The temporal relationships of the proestrous LH surge and the circadian locomotor activity rhythm were compared in hamsters entrained to four different 24-hr light-dark (LD) cycles. Animals were housed in cages equipped with running wheels to obtain continuous activity records. Stable entrainment of locomotor activity was complete within 3 weeks of exposure to each photoperiod at which time hamsters were randomly assigned to hourly sample groups. Serum was obtained by cardiac puncture under light ether anesthesia on the day of proestrous and was assayed by RIA for LH. A computer-based least-squares sine wave-fitting technique determined a single objective phase reference point for the time of the hormone maximum. In each photoperiod, precise temporal relationships were maintained between the LH surge and activity onset, whereas the phase relationship between the LH surge and the LD cycle was more variable. These data indicate that the environmental LD cycle entrains the circadian timing system which, in turn, provides temporal information to the rhythms of proestrous gonadotropin and locomotor activity.  相似文献

This study tested whether light pulses with a dusklike offset or a dawnlike onset caused phase shifts of different sizes in the circadian wheel-running activity of Syrian hamsters, Mesocricetus auratus. Six experiments were conducted, each with 30 hamsters; the hamsters received first one type of pulse and then the other type a few weeks later, allowing a paired comparison. The six experiments represented the combination of two maximum light intensities (150 and 250 lux) and three zeitgeber times (ZTs) at which the pulses were given (ZT13.5, ZT14.5, and ZT20). Pulses were 30 minutes long, a relatively short duration to minimize circadian time effects. Aschoff's type II method of measuring phase shifts was used. In none of the six experiments did a two-tailed paired t test detect a significant difference in the size of phase shifts caused by dusklike versus dawnlike pulses. A three-way analysis of variance (ANOVA) on the combined data from all six experiments (with pulse type, pulse intensity, and ZT as factors) also failed to detect a significant effect of pulse type. Statistical power was calculated and found to be reasonably good. These negative results are in line with those of a previous study in which a different methodology was used. Corticosterone alone was not able to stimulate release of luteinizing hormone (LH) from anterior pituitary cells invitro, but corticosterone in combination with luteinizing hormone releasing hormone (LHRH) augmented the release of LH into the culture media. Experiments were performed to study the responsiveness of the pituitary to gonadotropin-releasing hormone (GnRH) during the dynamic changes in gonadotropin secretion associated with the estrogen-induced luteinizing hormone (LH) surge in the ovariectomized (OVX) rhesus monkey. Silastic capsules filled with estradiol-17-beta were implanted subcutaneously in ovariectomized rhesus monkeys, resulting in an initial lowering of circulating LH and follicle-stimulating hormone (FSH) concentrations followed by an LH-FSH surge. GnRH was injected intravenously just before estrogen implantation, during the negative feedback response and during the rising, the peak, and the declining phases of the LH surge. The LH and FSH responses during the negative feedback phase were as large as those before estrogen treatment (control responses). During the rising phase of the LH surge, the acute response to GnRH injection did not differ significantly from the control response, but the responses 60 and 120 min after injection were somewhat increased. During the declining phase of the LH surge, the pituitary was not responsive to exogenous GnRH, although LH probably continued to be secreted at this time since the LH surge decreased more slowly than predicted by the normal rate of disappearance of LH in the monkey. We conclude that an increased duration of response to GnRH may be an important part of the mechanism by which estrogen induces the LH surge, but we do not see evidence of increased sensitivity of the pituitary to GnRH as an acute releasing factor at that time.  相似文献

