An intraperitoneal injection of an exogenous delta-sleep inducing peptide (DSIP) at a dose of 12 g/100 g body weight shifted the prooxidant–antioxidant balance of free radical process (FRP) in tissues and erythrocytes of rats: the activities of antioxidant enzymes (superoxide dismutase, catalase, glutathione peroxidase, and glutathione reductase) and the concentrations of antioxidants (reduced glutathione in particular) increased. The DSIP stimulated the myeloperoxidase activity in blood neutrophils and had no effect on the activity of xanthine oxidase, a prooxidant enzyme, in the brain and liver. Cold stress displaced the prooxidant–antioxidant balance by increasing the xanthine oxidase activity in tissues and decreasing the myeloperoxidase activity in blood neutrophils; it also inhibited the enzyme antioxidant activities in tissues and erythrocytes that was neutralized by an increased ceruloplasmin activity in blood plasma and by an elevated level of antioxidants in rat blood and tissues. Preliminary administration of DSIP to animals exposed to cold stress restored the prooxidant–antioxidant balance: it normalized the myeloperoxidase activity in blood neutrophils, decreased the xanthine oxidase activity, and increased the activity of antioxidant enzymes in tissues and erythrocytes restoring the antioxidant level. The molecular regulation mechanism of free radical processes by DSIP in tissues under stressful conditions is discussed.  相似文献

Peroral administration of salvipholin in a dose of 50 mg/kg to intact male rats (the body weight 180-220 g) had a positive effect on the carbohydrate-lipid metabolism in the liver and blood serum of animals. Administration of the same dose to rats with alloxan diabetes induced a significant decrease in the content of glucose, free fat acids, triglycerides and lysophospholipids of the liver and blood serum. The level of these components has sharply increased after subcutaneous alloxan administration in a dose of 150 mg/kg, the content of glycogen and pyruvic acid being normalized and insulin deficiency removed. These changes are closely related to salvipholin-promoted restoration of phospholipid spectra of the blood serum and liver of experimental animals disturbed under conditions of insulin deficiency. The possible mechanism of the salvipholin action is analyzed.  相似文献

Experiments were made with white random-bred rats (males) exposed to ethanol. The content of serotonin measured by spectrofluorometry was higher in the hypothalamus, brain stem and intestine, and was lower in the thalamus, striatum liver and blood in the animals predisposed to voluntary alcohol consumption and with lateral position duration 62 +/- 18 min as compared with the animals not predisposed to alcohol consumption and with lateral position duration 196 +/- 23 min, the dose of ethanol being 4.5 g/kg i. p. Thirty minutes after ethanol administration in a dose of 2.5 g/kg i. p. to the alcohol-predisposed rats there was a lowering of the serotonin content in the hypothalamus and an increase in the thalamus, brain stem, liver and blood. Meanwhile in the rats not predisposed to alcohol consumption, the serotonin content rose in the hypothalamus, brain stem, liver, intestine and blood and fell in the thalamus and striatum. It is assumed that the serotoninergic system of the brain may play a role in the formation of "positive" or "negative" attitudes to ethanol in the population of white random-bred rats.  相似文献

On the model of tumor growth of Guerin's carcinoma and experimental modeling of vitamin A provision the biochemical characteristics of tumor cachexia in animals with malignant tumors and different vitamin A provision status were studied. It is determined that tumor growth in the body, deprived of vitamin A, is characterized by negative nitrogen balance, decrease of glucose, free fatty acids and ketone bodies level in the blood serum, which indicates the increase of cancer cachexia.  相似文献

Previous studies have shown that the serum levels of the primary regulators of calcium (Ca) and phosphorus (P) metabolism, 1,25-dihydroxyvitamin D and parathyroid hormone, may change with age. Therefore, the effect of age on the ability of the rat to maintain a positive Ca and P balance was determined. Young (1.5 months) and old (18 months) rats were divided into three groups and fed either a low-Ca, high-P diet; a high-Ca, low-P diet; or a high-Ca, high-P diet. After 14 days, the young rats were in positive Ca and P balance regardless of diet. The old rats on the low-Ca, high-P diet were in negative Ca balance and positive P balance. The old rats on the other diets were in positive Ca and P balance. The negative Ca balance of the old rats was due to decreased intestinal absorption of Ca. Intestinal absorption was assessed by determining the percentage of dietary Ca absorbed in vivo and by measuring the active transport of Ca using the everted gut sac in vitro. Intestinal P absorption showed little change with age, except for a decrease in old rats on the high-Ca, low-P diet. Renal adaptation to dietary Ca and P restriction was similar in both young and old animals. Plasma Ca levels were unchanged with age, but plasma P levels decreased with age regardless of diet. These changes in Ca balance with age may reflect the reported decrease in serum 1,25-dihydroxyvitamin D3 levels and the slight increase in PTH levels with age. The inability of old rats to maintain a positive Ca balance in the face of Ca deprivation is consistent with a general characteristic of the aging process—the decreased ability of an organism to adapt to changes in the external environment.  相似文献

