An unusual species of Geastrum was found growing on decayed wood debris and leaves of Triplochiton scleroxylon in the Mbalmayo Forest Reserve, Cameroon. The species morphologically resembles G. saccatum and G. fimbriatum in having sessile endosperidium partly enclosed by the saccate base of the exoperidium. Microscopically, it is characterized by and distinguished from all other known species of the genus, in having subsmooth, punctate to moderately verruculose, slightly thick-to distinctly thick-walled polymorphous, constricted to eight-shaped, mostly oblong, ovoid, cylindrical, elliptic to clubshaped basidiospores. G. pleosporus was studied from a collection of about fifteen basidiomata covering different stages of development. It is described as new based on morphological analyses and phylogenetic inferences made from large ribosomal DNA sequence alignments. Phylogenetic relationship of G. pleosporus is investigated. In parsimony analyses of partial sequences of the large subunit rDNA from selected Gasteromycetes species, G. pleosporus is closely related to G. saccatum within the strongly supported clade of Geastrum species. The cluster of G. pleosporus and G. saccatum is well supported in parsimony analysis of the dataset with Geastrum species and related taxa using parsimony and maximum likelihood analysis.

The distribution of α-glucose 1-phosphate forming (α-type) trehalose phosphorylase and trehalase activities in various fungi was surveyed. α-Type phosphorylase occurred in the mycelia and fruit-bodies of Agaricales and Aphyllophorales in the Holobasidiomycetidae, and at least one species of Gasteromycetes, but not in Tremellaceae or Auriculariales of the Phragmobasidiomycetidae, Heterobasidiomycetes or Hemibasidiomycetes. The test fungi in the Ascomycotina and Deuteromycotina, and the yeasts of Basidiomycotina, showed different trehalase activities, but no trehalose phosphorylase activity. The test organisms showed different levels of trehalase activity. The fruit-bodies of most mushrooms showed higher activities of α-type trehalose phosphorylase than did the mycelia.

The compressed basidiocarp of a fungus belonging to Geasteraceae (Lycoperdales; Gasteromycetes) was recovered from the plant-bearing Cenozoic strata of the Pié de Vaca Formation, Puebla, Mexico. The fossil specimen consists of an open exoperidium with ten triangular rays, and a central, circular endoperidium with an eccentrical ostiole and irregular surface relief. Scanning electron microscopy revealed a delicate reticulum on the surface of rays and on samples of the endoperidium. An acetolyzed fragment of the endoperidium yielded a few fungal spores, and similar spores were also obtained from an acetolyzed rock fragment adjacent to the fossil specimen. The fossil fungus exhibits the characteristics of extant earthstars; morphological comparison between the extant and the fossil earthstar and the type of spores obtained from the endoperidium of the latter indicate its affinity with Geastrum, although uncertainty in diagnostic characters preclude its assignment to a living species. The report of a complete basidiocarp of Geastrum from the Cenozoic of Puebla may contribute greatly to the knowledge about the spatial distribution and evolutionary history of Geasteraceae and Lycoperdales.

Two new mesophilic, sporeforming, gram-positive, strictly anaerobic, rod-shaped bacteria were isolated which utilized betaine in the Stickland reaction. Strain M1 was obtained from pasteurized hypersaline sediments. Cells were motile rods and formed spherical terminal spores. Betaine was used with hydrogen and several amino acids as electron donors. In addition, several carbohydrates served as substrates. Growth required 1.5% NaCl with an optimum at 6.0% NaCl. The guanine plus cytosine content of the DNA was 26.9%. This strain is described as a new species, Clostridium halophilum.Strain W6 was isolated from marine sediments. Cells were motile rods and formed ovoid, subterminal spores. Betaine was used with hydrogen and several amino acids as electron donors. Carbohydrates were not fermented. Growth optimum was at 1.0% NaCl. The guanine plus cytosine content of the DNA was 26.1%. This strain is described as a new species, Clostridium litorale.Non standard abbreviations DMG N,N-dimethylglycine - TMA trimethylamine - PY peptone-yeast extract - PYG peptone-yeast extract-glucose  相似文献

