In 2002, postweaning multisystemic wasting syndrome (PMWS) was diagnosed in a European female wild boar (Sus scrofa), based on the detection of porcine circovirus 2 (PCV2) DNA in various organs, including the uterus, and on histopathologic lesions. This is the first detection of PCV2 DNA in the uterus of a wild boar. Three years later (2005), a wild boar < 6-8 mo of age was found moribund. It presented wasting and dyspnea and finally died. PCV2 DNA was detected in tissue samples, and histopathologic lesions consistent with PMWS were observed. Both wild boars were from neighboring hunting areas in central Greece. Two PCV2 strains from the wild boars were genetically characterized and compared to other reported PCV2 sequences from wild boars and domestic pigs. The PCV-2 sequences from the wild boars in this study were closely related to each other and were grouped with two isolates from wild boars from Hungary. The phylogenetic analysis revealed that the virus might be transmitted between hunting areas. In addition, PCV2 may spread from domestic pigs to wild boars and vice versa.

Since epidemiologically-based science on PCV2 in porcine semen is patchy, we investigated 806 Austrian (A) and German (G) AI boars from five studs, and boars from Austrian farms used for on-farm semen collection, for the presence for IgG/IgM in blood by ELISA (n=754) as well as for PCV2 DNA in semen (n=472) and if positive, also in blood of a few boars by nested PCR and sequencing. A total of 420 boars were tested for both PCV2 in semen and antibodies in blood. Boars were aged between 8 and 82 months at sampling. None of the boars tested positive for IgM but 60.1% did for IgG. PCV2 DNA was detected in 86 (18.2%) semen samples. Minor differences were found between boar populations with respect to the number of antibody positive boars and no differences for DNA in semen. Phylogenetic analysis of 28 sequences revealed a genetic diversity of PCV2 in semen within and between boar populations, with sequences belonging to both PCV2 genotypes 1 and 2. Mean nucleotide sequence identity was 95.7%, with maximum pairwise difference of 8.8%. Boars < or =16 months were tested more frequently positive for IgG (P<0.001) and for PCV2 DNA in semen (P<0.05) than older boars. Of 80 boars tested positive in semen, 34 (42.5%) were antibody negative. A total of 58 semen positive boars with (n=33) and without (n=25) IgG were all tested negative for PCV2 DNA in serum. In conclusion, this study demonstrated the ubiquity of PCV2 in the Austrian and German boar population. Genetically diverse PCV2 can be encountered in boar semen. Shedder boars cannot be detected on the basis of serology. There is an apparent possibility of PCV2 being transmitted through semen.

Tuberculosis (TB) in wild boar (Sus scrofa) may be affected by coinfections with other pathogens, such as porcine circovirus type 2 (PCV2). Therefore, sanitary measures focused on controlling PCV2 could be useful in reducing the impact of TB in this wild suid. The aim of this study was to explore whether vaccination against PCV2 targeting young animals affects TB prevalence and TB severity in wild boar. The study was conducted on a game estate in mid-western Spain. Seventy animals of ages ranging from 4 to 8 months were captured, individually identified, vaccinated against PCV2 and released, forming a vaccinated group. Not-captured animals cohabiting with the vaccinated wild boar constituted the control group. Animals from both groups were hunted between 2013 and 2016 and a TB diagnosis based on pathological assessment and microbiological culture was made in all of them. The effect of PCV2 vaccination on TB prevalence and severity was explored using generalized lineal models. Whereas TB prevalence was similar in vaccinated and control groups (54.55 vs. 57.78%), vaccinated animals showed less probabilities to develop generalized TB lesions. Furthermore, mean TB severity score was significantly lower in vaccinated animals (1.55 vs. 2.42) suggesting a positive effect of PCV2 vaccination.

Superficial inguinal lymph nodes from 72 wild boars examined in a previous immunohistochemical (IHC) study on porcine circovirus type 2 (PCV2) were selected for a PCV2 polymerase chain reaction (PCR) analysis. Four of these lymph nodes were PCV2-IHC strongly positive with PMWS histological lesions (outcome 1), 6 weak to mild PCV2-IHC positive without PMWS histological lesions (outcome 2) and 62 PCV2-IHC negative. Considering IHC the gold standard for diagnosis, the aims of the study were to evaluate the suitability of the PCV2-DNA extraction from formalin-fixed and paraffin-embedded (FFPE) tissue and the sensitivity and specificity of PCR under two IHC interpretations criteria: (A) the sample was considered positive if the result was outcome 1; (B) the sample was considered positive if the result was outcome 1 or 2. Under (A) criteria, sensitivity and specificity of PCR were 100% and 89.7%, respectively; the Cohen's Kappa coefficient was 0.49. Under (B) criteria, sensitivity and specificity of PCR were 80.0% and 95.2%, respectively; the Cohen's Kappa coefficient was 0.72. The high Cohen's Kappa coefficient under the (B) interpretative criteria indicates good agreement between the two methods. In conclusion, 1) DNA extracted from FFPE specimens of wild boar is suitable for PCR and further represents a screening test for PCV2/PCVD (PCV2 Diseases) investigations in wild boar as well; 2) routine histological sampling can also be useful for PCV2 virological studies in wild boar.

