After chronic administration of a dilute solution of silver nitrate in drinking water to rats, mice, and guinea pigs, granular deposits of metallic silver were detected in electron micrographs of the kidney, liver, thyroid, and pancreas. The silver deposits were in the form of extremely dense, angular particles with sharp outlines. They varied from aggregates a few microns in diameter down to granules at the limit of resolution of the electron microscope. The principal sites of deposition were (1) basement membranes, especially those of the renal glomeruli, proximal convoluted tubules, and various glands, and those associated with vascular endothelium, and (2) the cytoplasm of fixed and free macrophages. Both in Kupffer cells lining hepatic sinusoids and in the wandering macrophages of other tissues, the silver was segregated in discrete vacuoles. In addition, granular deposits were observed in occasional vesicular structures in the proximal convoluted tubules of the kidney, the hepatic cells, and the pancreatic acinar cell. These structures, in favorable preparations, contained an outer double layered membrane and internal folds similar to those of mitochondria, from which they appear to have been derived. The significance of these findings in heavy metal poisoning and in cellular physiology is briefly discussed.  相似文献

Experimental metastatic calcification in the proximal convoluted tubules of rat kidney, produced by large doses of vitamin D, has been studied with a variety of techniques. These techniques include the examination of thin sections of Araldite-embedded material under the electron microscope, selected area electron diffraction, and several histochemical methods. Two types of mineral are found in relation to the proximal convoluted tubule. The first form consists of aggregates of elongated crystals within cytoplasmic vacuoles of the proximal tubular cells. The dimensions of these crystals are consistent with those of hydroxyapatite. The other type of mineral deposit is found in and adjacent to the extracellular phase of the basal infoldings of these tubules. The latter deposits are made up of smaller crystals arranged in layers. These crystals could not be definitely identified by means of selected area electron diffraction. The observations are discussed in relation to calcium transport by the proximal convoluted tubule and also in terms of mechanisms of pathological calcification.  相似文献

Kallikrein has been localized in rodent kidney and salivary glands by means of an immunoglobulin-enzyme bridge technique. In sections of kidney, anti-kallikrein antibodies bound to the apical region of certain distal tubule segments in the cortex, to reabsorption droplets of proximal convoluted tubules, and to certain duct segments in the papilla. In salivary glands of both male and female rats and mice, and apical rim of most striated duct cells of submandibular, parotid and sublingual glands and granular tubules of submandibular glands exhibited immunoreactivity. Granular intercalated duct cells in female submandibular glands also displayed immunostaining for kallikrein. Phenylephrine administration resulted in loss of immunoreactive granules from the granular convoluted tubule cells of male mouse submandibular gland. This response was paralleled by a biochemically demonstrable decrease in kallikrein-like tosylarginine methyl ester (TAME) esterase activity.  相似文献

In the convoluted proximal tubules of the kidney of pregnant rats alkaline phosphatase activity decreases from 21 days gestation till the 2nd day after birth. During the same time the reaction product of the histochemical succinate dehydrogenase assay exhibits an atypical granular character in some of the proximal tubules, which sometimes additionally contain sudanophilic structures. Electron microscopy reveals dilated mitochondria, an increased number of autophagosomes and big lipid vacuoles of low density.  相似文献

Immunohistochemically detectable erythropoietin-like substance(Epo) in granular convoluted tubule(GCT) cells of submandibular glands (SMG's) was examined in mice in which hemolytic anemia had been induced by phenylhydrazine (ph), and in mice subjected to hypoxia, nephrectomy, or testosterone (TP) injections. Staining for Epo was negative in GCT cells of SMG's in normal mice, while positive staining occurred in GCT cells of the anemic mice and mice subjected to hypoxia or nephrectomy. A positive Epo reaction was also revealed at the luminal borders of distal tubules, and in cells of proximal and distal tubules in the kidney, and in some hepatic and spleen cells, of mice that had received combination regimens producing anemia and hypoxia, or had been nephrectomized. Increased staining of Epo was found in GCT cells of SMG's, and in proximal and distal kidney tubules of mice given the combination treatment plus TP injections. The detection of Epo in GCT cells suggests these extrarenal cells to be sites of accumulation or biosynthesis of the protein under certain specific conditions such as hemolytic anemia and hypoxia.  相似文献