1. The effects of water restriction and complete water deprivation on nitrogen retention and urea recycling were examined in camels, desert sheep and desert goats fed only dry desert grass of 3.2% crude protein content. 2. All three species were in negative nitrogen balance when water was available ad libitum. 3. In camels urea recycling was consistently high (94-97%) and nitrogen balance did not change with treatment. 4. In sheep and goats urea recycling increased from 75% to 94% (sheep) and from 79% to 95% (goats) during water deprivation, and nitrogen balance improved to positive values. 5. In all species water deprivation depressed dry matter intake and increased apparent digestibility of dry matter and nitrogen. 6. The effects of water restriction were generally intermediate. 7. These results suggest that nitrogen metabolism in adult domestic livestock raised by nomadic pastoralists in Sudan is not harmed, and may in fact be improved by short periods of water deprivation.  相似文献

In the present study, the protective effect of curcumin against sodium fluoride-induced nephrotoxicity was evaluated in rats. Renal injury was induced by daily administration of 600 ppm sodium fluoride in drinking water for 1 week. One week before the administration of fluoride, the animals selected as study group were given curcumin (10 and 20 mg/kg body weight, intraperitoneally). After 1 week, lipid peroxidation level, activities of superoxide dismutase, catalase, and level of glutathione in kidney homogenate were measured. Blood serum samples were examined for creatinine, serum urea, and blood urea nitrogen levels. Another group of rats received vitamin C (10 mg/kg) as standard antioxidant. The results show that curcumin and vitamin C treatment prior to fluoride administration normalized the levels of serum creatinine, serum urea, and blood urea nitrogen. Moreover, curcumin and vitamin C administrations prevented the antioxidant enzyme decreasing and lipid peroxidation levels imbalance. In conclusion, curcumin treatment at the doses of 10 and 20 mg/kg (intraperitoneally) showed significant nephroprotective effects.  相似文献

The administration to rats of amino acids (valine, leucine, and isoleucine) for 10 days after whole-body X-irradiation with a dose of 7 Gy increased the survival rate and average life of animals and normalized some carbohydrate and nitrogen metabolism indexes. The aim of the present experiment was to determine if a niacin supplementation of 6 g/d to lactating dairy cow diets can compensate negative effects of a rumen nitrogen balance (RNB) deficit. A total of nine ruminally and duodenally fistulated lactating multiparous German Holstein cows were successively assigned to one of three diets consisting of 10 kg maize silage (dry matter [DM] basis) and 7 kg DM concentrate: Diet RNB- (n = 6) with energy and utilisable crude protein at the duodenum (uCP) according to the average requirement of the animals but with a negative RNB (-0.41 g N/MJ metabolisable energy [ME]); Diet RNB0 (n = 7) with energy, uCP and a RNB (0.08 g N/MJ ME) according to the average requirement of the animals and, finally, Diet NA (n = 5), which was the same diet as RNB-, but supplemented with 6 g niacin/d. Samples of milk were taken on two consecutive days, blood samples were taken on one day pre- and post-feeding and faeces and urine were collected completely over five consecutive days. The negative RNB reduced milk and blood urea content and apparent total tract digestibility of DM, organic matter (OM) and neutral detergent fibre (NDF). Also N excretion with urine, the total N excreted with urine and faeces and the N balance were reduced when the RNB was negative. Supplementation of niacin elevated plasma glucose concentration after feeding and the N balance increased. Supplementing the diet with a negative RNB with niacin led to a more efficient use of dietary N thereby avoiding the negative effects of the negative RNB on the digestibility of DM, OM and NDF.  相似文献

Results of study of several blood biochemical parameters of the harp seal Pagophilus groenlandicus Erxleben, 1777, from birth to the adult state are presented. The following age groups of the animals have been studied—newborns, 1.5–2 months, 1 year, 2 years, 3 years, and adults (older than 6 years). The content of total protein and its fractions, free amine nitrogen, urea, creatinine, glucose, total lipids, total calcium, and inorganic phosphorus was determined in the seal blood plasma. Differences of the degree of expression of levels of all studied parameters in the course of early postnatal ontogenesis of the animals have been established. The most significant metabolic changes in the harp seals were revealed at the period of end of weaning and transition to independent ingestion and then after the first year of life.  相似文献

