Ammonium hydroxide treatment of 1,6:2,3-dianhydro-4-O-benzyl-β-D-mannopyranose, followed by acetylation, gave 2-acetamido-3-O-acetyl-1,6-anhydro-4-O-benzyl-2-deoxy-β-D-glucopyranose which was catalytically reduced to give 2-acetamido-3-O-acetyl-1,6-anhydro-2-deoxy-β-D-glucopyranose (6), the starting material for the synthesis of (1→4)-linked disaccharides bearing a 2-acetamido-2-deoxy-D-glucopyranose reducing residue. Selective benzylation of 2-acetamido-1,6-anhydro-2-deoxy-β-D-glucopyranose gave a mixture of the 3,4-di-O-benzyl derivative and the two mono-O-benzyl derivatives, the 4-O-benzyl being preponderant. The latter derivative was acetylated, to give a compound identical with that just described. For the purpose of comparison, 2-acetamido-4-O-acetyl-1,6-anhydro-2-deoxy-β-D-glucopyranose has been prepared by selective acetylation of 2-acetamido-1,6-anhydro-2-deoxy-β-D-glucopyranose.Condensation between 2,3,4,6-tetra-O-acetyl-α-D-glucopyranosyl bromide and 6 gave, after acetolysis of the anhydro ring, the peracetylated derivative (17) of 2-acetamido-2-deoxy-4-O-β-D-glucopyranosyl-α-D-glucopyranose. A condensation of 6 with 3,4,6-tri-O-acetyl-2-deoxy-2-diphenoxyphosphorylamino-α-D-glucopyranosyl bromide likewise gave, after catalytic hydrogenation, acetylation, and acetolysis, the peracylated derivative (21) of di-N-acetylchitobiose.  相似文献

Condensation of benzyl 2-acetamido-3,6-di-O-benzyl-2-deoxy-α-D-glucopyranoside with 2,3,4,6-tetra-O-benzyl-1-O-(N-methyl)acetimidoyl-β-D-glucopyranose gave benzyl 2-acetamido-3,6-di-O-benzyl-2-deoxy-4-O-(2,3,4,6-tetra-O-benzyl-α-D-glucopyranosyl)-α-D-glucopyranoside which was catalytically hydrogenolysed to crystalline 2-acetamido-2-deoxy-4-O-α-D-glucopyranosyl-α-D-glucopyranose (N-acetylmaltosamine). In an alternative route, the aforementioned imidate was condensed with 2-acetamido-3-O-acetyl-1,6-anhydro-2-deoxy-β-D-glucopyranose, and the resulting disaccharide was catalytically hydrogenolysed, acetylated, and acetolysed to give 2-acetamido-1,3,6-tri-O-acetyl-2-deoxy-4-O-(2,3,4,6-tetra-O-acetyl-α-D-glucopyranosyl)-α-D-glucopyranose Deacetylation gave N-acetylmaltosamine. The synthesis of 2-acetamido-2-deoxy-4-O-β-D-glucopyranosyl-α-D-glucopyranose involved condensation of benzyl 2-acetamido-3,6-di-O-benzyl-2-deoxy-α-D-glucopyranoside with 2,3,4,6-tetra-O-acetyl-α-D-glucopyranosyl bromide in the presence of mercuric bromide, followed by deacetylation and catalytic hydrogenolysis of the condensation product.  相似文献

Treatment of benzyl 2-acetamido-3-O-benzyl-2,6-dideoxy-4-O-(methylsulfonyl)-α-D-glucopyranoside (1) with sodium azide in hexamethylphosphoric triamide gave the 4-azido-α-D-galacto derivative (2), which was converted into benzyl 2,4-di-acetamido-3-O-benzyl-2,3,6-trideoxy-α-D-galactopyranoside (3) by hydrogenation and subsequent acetylation. Hydrogenolysis of 3 at atmospheric pressure afforded benzyl 2,4-diacetamido-2,4,6-tridcoxy-α-D-galactopyranoside (4), which was acetylated to give the 3-O-acetyl derivative (5). The n.m.r. spectrum of 5 was in agreement with the assigned structure and different from that of benzyl 2,4-di-acetamido-3-O-acetyl-α-D-glucopyranoside (9), which was prepared from the known benzyl 2,4-diacetamido-3-O-benzyl-2,4,6-trideoxy-α-D-glucopyranoside. Catalytic hydrogenolysis of 4 gave 2,4-diacetamido-2,4,6-trideoxy-D-galactose (6).  相似文献

