The effects of growth and assay temperature on unidirectionalK+ fluxes in excised roots of rye (Secale cereale cv. Rheidol)were studied using 86Rb+ as a tracer. Both K+ influx to thevacuole, estimated as K+ uptake between 3 and 12 h after transferof unlabelled roots to radioactive solution, and movement ofK+ to the xylem were determined directly. Other fluxes weredetermined on excised roots of plants, which had been labelledwith 86Rb+ since germination, by conventional triple exponentialefflux analysis. When assayed at 20°C, roots of plants previously grown at20°C(WG roots) had lower rates of net K+ uptake than rootsof low temperature-acclimated plants, grown with a temperaturediferential between roots (87°C) and shoots (20°C) eithersince germination (DG roots) or for 3 d prior to experiments(DT roots). This resulted from a greater unidirectional K+ effluxacross the plasma membrane and a reduced K+ flux to the xylemin WG roots, compared to DG or DT roots, rather than a decreasein unidirectional K+ influx or a decrease in the net K+ fluxto the vacuole. Indeed, although WG roots had lower rates ofK+ influx and K+ efflux across the tonoplast at 20°C thanDG or DT roots, roots of plants from all growth temperaturetreatments showed an equivalent net K+ flux to the vacuole. Although all unidirectional K+ fluxes in roots from plants grownunder all temperature regimes were reduced by lowering the temperatureof the root, these fluxes were differentially affected in rootsof plants from contrasting growth temperature treatments. Rapidcooling to 8°C of WG roots resulted in a lower rate of K+influx and a transient increase in K+ efflux across both theplasma membrane and tonoplast, compared to DG and DT roots.Furthermore, since the K+ flux to the xylem was lower in WGroots, the net K+ uptake at 8°C into WG roots was considerablyreduced compared to DG and DT roots. These results suggest thatlow temperature-acclimation of K+ fluxes in rye roots may involvea reduction in the temperature sensitivity of K+ influx anda curtailment of K+ efflux across both the plasma membrane andtonoplast at low temperatures. Kennedy, C. D. and Gonsalves, F. A. N. 1988. H+ efflux and trans-rootpotential measured while increasing the temperature of solutionsbathing excised roots of Zea mays.—J. exp. Bot. 39: 37–49. Novel temperature-ramp procedures have been used to measureH+ efflux and trans-root potential of excised roots of Zea mays(var. Fronica). Two types of experiment were performed: (1),increasing temperature from 17°C, and (2), pre-cooling theroots to 1°C before starting the temperature ramp. The ratesof increase of temperature for H+ efflux and trans-root potentialexperiments were 0·5 and 2·1°C min–1respectively The H+ scans revealed strong sharp maxima at 30°C and 32°C,for non-pre-cooled and pre-cooled roots respectively, the latterbeing significantly smaller. The trans-root potential scansfor the pre-cooled roots showed a corresponding maximum at 30°C,which was inhibited by KCN (1-0 mmol dm–3) with or withoutSHAM (10 mmol dm–3), or Hg2+ (1, 10, 100 µmol dm–3)in the bathing solutions. Some of the evidence suggests thatthese maxima are associated with electrogenic H+ pumping, mediatedby a plasma membrane-bound ATPase. However, no correspondingmaximum was observed in the trans-root potential scans for non-pre-cooledroots, the potential remaining at about — 75 m V from20°C to 35°C. As there is a 7-fold increase in H+ effluxbetween 20°C and 30°C, the relationship between netH+ efflux and electrogenic proton pumping in these roots isby no means clear. Some possibilities are considered here. Pre-cooled and non-pre-cooled roots show clear maxima in thetrans-root potential scans at about 46°C, at which temperaturethere is a slight net H+ influx. This paper examines the ionic composition of wound-induced electricalcurrents in higher plant tissue, using two non-injurious electrophysiologicaltechniques. By simultaneous recording of K+, H+ , and Ca2+ ionfluxes with extracellular ion-selective microelectrodes, wehave determined that a Ca2+ influx (2.4 µA cm–2),a small H+ influx (0.17 µA cm–2) and a large K+efflux (16 µA cm–2) occur immediately after woundingin roots of Pisum sativum L. var. Greenfeast. Using an extracellularvibrating probe at the wound site, net ion currents of 26 µAcm–2 were measured 5 min after wounding. In a more concentratedbathing medium (1/4 rather than 1/16 strength Hoagland's solution),net ion currents of 59 µA cm–2 were measured, andthese would appear