It is widely accepted that amphotericin B (AmB) together with sterol makes a mixed molecular assemblage in phospholipid membrane. By adding AmB to lipids prior to preparation of large unilamellar vesicles (LUV), we directly measured the effect of cholesterol on assemblage formation by AmB without a step of drug's binding to phospholipid bilayers. Potassium ion flux assays based on 31P-nuclear magnetic resonance (NMR) clearly demonstrated that cholesterol markedly inhibits ion permeability induced by membrane-bound AmB. This could be accounted for by a membrane-thickening effect of cholesterol since AmB actions are known to be markedly affected by the thickness of membrane. Upon addition of AmB to an LUV suspension, the ion flux gradually increased with increasing molar ratios of cholesterol up to 20 mol%. These biphasic effects of cholesterol could be accounted for, at least in part, by the ordering effect of cholesterol.  相似文献

Amphotericin B (AmB) is the most widely used antibiotic to treat systemic fungal infections. However, the molecular mechanism of its activity is still not completely understood. In the present work we have used FTIR spectroscopy to investigate the conformational state of the aliphatic chains of DPPC liposomes using the 2850 cm(-1) band, associated with the methylene symmetric stretching mode. The liposomes were either binary mixtures of the lipid with AmB, cholesterol or ergosterol, or ternary systems of these constituents. The two sterols contribute to an ordering of the aliphatic chains of the lipid, this ordering being slightly more important with ergosterol. In the gel state, AmB does not change the conformational order of DPPC even at high concentration. In the fluid phase, however, the drug clearly structures its lipid environment. Our results show that AmB can initiate a redistribution of the ergosterol in the plane of the membrane, but not of the cholesterol molecules, which might constitute an additional mechanism to explain the activity of the antibiotic.  相似文献

摘要 目的:建立高效液相色谱(HPLC)-电喷雾检测器(CAD)法测定两性霉素B脂质体(L-AMB)中氢化大豆磷脂酰胆碱(HSPC)和磷脂二硬酯酰磷脂酰甘油(DSPG)含量的方法。方法:采用Venusil XBP C 8(4.6 mm×250 mm,5 μm)色谱柱,柱温:30℃;以甲醇-水作为流动相;喷雾压力:56.4 psi,喷雾温度:55℃;梯度洗脱,流速1.0 mL?min-1结果:L-AMB各磷脂在 5~60 mg?mL-1内与峰面积呈良好的线性关系。HSPC平均回收率为100.06%,平均RSD为0.42;DSPG平均回收率100.03%,平均RSD为0.55。按HPLC-CAD 色谱条件测定L-AMB中HSPC、 DSPG含量分别为1497.66 mg?L-1、4710.93 mg?L-1,且无溶血磷脂检出。采用HPLC-ELSD 法验证,HSPC、 DSPG含量分别为1495.28 mg?L-1、4712.71 mg?L-1, 两种方法测定HSPC、 DSPG含量基本一致。结论:HPLC-CAD 测定L-AMB中HSPC、DSPG含量操作简单、准确,可准确测定L-AMB 中脂质体的质量。  相似文献

Recent studies surprisingly show that dietary inorganic nitrate, abundant in vegetables, can be metabolized in vivo to form nitrite and then bioactive nitric oxide. A reduction in blood pressure was recently noted in healthy volunteers after dietary supplementation with nitrate; an effect consistent with formation of vasodilatory nitric oxide. Oral bacteria have been suggested to play a role in bioactivation of nitrate by first reducing it to the more reactive anion nitrite. In a cross-over designed study in seven healthy volunteers we examined the effects of a commercially available chlorhexidine-containing antibacterial mouthwash on salivary and plasma levels of nitrite measured after an oral intake of sodium nitrate (10 mg/kg dissolved in water). In the control situation the salivary and plasma levels of nitrate and nitrite increased greatly after the nitrate load. Rinsing the mouth with the antibacterial mouthwash prior to the nitrate load had no effect on nitrate accumulation in saliva or plasma but abolished its conversion to nitrite in saliva and markedly attenuated the rise in plasma nitrite. We conclude that the acute increase in plasma nitrite seen after a nitrate load is critically dependent on nitrate reduction in the oral cavity by commensal bacteria. The removal of these bacteria with an antibacterial mouthwash will very likely attenuate the NO-dependent biological effects of dietary nitrate.  相似文献