2-Methyl-(2-acetamido-3,4,6-tri-O-benzyl-1,2-dideoxy-α-D-glucopyrano)-[2,1-d]-2-oxazoline,2-methyl-(2-acetamido-6-O-acetyl-3,4-di-O-benzyl-1,2-dideoxy-α-D-glucopyrano)-[2,1-d]-2-oxazoline,and 2-methyl-(2-acetamido-4-O-acetyl-3,6-di-O-benzyl-1,2-dideoxy-α-D-glucopyrano)-[2,1-d]-2-oxazoline were synthesized from the allyl 2-acetamido-3,4,6-tri-O-benzyl-2-deoxy-D-glucopyranosides, and from the 3,4-di-O-benzyl or 3,6-di-O-benzyl analogs, respectively, both the α and β anomer being used in each case. The preparation of allyl 2-acetamido-3,4,6-tri-O-benzyl- and 3,6-di-O-benzyl-2-deoxy-β-D-glucopyranoside is also described. Treatment of the tri-O-benzyl oxazoline with dibenzyl phosphate gave a pentabenzylglycosyl phosphate, from which all the benzyl groups were removed by catalytic hydrogenation, giving 2-acetamido-2-deoxy-α-D-glucopyranosyl phosphate. The corresponding β anomer was not detectable. Treatment of the 3,4-, or 3,6-, di-O-benzyl oxazoline with allyl 2-acetamido-3,4-di-O-benzyl-α-D-glucopyranoside readily gave disaccharide products from which the protecting groups were removed, to give the (1→6)-linked isomer of di-N-acetylchitobiose. Under both acidic and basic conditions, this isomer was less stable than the (1→4)-linked compound.Attempts to employ the 3,6-di-O-benzyl oxazoline for the formation of (1→4)-linked disaccharides, by treatment with either anomer of allyl 2-acetamido-3,6-di-O-benzyl-2-deoxy-D-glucopyranoside, were not very successful, presumably owing to hindrance by the bulky benzyl groups.  相似文献

Ethyl 6-O-benzyl-2,3-dideoxy-α-d-erythro-hex-2-enopyranoside (2) was converted, in three steps and in 73% overall yield, into ethyl 6-O-benzyl-2,3-dideoxy-3-C-(hydroxymethyl)-α-d-ribo-hex-2-enopyranoside. This transformation involved silylation of 2 with (bromomethyl)chlorodimethylsilane and application of the Nishiyama-Stork radical cyclisation, followed by Tamao oxidation of the sila cycle. Ethyl 6-O-benzyl-2,3-dideoxy-α-d-threo-hex-2-enopyranoside and benzyl 2,6-di-O-benzyl-α-l-threo-hex-4-enopyranoside were similarly transformed into, respectively, ethyl 6-O-benzyl-2,3-dideoxy-3-C-(hydroxymethyl)-α-d-lyxo-hex-2-enopyranoside (50%), and benzyl 2,6-di-O-benzyl-4-deoxy-4-C-(hydroxymethyl)-β-d-galactopyranoside (71%).  相似文献

An approach to stereoselective synthesis of α- or β-3-C-glycosylated l- or d-1,2-glucals starting from the corresponding α- or β-glycopyranosylethanals is described. The key step of the approach is the stereoselective cycloaddition of chiral vinyl ethers derived from both enantiomers of mandelic acid. The preparation of 1,5-anhydro-4,6-di-O-benzyl-2,3-dideoxy-3-C-[(2,3,4,6-tetra-O-benzyl-β-d-glucopyranosyl)methyl]-l-arabino-hex-1-enitol, 1,5-anhydro-4,6-di-O-benzyl-2,3-dideoxy-3-C-[(2,3,4,6-tetra-O-benzyl-β-d-glucopyranosyl)methyl]-d-arabino-hex-1-enitol, and 1,5-anhydro-4,6-di-O-benzyl-2,3-dideoxy-3-C-[(2,3,4-tri-O-benzyl-α-l-fucopyranosyl)methyl]-d-arabino-hex-1-enitol serves as an example of this approach.  相似文献

