中国香菇自然群体的交配型因子分析*   总被引:2,自引:1,他引:2  
以自收集于中国14个省、区的53个野生香菇菌株分离得到的94个单核体为材料,进行了交配型因子分析。从该样本中鉴定出66个不同的A因子和72个不同的B因子,而且A、B不亲和性因子系列中的特异性因子均呈等概率分布。据此估算在中国香菇自然群体中存在121个特异的A因子和151个特异的B因子,表明中国香菇自然种质中蕴藏着丰富的遗传多样性。  相似文献

Polyphenylalanine synthesis with ribosomes and two separated, partially purified elongation factors (EF) was measured in cell-free systems from the archaebacteria Thermoplasma acidophilum and Methanococcus vannielii, in an eukaryotic system from rat liver and an eubacterial one with Escherichia coli ribosomes and factors from Thermus thermophilus. By substitution of heterologous EF-2 or EF-G, respectively, for the homologous factors, ribosome specificity was shown to be restricted to factors from the same kingdom. In contrast, EF-1 from T. thermophilus significantly cooperated with ribosomes from T. acidophilum.  相似文献

Seven protein synthesis initiation factors were isolated from Krebs II ascites cells using the procedures developed for the purification of the corresponding factors from rabbit reticulocytes. The ascites factors display identical characteristics in ion exchange chromatography and sucrose density gradient sedimentation. Based on their profiles in SDS polyacrylamide gels, the ascites factors have polypeptide profiles and molecular weights identical to those of the reticulocyte factors. Most significantly, each ascites factor is competent in replacing its corresponding reticulocyte factor in a reconstituted in vitro protein synthesizing system which is dependent on all seven factors.  相似文献

中国白灵侧耳自然群体的交配型因子分析   总被引:1,自引:0,他引:1  
为了研究我国新疆白灵侧耳自然种群的遗传多样性,估测自然种群的遗传丰度,对51株新疆野生白灵侧耳的84个原生质体单核体的交配型因子进行分析。在84个单核体中存在54个不同的A因子和59个不同的B因子。x2检测表明交配型因子A和B的系列因子均为等概率分布。据此估算我国白灵侧耳自然群体中有79个A因子,100个B因子,自然群体中的单核交配型总数79×100=7,900个,双核交配型总数31,201,050个。可见,新疆白灵侧耳遗传多样性丰富,新疆是我国白灵侧耳的野生种质资源宝库。  相似文献

Summary Previous studies have shown that extracts from mouse embryos at mid and late stages of development contain factors that exhibit transforming growth factor activity. The work reported here demonstrates that cultured mouse embryos at significantly earlier stages of development produce and release factors that exhibit the characteristic property of transforming growth factors. Specifically, the data demonstrate that embryos cultured from the blastocyst stage in serum-containing medium or in serum-free medium release factors that promote the anchorage-independent growth of normal rat kidney fibroblasts. It is shown that these factors are produced and released by cells derived from the inner cell mass and by trophoblasts. The precise developmental stage when production of these factors first begins has not been determined but our findings suggest that these factors are produced by cell types associated with early postimplatation embryos. The roles of climatic factors in plant invasions have drawn intense attention in the past. America was one of the major donors of the invasive plants in China. In this study, we investigated the roles of climatic factors in plant invasions from America into China through Akaike’s information criterion model analysis in terms of specific geographical origins, respectively. From south to north, decreasing trends of species diversity were observed on the plants from Central-South America and Mexico, while the greatest alien species diversity in mid-Chinese latitudes was observed on the invaders from North America; For the invaders from Central-South America and Mexico, climatic factors explained most of the spatial variations, while for those from North America, the roles of the climatic factors were weak. The role of the climatic factors in plant invasions may, in some extent, depend on the ecological characters inherited form the geographical origins and their accommodation to the climate of the invaded regions. If the invasive plants were introduced into the region with similar environment to their area of origin, the role of the climatic factors may be shadowed by other factors. However, for the invaders whom were introduced into the regions vastly different from their areas of origin, climatic barriers may be responsible for most of the spatial variations. The invasive plants from Central-South America and Mexico may have strong potential to invade regions at higher latitude in China in the scenario of global warming, while for the invaders from North America, the impacts of global warming may be shadowed by other factors.  相似文献

Drug-resistant gram-negative bacilli conferred with R factors were isolated with high frequencies from the intestinal tracts of ayu (Plecoglossus altivelis) cultured in ponds, in which chemotherapeutics had often been used, and with relatively low frequencies from ayu which received no administration of chemotherapeutics. Drug-resistant bacteria were also isolated at low frequencies from the intestinal tracts of wild ayu in rivers, as well as from the water of ayu-culturing ponds and some of them carried R factors. The drug-resistant bacteria carrying R factors were Aeromonas liquefaciens, Citrobacter, Enterobacter cloacae, Escherichia coli, Hafnia and unidentified strains. All the R factors were classified as the Fi-(F) type, except the two R factors detected in an E. coli strain and in an unidentified strain.  相似文献

