The accumulation of microbial biomass in a porous medium can lead to a reduction of pore space and an associated decrease in the hydraulic conductivity of the medium – an effect called bioclogging. This phenomenon may occur in several natural and engineered subsurface systems and can be relevant in fields ranging from contaminant hydrology to civil and environmental engineering, as well as for enhanced oil recovery. During the last decades bioclogging has been studied in various laboratory and theoretical studies. Most of these studies considered only one-dimensional flow fields inside a porous medium. Although these studies provided valuable information on bioclogging and factors controlling it, recent studies showed that an extrapolation of these results to multi-dimensional flow fields is not straight forward. This paper reviews the experimental results obtained for one- and two-dimensional flow fields and compares the modeling results obtained using different conceptualizations of the pore space.  相似文献

Aim Variation partitioning based on canonical analysis is the most commonly used analysis to investigate community patterns according to environmental and spatial predictors. Ecologists use this method in order to understand the pure contribution of the environment independent of space, and vice versa, as well as to control for inflated type I error in assessing the environmental component under spatial autocorrelation. Our goal is to use numerical simulations to compare how different spatial predictors and model selection procedures perform in assessing the importance of the spatial component and in controlling for type I error while testing environmental predictors. Innovation We determine for the first time how the ability of commonly used (polynomial regressors) and novel methods based on eigenvector maps compare in the realm of spatial variation partitioning. We introduce a novel forward selection procedure to select spatial regressors for community analysis. Finally, we point out a number of issues that have not been previously considered about the joint explained variation between environment and space, which should be taken into account when reporting and testing the unique contributions of environment and space in patterning ecological communities. Main conclusions In tests of species‐environment relationships, spatial autocorrelation is known to inflate the level of type I error and make the tests of significance invalid. First, one must determine if the spatial component is significant using all spatial predictors (Moran's eigenvector maps). If it is, consider a model selection for the set of spatial predictors (an individual‐species forward selection procedure is to be preferred) and use the environmental and selected spatial predictors in a partial regression or partial canonical analysis scheme. This is an effective way of controlling for type I error in such tests. Polynomial regressors do not provide tests with a correct level of type I error.  相似文献

The submerged roots and rhizomes of the aquatic vascular macrophyteNuphar lutea (L.) Sm. are aerated, at least in part, by pressurizedventilation. Depending on temperature differences of up to 5K between the inside of young, just-emerged leaves and the surroundingair, pressure differences of 79 to 100 Pa higher than atmosphericare detectable inside the lacunuous spongy parenchyma of theleaf blades. The pressurization is a consequence of structuralfeatures of leaf tissues separating the air filled spaces ofthe spongy parenchyma from the atmosphere. These tissues areacting as thermo-osmotic partitions. Whereas the dimensionsof the stomatal openings (about 5·6 x 2·4 µm)and of the intercellular spaces of the palisade parenchyma (diametersabout 15 µm) are too large, those of the monolayers ofcells separating the palisade and the spongy parenchyma (diameters:0·7–1·2 µm) are small enough to impedefree gaseous diffusion. This inner non-homogeneous partitioninggives rise to the so-called Knudsen diffusion, a physical phenomenonleading to pressurization of the warmer air inside the spongyparenchyma. The rising pressure difference is strong enoughto establish an air flow through the aerenchyma of the wholeplant and out of the most porous older leaves in which a temperatureinduced pressurization is never detectable. These thermo-osmoticallyactive leaves enhance the influx of air to the rhizome and thediffusion path for oxygen to the roots is shortened to the distancebetween rhizome and root tips. Therefore, pressurized ventilationin Nuphar is seen to be of considerable ecological importancefor plant life in anaerobic environments. Our study is one of the very few cases of speleomycological research in recently discovered caves. The aim of this research was to assess the population size of fungal colonies and their species composition in the Jarkowicka cave, discovered in 2012. The air samples were taken from one location outside the cave and from two locations inside of it. Mycological evaluation of the rocks inside the cave was performed usingswab sampling procedure. In the Jarkowicka cave we found 22 species of fungi, including 13 isolated from air at the entrance and from the walls, and 8 species from air inside the cave. Cladosporium spp. were the fungi most frequently isolated from internal atmosphere of the Jarkowicka cave, and from the external air. On the other hand, the fungi most frequently isolated from the rocks were Mucor spp. We found several species not yet described as cave inhabitants: Hypocrea pachybasioides, Cladosporium uredinicola, and Embellisia abundans. Our study may provide a basis for comparison to other similar studies conducted in frequently visited caves by tourists.  相似文献