The lesions observed in 16 wild boars, hunted in central Italy, led to the suspect that could be related to the infection by porcine circovirus type 2 (PCV2). The animals had macroscopic and histological lesions in the lungs, tonsils, and bronchial lymph nodes. PCV2 was detected in tissue samples by polymerase chain reaction (PCR) and immunohistochemistry and it was isolated in newborn swine kidney cell cultures. From the infected cell culture supernatant, the presence of PCV2 DNA was confirmed by real-time PCR whereas virus particles were observed by electron microscopy. These diagnostic data indicate that PCV2 can infect and cause disease in Sus scrofa subspecies other than domestic swine and it is present in the wild boar population in central Italy.

Co-infections with parasites or viruses drive tuberculosis dynamics in humans, but little is known about their effects in other non-human hosts. This work aims to investigate the relationship between Mycobacterium bovis infection and other pathogens in wild boar (Sus scrofa), a recognized reservoir of bovine tuberculosis (bTB) in Mediterranean ecosystems. For this purpose, it has been assessed whether contacts with common concomitant pathogens are associated with the development of severe bTB lesions in 165 wild boar from mid-western Spain. The presence of bTB lesions affecting only one anatomic location (cervical lymph nodes), or more severe patterns affecting more than one location (mainly cervical lymph nodes and lungs), was assessed in infected animals. In addition, the existence of contacts with other pathogens such as porcine circovirus type 2 (PCV2), Aujeszky''s disease virus (ADV), swine influenza virus, porcine reproductive and respiratory syndrome virus, Mycoplasma hyopneumoniae, Actinobacillus pleuropneumoniae, Haemophilus parasuis and Metastrongylus spp, was evaluated by means of serological, microbiological and parasitological techniques. The existence of contacts with a structured community of pathogens in wild boar infected by M. bovis was statistically investigated by null models. Association between this community of pathogens and bTB severity was examined using a Partial Least Squares regression approach. Results showed that adult wild boar infected by M. bovis had contacted with some specific, non-random pathogen combinations. Contact with PCV2, ADV and infection by Metastrongylus spp, was positively correlated to tuberculosis severity. Therefore, measures against these concomitant pathogens such as vaccination or deworming, might be useful in tuberculosis control programmes in the wild boar. However, given the unexpected consequences of altering any community of organisms, further research should evaluate the impact of such measures under controlled conditions. Furthermore, more research including other important pathogens, such as gastro-intestinal nematodes, will be necessary to complete this picture.

Enzootic pneumonia (EP) caused by Mycoplasma hyopneumoniae has a significant economic impact on domestic pig production. A control program carried out from 1999 to 2003 successfully reduced disease occurrence in domestic pigs in Switzerland, but recurrent outbreaks suggested a potential role of free-ranging wild boar (Sus scrofa) as a source of re-infection. Since little is known on the epidemiology of EP in wild boar populations, our aims were: (1) to estimate the prevalence of M. hyopneumoniae infections in wild boar in Switzerland; (2) to identify risk factors for infection in wild boar; and (3) to assess whether infection in wild boar is associated with the same gross and microscopic lesions typical of EP in domestic pigs. Nasal swabs, bronchial swabs and lung samples were collected from 978 wild boar from five study areas in Switzerland between October 2011 and May 2013. Swabs were analyzed by qualitative real time PCR and a histopathological study was conducted on lung tissues. Risk factor analysis was performed using multivariable logistic regression modeling. Overall prevalence in nasal swabs was 26.2% (95% CI 23.3–29.3%) but significant geographical differences were observed. Wild boar density, occurrence of EP outbreaks in domestic pigs and young age were identified as risk factors for infection. There was a significant association between infection and lesions consistent with EP in domestic pigs. We have concluded that M. hyopneumoniae is widespread in the Swiss wild boar population, that the same risk factors for infection of domestic pigs also act as risk factors for infection of wild boar, and that infected wild boar develop lesions similar to those found in domestic pigs. However, based on our data and the outbreak pattern in domestic pigs, we propose that spillover from domestic pigs to wild boar is more likely than transmission from wild boar to pigs.

