Previously, we used computer-controlled fermentation technology to improve the yield of filamentous phage produced in Escherichia coli by 10-fold (Grieco et?al., Bioprocess Biosyst Eng 32:773-779, 2009). Purpose

To conduct high-resolution imaging of the retinal nerve fiber layer (RNFL) in normal eyes using adaptive optics scanning laser ophthalmoscopy (AO-SLO).

Methods

AO-SLO images were obtained in 20 normal eyes at multiple locations in the posterior polar area and a circular path with a 3–4-mm diameter around the optic disc. For each eye, images focused on the RNFL were recorded and a montage of AO-SLO images was created.

Results

AO-SLO images for all eyes showed many hyperreflective bundles in the RNFL. Hyperreflective bundles above or below the fovea were seen in an arch from the temporal periphery on either side of a horizontal dividing line to the optic disc. The dark lines among the hyperreflective bundles were narrower around the optic disc compared with those in the temporal raphe. The hyperreflective bundles corresponded with the direction of the striations on SLO red-free images. The resolution and contrast of the bundles were much higher in AO-SLO images than in red-free fundus photography or SLO red-free images. The mean hyperreflective bundle width around the optic disc had a double-humped shape; the bundles at the temporal and nasal sides of the optic disc were narrower than those above and below the optic disc (P<0.001). RNFL thickness obtained by optical coherence tomography correlated with the hyperreflective bundle widths on AO-SLO (P<0.001)

Conclusions

AO-SLO revealed hyperreflective bundles and dark lines in the RNFL, believed to be retinal nerve fiber bundles and Müller cell septa. The widths of the nerve fiber bundles appear to be proportional to the RNFL thickness at equivalent distances from the optic disc.  相似文献

Hyperspectral reflectance (350–2500 nm) measurements were made over two experimental rice fields containing two cultivars treated with three levels of nitrogen application. Four different transformations of the reflectance data were analyzed for their capability to predict rice biophysical parameters, comprising leaf area index (LAI; m2 green leaf area m−2 soil) and green leaf chlorophyll density (GLCD; mg chlorophyll m−2 soil), using stepwise multiple regression (SMR) models and support vector machines (SVMs). Four transformations of the rice canopy data were made, comprising reflectances (R), first-order derivative reflectances (D1), second-order derivative reflectances (D2), and logarithm transformation of reflectances (LOG). The polynomial kernel (POLY) of the SVM using R was the best model to predict rice LAI, with a root mean square error (RMSE) of 1.0496 LAI units. The analysis of variance kernel of SVM using LOG was the best model to predict rice GLCD, with an RMSE of 523.0741 mg m−2. The SVM approach was not only superior to SMR models for predicting the rice biophysical parameters, but also provided a useful exploratory and predictive tool for analyzing different transformations of reflectance data.  相似文献

Measuring Raman spectra through an optical fibre is usually complicated by the high intrinsic Raman scatter of the fibre material. Common solutions such as the use of multiple fibres and distal optics are complex and bulky. We demonstrate the use of single novel hollow‐core negative‐curvature fibres (NCFs) for Raman and surface‐enhanced Raman spectroscopy (SERS) sensing using no distal optics. The background Raman emission from the silica in the NCF was at least 1000× smaller than in a conventional solid fibre, while maintaining the same collection efficiency. We transmitted pump light from a 785‐nm laser through the NCF, and we collected back the weak Raman spectra of different distal samples, demonstrating the fibre probe can be used for measurements of weak Raman and SERS signals that would otherwise overlap spectrally with the silica background. The lack of distal optics and consequent small probe diameter (<0.25 mm) enable applications that were not previously possible.   相似文献

1,3-propanediol (1,3-PD) is a chemical compound of immense importance primarily used as a raw material for fiber and textile industry. It can be produced by the fermentation of glycerol available abundantly as a by-product from the biodiesel plant. The present study was aimed at determination of key kinetic parameters of 1,3-PD fermentation by Clostridium diolis. Initial experiments on microbial growth inhibition were followed by optimization of nutrient medium recipe by statistical means. Batch kinetic data from studies in bioreactor using optimum concentration of variables obtained from statistical medium design was used for estimation of kinetic parameters of 1,3-PD production. Direct use of raw glycerol from biodiesel plant without any pre-treatment for 1,3-PD production using this strain investigated for the first time in this work gave results comparable to commercial glycerol. The parameter values obtained in this study would be used to develop a mathematical model for 1,3-PD to be used as a guide for designing various reactor operating strategies for further improving 1,3-PD production. An outline of protocol for model development has been discussed in the present work.  相似文献