Circadian rhythms in Syrian hamsters can be phase advanced by activity or arousal stimulated during the daily rest phase ("subjective day"). A widely used method for stimulating activity is confinement to a novel wheel. Some hamsters decline to run, and some procedures may reduce the probability of running. The authors evaluated food deprivation (FD) as a method to promote running. Given evidence that perturbations of cell metabolism or glucose availability may affect circadian clock function in some tissues or species, they also assessed the effects of FD on free-running circadian phase, resetting responses to photic and nonphotic stimuli and plasma glucose. In constant light, a 27-h fast significantly increased running in a novel wheel and marginally increased the average size of resulting phase shifts. FD, without novel wheel confinement, was associated with some very large phase shifts or disruption of rhythmicity in hamsters that spontaneously ran in their home wheels during the subjective day. Hamsters that ran only during the usual active phase (subjective night) or that were prevented from running did not exhibit phase shifts, despite refeeding in the mid-subjective day. Using an Aschoff Type II design for measuring shifts, a 27-h fast significantly increased the number of hamsters that ran continuously when confined to a novel wheel but did not affect the dose-response relation between the amount of running and the size of the resulting shift. A day of fasting also did not affect the size of phase delay or advance shifts to 30-min light pulses in the subjective night. Plasma glucose was markedly reduced by wheel running in combination with fasting but was increased by running in nonfasted hamsters. These results establish FD as a useful tool for stimulating activity in home cage or novel wheels and indicate that in Syrian hamsters, significant alterations in glucose availability, associated with running, fasting, and refeeding, have surprisingly little effect on circadian pacemaker function.  相似文献

The goal of this study was to provide an example of nonsocial and nonphotic entrainment in Syrian hamsters, together with a corresponding phase response curve (PRC). Fourteen male hamsters were given 2-hr bouts of induced activity (mostly wheel running) at 23.83-hr intervals in constant darkness (DD). The activity onsets of 10 hamsters entrained to this manipulation, with no anticipatory activity present. After entrainment, the rhythms resumed free-running from a time 0.66-3.91 hr after the onset of the last bout of induced activity. Postentrainment free-running periods were shorter than pre-entrainment values. The PRC for 2-hr pulses of induced activity in DD revealed phase advances induced in some animals between circadian time (CT) 4 and CT 11 (approximately the last half of the hamsters' rest period), and delays between CT 23 and CT 3 and between CT 17 and CT 20. The CTs for phase advances are compatible with the phase angle differences observed between rhythm and zeitgeber at the end of entrainment. Many features of the results (not all animals entraining, PRC characteristics, lack of observable anticipation to the daily stimuli, phase relationship between zeitgeber and activity rhythms) are similar to those from a previous study on social entrainment in this species (Mrosovsky, 1988). These similarities reinforce the idea that induced activity and social zeitgebers act on activity rhythms via a common mechanism.  相似文献

Circadian rhythms of body temperature, heart rate, and locomotor activity were observed in the unanesthetized and unrestrained Syrian hamsters, Djungarian hamsters and Chinese hamsters, and the differences in these biological characters among the three species of hamster were investigated. In each species, body temperature, heart rate, and locomotor activity in the dark period were higher than those in the light period. Heart rate of Chinese hamsters was higher than that of the others in both the light and dark periods. In addition, it was found that the body temperature of Djungarian hamsters decreased rapidly one time a day. These results show species differences in body temperature, heart rate and locomotor activity of Syrian, Djungarian and Chinese hamsters.  相似文献

Testosterone (T) and luteinizing hormone (LH) in the peripheral plasma of 6 young postpubertal (52 weeks of age) bulls were measured by radioimmunoassay. For 3 bulls blood samples were collected at half-hour intervals for 6 hours one day before dexamethasone (DXM) injection (20 mg) and two days after. For the 3 others blood collection occurred two days before injection and two days after. On the days before treatment, T and LH concentrations fluctuated similarly to what was previously observed. After treatment LH decreased rapidly and remained between 0.25 and 1.0 ng/ml until the end of the experiment. We observed a small peak of T (between 1.9 and 6.1 ng/ml depending on the animal) immediately after DXM injection; this peak was followed by a decrease to low values (0.25 to 0.5 ng/ml) as soon as 4 hours after injection. It is concluded that DXM suppresses the testosterone secretion. Since we observed a large decrease of LH, we postulate that DXM lowers LH release and therefore indirectly lowers the T synthesis and/or release.  相似文献

