Pathogenic bacteria, such as multidrug-resistant Staphylococcus aureus (MRSA), which are not susceptible to most conventional antibiotics, are causing increased concern in healthcare institutions worldwide. The discovery of novel antibacterial compounds for biomedical exploitation is one avenue that is being pursued to combat these problematic bacteria. Marine eukaryotic microalgae are known to produce numerous useful products but have attracted little attention in the search for novel antibiotic compounds. Cell lysates of the marine diatom, Phaeodactylum tricornutum Bohlin, have been reported to display antibacterial activity in vitro, but the compounds responsible have not been fully identified. In this paper, using column chromatography and reversed-phase high-performance liquid chromatography, we report the isolation of an antibacterial fatty acid. Mass spectrometry and 1H-nuclear magnetic resonance spectroscopy revealed it to be the polyunsaturated fatty acid, eicosapentaenoic acid (EPA). We show that EPA is active against a range of both Gram-positive and Gram-negative bacteria, including MRSA, at micromolar concentrations. These data indicate that it could find application in the topical and systemic treatment of drug-resistant bacterial infections.  相似文献

Antibodies are important drugs for treating cancer and there is strong rationale for using multiple antibodies to improve outcomes. We labeled two breast cancer binding antibodies, anti-ErbB2 and anti-EpCAM, with infrared fluorescence dyes of different wavelengths and determined their in vivo distribution in a breast cancer xenograft model using a near-infrared (NIR) fluorescence imaging system. Our data show that these two antibodies can be readily assessed simultaneously in mouse xenograft model. This will help guide design of dosing strategies for multiple antibodies and identify potential interaction that could affect pharmacokinetics and possible side effects.  相似文献

A general purpose transformation vector, designated pPha-T1, was constructed for use with the diatom Phaeodactylum tricornutum Bohlin. This vector harbors the sh ble cassette for primary selection on medium containing the antibiotic zeocin, and a multiple cloning site flanked by the P. tricornutum fcp A promoter. pPha-T1 was used to establish the utility of three selectable marker genes and two reporter genes for P. tricornutum transformation. The nat and sat-1 genes confer resistance to the antibiotic nourseothricin, and npt II confers resistance to G418. Each of these genes was effective as a selectable marker for identifying primary transformants. These markers could also be used for dual selections in combination with the sh ble gene. The reporter genes uid A and gfp were also introduced into P. tricornutum using pPha-T1. Gus expression in some transformants reached 15 μg·μg−1 of total soluble protein and permitted excellent cell staining, while GFP fluorescence was readily visible with standard fluorescence microscopy. The egfp gene, which has optimal codon usage for expression in human cells, was the only version of gfp that produced a strong fluorescent signal in P. tricornutum. The codon bias of the egfp gene is similar to that of P. tricornutum genes. This study suggests that codon usage has a significant effect on the efficient expression of reporter genes in P. tricornutum. The results presented here demonstrate that a variety of selectable markers and reporter genes can be expressed in P. tricornutum , enhancing the potential of this organism for exploring basic biological questions and industrial applications.  相似文献

ABSTRACT

Both Heterosigma akashiwo and Phaeodactylum tricornutum have been reported to produce allelochemicals capable of inhibiting the growth of co-occurring microalgae. Here, potential allelopathy between H. akashiwo and P. tricornutum was evaluated using bi-algal culture, cell-free culture filtrate and a no-contact co-culturing system in nutrient-replete media. Experiments were also conducted in the no-contact co-culturing system under nutrient-limited conditions. In nutrient-replete bi-algal culture, the growth of P. tricornutum and H. akashiwo each tended to be strongly suppressed when the other species was inoculated at high cell densities. A mathematical model was used to simulate the growth interactions of the two species in bi-algal culture and showed that P. tricornutum outcompeted H. akashiwo over time with different initial cell densities under nutrient-sufficient conditions, indicating that P. tricornutum was more potent in allelopathy. Heterosigma akashiwo growth was inhibited both in the P. tricornutum culture filtrate and no-contact co-culturing system. This confirmed that the extracellular allelopathic compounds released by P. tricornutum were one of the sources of the H. akashiwo growth inhibition. Nutrient deficiency did not increase the extent of allelopathic activity of allelochemicals.  相似文献