The effects of amphotericin B upon the organization and dynamics of multibilayer membranes of dimyristoylphosphatidylcholine (DMPC) were investigated by means of 2H-NMR. At high amphotericin B concentrations (30 mol% with respect to the lipid) and at temperatures above 25°C, DMPC experiences two different environments which are in slow exchange on the 2H-NMR time scale. In one of these, the lipid is immobilized by the antibiotic, in a molar ratio of approximately 1:1, whereas the lipid unsequestered by amphotericin B is more ordered than in its pure state. This ordering effect is perceived at relatively low antibiotic doses (4%). The local lipid order, and the relative percentage, of sequestered DMPC, are temperature-independent (up to 65°C), whereas the ordering of the unsequestered lipid domain is not. The perturbation induced by amphotericin B is manifest similarly at the edges as well as in the center of the bilayer. Antibiotic addition leads to large decreases in the transverse relaxation time, T2, of the labelled lipid, but not in the spin-lattice relaxation time, T1. This indicates an increased density of slow motional modes and little change in rapid motions.  相似文献

Resonance energy transfer (RET) between anthrylvinyl-labeled phosphatidylcholine (AV-PC) or phosphatidylglycerol (AV-PG) as donors and the heme groups of cytochrome c (cyt c) as acceptors was examined in PC/PG model membranes containing 10, 20 or 40 mol% PG with an emphasis on evaluating lipid demixing caused by this protein. The differences between AV-PC and AV-PG RET profiles observed at PG content 10 mol% were attributed to cyt c ability to produce segregation of acidic lipids into lateral domains. The radius of lipid domains recovered using Monte-Carlo simulation approach was found not to exceed 4 nm pointing to the local character of cyt c-induced lipid demixing. Increase of the membrane PG content to 20 or 40 mol% resulted in domain dissipation as evidenced by the absence of any RET enhancement while recruiting AV-PG instead of AV-PC.  相似文献

The issue of whether d-tubocurarine, the classical acetylcholine receptor inhibitor, inhibits the receptor by a competitive or noncompetitive mechanism has long been controversial. d-Tubocurarine, in this study, has been found to be both a competitive (KC = 120 nM) and a noncompetitive (KNC = 4 microM) inhibitor of receptor-mediated ion flux at zero transmembrane voltage in membrane vesicles prepared from Electrophorus electricus electroplax. A spectrophotometric stopped-flow method, based on fluorescence quenching of entrapped anthracene-1,5-disulfonic acid by Cs+, was used to measure both the rate coefficient of ion flux prior to receptor inactivation (desensitization) and the rate coefficient of the rapid inactivation process. Inhibition by d-tubocurarine of the initial rate of ion flux decreased with increasing acetylcholine concentration, consistent with competitive inhibition, but the inhibition by micromolar concentrations of d-tubocurarine could not be overcome with saturating concentrations of acetylcholine, consistent with noncompetitive inhibition. A minimum mechanism is proposed in which d-tubocurarine competes for one of the two acetylcholine activating sites and also binds to a noncompetitive site. The present data do not distinguish between one or two competitive sites, although one successfully accounts for all of the data. By variation of the acetylcholine concentration, the two types of sites could be studied in isolation. Binding of d-tubocurarine to the noncompetitive site does not change the rate of rapid receptor inactivation, whereas binding of d-tubocurarine to the competitive site decreases the rate of rapid inactivation by displacing acetylcholine, in agreement with the observation that d-tubocurarine does not inactivate (desensitize) the E. electricus receptor by itself.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

Summary Microvillar membrane vesicle preparations with varying content of aminopeptidase N were prepared from enterocytes of the pig small intestine. Postembedding immunogold labelling of aminopeptidase N was performed on these glutaraldehyde/paraformaldehyde-fixed, osmium tetroxide-treated and Epon-embedded microvillar membrane vesicles. The number of gold particles per micrometre microvillar membrane (labelling intensity) was calculated and compared to the corresponding enzymatic activity. A very close relationship was found between labelling intensity and aminopeptidase N activity, demonstrating that postembedding immunogold labelling can be used quantitatively.  相似文献

The meso-tetra(α,α,α,α(o-pivalamidophenyl))porphinato iron-mono(1-lauryl-2-methylimidazole) complex embedded in the bilayer of dimyristoylphosphatidylcholine (liposomal heme) binds molecular oxygen reversibly at pH 7 and 37°C. Orientation of the iron porphyrin complex in the phospholipid bilayer was studied by electric birefringence and dichroism. It was observed that both the phospholipid bibilayer of liposome and the porphyrin plane are oriented nearly in parallel to the electric field. Therefore the angle between the porphyrin plane and the bilayer is considered to be practically small.  相似文献

The microbial community structure of two biofilters used for the oxidation of methane and organic trace gases generated in landfills was analysed by phospholipid fatty acid composition. Community structure varied with biofilter depth, reflecting varying conditions of substrate supply as well as of organic carbon content, nutrient status and osmotic stress determined by the different materials used for the individual biofilter layers. Both biofilters were dominated by type II methanotrophs. In the biofilter charged with landfill gas containing significant amounts of trace organics, fatty acid 18:1omega7c constituted 87% of the methanotrophic PLFA, while the recognised signature fatty acids 16:1omega8 and 18:1omega8, which were well represented in the other biofilter, were entirely absent. This indicates the development of a highly specific methanotrophic population, presumably as a result of the adaption to continuous organic trace gas exposure.  相似文献

