Aflatoxin B1 (AFB1) is a carcinogenic mycotoxin found in feeds and in airborne grain dusts. Aflatoxin B1 requires biotransformation to the AFB1-8,9 epoxide (AFBO) by a bioactivation system and subsequent covalent binding to DNA or proteins, to exert its carcinogenic potential. The lung contains cytochrome P450, prostaglandin-H-synthase, lipoxygenase, epoxide hydrolase and other bioactivation enzymes, and is thus a potential target for the effects of AFB1 via the routes of inhalation and ingestion. The A549 human epithelioid lung cell line and the methylthiazol tetrazolium (MTT) bioassay were used to investigate the cytotoxicity of AFB1 and its chemically synthesised epoxide (AFBO) in vitro. Statistical analysis of the MTT results indicated that there were overall significant differences between the control and both the AFB1-treated (p < 0.0001) and AFBO-treated cells (p = 0.00 2). However, there was no significant difference between AFB1 and AFBO-treated cells, when the entire range of concentrations were assessed against each other (p = 0.2877). Whenanalysed at each concentration, only at 0.01 mM was there a significant difference between the effects of AFB1 and AFBO (p = 0.0358). The intake of mycotoxin-contaminated feeds can lead to nutrient losses and may have adverse effects on animal health and on productivity. The aims of this study were (1) to determine the mycobiota present in poultry feed samples, and (2) to evaluate the natural occurrence of aflatoxin B1, fumonisin B1 and zearalenone. Fungal counts were similar between all culture media tested (103 CFU g−1). The most frequent genus isolated was Penicillium spp. (41.26%) followed by Aspergillus spp. (33.33%) and Fusarium spp. (20.63%). High precision liquid chromatography was applied to quantify aflatoxin B1 and fumonisin B1. Thin layer chromatography was used to determine zearalenone levels. Aflatoxin B1 values ranged between 1.2 and 17.5 μg kg−1. Fumonisin B1 levels ranged between 1.5 and 5.5 μg g−1. Zearalenone levels ranged between 0.1 and 7 μg g−1. The present study shows the simultaneous occurrence of two carcinogenic mycotoxins, aflatoxin B1 and fumonisin B1, together with another Fusarium mycotoxin (zearalenone) in␣feed intended for poultry consumption. Many samples contained AFB1 levels near the permissible maximum and it could affect young animals. A synergistic toxic response is possible in animals under simultaneous exposure.  相似文献

In Argentina, there is rather little information about the natural occurrence of mycotoxins in feedstuffs. The aim of this work was to determine the fungal flora and natural incidence of aflatoxin B1 (AFB1), zearalenone (ZEA) and deoxynivalenol (DON) in poultry feeds from 5 factories of Río Cuarto, Córdoba. Three hundred samples were taken from May 1995 to May 1996. Fungal counts of poultry feeds ranged 104 to 106 CFU g-1. The lowest counts were obtained on the first months from the sampling (May to September 1995) with mean values significantly different from those found at the last of the sampling (October 1995 to April 1996). The most prevalent species isolated of poultry feed samples belonged to the genera Penicillium that was present in 98% of the samples, Fusarium (87%) and Aspergillus (52%). Fusarium species isolated were: F moniliforme in 73% of the samples, F subglutinans (35%), F graminearum (20%) and within Aspergillus species: A. parasiticus (33%) and A. flavus (8%) were identified. In poultry feeds aflatoxin B1 (AFB1) was the most significant mycotoxin with levels ranging from 17 to 197 ng/g. For deoxynivalenol (DON) the levels ranged from 240 to 410 ng/g. Only three out of 300 samples were contaminated with zearalenone (ZEA) in concentrations of 30, 120 and 280 ng/g. These are preliminary data on this subject in our region. Fermentation of yoghurt and acidified milks containing aflatoxin B1 (AB1) were studied. AB1 added to milk before fermentation at concentrations of 600, 1000 and 1400 g/kg was reduced in yoghurts (pH 4.0) by 97, 91 and 90%, respectively. Coagulation time was approximately the same as in the controls. Streptococci had longer chains than those in the controls. The main decrease of AB1 occurred during the milk fermentation. A decrease of AB1 (conc. 1000 g/kg) in milks acidified with citric, lactic and acetic acids (pH 4.0) was 90, 84 and 73%, respectively.  相似文献