to be the largest extracellular currentsthat have been measured in plants. We made a quantitative comparisonof the summed ion fluxes with the net ion currents and thisrevealed that ion fluxes, in addition to those measured here,occur after wounding. ATP-dependent Ca2+-uptake was investigated in sealed plasmamembrane vesicles isolated from corn roots (Zea mays L. cv.Hybrid-3352/Palma-Pioneer). In a chloride-containing medium,at high calcium concentrations, about 30% of the total Ca2+accumulation ({small tilde}4 nmol Ca2+ mg–1 protein) wasshown to be protonophore-sensitive and corresponded to the fractionof Ca2+ not accumulated in a sulphate-containing medium. Furthermore,vesicles in the presence of nitrate, which stimulates H+ transport,or vesicles preloaded with H+, take up Ca2+ more rapidly, suggestingthat, at high calcium concentrations, there is a mechanism forCa2+ transport which depends on the magnitude of the protongradient across the membrane. The fraction of Ca2+ uptake shownto be sensitive to the protonophore CCCP increased by about150–200% as the Ca2+ concentration in the medium increasedfrom 50µM to 250µM. Under the same conditions, theCCCP-insensitive fraction of Ca2+ accumulated was reduced byabout 25–30% suggesting that different Ca2+ affinitiesexist in the two Ca2+ uptake processes. Although calmodulinstimulation was not observed, the sensitivity to Ca2+ and externalpH indicates that H+ gradient-independent Ca2+ accumulationreflects activity of the Ca2+–pump. These results indicatethat the plasma membrane of corn roots contain two distinctmechanisms of Ca2+ transport: a high Ca2+ affinity, proton gradient-independentCa2+ pump and a low Ca2+ affinity, proton gradient-dependentCa2+/H+ antiport, which have greatest activity at concentrationsof Ca2+ below and above 50+M, respectively. Plasma membrane vesicles of high purity, determined by markerenzyme assays, were obtained by phase partitioning microsomalfractions from stelar and cortical tissues of Zea mays (cv.LG11) roots. ATP hydrolytic activities in both of the plasmamembrane fractions were inhibited by vanadate, SW26 and erythrosinB, but were insensitive to nitrate. Activity in both fractionsexhibited a marked pH optimum of 6·5 and displayed typicalMichaelis-Menten kinetics. A high substrate specificity wasapparent in both the stele and cortex plasma membrane fractions,while the lower fractions, after phase partitioning, showedlower specificity for nucleotide substrates. Specific activitiesof the stele (67·8 µmol Pi mg–1 h–1)and cortex (78·4 µmol Pi mg–1 h–1)plasma membrane H+ -ATPases were very similar. Proton pumping activities in microsomal membrane fractions fromstele and cortex were inhibited by nitrate and insensitive tovanadate. Homogenization of stele and cortex tissue in the presenceof 250 mol m–3 KI resulted in microsomal fractions exhibitingvanadate-sensitive, nitrate-insensitive proton pumping activity,suggesting a plasma membrane origin for this activity. SW26was also an effective inhibitor of proton pumping activity,although results indicated an interaction between SW26 and thefluorescent probes quinacrine and acridine orange. The results are discussed in relation to models for the transportof ions into the stele and are consistent with a role for theH+ -ATPase activity in this process. Roots of Allium cepa L. cv. Yellow are differentially responsiveto gravity. Long (e.g. 40 mm) roots are strongly graviresponsive,while short (e.g. 4 mm) roots are minimally responsive to gravity.Although columella cells of graviresponsive roots are largerthan those of nongraviresponsive roots, they partition theirvolumes to cellular organelles similarly. The movement of amyloplastsand nuclei in columella cells of horizontally-oriented rootscorrelates positively with the onset of gravicurvature. Furthermore,there is no significant difference in the rates of organellarredistribution when graviresponsive and nongraviresponsive rootsare oriented horizontally. The more pronounced graviresponsivenessof longer roots correlates positively with (1) their caps being9.6 times more voluminous, (2) their columella tissues being42 times more voluminous, (3) their caps having 15 times morecolumella cells, and (4) their columella tissues having relativevolumes 4·4 times larger than those of shorter, nongraviresponsiveroots. Graviresponsive roots that are oriented horizontallyare characterized by a strongly polar movement of 45Ca2+ acrossthe root tip from the upper to the lower side, while