Monoclonal antibody specific for T cell-derived human IgE binding factors   总被引:3,自引:0,他引:3  
A B cell hybridoma secreting monoclonal antibody against human IgE binding factors was obtained by immunization of BALB/c mice with partially purified IgE binding factors, and fusion of their spleen cells with SP-2/0-AG14 cells. The monoclonal antibody bound all of the 60,000, 30,000, and 15,000 dalton IgE binding factors from two T cell hybridomas and those from activated T cells of a normal individual. The antibody bound both IgE-potentiating factors, which had affinity for lentil lectin, and IgE-suppressive factors, which had affinity for peanut agglutinin. However, the monoclonal anti-IgE-binding factor bound neither Fc epsilon R on RPMI 8866 cells nor IgE binding factors from the B lymphoblastoid cells. A monoclonal antibody against Fc epsilon R on B cells (H107) bound the 60,000 and 30,000 dalton IgE binding factors from both T cell hybridomas and RPMI 8866 cells but did not bind the 15,000 dalton IgE binding factors from either T cells or B cells. The results indicate that T cell-derived IgE binding factors have a unique antigenic determinant that is lacking in both Fc epsilon R on B cells and B cell-derived IgE binding factors. The anti-IgE binding factor and anti-Fc epsilon R monoclonal antibodies both failed to stain cell surface components of IgE binding factor-producing T cell hybridomas. However, both antibodies induced the T cell hybridoma to form IgE binding factors. The results suggest that the T cell hybridomas bear low numbers of Fc epsilon R that share antigenic determinants with IgE binding factors secreted from the cells.  相似文献

研究城市扩展驱动力对于准确判断城市发展规律,剖析演化过程和预测城市未来扩展趋势具有重要意义,同时也能为制定合理的调控政策提供指导。以沈阳市1997—2010年城市建成区变化作为因变量,选取三大类10种驱动因子,利用增强回归树(BRT)和Logistic回归两种方法分析影响城市扩展的主要驱动力。结果表明:(1)BRT分析得到影响沈阳城市扩展的驱动因子从大到小依次是:距1997年城区距离、距河流距离、数字高程模型DEM、距高速公路和铁路距离、土地利用类型、开发区规划、GDP、人口密度、坡向和坡度;(2)Logistic回归分析得到影响沈阳城市扩展的前8位驱动因子依次是:开发区、距1997年城区距离、DEM、距高速公路和铁路距离、人口密度、距河流距离、农村居民点和坡度;(3)距1997年城区距离、DEM、距高速公路和铁路距离是影响沈阳城市扩展的主要驱动力,均位于主要因子的前四位;(4)总体来说,沈阳城市扩展受邻域因子影响最大,而自然因子的影响相对较小,社会经济因子则只有开发区和农村居民点对城市扩展影响较大。  相似文献

应用体细胞不亲和性反应、交配型因子分析和基因组DNA的RAPD分析,研究了一个分布于方圆约1km的6根倒木上的18个香菇野生菌株间的遗传差异。结果表明,该群体大多数菌株间配对(80.4%)体细胞不亲和,而同一倒木菌株间配对的体细胞亲和率达 62.5%。不同倒木菌株间未发现体细胞亲和的配对。该群体存在 11个特异的A因子和7个特异的B因子。同一倒木的菌株有的交配型因子相同,有的则不同。不同倒木的菌株大多数交配型因子不同,未发现交配型因子完全相同的菌株。RAPD分析显示,体细胞亲和的菌株,交配型因子完全相同的菌株,在基于DNA相似系数的遗传相关聚类中,首先聚为小类。总起来看,在自然群体中,香菇个体间的遗传差异与其空间分布之间存在一定的联系,随着空间距离的增大,菌株间的异质性相应增高。  相似文献

Two protein factors (A and B) have been partially purified from Escherichia coli supernatant which, in combination, are more effective than 0.5 M NH4Cl in stimulating ribosomes for AcPhe-tRNA and fMet-tRNA binding, for the puromycin reaction, and for incorporating acetylphenylalanine from AcPhe-tRNA into polypeptide. The factors appear to differ from the initiation factors, the elongation factor EF-T, and ribosomal proteins. Some uncertainty exists as to whether factor B is different from EF-G. To maximize the effect of the factors in initiator tRNA binding, we preincubated the ribosomes with the factors and carried out the binding assay for a short period at 15 degrees C. Maximal stimulation of binding occurred after about a 2-min preincubation at 37 degrees C. Longer preincubation times were required at 15 degrees C, and only slight stimulation was observed after preincubation at 0 degrees C. The extent of stimulation by the factors was not affected when the NH4Cl concentration was increased from 40 to 500 mM in the preincubation. The presence of both the 30S and 50S ribosomal subunits is required for the enhancement of AcPhe-tRNA binding. Polyphenylalanine synthesis carried out without AcPhe-tRNA is inhibited by the factors. It is suggested that the factors may act by inducing a structural rearrangement of the ribosomes.  相似文献