Physiological processes of mung seeds (Vigna radiata variety Pusa Baishakhi) and their germination were found to be affected by different concentrations of aflatoxin B1. Inhibition in seed germination, seedling growth, chlorophyll, protein and nucleic acid syntheses was found to be due to aflatoxin B1. The aim of the study was to determine the effect of repeated applications of aflatoxin B1 (AFB1) on immunocompetent cells (CD3 T cells) and alkaline phosphatase in the intestinal mucosa. Mice were orally treated with AFB1 for 24 days. The mucosa of the intestine showed a significant decrease in the number of CD3 T cells and a significantly lower level activity of alkaline phosphatase on day 24 in AFB1 treated mice. Similarly, with changes in the small intestine, qualitative haematological parameters were modified in systemic immunity as lymphopenia, and neutropenia, monocytopenia. AFB1 treated animals showed reduction in body weight gain and increased liver weight. We supposed that changes found in the small intestine are secondary to primary systemic haematological lesions. The decrease in CD3 T cells suggests a connection with the decrease in the host's resistance to infectious diseases. Anin vitro study using single concentration and isotherm adsorption was carried out to evaluate the capacity of Vietnamese produced zeolite and bentonite to adsorb aflatoxin B1 (AFB1) in simulated gastrointestinal fluids (SGFs), and a commercial sorbent hydrated sodium calcium aluminosilicate (HSCAS) was used as reference. In this study, AFB1 solution was mixed with sorbents (0.3, 0.4 and 0.5% w/v) in SGFs at pH 3 and pH 7 and shaken for 8 h, centrifuged and the supernatant measured by Vicam fluorometer. Adsorption of AFB1 onto zeolite and bentonite varied according to the pH of SGFs and was lower than HSCAS. Linearity between the increased amount of AFB1 adsorbed on sorbents and the decrease of sorbent concentration was observed for bentonite and HSCAS, except for zeolite in SGFs at pH 7. The observed maximum amounts of AFB1 adsorbed on bentonite and HSCAS were 1.54 and 1.56 mg/g, respectively. The adsorption capacities of bentonite and HSCAS for AFB1 were 12.7 and 13.1 mg/g, respectively, from fitting the data to the Freundlich isotherm equation. Improvement in processing and purification for bentonite is needed to enhance the surface area, which would probably result in better adsorptive capacity for this sorbent.  相似文献

Within this research project, the LCI (Lebensmittelchemisches Institut des Bundesverbandes der Deutschen Süßwarenindustrie e. V.) conducted systematic studies to determine the thermal stability of the mycotoxins ochratoxin A (OTA) and aflatoxin B1, as the available literature provides contradictory data. Firstly, the said mycotoxins in pure form were subjected to thermal treament. Secondly, tests were conducted to determine the influence of certain matrix substances, including carbohydrates and proteins, on the thermal decomposition behaviour of the mycotoxins. As a result it can be said that OTA seems to be stable up to 180 °C; however aflatoxin B1 was almost completely degraded at heating temperatures of 160 °C and above. In several model assays it could further be shown that the degradation of mycotoxins is improved by the existence of certain matrix compounds.  相似文献