We studied the seroprevalence of six reproductive pathogens in Spanish hunter-harvested wild boar females. The sample was representative of the hunting harvest in the studied hunting estates. Mean antibody prevalences were: 60.6+/-0.06% for Aujeszky's disease virus (ADV), 56.6+/-0.09% for porcine parvovirus (PPV), 51.8+/-0.06% for porcine circovirus type 2 (PCV2), 29.7+/-0.09% for Brucella spp. and 36.3+/-0.1% for Toxoplasma gondii. We did not detect antibodies against porcine reproductive and respiratory syndrome virus (PRRSv). ADV seroprevalence was associated with PPV and PCV2 seroprevalence in Spanish wild boar females. Ovulation rate in the studied wild boar females was 4.41+/-0.16 (n=120), mean litter size was 3.91+/-0.16 (n=82) and the partial resorption index 0.92+/-0.17 (n=66). Ovulation rate and litter size were statistically associated with age. T. gondii seroprevalence was negatively related to ovulation rate and partial resorption index. Wild boars from managed fenced estates had antibodies against more pathogens than wild boars from open estates. Potential relations between management of wild boar populations and exposure of individuals to different reproductive pathogens are discussed.

Wild boar (Sus scrofa ssp scrofa) is a host of porcine circovirus 2 (PCV-2), and infection in this species has been reported in many countries. This study sampled tissues from 348 wild boars in Italy in the provinces of Bologna, Padua and Pisa. No clinical signs or macroscopic lesions were recorded in the sampled animals. Immunolocalisation for PCV-2 was performed on tissues, and viral antigen was disclosed in 38 animals, 11 immunohistochemistry (IHC) positive out of 148 (7.43%) sampled in northern Italy and 27 immunofluorescence (IF) positive out of 200 (13.5%) sampled in central Italy, respectively. Histologically, 19 cases displayed post-weaning multisystemic wasting syndrome (PMWS) histological lesions, but PCV2 was only proved in nine of them (three from Bologna and Padua by IHC and six from Pisa by IF). Animals aged 7 to 8 months were the most affected by PCV2 infection, suggesting an important role of this age class in the epidemiology of the virosis in the tested populations. Moreover, the study confirmed that wild boar can be a host of PCV-2, and showed that the virus and PMWS is widespread and endemic in wild boar in Italy as in other countries.

A large number of protected areas worldwide have been impacted by biological invasions, threatening the biodiversity they aim to protect. The wild boar (Sus scrofa) is one of the most threatening invasive species in Argentina, already occupying many ecoregions, including the central Monte Desert. However, there are no studies regarding the use that wild boars make of this invaded biome and what factors (climate or landscape) determine or contribute to the establishment of this species. The objectives of this study were to assess habitat use of the wild boar at spatial and temporal scales in the central Monte Desert, and to assess if climatic factors influence its abundance. Our results show that, at habitat-level the wild boar exhibited preferences for a particular habitat (Larrea shrubland) for feeding. At microhabitat-level, we found a positive association between herb cover and wild boar presence. In addition, we found a strong and positive association between the number of days with low temperatures and the number of wild boar signs registered. Therefore, we consider that in the central Monte Desert, habitat selection by wild boars is most likely determined by a maximization of food intake and a minimization of exposure to high temperature.

We determined prevalence of antibody to selected viral pathogens important for domestic pigs and livestock in 556 wild boar (Sus scrofa) sera collected during 2005-06 and 2009-10 in four counties in Croatia. These counties account for an important part of the Croatian commercial pig production and have a high density of wild boars. Samples were tested for antibodies to porcine parvovirus (PPV), Aujeszky's disease virus (ADV), porcine circovirus type 2 (PCV2), swine influenza virus, porcine respiratory and reproductive syndrome virus (PRRSV), porcine respiratory coronavirus (PRCV), transmissible gastroenteritis virus, and swine vesicular disease virus (SVDV). Antibodies to all of the infectious pathogens except SVDV were detected. There was a statistically significant difference in prevalence between the two periods for PPV, ADV, PCV2, PRRSV, and PRCV, with a higher prevalence of PPV and ADV in the 2009-10 period (P<0.05). During the same period, the prevalence of PCV2, PRRSV, and PRCV was lower (P<0.05). Our results provide information on the current disease exposure and health status of wild boars in Croatia and suggest that wild boars may act as a reservoir for several pathogens and a source of infection for domestic pigs and other livestock as well as humans, especially for ADV.

