应用Wolbachiawsp基因特异引物, 通过PCR扩增法检测了二斑叶螨Tetranychus urticae Koch 12个地理种群(辽宁兴城、河北昌黎、湖南长沙、河南郑州、江苏徐州、甘肃兰州、云南昆明、山东泰安、新疆石河子、宁夏银川、陕西眉县和上海)感染Wolbachia的状况。结果表明,12个地理种群全部感染了Wolbachia。克隆分离得到了它们的wsp基因序列,在GenBank的登录号依此为: AY585714、 AY585712、AY785376、AY785375、AY585713、AY785374、AY785371、AY712954、AY712955、AY785372、AY785377和AY785373。对序列进行系统发育分析,发现它们全部与B大组Ori组的Wolbachia株系十分相近或完全相同。总体来看,它们的聚类关系基本上呈平行交叉分布,在不同种群之间没有构成明显的地理分布聚集性。  相似文献

Wolbachia是一种广泛分布于节肢动物体内的共生细菌, 该共生菌经寄主卵的细胞质传播并参与多种寄主生殖活动调控, 对节肢动物的物种进化有着重要意义并可能应用于害虫的生物防治。本研究以柑桔黄龙病(Huanglongbing, HLB)的主要传播媒介——柑桔木虱Diaphorina citri为研究对象, 通过Wolbachia的16S rDNA、 细胞分裂基因(ftsZ)、 表面蛋白基因(wsp)的特异引物对来自于中国广西北海、 广西柳州、 广东深圳、 广东阳春、 福建福州5个地区的柑桔木虱进行PCR扩增, 并对扩增产物进行克隆测序, 与GenBank中相关序列进行比对分析, 建立了柑桔木虱共生Wolbachia的系统发育树。结果显示上述5个地区的柑桔木虱均存在Wolbachia感染, 通过Wolbachia的16S rDNA, ftsZ基因和wsp基因DNA序列的系统发育分析表明, 5个地区的柑桔木虱体内的Wolbachia均属于B组; 基于wsp基因的系统发育分析进一步表明5个地区的亚洲柑桔木虱体内的Wolbachia属于B组的Con亚组, 且不同地区柑桔木虱体内的Wolbachia的16S rDNA, ftsZ基因和wsp基因序列差异不明显。研究结果为认识柑桔黄龙病传播媒介昆虫的进化及其综合防治提供了重要的参考依据。  相似文献

Wolbachia是广泛分布于节肢动物体内一类共生细菌,它能够通过多种机制调控宿主的生殖方式。近年来的研究表明,Wolbachia与许多外来生物的成功入侵相关。本文利用长PCR的方法特异扩增了不同生物型烟粉虱(共24个种群)体内Wolbachiawsp基因,结果发现B型和Q型烟粉虱入侵种群体内均未检测到Wolbachia,而在非B/Q型的浙江种群和肯尼亚种群体内检测到了Wolbachia。对该wsp基因进行测序并和已知序列进行同源性分析发现,浙江烟粉虱种群的Wolbachia属于B组Con/Rug亚种群,而肯尼亚种群属于B组Btab1亚种群。Wolbachia的存在与否可能与烟粉虱的成功入侵有一定的关系。图2表2参23  相似文献

应用Wolbachiawsp基因特异引物,通过PCR扩增法对我国朱砂叶螨Tetranychus cinnabarinus7个地理种群进行了检测。在采自黑龙江佳木斯、安徽安庆、江苏镇江和浙江慈溪的4个地理种群中扩增出了596bp左右的Wolbachiawsp基因片段,而在河北威县、山东滨州和湖北赤壁3个地理种群中未发现这个Wolbachia特征基因片段,表明 Wolbachia在我国朱砂叶螨中的侵染较为普遍。通过对我国朱砂叶螨体内感染的 Wolbachiawsp基因序列进行系统发育分析,得出它们全部与B大组的Ori组的Wolbachia株十分相近或完全相同,提示它们可能是相近或相同的株。  相似文献

Wolbachia是一类广泛分布于节肢动物体内,可以引起节肢动物生殖行为改变的细胞内共生细菌。亚洲玉米螟Ostrinia furnacalis (Guenée)是我国常见的鳞翅目害虫,尤其对玉米危害严重。本文通过对Wolbahciawsp基因的PCR扩增及克隆测序,明确了Wolbachia在北京地区亚洲玉米螟野生种群中的分布情况。结果表明,在检测的种群中,存在两种类型的Wolbahcia,分别将其命名为wFurA(GenBank注册号为EU294311)和wFurB(GenBan k注册号为EU294312);系统树分析表明其分别属于Wolbahcia A组和B组。本研究首次明确了在我国亚洲玉米螟的野生种群内存在着Wolbachia的超感染现象。  相似文献