The growth, photosynthesis, and respiration of the marine diatom Phaeodactylum tricornutum were examined under photoautotrophic and mixotrophic conditions. 100 mM glycerol, acetate, and glucose significantly increased specific growth rate, and mixotrophic growth achieved higher biomass concentrations. Under mixotrophic conditions, respiration rate (R d) and light compensation irradiance (I c) were significantly higher, but net maximum photosynthetic O2 evolution rate (P m) and saturation irradiance (I k) were depressed. Organic carbon sources decreased the cell photosynthetic pigment content and chlorophyll a to c ratio, but with a higher carotenoid to chlorophyll a ratio. Ratios of variable to maximum chlorophyll fluorescence (F v/F m) and 77 K fluorescence spectra of mixotrophic cells indicated a reduced photochemical efficiency of photosystem II. The results were accompanied by lower electron transport rate. Therefore, organic carbon sources reduced the photosynthesis efficiency, and the enhancement of biomass of P. tricornutum implied that organic carbon sources had more pronounced effects on respiration than on photosynthesis.  相似文献

Mixotrophic growth of the eicosapentaenoic acid (EPA)producing diatom Phaeodactylum tricornutum UTEX640 was carried out in 1-L batch cultures under anexternal irradiance of 165 mol photons m-2s-1 by supplementing the inorganic culture mediumwith glycerol. The effect on the growth and the fattyacid profile was studied for different initialglycerol concentrations (0–0.1 M). The optimalglycerol concentration was 0.1 M.A lag phase was observed at high glycerolconcentrations. The present study also shows thatsuccessive additions of glycerol at 0.1M concentrationand using ammonium chloride as a nitrogen sourceremarkably increased the maximum biomass concentration(16.2 g L-1) and maximum biomass productivity(61.5 mg L-1 h-1). These values wererespectively 9 and 8-fold higher than in thephotoautotrophically grown control. The level ofsaponifiable lipids in mixotrophically cultured cellswas significantly higher than in photoautotrophicallycultured cells and increased with the glycerolconcentration in the medium. The concentration ofstorage lipids, saturated and monounsaturated fattyacids, were enhanced but the EPA content did notchange significantly. The EPA content was around 2.2%of biomass dry weight. The maximum EPA yield was33.5 mg L-1 d-1 and was obtained in aculture containing 0.1 M glycerol, supplementedperiodically by ammonium chloride. This productivitywas 10-fold higher than the EPA productivity obtainedunder mixotrophic conditions.  相似文献

The purpose of this study was to explore efficient methods of harvesting the microalga Phaeodactylum tricornutum. Natural sedimentation experiments, performed at different light and temperature conditions, did not yield significant improvements in efficiency even after 1?week. When alkalinity-induced flocculation was performed, both the flocculation efficiency and the concentration factor dramatically improved at pH?=?9.75 (0.5–0.7 units over the original pH of the culture) after 10?min settling time. Sedimentation rates are documented at pH ranging between pH?9.75 and 11.0. The results of the application of two conventional flocculants used in wastewater treatment, polyaluminium chloride and aluminium sulphate, are also presented. Chitosan was also used as a natural flocculating agent to improve possible contamination problems in the downstream process. pH was adjusted in order to determine optimum flocculation efficiency of chitosan in combination with a high concentration factor. Satisfactory results were found with chitosan at an adjusted pH of 9.9 using concentrations as low as 20?mg?L?1, after testing a flocculant range of 5–200?mg?L?1.  相似文献

The extracellular matrix of the ovoid and fusiform morphotypes of Phaeodactylum tricornutum (Bohlin) was characterized in detail. The structural and nanophysical properties were analyzed by microscopy. Of the two morphotypes, only the ovoid form secretes adhesive mucilage; light microscopy and scanning electron microscopy images showed that the mucilage was secreted from the girdle band region of the cell as cell‐substratum tethers, accumulating on the surface forming a biofilm. After 7 d, the secreted mucilage became entangled, forming adhesive strands that crisscrossed the substratum surface. In the initial secreted mucilage atomic force microscopy identified a high proportion of adhesive molecules without regular retraction curves and some modular‐like adhesive molecules, in the 7 d old biofilm, the adhesive molecules were longer with fewer adhesive events but greater adhesive strength. Chemical characterization was carried out on extracted proteins and polysaccharides. Differences in protein composition, monosaccharide composition, and linkage analysis are discussed in relation to the composition of the frustule and secreted adhesive mucilage. Polysaccharide analysis showed a broad range of monosaccharides and linkages across all fractions with idiosyncratic enrichment of particular monosaccharides and linkages in each fraction. 3‐linked Mannan was highly enriched in the cell frustule fractions indicating a major structural role, while Rhamnose and Fucose derivatives were enriched in the secreted fractions of the ovoid morphotype suggesting involvement in cell adhesion. Comparison of SDS‐PAGE of extracellular proteins showed two major bands for the ovoid morphotype and four for the fusiform morphotype of which only one appeared to be common to both morphotypes.  相似文献