We examined the effect of environmental patchiness on the spatial segregation of the sexes in the dioecious anemophilus grass Poa ligularis. Because the species is sensitive to grazing, a better understanding of environmental factors that control its spatial distribution and abundance could improve conservation efforts. We hypothesized that (i) males and females are spatially segregated in the microenvironments created by plant patches as the result of sexual specialization in habitat and/or resources use, (ii) sexual specialization is related to different tolerance to competition and reproductive costs of males and females, and (iii) changes in patch structure affect the microenvironment and the intensity of spatial segregation of the sexes. We analyzed the spatial distribution of sexes at three sites with different plant and micro-environmental patchiness and performed a controlled competition experiment with different substitution of males and females. Our results showed that large plant patches created larger sheltered soil fertility islands than small patches. As patch size and their area of influence increased, the density and the spatial segregation of the sexes of P. ligularis also increased, resulting in biased habitat-specific sex ratios. In accordance with their higher reproductive costs, females were more frequent in sheltered (low air evaporative demand) and nitrogen-rich areas inside patch perimeters than males. Females were also better able to tolerate inter-sexual competition than males. In contrast, males tolerated low nitrogen concentration in soil and low sheltering, probably gaining advantage in pollen dispersal. Inter- and intra-sexual competition, however, affected the reproductive output of both sexes. From the point of view of conservation, environmental patchiness is important to the status of P. ligularis populations. The reduction of patch size limits the available microsites, biases the sex ratio towards males inside patches, increases inter- and intra-sexual competition, and it might be expected to decrease overall seed and pollen production and consequently potential recruitment.  相似文献

Biological macromolecules have evolved over billions of years to function inside cells, so it is not surprising that researchers studying the properties of such molecules, either in extracts or in purified form, take care to control factors that reflect the intracellular environment, such as pH, ionic strength and composition, redox potential and the concentrations of relevant metabolites and effector molecules. There is one universal aspect of the cellular interior, however, that is largely neglected--the fact that it is highly crowded with macromolecules. It is proposed that the addition of crowding agents should become as routine as controlling pH and ionic strength if we are to meet the objective of studying biological molecules under more physiologically relevant conditions.  相似文献

Two-dimensional air current speeds in the culture vessel were measured using a tracer-based visualization technique and the effect of the air movement in the culture vessel on the photosynthesis of in vitro potato plantlets was assessed under a photoautotrophic culture condition. The air current speeds inside the vessel were varied by controlling free convection induced by spatial variations of temperatures in the culture vessel. For all conditions examined, upward air currents were observed around the plantlets in the central part of the culture vessel and downward air currents were observed near inside walls in the culture vessel. The upward and downward air currents were restricted by the presence of the plantlet. The upward air current speeds were affected by plantlet size inside the vessel and it was 24, 8 and 4 mm s−1 in culture vessels with no plantlets, a 10-mm-tall plantlet and a 60-mm-tall plantlet cultured inside the vessel, respectively. The upward air current speed was increased by 2 times by increasing wind velocity above the culture vessel from 0.1 to 1.0 m s−1. Placing the black plate on the medium also increased the air current speeds by 1.5 times. The net photosynthetic rates of the plantlets increased from 2.0 to 2.5 μmol m−2 s−1 as the upward air current speed in the culture vessel increased from 2.4 to 8.0 mm s−1. The air current speeds in the culture vessel were significantly slow. Enhancement of the air movement in the culture vessel is important to promote photosynthesis of the in vitro plantlets.  相似文献