Silver trifluoromethanesulfonate-promoted condensation of 3,4,6-tri-O-acetyl-2-deoxy-phthalimido-β-d-glucopyranosyl bromide with benzyl 3,6-di-O-benzyl-α-d-mannopyranoside and benzyl 3,4-di-O-benzyl-α-d-mannopyranoside gave the protected 2,4- and 2,6-linked trisaccharides in yields of 54 and 32%, respectively. After exchanging the 2-deoxy-2-phthalimido groups for 2-acetamido-2-deoxy groups and de-blocking, the trisaccharides 2,4-di-O-(2-acetamido-2-deoxy-β-d-glucopyranosyl)-d-mannose and 2,6-di-O-(2-acetamido-2-deoxy-β-d-glucopyranosyl)-d-mannose were obtained. Similar condensation of 3,6-di-O-acetyl-2-deoxy-2-phthalimido-4-O-(2,3,4,6-tetra-O-acetyl-β-d-galactopyranosyl)-β-d-glucopyranosyl bromide with benzyl 3,4-di-O-benzyl-α-d-mannopyranoside gave a pentasaccharide derivative in 52% yield. After transformations analogous to those applied to the trisaccharides, 2,6-di-O-[β-d-galactopyranosyl-(1→4)-O-(2-acetamido-2-deoxy-β-d-glucopyranosyl)]-d-mannose was obtained.  相似文献

Acetylation of benzyl 6-deoxy-3,4O-isopropylidene-β-L-galactopyranoside gave benzyl 2-O-acetyl-6-deoxy-3,4-O-isopropylidene-β-L-galactopyranoside (1). Removal of the isopropylidene group afforded benzyl 2-O-acetyl-6-deoxy-β-L-galactopyranoside (2), which was converted into benzyl 2-O-acetyl-6-deoxy-3,4-di-O-(methyl-sulfonyl)-β-L-galactopyranoside (3). Benzyl 2,3-anhydro-6-deoxy-4-O-(methyl-sulfonyl)-β-L-gulopyranoside (4) was obtained from 3 by treatment with alkali. Reaction of 4 with sodium azide in N,N-dimethylformamide gave a mixture of two isomeric benzyl 2,4-diazido-2,4,6-trideoxy hexoses, the syrupy diazido derivative 5 and the crystalline benzyl 2,4-diazido-2,4,6-trideoxy-β-L-idopyranoside (6). Acetylation of 6 afforded a compound whose n.m.r. spectrum was completely first order and in agreement with the structure of benzyl 3-O-acetyl-2,4-diazido-2,4,6-trideoxy-β-L-idopyranoside (7). Lithium aluminium hydride reduction of 5, followed by acetylation, afforded a crystalline product (8), shown by n.m.r. spectroscopy to be benzyl 2,4-diacetamido-3-O-acetyl-2,4,6-trideoxy-β-L-altropyranoside. Similar treatment of the diazido derivative 6 afforded benzyl 2,4-diacetamido-3-O-acetyl-2,4,6-trideoxy-β-L-idopyranoside (9). Compounds 8 and 9 could also be obtained from 4 by treatment of the crude diazido mixture with lithium aluminium hydride, with subsequent N-acetylation. The syrupy benzyl 2,4-diacetamido-2,4,6-trideoxy-β-L-altropyranoside (10) and the crystalline benzyl 2,4-diacetamido-2,4,6-trideoxy-β-L-idopyranoside (11) thus obtained were then O-acetylated to give 8 and 9 respectively. Benzyl 2,4-diacetamido-2,4,6-trideoxy-β-L-talopyranoside (15) was obtained from 11 by treatment with methanesulfonyl chloride and subsequent solvolysis. Compound 15 was O-acetylated to yield benzyl 2,4-diacetamido-3-O-acetyl-2,4,6-trideoxy-β-L-talopyranoside (16). the n.m.r. spectrum of which was in full agreement with the assigned structure. The mass spectra of compounds 8–11, 15, and 16 were also in agreement with their proposed structures. Removal of the benzyl groups from 10, 11 and 15 afforded the corresponding 2,4-diacetamido-2,4,6-trideoxyhexoses 12, 13, and 17, having the L-altro, L-ido, and L-talo configurations, respectively.  相似文献

