terms of strontium (Sr) and calcium (Ca) uptake in the otolith by using wavelength dispersive X-ray spectrometry on an electron microprobe. Otolioth Sr:Ca ratios of freshwater-reared samples remained consistently at low levels throughout the otolith. The Sr:Ca ratios of samples from Lake Aynskoye of Sakhalin Island showed a low value from the core up to a point of 700–2140µm. Thereafter, the ratios increased sharply and remained at higher levels up to the outermost regions. The difference in Sr:Ca ratio might be the result of the presence of individuals that underwent seawater and freshwater life history phases, probably reflecting the ambient salinity or the seawater–freshwater gradient in Sr concentration. The ratio of strontium to calcium concentration in the otolith of four species of Tribolodon (Cyprinidae) was examined to reveal their migratory histories. The ratio was relatively stable from the core to the edge of the otolith in fluvial Tribolodon ezoe and T. hakonensis, whereas it changed from a low level to a higher level in anadromous T. brandtii and T. hakonensis. Four specimens of T. nakamurai from the Mogami River, Yamagata Prefecture, northern Japan, showed relatively stable ratios, indicating a fluvial life mode. On the other hand, the ratio increased once in only one specimen of T. nakamurai from the Koyoshi River of Akita Prefecture, suggesting the possibility that it was born in the Mogami River, the nearest major habitat of this species, and accidentally migrated to the Koyoshi River through the sea.  相似文献

 We tried to elucidate how females of a paternal nest brooding goby Rhinogohius sp. OR deposit their eggs in a nest, using a marking technique for live eggs under laboratory conditions in which male somatic condition, nest space, and mating pattern (monogamous or bigamous) were controlled. Whether females rejected mating was independent of either male quality, such as body size and somatic condition, or nest space. In a situation in which two females were allowed to spawn sequentially with a male, however, females rejected mating at a higher rate when they were the first to spawn than when they were the second to spawn; this is because eggs from first females were more vulnerable to cannibalism by parental males and second females. Even when nest space was limited and thus was occupied by eggs from the first females, second females could deposit all their eggs in the nest by using the minute interspace of existing eggs. In the presence of the parental male, such a female seemed less likely to suffer a cost from increased egg mortality due to crowdedness, still holding the advantage of being the second spawner. Finally, we extrapolated the field breeding ecology of this fish from the laboratory data. It was suggested that a single monolayer brood of the same age usually consisted of eggs from multiple females and thus the mating pattern would be more polygynous than previously expected. The CO2 fixation rate of salt-stressed maize leaves was influenced by the Ca∶Na ratio in the solution cultures. At an osmotic potential of −0.4 MPa in the root media, both the photosynthetic rate and the water-use efficiency declined as substrate Ca increased. Blade-Ca concentration also increased, while blade-Na and-Mg decreased. Apparently photosynthetic activity was inhibited in part by internal Mg deficiency rather than by Na toxicity or by Na-induced Ca deficiency. Reduction of the Ca∶Mg ratio in the culture stimulated the CO2 fixation rate.  相似文献

This paper presents the basis of DNA genealogy, a new field of science, which is currently emerging as an unusual blend of biochemistry, history, linguistics, and chemical kinetics. The methodology of the new approach is comprised of chemical (biological) kinetics applied to a pattern of mutations in non-recombinant fragments of DNA (Y chromosome and mtDNA, the latter not being considered in this overview). The goal of the analysis is to translate DNA mutation patterns into time spans to the most recent common ancestors of a given population or tribe and to the dating of ancient migration routes. To illustrate this approach, time spans to the common ancestors are calculated for ethnic Russians, that is Eastern Slavs (R1a1 tribe), Western Slavs (I1 and I2 tribes), and Northern (or Uralic) Slavs (N1c tribe), which were found to live around 4600 years before present (R1a1), 3650 ybp (I1), 3000 and 10,500 ybp (I2, two principal DNA lineages), and 3525 ybp (N1c) (confidence intervals are given in the main text). The data were compared with the respective dates for the nearest common ancestor of the R1a1 “Indo-European” population in India, who lived 4050 years before present, whose descendants represent the majority of the upper castes in India today (up to 72%). Furthermore, it was found that the haplotypes of ethnic Russians of the R1a1 haplogroup (up to 62% of the population in the Russian Federation) and those of the R1a1 Indians (more than 100 million today) are practically identical to each other, up to 67-marker haplotypes. This essentially solves a 200-year-old mystery of who were the Aryans who arrived in India around 3500 years before the present. Haplotypes and time spans to the ancient common ancestors were also compared for the ethnic Russians of haplogroups I1 and I2, on one hand, and the respective I1 and I2 populations in Eastern and Western Europe and Scandinavia, on the other. It is suggested that the approach described in this overview lays the foundation for “molecular history”, in which the principal tool is high-technology analysis of DNA molecules of both our contemporaries and excavated ancient DNA samples, along with their biological kinetics.  相似文献

