Effect of estradiol-17 beta on rat vaginal epithelial cells (VEC) was studied by transmission electron microscopy. During the normal estrous cycle in adult rats keratinization and exfoliation of VEC was observed only in the estrus phase, though tonofilament bundles were seen distributed in the cytoplasm of the VEC during the proestrus and, to a lesser extent, in the diestrus phase. Electron microscope data on adult ovariectomised, immature and neonatally estradiol-primed rats demonstrated keratinization of the VEC within 24 h of injection. Proliferative activity was observed in the basal cells in primed animals. The basal cells remained cuboidal whereas cells of intermediate and luminal layers became flattened. These cells showed irregular profiles and, therefore, enhanced numbers of cellular junctions (desmosomes) were observed compared to control groups. Many tonofilament bundles were seen in the cytoplasm of cells belonging to intermediate and luminal layers in primed animals.  相似文献

Summary The epithelium of normal human hard palate was subjected to stereologic analysis. Ten biopsies were selected from a total of twenty specimens collected from 9 to 16 year old females, and processed for light- and electron microscopy. At two levels of magnification, electron micrographs were sampled from three strata (basale, spinosum, granulosum) in two locations (epithelial ridges and portions over connective tissue papillae). Stereologic point counting procedures were employed to analyse a total 1560 electron micrographs. In general, the thickness of the palate epithelium was 0.12 mm (over papillae) and 0.31 mm (in ridges), the epithelium is distinctly stratified, and homogeneously ortho-keratinized. From basal to granular layers, the composition of strata revealed decreasing densities of nuclei, mitochondria, membrane-bound organelles and aggregates of free ribosomes. Keratohyalin bodies and membrane coating granules increased, and cytoplasmic filaments with a constant diameter of about 85 Å increased from 14 to 30% of cytoplasmic unit volume. The cytoplasmic ground substance occupied a stable 50% of the epithelial cytoplasm in all strata. The composition of basal layers in ridges differed from that over connective tissue papillae. The data are discussed in relation to the observations that (1) an increasing gradient of filament density is not the most characteristic feature of ortho-keratinizing oral epithelium and (2) differences in the degree of differentiation in cells of the stratum basale coincided with the comparable frequency distribution pattern of dividing cells.The authors are thankful to Miss K. All stages of regeneration in hamster tracheal epithelium were studied following a denuding mechanical injury. At 1 h all the cells had sloughed from the wound site leaving a bare and sometimes disrupted basal lamina. Viable cells at the wound margins rapidly changed shape, flattened and migrated to cover the denuded lesion by 12 h. In addition, epithelial cells that remained viable demonstrated sublethal changes that included the rapid discharge of mucous granules from secretory cells, internalization of cilia by ciliated cells and evidence of heterophagy in both cell types. By 24 h a wave of epithelial cell divisions occurred, primarily by secretory cells. This produced a multilayered epidermoid metaplasia that was best developed at 48 h. The metaplastic epithelium was largely composed of cells with both secretory (mucous granules) and epidermoid (tonofilament bundles and numerous desmosomes) characteristics. The peroxidase-antiperoxidase (PAP) method demonstrated a few keratin-positive cells in the wound as early as 12 h post-wounding and keratin was demonstrated in more cells by 24 h. All cells in the metaplastic wound epithelium were keratin-positive by 48 h. Following 48 h some of the most superficial keratinized cells sloughed from the epithelium and the keratin content of the remaining cells began to decline. At 72 h pre-ciliated and pre-secretory cells were seen in the wound. Pre-ciliated cells were characterized by an abundant electron-lucent cytoplasm, large pale nucleus, filiform apical microvilli and evidence of ciliogenesis, similar to that seen during fetal development. Pre-ciliated cells often contained apical mucous granules, apparently carried over from the parent secretory cells. With the appearance of these columnar cells the normal mucociliary morphology was restored in small wounds by 120 h, but some persistent epidermoid metaplasia remained in the large wounds through 168 h post-wounding. These data provide further evidence for the important role of secretory cells in the histogenesis of epidermoid metaplasia and the regeneration of normal morphology following injury. The implications of these findings in understanding the histogenesis of other lesions in the tracheo-bronchial epithelium are discussed.  相似文献

