Possible application of a modified procedure for treatment of soil samples with calcium carbonate under humid conditions to isolation of actinomycetes was studied. The procedure proved to be rather efficient in regard to increasing the number of the isolates including representatives of rare genera as compared with the routine methods. The number of microorganisms isolated from the soil samples treated with calcium carbonate under humid conditions was 2 orders higher than that from untreated soil samples. 1033 actinomycete cultures were isolated from the soil samples treated in accordance with the above procedure and only 597 cultures were isolated from the untreated soil samples. The antibiotic activity of the isolates was studied and their taxonomic position at the genus level was determined. The preliminary treatment of soil samples equally stimulated development of actinomycetes belonging to different genera. It is advisable that the described procedure for isolation of actinomycetes from soil samples in screening of cultures producing new antibiotics be used in combination with other selective methods of isolation.  相似文献

AIMS: To devise and evaluate a method for isolating the rare, zoosporic actinomycetes, Actinokineospora spp. in soil and plant litter. METHODS AND RESULTS: The newly developed method consists of two enrichment stages followed by plating on a selective medium. The source material is initially incubated with calcium carbonate to multiply the population of Actinokineospora spp., and is then air-dried. The second stage consists of rehydration-centrifugation, in which the amended substrate is immersed in phosphate buffer-soil extract to liberate actinomycete zoospores, and nonmotile microbial associates are then eliminated by centrifugation. Portions of the supernatant enriched with zoospores are plated on humic-acid vitamin agar supplemented with fradiomycin, kanamycin, nalidixic acid and trimethoprim. We examined 39 soil and plant-litter samples taken from fields, forests and stream banks. The proposed method consistently enriched and selectively isolated Actinokineospora spp. in 17 samples. Evidence for antimicrobial activity was found in most of the isolates. CONCLUSION: A combination of enrichment and a medium containing selective antibiotics can be used successfully for efficient isolation of certain rare actinomycete taxa. SIGNIFICANCE AND IMPACT OF THE STUDY: The development of new methodologies with which to isolate rare actinomycetes is of great importance to extend our understanding of their ecology, taxonomy and bioactivity.  相似文献

The present paper describes a simple enrichment technique which enables rapid and selective isolation of diverse zoosporic actinomycete genera directly from soil and plant litter. This technique, designated the rehydration and centrifugation (RC) method, consists of immersing the air-dried source material in 10 mM phosphate buffer containing 10% soil extract, letting the preparation stand at 30 °C for 90 min, followed by centrifugation of the fluid at 1,500×g for 20 min. Portions of the supernatant containing actinomycete zoospores are plated on the humic acid-vitamin agar which is supplemented with nalidixic acid and trimethoprim as the selective inhibitors for Gram-negative bacteria and bacilli. The phosphate buffer-soil extract solution significantly promoted liberation of motile zoospores from the source material. The centrifugation stage greatly eliminated streptomycetes and other non-motile actinomycetes from the liquid phase, thereby facilitating selective growth of rare, motile actinomycetes on the isolation plates subsequent to inoculation. Ten different soil and leaf-litter samples, taken from fields, forests, and stream banks, were examined. The RC method consistently achieved preferential isolation of motile actinomycetes in all samples, which accounted for 37–86% of the total microbial population recovered. The most frequently isolated motile actinomycetes were Actinoplanes and Dactylosporangium. Strains of Actinokineospora, Catenuloplanes and Kineosporia were also recovered, depending on the nature of the samples examined. Other motile actinomycetes that were occasionally isolated in small numbers included Actinosynnema, Geodermatophilus and Sporichthya.  相似文献

The composition of an arginine-glycerol-salt medium (AGS), suitable for the selective isolation of aerobic actinomycetes, was given. When soil samples were treated with calcium carbonate and plated on the AGS medium, higher total and relative plate counts of actinomycetes were obtained than when other media and methods were used.  相似文献

The rare actinomycete Actinomadura viridis was efficiently isolated from a variety of field soils by a new plate culture method in which a water suspension of dry-heated (110°C, 1h) soil was treated with 1.0% phenol and cultured on Humic acid-vitamin agar supplemented with kanamycin, josamycin, lysozyme and nalidixic acid. The method significantly eliminated unicellular bacteria and undesirable actinomycetes contaminants from the isolation plates, thereby achieving the highly selective isolation of A. viridis.  相似文献