The cytoplasmic uptake of 3H-L-leucine and 3H-L-proline by hepatocytes and cells of the proximal and distal convoluted and of the collecting tubules of the kidney was compared with that of 3H-DL-leucine and 3H-DL-proline in an autoradiographic study. 34 male white Sprague-Dawley rats were killed 1, 2, 6, and 24 hours after the intraperitoneal injection of these amino acids. The rate of incorporation of 3H-L-leucine in the liver and in the renal tubules, as judged by the number of silver grains counted, was about twice that of 3H-L-proline. In the tubules of the kidney the intensity of labelling progressively declined from the proximal convoluted to the collecting tubules. When the two 3H-DL-amino acids were used, almost identical rates of incorporation were found in the liver as well as in the kidney. The only exception was the pars recta of the proximal tubule: Here there could be found an unusually high uptake of 3H-DL-proline. The values were not only higher than those found for the uptake of 3DL-leucine in this particular segment, but they also surpassed those due to 3H-DL-proline and 3DL-leucine in the other parts of the renal tubules, as well as in the liver. The conspicuously high labelling seen in the pars recta after the injection of 3H-DL-proline suggests that there is present in the cells of this segment a d-amino acid oxidase, which may be relatively specific for D-proline. The possibility is considered that this enzyme may participate in a detoxifying function of the pars recta.  相似文献

Recent evidence suggests that a local reninangiotensin system is operational in the kidney and that it mediates some of the actions of angiotensin II on renal tubules. In this study the ontogeny and renal distribution of the unique precursor to angiotensin II formation, angiotensinogen, was investigated in rats by use of immunohistochemistry, immuno-electron microscopy and non-isotopic hybridization histochemistry. At the light-microscopic level, intense staining for angiotensinogen was found in the proximal convoluted tubules of the cortex, with lighter staining in the straight proximal tubules of the outer stripe. The strongest immunostaining was found in the kidneys of neonatal rats, where glomerular mesangial cells and medullary vascular bundles were also immunopositive. The angiotensinogen content of the kidneys in late gestation embryos and neonates showed the presence of angiotensinogen by day E18 and a peak content in the neonate. Non-isotopic hybridization histochemistry with biotinylated oligodeoxynucleotide probes confirmed the presence of angiotensinogen mRNA expression in the proximal convoluted tubules of the renal cortex. Electron-microscopic immunohisto-chemistry showed staining of relatively few electron-dense structures close to the apical membrane of proximal convoluted tubule cells in the adult kidney. In the neonatal rat kidney, angiotensinogen immunostaining at the electron-microscopic level was found throughout the proximal tubule cells and was markedly stronger than that seen in adult kidney. The presence of angiotensinogen, from embryonic day 18, in the proximal tubules, mesangial cells and vasculature of the kidney suggests multiple potential sites of intrarenal angiotensin II generation with an ontogeny in late gestation.  相似文献

The size, number, and location of lysosomes, phagosomes, and phago-lysosomes in different segments of the proximal and distal tubules, in the collecting tubules, and in invading macrophages of the kidneys of rats were compared by staining lysosomes (acid phosphatase) red, and phagosomes (injected horseradish peroxidase) blue in separate sections, and by staining phago-lysosomes purple by successive application of the reactions for the two enzymes in the same sections. It was concluded from these observations that the absorption of the foreign protein from the lumen and its gradual digestion in large phago-lysosomes took place mainly in the cells of the proximal convoluted tubules of the outer cortex. Several segments of the proximal convoluted tubules were distinguished on the basis of differences in the size and location of the phago-lysosomes and the amounts of peroxidase ingested. The distal tubules showed, in addition to moderate numbers of phago-lysosomes, many small phagosomes in the apical and basal zones of the cells. Moderate numbers of phagosomes and phago-lysosomes were observed in the cells of the collecting tubules. Macrophages showing very large phago-lysosomes were seen in the peritubular capillaries of the medulla, after injection of peroxidase. When high doses of peroxidase were administered, enlarged phago-lysosomes, parts of which seemed to be extruded into the lumen, were formed in the terminal segments of the proximal convoluted tubules.  相似文献