The spontaneous development of a cell line of neonatal mouse C3H/He epidermal cells is described. The culture has been serially passaged at 29 °C over 18 months in the absence of any dermal support. The cell morphology of the 18th passage is reported. During early growth phase, the morphology of the cell layers was similar to that observed in the basal and differentiating strata of the epidermis: numerous tonofilament bundles and desmosome-filament complexes were observed. During late growth phase, maturation and vertical stratification occurred: demonstrated by the tonofilament accumulation, cell organelle degradation, nuclear pyknosis, presence of keratohyalin granules and horny cell layers with thickened membranes. Hemidesmosome-like structures were shown. No basal lamina or membrane coating granules were detectable. The 18th passage cultured cells did not induce tumors in nude mice. This keratinocyte cell line is not permanent, however: a malignant transformation occurred after 25 subcultures which resulted in an undifferentiated cell population.  相似文献

The influence of a 90% jejunoileal bypass on the rat exocrine pancreas was studied by morphometrical procedures. In sham-operated animals exocrine acinar cells accounted for 80.3% of the pancreas volume. These cells are composed of 9.9% nuclei, 8.4% mitochondria, 12.2% zymogen granules, 0.3% lipid droplets and 69.2% of a compartment ("ERGLS") composed of endoplasmic reticulum, ribosomes, Golgi areas, lysosomes and the cytoplasmic ground substance. Intestinal bypass did not change the volume density of exocrine cells nor that of nuclei in the cells during the three postoperative months. The means nuclear diameter was approximately the same in both groups. However, the volume density of secretory granules diminished by 50%. This was accompanied by a decrease in mean granular diameter, but not in their numerical density. The volume density of lipid droplets increased 10 fold, that of mitochondria increased slightly from the 15th postoperative day but significantly from the 45th day. The remaining cellular compartment composed of "ERGLS" was not modified by intestinal bypass. These findings suggest that a 90% jejunoileal bypass induces major changes in the composition of pancreatic acinar cells but not in their size.  相似文献

In Phestilla, the attachment of the larval body to shell and operculum occurs at muscle insertion sites. Attachment zones are specialized areas of squamous epithelium wherein the cells contain structures considered to be cytoplasmic anchors. The anchors are intracellular organelles consisting of apical and basal hemidesmosomal plaques connected by bundles of tonofilaments which traverse the cells. Muscle-to-epithelium and epithelium-to-shell adhesion is probably due to an extracellular cement. At metamorphosis, both shell and operculum are lost. Electron microscopic investigation of shell and opercular attachment sites during metamorphosis has demonstrated that apical hemidesmosomes lose their integrity and tonofilament bundles pull away from the apical plasmalemma of the epithelial cells as the cells lose contact with the shell or operculum.  相似文献

An electron microscopic study of satellite cells in the sensory lumbar ganglia from 12 pre- and post-hatching stages of Gallus gallus domesticus L., between the 5th and 120th d, was undertaken. A combined light and electron microscopic morphometric analysis was made in 10 and 18 d embryos and in 8, 35, 61, and 120 d post-hatching specimens. RER, SER, Golgi complex, mitochondria, free ribosomes, cell expansions, pinocytotic vesicles, subsurface cisternae and adhering, gap, and tight junctions are described from the different stages considered. Although a continuous increase in nuclear and cytoplasmic volumes is observed, there is a tendency towards a decrease in the cytoplasm/nucleus and nerve cell/satellite cell volume ratios.  相似文献