A new pleurostomatid genus Apoamphileptus is described, which is diagnosed as: Belonging to the Amphileptidae with spica on right side; on each side of the cell, a single perioral kinety, which encircles the cytostome and does not extend to the posterior end of the cell; somatic kineties of both sides near ventral margin shortened and forming a postoral suture; two to several extra fragments with densely arranged dikinetids located in anterior portion of left side. As the type species, the morphology and infraciliature of Apoamphileptus robertsi nov. spec., isolated from a shrimp-farming pond near Qingdao (Tsingtao), China, have been investigated using living observations and the protargol silver impregnation method. The diagnosis for this new species is: Apoamphileptus 90-180 x 30-60 microm in vivo, body elongate pyriform-shaped and slightly flattened; with one cross-striated band along the cytostome; 2-6 (generally 4) large macronuclear nodules, one micronucleus; 33-43 right somatic kineties; left side 6-8 kineties; two extra anterior fragments on left side; about 13 contractile vacuoles dispersed throughout whole body; extrusomes absent or not recognizable; marine habitat. Some morphologically related morphotypes are discussed and tabulated. Regarding the pattern of infraciliature and other morphological features, the well-described fresh-water species, Amphileptus claparedii Stein, 1867 is believed to be a member of this new genus, hence a new combination is suggested: Apoamphileptus claparedii (Stein, 1867) nov. comb.

A new species of entomopathogenic nematode, Steinernema aciari sp. n. was described. It was recovered from a soil sample collected from Haimen town, Shantou district in the eastern coast of Guangdong province, the People's Republic of China during a survey for entomopathogenic nematodes. S. aciari sp. n. belongs to the Steinernema glaseri group. It can be separated from all described Steinernema species by the combined morphological and morphometrical characters of various stages of the nematodes. For male, the new species can be recognized by spicule length (86+/-6.3 microm); spicule tip blunt with a hook-like structure; gubernaculum with a short and Y-shaped cuneus and corpus well-separated posteriorly. For infective juvenile, the combination of the following characters: body length (1113+/-68 microm), distance from anterior end to excretory pore (95+/-3.7 microm), tail length (78+/-5.2 microm), and E % (123+/-7) can be used to differentiate the new species from other nematodes. For female, the tail (conoid with a long mamillate terminus and a distinct postanal swelling) and vulva (slightly protruding from body surface with conspicuous double flapped epiptygma) shapes can be used as diagnostic characters for the new species. The new species can also be distinguished from other Steinernema species by DNA sequences of either a partial 28S rDNA or the internal transcribed spacer regions of rDNA, and from the close related species S. glaseri, Steinernema longicaudum CWL05, and Steinernema guangdongense by cross-breeding test.

Leptoamphisiella vermisGruber, 1888 n. g., n. comb. (basionym Epiclintes vermisGruber, 1888) is an extraordinarily large and worm-like marine stichotrichous ciliate. Based on a population isolated recently from coastal waters of Qingdao, China, the living morphology and infraciliature are redescribed and its taxonomic position is defined. Accordingly, a new diagnosis for this species is suggested: large, marine Leptoamphisiella with a conspicuous layer of pellicular alveoli; 400-1000 microm x 40-70 microm in vivo; body band-like, highly flexible; about 40 membranelles; always three frontal and two buccal cirri; 52-80 transverse cirri (TC) extending to the posterior end of the buccal field; 57-79 left midventral and 44-62 right midventral cirri; 62-102 cirri in left and 63-91 cirri in right marginal rows (MR); 9-13 dorsal kineties that extend the full body length; about 100 macronuclear nodules and 5-13 micronuclei. The diagnosis for the new genus is as follows: vermiform Pseudoamphisiellidae with strongly contractile body, differentiated frontal, buccal, and highly developed TC; two remarkably separated midventral rows; one MR on each side of the body; frontoterminal and caudal cirri absent. Leptoamphisiella vermis n. comb. is fixed as the type species of the new genus.