similarlyoriented nongraviresponsive roots exhibit only a minimal polartransport of 45Ca2+. These results indicate that the differentialgraviresponsiveness of roots of A. cepa is probably not dueto either (1) ultrastructural differences in their columellacells, or (2) differences in the rates of organellar redistributionwhen roots are oriented horizontally. Rather, these resultsindicate that graviresponsiveness may require an extensive columellatissue, which, in turn, may be necessary for polar movementof 45Ca2+ across the root tip. Simultaneous measurements of the extracellular potential andthe K+(86Rb) efflux, and of the intracellular and extracellularpotentials of the cortical cells were used to study the effectsof external Ca2+ on the plasma membrane K+(86Rb) permeabilityin two-day-old mung bean (Vigna mungo L. Hepper, ‘Blackmatpe’) roots under high KCl stress. The K+ efflux wasenhanced by a high KCl solution (>7.5 mM), and addition of0.5 mM Ca2+ could suppress this efflux. The removal of membrane-associatedCa+ from the root surface with EDTA led to a recovery of theK+ efflux along with a marked decrease in the extracellularpotential. Proton Fluxes and the Activity of a Stelar Proton Pump in Onion Roots   总被引:6,自引:2,他引:4  
The xylem vessels of excised adventitious roots of onion, Alliumcepa, were perfused with unbuffered nutrient solution adjustedinitially to either pH 9·3 or 3·9; the pH of thesolution after passage through the xylem, at rates not lessthan 2 xylem volume changes min–1, was close to pH 6·5in both instances. The flux of H+ across the xylem/symplastboundary into mildly alkaline, phosphate-buffered solutionsperfusing the vessels could be increased greatly with increasingbuffer strength, up to a maximum value between 0·5–1·0pmol H+ mm–2 s–1. The apparent neutralization ofacidic malic acid buffers had a slightly lower maximum capacity,equivalent to –0·3 to –0·5 pmol H+mm–2 s–1. The addition of 5·0 pmol m–3fusicoccin (FC) to the xylem perfusion solution stimulated theentry of H+ into the xylem; in unbuffered perfusion solutionsthe pH fell to pH 3·6 after a lag of 25–35 min.FC additions to phosphate-buffered solutions also stimulatedthe H+ flux to an extent similar to that in unbuffered solution,viz. 0·2–0·4 pmol mm–2 s–1. The release of K+ (36Rb-labelled) into xylem sap transientlyincreased as the [K+] in weakly buffered perfusion solutionswas raised stepwise; a very marked increase being seen whenthe concentration was raised to 100 mol m–3 from 40 molm–3. The addition of 5·0 mmol m–3 FC to theperfusing solution containing 100 mol m–3 K+ rapidly decreasedthe K+ flux to the xylem as the H+ flux increased. Fusicoccinalso inhibited the flux of K+ into unbuffered perfusion solutionsbut the effect appeared reversible. Addition of 10 mmol m–3abscisic acid (ABA) to the perfusion solution quickly producedtransient increases in both K+ and H+ fluxes into the xylem.In this and other experiments using weakly phosphate-bufferedperfusing solutions, H+ fluxes were comparable in size to thoseof K+ The results are consistent with the idea that the stele of onionroots contains a proton trarislocating ATPase whose activityresponds to the pH of the xylem sap. It is evident that theactivity of the proton secreting and proton neutralizing mechanismsin the xylem parenchyma control the movement of other ions acrossthe xylem/symplast boundary. In a previous work the involvement of a fenamate-sensitive Ca2+-activated nonselective cation channel (NSCC) in free radical-induced rat liver cell necrosis was demonstrated (5). Therefore, we studied the effect of radical oxygen species and oxidizing agents on the gating behavior of a NSCC in a liver-derived epithelial cell line (HTC). Single-channel currents were recorded in HTC cells by the excised inside-out configuration of the patch-clamp technique. In this cell line, we characterize a 19-pS Ca2+-activated, ATP- and fenamate-sensitive NSCC nearly equally permeable to monovalent cations. In the presence of Fe2+, exposure of the intracellular side of NSCC to H2O2 increased their open probability (Po) by 40% without affecting the unitary conductance. Desferrioxamine as well as the hydroxyl radical (·OH) scavenger MCI-186 inhibited the effect of H2O2, indicating that the increase in Po was mediated by ·OH. Exposure of the patch membrane to the oxidizing agent 5,5'-dithio-bis-2-nitrobenzoic acid (DTNB) had a similar effect to ·OH. The increase in Po induced by ·OH or DTNB was not reverted by