The effect of experimental metabolic acidosis and its correction for nitrogen and energy metabolism was studied in new-born calves. It was discovered that a change in the acid-base balance towards acidosis causes a sharp increase in "ammoniogenesis", urea formation and inhibition of the tricarboxylic acid cycle, which is also observed in calves suffering from dyspepsia with symptoms of acute diarrhea. Alongside with the use of therapeutic measures for treating dyspepsia of new-born calves, it is necessary to control the acid-base balance of blood in the calves and in case of revealing the acidosis state to use means of its correction.  相似文献

During moderate calorie restriction (CR) the heterotherm Microcebus murinus is able to maintain a stable energy balance whatever the season, even if only wintering animals enter into torpor. To understand its energy saving strategies to respond to food shortages, we assessed protein and energy metabolisms associated with wintering torpor expression or summering torpor avoidance. We investigated body composition, whole body protein turnover, and daily energy expenditure (DEE), during a graded (40 and 80%) 35-day CR in short-days (winter; SD40 and SD80, respectively) and long-days (summer; LD40 and LD80, respectively) acclimated animals. LD40 animals showed no change in fat mass (FM) but a 12% fat free mass (FFM) reduction. Protein balance being positive after CR, the FFM loss was early and rapid. The 25% DEE reduction, in LD40 group was mainly explained by FFM changes. LD80 animals showed a steady body mass loss and were excluded from the CR trial at day 22, reaching a survival-threatened body mass. No data were available for this group. SD40 animals significantly decreased their FM level by 21%, but maintained FFM. Protein sparing was achieved through a 35 and 39% decrease in protein synthesis and catabolism (protein turnover), respectively, overall maintaining nitrogen balance. The 21% reduction in energy requirement was explained by the 30% nitrogen flux drop but also by torpor as DEE FFM-adjusted remained 13% lower compared to ad-libitum. SD80 animals were unable to maintain energy and nitrogen balances, losing both FM and FFM. Thus summering mouse lemurs equilibrate energy balance by a rapid loss of active metabolic mass without using torpor, whereas wintering animals spare protein and energy through increased torpor expression. Both strategies have direct fitness implication: 1) to maintain activities at a lower body size during the mating season and 2) to preserve an optimal wintering muscle mass and function.  相似文献

It was shown that administration in the course of one week, before or after a single use of killed or chemical typhoid vaccine of dibiomycin, biomycin, or biomycin in combination with erythromycin in comparatively high doses produced no negative effect of the production of typhus antibodies and the intensity of antitoxic immunity in albino mice. The same antibiotics failed to influence the antibody formation in guinea pigs if they produced no toxic effect on the animals; but in case of development of toxic phenomena connected with the administration of the mentioned antibiotics a strong depression of antibody production was observed in guinea pigs.  相似文献

Objective To examine the associations between positive and negative affect and subsequent coronary heart disease events independently of established risk factors.Design Prospective cohort study with follow-up over 12 years.Setting 20 civil service departments originally located in London.Participants 10 308 civil servants aged 35-55 years at entry into Whitehall II study in 1985.Main outcome measures Fatal coronary heart disease, clinically verified incident non-fatal myocardial infarction, and definite angina (n=619, mean follow-up 12.5 years).Results In Cox regression analysis adjusted for age, sex, ethnicity, and socioeconomic position, positive affect (hazard ratio=1.01, 95% confidence interval 0.82 to 1.24) and the balance between positive and negative affect, referred to as the affect balance score (hazard ratio=0.89, 0.73 to 1.09), were not associated with coronary heart disease. Further adjustment for behaviour related risk factors (smoking, alcohol consumption, daily fruit and vegetable intake, exercise, body mass index), biological risk factors (hypertension, blood cholesterol, diabetes), and psychological stress at work did not change these results. However, participants in the highest third of negative affect had an increased incidence of coronary events (hazard ratio=1.32, 1.09 to 1.60), and this association remained unchanged after adjustment for multiple confounders.Conclusions Positive affect and affect balance did not seem to be predictive of future coronary heart disease in men and women who were free of diagnosed coronary heart disease at recruitment to the study. A weak positive association between negative affect and coronary heart disease was found and needs to be confirmed in further studies.  相似文献

