Group structure and cohesion along with their changes over time play an important role in the success of missions where crew members spend prolonged periods of time under conditions of isolation and confinement. Therefore, an objective system for unobtrusive monitoring of crew cohesion and possible individual stress reactions is of high interest. For this purpose, an experimental wireless group structure (WLGS) monitoring system integrated into a mobile psychophysiological system was developed. In the presented study the WLGS module was evaluated separately in six male subjects (27–38 years old) participating in a 520-day simulated mission to Mars. Two days per week, each crew member wore a small sensor that registered the presence and distance of the sensors either worn by the other subjects or strategically placed throughout the isolation facility. The registration between two sensors was on average 91.0% in accordance. A correspondence of 95.7% with the survey video on day 475 confirmed external reliability. An integrated score of the “crew relation time index” was calculated and analyzed over time. Correlation analyses of a sociometric questionnaire (r = .35-.55, p< .05) and an ethological group approach (r = .45-.66, p < 05) provided initial evidence of the method''s validity as a measure of cohesion when taking behavioral and activity patterns into account (e.g. only including activity phases in the afternoon). This confirms our assumption that the registered amount of time spent together during free time is associated with the intensity of personal relationships.  相似文献

To understand whether prolonged confinement results in reductions in physical activity and adaptation in the musculoskeletal system, six subjects were measured during 520 d isolation in the Mars500 study. We tested the hypothesis that physical activity reduces in prolonged confinement and that this would be associated with decrements of neuromuscular performance. Physical activity, as measured by average acceleration of the body’s center of mass (“activity temperature”) using the actibelt® device, decreased progressively over the course of isolation (p<0.00001). Concurrently, countermovement jump power and single-leg hop force decreased during isolation (p<0.001) whilst grip force did not change (p≥0.14). Similar to other models of inactivity, greater decrements of neuromuscular performance occurred in the lower-limb than in the upper-limb. Subject motivational state increased non-significantly (p = 0.20) during isolation, suggesting reductions in lower-limb neuromuscular performance were unrelated to motivation. Overall, we conclude that prolonged confinement is a form of physical inactivity and is associated with adaptation in the neuromuscular system.  相似文献

EXEMSI'92 was a 60-day isolation and confinement experiment with an international crew. During this second experiment of the European Space Agency (the first one was a 28-day confinement with 6 men: ISEMSI'90), blood volume regulating hormones and water balance were studied. During ISEMSI'90, stress level was elevated as shown by specific markers and we observed some interesting results which have been already observed before, during bed-rest experiments. Thus, our conclusion was that some of the physiological effects observed during bed rest could be the consequence of the "confinement effect" and/or stress imposed to the subjects during such experiments. The aim of the present physiological study was: 1/to define the origin of the blood pressure increase seen during ISEMSI'90; 2/ to study the blood volume regulating hormones (ANP, renin, aldosterone) variations; 3/ to determine the dehydration level by measuring the total body water with the doubly labeled water (DLW) method and fluid balance in general.  相似文献

The characteristics of psychophysiological adaptation to the simulation of the extended autonomous manned mission to Mars with limited resources and external communication were studied. The behavioral (communication with the external world) and physiological correlates of 105-day life and work in a pressurized compartment of the crew of six male subjects representing Russian and European space agencies under the conditions of sensory deprivation, confinement, monotony, and high autonomy were studied. Psychological, physiological, and biochemical (urinary cortisol and gonadal hormones) testing together with content analysis of the crew??s communication with the external world were made in parallel. The results confirmed, to a considerable extent, the preliminary hypothesis about the serious impact of stressful factors, including conflict, the significant decrease of the variety and scope of communication with the outer world, and social pressure on the verbal behavior and hormone levels in the future Martian crew. The correlation between the metabolism of the gonadal hormones and the volume, content, and creativity of the verbal human behavior during various stages of adaptation to the extended isolation in pressurized compartments has been discovered.  相似文献