When rat kidney slices were incubated in the presence of horseradish peroxidase, there was an energy-dependent uptake of the protein by the cells of the kidney tubules. The uptake was greatest in the proximal convoluted tubules and in the thick ascending limbs of the loops of Henle; it was abolished by cold, anoxia, 2,4-dinitrophenol, and fluoroacetate, and was more readily depressed by unfavorable metabolic conditions in the proximal convoluted tubules than in the thick ascending limbs. Protein uptake was inhibited when the kidney slices were incubated in electrolyte-free media. In sodium chloride solutions, uptake was reduced as sodium was progressively replaced by choline, and ouabain inhibited uptake in the proximal convoluted tubules, but not in the thick ascending limbs. To a limited extent, lithium could replace sodium in the incubation medium with no depression of peroxidase uptake. These results suggest that a sodium-stimulated, ouabain-sensitive ATPase may be involved in the uptake of protein by cells of the kidney tubule. The intracellular transport of peroxidase in cells of the proximal convoluted tubules was abolished by cold, anoxia, and 2,4-dinitrophenol, but it was not affected by concentrations of ouabain which inhibited the uptake of the protein.  相似文献

The cellular and subcellular localization of epidermal growth factor in the submandibular glands of male and female adult mice was established by immunoperoxidase techniques. In light microscopic preparations epidermal growth factor was found exclusively in the granular convoluted tubules of the gland. The intensity of staining for epidermal growth factor varied from cell to cell, and some cells apparently were negative. The pattern of staining was similar in the glands of male and female mice; however, the granular convoluted tubules are androgen-responsive, and thus more extensive and composed of larger cells in males. In thin sections epidermal growth factor was most heavily concentrated in the secretion granules of the granular convoluted tubule cells. Within a given cell there was variation in intensity of staining of individual secretion granules, with some granules appearing minimally reactive or negative. The only other cell component with deposits of reaction product was the ribosomes.  相似文献

Such ordered arrangement of dense particles was encountered inside as well as outside of the cytoplasmic organelles for which the name "siderosomes" has been proposed previously, and which may be derived from mitochondria. Study of hemosiderin granules in hepatic parenchymal and reticuloendothelial cells of human beings yielded similar results. Membrane traffic after inhibition of endocytosis in renal proximal tubules   总被引:3,自引:0,他引:3  
This study was performed to examine quantitatively the cellular organelles involved in membrane recycling after inhibition of luminal endocytosis in renal proximal tubules. Paraffin oil was microinfused into rat renal proximal convoluted tubules to prevent luminal endocytosis. After 1-2 hr the kidneys were fixed by perfusion and prepared for electron microscopy. Segment 1 proximal tubules infused with paraffin oil and control tubules from the same kidney were studied. In addition we examined proximal tubules from kidneys fixed by immersion 30 sec after removal of the kidney. In the oil-infused tubules the large endocytic vacuoles (greater than 0.5 micron) disappeared, the amount of small endocytic vacuoles (less than 0.5 micron) was reduced to about 10%, and the amount of dense apical tubules was significantly increased. The dense apical tubules were very seldom seen connected to the apical plasma membrane in controls but this was occasionally observed in tubules fixed by immersion and relatively often in oil-infused tubules. An ultrastructural morphometric analysis substantiated and extended the qualitative observations and provided quantitative estimates of volumes and surface areas for large endocytic vacuoles, lysosomes, mitochondria, small endocytic vacuoles, and dense apical tubules in control and experimental tubules. The results strongly support the suggestion that the dense apical tubules located in the apical cytoplasm represent the vehicle for the recycling of membrane from endocytic vacuoles back to the plasma membrane, and show that in renal proximal tubule cells small and large endocytic vacuoles are transformed into dense apical tubules when endocytosis is stopped.  相似文献