preventing formation or by DTNB washout, respectively. However, the reducing agent dithiothreitol completely reversed the effects on Po of both ·OH and DTNB. A similar increase in Po was observed by applying the physiological oxidizing molecule GSSG. Moreover, GSSG-oxidized channels showed enhanced sensitivity to Ca2+. The effect of GSSG was fully reversed by GSH. These results suggest an intracellular site(s) of action of oxidizing agents on cysteine targets on the fenamate-sensitive NSCC protein implicated in epithelial cell necrosis. The phospholipid and sterol contents of plasma membranes isolatedfrom stele and cortex of maize roots were compared. The majorsterol present in both tissues was stigmasterol which was foundin significantly higher quantities in the cortex (27·4µg mg–1 membrane protein) compared to the stele(17·4 µg mg–1). Other sterols detected includedsitosterol, campesterol and small quantities of cholesterol.The major phospholipids found were phosphatidylethanolamine(PE) and phosphatidylcholine (PC), with PC more abundant inthe stele. The fatty acid composition indicated that the majorfatty acid in both stele and cortex was 16:0 (palmitic), withothers found in lesser amounts. Effluxes of K+ and Ca2+ from root segments of both wheat, Triticunaestivum L. cv. Capelle and mung bean, Vigna radiata (L.) Wilczek,were measured in the presence or absence of 20 mol m–3para-fluorophenylalanine (p-FPA). The results were used to estimatethe compartment contents and transmembrane K+ and Ca2+ fluxesin root cortex cells. Using the Ussing-Teorell flux equationas the criterion, it was concluded that entry of K+ from theoutside solution to the cytoplasm, and from the cytoplasm tothe vacuole were active in both wheat and mung bean. Also, inboth species, Ca2+ entered the cytoplasm passively across theplasmalemma and was actively pumped back to the external solution.However, interpretation of the direction of active transportacross the tonoplast depends upon an assumption about Ca2+ activityin the cytoplasm. The only qualitative effect of p-FPA was to alter the drivingforce for K+ influx, across the plasmalemma in wheat, from anactive to a passive one. Quantitative effects of the analoguewere seen for K+ fluxes in both wheat and mung bean and forCa2+ fluxes in wheat. The p-FPA reduced transport of K+ in bothspecies, while transport of Ca2+ was unaffected. The implicationsof these results for the ‘two pump hypothesis’ arediscussed. This paper deals with the responses of roots of wheat {Triticumaestivum L.) to hypoxia with special emphasis on the effectsof severe O2 deficiency on membrane integrity, loss of K+ fromthe root and root membrane potentials. Seminal and crown roots of 26-d-old plants exposed to severehypoxia (0.003 mol O2 m–3) for 3 h or 10 d prior to excisionand subsequently exposed to hypoxic solutions, had slightlylower rates of sorbitol influx and a slightly smaller apparentfree space than roots in aerated solutions. These results indicatethat neither a few hours nor a 10-d exposure to hypoxia hadadverse effects on the membrane integrity of the bulk of thecells in the roots. However, both 6-d-old seedlings and 26-d-oldplants lost K+ from the roots following their transfer fromaerated to hypoxic nutrient solutions. In the 26-d-old plants,which were of high nutritional status, there was a net K+ effluxfrom the roots to the external solution. In contrast, with the6-d-old seedlings, which were of low nutritional status, thedecrease in the K+ content of the roots was smaller than thenet K+ uptake to the shoots. Exposure of excised roots to 0.008 mol O2 m3caused arapid and reversible membrane depolarization from –120to ––80 mV. These data and the magnitude of thenet effluxes strongly suggest that K+ losses during the earlystages of hypoxia are due to membrane depolarization ratherthan to increases in the permeability of membranes to K +. Plasma membrane sheets from Xenopus oocytes have been isolated for use in fluorescence resonance energy transfer (FRET) measurements. This system has the following advantages: 1) fluorescent recordings from a large surface area to maximize the signal-to-noise ratio, 2) reduction in background fluorescence from proteins retained in intracellular compartments, and 3) access to the cytoplasmic surface of the plasma membrane for rapid solution changes. To demonstrate the utility of this approach, we have examined a previously published FRET-based Ca2+ sensor, namely, the Cameleon-PM. This construct targets