Association studies in environmental statistics often involve exposure and outcome data that are misaligned in space. A common strategy is to employ a spatial model such as universal kriging to predict exposures at locations with outcome data and then estimate a regression parameter of interest using the predicted exposures. This results in measurement error because the predicted exposures do not correspond exactly to the true values. We characterize the measurement error by decomposing it into Berkson-like and classical-like components. One correction approach is the parametric bootstrap, which is effective but computationally intensive since it requires solving a nonlinear optimization problem for the exposure model parameters in each bootstrap sample. We propose a less computationally intensive alternative termed the "parameter bootstrap" that only requires solving one nonlinear optimization problem, and we also compare bootstrap methods to other recently proposed methods. We illustrate our methodology in simulations and with publicly available data from the Environmental Protection Agency.  相似文献

交通出行的便捷和绿色程度是城市生态社区评价的关键内容。基于生态社区评价研究中交通出行相关指标的筛选及分析,选择了评价社区交通出行生态水平的5项关键指标,即居民工作出行时长,社区停车状况,绿色出行比例,公共交通满意度,社区停车环境满意度,建立了基于交通出行数据采集的客观评价和基于居民满意度问卷调查的主观评价相结合的评价方式。选择北京市城区和近郊的4个不同社区案例进行调查、评价和比较,验证评价指标的有效性及北京典型社区的交通出行生态水平。北京居民工作出行时长在60 min以内的占样本总量的75%左右,7%的居民出行时长在90 min以上;居民平均绿色出行比例为69.5%;北京居民对公共交通的满意度约为46%,对社区停车环境满意度则不足19%。居民工作出行时长指标可以更直接的反应交通便捷程度,而公共交通的便利程度会影响到居民选择绿色出行方式的比例。  相似文献

The analysis of exhaled air has several advantages since it is a noninvasive method applicable to a large number of toxic agents, in addition to being a simpler matrix than those of other biological samples such as urine and blood. However, it presents some challenges, such as the necessity of a more sensitive sampling procedure, since the chemical substances eliminated through exhaled air are unchanged in form, not being metabolized, and exhaled compounds are present at extremely low concentrations, i.e. in the nanomolar range. To improve the sensitivity and precision of measurement of the concentration of these substances in exhaled air, the sample usually has to be concentrated before assay by gas chromatography. To this end, the use of the solid-phase microextraction (SPME) technique has been proposed as an efficient sampling method. This paper presents a revision of breath analysis as a biomarker for occupational and environmental exposure to chemicals. The sampling methods and the potential use of SPME for determining chemical substances in exhaled air are discussed.  相似文献

We have developed a technique in which transglutaminase is used to measure the penetration of terminal complement proteins across the erythrocyte membrane into the cytoplasmic space. Penetration of a given terminal complement protein into the cytoplasmic space was assessed by labeling the protein of interest with radioactive iodine, forming the complement channel using the labeled protein, adding transglutaminase to only one side of the membrane, and allowing the enzyme to cross-link the susceptible proteins on that side of the membrane. Cross-linking was assessed by measuring the increase in molecular weight of the appropriate molecule on sodium dodecyl sulfate gels under reducing conditions. The results of these experiments indicate that C8 and C9 are rapidly cross-linked to high molecular weight from either the interior or the exterior of the membrane. In order to determine whether the cross-linking mediated by enzyme on the interior was occurring from within the ghosts and not via enzyme that had leaked into the extracellular medium, experiments were performed with dimethylcasein in the extracellular medium. In the presence of this protein, cross-linking of C8 and C9 from outside was negligible. Hence, if cross-linking occurs when transglutaminase is trapped inside the ghosts, it cannot be due to leakage of enzyme, but must be attributable to cross-linking from the inside. The results show that C9 definitely penetrated across the membrane into the intracellular space. With respect to C8, statistical evaluation indicates that C8 probably penetrated into the intracellular space.  相似文献

