The occurrence of culturable yeasts in glacial meltwater from the Frías, Casta?o Overo and Río Manso glaciers, located on Mount Tronador in the Nahuel Huapi National Park (Northwestern Patagonia, Argentina) is presented. Subsurface water samples were filtered for colony counting and yeast isolation. The total yeast count ranged between 6 and 360 CFU L(-1). Physiologic and molecular methods were employed to identify 86 yeast isolates. In agreement with yeast diversity data from studies for Antarctic and Alpine glaciers, the genera Cryptococcus, Leucosporidiella, Dioszegia, Rhodotorula, Rhodosporidium, Mrakia, Sporobolomyces, Udeniomyces and Candida were found. Cryptococcus and Leucosporidiella accounted for 50% and 20% of the total number of strains, respectively. Among 21 identified yeast species, Cryptococcus sp. 1 and Leucosporidiella fragaria were the most frequent. The typically psychrophilic Mrakia yeast strain and three new yeast species, yet to be described, were also isolated. All yeast strains were able to grow at 5, 10, and 15 degrees C. Among yeast strains expressing extracellular enzymatic activity, higher proteolytic and lipolytic activities were obtained at 4 degrees C than at 20 degrees C.  相似文献

Fifteen aquatic environments (lakes, lagoons and rivers) of glacial origin in the northern Andean Patagonia (Argentina) were surveyed for the occurrence of red yeasts. Subsurface water samples were filtered and used for colony counting and yeast isolation. A preliminary quantitative analysis indicated that total yeast counts ranged between 0 and 250 cells l−1. A polyphasic approach including physiological and molecular methods was used for the identification of 64 carotenogenic yeast strains. The molecular characterisation of the isolates was based on the mini/microsatellite-primed PCR technique (MSP-PCR) employing the (GTG)5 and the M13 primers. Comparison of representative fingerprints of each group with those of the type strains of pigmented yeasts allowed the expeditious identification of 87.5% isolates. The sequence analysis of the D1/D2 domains of the 26S rDNA was employed to confirm identifications and in the characterization of the unidentified MSP-PCR groups. Teleomorphic yeast species were detected by performing sexual compatibility assays. The isolates corresponded to 6 genera and 15 yeast species, including four new yeast species of the genera Cryptococcus (1), Rhodotorula (1) and Sporobolomyces (2). Rhodotorula mucilaginosa was found in the majority of the samples and represented ca. 50% of the total number of isolates. However, this yeast was not detected in aquatic environments with very low anthropic influence. Other frequent yeast isolates were teleomorphic yeast species of Rhodosporidium babjevae, R. kratochvilovae and Sporidiobolus salmonicolor. This study represents the first report on red yeast occurrence and biodiversity in northwestern Patagonia. Seven strains with similar characteristics from the laboratory collection of yeasts isolated from forest substrates collected in the central part of European Russia corresponded to none of the known yeast species. Based on the study of their life cycle, physiological characteristics, and the nucleotide composition of their DNA and taking into account the data of PCR analysis with universal primers, the strains were ascribed to a new psychrophilic yeast species, Mrakia curviuscula sp. nov.  相似文献

Mechanisms of survival during the Pleistocene glaciation periods have been studied for more than a century. Until now, molecular studies that confirmed animal survival on Alpine nunataks, that is, ice‐free summits surrounded by glaciers, were restricted to peripheral areas. Here, we search for molecular signatures of inner‐Alpine survival of the narrow endemic and putatively parthenogenetic Alpine jumping bristletail Machilis pallida combining mitochondrial and AFLP data from its three known populations. The mitochondrial data indicate survival on both peripheral and central nunataks, the latter suggesting that refugia in the centre of the Alpine main ridge were more widespread than previously recognized. Incongruences between mitochondrial and AFLP patterns suggest a complex evolutionary history of the species and may be explained via parallel fixation of parthenogenesis of different origins during the last glacial maximum. We suggest that the inferred parthenogenesis may have been essential for central nunatak survival, but may pose a serious threat for M. pallida in consideration of the present climatic changes.  相似文献

Many mountain ranges have been strongly glaciated during the Quaternary ice ages, and the locations of glacial refugia of mountain plants have been debated for a long time. A series of detailed molecular studies, investigating intraspecific genetic variation of mountain plants in the European Alps, now allows for a first synopsis. A comparison of the phylogeographic patterns with geological and palaeoenvironmental data demonstrates that glacial refugia were located along the southwestern, southern, eastern and northern border of the Alps. Additional glacial refugia were present in central Alpine areas, where high-elevation plants survived the last glaciation on ice-free mountain tops. The observed intraspecific phylogeographies suggest general patterns of glacial survival, which conform to well-known centres of Alpine species diversity and endemism. This implies that evolutionary or biogeographic processes induced by climatic fluctuations act on gene and species diversity in a similar way.  相似文献

