The binding of plasmin to Streptococcus uberis strain 0140 J was optimal in the pH range 5·0–5·5. Plasmin binding decreased exponentially with increasing NaCl concentration (0–0·8 mol l−1), reaching a minimum at NaCl concentrations exceeding 0·55 mol l−1. Neither K+, Mg2+ nor the metal chelator EDTA had any effect on the interaction. Plasmin binding was prevented, in a concentration-dependent manner, by the amino acids lysine, arginine and ε-aminocaproic acid. Bound plasmin was also eluted from the bacterial cell using the same amino acids. Bound plasmin was lost from the bacterium in a time- and temperature-dependent fashion, the rate of plasmin loss increased with increasing temperature over the range 4–55 °C, and the elution of plasmin from live and heat-killed bacteria was similar. Cell-bound plasmin was only partially inhibited by the physiological inhibitor α2-antiplasmin whereas the serine protease inhibitor aprotinin, and the active site titrant p -nitrophenyl- p -guanidiniobenzoate, inhibited the activity of the cell-bound plasmin by more than 95%.  相似文献

The interaction of temperature and fish size on growth of juvenile halibut   总被引:3,自引:0,他引:3  
Growth rate of individually tagged juvenile halibut was influenced significantly by the interaction of temperature and fish size. The results suggest an optimum temperature for growth of juvenile halibut in the size range 5–70 g between 12 and 15° C. Overall growth rate was highest at 13° C (1·62% day −1). At c. 5 g at the beginning of the experiment, fish at 16° C had the highest growth rate (3·2% day −1), but reduced this rate as they grew bigger. At 9 and 11°p C, growth rates were equal or only slightly lower during the later stages of the experiment, while the fish at 6° C showed significantly lower overall growth rate (0·87% day−1). Optimal temperature for growth decreased rapidly with increasing size, indicating an ontogenetic reduction in optimum temperature for growth. Moreover, a more flattened parabolic regression curve between growth and temperature as size increased indicated reduced temperature dependence with size. Although individual growth rates varied significantly at all times within the experimental temperatures, significant size rank correlations were maintained during the experiment. This indicated an early establishment of a stable size hierarchy within the fish groups. Haematocrit was highest at the highest temperature while Na+/K+-ATPase activity was inversely related to temperature. There was no difference in plasma Na+, Cl and K+ concentrations among the temperature groups.  相似文献

The effect of chlorine on β- D- galactosidase activity of sewage bacteria and Escherichia coli was studied. β- D- galactosidase activity of sewage was more resistant to chlorine than faecal coliform cultivability. At low initial dosage (0·05 mg Cl2 l−1) neither cultivability (colony-forming units (cfu)), nor enzyme activity of E. coli suspensions were severely impaired. When initial chlorine concentration was increased to 0·1 mg Cl2 l−1, the cfu number decreased whereas enzyme activity remained high, i.e. the enzyme activity calculated cfu−1 increased. At higher chlorine doses both cfu and enzyme activity were reduced, but non-cultivable cells retained assayable activity after chlorination. Mean values of the enzyme activity calculated cfu−1 decreased when the chlorine dosage was increased from 0·1 to 0·5 mg Cl2 l−1, but were not significantly different ( P > 0·05) for dosages of 0·2–0·7 mg Cl2 l−1. After chlorination, β- D- galactosidase activity of E. coli was less reduced than cfu and direct viable count numbers, but more reduced than 5-cyano-2-3, ditolyl tetrazolium chloride and total cell counts, and the enzyme activity represented an alternative activity parameter of chlorinated samples.  相似文献

A method for measuring oxygen consumption in isolated perfused gills   总被引:1,自引:0,他引:1  
A method is described for measuring respiration in isolated perfused flounder gills experiencing pressures and flows similar to those seen in vivo . Mean oxygen consumption of 13 preparations bathed and perfused in identical saline was 5·00 ± 0·75 (s.e.) μ mol h−1 g wet−1, whilst that of five preparations perfused with saline but bathed in sea water (32 mg l−1) was 12·06±2·39 (s.e.) μmol h−1 g wet−1. The oxygen consumption of the seawater bathed gills was significantly higher (P<0·05) than that in saline bathed gills. These results provide direct evidence both of the high metabolic activity of the gill under normal perfusion conditions and of the increased energy expenditure of the giil in hyperosmotic, compared to isosmotic, environments.  相似文献

