Differential effects on proliferation of individual vs. combined administration of high- and low-molecular-weight microbial autoregulators (extracellular RNase from Bacillus subtilis and anabiosis-inducing factor d1) are reported for the first time for cultured cells of higher eukaryotes. Proliferation of ras-transformed mouse fibroblasts was affected by both autoregulators dose-dependently. The cytotoxic activity of individual regulators was directly related to their concentration. Unlike RNase, factor d1 (which functions as a chemical chaperone) exerted reversible effects. Studies of the effects of combined administration of the autoregulators demonstrated that pretreatment of the cells with low-dose d1 decreased the toxicity of RNase. Higher doses of d1 were required to attenuate the effects of toxic agents with more pronounced membrane tropism. The results obtained suggest that a universal system regulating the physiological activity of cells is operative in taxonomically remote organisms. The operation of the system is based on sequential changes in the structural organization and function of subcellular structures, induced by low- and high-molecular-weight autoregulators.  相似文献

Extracellular, stylar RNases (S-RNases) are produced by self-incompatible, solanaceous plants, such asNicotiana alata, and are thought to be involved in selfpollen rejection by acting selectively as toxins to selfpollen. In this study, the toxicity of RNases to other plant cells was tested by culturing cells ofN. alata andN. plumbaginifolia in the presence ofS-RNases fromN. alata. The growth of cultured cells ofN. plumbaginifolia was inhibited by theS-RNases, but viability was not affected. Growth of cultured cells of oneN. alata selfincompatibility genotype was inhibited by twoS-RNases, indicating that inhibition was not allele specific. Comparisons with the effects of inactivated RNase and other proteins, suggest that the inhibition of growth byS 2-RNase was partly, but not wholly, due to RNase activity. Heat-denaturedS 2-RNase was a very effective inhibitor of cell growth, but this inhibitory activity may be a cell surface phenomenon.  相似文献

The haloalkaliphilic, lithoautotrophic, sulfur-oxidizing gram-negative bacteria Thioalkalivibrio versutus and Thioalkalimicrobium aerophilum were found to possess a special system for the autoregulation of their growth. The system includes the extracellular autoinducers of anabiosis (the d1 factor) and autolysis (the d2 factor). The principal components of the d1 factor are alkylhydroxybenzenes, as evidenced by specific qualitative reactions. The principal components of the d2 factor are free unsaturated fatty acids dominated by oleic acid isomers. Like the respective autoregulators of neutrophilic bacteria, the d1 factor of haloalkaliphilic bacteria presumably controls their growth and transition to anabiotic state, while the d2 factor controls autolytic processes. Alkylhydroxybenzenes of both microbial and chemical origin were found to influence bacterial respiration. The low-molecular-weight osmoprotectant glycine betaine enhanced the thermostability of trypsin. This suggests that both glycine betaine and the d1 factor serve as chemical chaperones.  相似文献

A whole-cell biotransformation system for the conversion of d-fructose to d-mannitol was developed in Escherichia coli by constructing a recombinant oxidation/reduction cycle. First, the mdh gene, encoding mannitol dehydrogenase of Leuconostoc pseudomesenteroides ATCC 12291 (MDH), was expressed, effecting strong catalytic activity of an NADH-dependent reduction of d-fructose to d-mannitol in cell extracts of the recombinant E. coli strain. By contrast whole cells of the strain were unable to produce d-mannitol from d-fructose. To provide a source of reduction equivalents needed for d-fructose reduction, the fdh gene from Mycobacterium vaccae N10 (FDH), encoding formate dehydrogenase, was functionally co-expressed. FDH generates the NADH used for d-fructose reduction by dehydrogenation of formate to carbon dioxide. These recombinant E. coli cells were able to form d-mannitol from d-fructose in a low but significant quantity (15 mM). The introduction of a further gene, encoding the glucose facilitator protein of Zymomonas mobilis (GLF), allowed the cells to efficiently take up d-fructose, without simultaneous phosphorylation. Resting cells of this E. coli strain (3 g cell dry weight/l) produced 216 mM d-mannitol in 17 h. Due to equimolar formation of sodium hydroxide during NAD+-dependent oxidation of sodium formate to carbon dioxide, the pH value of the buffered biotransformation system increased by one pH unit within 2 h. Biotransformations conducted under pH control by formic-acid addition yielded d-mannitol at a concentration of 362 mM within 8 h. The yield Y D-mannitol/D-fructosewas 84 mol%. These results show that the recombinant strain of E. coli can be utilized as an efficient biocatalyst for d-mannitol formation.  相似文献