Several papers reported the role of TASK2 channels in cell volume regulation and regulatory volume decrease (RVD). To check the possibility that the TASK2 channel modulates the RVD process in kidney, we performed primary cultures of proximal convoluted tubules (PCT) and distal convoluted tubules (DCT) from wild-type and TASK2 knockout (KO) mice. In KO mice, the TASK2 coding sequence was in part replaced by the lac-Z gene. This allows for the precise localization of TASK2 in kidney sections using beta-galactosidase staining. TASK2 was only localized in PCT cells. K+ currents were analyzed by the whole-cell clamp technique with 125 mM K-gluconate in the pipette and 140 mM Na-gluconate in the bath. In PCT cells from wild-type mice, hypotonicity induced swelling-activated K+ currents insensitive to 1 mM tetraethylammonium, 10 nM charybdotoxin, and 10 microM 293B, but blocked by 500 microM quinidine and 10 microM clofilium. These currents were increased in alkaline pH and decreased in acidic pH. In PCT cells from TASK2 KO, swelling-activated K+ currents were completely impaired. In conclusion, the TASK2 channel is expressed in kidney proximal cells and could be the swelling-activated K+ channel responsible for the cell volume regulation process during osmolyte absorptions in the proximal tubules.  相似文献

In the metanephros of foetuses beginning from the 14 days of development and in the newborn mice from females with injured kidneys, marked defects of proximal convoluted tubules and less marked defects of glomeruli were noted what corresponded to the character of kidney injury in their mothers. At the same time the diameter of glomeruli and convoluted tubules, the height and width of their cells increased, but the diameter of their nuclei decreased. These changes are accompanied by distrophic changes, mainly in the cells of convoluted tubules, the most markedly expressed in the newborn mice. No marked histological changes were found in the metanephros of two months old experimental mice.  相似文献

Proximal convoluted, proximal straight, and cortical collecting tubular segments isolated from rabbit kidney were perfused with I 125-labeled rabbit serum albumin (RSA-I 125) in ultrafiltrate of serum for up to 3 hr After perfusion, the segments were fixed with glutaraldehyde, embedded in Epon, and either counted with a gamma spectrometer to quantitate protein accumulation or analyzed by electron microscope radioautography to sequentially localize radioactivity Proximal convoluted and proximal straight segments accumulate RSA-I 125 nearly linearly as a function of time whereas cortical collecting segments do not accumulate measurable amounts of protein. The rate of accumulation of RSA-I 125 in the proximal convoluted tubule is 2 6 times as great as that in the proximal straight tubule. Electron microscope radioautography of the isolated proximal tubule demonstrated that RSA-I 125 is taken up via small apical vesicles and tubular invaginations, released into large cytoplasmic vacuoles, and finally concentrated in membrane-bounded structures, some of which are acid phosphatase positive These results show that albumin is absorbed by proximal tubules and may be degraded intracellularly within lysosomes. In addition, less radioactivity was located at all times over the lateral intercellular and basilar labyrinthine spaces, suggesting that labeled albumin and/or its breakdown products may be transported across the peritubular cell membrane.  相似文献

Silver nitrate staining of decalcified bone sections is known to reveal osteocyte canaliculi and cement lines. Nucleolar Organising Regions (NOR) are part of the nucleolus, containing argyrophilic proteins (nucleoclin/C23, nucleophosmin/B23) that can be identified by silver staining at low pH. The aim of this study was to clarify the mechanism explaining why AgNOR staining also reveals osteocyte canaliculi. Human bone and kidney sections were processed for silver staining at light and electron microscopy with a modified method used to identify AgNOR. Sections were processed in parallel for immunohistochemistry with an antibody direct against osteopontin. Protein extraction was done in the renal cortex and decalcified bone and the proteins were separated by western blotting. Purified hOPN was also used as a control. Proteins were electro-transferred on polyvinylidene difluoride membranes and stained for AgNOR proteins. In bone, Ag staining identified AgNOR in cell nuclei, as well as in osteocyte canaliculi, cement and resting lines. In the distal convoluted tubules of the kidney, silver deposits were also observed in cytoplasmic granules on the apical side of the cells. Immunolocalization of osteopontin closely matched with all these locations in bone and kidney. Ag staining of membranes at low pH revealed bands for NOR proteins and 56 KDa (kidney), 60KDa (purified hOPN) and 75 KDa (bone) bands that corresponded to osteopontin. NOR proteins and osteopontin are proteins containing aspartic acid rich regions that can bind Ag. Staining protocols using silver nitrate at low pH can identify these proteins on histological sections or membranes.  相似文献