An improved method for purification of pectate lyases (PLI and PLII) from culture fluids of Pseudomonas fluorescens CY091 and Ps. viridiflava PJ-08-6 by using a phosphocellulose cation exchanger was described. Analysis of purified PLI and PLII by sodium dodecyl sulphate-polyacrylamide and isoelectric focusing gel electrophoresis revealed that both enzymes had been purified to near homogeneity. Optimal Ca2+ concentration required for PLI and PLII activity was determined to be 0·5 mmol l−1. The Ca2+ requirement could not be replaced by other metal cations such as Mg2+, Cu2+, Zn2+, Fe3+ and Co2+. Optimal pH for activity was determined to be between 8·5 and 9·0. The K m values for sodium polygalacturonate were 1·28 and 1·11 mg ml−1 for PLI and PLII, respectively. Both PLI and PLII were stable at low temperatures (25°C or below) for at least 1 month. However, at 37°C, the activity decreased 50% in 36 h. Optimal temperatures for activity were estimated to be 46° and 52°C for PLI and PLII, respectively. Thermal stability of both enzymes at elevated temperatures (48°C or higher) increased when CaCl2 or a positively charged molecule such as polylysine was present, but decreased when polygalacturonate or a negatively charged molecule such as heparin was present. PLI and PLII exhibit differential degrees of sensitivity to group-specific inhibitors, including iodoacetic acid and diethylpyrocarbonate. This result suggests that both sulphydryl and imidazole groups are important for the catalytic function of PLI and PLII.  相似文献

Juvenile yellowtail Seriola quinqueradiata (initial body mass of 22 g) were fed either a commercial diet (control, diet 1) or diets supplemented with histidine (diet 2), histidine+β-alanine (diet 3), or histidine+β-alanine+thyroxine (diet 4), for 6 weeks. The dietary treatment did not affect the final body mass. Free histidine levels of white muscle in the fish fed the diets supplemented with histidine (diets 2-4) were significantly higher (>62 mmol kg−1 of wet tissue) than that of control group (42 mmol kg−1 of wet tissue). Dietary supplementation of β-alanine (diet 3) or β-alanine+thyroxine (diet 4) failed to increase muscle anserine (β-alanyl-π-L.-histidine) level. Muscle buffering capacity of the range from pH 6·0 to 7·5 of the fish fed the diets 2-4 (41·6-42·7 mmol NaOH pH−1 kg muscle−1) reflected the increase of muscle histidine level, having slightly but significantly intensified compared to control fish (36·6 mmol NaOH pH−1 kg muscle−1). Most of the free amino acids other than histidine were significantly lower in the fish fed the diets 2-4 than in control fish. Thus, crystalline histidine supplemented to diets appears to be deposited in muscular tissue, and consequently enhance muscle buffering capacity in this species.  相似文献

1. Termite nitrogenase activity was highest in autumn and spring (≈ 3 μg N2 fixed termite fresh mass (g)–1 day–1) and lowest in winter and summer (≈ 0·8 μg N2 fixed termite fresh mass (g)–1 day–1).
2. The nitrogenase activity of worker termites was significantly higher than all other castes (1·58 ± 0·27 μg N2 fixed termite fresh mass (g)–1 day–1).
3. Worker termites constituted the largest proportion of all the castes throughout the study period (≈ 90%).
4. The localized input of fixed nitrogen by termites may reach 15·3 mg N log–1 day–1 and 5·6 g N log–1 year–1.  相似文献

Addition of small amounts of Fe2+, Zn2+, Cu2+ and thiamine-HCl to the culture medium was required for promoting the galacto-oligosaccharide (Gal-OS)-producing activity of Sterigmatomyces elviae CBS8119, when the concentration of yeast extract in the medium was lowered to 0·1 g l−1. Galacto-oligosaccharide production using a recycling cell culture was performed in a medium containing 360 mg ml−1 of lactose supplemented with optimal concentrations of Fe2+ (1·5 mg l−1 of FeSO4.7H2O), Zn2+ (15 mg l−1 of ZnSO4.7H2O), Cu2+ (0·5 mg l−1 of CuSO4.5H2O) and thiamine-HCl (1 mg l−1 ) . Galacto-oligosaccharide production was maintained at high levels during six cycles of production, with the amount of Gal-OS produced in each cycle being more than 216 mg ml−1 (weight yield of more than 60%).  相似文献