Three different approaches starting from 1,2-O-isopropylidene-α-d-glucofuranose were tested for the synthesis of daunosamine hydrochloride (24), the sugar constituent of the antitumor antibiotics daunomycin and adriamycin. The third route, affording 24 in ~5% overall yield in 11 steps, constitutes a useful, preparative synthesis, 3,5,6-Tri-O-benzoyl-1,2-O-isopropylidene-α-d-glucofuranose was converted via methyl 2,3-anhydro-β-d-mannofuranoside into methyl 2,3:5,6-dianhydro-α-l-gulofuranoside, the terminal oxirane ring of which was split selectively on reduction with borohydride, to afford methyl 2,3-anhydro-6-deoxy-α-l-gulofuranoside (31). Compound 31 was converted into methyl 2,3-anhydro-5-O-benzyl-6-deoxy-α-l-gulofuranoside, which was selectively reduced at C-2 on treatment with lithium aluminum hydride, affording methyl 5-O-benzyl-2,6-dideoxy-α-l-xylo-hexofuranoside. Subsequent mesylation, and replacement of the mesoloxy group by azide, with inversion, afforded methyl 3-azido-5-O-benzyl-2,6-dideoxy-α-l-lyxo-hexofuranoside, which could be converted into either 24 or methyl 3-acetamido-5-O-acetyl-2,3,6-trideoxy-α-l-lyxo-hexofuranoside, which can be used as a starting material for the synthesis of daunomycin analogs.  相似文献

2-O-Benzoyl-3,6-di-O-benzyl-4-O-(chloroacetyl)-, 4-O-acetyl-2-O-benzoyl-3,6-di-O-benzyl-, and 2-O-benzoyl-3,4,6-tri-O-benzyl-α-d-galactopyranosyl chloride were converted into the corresponding 2,2,2-trifluoroethanesulfonates, and these were treated with allyl 2-O-benzoyl-3,6-di-O-benzyl-α-d-galactopyranoside, to give allyl 2-O-benzoyl-4-O-[2-O-benzoyl-3,6-di-O-benzyl-4-O-(chloroacetyl)-β-d-galactopyranosyl]-3,6-di-O-benzyl- α-d-galactopyranoside (26; 41% yield), allyl 4-O-(4-O-acetyl-2-O-benzoyl-3,6-di-O-benzyl-β-d-galactopyranosyl)-2-O-benzoyl-3,6-di-O-benzyl- α-d-galactopyranoside (27; 62% yield), and allyl 2-O-benzoyl-4-O-(2-O-benzoyl-3,4,6-tri-O-benzyl-β-d-galactopyranosyl)-3,6-di-O-benzyl-α-d-galactopyranoside (28; 65% yield). All disaccharides were free from their α anomers. Disaccharides 26 and 27 were found to be base-sensitive, and were de-esterified by KCN in aqueous ethanol, and debenzylated with H2-Pd. Attempts to produce (1→4)-β-d-galactopyranosides from the coupling of a number of fully esterified d-galactopyranosyl sulfonates to allyl 2,3,6-tri-O-benzoyl-α-d-galactopyranoside were unsuccessful.  相似文献

Syntheses are described of di- and tri-saccharides required for studies of the inhibition of adhesion by means of fimbriae of pyelonephritogenic E. coli bacteria to epithelium cells in the urinary tract containing suitable receptor sites. The disaccharides are the p-nitrophenyl and methyl glycosides of 4-O-α-d-galactopyranosyl- β-d-galactopyranose, and the trisaccharides are the p-nitrophenyl and methyl glycosides of 4-O-(4-O-α-d-galactopyranosyl-β-d-galactopyranosyl)-β-d-glucopyranose. The key aglycons were the 1,2,3,6-tetrabenzoate of α-d-galactose and the 1,2,3,6,- 2′,3′,6′-heptabenzoate of α-lactose. Glycosidations were performed with 2,3,4,6- tetra-O-benzyl-α-d-galactopyranosyl chloride as glycosylating agent and silver triflate as promoter.  相似文献