【背景】红树林生境拥有丰富的微生物资源,分离获得更多的纯培养放线菌为新型抗生素的发现提供菌种资源。【目的】使用新型放线菌分离培养基,发掘广西北部湾茅尾海红树林生境放线菌资源,评估新型放线菌分离培养基分离效果。【方法】设计出新型放线菌分离培养基——椰子汁-海藻糖培养基,对广西北部湾茅尾海红树林根际土壤和红树林根皮放线菌进行分离。基于分子生物学鉴定方法,对获得的纯培养放线菌进行多样性分析。【结果】从红树林根际土壤样品中分离获得65株放线菌,分布在5个目10个科15个属,使用YIM171培养基分离到7个属,MA培养基分离到5个属,椰子汁-海藻糖培养基分离到11个属,其中包括4株潜在新种;从红树林根皮样品中分离获得19株放线菌,分布在4个目7个科10个属,使用YIM171和椰子汁-海藻糖培养基分别分离到7个属,包括4株潜在新种;使用椰子汁-海藻糖培养基在红树林生境两类样品中获得3株潜在新种。【结论】广西北部湾茅尾海红树林根际土壤和根皮中放线菌多样性丰富,使用椰子汁-海藻糖培养基分离放线菌效果较佳,可用作放线菌的分离培养基。  相似文献

A method for selective isolation of gramnegative nonsporulating bacteria from soil substrates was developed. The method consists of plating out the substrates on a glucose-yeast medium with addition of benzylpenicillin and nalidixic acid. The isolation frequency of gramnegative nonsporulating bacteria increased under such conditions from 9-16 (control) to 80-100 per cent. At the same time the isolation frequency of gram-positive bacteria decreased from 88.5 to 9.6 per cent.  相似文献

摘要:【目的】针对酸性土壤中的嗜酸丝状放线菌,建立有效的选择性分离方法,并了解其多样性。【方法】用不同的样品预处理方式和分离培养基,并添加不同的抑制剂进行分离;根据放线菌的菌落数和出菌率确定最佳分离方法组合。采用最佳分离方法对从江西采集的17份酸性土壤样品进行分离;根据培养特征对分离菌株进行分群,进一步通过对各类群的显微形态观察和pH梯度生长实验确定代表菌株;对代表菌株进行16S rRNA基因序列分析研究其多样性。【结果】嗜酸丝状放线菌的最佳分离方法为:土壤样品经分散差速离心预处理后,涂布添加了放线菌酮、制霉菌素和萘啶酮酸(各50 mg/L)的GTV培养基。用此方法共分离到放线菌369株,归为10个不同的颜色类群,其中6.6%为严格嗜酸放线菌,72.4%为中度嗜酸放线菌,21.0%为耐酸放线菌。52株嗜酸放线菌代表菌株分布于放线菌目中的12个属:链霉菌属(Streptomyces)、小单孢菌属(Micromonospora) 、诺卡氏菌属(Nocardia)、野野村菌属(Nonomuraea) 、韩国生工属(Kribbella) 、小双孢菌属(Microbispora)、马杜拉菌属(Actinomadura)、拟无枝菌酸菌属(Amycolatopsis)、指孢囊菌属(Dactylosporangium)、伦茨氏菌属(Lentzea)、游动四孢菌属(Planotetraspora) 和链嗜酸菌属(Streptacidiphilus),其中链霉菌分离菌株在系统发育树上形成12个不同的进化类群。【结论】所建立的选择性分离方法可用于土壤嗜酸丝状放线菌的高效分离;江西酸性土壤含有丰富多样的嗜酸丝状放线菌种属。  相似文献

An effective plate culture method for the isolation and enumeration of four-spored actinomycetes belonging to the Microtetraspora glauca group of Thiemann et al. is described, in which a water suspension of a soil sample given dry heat (110°C, 1 h) is subjected to a treatment with 0·05% benzethonium chloride (BC) and cultured on the new medium LSV-SE agar which is supplemented with kanamycin, norfloxacin and nalidixic acid. The LSV-SE agar, based on a commercial lignin (dealkaline; 1 g 1-1 as the major carbon source, specifically supported adequate growth and good sporulation for the M. glauca -group species, thus facilitating the detection and enumeration of this rare actinomycete group in the sample. The addition of the antibacterial agents to the LSV-SE agar, as well as the dry heat and BC treatment, provided a selective effect of reducing the nonfilamentous bacteria and undesirable actinomycetes associates occurring on the isolation plates. A total of 26 different natural samples were examined. From 18 samples (16 of field and forest soils, and two of stream and lake sediments), the new method consistently achieved the selective isolation of the M. glauca group, which accounted for 2–27% of the total population recovered. Definite evidence of biological activity which degrades the grass lignocellulose was found in all of the 39 test M. glauca -group isolates. Of these, 14, or 36%, possessed antimicrobial activity.  相似文献

A selective medium containing Todd-Hewitt broth, sheep blood, nalidixic acid, and gentamicin sulfate was found to enhance significantly the isolation of group B streptococci from vaginal cultures. Preparation of the medium, which is stable for up to 4 weeks at 4 C, is simple and inexpensive. Use of such a medium should facilitate identification of vaginal colonization with group B streptococci.  相似文献