Two nematode species of the genus Enchodelus Thorne, 1939, one new and one known from Arctic polar deserts were studied. Enchodelus makarovaesp. n. is an amphimictic species, characterised by females with body length of 1.57-2.00 mm, lip region 15-17.5 μm wide, amphid duplex, odontostyle 38-43 μm long or 2.3-2.8 times lip region diam. Odontophore with flanges, 1.2-1.4 times as long as odontostyle; pharynx length 320-377 μm, pharyngeal expansion 113-130 μm long or 32-37% of total pharynx length; female genital system amphidelphic, uterus tripartite, pars refringens vaginae with two trapezoid sclerotisations, vulva a transverse slit (V=45-51%); tail bluntly conoid (25-35 μm, c=45.8-70.3, c'=0.6-0.9 in females, and 29-33 μm, c=46.4-58.9, c'=0.7-0.8 in males). Males with 65-74 μm long spicules and 10-12 spaced ventromedian supplements. During a recent soil sample survey in Eastern China, a new entomopathogenic nematode species, collected from the Chongming Islands in the southern-eastern area of Shanghai, was discovered. Morphological characteristics of different developmental stages of the nematode combined with molecular data showed that this nematode is a new genus of Rhabditidae, and described as Heterorhabditidoides chongmingensis gen. nov., sp. nov., for that it shares more morphological characteristics with heterorhabditids than with steinernematids. For males, the papillae formula of bursa is 1, 2, 3, 3, with constant papillae number in the terminal group, stoma tubular-shaped and about 1.5 head width; cheilorhabdions cuticularized, esophageal collar present and long, median bulb present. For infective juveniles, EP = 90 (80-105) μm, ES = 104 (92-120) μm, tail length = 111 (89-159) μm, and a = 19.1 (15-21). The percentages of the nucleotides A, T, C and G in the ITS1 regions of the new species are significantly different from those of heterorhabditids and other rhabditids. Molecular phylogenetic trees based on 18S rDNA and the internal transcribed spacer (ITS) sequences data revealed that the new entomopathogenic nematode species forms a monophyletic group, which is a sister group of the clade comprised of some genera of Rhabditidae.

Nineteen bacteria isolates recovered from shellfish samples (mussels and oysters) showed a new and specific 16S rDNA-RFLP pattern with an Arcobacter identification method designed to recognize all species described up to 2008. These results suggested that they could belong to a new species. ERIC-PCR revealed that the 19 isolates belonged to 3 different strains. The sequence of the 16S rRNA gene of a representative strain (F98-3T) showed 97.6% similarity with the closest species Arcobacter marinus followed by Arcobacter halophilus (95.6%) and Arcobacter mytili (94.7%). The phylogenetic analysis with the16S rRNA, rpoB, gyrB and hsp60 genes placed the shellfish strains within the same cluster as the three species mentioned (also isolated from saline habitats) but they formed an independent phylogenetic line. The DDH results between strain F98-3T and A. marinus (54.8% ± 1.05), confirmed that it represents a new species. Several biochemical tests differentiated the shellfish isolates from all other Arcobacter species. Although the new species was different from A. mytili, they shared not only the same habitat (mussels) but also the characteristic of being so far the only Arcobacter species that are simultaneously negative for urea and indoxyl acetate hydrolysis. All results supported the classification of the shellfish strains as a new species, for which the name Arcobacter molluscorum sp. nov. with the type strain F98-3T is proposed (=CECT 7696T = LMG 25693T).

Three new yeast species, Candida kashinagacola (JCM 15019(T) = CBS 10903(T)), C. pseudovanderkliftii (JCM 15025(T) = CBS 10904(T)), and C. vanderkliftii (JCM 15029(T) = CBS 10905(T)) are described on the basis of comparison of nucleotide sequences of large subunit ribosomal DNA D1/D2 region (LSU rDNA D1/D2). The nearest assigned species of the three new species was Candida llanquihuensis. Candida kashinagacola and C. pseudovanderkliftii differed from C. llanquihuensis by 3.8% nucleotide substitution of the region, while C. vanderkliftii did by 4.4%. Three new species differed in a number of physiological and growth characteristics from any previously assigned species and from one another. A phylogenetic tree based on the sequences of LSU rDNA D1/D2 showed that these new species together with Candida sp. ST-246, Candida sp. JW01-7-11-1-4-y2, Candida sp. BG02-7-20-001A-2-1 and C. llanquihuensis form a clade near Ambrosiozyma species. The new species did not assimilate methanol as a sole source of carbon, which supported the monophyly of these non methanol-assimilating species which are closely related to the methylotrophic yeasts. Candida kashinagacola was frequently isolated from the beetle galleries of Platypus quercivorus in three different host trees (Quercus serrata, Q. laurifolia and Castanopsis cuspidata) located in the sourthern part of Kyoto, Japan, thus indicating that this species may be a primary ambrosia fungus of P. quercivorus. On the other hand, C. pseudovanderkliftii and C. vanderkliftii were isolated only from beetle galleries in Q. laurifolia. Candida vanderkliftii was isolated from beetle gallery of Platypus lewisi as well as those of P. quercivorus. Candida pseudovanderkliftii and C. vanderkliftii are assumed to be auxiliary ambrosia fungi of P. quercivorus.