Males of the stream goby Rhinogobius sp. DA (dark type) court females in deep pools and care for the eggs under stones in shallow riffles. We studied male–male competition for access to females and nest sites to understand how male size influences the mating success of this species. In field observations, larger males won in fighting with other males. However, large males did not tend to monopolize courtship opportunities, and the frequency of successful courtships, after which males led the females to the nests, was not related to male body size. The fact that courted females always escaped from the fighting sites once males began fighting likely explains why male size was not positively related to courtship success. Large males occupied large nest stones, and the number of eggs received in the nest was correlated positively with nest size. In aquarium experiments with two tiles of different sizes provided as nesting materials, males always chose the larger nest and, when two males were introduced simultaneously, the larger one occupied the larger nest. These results suggested that male mating success of this goby is determined by male–male competition for large nests rather than for access to females. Summary To distinguish ligand-induced structural states of the (Na+–K+)-ATPase, the purified membrane-bound enzyme isolated from rat kidneys was digested with trypsin in the presence of various combinations of Na+, K+, Mg++ and ATP. It was found that first the large and then the small polypeptide chain of the (Na+–K+)-ATPase was degraded, indicating that the lysine and arginine residues of the large chain are more exposed than are those of the small one. The (Na+–K+)-ATPase activity was inactivated in parallel with the degradation of the large polypeptide chain. After the degradation of the large polypeptide chain, about 75% of the (Na+–K+)-ATPase protein remained bound to the membrane, demonstrating that the split protein segments were only partially released.It was found that the combinations of ATP, Mg++, Na+ and K+ present during trypsin digestion influenced the time course and degree of degradation of the (Na+–K+)-ATPase protein. The degradations of the large and the small polypeptide chain were affected in parallel. Thus, certain ATP and ligand combinations influenced neither the degradation of the large nor the degradation of the small polypeptide chain, whereas by other combinations of ATP and ligands the degree of susceptibility of both polypeptide chains to trypsin was equally increased or reduced.In the absence of ATP the time course of trypsin digestion of the (Na+–K+)-ATPase was the same, whether Na+ or K+ was present. With low ATP concentrations (e.g., 0.1mm), however, binding of Na+ or K+ led to different degradation patterns of the enzyme. If a high concentration of ATP (e.g., 10mm) was present, Na+ and K+ also influenced the degradation pattern of the (Na+–K+)-ATPase, but differentially compared to that at low ATP concentrations, since the effects of Na+ and K+ were reversed. Furthermore, it was found that the degradation of the small chain was only influenced by certain combinations of ATP, Mg++, Na+ and K+ if the large chain was intact when the ligands were added to the enzyme.The described results demonstrate structural alterations of the (Na+–K+)-ATPase complex which are supposed to include a synchronous protrusion or retraction of both (Na+–K+)-ATPase subunits. The data further suggest that ATP and other ligands primarily alter the structure of the large (Na+–K+)-ATPase subunit. This structural alteration is presumed to lead to a synchronous movement of the small subunit of the enzyme. The structural state of the (Na+–K+)-ATPase is regulated by binding of Na+ or K+ to the enzyme-ATP complex. The effects of Na+ and K+ on the (Na+–K+)-ATPase structure are modulated by the ATP binding to high affinity and to low affinity ATP binding sites.  相似文献