Summary Nerve cells of the superior cervical ganglion of young rats (20 g body weight) were investigated electron microscopically 6 h, 24 h, 48 h and 5 days after subcutaneous injection of nerve growth factor (10 /g body weight every 24 h). By means of a planimetric method with high accuracy significant changes of the Nissl substance and the Golgi apparatus could be demonstrated as early as 6 h after injection. Within the Nissl bodies both the density of bound ribosomes on the cisternae of the rough endoplasmic reticulum and the density of free ribosomes and polysomes decreased in a given field. This finding reflects a rearrangement of the Nissl substance and a spreading over larger areas of the cytoplasm, indicating an activation of the ribosomal system. The Golgi apparatus, in particular its outer part, increases in volume with time of nerve growth factor treatment. On the other hand, the total cell volume does not show significant changes before 48 h of nerve growth factor treatment. At this time an increase in the cytoplasmic volume can be seen, whereas the nuclear volume remains unchanged. The possibility of correlations of the present findings with data from biochemical studies done under similar experimental conditions is discussed.We thank Mrs. M. Summary The epithelium of intact guinea pig palate was subjected to stereologic analysis in a study of structural alterations in the keratinizing epithelium in response to wounding. Point counting procedures were employed to analyse electron micrographs sampled from three epithelial strata in biopsies collected from five animals. The differentiation pattern of the guinea pig palate epithelium displayed the following structural density gradients from basal to granular layers: descending gradients of metabolically active organelles, ascending gradient of bundled filaments coupled with the appearence of membrane coating granules and keratohyalin granules, and a plateau-like gradient of cytoplasmic ground substance. This pattern of epithelial differentiation is basically identical to that of human hard palate epithelium and epidermis. Two kinds of epithelial cells, dark and light types, are alternately arranged in the gill of Daphnia magna. The dark cell has numerous mitochondria and an elaborate tubular system containing two kinds of cytoplasmic tubules, small about 70 nm in diameter, and large about 130 nm in diameter. The former occur in bundles and seem to be smooth-surfaced endoplasmic reticulum. The latter, lined with a ridged surface coat and frequently open at the lateral and basal cell membrane, are regarded as extensions of the cell membrane. The atypical cell membrane of the dark cell is modified by repeated subunits of a cytoplasmic coat on the inner leaflet of the unit membrane. The light cell exhibits a high degree of basal infoldings of the cell membrane, which represent a magnification of the surface area of the cell. Large mitochondria between the infoldings often come into intimate association with the infolded cell membrane to form a regular array of parallel mitochondria interposed with the double cell membranes. The results suggest that at least the dark epithelial cells play an important role in the osmoregulation of this animal.  相似文献

The ultrastructure of the adult frog ciliary epithelium cells has definite regional differences. Cells of ciliary epithelium folds near the iris display morphological features characterizing its barrier and secretory functions which lead to the formation of aqueous humor. These are junctional complexes with tight junctions (zonula occludents) in the apical parts of contacting sides of cells of the inner leaf: a great quantity of mitochondria, ribosomes and various vesicles, well developed endoplasmic reticulum in the cytoplasm, much folded basal surface, gap junctions between cells of external and internal leaflets. In the mammalian inner epithelial layer different cell junctions are known to be arranged in a fixed spatial fashion. Unlike, in the frog's epithelium both zonula adherent and desmosomes may be found in any sequence. Tight junctions are formed during metamorphosis, on the place of focal junctions, whereas gap junctions, referred to earlier as "extended", start functioning between cells just on the very early stages of eye morphogenesis (Dabagyan et al., 1979). The epithelium of the posterior part of the ciliary fold and pars plana of the ciliary body have, in addition, the number of morphological sign indicating the cell involvement in the accomodational function of any eye (i. e. a majority of desmosomes binding all cells together and of zonulae adherentes, well developed intracellular skeleton of tonofilament bundles). These features are characteristic of the whole distal part of ciliary epithelium rather than of the place of attachment of zonula fiber only.  相似文献