In conditions of constant darkness, interindividual variability in the clarity of circadian rhythmicity was observed in sexually immature young quail, with birds classified as more or less rhythmic or arrhythmic. The relative clarity of this circadian rhythm was observed on the actograms by measurement of the autocorrelation coefficient ratio over 12 cycles. Autocorrelation coefficients were calculated from sequential series of total activity over 12-minute periods. Crosses of selected phenotypes with different clarities of rhythmicity were conducted in order to study the possibility of selection of this characteristic. From a random population (N = 42, twice), pairs of the most rhythmic birds (3 families), and pairs of arrhythmic birds (4 families) were reared. Autocorrelation coefficient ratios of Fl birds from rhythmic families (N = 54) were greater than those of Fl birds (N = 48) from arrhythmic families (t-test, p <. 0001). These ratios in offspring were significantly correlated with that of the mean parent of each clutch of siblings (N= 102, r =.35, p =. 0003). This result was maintained in a second generation (F2) of birds, for which significant differences in expressed rhythmicity were observed. That is, autocorrelation coefficient ratios of F2 birds from two rhythmic families (TV = 30) were greater than those of F2 birds from arrhydimic families (N = 20) (t-test, p =. 039). Comparison of F2 outbred and inbred birds from rhythmic pairs showed greater values of autocorrelation coefficient ratios in the case of inbred birds (N = 16) than for outbred birds (N = 30; t-test, p =. 036). There was no difference between outbred (N = 20) and inbred birds (N = 15) from arrhythmic pairs. Therefore, selection of a rhythmic strain seems possible, whereas crosses between two arrhythmic birds may also give rise to rhythmic birds. Comparisons between rhythmic birds of different families did not show differences in the free-running period of the circadian rhythm, which is true also for rhythmic birds bred from two arrhythmic parents. Therefore, our selection procedure did not seem to be based on the characteristics of the pacemaker itself, but rather on a downstream event. Although the parents were not selected on the basis of quantity of activity per cycle or on the duration of the active phase, significant differences among the offspring of different families were shown. In a preliminary study, 256 feed samples collected from different parts of Northern India were examined for the presence of aflatoxigenic strains of Aspergillus flavus/parasiticus and for detection of Aflatoxin B1 (AFB1). Out of 198 A. flavus and 15 A. parasiticus strains isolated, 76% and 86% respectively, were found to be toxigenic. Aflatoxin B1 content of these feeds, as estimated by thin layer chromatography (TLC) and enzyme linked immunosorbent assay (ELISA) were very high (average 0.412 ± 0.154 ppm) in comparison to the permissible Indian regulation level (0.03 ppm). Many types of aflatoxin cause problems for both public and animal health. Aflatoxin B1 (AFB1) is the most toxic and commonly encountered fungal toxin that appears in poultry feed and in feeds stored under unsuitable conditions. AFB1 decreases feed quality, egg production and fertility of hatching eggs. Also, AFB1 alters the development of embryos by infecting eggs. We investigated using sequence analysis the changes caused by different concentrations of AFB1 on the promoter sequences of the growth hormone regulated gene-1 (GHRG-1) in chick embryo at 13, 17, 19 and 21 days incubation. DNA isolated from the liver of chick embryos treated with different concentrations of AFB1 was separated using agarose gel electrophoresis to detect apoptosis, and DNA interaction with AFB1 was investigated using plasmids to detect changes in electrophoretic mobility and their effects on DNA. Base changes of the promoter sequences of GHRG-1 in 5 ng/egg, 15 ng/egg and 40 ng/egg doses of AFB1 were increased on day 19 compared to base changes of the same AFB1 doses on day 13. We also found that AFB at different concentrations changed the mobility of DNA by binding to it, and that high doses of AFB1 destroyed DNA. The DNA interaction study using plasmid demonstrated that AFB1 at high doses was bound to plasmid DNA, slowed its mobility and inhibited restriction cuts.  相似文献

