Interactions between a female-specific lethal mutant, Sxlf#1, and each of three male-specific lethal mutants, mle(3)132, msl-227 and mle, of Drosophila melanogaster were observed to produce morphological changes in various sexually dimorphic external characters. Chromosomal females heterozygous for Sxlf#1 and homozygous for any one of the male-specific lethals (and to a lesser degree heterozygous for male-specific lethals) sometimes had sex combs, male-type tergites, male-type sternites, male-type anal plates or male-type external genitalia. Penetrance was not high and expression was often incomplete; single individuals never had all the sexually dimorphic structures transformed. When mothers were homozygous for male-specific lethals, higher proportions of female progeny were affected than when mothers were heterozygous, suggesting a maternal effect.  相似文献

Effects of a second chromosome male-specific lethal gene, maleless (mle), of Drosophila melanogaster were further studied. It was shown that, although no maternal effect was seen with respect to the male-specific lethality, the lethal stage was influenced by whether parental females were homozygous or heterozygous for mle. Thus, in the former mle/mle males died mostly in the late third instar larval stage, while in the latter practically all males survived to the pupal stage. In the dying mle/mle male pupae complete differentiation of adult external head and thorax structures was often observed but that of abdominal structures was incomplete forming only a few segments in most cases. Imaginal discs from third instar mle/mle male larvae which were produced by mle/mle mothers and were destined to die as larvae were able to differentiate into adult structures upon transplantation into normal third instar larval hosts.——A somewhat elaborated version of the previously presented hypothesis (Fukunaga, Tanaka and Oishi 1975) was discussed as to the possible presence of a class of sex-specific lethals which are not related to the process of primary sex differentiation.  相似文献

Ethyl methanesulfonate-treated third chromosome of Drosophila melanogaster were tested for the presence of dominant and recessive temperature-sensitive lethal mutations at 17 degrees , 22 degrees and 29 degrees C. Out of 1,176 chromosomes tested, no dominant ts lethals, 21 heat-sensitive, 22 cold-sensitive and 10 heat-cold-sensitive lethals were recovered. Heat-cold sensitivity was produced by a single mutation in all cases. Sixty-two percent of the ts lethals were fertile as homozygotes in both sexes. Surprisingly, 88% of the ts lethals mapped between st and Sb, a region straddling the centromere and estimated to comprise 12.9% of the genetic length and 55% of the physical length of chromosome 3. All but one of the heat- and cold-sensitive lethals complemented with each other at their respective restrictive temperatures.  相似文献

The effects of SRO (Sex-ratio organism; previously called SR spirochetes) infection on the viability of gynandromorphs of D. melanogaster were examined. A fraction of gynadromorphs, most of them with small areas of XO tissue, survived, but the majority were not viable when XO nuclei or cells are affected by the infecting SRO. Apparently the replacement of lethal XO nuclei or cells by adjacent XX ones does not take place. Some possibilities are discussed as to the time and the primary site of the lethal action of SRO.  相似文献

Ethyl methanesulfonate-treated autosomes were screened for the presence of dominant cold-sensitive (DCS) lethal mutations in Drosophila melanogaster. None was found among 6,552 treated and 168 untreated third chromosomes. Twenty-three DCS-L chromosomes which caused death at 17 degrees C but survived at 22 degrees C and 29 degrees C were recovered from 5,046 mutagenized chromosome 2's.-The DCS-L mutations all mapped around dp and appeared to be functionally allelic. Lethality of heterozygotes for most of the DCS-L's occurred over a prolonged interval from the embryonic through the larval instars. Prolonged incubation at 17 degrees C did not demonstrate any maternal effect on zygotic survival.  相似文献

Out of 25,000 EMS-treated third chromosomes examined, ten dominant temperature-sensitive (DTS) lethal mutations which are lethal when heterozygous at 29 degrees C but survive at 22 degrees C were recovered. Seven of the eight mutations mapped were tested for complementation; these mutants probably define eight loci. Only DTS-2 survived in homozygous condition at 22 degrees C; homozygous DTS-2 females expressed a maternal effect on embryonic viability. Two of the mutant-bearing chromosomes, DTS-1 and DTS-6, exhibited dominant phenotypes similar to those associated with Minutes. Each of the seven mutants examined exhibited a characteristic phenotype with respect to the time of death at 29 degrees C and the temperature-sensitive period during development. Only DTS-4 exhibited dominant lethality in triploid females.  相似文献

Every aspect of the phenotype of the daughterless (2–41.5) maternal effect mutation was found to be strongly temperature sensitive. Above 22°, da/da mothers produced no daughters; however female progeny did survive the da maternal effect if the last 60 hours of oogenesis and the first three hours of embryonic development took place at 18°. The females which survived under these conditions displayed morphological abnormalities in a variety of adult cuticular structures, characteristic of cell death during development. In contrast, their male siblings were morphologically normal. Upon prolonged exposure to 29°, da/da females became sterile but continued to lay eggs. Some sexually mosaic progeny from da/da mothers survived even at 25°, but the distribution and development of the female tissue in these mosaics were abnormal. It is suggested that there are multiple functions of the da+ gene during oogenesis, one of which may be required specifically for the subsequent survival of female cells throughout the embryo. In addition to and distinct from its effect during oogenesis, the da mutation acted in both sexes of progeny as a recessive temperature-sensitive lethal mutation with a TSP during the first half of embryonic development.  相似文献