Hybridization between wild boar (Sus scrofa) and domestic pig occurred in the past and still occurs today, having great evolutionary and management implications. In fact, genetic introgression from the domestic form may alter traits like behavior, reproduction rate, and immunology in wild populations, with likely demographic impacts. Thus, it is crucial to understand under what conditions hybridization occurs in S. scrofa. Captive crosses with domestic pigs (released or escaped) have been suggested to constitute the major source of the spread of domestic genes into wild boar populations. However, to date, few studies have assessed the degree of admixture in farmed animals in comparison to the surrounding wild populations. With this purpose, we analyzed microsatellite loci in wild boar sampled in breeding stations and in the local wild population in two Italian regions (Sardinia and Piedmont). Both captive populations had lower allelic richness than the corresponding wild population, but a similar expected heterozygosity. In Piedmont, introgression from the domestic form into the wild population seems to be extremely low, while there are significant signs of admixture in the sampled breeding stations. In Sardinia, instead, the captive sample did not differ significantly from the wild population, which showed moderate signs of introgression. We conclude that hybridization in nature seems to play the key role in Sardinia, while intentional hybridization in captivity is the major source of introgression in Piedmont. Our findings emphasize the need for a routine genetic monitoring of wild boar captive populations, coupled with reference data on the neighboring wild populations.

The coexistence of wild boars and domestic pigs across Eurasia makes it feasible to conduct comparative genetic or genomic analyses for addressing how genetically different a domestic species is from its wild ancestor. To test whether there are differences in patterns of genetic variability between wild and domestic pigs at immunity-related genes and to detect outlier loci putatively under selection that may underlie differences in immune responses, here we analyzed 54 single-nucleotide polymorphisms (SNPs) of 19 immunity-related candidate genes on 11 autosomes in three pairs of wild boar and domestic pig populations from China, Iberian Peninsula, and Hungary. Our results showed no statistically significant differences in allele frequency and heterozygosity across SNPs between three pairs of wild and domestic populations. This observation was more likely due to the widespread and long-lasting gene flow between wild boars and domestic pigs across Eurasia. In addition, we detected eight coding SNPs from six genes as outliers being under selection consistently by three outlier tests (BayeScan2.1, FDIST2, and Arlequin3.5). Among four non-synonymous outlier SNPs, one from TLR4 gene was identified as being subject to positive (diversifying) selection and three each from CD36, IFNW1, and IL1B genes were suggested as under balancing selection. All of these four non-synonymous variants were predicted as being benign by PolyPhen-2. Our results were supported by other independent lines of evidence for positive selection or balancing selection acting on these four immune genes (CD36, IFNW1, IL1B, and TLR4). Our study showed an example applying a candidate gene approach to identify functionally important mutations (i.e., outlier loci) in wild and domestic pigs for subsequent functional experiments.

Mycoplasma hyopneumoniae (Mhyo) is the principal etiological agent of enzootic pneumonia (EP), one of the most economically important diseases in the pig production industry worldwide. Although swine and wild boars (Sus scrofa) share susceptibility to Mhyo infection, information regarding the dynamics of Mhyo infections and pathology in wild boars is currently limited. Therefore, the aim of this study was to determine the spatiotemporal dynamics of Mhyo infections in free-living wild boars through serological ELISA testing. Additionally, the presence of EP-like gross lesions and their association with the presence of Mhyo, based on a PCR assay, were assessed. Over a period of 5 years, antibodies against Mhyo were detected in 655 (30.0 %) of the 2,177 analyzed wild boars. A generalized temporal increase of seroprevalence coupled with an increase of mean antibody titers of seropositive individuals was found. Moreover, a similar seroprevalence between age classes associated with higher antibody titers of younger individuals indicated the wild boars were infected during the early stages of their life. Out of 99 lungs tested, 43 showed EP-like lesions and 45 were PCR positive for the presence of Mhyo DNA. The lung lesion scores were related to the sex and age of the wild boars, with young individuals having higher lung scores than others, and there was a positive association with the Mhyo PCR-positive status. The temporal increase of the Mhyo seroprevalence and infection association with EP-like lesions, when coupled with the spreading of wild boar populations, raises concerns on the epidemiological role of this species.