Periplasmic space in Salmonella typhimurium and Escherichia coli.   总被引:74,自引:0,他引:74  
The volume of the periplasmic space in Escherichia coli and Salmonella typhimurium cells was measured. This space, in cells grown and collected under conditions routinely used in work with these bacteria, was shown to comprise from 20 to 40% of the total cell volume. Further studies were conducted to determine the osmotic relationships between the periplasm, the external milieu, and the cytoplasm. Results showed that there is a Donnan equilibrium between the periplasm and the extracellular fluid, and that the periplasm and cytoplasm are isoosmotic. In minimal salts medium, the osmotic strength of the cell interior was estimated to be approximately 300 mosM, with a net pressure of approximately 3.5 atm being applied to the cell wall. A corollary of these findings was that an electrical potential exists across the outer membrane. This potential was measured by determining the distributions of Na+ and Cl- between the periplasm and the cell exterior. The potential varied with the ionic strength of the medium; for cells in minimal salts medium it was approximately 30 mV, negative inside.  相似文献

Summary Sections of metal impregnated tissue and freeze-fracture have been used to examine intracellular membrane systems in marginal cells of the stria vascularis in mammalian cochleae. A continuous network of elements of the smooth endoplasmic reticulum was revealed. Notable features of this system were a series of flattened cisternae just inside and parallel with the lateral plasma membrane in continuity with an apical network of tubules, cisternae and sheets oriented in parallel with the luminal membrane. The whole system was closely associated with mitochondria. These characteristics suggest that the potassium transporting marginal cells possess a tubulo-cisternal endoplasmic reticulum (TER) like that found in many sodium transporting epithelial cells. The lateral elements of the TER dilated, appearing like vacuoles, and opened to the lateral extracellular space in response to the effects of ethacrynic acid. This diuretic impairs ion transport in the stria vascularis. It is suggested that the TER in marginal cells is involved in the transport of ions and fluid from the cell to the intercellular space when ion balance is disturbed and may play a role in cell volume regulation.This work was supported by the Medical Research CouncilPart of this work was presented at the 18th Workshop on Inner Ear Biology, Montpellier, September, 1981  相似文献

Abstract. Seasonal data on leaf conductance (gl) for three different apple cultivars grown in four separate orchards with different aged trees was studied between 1979 and 1985. A number of empirical models for predicting leaf conductance from environmental measurements were compared using this data and a general method for adapting such models for the prediction of different data sets is proposed. Although stepwise multiple regression identified relative humidity or vapour pressure as important variables, it frequently did not identify the optimal set of independent variables, which often did not include either of these. There was no advantage in regressing gl against principal components of the environment, rather than against the raw environmental variables. A simple model involving air vapour pressure deficit, air temperature and a hyperbolic function of irradiance was found to explain between 32 and 62% of the variance in gl for the different data sets. Parameters fitted for one data set led to the effective prediction of gl in other years or plots. The model fit could generally be improved significantly by including soil moisture deficit among the independent variables.  相似文献

Previously, we, and others found that cryoablation on normal lung produced localized pulmonary hemorrhage and edema, causing obliteration of air space. Therefore, we hypothesized that lung air leakage may be diminished by this procedure. In the present study, we examined if cryoablation can attenuate experimentally created lung air leakage. Male domestic pigs (n = 4) underwent a thoracotomy. The lung was resected approximately 5 mm in diameter and 1 mm in depth to create air leakage lesions. An argon gas cryoprobe with a copper plate attached to its tip was used to cryoablate the lesions superficially. After cryoablation, the positive airway pressure that produced macroscopic bubbles from each lesion site was compared between cryoablated and untreated lesions. Also, cryoablation of the lung surface was carried out in male Donryu rats (n = 20) which were sequentially sacrificed to observe the histological changes over a time course. In the pigs, the air leakage pressure was significantly increased with cryoablation (40 cmH2O <) compared to no treatment (19 ± 5 cmH2O) (p = 0.021, Mann–Whitney U test). Histologically, cryoablation produced acute pulmonary hemorrhage and edema. In the rats, the region with extensive hemorrhage progressed to fibrosis in 1 month, and the areas with edema recovered. This study provides supportive evidence that cryoablation has the potential to stop air leakage from surface pulmonary injury. This procedure may provide a useful adjunct to surgical resection for spontaneous pneumothorax, and the control of air leakage from dissected raw lung surfaces during lung resection.  相似文献