Wolbachia是一类广泛存在于节肢动物体内的共生菌。玉米螟赤眼蜂Trichogramma ostriniae是我国玉米田间的优势赤眼蜂种, 据报道, 赤眼蜂种内有Wolbachia感染。本文利用Wolbachia的16s rDNA和wsp基因引物通过PCR方法对玉米螟赤眼蜂的野生种群进行了调查, 发现以wsp基因为鉴定依据, 检测的所有个体都感染了3种Wolbachia [wOstGDAa (GenBank accession no. EU157103), wOstGDAb (GenBank accession no. EU157104) 和 wOstGDB (GenBank accession no. EU157105)]。本文首次报道了野生赤眼蜂种群内Wolbachia的三重感染率几乎为100%。根据本研究的结果, 可以推测当不同种赤眼蜂寄生同一寄主时, Wolbachia可能会在不同赤眼蜂种间进行横向传播。  相似文献

Wolbachia属是广泛分布于节肢动物生殖组织内的一类共生细菌。这些共生菌通过卵的细胞质传播并参与多种调控其宿主生殖活动的机制,包括:诱导生殖不亲和、诱导孤雌生殖、雌性化、雄性致死和调节繁殖力。Wolbachia被认为与性别决定、共生关系和物种形成等重要生物学问题密切相关,是探索这些研究领域的新线索。而且Wolbachia可作为特定的载体对其宿主种群进行遗传调控,如增强寄生蜂在害虫生物防治中的作用,控制线虫引起的疾病传播。该文综述了Wolbachia的形态学及存在部位、基因组结构、系统发育、种的命名、水平传递和Wolbachia对其宿主生殖活动的调控作用,并分析了Wolbachia研究的科学意义和发展趋势,以期引起我国生物学家对Wolbachia研究的注意和快速切入。  相似文献

测定了我国尖音库蚊复合组和白纹伊蚊蚊虫体内感染的Wolbachia株的wsp基因序列。核苷酸和氨基酸的同源性及系统关系分析表明,我国尖音库蚊复合组和白纹伊蚊中Wolbachia株的wsp基因序列与Pip组其它株的核苷酸及氨基酸同源性分别为98%~100%和97%~100%, 属B大组Wolbachia中的Pip组。  相似文献

Wolbachia是专性的细胞内细菌,广泛存在于节肢动物生殖组织。已有的研究结果表明,节肢动物中存在A组和B组Wolbachia,而烟粉虱Bemisia tabaci中主要检测到了B组Wolbachia。本研究从福建省采集到17个不同烟粉虱地理种群,首先通过rDNA-ITS1克隆测序鉴定了不同烟粉虱地理种群的生物型,然后采用Wolbachia 16S rDNA的特异引物,并通过PCR-RFLP技术分析了不同烟粉虱地理种群中Wolbachia的感染特点。结果表明:从福建省闽侯、平潭、南平、来舟、漳平和沙县采集到的烟粉虱自然种群属于非B型,而非B型烟粉虱种群中存在广泛的超感染现象,即单个非B型烟粉虱个体中同时感染了不同型Wolbachia。相反,B型烟粉虱自然种群的个体中只感染A组Wolbachia。该研究依据密集采样的数据进一步证实了Wolbachia在烟粉虱自然种群中的分布确实与宿主的生物型密切相关,提示Wolbachia可能在烟粉虱的种群分化中发挥作用。  相似文献

Wolbachia是广泛分布于节肢动物体内的一类共生细菌。采用16S rDNA特异片段的PCR-RFLP方法对烟粉虱Bemisia tabaci (Gennadius)不同生物型及米蛾Corcyra cephalonica (Stainton)共生菌Wolbachia进行了检测与分型分析。基于wsp基因对烟粉虱共生菌B组Wolbachia以及米蛾共生菌Wolbachia进行了系统树分析,并对相应的Wolbachia16S rDNA特异片段进行了克隆、测序以及序列比对。结果表明:16S rDNA的特异片段经NheⅠ酶切后RFLP图谱可有效检测与鉴别Wolbachia。烟粉虱共生菌Wolbachia的16S rDNA特异片段经VspⅠ酶切后可得到预期RFLP图谱,而米蛾共生菌B组Wolbachia (基于wsp序列分析为B组)则产生不同的RFLP图谱。序列分析表明,Nauru型烟粉虱体内B组Wolbachia的16S rDNA片段序列与已知B组Wolbachia对应序列(DQ278884)同源性为100%;米蛾体内B组Wolbachia 16S rDNA特异片段有碱基变异,并存在于VspⅠ识别位点内,这是导致VspⅠ酶切后RFLP图谱不同的原因。结果提示,B组Wolbachia 16S rDNA特异片段经VspⅠ酶切的RFLP图谱存在多态性。本研究结果可为今后Wolbachia的检测与分型提供借鉴。  相似文献