The isiAB genes have proven to be highly stress-responsive under a variety of environmental conditions, including iron deficiency, high salt and oxidative stress. In order to understand the function of IsiA and its importance in oxidative stress, we constructed a knock out mutant of the isiA gene and compared differential gene expression of the DeltaisiA strain in the presence and absence of H2O2. We used the full genome microarray for the cyanobacterium Synechocystis sp. PCC 6803 as previously described [Postier BL, Wang HL, Singh A, Impson L, Andrews, HL, Klahn J, Li H, Risinger G, Pesta D, Deyholos M, Galbraith DW, Sherman LA and Burnap RL (2003) BMC Genenomics 4: 23-34]. We determined that one of the main differences in DeltaisiA compared to wild-type (in the absence of peroxide) was the induction of a gene cluster (sll1693-sll1696) that encoded genes resembling pilins or general secretory proteins (Gsp). These proteins are targeted to the cytoplasmic membrane and we suggest that they may be involved in the assembly of membrane complexes, including pigment-protein complexes. The DeltaisiA strain was more resistant to H2O2 compared to the wild-type. In the presence of 1.5 mM H2O2 for 30 min, a cluster of genes that includes a peroxiredoxin was induced 7- to 8-fold and we suggest that this peroxide scavenging enzyme is responsible for the increased peroxide resistance of the DeltaisiA strain.  相似文献

The ubiquitous alpha-proteobacteria of the order “Candidatus Pelagibacterales” (SAR11) are highly abundant in aquatic environments, and among them, members of the monophyletic lineage LD12 (also known as SAR11 clade IIIb) are specifically found in lacustrine ecosystems. Clade IIIb bacteria are some of the most prominent members of freshwater environments, but little is known about their biology due to the lack of genome representatives. Only recently, the first non-marine isolate was cultured and described as “Candidatus Fonsibacter ubiquis”. Here, we expand the collection of freshwater IIIb representatives and describe a new IIIb species of the genus “Ca. Fonsibacter”. Specifically, we assembled a collection of 67 freshwater metagenomic datasets from the interconnected lakes of the Chattahoochee River basin (GA, USA) and obtained nearly complete metagenome-assembled genomes (MAGs) representing 5 distinct IIIb subclades, roughly equivalent to species based on genomic standards, including the previously described “Ca. F. ubiquis”. Genomic comparisons between members of the IIIb species revealed high similarity in gene content. However, when comparing their abundance profiles in the Chattahoochee basin and various aquatic environments, differences in temporal and spatial distributions among the distinct species were observed implying niche differentiation might be underlying the coexistence of the highly functionally similar representatives. The name Ca. Fonsibacter lacus sp. nov. is proposed for the most abundant and widespread species in the Chattahoochee River basin and various freshwater ecosystems.  相似文献

Coralligenous habitat and rhodoliths beds are very important in terms of biodiversity in the Mediterranean Sea. During an oceanographic campaign, carried out in northern Cap Corse, new coralligenous structures have been discovered. These structures, never previously identified in the Mediterranean Sea, are named “coralligenous atolls” because of their circular shape. The origin and growth dynamics of these atolls are still unknown but their form does not appear to result from hydrodynamic action and an anthropogenic origin also seems unlikely. However, this kind of shape seems rather closer to that of other circular structures (e.g. pockmarks) the origin of which is related to gaseous emissions. Further studies are needed to confirm this hypothesis through chemical analysis.  相似文献

Abstract

Chromosome number knowledge of the Italian vascular flora is stored in the online database Chrobase.it, which includes 6723 records, referable to 3428 taxa, 2799 accepted species and subspecies (about 35% of the national flora), and 3410 different chromosome countings (cytotypes). Appropriate queries to Chrobase.it allowed us to calculate mean, modal and median chromosome numbers for the Italian vascular flora, for geographical subgroups (islands, south, centre, north) and for selected orders, families and genera. Chromosome number data were available for 41 out of 55 orders (74%) and 107 out of 428 families (67%), represented by 664 out of 1297 genera (51%). The most studied administrative regions are Sicily (844 taxa), Tuscany (592 taxa), and Sardinia (390 taxa), while the most studied families are Asteraceae (465 taxa), Fabaceae (266 taxa), Brassicaceae (158 taxa), and Poaceae (144 taxa). Chromosome numbers range from 2n = 6, occurring in several species of Hypochaeris (Asteraceae), to 2n = 240, occurring in Ophioglossum (Ophioglossaceae), Dryopteris (Dryopteridaceae) and Arenaria (Caryophyllaceae) (mode is 2n = 18, and median is 2n = 24). Chromosome number variability was analyzed by frequencies (linear plots) and ANOVA, resulting in significant differences among geographical groups (mean chromosome number increasing from islands-south to centre-north) and selected taxa. B-chromosomes occur in 5.3% of data (148 taxa) and their number is not significantly different among geographical areas, while they occur only in 14 orders, 17 families, and 56 genera. Genome evolution arises from two main ways of duplication and reduction. Fish specific genome duplication (FSGD) may have occurred before the radiation of the teleosts. Common carp (Cyprinus carpio L.) has been considered to be a tetraploid species, because of its chromosome numbers (2n=100) and its high DNA content. Using 69 microsatellite primer pairs, the variations were studied to better understand the genome evolution (genome duplication and diploidization) of common carp from a gynogenetic family. About 48% of primer pairs were estimated to amplify duplicates based on the number of PCR amplification per individual. Segregation patterns in the family suggested a partially duplicated genome structure and disomic inheritance. This indicates that the common carp is tetraploid and polyploidy occurred by allotetraploidy. Two primer pairs (HLJ021 and HLJ332) were estimated to amplify reduction based on the number of PCR amplification per individual. One allele in HLJ002 locus and HLJ332 locus was clearly lost in the gynogenetic family and the same as in six wild populations. Segregation patterns in the family suggested a partially diplodization genome structure. A hypothesis transition (dynamic) and equilibrium (static) were proposed to explain the common carp genome evolution between genome duplication and diploidization.  相似文献