In contrast to what it is observed during starvation, animals maintained on a protein-free isocaloric diet showed an increase in the rate of hepatic peptide chain elongation as determined by measuring the ribosomal transit time in vivo. The loss of body nitrogen per se is insufficient to generate the signal(s) which arrests hepatic peptide chain elongation. This observation suggests that it is an increase in gluconeogenic demand, and not the negative nitrogen balance, which is implicated in determining reciprocal changes in the rate of protein synthesis.The rate of protein synthesis, as expressed per mg of DNA, does not change in protein deprived animals, while the RNA to DNA ratio decreased. These data also agree with a higher ribosomal efficiency at the elongation step. The animals maintained on a protein-free diet have a decreased hepatic content of protein and an increased concentration of valine, indicating an increased proteolysis.The enhanced rate of polypeptide elongation observed in animals kept on a protein-free diet was accompanied by decreases in the state of aggregation of polyribosomes and in the ability of liver extracts to form eIF-2 catalyzed ternary complexes. These observations suggest that the activity of the hepatic initiation factor in vivo may not be rate limiting.The administration of alanine in vivo to animals maintained on a protein-free diet showed a preferential effect in reaggregating polyribosomes. This action was neither accompanied by detectable effects on the rate of eIF-2 catalyzed ternary complexes formation nor by significant changes in the rate of elongation. It is concluded that factors other than eIF-2 activity or the rate of polypeptide elongation must be controlling the hepatic polyribosomal state of aggregation.  相似文献

Effect of interaction of melatonin and caffeine on caffeine induced oxidative stress and sleep disorders was studied. Fifteen wistar rats were randomly assigned into three study groups. The animals in group 1 (the control) received a placebo of 10.0 ml distilled water via gastric intubation. The hosts in groups 2 and 3 were treated with 100 mg caffeine/ kg, or melatonin/ kg, respectively, in a total volume of 10.0 ml vehicle. The experiment lasted for 30 days. One day after the final exposure, the animals were euthanized by inhalation of overdose of chloroform. Blood was collected by cardiac puncture. Serum was obtained by centrifugation (6000 Xg, 30 mins), and used for serum total protein and serum blood urea nitrogen levels. The brain of each rat was also harvested and processed into whole homogenate, frozen in liquid nitrogen (N2), and maintained at -80oC until used for total brain cholesterol and tryptophan levels. The results showed that interaction of melatonin and caffeine enhanced protein synthesis; stimulated gonadotrophin release, and could be used as oral contraceptive for women, and may be beneficial in the treatment of impotence (androgen depression), leading to improved reproductive and sex life; stimulated tryptophan metabolism, which prevents vitamin B6 deficiency, anemia, negative nitrogen balance, tissue wasting and accumulation of xanthurenic acid, which promotes sleep; and could be beneficial in the treatment of hyper cholesterolemia, thereby preventing coronary heart disease, and post menopausal osteoporosis.  相似文献

Studies of the mechanism of lipid-induced regulation of the microsomal enzyme UDP-glucuronosyltransferase have been extended by examining the influence of charge within the polar region on the ability of lipids to activate delipidated pure enzyme. The effects of net negative charge, of charge separation in phosphocholine, and of the distribution of charge in the polar region of lipids were studied using the GT2p isoform isolated from pig liver. Prior experiments have shown that lipids with net negative charge inhibit the enzyme (Zakim, D., Cantor, M., and Eibl, H. (1988) J. Biol. Chem. 263, 5164-5169). The current experiments show that the extent of inhibition on a molar basis increases as the net negative charge increases from -1 to -2. The inhibitory effect of negatively charged lipids is on the functional state of the enzyme and is not due to electrostatic repulsion of negatively charged substrates of the enzyme. Although the inhibitory effect of net negative charge is removed when negative charge is balanced by a positive charge due to a quaternary nitrogen, neutrality of the polar region is not a sufficient condition for activation of the enzyme. In addition to a balance of charge between Pi and the quaternary nitrogen, the distance between the negative and positive charges and the orientation of the dipole created by them are critical for activation of GT2p. The negative and positive charges must be separated by the equivalent of three -CH2- groups for optimal activation by a lipid. Shortening this distance by one -CH2- unit leads to a lipid that is ineffective in activating the enzyme. Reversal of the orientation of the dipole in which the negative charge is on the polymethylene side of the lipid-water interface and the positive charge extends into water also produces a lipid that is not effective for activating GT2p. On the other hand, lipids with phosphoserine as the polar region, which has the "normal" P-N distance but carries a net negative charge, do not inhibit GT2p. This result again illustrates the importance of the dipole of phosphocholine for modulating the functional state of GT2p.  相似文献

It has been established that data of acid-base balance in cows' and newborn calves' blood closely correlate with physiological status of animals during their initial days of postnatal ontogenesis and depend on the peculiarities of metabolism in fetus and newborn organisms. Data on the acid-base balance in blood of newborn calves under diarrhea syndrome are discussed from the point of view of the influence of rehydration therapy and "Namacit" preparation on clinical status of animals.  相似文献