Wolbachia are symbiotic bacteria altering reproductive characters of numerous arthropods. Their most recent phylogeny and classification are based on sequences of the wsp gene. We sequenced wsp gene from six Wolbachia strains infecting six Trichogramma species that live as egg parasitoids on many insects. This allows us to test the effect of the addition of sequences on the Wolbachia phylogeny and to check the classification of Wolbachia infecting Trichogramma. The six Wolbachia studied are classified in the B supergroup. They confirm the monophyletic structure of the B Wolbachia in Trichogramma but introduce small differences in the Wolbachia classification. Modifications include the definition of a new group, Sem, for Wolbachia of T. semblidis and the merging of the two closely related groups, Sib and Kay. Specific primers were determined and tested for the Sem group.  相似文献

The alpha‐proteobacteria of the genus Wolbachia is a widespread group of maternally inherited endosymbionts of arthropod and nematode hosts. Wolbachia infection induces a range of host phenotypes, including cytoplasmic incompatibility, male killing, feminization, and induction of thelytokous parthenogenesis. Heterogony (cyclical parthenogenesis) is a remarkable characteristic of oak gallwasps, Cynipini, the largest tribe of the Cynipidae. A few species of Cynipini are exceptional in that they are univoltine and exhibit thelytokous parthenogenesis, probably because they lost the arrhenotokous generation of their heterogonic ancestor species due to Wolbachia infection. In this study, the presence of Wolbachia was detected using polymerase chain reaction primers for the wsp genes in a thelytokous parthenogenetic species [Dryocosmus kuriphilus (Yasumatsu)] (Hymenoptera: Cynipidae: Cynipini). Approximately 29.8 and 87.1% of adults of the Zhuzhou and Fuzhou strains, respectively, were infected with Wolbachia while all females of the remaining four strains collected from other localities in China were Wolbachia free. The length of the wsp fragment of Zhuzhou and Fuzhou strains was found to be 573 and 561 bp, respectively. The nucleotide sequence of the bacterial wsp fragment indicated that the endosymbiotic bacteria of the Zhuzhou and Fuzhou strains are members of supergroup A, but belong to different clades; they probably originated from two independent infection events. In conclusion, thelytokous parthenogenesis of D. kuriphilus is not caused by Wolbachia infection and the deletion of the arrhenotokous generation is thus not associated with such an infection.  相似文献

The inherited bacterium Wolbachia spreads through the manipulation of host reproduction, and has been suggested to be an important factor in arthropod evolution, from host speciation to the evolution of sex-determination systems. Past work has shown that members of this group may produce cytoplasmic incompatibility, feminize genetically male hosts, and induce host parthenogenesis. Here, we report an expansion of the range of reproductive manipulations produced by members of this clade, recording Wolbachia strains that kill male hosts during embryogenesis in two host species, the ladybird Adalia bipunctata, and the butterfly Acraea encedon. Both male-killing bacteria belong to the B group of Wolbachia. However, phylogenetic analyses were unable to resolve whether the bacteria in the two species are monophyletic, or represent independent origins of male-killing among the B-group Wolbachia. We also found significant divergence within the wsp gene of Wolbachia strains found in different A. bipunctata individuals, suggesting this host species contains two Wolbachia strains, diverged in wsp sequence but monophyletic. Our observations reinforce the notion that Wolbachia may be an important agent driving arthropod evolution, and corroborates previous suggestions that male-killing behaviour is easily evolved by invertebrate symbionts.  相似文献

Wolbachia是广泛分布于节肢动物体内的一类共生菌, 它们参与多种调控寄主的生殖活动机制。通过对wsp基因的特异性扩增和测序,发现了Wolbachia在米蛾Corcyra cephalonica (Stainton)体内的感染。利用所测序列和其他已发表的序列建立系统树,结果表明米蛾体内Wolbachia属于B大组的Pip类群,与其寄生物茧蜂及赤眼蜂中的Wolbachia各株系遗传距离相差较远。据此推测米蛾体内感染的Wolbachia不是由寄生物(茧蜂、赤眼蜂)水平传播所致。  相似文献