通过考察氯过氧化物酶(CPO)发酵相关参数的动力学特征,探究酶产量与参数之间的关联性。结果表明:CPO发酵过程中慢速C源麦芽糖比葡萄糖更有利于调控CPO的稳定合成,前者CPO最高比酶活(179.50 U/m L)比后者(135 U/m L)高出44.5 U/m L,而且产酶高峰期延迟1~2 d;发酵过程p H波动与C源消耗速率密切相关,且对CPO合成具有明显的指标性作用。通过生物量曲线及糖消耗曲线与产酶特征对比判断,菌株合成CPO为中期合成类型。副产物黑色素是菌体成熟时期的一种次生代谢物质,与酶的生物合成存在时间上的同步性。控制C源基质和p H对提高CPO稳定化生产具有一定成效。  相似文献

Fermentation parameters for phytase production in column-type bioreactor were monitored using a new data acquisition system. There are a number of studies reporting phytase production in flasks, but a lack of data about microorganism respiration behaviour during phytase production using column bioreactor. The objectives of this work were the monitoration of fermentation parameters during phytase production and its relation with fungal growth and forced air. Phytase production by A. niger FS3 increased with forced air. The O2 consumption and CO2 production during solid-state fermentation were monitored by sensors (in the bottom and top of the columns) linked to controllers, recorded by acquisition software and processed by Fersol2® software tool. Phytase synthesis was associated with fungal growth. Therefore, phytase could be used to estimate FS3 biomass formed in citric pulp degradation.  相似文献

Aspergillus oryzae MTCC 5341, when grown on wheat bran as substrate, produces several extracellular acid proteases. Production of the major acid protease (constituting 34% of the total) by solid-state fermentation is optimized. Optimum operating conditions obtained are determined as pH 5, temperature of incubation of 30°C, defatted soy flour addition of 4%, and fermentation time of 120 h, resulting in acid protease production of 8.64 × 105 U/g bran. Response-surface methodology is used to generate a predictive model of the combined effects of independent variables such as, pH, temperature, defatted soy flour addition, and fermentation time. The statistical design indicates that all four independent variables have significant effects on acid protease production. Optimum factor levels are pH 5.4, incubation temperature of 31°C, 4.4% defatted soy flour addition, and fermentation time of 123 h to yield a maximum activity of 8.93 × 105 U/g bran. Evaluation experiments, carried out to verify the predictions, reveal that A. oryzae produces 8.47 × 105 U/g bran, which corresponds to 94.8% of the predicted value. This is the highest acid protease activity reported so far, wherein the fungus produces four times higher activity than previously reported [J Bacteriol 130(1): 48–56, 1977].  相似文献

In a fermentation process, the establishment of gas mass balances provides valuable information and allows both measurements concerning the characteristics of the biomass itself and the monitoring of a cultivation process. If the quantity and oxidation level of substances excreted into the fermentation broth are known or constant, the yield factor and the dry cell-weight production are stoichiometrically related to the quantity of CO2 evolved and to the quantity of O2 consumed. Where frequent measurements of both yield factor and dry cell-weight production are desirable or where rapid adjustment of the parameters is necessary, on-line identification of these parameters is required, An algorithm allowing the identification of the specific growth rate is presented. Molecular biosensors are devices of molecular size that are designed for sensing different analytes on the basis of biospecific recognition. They should provide two coupled functions - the recognition (specific binding) of the target and the transduction of information about the recognition event into a measurable signal. The present review highlights the achievements and prospects in design and operation of molecular biosensors for which the transduction mechanism is based on fluorescence. We focus on the general strategy of fluorescent molecular sensing, construction of sensor elements, based on natural and designed biopolymers (proteins and nucleic acids). Particular attention is given to the coupling of sensing elements with fluorescent reporter dyes and to the methods for producing efficient fluorescence responses.  相似文献

We have numerically solved the photon diffusion equation to predict the distribution of light in a tissue model system with a uniform concentration of fluorophore. Our results show that time-dependent measurements of light propagation can be used to monitor the fluorescent lifetimes of a uniformly distributed fluorophore in tissues. With proper referencing, frequency-domain measurements of phase-shift, theta, may allow quantitation of fluorescent lifetimes, tau, independent of changes in the local absorption and scattering properties. These results point to a new approach for noninvasive diagnostic monitoring through quantitation of fluorescent lifetime, tau, when the lifetime of the fluorophore is comparable with photon migration times.  相似文献

The final parameters of tetracycline biosynthesis in different fermentation apparatus were analysed comparatively with an account of possible changing of the fermentation broth volume against the initial one due to water evaporation. It was shown that for comparison of the biosynthesis parameters in different fermentation apparatus it is necessary to reestimate the activity values or the antibiotic concentration in a unit of the medium volume with respect to the initial volume of the fermentation medium charged into the apparatus.  相似文献