This paper continues the studies of vesicle-bound flavins (‘anisotropic flavin chemistry’). It is possible to anchor the flavin nucleus in various modes within the lipid/water interface by means of long aliphatic chains and using different saturated lipids, thereby mimicking the specific binding of the coenzyme to the apoprotein in flavoproteins. Based on absorption spectroscopy and EPR spectroscopy studies we explored the rotational mobility and the microenvironment of membrane-bound amphiflavin radicals. N(5)-unsubstiluted amphiflavin radicals exhibit a similarly high disproportionation constant as known from isotropic flavin chemistry. However, reasonable stabilization of the radical was achieved by introduction of an alkyl group in position 5 in the reduced state prior to the one-electron oxidation. Adopting the fine structure of the corresponding EPR spectra as assay for the mobility of the semiquinone, we determined rotational relaxation times ranging from 60 ns in the crystalline state down to 10 or 15 ns in the liquid-crystalline state of the membrane. The solvatochromic effect shown by absorption spectra of the membrane-bound flavin radicals reflects a dielectric constant of the microenvironment of ? = 30–40, corresponding to the lipid/water interface region. The results obtained in this study are consistent with those obtained previously, from fluorescence analyses, supporting our former conclusions.  相似文献

Accurate knowledge of the response of root turnover to a changing climate is needed to predict growth and produce carbon cycle models. A soil warming system and shading were used to vary soil temperature and received radiation independently in a temperate grassland dominated by Holcus lanatus L. Minirhizotrons allowed root growth and turnover to be examined non‐destructively. In two short‐term (8 week) experiments, root responses to temperature were seasonally distinct. Root number increased when heating was applied during spring, but root death increased during autumnal heating. An experiment lasting 12 months demonstrated that any positive response to temperature was short‐lived and that over a full growing season, soil warming led to a reduction in root number and mass due to increased root death during autumn and winter. Root respiration was also insensitive to soil temperature over much of the year. In contrast, root growth was strongly affected by incident radiation. Root biomass, length, birth rate, number and turnover were all reduced by shading. Photosynthesis in H. lanatus exhibited some acclimation to shading, but assimilation rates at growth irradiance were still lower in shaded plants. The negative effects of shading and soil warming on roots were additive. Comparison of root data with environmental measurements demonstrated a number of positive relationships with photosynthetically active radiation, but not with soil temperature. This was true both across the entire data set and within a shade treatment. These results demonstrate that root growth is unlikely to be directly affected by increased soil temperatures as a result of global warming, at least in temperate areas, and that predictions of net primary productivity should not be based on a positive root growth response to temperature.  相似文献

Phosphodiesterase 3B (PDE3B), a major PDE isoform in adipocytes, plays a pivotal role in the anti-lipolytic action of insulin. Insulin phosphorylates and activates PDE3B in a phosphatidylinositol 3-kinase-dependent manner. We identified a new 50 kDa protein that is phosphorylated by insulin and is co-immunoprecipitated with PDE3B by anti-PDE3B antibodies in rat adipocytes. The insulin-induced phosphorylation of the 50 kDa protein was also detected in a cell free system against the N-terminal and the catalytic regions, which are more than 700 amino acids apart recognize the 50 kDa protein, suggesting that it is not a proteolytic product, but an associated protein with PDE3B. Phosphoamino acid analysis indicated that both serine and threonine residues in the 50 kDa protein were phosphorylated, but only serine residues in PDE3B were phosphorylated. Therefore, it appears likely that this is a new protein which is associated with PDE3B.  相似文献

Chloroplast DNA polymorphisms were analysed to infer the post-glacial history of Nothofagus nervosa (Phil.) Dim. et Mil., a species endemic to South American Temperate Forests. Two hypotheses were postulated to explain the current distribution of the species in relation to the refugia proposed after palynological studies and the time elapsed since Last Glacial Maximum. If refugia were located only in the Coastal Mountains, as pollen records suggests, long-distance dispersal events should be invoked to explain the current occurrence of the species in southern latitudes. The alternative hypothesis is the existence of cryptic refugia in southern latitudes. A total of 26 populations covering the entire geographical range of the species in Chile and Argentina were analysed through PCR–RFLP. Five haplotypes were identified, and a very low intrapopulation variation was observed together with a high gene differentiation (G ST=0.93). The haplotypes showed a highly structured geographic distribution, separating populations located in the Pacific Coastal Mountains from those coming from the easterly located Andes Mountains. Moreover, among the populations of the Andes Mountains a north–south variation in the distribution of haplotypes was found. The results strongly suggest the persistence of the species in several ice-age refugia. The possible location of such refugia is discussed in combination with the available palynological data.  相似文献