The influence of osmotic stress on proline catabolism in the diatom Phaeodactylum tricornutum and the fate of proline after relaxation of osmotic stress are described. The conversion of 14C-proline into metabolic products is greatly inhibited during water stress conditions. A sudden reversal of osmotic stress from 1.9 to 0.77 osmolar in the algal medium leads to a rapid decrease of the cellular proline concentration. This is mainly due to a release of proline into the outside medium of the algal cells, caused by a temporary breakdown of selective permeability in the plasmamembrane. The magnitude of membrane leakage depends on the concentration differences applied. It is supposed that the altered barrier properties reflect an increase in membrane fluidity, as a consequence of osmotic downshock. A replacement of NaCl by KCl, during continued osmotic stress, leads to a rapid and irreversible breakdown of selective permeability in the plasmamembrane. This effect might be due to a special lipid composition of the plasmamembrane or caused by an influence on the membrane potential.  相似文献

Effects of copper at concentrations of 0.13 and 0.25 mg/l on content of photosynthetic pigments, growth, size characteristics, and cell shape of Phaeodactylum tricornutum (Bacillariophyta) microalga were studied. The inhibitory effect of copper on growth parameters was shown to become stronger with increasing concentration of copper in the growth medium. In spite of considerable retardation of cell growth, the concentrations examined had no effect on the content of photosynthetic pigments though affected some morphometric parameters.  相似文献

Three pigment-protein complexes were isolated from the marine diatom Phaeodactylum tricornutum (Bohlin) by treatment of thylakoid membrane fragments with 1% Triton X-100 at 4°C followed by centrifugation on sucrose density gradients. The major complex contains chlorophyll a, c1, c2, and the carotenoid fucoxanthin (chlorophyll a: c1: c2: fucoxanthin = 1.0: 0.09: 0.28: 2.22) bound to an apoprotein doublet of 16.4 and 16.9 kilodaltons. This complex accounts for >70% of the total pigment and 20 to 40% of the protein in the thylakoid membranes. Efficient coupling of chlorophyll c and fucoxanthin absorption to chlorophyll a fluorescence supports a light-harvesting function for the complex. A minor light-harvesting complex containing chlorophyll a, c1, and c2 but no fucoxanthin (chlorophyll a: c1: c2 = 1.0: 0.23: 0.26) was also isolated at Triton: chlorophyll a ratios between 20 and 40. These pigments are bound to a similar molecular weight apoprotein doublet. The third complex isolated was the P700-chlorophyll a protein, the reaction center of photosystem I, which showed characteristics similar to those isolated from other plant sources. The yield of the chlorophyll a/c-fucoxanthin complex was shown to respond strongly to changes in light intensity during growth, accounting for most of the changes in cellular pigmentation.  相似文献

In Phaeodactylum tricornutum cells kept 30 min in the dark, induction of fluorescence showed the well-known levels OIDPSMT. The decrease of MT was the most important when the intensity of excitation light was high. It was mainly due to the photochemical quenching. After addition of DCMU (2 to 20 M), a quenching qE was still observed: this quenching, cancelled by NH4Cl (2 to 20 mM) is attributed to pH. This qE was also inhibited by antimycin, an inhibitor of cyclic phosphorylation and may be of chlororespiration above plastoquinones. Anaerobiosis also decreased it. We can infer that chlororespiration also plays a part in the formation of the pH in the presence of DCMU. After 30 mn of preillumination in red light, the levels P and M were lower and the quenching in presence of DCMU was no more observed: thus, neither the chlororespiration nor the cyclic phosphorylation were active, unless the activity of ATPase was much more important. So, in diatoms, one at least of the above cited phenomena can be modulated by light.  相似文献

The extractability of chlorophyllase is used as an indicator for the way in which the enzyme is incorporated in Phaeodactylum tricornutum photosynthetic membranes. Whereas with various aqueous solutions no appreciable amount of chlorophyllase is washed from the membranes even after chloroform-pretreatment. the enzyme can be extracted with aqueous solutions after practically all the lipids have been removed by means of 80% acetone. The proteins in an aqueous extract of the membranes thus treated were separated by electrophoresis on a 4% polyacrylamide gel; extraction of the active protein from several gels yielded a purified chlorophyllase solution. As with the intramembraneous enzyme, activity of purified, solubilized chlorophyllase depends upon the combined presence of MgCl2 and dithiothreitol. The enzyme is inactivated upon heating at 56°C for 5 min. After SDS-gel-electrophoresis of an enriched extract, enzyme activity could be localized in the gels. The molecular weight of SDS-treated chlorophyllase, or of its principal subunit, was estimated to be about 38 kilodaltons. The results are discussed in terms of the identity of prochlorophyllase.  相似文献