After fertilization, the development of a zygote depends upon both gene products synthesized by its maternal parent and gene products synthesized by the zygote itself. To analyze genetically the relative contributions of these two sources of gene products, several laboratories have been isolating two classes of mutants of Drosophila melanogaster: maternal-effect lethals and zygotic lethals. This report concerns the analysis of two temperature-sensitive mutants, OX736hs and PC025hs, which were isolated as alleles of a small-disc mutant, l(3)1902. These alleles are not only zygotic lethals, but also maternal-effect lethals. They have temperature-sensitive periods during larval life and during oogenesis. Mutant larvae exposed continuously to restrictive temperature have small discs. One-or two-day exposures to the restrictive temperature administered during the third larval instar lead to a homeotic transformation of the midlegs and hindlegs to the pattern characteristic of the forelegs. Mutant females exposed to the restrictive temperature during oogenesis produce eggs that can develop until gastrulation, but do not hatch.--The existence of these mutants, and one that was recently described by another group, implies that there may be a class of genes, heretofore unrecognized, whose products are synthesized during oogenesis, are essential for embryogenesis and are also synthesized during larval stages within imaginal disc cells.  相似文献

We show that six mapped recessive lethal point mutations of the Notch locus affect mitochondrial enzyme activities: NADH oxidase, NADH dehydrogenase, succinate dehydrogenase and α-glycerophosphate dehydrogenase. The mutant N264-40, which has the same morphological and embryological effects as the Notch8 deletion, demonstrates the same biochemical effects and dosage relations as Notch8. The other five mapped recessive lethals also affect four enzymic activities. They show specific patterns of activity that depend in several cases on the wild-type chromosome in the heterozygous females. That effect occurs with mutants located in the extreme right part of the Notch locus where some mutations, according to other authors, show temperature-sensitive expression.  相似文献

Three dominant female-sterile mutations were isolated following ethyl methanesulfonate (EMS) mutagenesis. Females heterozygous for two of these mutations show atrophy of the ovaries and produce no eggs (ovo D1) or few eggs (ovoD2); females heterozygous for the third mutation, ovoD3, lay flaccid eggs. All three mutations are germ line-dependent and map to the cytological region 4D-E on the X chromosome; they represent a single allelic series. Two doses of the wild-type allele restore fertility to females carrying ovoD3 and ovoD2, but females carrying ovoD1 and three doses of the wild-type allele remain sterile. The three mutations are stable in males but are capable of reversion in females; reversion of the dominant mutations is accompanied by the appearance, in the same region, of a recessive mutation causing female sterility. We discuss the utility of these mutations as markers of clones induced in the female germ line by mitotic recombination as well as the nature of the mutations.  相似文献

Region 98EF-100F in chromosome 3 is interesting for genetic analysis because it contains a number of genes of developmental importance. Although there are no preexisting simple deficiency stocks, this region is amenable to genetic manipulation using other types of rearrangements. In the present investigation we obtained deficiencies by combining the terminal deficiencies formed by segregation of Y;3 translocations with a series of duplications of the tip of 3R, both from Y;3 translocations with different breakpoints and from 3;1 duplications in which the 3R tip is carried as a second arm on the X chromosome. Analysis of such synthetic deficiencies reveals five haplo-abnormal loci in the 98A-100F interval. These include a haplolethal site, a newly described Minute and three previously reported Minute mutations. The newly discovered Minute has been designated M(3)99D and is localized cytologically to bands 99D1-9. The three previously reported Minute loci in the region have been localized more precisely: M(3)1 to bands 99B5-9, M(3)f to bands 99E4-F1 and M(3)g to region 100C-F. In addition, we have been able to obtain synthetic deficiencies uncovering all of the intervals from 99B5 to 100B. These deficiencies will be useful for future genetic and molecular analyses of the genes that map within the right tip of chromosome 3.  相似文献

To test the validity of previous results the inducibility of amylase as well as other biochemical parameters was measured using 45 homozygous strains of Drosophila melanogaster from Akayu, Japan. Only the inducibility (but not protein contents or specific activity of the enzyme) was highly correlated with productivity measured using a starch food regime (rp = 0.41, P < 0.005, rg = 0.73 ± 0.21). Inducibility was also negatively correlated with developmental time using starch food; namely, the one with high inducibility developed the fastest. Population cage experiments using 1600 genomes from the same natural population showed that the inducibility responded positively to natural selection (1.6-fold increase in inducibility in cages using starch food relative to those using normal food), but little frequency change of allozymes was observed. All of these results were consistent and indicated that polymorphisms of inducing factors or regulatory genes were major determinants of fitness differences in a particular environment and may be the genetic materials responsible for the adaptive evolution of organisms, at least in amylase loci.  相似文献