The possible relationship between the circadian rhythm of blood levels of melatonin and corticosterone in ring dove (Streptopelia risoria) subjected to both immobilization stress and immobilization stress plus dexamethasone treatment were studied. The results show changes in the circadian rhythm of melatonin, with increased day-time levels in situations of stress accompanied by increased corticosterone levels. The highest blood melatonin levels over the 24 h of the study were obtained when the animals were treated with dexamethasone and then subjected to stress. Given the antioxidant role of melatonin, our results support the idea of melatonin-corticosterone coupling with the possibility that melatonin released in situations of stress counteracts the adverse effects of glucocorticoids on the organism.  相似文献

The locomotor activity of the millipede Glyphiulus cavernicolus (Spirostreptida), which occupies the deeper recesses of a cave, was monitored in light-dark (LD) cycles (12h light and 12h darkness), constant darkness (DD), and constant light (LL) conditions. These millipedes live inside the cave and are apparently never exposed to any periodic factors of the environment such as light-dark, temperature, and humidity cycles. The activity of a considerable fraction of these millipedes was found to show circadian rhythm, which entrained to a 12:12 LD cycle with maximum activity during the dark phase of the LD cycle. Under constant darkness (DD), 56.5% of the millipedes (n = 23) showed circadian rhythms, with average free-running period of 25.7h ± 3.3h (mean ± SD, range 22.3h to 35.0h). The remaining 43.5% of the millipedes, however, did not show any clear-cut rhythm. Under DD conditions following an exposure to LD cycles, 66.7% (n = 9) showed faint circadian rhythm, with average free-running period of 24.0h ± 0.8h (mean ± SD, range 22.9h to 25.2h). Under constant light (LL) conditions, only 2 millipedes of 11 showed free-running rhythms, with average period length of 33.3h ± 1.3h. The results suggest that these cave-dwelling millipedes still possess the capacity to measure time and respond to light and dark situations. Prostacyclin (PGI2) or its stable metabolite, 6-keto-PGF1 alpha (1-5 micrograms) in 2.5 microliter 0.05 M phosphate buffer (pH 7.4), was injected into the third ventricle (3 V) of ovariectomized (OVX), freely moving rats. Control animals received 2.5 microliter of buffer. In the initial experiments a control blood sample was taken and then the PGI2 was injected and frequent samples taken thereafter. With this protocol injection of 2 micrograms of PGI2 produced a significant decrease in mean plasma LH only at 60 min after its injection (p less than .05), while the higher dose (5 micrograms) decreased plasma LH concentrations at 30 and 60 min (p less than .01 and p less than .001, respectively). In subsequent experiments, blood was removed from indwelling external jugular vein cannulae every 5-6 min during 2 hours and plasma LH and PRL levels were determined by radioimmunoassay. LH pulses were monitored and several parameters of LH pulsation were calculated during the hour before and after injection of phosphate buffer, PGI2 or 6-keto-PGF1 alpha. Intraventricular injection of phosphate buffer failed to modify the characteristic pulsatile release of LH and did not alter plasma PRL levels. The amplitude of LH pulses was significantly reduced by PGI2 and the inhibitory effect was dose-related. Even a dose of 1 microgram produced a significant reduction in pulse height and the response was graded with maximal reduction occurring with the 5 microgram dose which essentially abolished the LH pulses. Following the microinjection of 6-keto-PGF1 alpha, no significant changes were observed in plasma LH values and the pulses of the hormone. Five micrograms PGI2 considerably elevated plasma PRL values during the 20-25 min following its 3V injection, whereas the same dose of 6-keto-PGF1 alpha produced only a very slight stimulatory effect. Since PGI2 had no effect to alter LH release by cultured pituitary cells in vitro, it is concluded that PGI2 can act on structures near the 3V to inhibit pulsatile release of LHRH.  相似文献