Thymidine incorporation into DNA is widely used to estimate rates of bacterial growth and secondary production in aquatic systems. The procedure requires the use of several conversion factors and assumptions to convert rates of thymidine uptake to rates of carbon production. Perhaps the most controversial of the conversion factors is that which converts rates of incorporation to the rate of cell production. During a year-long study in Lake Arlington, Texas, we empirically determined conversion factors from bacterial growth in filtered (1.0 m porosity) and diluted (1:9) lake water. Bacterial growth rate constants determined from changes in cell abundance were compared to growth rate constants estimated from empirically derived (both instantaneous and annually averaged), theoretical, and modeled conversion factors. Single value conversion factors (i.e., theoretical, or average of 19 empirically determined conversion factors) did not yield estimates of bacterial growth rate constants that compared favorably to growth estimates from changes in bacterial abundance. Conversion factors, determined from a regression model based on empirically determined conversion factors, gave annual growth estimates that were similar to those obtained from changes in cell abundance. 应用逐步回归法,分析了5~9月松嫩盐碱草地植物群落的生物量、丰富度、多样性和均匀度与同月土壤因子间的关系。松嫩盐碱草地植物群落特征受若干土壤因子的共同影响,不同月份影响因子的变化较复杂。5~7月土壤盐分和养分共同影响着松嫩盐碱草地植物群落生物量,但是土壤盐分因子与群落生物量的相关性更明显(5月的Ca2+质量分数,6月和7月的Mg2+质量分数)。土壤盐分因子在整个生长季内对松嫩盐碱草地植物群落生物量的直接作用都大于土壤养分因子的。6月时影响松嫩盐碱草地植物群落丰富度、Simpson指数和Shannon指数的土壤养分因子增加,其中土壤全氮质量分数的直接作用是所有土壤因子中最大的。8月植物群落生物量、Simpson指数和Shannon指数仅受土壤盐分因子影响,其中生物量与土壤pH值以及Simpson指数与土壤碱化度都是极显著负相关,Shannon指数与土壤含盐量显著负相关。6~9月的松嫩盐碱草地植物群落均匀度也同时受土壤养分和盐分因子的作用,其中6月和9月时土壤盐分因子对群落均匀度的直接作用更突出。生长末期(9月)松嫩盐碱草地植物群落生物量与土壤因子间的关系以及生长初期(5月)植物群落均匀度与土壤因子间的关系都不明显。  相似文献

Soluble factors obtained from human, rat and rabbit erythroid cell lysates are capable to stimulate basal and hormone activated adenylate cyclase of erythroid cell membranes from homologous sources. Extensive dialysis and removal of hemoglobin from the soluble factors do not modify their activity. Human erythrocyte soluble factors stimulate the human reticulocyte enzyme. Nevertheless human erythrocyte adenylate cyclase is not stimulated by either of the soluble factors. The presence of active soluble factors in human erythrocytes where the adenylate cyclase is no longer sensitive to these factors, as well as to guanylnucleotides or protaglandins, indicates that the enzyme has been altered during the maturation processes.  相似文献

Incubation of normal mouse spleen cells with homologous IgE resulted in the formation of soluble factors that inhibited rosette formation of mouse Fc epsilon R+ cells with IgE-coated ox erythrocytes. The soluble factors could be absorbed with mouse or rat IgE coupled to Sepharose and recovered from the beads by acid elution. However, the factors had no affinity for either human IgE or mouse IgG. The IgE-binding factors were derived from T cells. Production of the factors required Lyt1+ T cells and Fc gamma R+ cells, which suggests that the factors are derived from Fc gamma R+ Lyt 1+ T cells. The molecular size of IgE-binding factors was approximately 15,000 daltons. When IgE-binding factors were formed by BALB/c spleen cells, nearly one-half of the factors had affinity for lentil lectin, and the remaining half of the factors failed to bind to the lectin. The proportion of the two species of IgE-binding factors differed depending on mouse strains. The majority of the factors formed by B6D2F1 spleen cells had affinity for lentil lectin, but those formed by SJL spleen cells failed to bind to the lectin. The IgE-binding factors were also induced by incubation of normal spleen cells with polyinosinic-polycytidylic acid (pI:pC). The nucleotide stimulated splenic adherent cells to form "inducers" of IgE-binding factors, which in turn induced normal lymphocytes to form IgE-binding factors. The inducers of IgE-binding factors were inactivated (or neutralized) by antibodies specific for mouse Type I interferon. It was also found that purified mouse beta interferon could induce the formation of IgE-binding factors. IgE-binding factors induced by pI:pC consisted of two different molecules: one had a m.w. of 15,000 daltons, and another had a m.w. of between 40,000 and 60,000 daltons.  相似文献
