目的:建立固体制剂中包合型维生素K1的含量测定方法。方法:采用μBondapak C18,3.9×300mm,10μm,以甲醇-水(95:5)为流动相,流速1.0mL/min,检测波长248nm,进样20μL。结果:线性:维生素K1在6.4205~51.3640μg/mL浓度范围内与峰面积呈良好的线性关系;回收率:平均回收率为99.95%,RSD为1.03%。结论:本法简便、快速、准确,可作为固体制剂中维生素K1含量测定方法。  相似文献

葛根素注射液中葛根素含量的HPLC测定   总被引:4,自引:0,他引:4  
建立了一种简便、准确、可靠的葛根素测定方法。采用SymmetryC18(3.9mm×150mm,5μm)柱,流动相采用甲醇-水(体积比为3525),流速为1.00ml·min-1,检测波长为250nm。进样量在0.0368μg~0.589μg范围内线性关系良好,(r=0.9994)。该方法简便快速,准确可靠,可用于葛根素注射液的含量测定。同时,测定结果表明两种产品在含量上有差别,因此,保证和进一步提高葛根素注射液质量,并且规范质量标准势在必行。  相似文献

刺玫根对肾炎有显著疗效,从其中分离出一单株成分,经理化性质和光谱分析鉴定为齐墩果酸。同时建立了用双波长薄层扫描测定刺玫根及刺玫果中齐墩果酸含量的方法。结果表明,本法简便快速、精密度高,回收率好,为山刺玫的开发利用及其质量标准的制定提供依据。  相似文献

目的:建立妇血荣胶囊中芍药苷含量的HPLC检测方法.方法:采用甲醇提取样品;色谱条件:Alltech ODS柱(5μm,250mm×4.6 mm);流动相为乙腈-水(16:84,v/v);检测波长为230 nm.结果:线性范围为24.3~218.7 μg.mL-1(r=0.9999),平均回收率为101.21%,RSD为0.80%.结论:高效液相色谱法简单易行,准确,灵敏度高,适用于妇血荣胶囊中芍药苷的含量测定.  相似文献

HPLC法同时测定千斤拔属植物7种黄酮的含量   总被引:1,自引:0,他引:1  
目的:测定千斤拔属7种植物中的7种黄酮类的含量,为千斤拔类药材质量控制提供依据。方法:色谱条件为Hedera C18色谱柱(250 mm×4.6 mm,5.0μm):流速1.2 mL/min,流动相为0.5%HAc水和乙腈梯度洗脱,柱温35℃,检测波长263 nm。结果:七种黄酮类成分在90 min内达到现行分离,线性关系良好(,R2>0.9995),日内、日间精密度、稳定性和重复性的RSD≤3.0%。该法成功的应用于千斤拔药材7种黄酮的评均回收率分别为96.94%~105.29%(RSD≤2.98%)结论:本方法准确、快速、重现性好,不同品种的千斤拔药材质量存在差异,可以为研究千斤拔药材的质量标准提供依据。  相似文献

Plasminogen activator "Ahh-32" from Agkistrodon halys halys venom has been isolated and purified using affinity and ion-exchange chromatography. The purified enzyme consists of the single peptide-chain with molecular weigth of 32 kDa. It can convert free plasminogen into active form--plasmin. "Ahh-32" was inhibited by DFP and benzamidine. Besides, the enzyme influences significantly the activation of plasminogen by streptokinase without having effect on analogical process in case of usage of tissue tipe plasminogen activator. The obtained protein can be used as an instrument under investigation of protein-protein interactions in haemostasis system.  相似文献

By Q-sepharose column ion-exchange chromatography, alkyl-sepharose column hydrophobic chromatography the purified fibrinogenolytic enzyme was obtained from Agkistrodon halys halys venom. It is a single peptide-chain with molecular weight about 28 kDa. It was founded that this enzyme cleaved A alpha-chain of fibrinogen, pH-optimum was determined in the range of 7.5-8.0. Its fibrinogenolytic activity was estimated 15.6 mM fibrinogen/min per mg protein; caseinolytic activity was estimated 7.5 c.u., and amidolytic activity was 0.325 mM pNA/min/mg and 0.175 mM pNA/min/mg for S2238 and S2251 respectively; K(m) was 5.6 mM. The enzyme activity was inhibited by DFP and benzamidine. These results suggest that the enzyme is serine protease. It inhibited the platelet-aggregation.  相似文献

从蝮蛇毒腺中抽提总RNA.利用人工合成寡核苷酸引物作逆转录,以cDNA为模板进行体外扩增,获得磷脂酶A2(简称PLA2)基因,克隆至pBS-ks载体中。通过对3个碱性PLA2(简称BPLA2)基因单独克隆分别作DNA全序列分析,推导pro-BPLA2由138个氨基酸残基构成,与已测定的部分氨基酸序列比较,基本相符。该基因成功的克隆,不仅推导出BPLA2的蛋白质全序列,也为进一步开展蛇毒功能肽蛋白质工程的研究工作打下了良好的基础。  相似文献