Wolbachia are a common and widespread group of symbiotic bacteria found in the reproductive tissues of arthropods. Bactrocera dorsalis (Hendel) is an important pest causing considerable economic losses of fruits and vegetables in several southern provinces of China. In this study, polymerase chain reaction (PCR) with general Wolbachia surface protein (wsp) primers was used to test the presence of Wolbachia in 1,500 individuals of B. dorsalis from five geographical populations of China. We detected 19 individuals of B. dorsalis infected by Wolbachia, and the infection rates of different populations varied. Comparison of wsp gene sequences from 19 individuals and search of the GenBank identified four new sequences, probably representing four Wolbachia strains. Sequence comparison showed that the four Wolbachia strains from B. dorsalis in China belonged to three groups (Kue, Mel, and Cuc). Phylogenetic analysis of the wsp sequences suggests that geographical isolation of Wolbachia exists among the populations of B. dorsalis in China, and gene flow of Wolbachia might have occurred between B. dorsalis populations of China and Thailand. Phylogenetic analysis performed on the host mitochondrial cytochrome oxidase I (COI) gene and wsp gene suggests that host has coevolved with Wolbachia.  相似文献

Endosymbionts of the genus Wolbachia were efficiently cured from Trichogramma species by incorporating 0.02% tetracycline into the artificial diet used to rear larvae. Use of this technique yielded stable cured lines (bisexual and arrhenotokous lines) in which no Wolbachia organisms were detected by PCR for up to 14 generations after curing. Four cured strains of Trichogramma pretiosum showed a significantly lower total fecundity compared to their Wolbachia-infected counterpart. However, the fecundity of a single cured strain of Trichogramma evanescens was similar to its Wolbachia-infected counterpart. These differences in the effect on fecundity may be due to differences between the Wolbachia strains infecting T. pretiosum or T. evanescens, providing additional evidence for the hypothesis that a specific interaction exists between some Trichogramma species and their Wolbachia symbionts. Tetracycline in larval diet was also used to generate bisexual strains of Trichogramma oleae and Trichogramma cordubensis so that these species could be crossed with the closely related species, respectively, T. pretiosum and T. evanescens, to test their compatibility. These crosses showed a lack of compatibility, validating maintenance of these as distinct species.  相似文献

Wolbachia are a group of intracellular inherited bacteria that infect a wide range of arthropods. They are associated with a number of different reproductive phenotypes in their hosts, such as cytoplasmic incompatibility, parthenogenesis and feminization. While it is known that the bacterial strains responsible for these different host phenotypes form a single clade within the alpha-Proteobacteria, until now it has not been possible to resolve the evolutionary relationships between different Wolbachia strains. To address this issue we have cloned and sequenced a gene encoding a surface protein of Wolbachia (wsp) from a representative sample of 28 Wolbachia strains. The sequences from this gene were highly variable and could be used to resolve the phylogenetic relationships of different Wolbachia strains. Based on the sequence of the wsp gene from different Wolbachia isolates we propose that the Wolbachia pipientis clade be initially divided into 12 groups. As more sequence information becomes available we expect the number of such groups to increase. In addition, we present a method of Wolbachia classification based on the use of group-specific wsp polymerase chain reaction (PGR) primers which will allow Wolbachia isolates to be typed without the need to clone and sequence individual Wolbachia genes. This system should facilitate future studies investigating the distribution and biology of Wolbachia strains from large samples of different host species.  相似文献

Wolbachia是一类在节肢动物中广泛感染的胞内共生菌。为了了解其在我国蚜虫中的感染情况, 本研究通过扩增wsp基因片段对采集自我国多个地区的3种小麦蚜虫(荻草谷网蚜Sitobion miscanthi、 麦二叉蚜Schizaphis graminum和禾谷缢管蚜Rhopalosiphum padi)和1种大豆蚜虫(大豆蚜Aphis glycines)样品进行了内共生菌Wolbachia的感染检测。结果显示: 3种小麦蚜虫中均未检测出Wolabchia。大豆蚜也仅在采集自北京和杭州的种群中发现了Wolbachia的感染, 感染率分别为95.8%和22.9%, 并且所检测的个体均为单株系感染。wsp基因序列的比对分析显示, 大豆蚜感染的Wolbachia株系与多个亲缘关系较远的昆虫物种中所感染的Wolbachia株系间具有高度一致的基因序列。wsp基因序列构建的系统发育关系和序列一致性均表明大豆蚜感染的Wolbachia株系属于B大组CauB组。本研究为今后探讨Wolbachia在我国蚜虫中的寄主范围和株系多样性提供了数据支持。  相似文献