Stepwise discriminant analysis was used to identify the most powerful selective substrates which could be used to formulate media capable of enriching for antibiotic-producing soil isolates. This was achieved by characterizing a collection of 74 soil bacteria, including eubacteria and actinomycetes, according to their ability to produce antibacterial antibiotics and their growth responses to 43 physiological and nutritional tests. The characters which were selective for actinomycetes relative to eubacteria included growth on proline (1%, w/v) and humic acid (0.1%) as sole sources of both carbon and nitrogen, growth on nitrate as a nitrogen source, and growth at pH 7.7-8.0. Growth on proline (1%) and humic acid (0.1%) as sole carbon/nitrogen sources, growth on asparagine as a nitrogen source, and growth in the presence of vitamins were among the characteristics which allowed antibiotic-producing actinomycetes to be differentiated from non-antibiotic-producing strains. Several simple isolation media which incorporated the selective substrates identified by discriminant analysis succeeded in increasing the proportion of actinomycetes isolated from soil samples. Furthermore, the percentage of isolates capable of antibiotic production was considerably increased.  相似文献

Tobramycin and dioxidine sensitivity of 57 strains belonging to 14 actinomycetes genera was studied. The cultures of Streptomyces were much more sensitive to tobramycin than the cultures of rare genera. The majority of the Streptomyces cultures showed a high resistance to dioxidine (MIC greater than or equal to 50 micrograms/ml). At the same time the majority of the cultures of rare genera were inhibited by low concentrations of dioxidine (no more than 50 micrograms/ml). For isolation of actinomycetes from soil samples, tobramycin, dioxidine, ceftriaxone and novobiocin were used. Tobramycin added in a concentration of 10 micrograms/ml to the Gauze agar organic medium No. 2 promoted a 2-fold increase in detection of actinomycetes of the rare genera as compared to the control. It was especially favourable for detection of cultures belonging to Micromonospora, Amycolatopsis, Streptosporangium and Nocardiopsis. Dioxidine in concentrations of 10 and 50 micrograms/ml inhibited the growth of the cultures belonging to rare genera. Ceftriaxone in the same concentrations inhibited the growth of the cultures of both Streptomyces and the rare genera. Novobiocin favoured detection of the cultures belonging to Amicolatopsis and Micromonospora. Therefore, among the tested compounds tobramycin and novobiocin appeared to be the most useful selective agents for isolation of actinomycetes of rare genera.  相似文献

Summary The isolation and pure culture of the symbiotic nitrogen-fixing frankiae has always been difficult. In the past the isolation of these actinomycetes directly from soil samples has proven impossible and isolations from root nodules of many genera has been only poorly successful. We report here a modified sucrose fractionation procedure which increased the success of isolations from root nodules and which permitted the isolation ofFrankia directly from soil samples. Crushed nodule suspensions or soil suspensions were incubated briefly in 0.7% phenol (carbolic acid) just before application to a sucrose density gradient. This phenol incubation decreased the number of contaminating eubacteria and fungi but more importantly increased the number ofFrankia developing on the isolation plates. If the phenol incubation was used solely without sucrose fractionation noFrankia were isolated, suggesting the death of the organisms due to phenol toxicity. The use of selective nitrogen-deficient media proved important for the isolation of frankiae from soils.  相似文献

In order to facilitate the discovery of novel actinomycetes from the Egyptian deserts, which can be useful as new sources for bioactive metabolites, different media for enumeration and isolation of desert actinomycetes have been tested. For this purpose, 30 soil samples from different six sites representing the Western and Eastern deserts of Egypt were collected. The two deserts are considered hyper-arid and the soil characteristics were determined. The media used were glucose–yeast extract agar, soil extract agar and a new minimal medium (MM) containing glucose, yeast extract and mineral salts. The effects of the soil characteristics on the total viable actinomycete counts on the three media were evaluated. The results showed that the highest actinomycete count in samples from five out of six sites was obtained on MM. Also MM was more selective for actinomycetes and significantly decreased the number of fungal colonies and to a lower extent the number of bacterial colonies. Moreover, it supported the development of different and diverse groups of actinomycetes. From the results obtained in this study, MM is a new useful medium for enumeration and selective isolation of actinomycetes from the desert soils.  相似文献

Petuely's selective medium for Bifidobacterium was improved by addition of riboflavin, nucleic acid bases, pyruvic acid, and nalidixic acid. The modified medium, when examined under strictly anaerobic conditions for efficient isolation of Bifidobacterium from human fecal samples, exhibited selective and high viable counts that were close to those found on the usual nonselective medium.  相似文献

从辐射污染区采集42份土样, 分别采用6种分离培养基进行放线菌的分离, 共获得152株放线菌。经形态、核糖体DNA扩增片段限制性内切酶(Amplifed ribosomal DNA restriction analysis, ARDRA)分析比较, 选取其中的60株进行16S rRNA基因测序。通过序列比对、聚类分析, 60株菌分布在放线菌纲中的12个属, 链霉菌属占大多数, 有大量稀有放线菌, 这表明辐射污染区具有较丰富的放线菌属种多样性。  相似文献