to the plasma membrane and, upon various Ca2+ additions to the cytoplasmic face of the membrane, shows ratiometric FRET changes. From the ratiometric changes recorded, an apparent Ca2+ affinity of 1.65 µM was determined. Two membrane fractions were obtained from 16%/26% and 34%/40%interfaces following discontinuous sucrose density gradientcentrifugation of a 10,000–80,000xg pellet from mung bean(Phaseolus mungo L.) roots. The ATPases in the fractions differedfrom each other in their sensitivity toward various inhibitors,activation with salts, dependence of activity on pH, and Kmfor ATP.Mg2+. Judging from their sensitivity toward inhibitors,the ATPases in the low and high density membranes are consideredmainly of tonoplast and plasma membrane origin, respectively.Both ATPases were activated by gramicidin D and nigericin. ATP-inducedquenching of quinacrine fluorescence in both fractions requiredMg2+ and permeant anions such as Cl and quenching wascollapsed by carbonylcyanide p-trifluoromethoxyphenyl hydrazone.The sensitivities of quenching to the inhibitors were essentiallythe same as those of ATPase activity in the membranes. The method of compartmental analysis was applied to study sodiumfluxes in roots of intact seedlings of Helianthus annuus L.By measuring sodium uptake and transport to the shoots of theseedlings in parallel experiments, transport of tracer sodiumto shoots and net accumulation of Na+ in the roots during theflux measurements was accounted for. The steady-state sodiumfluxes in the intact sunflower roots were similar in size tothose in excised roots but in general they were somewhat higher.This indicates more metabolic activity in the intact tissues.Using whole plants it is possible to study the response of ionfluxes in roots to ecophysiological stimuli received by theshoots, and in the present experiments the effect of continuouslight versus long-day growth conditions was investigated. Potassium,when continually present, depressed all fluxes and the cytoplasmiccontent of sodium but tended to increase the vacuolar sodiumcontent, in particular when this was related to the cytoplasmiccontent. When added to sodium-loaded roots, potassium stimulatedthe plasmalemma sodium efflux but slightly, suggesting a lowefficiency of K+-Na+ exchange across the plasmalemma in intactas well as excised sunflower roots. Subsequently, however, potassiuminduced a transient decrease in the 22Na efflux that was followedby oscillations in tracer efflux. These changes were attributedto potassium-induced transfer of sodium to vacuoles. Moreover,the oscillations seem to indicate the operation of negativefeedback control of sodium fluxes.  相似文献

Net fluxes of H+and Ca2+were measured in the mesophyll tissueof broad bean (Vicia faba L.) leaves and in protoplasts derivedfrom these cells. NaCl at 90 m M enhanced H+extrusion in bothprotoplasts and tissue, but in different ways. Proton extrusionwas inhibited by vanadate, suggesting the involvement of theplasma membrane H+-ATPase in cell responses to salinity. Therewas virtually no effect of NaCl on the net Ca2+flux in protoplasts,while in the tissue a large transient Ca2+efflux followed thesalt treatment. Salt-induced Ca2+efflux was essentially independentof external Ca2+concentrations in the range 0.1 to 10 m M. Also,Ca2+flux responses were ‘saturated’ above 50 m MNaCl. It is suggested that almost all the measured Ca2+fluxoriginates from Na+/Ca2+and H+/Ca2+ion exchange in the cellwall. This conclusion was supported by the results of modellingcation exchange in the cell wall. Respiratory oxygen consumption by roots was 1·4- and1·6-fold larger in NH+4-fed than in NO-3-fed wheat (Triticumaestivum L.) and maize (Zea mays L.) plants respectively. Higherroot oxygen consumption in NH+4-fed plants than in NO-3-fedplants was associated with higher total nitrogen contents inNH+4-fed plants. Root oxygen consumption was, however, not correlatedwith growth rates or shoot:root ratios. Carbon dioxide releasewas 1·4- and 1·2-fold larger in NO+3-fed thanin NH+4-fed wheat and maize plants respectively. Differencesin oxygen and carbon dioxide gas exchange rates resulted inthe gas exchange quotients of NH-4-fed plants (wheat, 0·5;maize, 0·6) being greatly reduced compared with thoseof NO-3-fed plants (wheat, 1·0; maize, 1·1). Measuredrates of HCO-3 assimilation by PEPc in roots were considerablylarger in 4 mM NH+4-fed than in 4 NO-3 plants (wheat, 2·6-fold;maize, 8·3-fold). These differences were, however, insufficientto