Bacterial plasmids and the chromosomal DNA of many organisms adopt naturally the negatively supercoiled conformation. Therefore, the irradiation of such plasmids could be used to model conformational changes of chromosomal DNA associated with externally-induced damage. We have applied dynamic size-sieving capillary electrophoresis (CE) to monitor the damage of three DNA plasmids, over an unprecedented base pair (bp) size range (2870–27 500 bp), upon exposure to γ-radiation (20–400 Gy). Predominantly, CE with UV absorbance detection in the absence of DNA intercalating dyes was employed to preclude undesirable, induced plasmid conformational changes. Plasmid samples and their enzymatic digestion products were analyzed using both CE and slab gel electrophoresis (SGE) in order to verify the conformation of sample components. Relative to SGE, CE analyses revealed more fine structural features of plasmid degradation.  相似文献

For future interplanetary manned spaceflight, mental issues, as well as physiological problems, must inevitably be considered and solved. Mars500 is a high-fidelity ground simulation experiment that involved 520 days of confined isolation for six multinational crewmembers. This experiment provided a good opportunity to perform psycho-physiological and psycho-social researches on such missions. To investigate emotional responses and psychological adaptation over long-term confinement, the International Affective Pictures System (IAPS) was selected as the visual emotional stimuli in this study. Additional data collected and analyzed included the Profile of Mood States (POMS) questionnaire and the levels of four types of plasma hormones: cortisol, 5-hydroxy tryptamine, dopamine, and norepinephrine. The results demonstrated an obvious bias on valence rating for unpleasant stimuli with time (p<0.05), and the correlation between psychological and biochemical data was identified (p<0.05). Overall, we concluded that the confined crew tended to assign positive ratings to negative pictures with time, which might be driven by a defensive system. There was a stage-changing pattern of psychological adaptation of the Mars500 crew, which is similar to the third-quarter phenomenon.  相似文献

Extinction     
A significant proportion of conservationists' work is directed towards efforts to save disappearing species. This relies upon the belief that species extinction is undesirable. When justifications are offered for this belief, they very often rest upon the assumption that extinction brought about by humans is different in kind from other forms of extinction. This paper examines this assumption and reveals that there is indeed good reason to suppose current anthropogenic extinctions to be different in kind from extinctions brought about at other times or by other factors. Having considered – and rejected – quantity and rate of extinction as useful distinguishing factors, four alternative arguments are offered, each identifying a way in which anthropogenic extinction is significantly different from other forms of extinction, even mass extinction: (1) Humans are a different kind of natural cause from other causes of extinction; (2) Extinctions brought about by humans are uniquely persistent; (3) Anthropogenic extinctions are effectively random whereas past mass extinctions are rule-bound; (4) The impact of the current anthropogenic extinction event differs from the impact of other extinction events of the past, such that future recovery may not follow past patterns. Together, these four arguments suggest that the present-day extinction event brought about by humans may be unprecedented and that we cannot clearly extrapolate from past to present recovery from extinctions. Although insufficient as justification for the claim that present-day extinctions are undesirable, the arguments provide some ammunition for conservationists' conviction that species extinction – in which humans play an accelerating role – ought to be prevented.  相似文献

Methods to provide accurate accumulation rates for lake models are discussed. Cores were taken in 1979 in two basins of Lake Lucerne, Switzerland, and accumulation rates were calculated by using Pb-210 dating and by a natural landslide marker of 1795 in one basin (Weggis). In the other basin (Horw Bay) the sediment accumulation rates based on the lead method were compared with yearly sedimentation rates measured by sediment traps in 1969/70. At the Weggis station, the core dating yielded sediment accumulation rates of about 400 g dry wt. m–2 y–1 with the lead method, averaged over a sediment depth of 4–20 cm; accumulation was about 700 g dry wt. m–2 y–1 with the marker method, averaged over 0–33 cm. In Horw Bay, the trap method yielded about 1300 g dry wt. M–2 y–1 compared with 400–1000 g dry wt. m–2 y–1 obtained with the lead method and related to various depth intervals. The characteristic sources of error of the three methods as well as several hypotheses for these discrepancies are discussed.  相似文献