The protective effects of dietary Ca2+ supplementation against Cd accumulation in rainbow trout Oncorhynchus mykiss fed with Cd-contaminated food were evaluated in relation to chronic changes in intestinal absorption rates. The changes were measured ' in vitro '. The control diet contained c. 20 mg Ca2+ g−1 food and 0·25 μg Cd g−1 food; the experimental diets were supplemented with CaCO3 and Cd(NO3)2·4H2O to levels of 50 mg Ca2+ g−1 food and 300 μg Cd g−1 food, alone and in combination. The Ca2+ and Cd absorption rates were measured using radiotracers (45Ca, 109Cd) at total Ca2+ and Cd concentrations of 3·0 and 0·12 mmol l−1, respectively in the intestinal saline. Chronically elevated dietary Cd caused a significant increase in Cd absorption rate by up to 10-fold at 30 days in the mid-intestine. The high Ca2+ diet prevented this up-regulation of Cd transport rate. Conversely, intestinal Ca2+ absorption was significantly increased by two- to five-fold by the Ca2+-supplemented diet at 30 days in both the mid- and posterior intestine, and this effect was eliminated when Cd was simultaneously elevated in the diet. Ca2+ and Cd probably interact at common pathways and transport mechanisms in the intestine, though independent pathways may also exist.  相似文献

The major β-1,4-endoglucanase (EG) of the thermophilic actinomycete, Thermomonospora curvata , contributed over 80% of the total EG activity recovered from cell-free culture fluid after growth on cellulose. The enzyme was purified to electrophoretic homogeneity by ammonium sulphate precipitation, ion-exchange chromatography and size exclusion HPLC. This monomeric enzyme had a specific activity of 750 IU mg−1 when assayed with 2.5% (w/v) carboxymethyl cellulose (CMC) at 70°C, pH 6.0. Highest activity was observed on CMC with a degree of polymerization of 3200. The EG was stable for 48 h at 60°C, pH 6.0 and had a half-life of 30 min at 80°C; temperature and pH optima were 70–73°C and 6.0–6.5, respectively. The mol. wt was 100000 and the pI was 4.0. The K m and V max values were 7.33 mg ml−1 and 833 μmol min−1, respectively. EG activity was inhibited by Fe2 +, Hg2 +, Ag+ and Pb2 +, and enhanced by dithiothreitol and Zn2 +. The first 12 amino acid residues at the N -terminus were: Asp-Glu-Val-Asp-Glu-Ile-Arg-Asn-Gly-Asp-Phe-Ser. Glutamic and aspartic acid constituted 24% of the total amino acid composition; no amino sugar was found.  相似文献

Long-term ammonia exposure of turbot: effects on plasma parameters   总被引:2,自引:0,他引:2  
Turbot juveniles were exposed to four ammonia concentrations [0·17 (L), 0·34 (M), 0·73 (MH) and 0·88 (H) mg l−1 NH3-N] for different exposure durations (28 days minimum to 84 days). Their physiological status and growth performances were compared to a control group [0·004 (C) mg l−1 NH3-N]. No growth was observed in the H group, and by day 57, mass increase in the MH group was only 15% of that in group C. During the first month growth in the L group was similar to that in control group while it was lower (33%) in the M group; afterwards the L and M groups had a similar growth (half that of controls). Accumulation of total ammonia nitrogen (TA-N) in plasma was dependent on ambient ammonia concentrations. Plasma urea levels in ammonia-exposed fish were lower, similar or greater than in controls (depending on ammonia concentration or exposure duration). Osmolarity, Cl and Na+ plasma concentrations were stable in the L and M groups. The increases in Na+, Cl, K+ and total Ca concentrations observed by the end of the experiment in the H and MH groups suggest that fish failed to adapt. There was an initial rise in plasma cortisol in all ammonia-exposed groups followed by a return to basal level (1·7–4 ng ml−1) in the L and M groups. In group MH, plasma cortisol peaked at 42 ng ml−1 by day 14, and after a decline at c . 1 month (14 ng ml−1), it rose again.  相似文献

Quantitative analyses of the utilization of amino acids by Lactococcus lactis subsp. cremoris FD1 in yeast extract medium (YE) and in casein peptone medium (CP) have been performed. Both free and peptide-bound amino acids were measured. In the CP most amino acids are peptide-bound and some amino acids are virtually only present in peptides. Thirty-six per cent of all peptide bonds in CP are hydrolysed during fermentation (6·3 mmol peptide bonds per gram biomass formed) and there is a transition of the growth rate related ATP consumption Y xATP (mmol ATP g biomass-1) from 25 mmol g-1 to 71 mmol g-1 coincident with a decrease of the peptide consumption. In YE most of the amino acids are on the free form and only 26% of the peptide bonds are hydrolysed during fermentation (1·5 mmol peptide bonds per gram biomass formed). A constant Y xATP= 38 mmol g-1 prevails throughout the fermentation in YE.  相似文献