从江浙蝮蛇粗毒中经SephadexG 15 0凝胶过滤、DEAE SepharoseCL 6B以及FPLCSuperose 12分离出一种蛋白质。该蛋白质经SDS PAGE测定在非还原和还原条件下分子量均为 5 7kD左右 ,等电聚焦测得其等电点约为 4.9。生物活性测定表明该蛋白质具有L 氨基酸氧化酶活性 ,能够抑制由ADP和胶原所引起的血小板聚集 ,具有抑制革兰氏阴性菌的作用 ;并且具有细胞毒性 ,能够诱导细胞凋亡  相似文献

Basic phospholipase A2 (BPLA2) from the venom of Agkistrodon halys pallas has a strong ability to hemolyze erythrocytes. The asymmetrical unit of P212121 crystal of BPLA2 contains two molecules. Self-rotation function was used to study the orientation relationship of these two molecules. Cross-rotation and translation functions were then used to determine the orientations and positions of the two molecules in the unit cell. The model building and preliminary structure refinement were carried out. The result shows that the two molecules in the asymmetrical unit of orthorhombic crystal are related by a non-crystallographic 2-fold symmetry axis.  相似文献

从长白蝮蛇(Agkistrodon halys Ussuriensis)毒腺中抽提总RNA,采用RT-PCR扩增其类凝血酶基因,经全序列测定,类凝血酶基因Ussurin全长为708个核苷酸,即编码236个氨基酸;根据同源性,推测它的活性中心为His^43,Asp^88和Ser^182;二硫键为Cys^7-Cys^141,Cys^28-Cys^44,Cys^76-Cys^234,Cys^120-Cys^188,Cys^152-Cys^167和Cys^178-Cys^203。该蛇毒类凝血酶cDNA序列及推导的氨基酸序列均为首次报道。  相似文献

Agkistin was a new snake venom metalloproteinase (SVMP) gene which was cloned from Agkistrodon halys. Its deduced amino acid sequence has two additional cysteines (Cys407 and Cys426) in the disintegrin domain compared to other RGD containing SVMPs. The full-length gene (Agkistin) and its disintegrin region (named Agkistin-s) were expressed by baculovirus expression system (pFastBac-Htb vector) with His-tag, and their platelet aggregation-inhibition activity was evaluated. The expressed protein Agkistin can also induce apoptosis of HMEC cells in the basal medium after incubated at 37 degrees C for 20 h.  相似文献

我们利用简并引物从江浙蝮蛇腺总RNA经RP-PCR扩增磷脂酶A2(简称PLA2)基因,并以碱性PLA2(B-PLA2)基因为探针,分离出了酸性PLA2(A-PLA2)和两个未见报道的特征结构类同的基因,分别命名为Asn^48-PAL2和BA-PAL2。双向测序测定了这组PLA2同工酶(除信号肽外)基因的全序列,并由此推导编码的氨基酸序列。其中A-PLA2基因编码的氨基酸序列与较早报道的由蛇毒中分离  相似文献

目的:研究蝮蛇毒蛋白C激活物的分离纯化与理化性质。方法:经DE52-纤维素、CM-Sphadex C-50、G-75柱层析,从安徽芜湖产蝮蛇(Agkistrodon halys)蛇毒中纯化一种均一的蛋白C激活物(PCA)。结果:SDS-PAGE测定分子量约为155000Da,IEF-PAGE测定等电点为4.8。它能使人血浆的KPTT明显延长,显示出强烈的抗凝活性。通过中和试验与显色肽定量实验表明,  相似文献

Agkistrodotoxin, a neutral phospholipase A2 with high presynaptic neurotoxicity from the venom of Agkistrodon halys Pallas, has been crystallized by hanging drop vapor diffusion method. The crystal belongs to P21 space group with the cell dimensions a = 10.836 nm, b=8.486nm, c = 7.082nm, β=109.87$ showing C2 pseu-do-symmetry. Diffraction data to 0. 26 nm resolution have been collected on a Siemens X-200B area detector. C2 pseu-do-symmetry suggests that there exists a non-crystallographic two-fold axis parallel to crystallographic b axis. Self-rotation function calculation with different integrated radius and resolution ranges using the program POLARRFN yields four stable high peaks corresponding to three more non-crystallographic two-fold axis and one special non-crystal-lographic symmetry. The molecules in the asymmetric unit are suggested to be arranged in a manner of "dimer of dimers" by inference.  相似文献