A new histozoic species of myxosporean (Henneguya schizodon n. sp.) is described from the Amazon River teleost fish Schizodon fasciatus Spix & Agassiz, 1892 (Characiformes, Anostomidae). The plasmodia, which showed asynchronous development, were located in the kidney of the host. The spore body was ellipsoidal and was 13.1 (12-14) micron long by 3.3 (3-4) micron wide. The total length of the spore was 28.9 (27-30) micron, and each value had a caudal process measuring 16.3 (15-17) micron. The polar capsules were 5.4 (5-6) micron long by 1.3 (1-1.5) micron wide, and each had a polar filament with 8-10 coils. The characteristics of the species were compared with nearly all the species described so far, including all the species reported from South American fishes. This comparison allows to consider the materials as a new species, and the name Henneguya schizodon n. sp. is proposed.

The diagnosis of Theissenia is emended to include taxa that lack a definite central perithecial columella and taxa that feature ascospores with a germ slit. Theissenia cinerea is described as a new species that lacks a perithecial columella; T. eurima is described as a new species with ascospores having a germ slit. The type species, T. pyrenocrata, is redescribed and compared with the new species. The taxonomic position of Theissenia is discussed.

The Neotropical psittacine species Amazona aestiva, commonly known as the blue-fronted Amazon, is one of the most common and best-known psittacine birds kept as a pet worldwide. However, very little is known about the diseases or parasites of these birds. In this study, we describe a new species, Eimeria aestivae, associated with these parrots. The new species is characterized by: ovoid smooth oocysts (n = 60), 36.8 (33.2-41.5) × 23.7 (21.7-25.7) μm, length/width ratio = 1.55; polar granule present; ellipsoidal sporocysts (n = 25), 19.8 (17.5-21.6) × 9.3 (8.3-9.9) μm; Stieda, sub-Stieda body, and sporocyst residuum present. Sporozoites (n = 20), 2 per sporocyst, elongate and curved, 17.6 (15.8-19.2) × 3.8 (3.2-4.8) μm; each with 2 refractile bodies. The oocysts of the other 2 eimerian species described for Amazona are larger than those of the presented species, but they all seem to be closely related because of some similarities among them.

A new species of entomopathogenic nematode, herein described as Steinernema akhursti sp. n., was recovered from soil samples collected from Yunnan Province, the People's Republic of China. Both morphological and molecular data show congruently that S. akhursti sp. n. belongs to the Steinernema feltiae group. It can be separated from all described Steinernema species by the combined morphological and morphometrical characters of various stages of the nematodes. For the first generation male, the new species can be recognized by spicule length 90 +/- 4.6 microm, spicule tip blunt with an aperture on the ventral side, gubernaculum with a long and needle-shaped cuneus, and tail conoid with a prominent mucron on the tip and a concave on ventral side. For the infective juvenile, the combination of the following characters: body length 812 +/- 19 microm, distance from anterior end to excretory pore 59 +/- 1.5 microm, tail length 73 +/- 2.9 microm, E% 77 +/- 4.5, lateral field with six evenly distributed and identical ridges at the middle body portion, and tail with long and slightly constrict hyaline portion can be used to separate the new species from other nematodes. For the female, the new species is characterized by: tail conoid with a short mucron and slightly swelling anal portion and a symmetrical, slightly protruding vulva with conspicuous double-flapped epiptygma. The nematode can be separated from other described species of Steinernema by DNA sequences of either a partial 28S rDNA or the internal transcribed spacer regions of rDNA and from the closely related species S. feltiae and Steinernema oregonense by cross-breeding tests.