The objectives were to determine whether the amount of porcine circovirus type 2 (PCV2) shed in semen increased in boars experimentally coinfected with Mycoplasma hyopneumoniae (MHYO), and whether PCV2 vaccination of boars prior to PCV2 exposure reduced PCV2 viremia and virus shedding in semen. Twelve specific-pathogen-free PCV2- and MHYO-naïve boars were randomly and equally assigned to one of four groups. Six boars were vaccinated against PCV2 (VAC) on Day 0; three PCV2 vaccinated and three non-vaccinated boars were inoculated with MHYO on Day 21, and all boars were challenged with PCV2 on Day 35. The four treatment groups included PCV2-Infected (I), VAC-PCV2-I, MHYO-PCV2-Coinfected (CoI), and VAC-MHYO-PCV2-CoI. Semen, blood swabs, feces, and serum samples were collected weekly until Day 70. All vaccinated boars had seroconverted to PCV2 by Day 35. Between Days 28 and 35, MHYO boars developed moderate respiratory disease, characterized by coughing, respiratory distress, mucopurulent nasal discharge and loss of body condition. One MHYO-PCV2-CoI boar died on Day 50. Boars in the PCV2-I and MHYO-PCV2-CoI groups had significantly higher PCV2 DNA loads in blood swabs than the remaining boars. Moreover, PCV2 vaccination significantly reduced the incidence and amount of PCV2 shedding in semen and feces. In summary, although concurrent MHYO infection did not influence PCV2 shedding patterns, coinfection of boars with PCV2 and MHYO resulted in severe clinical disease and viral shedding was significantly decreased by PCV2 vaccination.

Eleven isozyme systems were investigated in wild boar (Sus scrofa ferus L.) by means of horizontal starch gel-electrophoresis using liver and kidney extracts. Samples of 103 specimen that originated from three adjunct localities from Rhineland-Palatinate (Fed. Rep. Germany) were analysed. AAT, ACO, GDH, GPDH, LDH, ME, MPI, MR, PGDH, and PGI were invariant. Genetic polymorphism is described for PGM. The genetic polymorphism at Pgm2 is explained by a biallelic model. Observed genotypic structure did not differ significantly from Hardy-Wein-berg-proportions at this gene locus. The degree of heterozygosity is 58,3 % and Wright's Fixation Index F =–0.166 at Pgm2. The allelic structure at this gene locus differed to a great extent from that found in domestic swine, i. e. in domestic swine the faster migrating Pgm2 allozyme was only found in relative low frequencies compared to wild boar. The genetic polymorphisms were moderate in the wild boar population compared to electrophoretic data of other animal species.

While motorways are often assumed to influence the movement behaviour of large mammals, there are surprisingly few studies that show an influence of these linear structures on the genetic make-up of wild ungulate populations. Here, we analyse the spatial genetic structure of red deer (Cervus elaphus) and wild boars (Sus scrofa) along a stretch of motorway in the Walloon part of Belgium. Altogether, 876 red deer were genotyped at 13 microsatellite loci, and 325 wild boars at 14 loci. In the case of the red deer, different genetic clustering tools identified two genetic subpopulations whose borders matched the motorway well. Conversely, no genetic structure was identified in the case of the wild boar. Analysis of isolation-by-distance patterns of pairs of individuals on the same side and on different sides of the motorway also suggested that the road was a barrier to red deer, but not to wild boar movement. While telemetry studies seem to confirm that red deer are more affected by motorways than wild boar, the red deer sample size was also much larger than that of the wild boars. We therefore repeated the analysis of genetic structure in the red deer with randomly sub-sampled data sets of decreasing size. The power to detect the genetic structure using clustering methods decreased with decreasing sample size.

The involvement of secretions from boar submaxillary salivary glands in mediating the induction of precocious puberty in the gilt was investigated as follows. Forty-eight Large White × (Large White × Landrace) prepubertal gilts from 12 litters were randomly allocated within litters by weight, to four treatment groups of six, in two replicates, at 145 days of age. Treatments commencing at a mean group age of 165 days, were: (1) control (no boar exposure); (2) gilts exposed to a mature sialectomised boar (submaxillary salivary glands were removed at 9 weeks of age); (3) gilts exposed to a mature sham-operated boar; (4) gilts exposed to a mature unoperated boar.Boar exposure occurred for 30 min per day for 75 days, or until pubertal oestrus was observed. Gilts showing pubertal oestrus were removed and slaughtered. Ovaries were examined to confirm reproductive status. Gilts failing to exhibit oestrus by 240 days of age were slaughtered and nominally ascribed a pubertal age of 245 days. Age at puberty was significantly earlier in all three boar-exposed treatments than in the control treatment (P<0.05 for treatments 2 and 3, P<0.001 for treatment 4; median ages at puberty being 227.0, 203.5 , 202.0 and 179.0 days for treatments 1 to 4 respectively). No frothy saliva was ever produced by the sialectomised boar, and chromatographic analysis of saliva produced by the sham-operated boar during mating revealed very low levels of 16-androstene pheromones, while levels in the unoperated boar's saliva were normal. These results provide further evidence for an important role of boar salivary pheromones in the induction of precocious puberty attainment in the gilt.