Alkyl-substituted hydroxybenzenes (AHBs), which are autoinducers of microbial dormancy (d 1 factors), were found to stabilize the structure of protein macromolecules and modify the catalytic activity of enzymes. In vitro experiments showed that C6-AHB at concentrations from 10−4 to 10−2 M, at which it occurs in the medium as a true solution and a micellar colloid, respectively, nonspecifically inhibited the activity of chymotrypsin, RNase, invertase, and glucose oxidase. C6-AHB-induced conformational alterations in protein macromolecules were due to the formation of complexes, as evidenced by differences in the fluorescence spectra of individual RNase and C6-AHB and their mixtures and in the surface tension isotherms of C6-AHB and trypsin solutions. Data on the involvement of dormancy autoinducers in the posttranslational modification of enzymes and their inhibition will provide further insight into the mechanisms of development and maintenance of dormant microbial forms.  相似文献

To understand the expression pattern of theS RNase gene in the floral tissues associated with self-incompatibility (SI), promoter region of S11 RNase gene was serially deleted and fused GUS. Five chimeric constructs containing a deleted promoter region of the S11 RNase gene were constructed, and introduced intoNicotiana tabacum using Agrobacterium-mediated transformation. Northern blot analysis revealed that the GUS gene was expressed in the style, anther, and developing pollen of all stages in each transgenic tobacco plant The developing pollen expressed the same amount of GUS mRNA in all stages in transgenic tobacco plants. In addition, histochemical analysis showed GUS gene expression in vascular bundle, endothecium, stomium, and tapetum cells during pollen development in transgenic plants. From these results, it is speculated that SI ofLycopersicon peruvianum may occur through the interaction ofS RNase expressed in both style and pollen tissues.  相似文献

The evolution of the ribonuclease A (RNase A) vertebrate-specific enzyme family is interesting in that specific gene lineages appear to be responding to unique selective pressures in wildly diverse manners to generate proteins that are capable of reducing the infectivity of viruses, killing systemic pathogens, and inducing the growth of blood vessels all while maintaining the signature motifs of a ribonuclease. In this paper, we present the DNA sequence and gene structure of Mus musculus RNase 6 and examine the expression pattern and enzymatic activity of the recombinant protein. M. musculus RNase 6 has a limited expression pattern compared to human RNase 6 and is an efficient ribonuclease, with a catalytic efficiency 17-fold higher than that of human protein. Evo- lutionary analysis reveals that RNase 6 was subject to unusual evolutionary forces (dN/dS=1.2) in an ancestral rodent lineage before the separation of Mus and Rattus. However, more recent evolution of rodent RNase 6 has been relatively conserved, with an average dN/dS of 0.66. The mutant R33 of the obligatory aerobic yeastRhodotorula glutinis exhibited a defect ind-glucose uptake. Detailed kinetic studies ofd-glucose andd-fructose transport in wild-type and mutant strains provided evidence for the existence in the plasma membrane of a carrier specific for fructose. The transport ofd-fructose in the mutant exhibited saturation kinetics up to 1 mmol/Ld-fructose; at higher concentrations the rate ofd-fructose uptake decreased. In the wild-type strain biphasicd-fructose uptake kinetics were observed; the low-affinity component was not found in the mutant, but the high-affinity transport system persisted. During the exponential phase of growth (ond-glucose) the high-affinityd-fructose system was repressed in the wild-type strain. Mutual competition betweend-fructose andd-glucose as well as the pH dependence of transport of the two hexoses further supported the following conclusion: In the wild-type strain,d-fructose is taken up both by the specific fructose carrier (K T=0.22 mmol/L) and the glucose carrier (K T=9.13 mmol/L). The former does not translocated-glucose, the latter is damaged by the mutation. Finally H+ co-transport and plasma membrane depolarization induced by the onset ofd-fructose transport indicated that the fructose carrier is an H+ symporter.  相似文献

Parabiosis of adult DBA/2J (H-2 d ) mice with adult (DBA/2J× CSH/HeJ)F1 (H-2 d /H-2 k ) mice results in survival beyond 100 days in 44% of such pairs, induction ofin situ unresponsiveness to C3H/HeJ skin, and the complete takeover of the erythroid system of the F1 by parental cells. However, in vitro responsiveness to C3H/HeJ cells remains. Dye exclusion cytotoxicity assays establish the absence ofF 1 lymphoid cells in the spleens and bone marrow of both partners. The parental takeover of the erythroid system of the F1 partner requires immune recognition of the hybrid's alloantigens, because this takeover is not seen with tolerant parental cells. PartialH-2 differences (on the C3H background) influence both survival and the takeover reaction when incorporated into parabioses with DBA/2J partners. When only theK andI subregions ofH-2 were targets of the parental response, 58% of parabionts survived, with complete parental hemopoietic takeover. When onlyH-2D was the target, 83% of parabionts survived, with incomplete hemopoietic takeover. Changing the non-H-2 background of the F1 target did not significantly affect survival or takeover, while substituting a differentH-2 d parental strain (BALB/c) eliminated survival altogether.Thus the parabiont takeover reaction encompasses the lymphoid and hemopoietic systems, requires immunorecognition of the target alloantigens, and seems to require a strongH-2 difference for its induction.  相似文献