Inhibition in the olfactory bulb of the carp was studied by recording potentials from secondary neurons intracellularly. Three types of inhibition — trace, early, and late — can arise in neurons of the olfactory bulb. Trace inhibition corresponds to hyperpolarization about 20 msec in duration, which is closely connected with the spike, but it is not after-hyperpolarization but an IPSP. Early and late inhibition correspond to IPSPs of different parameters. The first has a latency of 0–50 msec (relative to the spike) and a duration of 60–400 msec; the corresponding values for the second are 100–400 msec and 0.5–3 sec. Synopsis Effect of food deprivation and refeeding on metabolic parameters were studied in juvenile Rutilus rutilus, weighing 280–460 mg. Tissue hydration increased with the length of the starvation period, reaching a new steady state after 4–5 weeks. Total protein concentration remained constant at about 60% of dry body mass. The concentration of glycogen decreased during food deprivation, a new steady state being reached at about 30% of control values after 4 weeks. Refeeding caused a dramatic increase of glycogen concentration which exceeded the value in fed controls by 6- to 9-fold. This is seen as a tactic for rapid storage of food energy, to be used later for the synthesis of body materials. With respect to their responses to food deprivation the 12 enzymes investigated formed four groups: (1) activity unaffected by food deprivation or refeeding (COX, THIOL, CK, GOT); (2) activity drops to about 60% of control value during the initial phase of food deprivation but remains constant thereafter (PK, LDH, Pase); (3) slow but continuous decrease in activity during the whole period of starvation, i.e. up to 7 weeks (PFK, OGDH, CS, FBPase); (4) activity increases during food deprivation, decreases again upon refeeding (GPT). A model is discussed which distinguishes between four phases in the general response of young fish to food deprivation and refeeding: stress, transition, adaptation, and recovery.  相似文献

Since there is some question as to whether or not legumin is glycosylated, this storage protein was isolated by various procedures from developing cotyledons of Pisum sativum L. supplied with [14C]-labeled glucosamine and analyzed by sodium dodecylsulfate-polyacrylamide gel electrophoresis. Legumin isolated by the classical method of Danielsson [(1949) Biochem. J. 44, 387–400] a procedure in which globulins extracted with a buffered salt solution are precipitated with ammonium sulfate (70% saturation) and legumin separated from vicilin by isoelectric precipitation, was labeled. The glucosamine incorporated into legumin was associated with low-molecular-weight polypeptides. In contrast, legumin isolated by the method of Casey [(1979) Biochem. J. 177, 509–520], a procedure where legumin is prepared by zonal isoelectric precipitation from globulins precipitated with 40–70% ammonium sulfate, was not labeled. However, the globulin fraction precipitated with 40% ammonium sulfate was labeled and the radioactive glucosamine was associated with low-molecular-weight polypeptides. Legumin isolated from protein bodies [Thomson et al. (1978) Aust. J. Plant Physiol. 5, 263–279] was not extensively labeled. However, the saltinsoluble fraction of protein body extracts was labeled and the radioactivity was associated with low-molecular-weight polypeptides. These results indicate that protein bodies contain a glycoprotein of low-molecular-weight that co-purifies with legumin isolated by the method of Danielsson but that is discarded when isolation methods developed more recently are used.  相似文献