Summary Young rats were treated with high doses of parathyroid hormone (PTH). Osteoclasts from these animals revealed characteristic alterations in comparison to control cells: a) The cytoplasm contained large vacuoles with phagocytosed cells, some of which resembled osteoblasts or osteocytes. The vacuoles were interpreted as lysosomes because the engulfed cells often appeared partly digested and the vacuoles contained acid phosphatase as demonstrated histochemically, b) lipid droplets were present in the cytoplasm and usually located close to the endoplasmic reticulum and/or in regions with many free ribosomes, c) the Golgi complex was more frequently separated from the nuclei than in control cells, d) small coated cytoplasmic bodies were numerous in the peripheral cytoplasm, e) the membranes of the endoplasmic reticulum were fused in some places, f) cytoplasmic regions with numerous free ribosomes were frequent, g) large ring-shaped granules occurred in some mitochondria. Energy dispersive X-ray analysis of these granules provided evidence that they contained calcium and probably phosphorus, h) in some osteoclasts the mitochondria were enlarged. — The findings are consistent with an increased activity of osteoclasts and in particular a stimulation of the lysosomal system in these cells.This research was supported by grants no. 512-819, 512-1545 and 512-1912 from the Danish Research Council. Summary Stereological data of phytohaemagglutinin (PHA)-activated human T-lymphocytes were recorded at intervals (12 to 72 h) together with biochemical (isotope-uptake, lymphotoxin-release) and morphological measurements. About 98 % of the cells were activated 12 h after PHA-stimulation. The activation phase lasted less than 48 h, i.e., cells entering the activation phase within 12 h were at their activation maximum by 48 h. The activated cell increased in size. The nuclear/cytoplasmic-ratio decreased. Most of the cytoplasmic organelles developed in phase with the increase of cytoplasmic volume. After 48 h, mitotic figures were frequently seen. Due to the increasing number of secondary, activated daughter cells, parameters of most cytoplasmic components declined between 48 and 72 h. Structural changes in the nucleus preceded the 3H-leucine uptake, which had not reached its maximum after 72 h of incubation. The 3H-leucine uptake started as early as 12 h after culture initiation, and its increase was proportional to the increasing polyribosome density. No maximum uptake was reached up to 72 h, but the development of structural components related to this uptake was at its maximum at the end of the activation phase (48 h). The formation of bound ribosomes occurred subsequent to the enlargement of the surface of the rough endoplasmic reticulum. Initial polysome formation occurred at the expense of existing free ribosomes.  相似文献

The secretions of the salivary gland system are essential for the maintenance of oral health. The nature of cell-specific secretions of the various glands and their regulation is not completely understood. The objective of this study was to establish epithelial cell cultures from the human parotid gland that exhibit the tissue-specific function of α-amylase secretion. A specimen of normal human parotid gland was obtained at surgery and used to obtain primary cultures by the explant/outgrowth procedure. The cultures were maintained in keratinocyte basal medium, supplemented with insulin (5 μg/ml), EGF (10 ng/ml), hydrocortisone (0.5 μg/ml), bovine pituitary extract (25 μg/ml), and antibiotics. The cultures were passaged using 0.125% trypsin to dissociate the cells. Phase contrast and ultrastructural observations showed that the cells were polygonal and exhibited desmosomes. Their cytoplasm contained tonofilament bundles and abundant rough endoplasmic reticulum and Golgi complexes. Immunofluorescence studies showed that all cells were positive for cytokeratins. Immunoblot analysis revealed keratins with molecular weights of 58, 56, 52, 50, 48, 46, and 40 KD, which are characteristic of secretory epithelia. The cells have been passaged 35 times so far, undergoing a cumulative 120–140 population doublings. The serially passaged epithelial cell cultures produced and secreted α-amylase, a major component of parotid gland acinar cell secretion. The β-adrenergic agonist, isoproterenol (ISP), stimulated α-amylase secretion, which was accompanied by increased intracellular concentrations of cAMP. ISP-induced stimulation of amylase and cAMP was blocked by the β-adrenergic antagonist, propranolol. Further, dibutryl cAMP also enhanced the secretion of amylase. Thus we have established a long-term epithelial cell culture model of human parotid gland epithelial cells that exhibits differentiated function and retains the intact β-adrenergic receptor system. © 1993 Wiley-Liss, Inc.  相似文献