Varanus niloticus (Linnaeus, 1766), the Nile monitor lizard, is considered the largest lizard in Africa and one of the most widespread. The Egyptian Nile monitor lizard exists in variable habitats, from grasslands to rainforests. This study pointed to investigate the light and ultrastructural features of the renal tissue of this lizard. Microscopically, the lizard nephron deprived from loop of henle and no demarcation could be detected between the cortical and medullary tissues. The renal corpuscles were small, but complex structures. The proximal convoluted tubules were lined by acidophilic cuboidal cells with hemosiderin pigment in their apical cytoplasm. The cytoplasm of the distal convoluted tubular cells reacted strongly with alcian blue stain. The sexual segment of the lizard kidney was lined with high columnar cells with massive periodic acid–Schiff-positive/alcian blue-negative supra-nuclear granules. Ultrastructurally, the basal infoldings of the proximal convoluted cells were evident. Supra-nuclear electron-dense vesicles were detected in the cytoplasm of the sexual segment cells. In summary, the kidney of the Egyptian Nile monitor lizard shares many histochemical features with other reptiles. However, they own several structural specializations in order to adapt to their harsh environments. Future studies focusing on the histochemical components of the sexual segment secretion would be required.  相似文献

 The bifunctional protein PCD/DCoH is both a pterin-4α-carbinolamine dehydratase (PCD) involved in the recycling of tetrahydrobiopterin (BH4) and a dimerisation cofactor (DCoH) of the hepatic nuclear factor 1α (HNF-1α). An antiserum raised against rat PCD/DCoH was used to localise the protein in peripheral organs. In liver, all the hepatocytes but not the other cell types are immunoreactive. In kidney, the protein is prevalent in the proximal and distal convoluted tubules. In adrenals, all the cells of the medulla are labelled. Positive nerve cells occur in myenteric ganglia of the whole gastrointestinal tract and in the intestinal submucous ganglia. Many positive endocrine cells are present in the epithelium. The immunoreactivity is either cytoplasmic (hepatocytes, convoluted tubules of the kidney and part of the gastrointestinal endocrine cells) or prominently nuclear (kidney collecting tubules, adrenals, intestinal neural plexuses and part of the gastrointestinal endocrine cells). Our results show that PCD/DCoH is present in cells expressing enzymes that use BH4 as a cofactor and/or HNF-1α. In addition, PCD/DCoH is present in other cells, for example, neurons in the submucosal plexus. This fact and the prominent nuclear immunoreactivity found in all the positive cells derived from the neural crests argue in favour of a new, still unknown function for the protein. A number of organs from adult female mice were investigated after continuous application of the anticonvulsant drug valproic acid (VPA) by enzyme cytochemistry, light and electron microscopy, pharmacokinetics and clinical chemistry. VPA plasma levels were maintained between 55 micrograms/ml and 67 micrograms/ml for three days following subcutaneous implantation of drug reservoirs. Effects detectable by enzyme cytochemical or electron microscopical means were mainly observed in liver, kidney, thymus and spleen. A strict concentration-dependency of drug effects could not be found. In the liver, the activities of some surface-membrane hydrolases were increased at the biliary pole; the activities of other hydrolases were decreased or unchanged. Electron microscopically, number and length of microvilli of hepatocytes were increased and many of them showed fat inclusions, mitochondrial swellings and autophagic vacuoles. In some of the proximal convoluted tubules of the kidney, the reaction product originating from microvillous and lysosomal hydrolases was diffusely distributed and its amount lowered. This was paralleled by tubular cells with an increased number of fat droplets and swollen mitochondria or destroyed tubular cells, as demonstrated by electron microscopy. Additionally, peritubular endothelial cells were arranged in a garland-like pattern. Alkaline phosphatase was activated in the straight portion of the proximal tubules. Increased glucose, creatinine and total protein concentrations and increased gamma-glutamyl transpeptidase and alkaline phosphatase activities in the urine reflected well the damage of the proximal renal tubules. Cortical and medullary morphology varied considerably in the thymus. In extreme cases, the cortical zone was either reduced in size or the medulla showed a cortex-like structure or vice versa (inverted type of thymus). The thymic cortical reticular cells showed increased aminopeptidase A activity accompanied by a generalized aminopeptidase M and alkaline phosphatase reaction. Our data indicate that--in addition to the liver--also the kidney, thymus and spleen are target organs of VPA-induced toxicity in the mouse.  相似文献