We report the occurrence of an unidentified species of the wasp Microctonus Wesmael (Hymenoptera: Braconidae) parasitizing adults of the Fuller’s rose weevil Naupactus cervinus (Boheman) (Coleoptera: Curculionidae), a widespread pest of economically important crops included in the South American tribe Naupactini. Cytochrome c oxidase subunit I-based phylogenetic analysis indicates that the parasitoid is closely related to Microctonus hyperodae Loan. Their first instar larvae show slight morphological differences with this species. Superparasitism by first instar larvae occurred at low frequency. Some teratocytes were observed. Microctonus sp. and its host were infected with different strains of the reproductive parasite Wolbachia pipientis Hertig (Rickettsiales: Rickettsiaceae), although the bacterial lineage harbored by the wasp coincides with that infecting most parthenogenetic Naupactini. This multipartite association (weevil bearing both Wolbachia and Microctonus, and Microctonus bearing Wolbachia) opens challenging perspectives for future research on biological interactions and biological control.  相似文献

Bacterial endosymbionts belonging to the family Rickettsiaceae were recently identified in the unicellular green alga Carteria cerasiformis, providing the first molecular evidence of rickettsial endosymbionts within photosynthetic eukaryotes. However, previous morphological studies using transmission electron microscopy (TEM) with conventional chemical fixation did not demonstrate whether the endosymbionts of C. cerasiformis have the diagnostic characteristics of the family Rickettsiaceae. In this study, we observed the rickettsial endosymbionts “MIDORIKO” within C. cerasiformis cells by TEM with high-pressure freezing and freeze-substitution fixation. The rickettsial endosymbionts resided directly in the C. cerasiformis cytoplasm without engulfing or encompassing membranes or vacuoles. The endosymbionts had a Gram-negative cell envelope composed of outer and inner bilayer membranes. The thicknesses of the outer and inner leaflets of the bacterial cell wall were almost identical. These morphological characteristics are consistent with those of the genus Rickettsia, but the cell wall structure differed from that of the genus Orientia within the family Rickettsiaceae.  相似文献

The extreme climate of the Canadian Prairies poses a major chal enge to improve yield. Although it is possible to breed for yield per se, focusing on yield-related traits could be advantageous because of their simpler genetic architecture. The Canadian flax core col ection of 390 accessions was genotyped with 464 simple sequence repeat markers, and phenotypic data for nine agronomic traits including yield, bol s per area, 1,000 seed weight, seeds per bol , start of flowering, end of flowering, plant height, plant branching, and lodging col ected from up to eight environments was used for association mapping. Based on a mixed model (principal component analysis (PCA) t kinship matrix (K)), 12 significant marker-trait associations for six agronomic traits were identi-fied. Most of the associations were stable across environments as revealed by multivariate analyses. Statistical simulation for five markers associated with 1000 seed weight indicated that the favorable al eles have additive effects. None of the modern cultivars carried the five favorable al eles and the maximum number of four observed in any accessions was mostly in breeding lines. Our results confirmed the complex genetic architecture of yield-related traits and the inherent difficulties associated with their identification while il ustrating the potential for improvement through marker-assisted selection.  相似文献