Adventitious root formation in vitro in 1-mm stem slices cut from microshoots of apple cv. Jork 9 was studied using light and electron microscopy. When indole-3-butyric acid (IBA) had been added to the medium, starch grains accumulated during the first 24 h of culture in cells of the cambial region and in cells in the vicinity of vascular tissue and in the primary rays. This accumulation occurred only in the basal part of explants. After that, the nuclei in these cells were activated, and the density of the cytoplasm and the number of cell organelles increased, whereas starch was broken down. Cambium cells started to divide transversely and at 96 h, after several divisions, a continuous ring of isodiametric cytoplasmic cells had appeared around the xylem near the basal cutting surface. The cells in this ring were rich in cell structures, and did not contain large starch grains and a central vacuole. Root meristemoids regenerated from the portions of the ring that were localized in the primary rays. From the other cells in the ring, callus developed. The meristemoids did not grow into the direction of the epidermis as in shoots, but along the vascular bundles. After emergence from the cutting surface, the meristemoids were transformed into small, dome-like primordia. They developed a typical root apex with root cap, root ground meristem and tracheid connection with shoot vascular tissue. Taxol induces a vast increase in the number of microtubules (MTs) in functional chondroblasts. The drug also induces a marked change in MT distribution. In control cultures, anti-tubulin stains long, fine, sinuous filaments radiating from a perinuclear center. In taxol-treated cells, anti-tubulin stains stubby, straight, chevron-like structures that assume a striking antipodal distribution. Such MT-bundles are relatively stable: they persist for over 48 h after removal of taxol, and even for 16-24 h in Colcemid. Many of these supernumerary MTs bind to, and align on, the cytoplasmic face of the rough endoplasmic reticulum (RER). In binding, the MTs displace the numerous ribosomes that normally stud the surface of the cisternae of the RER. The bound MTs form a remarkably uniform layer with center-to-center spacings of 40 nm. The attached parallel arrays of MTs achieve lengths of over 10 microns. These bound MTs not only dislodge ribosomes from the RER surface, but they also zip together adjacent ER complexes, forming tiers of two to eight cisternae. Numerous cytoplasmic bundles of hexagonally-ordered MTs are also induced. When closely aligned, the MTs assume a crystalline configuration with a six-fold symmetry, a central MT being surrounded by six equidistant MTs. A single cell can have over 100 MT-bundles and the number of MTs per bundles varies from 2-30. The forces aggregating cytoplasmic MT-bundles probably differ from those that bind MTs to the RER. Taxol also fragments the prominent Golgi complex that characterizes actively secreting chondroblasts. No obvious morphologic relationship has yet been detected between these induced MTs and other organelles such as intermediate-sized filaments, microfilaments, mitochondria, Golgi cisternae, or secretory vesicles.  相似文献

A study was made of the ultrastructure of epithelial cells of the rat proximal tubules nephrons 25-30 seconds, and 15 and 30 minutes following penicillin injection. The morphological changes in the transporting cells that were observed involved the dilation of the inner cavities of canals and cisterns of the endoplasmic reticulum. The number of elements of the smooth endoplasmic reticulum concentrated on the periphery of the cell, near the basolateral border, was seen to increase. Microbodies appeared enlarged with the appearance of the nucleolus; the electron density of the cytoplasmic ground substance fell down. The space between cells augmented, and the interstitial space between infoldings of the basal plasmalemma was much broadened. Similar morphological changes but different in the intensity were found at either stage examined. The maximum changes were established 25-30 seconds following penicillin injection.  相似文献

A liver biopsy specimen from a case of primary amyloidosis was investigated by electron microscopy. The cytoplasmic periphery of the hepatocytes showed degenerativechanges which are interpreted as indicating shedding of peripheral parts of the cytoplasm. Two main variants of this process could be discerned: 1) Protrusion and sequestration of hernia-like blebs of cytoplasm, and 2) shedding of vesicles derived from degenerated endoplasmic reticulum. In the latter case transient defects of the plasma membrane seem to be relevance. Endoplasmic reticulum and cytoplasmic ground substance appeared to be shed preferentially, whereas mitochondria are retained within the cell. As a consequence the fractional volume of the mitochondria in the cytoplasm of atrophic cells is markedly increased. Shedding of peripheral cytoplasm, therefore, seems to be an effective mechanism enabeling the cell to adapt the mass and the composition of its cytoplasm to an unfavourable environment.  相似文献

在玉米(ZeamaysL.)叶片原形成层发育的不同阶段常出现细胞衰退的现象,其衰退方式可分为自溶型和凝缩型。自溶型衰退的特点是细胞发生一系列降解活动最后导致细胞自溶死亡。衰退早期的细胞出现明显的自体吞噬活动,伴随核糖体和细胞质电子密度下降。衰退最后阶段的细胞自溶导致所有原生质组分的解体和消失,其中细胞核和质膜是最后消失的组分。凝缩型衰退的起动则伴随细胞质密度的增大。随着衰退的进行,原生质组分以一定顺序逐步退化解体:首先是高尔基体和内质网,接着是质体、细胞核和部分线粒体。但到衰退后期当细胞被挤压塌陷时仍有质膜、核糖体和线粒体存在。  相似文献