Seed germination and seedling growth as well as chlorophyll and carotenoid contents of mustard and gram seeds were inhibited or reduced significantly due to the treatment of different concentrations (100, 250, 500, 1000 and 2000 ng/ml) of aflatoxin B1. The range of inhibition in all the parameters was found to be directly influenced by the concentrations of toxin applied. Summary Chronic exposure of a cloned rat hepatocyte culture (RL-PR-C) to a subtoxic, sublethal dose of aflatoxin B1 resulted in malignant transformation. Continuous exposure to aflatoxin B1 caused increasing tumorigenic potential as tested by back injection into isogenic animals. Control cultures exhibited spontaneous transformation, although approximately 20 passages beyond the chemically induced event. Neither aflatoxin-treated nor control cultures exhibited cytopathological morphology, formation of cell foci, growth in soft agar, or irregular fibroblast-like growth patterns that could be specifically related to the onset of tumorigenic potential. In general, those parameters commonly used to monitor fibroblast cultures for transformation in vitro were not applicable for assessing the tumorigenic potential of these epithelial cells. Karyotypic analyses revealed no specific chromosomal aberrations associated with aflatoxin treatment; however, chromosomal instability was a property of the tumorigenic cell populations. Injection of both aflatoxin-treated and control cultures at passage 56 resulted in tumors indicative of both carcinoma and sarcoma indicating to us the multipotency of these epithelial cells transformed in vitro.  相似文献

【背景】丁酸梭菌是专性厌氧的新一代芽孢益生菌,耐热、耐酸、抗逆性强,极具应用价值和开发前景。【目的】优化丁酸梭菌发酵培养基并初步研究其发酵液对黄曲霉菌的抑制作用和降解黄曲霉毒素B1 (aflatoxin B1, AFB1)的能力。【方法】利用响应面法对发酵培养基进行优化,采用牛津杯法对丁酸梭菌发酵液抑制黄曲霉菌生长进行研究,并通过酶联免疫法测定发酵液对AFB1的降解能力。【结果】优化后的发酵培养基为:葡萄糖18.1g/L,大豆蛋白胨29.7g/L,磷酸氢二钾3.8 g/L,氯化钠2.0 g/L,乙酸钠4.0 g/L,结晶硫酸镁1.2 g/L,L-半胱氨酸盐酸盐0.3 g/L。优化后的丁酸梭菌生物量由8.99×108个/mL提高至2.28×109个/mL,是优化前的2.54倍。丁酸梭菌发酵液对致病真菌黄曲霉菌的抑菌效果十分显著,其上清液经浓缩后对AFB1降解72h的降解率达到68.65%,初步分析表明上清液中对AFB1  相似文献

Levels were evaluated of Aflatoxin B1 (AFB1) exposure through the intake of AFB1-contaminated foods among Koreans. Average daily intake, excess cancer risk, and population risk of AFB1 from food intake were calculated from Koreans with age greater than 20 years. Average daily intake of AFB1 by food ingestion was 0.064 ~ 0.361 ng/kg bw/ day. Excess cancer risk of AFB1 in food with the surface antigen of the hepatitis B-virus negative (HBsAg?) and positive (HBsAg+) population was 5.76 × 10? 7~ 3.25 × 10? 6 and 1.47 × 10? 5~ 8.31 × 10? 5, respectively. Application of JECFA's two tentative maximum levels (MLs) (10 μ g/kg and 20 μ g/kg) resulted in the population risk of 0.0009 ~ 0.0075 and 0.0014 ~ 0.0079 cancers/year/100,000 people, respectively, indicating that lower ML does not give a significant reduction of the risk. However, because Korea has a high incidence of HBsAg+ individuals, adoption of lower ML may be a reasonable decision to protect a high risk group. This is the first report on the risk assessment of AFB1 in food among the Korean population. Our study will contribute to providing the solid basis of establishing the maximum levels of AFB1 exposure through food ingestion in Korea as well as other countries.  相似文献