Three new saponins, melongosides N, O and P, have been isolated from the methanolic extract of seeds of Solanum melongena and their structures elucidated. Melongoside N is 3-O-[β-D-glucopyranosy l-(1 → 2)-β-D-glucopyranosyl]-26-O-(β-D-glucopyranosyl)-(25R)-5α-furostan-3β,22 α,26-triol, whereas melongoside O is 3-O-[β-D-glucopyranosyl-(1 → 2)β-D-glucopyranosyl]- 26-O-(β-D-glucopyranosyl)-(25R)-furost-5-en-3β,22α,26-triol and melongoside P is 3-O- [β-D-glucopyranosyl-(1 → 2)]-[α-L-rhamnopyranosyl-(1 → 3)]-β-D-glucopyranosyl)-26-O- (β-D-glucopyranosyl)-(25 R)-5α-furostan-3β,22α,26-triol.  相似文献

The following ethers, of potential value for the synthesis of α-D-galactopyranosides, were prepared: 2-O-benzyl-D-galactose, 2,6-di-O-benzyl-D-galactose, and 2,3-di-O-benzyl-D-galactose. Isopropylidenation of methyl α-D-galactopyranoside in the presence of phosphorus pentaoxide gave its 3,4-, and 4,6-O-isopropylidene derivatives. Treatment of the 3,4-acetal with trityl chloride in pyridine produced the 6-trityl ether, which was benzylated with benzyl chloride and sodium hydride in N,N-dimethylformamide to yield the 2-benzyl ether. Acid hydrolysis of this product gave 2-O-benzyl-D-galactose. Benzylation of methyl 3,4-O-isopropylidene-α-D-galactopyranoside, followed by hydrolysis, gave 2,6-di-O-benzyl-D-galactose. Similarly, 2,3-di-O-benzyl-D-galactose was obtained by acid hydrolysis of methyl 2,3-di-O-benzyl-4,6-O-isopropylidene-α-D-galactopyranoside and of methyl 2,3-di-O-benzyl-4,6-O-benzylidene-β-D-galactopyranoside.  相似文献

A diastereoisomer of Kanamycin C has been synthesized by a modified Koenigs—Knorr reaction of 3,4,6-tri-O-acetyl-2-(2,4-dinitroanilino)-2-deoxy-α-D-glucopyranosyl bromide with 4-O-(3-acetamido-2,4,6-tri-O-benzyl-3-deoxy-α-D-glucopyranosyl)-N,N′-di[(benzyloxy)carbonyl]-2-deoxystreptamine. Several Kanamycin analogues were synthesized by a similar condensation reaction. Each of the condensed products was isolated as its crystalline tetra-N-acetyl derivative and was proved by n.m.r. spectroscopy in D2O to have the α-configuration.  相似文献

Conformational equilibria of derivatives of 2,4-diamino-2,4-dideoxy-α-d-idopyranose are investigated. In the case of methyl 2,4-diacetamido-3,6-di-O-acetyl-2,4-dideoxy-α-d-idopyranoside, the proportion of the 1C4 conformation is between 7 and 44%, depending upon the solvent, and increases with increasing polarity of the solvent. Methyl 2,4-di(N-acetyl-N-acylamino)-3,6-di-O-acyl-2,4-dideoxy-α-d-idopyranosides show such strong steric 1,3-diaxial interaction that the proportion of the 1C4 conformation is 93%. Methyl 2,4-diamino-2,4-dideoxy-α-d-idopyranoside and its bis(ammonium) salt show strong 1,3-diaxial interactions of polar nature, resulting in an increase of the 1C4 conformation up to 74% and 84%, respectively.  相似文献

Quaternary ammonium and triphenylphosphonium salts of 2,3,4-tri-O-benzyl-6-O-(N-phenylcarbamoyl)-D-glucopyranosyl bromide were readily prepared by reaction with tertiary amines and triphenylphosphine under anhydrous conditions. Methanolysis of these salts was studied to determine the conditions of solvent and temperature that would produce the highest yields of α-D-glucosides. The quaternary ammonium salts gave the highest yields with solvents of low dielectric constant and room temperature. The phosphonium salts gave moderate yields with diethyl ether at 50°. The synthesis of methyl 2,3,4-tri-O-benzyl-6-O-(2,3,4,6-tetra-O-benzyl-α-D-glucopyranosyl)-α-D-glucopyranoside by treatment of the quaternary ammonium salt of 2,3,4,6-tetra-O-benzyl-α-D-glucopyranosyl bromide with methyl 2,3,4-tri-O-benzyl-α-D-glucopyranoside was studied as a model for the synthesis of oligosaccharides. The anomeric composition of the disaccharide product could be easily determined from the optical rotation since the specific rotations of both the final product and of the gentiobioside analog are known. Under the best conditions, the yield of disaccharide was low (50%) and the reactions were not completely stereoselective.  相似文献