account for the observed differences in root carbon dioxideflux and it is probable that HCO-3 uptake is also importantin determining carbon dioxide fluxes. Previous papers have shown that abscisic acid can inhibit transportof ions across the root to the xylem vessels, resulting in reducedexudation from excised roots or inhibiting guttation from intactplants. However, it has not been established whether the inhibitionwas due to a reduction in salt transport (Js) or in permeabilityof the roots to water (Lp). This paper investigates the effectof ABA on Lp and Js separately. It is shown that Lp increasedin ABA and then fell, but was about the same as in control rootswhen transport was inhibited. The effect of ABA on exudationtherefore appeared to be mainly due to reduction in Js. Inhibitionof Js was also present in intact, transpiring plants and sowas not due to reduced water flow. The inhibition of ion releaseto the xylem affected Na+, Mg2+, Ca2+, and phosphate as wellas the major ion in the exudate, K+. It is concluded that ABAinhibits salt transport to the shoot by acting on ion transportinto the xylem, and not by reducing water flow coupled withsalt transport.  相似文献

Ca2+-channel blockers at concentrations greater than 1 mmolm–3, directly affect the activity of K +selective channelsin the plasma membrane of Amaranthus tricolor protoplasts. Theseeffects are not mediated by the blockade of Ca2+ channels. Blockers tested included 1, 4-dihydropyridines (nifedipine,nicardipine), verapamil, bepridil, Gd3+ and La3+, applied towhole-cell and detached outside-out patches of plasma membraneat concentrations from 50µmol m–3 to 100 mmol m–3.For certain experiments the concentration of Ca2+ on the cytoplasmicside of the plasma membrane ([Ca2+]cyt) was buffered at either50ftmol m–3 or 500 µmol m–3. The principal currents observed in whole-cells flowed throughcation outward rectifier (OR) channels. Each blocker causedan immediate reduction of time-dependent outward currents atdoses down to 1 mmol m–3 and produced a different, reversible,kinetic block of the outward current, independent of the levelof [Ca2+]cyt. Verapamil also activated a sustained inward cationcurrent at negative p.d. The same effects were found with individualchannels in detached outside-out patches. Conductance and selectivityof the cation OR channels were unchanged by the drugs. [Ca2+]ex, was varied over a range from 0 to 10 mol m–3.Progressively lower [Ca2+]eI, increasingly enhanced the maximumamplitude of the time-dependent currents. Time-constants fordecay of inward tail currents were increased at low [Ca2+]eit.These effects were rapidly reversible. Although there was noevidence that the cation ORs in plasma membrane of Amaranthustricolor were dependent on [Ca2+]cyl for their activation, theywere sensitive to the concentration of free Ca2+ in the extracellularmedium. We employed a glycogen-depleting session of exercise followed by a low-carbohydrate (CHO) diet to investigate modifications that occur in muscle sarcoplasmic reticulum (SR) Ca2+-cycling properties compared with low-CHO diet alone. SR properties were assessed in nine untrained males [peak aerobic power (O2 peak) = 43.6 ± 2.6 (SE) ml·kg–1·min–1] during prolonged cycle exercise to fatigue performed at 58% O2 peak after 4 days of low-CHO diet (Lo CHO) and after glycogen-depleting exercise plus 4 days of low-CHO (Ex+Lo CHO). Compared with Lo CHO, Ex+Lo CHO resulted in 12% lower (P < 0.05) resting maximal Ca2+-ATPase activity (Vmax = 174 ± 12 vs. 153 ± 10 µmol·g protein–1·min–1) and smaller reduction in Vmax induced during exercise. A similar effect was observed for Ca2+ uptake. The Hill coefficient, defined as slope of the relationship between cytosolic free Ca2+ concentration and Ca2+-ATPase activity, was higher (P < 0.05) at rest (2.07 ± 0.15 vs. 1.90 ± 0.10) with Ex+Lo CHO, an effect that persisted throughout the exercise. The coupling ratio, defined as the ratio of Ca2+ uptake to Vmax, was 23–30% elevated (P < 0.05) at rest and during the first 60 min of exercise with Ex+Lo CHO. The 27 and 34% reductions (P < 0.05) in phase 1 and phase 2 Ca2+ release, respectively, observed during exercise with Lo CHO were not altered by Ex+Lo CHO. These results indicate that when prolonged exercise precedes a short-term Lo CHO diet, Ca2+ sequestration properties and efficiency are improved compared with those during Lo CHO alone. calcium cycling; vastus lateralis; contractile activity; glycogen; phosphorylation potential  相似文献