Summary The effect of energy deprivation on the intracellular transport and secretion of thyroglobulin was studied in open follicles isolated from porcine thyroids. Follicles were pulse-labeled with 3H-leucine or 3H-galactose. Labeled thyroglobulin was secreted into the incubation medium where it was isolated by means of immunoprecipitation. Secretion was followed in chase incubations under various experimental conditions using CCCP (carbonyl-cyanide-mchlorophenylhydrazone) or DNP (dinitrophenol), both uncouplers of oxidative phosphorylation, or CN, which inhibits respiration. CCCP (1 M) was shown to inhibit exocytosis by about 80%, DNP (0.1–5 mM) by 45–85%, and CN (0.5–1.1 mM) by 5–55%. By combining CN with the ionophore monensin, which blocks transport through the Golgi complex but does not essentially interfere with exocytosis, evidence was obtained that CN also inhibits transport of thyroglobulin from the Golgi cisternae to the exocytic vesicles by 40%. Electron-miroscopic autoradiography of isolated thyroid lobes from the rat also indicated that transport of 3H-leucine label into the follicle lumen is inhibited in the presence of CCCP or CN. Intracellular ATP content was found to be about 40% of the control level in follicles incubated with CCCP (1 uM) or CN (0.9 mM). The results show that the transport of thyroglobulin from the Golgi complex to the exocytic vesicles as well as from the exocytic vesicles into the follicle lumen is dependent upon metabolic energy. A Space Molecular Sink Research Facility (Molsink) was used to evaluate the ability of microorganisms to survive the vacuum of outer space. this facility could be programmed to simulate flight spacecraft vacuum environments at pressures in the 10–10 torr range and thermal gradients (30 to 60°C) closely associated to surface temperatures of inflight spacecraft.Initial populations ofStaphylococcus epidermidis and aMicrococcus sp. were reduced approximately 1 log while exposed to –105 and 34°C, and approximately 2 logs while exposed to 59°C for 14 days in the vacuum environment.Spores ofBacillus subtilis var.niger were less affected by the environment. For many mammalian species short-term fasting is associated with intestinal atrophy and decreased digestive capacity. Under natural conditions, strictly carnivorous animals often experience prey scarcity during winter, and they may therefore be particularly well adapted to short-term food deprivation. To examine how the carnivorous gastrointestinal tract is affected by fasting, small-intestinal structure, brush-border enzyme activities and hepatic structure and function were examined in fed mink (controls) and mink that had been fasted for 1–10 days. During the first 1–2 days of fasting, intestinal mass decreased more rapidly than total body mass and villus heights were reduced 25–40%. In contrast, tissue-specific activity of the brush-border enzymes sucrase, maltase, lactase, aminopeptidase A and dipeptidylpeptidase IV increased 0.5- to1.5-fold at this time, but returned to prefasting levels after 6 days of fasting. After 6–10 days of fasting there was a marked increase in the activity of hepatic enzymes and accumulation of intra-hepatic lipid vacuoles. Thus, mink may be a useful model for studying fasting-induced intestinal atrophy and adaptation as well as mechanisms involved in accumulation of intra-hepatic lipids following food deprivation in strictly carnivorous domestic mammals, such as cats and ferrets.Communicated by I.D. Maximum activities of manganese-dependent peroxidase (MnP) and lignin peroxidase (LiP) in free cultures of Phanerochaete chrysosporium (ATCC 24725) were 258 U l–1 and 103 U l–1, respectively, in an airlift bioreactor. Immobilisation of the fungus on an inert carrier as well as several design modifications of the bioreactor employed gave MnP activities around 500–600 U l–1 during 9 days' operation. The continuous operation of the latter led to MnP and LiP activities about 140 U l–1 and 100 U l–1, respectively, for two months, without operational problems. Furthermore, the extracellular liquid secreted decolourised the polymeric dye Poly R-478 about 56%.  相似文献