A stylar S-RNase is associated with gametophytic self-incompatibility in the Rosaceae, Solanaceae, and Scrophulariaceae. This S-RNase is responsible for S-allele-specific recognition in the self-incompatible reaction, but how it functions in specific discrimination is not clear. Window analysis of the numbers of synonymous (dS) and non-synonymous (dN) substitutions in rosaceous S-RNases detected four regions with an excess of dN over dS in which positive selection may operate (PS regions). The topology of the secondary structure of the S-RNases predicted by the PHD method is very similar to that of fungal RNase Rh whose tertiary structure is known. When the sequences of S-RNases are aligned with the sequence of RNase Rh based on the predicted secondary structures, the four PS regions correspond to two surface sites on the tertiary structure of RNase Rh. These findings suggest that in S-RNases the PS regions also form two sites and are candidates for the recognition sites for S-allele-specific discrimination.  相似文献

Changes in RNase activity during sporulation of a homothallic diploid strain of Saccharomyces cerevisiae were measured in caffeine-treated and non-treated cells.
1.  In caffeine-treated cells soon after the transfer to the sporulation medium a significant increase in RNase activity was observed; in control cells the rise of RNase activity was less and started after a lag period of 5 h. The final activity of RNase was about twice as high in caffeine-treated cells as in control cells.
2.  Increase in RNase activity during sporulation was sensitive to cycloheximide in control cells, but insensitive in caffeine-treated cells.
3.  RNases from vegetative cells and from sporulating ones are different in their K m values.
Relation of the changes in RNase activity to premeiotic DNA synthesis is discussed.  相似文献

The mutagenic activity of chemical analogues of microbial anabiosis autoinducers (the autoregulatory d1 factors of cell differentiation), which act to inhibit cell proliferation, to enhance cell tolerance, and to induce the transition of cells to an anabiotic state, was studied using the Ames test. In the range of concentrations studied (0.1 to 100 g/ml), alkyl-substituted hydroxybenzenes (AHBs) differing in hydrophobicity, i.e., methylresorcinol (C1-AHB) and hexylresorcinol (C6-AHB), as well as unsubstituted resorcinol, showed different growth-inhibiting and mutagenic effects. C6-AHB was found to inhibit the growth of Salmonella typhimurium TA100 and to induce its mutagenesis at a rate of 1.8 revertants/nmol. C1-AHB taken at low concentrations not only failed to inhibit bacterial growth but even stimulated it and exerted an antimutagenic effect. Unsubstituted resorcinol virtually did not influence bacterial growth and showed weak mutagenic activity. The growth-inhibiting effect of elevated C6-AHB concentrations correlated with the degree of the transition of the original phenotype producing S-type colonies to a phenotype producing R-type colonies. The role of AHB homologues, as microbial autoregulators with mutagenic activity, in the regulation and correlation of two processes: the phenotypic dissociation of microbial populations and the formation of resting microbial forms, is discussed. The accumulation of AHBs in senescent microbial cultures may induce adaptive mutations, change the expression of genes, and promote the development of minor cell subpopulations (phenotypes), thus providing for the adaptation of these cultures to varying environmental conditions.  相似文献

Summary Adrenergic stimulation induces contraction of hypertrophied prostatic tissue via the α1 adrenoceptor, and the results of pharmacological studies suggested the existence of adrenoceptor subtypes. Recently three subtypes (α1a, α1b, and α1d) were cloned. Using probes for these subtypes, we demonstrated their expression in the tissues of ten cases of benign prostatic hypertrophy, usingin situ hybridization. To determine the ratio between these subtypes, an RNase protection assay was also performed in three cases. Expression of the α1a and α1d adrenoceptors was diffuse in the smooth muscles of the interstitium, but was absent in glandular epithelial cells. On the contrary, the α1b adrenoceptor was hardly detectable. The RNase protection assay confirmed the absence of the α1b adrenoceptor, the ratio of α1a and α1d being 4∶1. These results supported the idea that the differences in prostatic contractile response to several adrenergic drugs are based on the affinities of these drugs for the different subtypes.  相似文献