Since bioenergetics models for 0+ fish have seldom been validated by field consumption estimates, field-based and indirectly estimated daily food rations were compared in larval perch Perca fluviatilis and zander Stizostedion lucioperca. Field-based estimates were calculated with linear and exponential evacuation rates based on gut fullness data during a 24-h cycle, with hourly field samplings instead of the normally recommended 3-h intervals. Indirect calculations used bioenergetics modelling with variable activity multipliers ( A ). Field-based estimates of daily rations ranged between 0·21 and 0·27 g g−1 day−1 in perch (mean L T 13·1 mm) and 0·31–0·40 g g−1 day−1 in zander (mean L T 10·6 mm). The higher values were calculated by using the exponential model. Daily rations calculated by bioenergetics modelling with A = 1 were only slightly higher than direct estimates in both species. However, if A values >1 were used, calculated daily rations were substantially higher than direct estimates. Estimates of daily ration based only on every third value ranged between 41 and 72% compared with 1-h intervals, mainly because of lower estimates of evacuation rate.  相似文献

The sub-chronic (28–56 days) effects of exposure to low concentrations of cadmium (Cd; 0·05, 0·25, 0·50 and 2·50 μg l−1) shortly following fertilization on embryos, larvae and juvenile rainbow trout Oncorhynchus mykiss were examined. Premature hatching occurred at lower concentrations (0·05 and 0·25 μg l−1 Cd), however, delayed hatching was seen in the 2·50 μg l−1 Cd group, with >90% of hatching occurring on the last day of the hatching period. Larval growth was negatively affected by Cd exposure in a concentration-dependent manner. Larvae exposed to 2·50 μg l−1 Cd were 13·9 ± 0·8% shorter in total length ( L T) and weighed 22·4 ± 3·5% (mean ± s . e .) less than controls at the end of the exposure period. Plasma sex steroid concentrations (oestradiol in juvenile females and 11-ketotestosterone in juvenile males) were elevated (four- to 10-fold over controls) in exposed fish in both males and females, following 28 days of exposure to 0·05, 0·25 and 0·50 μg l−1 Cd, respectively. These results suggest that environmentally realistic concentrations (in the μg l−1 range) of Cd can affect the development of O. mykiss impacting embryos, larvae and juvenile fish.  相似文献

Abstract— Myelin, synaptosomal and mitochondrial fractions obtained from homogenates of whole mouse brain contain K+ which can exchange with 42K+ at 2º in 0·32 m -sucrose. The content and rates of exchange of K+ were greater at pH 8·2 than at 6·1. In the synaptosomal preparations, the rates of exchange and content of 42K+ and K+ declined progressively with decreasing pH.
Of the total synaptosomal K+, 95 per cent could exchange with external 42K+. At pH 7·5, 20 per cent of the K+ and 78 per cent of the Na+ appeared to reside in osmotically insensitive pools. Synaptosomal K+ at 2º was slowly displaced by NaCl (0·18 m ) and the rate of exchange between 42K+ and K+ was retarded. KCI (0·18 m ) did not readily displace endogenous Na+. Synaptosomal K+ exchanged with exogenous K+ more rapidly than with exogenous Na+.
These observations have been discussed in terms of possible roles for ion exchange as the principal means by which K+ traverses the plasma membrane at 2º.  相似文献

Physiological impact of sea lice on swimming performance of Atlantic salmon   总被引:6,自引:0,他引:6  
Atlantic salmon Salmo salar were infected with two levels of sea lice Lepeophtheirus salmonis (0·13 ± 0·02 and 0·02 ± 0·00 sea lice g−1). Once sea lice became adults, the ventral aorta of each fish was fitted with a Doppler cuff to measure cardiac output ( ̇ ), heart rate ( f H) and stroke volume ( V S) during swimming. Critical swimming speeds ( U crit) of fish with higher sea lice numbers [2·1 ± 0·1 BL (body lengths) s−1] were significantly lower ( P  < 0·05) than fish with lower numbers (2·4 ± 0·1 BL s−1) and controls (sham infected, 2·6 ± 0·1 BL s−1). After swimming, chloride levels in fish with higher sea lice numbers (184·4 ± 11·3 mmol l−1) increased significantly (54%) from levels at rest and were significantly higher than fish with fewer lice (142·0 ± 3·7 mmol l−1) or control fish (159·5 ± 3·5 mmol l−1). The f H of fish with more lice was 9% slower than the other two groups at U crit. This decrease resulted in ̇ not increasing from resting levels. Sublethal infection by sea lice compromised the overall fitness of Atlantic salmon. The level of sea lice infection used in the present study was lower than has previously been reported to be detrimental to wild Atlantic salmon.  相似文献