During expiration, the carbon dioxide (CO2) levels inside the dead space of a filtering facepiece respirator (FFR) increase significantly above the ambient concentration. To reduce the CO2 concentration inside the dead space, we attach an active lightweight venting system (AVS) comprising a one-way valve, a blower and a battery in a housing to a FFR. The achieved reduction is quantified with a computational-fluid-dynamics model that considers conservation of mass, momentum and the dilute species, CO2, inside the FFR with and without the AVS. The results suggest that the AVS can reduce the CO2 levels inside the dead space at the end of expiration to around 0.4% as compared to a standard FFR, for which the CO2 levels during expiration reach the same concentration as that of the expired alveolar air at around 5%. In particular, during inspiration, the average CO2 volume fraction drops to near-to ambient levels of around 0.08% with the AVS. Overall, the time-averaged CO2 volume fractions inside the dead space for the standard FFR and the one with AVS are around 3% and 0.3% respectively. Further, the ability of the AVS to vent the dead-space air in the form of a jet into the ambient – similar to the jets arising from natural expiration without a FFR – ensures that the expired air is removed and diluted more efficiently than a standard FFR.  相似文献

In the Atlantic Montane Rain Forest of south-eastern Brazil, a field study was carried out to describe the forest disturbance regime, analyse canopy gap composition and evaluate the influence of habitat parameters on gap tree species composition. We characterized canopy gaps considering the group of variables as follows: area, type and number of tree/branch falls, topographic position, soil coverage and surrounding canopy trees. Gap composition was assessed at species level by measuring all individuals inside gaps higher than one meter. Mean gap area of the 42 canopy gaps analysed was 71.9 ± 9.0 m2 (mean ± SE). Out of the studied gaps, 35.7% were created by uprooted and by snapped trees, 16.7% by dead-standing trees and 11.9% by the fall of large branches. The disturbance regime was characterized by gap openings predominantly smaller than 150 m2 and by spatial patterning related to topography. Ridges had smaller gaps and higher proportions of gaps created by branch falls; slopes had bigger gaps generally created by uprooting events. The more abundant and frequent species were shade tolerant and the more species-rich families found inside gaps did not differ from the forest as a whole. Pioneer species were rare and restricted to medium and large size classes. The Indicator Species Analysis and the Canonical Correspondence Analysis indicated gap area, topography and the percentage of soil cover by the genera Calathea and Ctenanthe were the predominant variables correlated with woody species distribution. So, topography emerged as an important issue not only to the gap disturbance regime, but also to gap colonization. In respect to the influence of gap processes on the Atlantic Montane Rain Forest regeneration, our results support the view that canopy gap events may not be working as promoters of community wide floristic shifts.  相似文献

平衡pH是培养基中二氧化碳分压与空气中二氧化碳分压平衡时的pH,是微藻培养基的一个重要特征参数。选用常用的三种微藻培养基:BG11培养基、BBM培养基和Zarrouk培养基,使用鼓泡通入空气的方法,研究碳源、氮源、磷源浓度和盐度对培养基平衡pH的影响。结果如下:在NaHCO3浓度016.8 g/L范围内,三种培养基的平衡pH都随碳源浓度的增加而升高;NaNO3浓度在02.5 g/L范围内,对培养基平衡pH没有影响;0.1 g/L KH2PO4使BG11培养基的平衡pH稍有升高,与之相反,BBM培养基的平衡pH稍有下降,进一步提高磷(P)浓度(最高0.4 g/L),BG11培养基和BBM培养基的平衡pH不再变化;P浓度变化对Zarrouk培养基的平衡pH没有影响;三种培养基的平衡pH都随盐度的升高而降低。碳源浓度是影响培养基平衡pH的最主要因素,平衡pH与碳源浓度的关系可以用回归方程y=0.3504ln(x)+8.9647(R2=0.9708)表示,氮源浓度对平衡pH没有显著影响,磷源浓度和盐度对培养基平衡pH有影响,但影响不大。控制藻液pH不低于平衡pH,可以提高微藻培养中CO2的利用效率,有利于实现微藻CO2生物固定的环境效益。    相似文献