A Pichia pastoris expression system for bovine pancreatic RNase A was constructed: the RNase A sequence was fused to the PHO1 signal and the AOX1 promoter was used for efficient secretion. Approximately 5 mg of soluble enzymes were secreted per liter of the culture, but one half of them were glycosylated. After a series of purifications by cation-exchange chromatography, the glycosylated enzyme was removed and the pure recombinant soluble unglycosylated RNase A was obtained in the final yield of 1 mg per liter of the culture. N-Terminal sequence, molecular weight, secondary structure, thermal stability, and activity were completely identical with those of commercial RNase A. Glycosylated RNase A had a decreased k cat, 60-70% of the activity of wild-type RNase A, as in the case of RNase B. Its carbohydrate moiety seemed to destabilize the enzyme differently from RNase B since T m of the glycosylated RNase A was decreased by 6°C. The carbohydrate moiety of the glycosylated enzyme contained no GlcNAc. The N34A mutant RNase A, in which the only potential N-glycosylation site, Asn34, is mutated to alanine, was also glycosylated, implying that glycosylation is not N-linked but O-linked.  相似文献

Incubation of excised Avena leaves in a wet chamber in darkness resulted in an increase in both soluble and particle-bound Rnase activities. Illumination promoted the increase in the total RNase which occurred upon leaf excision. The light-induced increase in total RNase was due to an increase in soluble RNase. The increase in RNase activity in the particulate fraction was inhibited by illumination. Feeding 2 per cent sucrose to the tissues in the dark increased the level of soluble RNase and decreased the activity found in the particulate fraction. Treatment of the illuminated tissues with 10?4M dichlorophenyldimethylurea (DCMU) inhibited the effects of light on the RNase level. It is concluded that the light-effect is explained at least in part by the photosynthetic production of sugars. In excised leaves kept in darkness the RNA content rapidly decreased. Feeding sugars to or illumination of the tissues lowered the rate of RNA breakdown due to leaf excision. DCMU counteracted the light effect. In general, the decrease of RNA was repressed by all treatments leading to an inhibition of the increase of particulate RNase. On the other hand, the observed changes of the soluble RNase were not related with the variations of RNA. Treatment with 3 M urea increased the RNase activity both in the particulate and the soluble fractions. The RNase activity of soluble preparations, partially purified on a Sephadex G-50 column or by (NH4)2SO4 fractionation, was also stimulated by 3 M urea. Treatment with 10?5M kinetin repressed the increase in RNase activity due to leaf excision both in the soluble and the particulate fractions.  相似文献

The fermentation ofd-arabinose byBacteroides ruminicola strain B14 occurs in a manner similar to or identical with that shown previously forl-arabinose metabolism by the organism, a combination of hexose resynthesis and the Embden-Meyerhof sequence. The use ofd-arabinose by strain B14 was repressed by prior growth in medium containingd-glucose and induced by prior growth in the presence ofl-arabinose ord-xylose. The use ofd-ribose andd-xylose by strain B14 is different from that ford-arabinose. During growth in the presence of 1-14C-d-arabinose, labeled acetate, propionate, and succinate were formed, whereas during 1-14C-d-ribose growth only labeled acetate and propionate were obtained. Under the conditions used,d-xylose growth failed to allow formation of acetate, propionate, or succinate. Strain B14 incorporates label from 1- or 2-labeled glycine into acetate, propionate, and succinate by a mechanism involving the cleavage of glycine and equilibration of glycine carbons 1 and 2 with different metabolic pools.  相似文献

Summary Cell extracts ofCandida guilliermondii grown ind-xylose,l-arabinose,d-galactose,d-glucose,d-mannose and glycerol as sole carbon sources possessed NADPH-dependent aldose reductase activity, but no NADH-dependent activity was detected.d-xylose andl-arabinose were the best inducers of aldose reductase activity. The highest enzyme activity ind-xylose orl-arabinose-grown cells was observed first withl-arabinose followed byd-xylose as substrates of the enzymatic reaction. However, only low activity was found ind-glucose,d-mannose andd-galactose-grown cells, indicating that these carbon sources cause catabolite repression. Enzyme activities induced ind-xylose-grown cells were twice as high as those obtained from the cells under resting conditions. Furthermore, the level of induction of aldose reductase activity depended on the initial concentration ofd-xylose. The present study shows that aldose reductase activity may be efficiently induced by pentose sugars of hemicellulosic hydrolysates and weakly by hemicellulosic hexoses.  相似文献

Stationary unaerated cells ofS. pombe containing endogenous substrates but not energized by any exogenous ones take up 2-deoxy-d-glucose, 6-deoxy-d-glucose,d-xylose andd-arabinose actively over diffusion equilibrium. The active uptake is inhibited by 20–100 mmol/L H2O2 which causes an increase inK T but has no effect onJ max. This “competitive inhibition” indicates that H2O2 affects directly the sugar binding sites of the transporters. The ATP-binding site of the plasma membrane H+-ATPase is also affected by 100 mmol/L H2O2; theK T decreases 7-fold,J max about 2.5-fold. These effects are not likely to be mediated by membrane lipid peroxidation which appears to be lacking inS. pombe, and this lack may be one of the reasons for the high resistance of this yeast to H2O2. Because of thisS. pombe represents a suitable system for studying direct effects of oxidants on membrane proteins.  相似文献