Abstract— Slices of cerebral cortex were incubated in medium containing 0·75 or 2·8 mM 45CaCl2, in the presence or absence of 0·01–0·1 m m -ouabain. Ouabain induced accumulation of calcium by slices to a maximum of 4 μmoles/g of tissue/hr (0·75 m m -CaCl2 in the medium) and to 8 μmoles/g of tissue/hr (2·8 m m -CaCl2 in the medium). Accumulation of Ca2+ occurred more slowly than loss of K+ from the slices and more closely resembled the pattern of Na+ uptake.
Mitochondrial fractions isolated from ouabain-treated slices contained significantly more calcium than controls. Inclusion of EDTA in the homogenization medium resulted in decreased amounts of particulate-bound calcium.
The effect of ouabain on accumulation of calcium is discussed with regard to possible relationships to processes of active and passive transport.  相似文献

Backcalculated lengths at the end of the first growth season in wild Atlantic salmon Salmo salar differed significantly between parr smolting at age 1, 2 and 3 years over a period of 11 years (i.e. 1983–1993). Mean body lengths of the respective age groups at the end of the first growth period were 11·1, 6·2 and 4·7 cm, respectively. The mean percentage distribution of fish smolting at age 1, 2 and 3 was 14, 78 and 7%, and the mean smolt age was 1·95 years. Mean lengths at smolting of age groups 1, 2 and 3 were 13·6, 15·8 and 17·5 cm, respectively. Females outnumbered males among the downstream migrating smolts with a mean sex ratio (females/ males) estimated at 1·61, with a significant female surplus in 7 of the 11 years sampled. Of the smolts sampled, 14% exhibited enlarged gonads indicative of parr maturation, and all were males (37% of the parr males sampled). Mean annual smolt density from 1975 to 1996 was 13·4 individuals 100 m−2 ranging between 0·3–31 smolts 100 m−2. Mean densities (100 m−2) of the smolts aged 1, 2 and 3 years were 1·5, 9·3 and 0·9 fish, respectively. Mean annual biomass for the 22-year period (1975–1996) was estimated at 437 g 100 m−2, with a range of variation from 136 to 683 g 100 m−2. Smolt age 2 made up 81% of the mean annual biomass (355 g 100 m−2) and smolt age 1 and 3, 8% (35 g 100 m−2) and 11% (47 g 100 m−2), respectively.  相似文献

The antagonistic effects of ethylene and Ag+ on the metabolism of [1-14C]indole-3-acetic acid (IAA) and on the rates of ethylene production were studied in tobacco leaf discs ( Nicotiana rustica var. Brasilia ). During the first 10 h of incubation, Ag+-pretreated leaf discs contained more free [14C]IAA than untreated ones due to decreased oxidative decarboxylation, and the discs also produced more ethylene. Exogenously supplied ethylene nullified these effects of Ag+. However, the most pronounced effect of Ag+ in increasing ethylene production, as well as the strongest antagonistic effect of exogenous ethylene, were found between 24 and 48 h of incubation. During this time span no effect on the level of free IAA and on its decarboxylation could be observed. It is suggested that ethylene exerted its autoinhibitory effect by a feedback control on the IAA-induced ethylene biosynthesis. Possible mechanisms for the autoinhibitory effect of ethylene are discussed.  相似文献

This study aimed to measure protein synthesis using a stable isotope method, investigate protein-nitrogen flux in a flatfish Pleuronectes flesus , and use the data to test the hypothesis that individual differences in growth efficiency were related to individual differences in protein-nitrogen flux mediated through differences in protein synthesis and degradation. Three measurements of protein-nitrogen flux via consumption, protein synthesis and nitrogenous excretion were made for individual flounder during a 212-day period and fractional rates of protein-nitrogen flux were scaled for a 50–g flounder to provide mean values for protein consumption (2·11 ± 0·21% day−1), protein synthesis (2·08±0·23% day−1), protein growth (0·71±0·06% day−1) and protein degradation (1·37±0·24% day−1). Mean rates of nitrogenous excretion were 0·142 mg N g−1 day−1 and 0·047 mg N g−1 day−1 for ammonia and urea, respectively. Individual flounder had different protein growth efficiencies and this was correlated negatively and significantly with mean rates of protein synthesis ( r - 0·70; P <0·05) and degradation ( r - 0·67; P < 0·05) and correlated positively and significantly with the efficiency of retaining synthesized protein ( r +0·63, P <0·05). This supported the proposed hypothesis that flounder which grow more efficiently achieve this through adopting a low protein turnover strategy.  相似文献