The reaction of phenyl 2-acetamido-2-deoxy-4,6- O-(p-methoxybenzylidene)-β-d-glucopyranoside with 2,3,4-tri-O-benzyl-α-l-fucopyranosyl bromide under halide ion-catalyzed conditions proceeded readily, to give phenyl 2-acetamido-2-deoxy-4,6-O-(p-methoxybenzylidene)-3-O-(2,3,4-tri-O-benzyl-α-l-fucopyranosyl)-β-d-glucopyranoside (8). Mild treatment of 8 with acid, followed by hydrogenolysis, provided the disaccharide phenyl 2-acetamido-2-deoxy-3-O-α-l-fucopyranosyl-β-d-glucopyranoside. Starting from 6-(trifluoroacetamido)hexyl 2-acetamido-3,4,6-tri-O-acetyl-2-deoxy-β-d-glucopyranoside, the synthesis of 6-(trifluoroacetamido)hexyl 2-acetamido-2-deoxy-3-O-β-l-fucopyranosyl-β-d-glucopyranoside has been accomplished by a similar reaction-sequence. On acetolysis, methyl 2-acetamido-2-deoxy-3-O-α-l-fucopyranosyl-α-d-glucopyranoside gave 2-methyl-[4,6-di-O-acetyl-1,2-dideoxy-3-O-(2,3,4-tri-O-acetyl-α-l-fucopyranosyl)-α-d-glucopyrano]-[2, 1-d]-2-oxazoline as the major product.  相似文献

Methyl 2-acetamido-3-O-allyl-2-deoxy-4-O-methyl-α-D-glucopyranoside, methyl 2-acetamido-2-deoxy-4-O-methyl-α-D-glucopyranoside, and methyl 2-acetamido-3,4-di-O-allyl-2-deoxy-α-D-glucopyranoside, prepared from methyl 2-acetamido-2-deoxy-α-D-glucopyranoside, were coupled with 2,3,4,6-tetra-O-acetyl-α-D-glucopyranosyl phosphate (13), to give the phosphoric esters methyl 2-acetamido-3-O-allyl-2-deoxy-4-O-methyl-α-D-glucopyranoside 6-(2,3,4,6-tetra-O-acetyl-α-D-glucopyranosyl phosphate) (16), methyl 2-acetamido-2-deoxy-4-O-methyl-α-D-glucopyranoside 6-(2,3,4,6-tetra-O-acetyl-α-D-glucopyranosyl phosphate) (23), and methyl 2-acetamido-3,4-di-O-allyl-2-deoxy-α-D-glucopyranoside 6-(2,3,4,6-tetra-O-acetyl-α-D-glucopyranosyl phosphate) (17). Compound 13 was prepared from penta-O-acetyl-β-D-glucopyranose by the phosphoric acid procedure, or by acetylation of α-D-glucopyranosyl phosphate. Removal of the allyl groups from 16 and 17 gave 23 and methyl 2-acetamido-2-deoxy-α-D-glucopyranoside 6-(2,3,4,6-tetra-O-acetyl-α-D-glucopyranosyl phosphate) (19), respectively. O-Deacetylation of 23 gave methyl 2-acetamido-2-deoxy-4-O-methyl-α-D-glucopyranoside 6-(α-D-glucopyranosyl phosphate) (26) and O-deacetylation of 19 gave methyl 2-acetamido-2-deoxy-α-D-glucopyranoside 6-(α-D-glucopyranosyl phosphate) (24). Propyl 2-acetamido-2-deoxy-α-D-glucopyranoside 6-(α-D-glucopyranosyl phosphate) (25) was prepared by coupling 13 with allyl 2-acetamido-3,4-di-O-benzyl-2-deoxy-α-D-glucopyranoside, followed by catalytic hydrogenation of the product to give the propyl glycoside, which was then O-deacetylated. Compounds 24, 25, and 26 are being employed in structural studies of the Micrococcus lysodeikticus cell-wall.  相似文献
