大花蕙兰茎尖组织培养及其形态建成的研究   总被引:23,自引:0,他引:23  
以大花蕙兰(Cymbidium grandi,lorium)茎尖为外植体,在改良的Vacin和Went培养基上诱导形成了原球苹,并且在添加有1 mg/1BA的‘Vacin和Went培养基上获得了快速繁殖无性系,将原球茎转移到加有香蕉匀浆汁的Knudson c培养基上,很快长出叶片和根,形成完整的再生植株。较高浓度的BA(1.O mg/1)能促进原球茎的增殖;较低浓度的BA(O.4mg/1)能促进原球茎的分化。形态发生的显微观察表明原球茎来源于体细胞胚,它的发育过程与合子胚十分相似。  相似文献

黑节草(Dendrobium candidum Wall ex Lind.)2—6个月种龄的胚均处于球形胚阶段,不同种龄的胚在体积大小、胚细胞数目、胚细胞内的淀粉粒含量和超微结构上有差异。在离体培养条件下黑节草种子萌发率可达95%,种子萌发后形成原球茎,原球茎可以直接发育形成幼苗,又可以由原球茎产生大量愈伤组织,由愈伤组织再分化发育成幼苗。种子萌发过程中,胚顶端分生组织细胞的淀粉逐渐消耗,淀粉的变化与分生组织和子叶的形成有明显的相关性。  相似文献

Micropropagated plants of two annual haloxerophytic Asiatic Salsola species (S. pestifer and S. lanata) were obtained from zygotic embryos cultured on Murashige and Skoog (MS) agar medium supplemented with 0.5 μM benzylamino-purine (BAP) and 0.3 μM indole-3-acetic acid (IAA) or with 0.5 μM 6 γ, γ-dimethylallylaminopurine and 0.3 μM IAA. The callus induction from shoot and leaf explants derived from plants propagated in vitro were obtained on MS agar medium with various concentration of auxins and cytokinins. The best medium for growth and proliferation of calluses of both studied species was MS medium containing 9.0 μM 2,4-dichlorophenoxyacetic acid. It was also determined that beginning of plant regeneration from callus of S. lanata was induced by 8.8 μM BAP. 利用农杆菌介导法将白细胞介素-2基因(il-2)导入番茄中,对影响其转化的因素进行了分析。结果表明:农杆菌菌种(EHA105和C58C1)、外植体类型(子叶和下胚轴)、带有不同筛选标记(Kanr、PPTr、Hygr)的载体质粒几个因素对芽诱导分化及转化均有影响。实验共接种转化2018个子叶和下胚轴外植体,获得了47株抗性再生株,对其进行il-2的PCR扩增检测,有44株呈阳性。PCR-Southern杂交证实PCR结果可靠,显示il-2基因已导入到番茄中。  相似文献

天麻Gastrodiaelata种子与石斛小菇Mycenadendrobii的共生萌发试验表明,石斛小菇可与天麻共生,促进天麻种子发芽并形成原球茎。菌丝主要分布于原球茎的柄状细胞、外皮层细胞和内皮层细胞,在外皮层细胞中形成菌丝结,内皮层细胞中的菌丝则被消化。原球茎细胞中的菌丝均被电子透明物质和原球茎细胞质膜包围而与原球茎细胞质相隔离,菌丝进一步液泡化并最终被水解。含有衰败菌丝的原球茎细胞常被菌丝重新定殖。这一菌丝被消化及菌丝的重新定殖过程在整个原球茎发育过程中可不断重复发生。  相似文献

改良菜心离体培养植株再生体系的研究(简报)   总被引:4,自引:0,他引:4  
This investigation has developed an efficient and fast method for plant regeneration from petiole of cotyledon explants of Brassica campestris L. subsp. chinensis Makino var. parachinensis Tsen et Lee. A medium was designed for B. campestris subsp. chinensis var. parachinensis to obtain the high frequency of shoot regeneration, which contained BAP 2 mg/L, NAA 0.75-1.0 mgL and 7.5 mg/L AgNO3 solution to the half of NH4+ concentration's MS basic medium. 60 mL/L coconut milk were added to all of media. In this method, frequency of shoot regeneration of "youqing caixin" reached as high as 91.2% and the number of shoots per explant reached as high as 4.7 plants. The result showed that there was a positive correlation between frequency of shoot regeneration and number of shoots per explant. The little shoots could be observed five days after inoculation and were formed directly. The inducing rate of roots of the shoots reached as high as 100% and the rate of viability of transferred mature plant reached higher than 95%. The regeneration period from petiole with cotyledon to a seedling was shorten to about 49 days. Factors influencing in vitro explant regeneration were studied.  相似文献

Multiple shoots were induced on Valenciatype peanut (Arachis hypogaea L.) explants cultured in vitro on a nutrient medium supplemented with thidiazuron. Zygotic embryos excised from mature seeds were germinated on Murashige-Skoog nutrient medium, and the resulting plantlets (8 days-old) were used as a source of explants. When cultured on a nutrient medium with increasing levels of thidiazuron (0.5 to 30 mg/l), expiants from various parts of the peanut plant (except the root) produced multiple shoot primordia which subsequently developed into individual shoots. Hypocotyl and cotyledon explants produced shoots in higher numbers than other explants (20 shoots per hypocotyl explant at all thidiazuron concentrations and 15 shoots per cotyledon explant at 30 mg/l). 将枳和酸橙的实生试管苗上、下胚轴和子叶切割成若干段分别进行离体培养,结果表明,枳的各个切段在添加10-4或5×10-5摩尔BA的MS培养基中,不定芽形成率最高,平均每个外植体不定芽发生总量分别达41个和33.5个,其中以上胚轴发生的不定芽最多,占全部不定芽总量的66.6%。在上、下胚轴中,则以第一切段发生的不定芽最多。添加KT的所有处理,不定芽均较少,形成不定芽最多的处理也仅及N0B1处理的四分之一。发根量则以添加10-5摩尔NAA的培养基最多。将枳的不定芽扦插于添加5×10-6或10-6摩尔NAA的MS培养基中,15-20天即可发根成苗。在完全相同的各个处理中,酸橙的不芽定形成量明显少于枳。  相似文献