During hot and humid seasons, extensive rot of sour lime was observed to be caused by Aspergillus flavus. In view of this, investigations were undertaken to obtain data on the production of various toxins by A. flavus during post harvest pathogenesis of sour lime. Sixty percent of the pathogenic A. flavus isolates were detected to be aflatoxin B1 producers in sour lime tissue. It was also noted that thirty three percent of aflatoxigenic A. flavus isolates had the potential to coproduce cyclopiazonic acid (CPA). Such aflatoxigenic isolates produced quantitatively more CPA (ranging from 250.0 to 2501.3 g/kg) than aflatoxin B1 (ranging from 141.3 to 811.7 g/kg) in the affected sour lime. This study demonstrates for the first time that sour lime are a favourable substrate for aflatoxin B1 and cyclopiazonic acid production by A. flavus isolates. On administration of aflatoxin B1 to whole parsley (Petroselinum crispum) plants, a derivative was formed, which was shown to be aflatoxicol by its chromatographic properties and mass spectrometry. Optimum conditions for the production of the derivative was on the second day after administration of the toxin to the plants, which were 90 days old after germination. Cell-free preparations of parsley were found not to produce aflatoxicol A from added aflatoxin B1; instead they formed two new derivatives, which from chromatographic properties, were shown to be more polar than either aflatoxin B1 or aflatoxicol A.  相似文献

In mice kept on a diet with no vitamin B1, the total content of thiamine in the brain decreased, and muscle contractions evoked by stimulation of the nerve in phrenico-diaphragmatic preparations obtained from such animals became weaker. The measurements were performed in Krebs solution with a decreased content of Ca2+ and increased concentration of Mg2+; values of the developed force were normalized with respect to those in normal Krebs solution. Thus, B1 hypovitaminosis results in a decrease in the efficacy of neuromuscular synaptic transmission. Some neurological symptoms typical of a deficiency of vitamin B1 in the organism can be related to this effect. In most organisms living in temperate zones, reproduction is under photoperiodic control. Although photoperiodic time measurement has been studied in organisms ranging from plants to vertebrates, the underlying molecular mechanism is not well understood. The Japanese quail (Coturnix japonica) represents an excellent model to study this problem because of the rapid and dramatic photoperiodic response of its hypothalamic‐pituitary‐gonadal axis. Recent investigations of Japanese quail show that long‐day‐induced type 2 deiodinase (Dio2) expression in the mediobasal hypothalamus (MBH) plays an important role in the photoperiodic gonadal regulation by catalyzing the conversion of the prohormone thyroxine (T4) to bioactive 3,5,3′‐triiodothyronine (T3). The T3 content in the MBH is approximately 10‐fold higher under long than short days and conditions, and the intracerebroventricular infusion of T3 under short days and conditions mimics the photoperiodic gonadal response. While Dio2 generates active T3 from T4 by outer ring deiodination, type 3 deiodinase (Dio3) catalyzes the conversion of both T3 and T4 into inactive forms by inner ring deiodination. In contrast to Dio2 expression, Dio3 expression in the MBH is suppressed under the long‐day condition. Photoperiodic changes in the expression of both genes during the photoinduction process occur before the changes in the level of luteinizing hormone (LH) secretion, suggesting that the reciprocal changes in Dio2 and Dio3 expression act as gene switches of the photoperiodic molecular cascade to trigger induction of LH secretion.  相似文献

The aim of the present study was to investigate the influence of nutritional deficiency and dietary addition of vitamins (B2, B6, and folate) on hepatic concentration of zinc and copper in rats. The experiment was performed on 260 growing male Wistar rats divided into 13 groups. Animals of 11 groups were fed isocaloric diets (14.7 MJ/kg) in which the 20% of energy was derived from protein. Another two groups of rats were offered diets with 9% or 4.5% of energy originating from protein. Animals of both mentioned groups and of the control group (20% of energy from protein) were offered diets ad libitum. The other 10 groups were offered 50% and 30% of the amount consumed in the control group. Eight groups, from those 10 restricted ones, were differentiated by dietary addition of vitamins B2 and B6 and folate (300% addition). Restricted feed intake did not affect the liver zinc concentration but significantly increased the copper concentration. The addition of vitamin B6 decreased the liver Zn concentration. The highest liver Cu concentration was noted in rats offered restricted diets to only 30% of intake in the control group and high in vitamin B2 and in rats supplemented with all of studied vitamins together. It suggests that vitamin B2 had the strongest impact on liver Cu concentration in rats fed restricted diets.  相似文献