Simultaneous measurements of plasma LH, body temperature, and locomotor activity were made in laying turkey hens and are reported. Blood samples were remotely collected using a jugular cannula system, and body temperature and locomotor activity were remotely monitored using a radiotelemetry system in freely moving laying turkeys. Under a photoschedule of 14L:10D, the period for preovulatory surges of LH was 25.7 +/- 0.4 h while the periods for peak body temperature and onset of sustained locomotor activity were 24.9 +/- 0.4 and 25.7 +/- 0.5 h, respectively. During exposure to constant light, the periods for preovulatory surges of LH, peak body temperature, and onset of sustained locomotor activity increased to 27.9 +/- 0.9, 26.7 +/- 0.7, and 27.4 +/- 0.7 h, respectively. With the 14L:10D photoschedule, initiation of LH surges was restricted to the scotophase, but after 8 days of constant light, initiation of LH surges had dispersed throughout the 24-h subjective day and night. With constant light, the amplitude of the peak body temperature rhythm decreased, while the duration of the locomotor activity rhythm became broadened and, in some birds, disorganized. Peak body temperature and onset of locomotor activity rhythms and LH surges did not coincide, even though peak body temperature, onset of locomotor activity, and LH surges had similar periods. It is concluded that 1) the photoschedule influences the periods of the LH surge, peak body temperature, and onset of locomotor activity; and 2) a specific or direct relationship between the rhythms of LH surge, body temperature, and locomotor activity remains to be determined in laying turkey hens.  相似文献

Our previous work has suggested that glucocorticoid pretreatment suppresses the enhanced responsiveness to GnRH seen in serum LH 12 h after castration. By contrast, serum FSH continues to show the castration-induced hypersensitivity to GnRH. Our attempts to replicate this LH suppression in static pituitary culture in vitro were not successful. This suggested to us the possibility that corticoids in vivo might be preventing castration-induced increases in pituitary GnRH receptor levels. We tested this at 24 h post-castration and, in fact, corticoids did not suppress the increase in GnRH receptors. In addition to the aforementioned effects of corticoids, we have seen that cortisol reverses the castration-induced drop in pituitary FSH content. It does this for 7 days post-castration, even though it no longer has an effect in suppressing serum LH. Thus, our accumulated data reveal that glucocorticoids have a differential effect on LH and FSH synthesis and secretion. Further studies are needed to clarify the site(s) of action of glucocorticoids in gonadotropin secretion and synthesis. Glucocorticoids may well prove to be a key in unlocking the mystery of the mechanism of differential control of regulation of LH and FSH.  相似文献

To investigate daily feeding rhythms in zebrafish, the authors have developed a new self-feeding system with an infrared photocell acting as a food-demand sensor, which lets small-size fish such as zebrafish trigger a self-feeder. In this paper, the authors used eight groups of 20 fish. Locomotor activity rhythms were also investigated by means of infrared sensors. Under a 12?h:12?h light (L)-dark (D) cycle, zebrafish showed a clear nocturnal feeding pattern (88.0% of the total daily food-demands occurring in the dark phase), concentrated during the last 4?h of the dark phase. In contrast, locomotor activity was mostly diurnal (88.2% of total daily activity occurring in the light phase). Moreover, both feeding and locomotor rhythms were endogenously driven, as they persisted under free-running conditions. The average period length (τ) of the locomotor and feeding rhythms was shorter (τ?=?22.9?h) and longer (τ?=?24.6?h) than 24?h, respectively. During the time that food availability was restricted, fish could only feed during ZT0-ZT12 or ZT12-ZT16. This resulted in feeding activity being significantly modified according to feeding time, whereas the locomotor activity pattern remained synchronized to the LD cycle and did not change during this trial. These findings revealed an independent phasing between locomotor and feeding activities (which were mostly nocturnal or diurnal, respectively), thus supporting the concept of multioscillatory control of circadian rhythmicity in zebrafish.  相似文献