The effects of the watch system of job management on central hemodynamic parameters and the constant potential of the brain were studied in sailors during a long voyage. Twenty crew members were examined during a 120-day translatitudinal voyage. The studies were performed on days 4–6, 40–42, 73–75, and 107–109 of the voyage. It was found that the role of the peripheral regulatory loop in maintaining physiological functions increases during the second half of the voyage and that the functional activity before a watch determines the changes in the parameters by the end of the watch. For each stage of the study, the baseline value of the constant potential of the brain is an important element of functional activity. The system of ratios between parameters before and after a watch that is formed in the early period of the voyage breaks up during the second month, the general reactivity of the body after a watch being independent of the reactivity before the watch; the reactivity is stochastic and is mainly controlled by central mechanisms. In the second month of the voyage, the first signs of a strategy aimed at making the functioning more economical are observed in crew members. The second and third months of the voyage are characterized by a considerable strain of the mechanisms of adaptation, activity, and reactivity in sailors. High concentrations of 422 g sophorolipids l–1 were produced using a two-stage cultivation process: first deproteinized whey concentrate (DWC) containing 110 g lactose l–1 was used for cultivation of the yeast Cryptococcus curvatus ATCC 20509, resulting in 34 g dry weight l–1, 20 g single-cell oil l–1 and reducing the chemical oxygen demand (COD) from 159 g l–1 to 35 g oxygen l–1. Afterwards cells were disrupted by passing the cell suspension directly through a high pressure laboratory homogeniser. After autoclavation, the resulting crude cell extract containing the single-cell oil served as substrate for growth of Candida bombicola ATCC 22214 and for sophorolipid production in a second stage. When the single-cell oil was consumed, repeated feeding of 400 g rapeseed oil l–1 was started increasing the yield of sophorolipids to 422 g l–1. Type 4 glutathione peroxidase (GPx4) is a widely expressed mammalian selenoenzyme known to play a vital role in cytoprotection against lipid hydroperoxide (LOOH)-mediated oxidative stress and regulation of oxidative signaling cascades. Since prokaryotes are not equipped to express mammalian selenoproteins, preparation of recombinant GPx4 via commonly used bacterial transformation is not feasible. A published procedure for isolating the enzyme from rat testis employs affinity chromatography on bromosulfophthalein–glutathione-linked agarose as the penultimate step in purification. Since this resin is no longer commercially available and preparing it in satisfactory operational form is tedious, we have developed an alternative purification approach based on sequential anion exchange, size exclusion, and cation exchange chromatography. Final preparations were found to be essentially homogeneous in GPx4 (Mr  20 kDa), as demonstrated by SDS–PAGE with protein staining and immunoblotting. Specific enzymatic activity was determined using a novel thin-layer chromatographic approach in which the kinetics of phosphatidylcholine hydroperoxide loss or cholesterol-7α-hydroperoxide loss was monitored. A >400-fold purification of active enzyme has been attained. The relatively straightforward isolation procedure described should prove valuable for further functional studies on GPx4, e.g. how its ability to catalyze LOOH reduction compares with that of other LOOH detoxifying enzymes.  相似文献

Synopsis Genetic differentiation and patterns of variability in the endangered Iberian endemic,Aphanius iberus, were analyzed by allozyme electrophoresis as a valuable database for conservation purposes. Genetic variability values expressed as heterozygosity (H = 0.015–0.097) were close to the values found in other members of Cyprinodontidae (H = 0.012–0.123). Polymorphism values (P = 0.125–0.542) were higher than reported in the literature (P = 0.036–0.150) for Cyprinodontidae. Significant correlation existed between salinity values and genetic variability expressed as heterozygosity (r = – 0.76, p < 0.01) and polymorphism (r = – 0.60, p = 0.04). Low genetic variability values (H = 0.024–0.055, P = 0.125–0.292) were exhibited by populations which inhabit salty lagoons. The highest values were found in populations occurring in marshes and irrigation channels (H = 0.051–0.097, P = 0.250–0.542). Associations among genetic, geographic and ecological parameters were tested using a Mantel test indicated that most of the genetic distances were explained by geographic distances but not by ecological factors, suggesting that isolation by distance could be the main factor explaining the differentiation between sites. According to the genetic distances obtained, two mainA. iberus groups were discernible: the Atlantic and Mediterranean. Genetic distances between both groups (DRogers = 0.179–0.261) were higher than values between recognized species of other cyprinodontids (DRogers = 0.11–0.27). On the basis of genetic distances we have dated the fragmentation of both populations to the Upper Miocene-Pliocene when most of the Mediterranean sea dried up. Subsequently, gene flow between Mediterranean and Atlantic populations was interrupted. The results of our genetic analyses suggested the existence of five operational conservation units (OCUs) forA. iberus. These units are defined as a continuous area limited by geographical boundaries, and inhabited by one or more populations sharing the same genetic pattern.  相似文献