We isolated 99 yeast strains, including 40 red yeasts, from benthic animals and sediments collected from the deep-sea floor in various areas in the northwest Pacific Ocean. Comparing the yeast isolates from animals and sediments collected from shallow locations, the proportion of red yeasts differed considerably, comprising 81.5% and 10.6% of the isolates from animals and sediments, respectively. All of the red yeast isolates belonged to the genera Rhodotorula and Sporobolomyces. On the basis of morphological and physiological characteristics, the isolates were identified as R. aurantiaca, R. glutinis, R. minuta and R. mucilaginosa of the genus Rhodotorula, and S. salmonicolor and S. shibatanus of the genus Sporobolomyces. Only R. glutinis and R. mucilaginosa were isolated from sediments. All of the others were isolated from animal sources. Phylogenetic analyses based on internal transcribed spacer (ITS) regions and 5.8S rRNA gene sequences allowed us to establish the precise taxonomic placement of each of the isolates and thereby investigate the intraspecific relationships among the isolates. Twenty-two strains identified as members of R. glutinis, which showed a wide distribution in the deep-sea, and five isolates identified as R. minuta, which were isolated only from benthic animals, showed substantial heterogeneity within the species. The isolates phenotypically identified as Sporobolomyces species and R. mucilaginosa phylogenetically occupied the placements corresponding to these species. Some strains assigned to known species on the basis of phenotypic features should be regarded as new species as suggested by the results of molecular analysis.  相似文献

【目的】揭示乌鲁木齐河源天山1号冰川表面冰尘(CS)和底部沉积层(DS)可培养酵母菌系统发育类群及其结构组成差异,分析低温酵母菌代表菌株之间的生态、生理生化特性。【方法】利用4种培养基分离天山1号冰川可培养酵母菌,采用ITS基因序列分析确定菌种的系统进化地位。对分离菌株的最适生长温度、耐盐性和产酶等生态、生理学特性进行分析。【结果】从冰尘和底部沉积层中共分离出152株酵母菌菌株,通过ITSrRNA基因序列的NCBI比对和Rep-PCR指纹分型,结果表明酵母菌类群包括担子菌门(Basidiomycota)和子囊菌(Ascomycota),分属于14个属26种,其中担子菌门柄锈菌亚门(Pucciniomycotina)88株、伞菌亚门(Agariomycotina)24株,子囊菌门40株,冰川广布酵母菌Vishniacozyma victoriae为优势菌株(占比21.84%)。17种酵母的最适生长温度为15°C、2种为10°C、6种为20°C。25株代表酵母菌株产酶分析显示,产脂肪酶、淀粉酶、蛋白酶菌株分别为11株、11株、5株,6株3种酶都不产。【结论】天山1号冰川冰尘及底部沉积层可培养低温酵母系统发育类群结构存在差异,产低温酶活性高、稳定性好,为今后冰川低温酵母菌的研究提供有价值的数据支持。  相似文献

嗜冷产甲烷菌对于自然界的碳素循环具有非常重要的意义,近年来引起了国内外学者的广泛关注.利用嗜冷产甲烷菌实现低温厌氧生物处理过程,可从本质上突破低温厌氧工艺的技术瓶颈,进而大大拓展厌氧生物处理技术的应用范围并降低废水处理的成本.本文针对研究者广泛关注的热点问题,从分离培养及生理生化特性、适冷机制和分子生物学研究几个方面,对嗜冷产甲烷菌的研究进展进行了全面的综述,并对其在低温厌氧生物处理技术中的应用前景进行了分析和展望.  相似文献

从连云港海域、港口、远洋捕捞船及鱼市等地采集的海水和各类海鱼、贝类等样品中分离到217株产蛋白酶的细菌,并从中得到1株产低温碱性蛋白酶的海洋适冷细菌—SY。研究表明,该菌株最适生长温度和最适产酶温度均在15℃左右,0℃下仍可生长;具有一定的耐盐性和嗜盐性,无盐条件不能生长,在3%的NaCl盐浓度时,生长达到最高峰;最适生长和最适产酶pH均为8.0;SY菌所产的蛋白酶可能为一种丝氨酸蛋白酶,酶最适作用温度为50℃,最适作用pH为9.0;酶的热稳定性差,50℃保温20min,酶活下降40%。  相似文献

A total of 436 milk samples from non-infected and 80 from infected quarters were investigated: 24.5% of the samples collected from non-infected and 55% of those collected from infected quarters were positive. Normal milk yielded not less than 16 different species and among them many potentially pathogenic yeast species such as C. parapsilosis, C. guilliermondii, C. tropicalis, C. glabrata and T. asahii, all five able to grow at 40 ° C. In contrast, the yeasts isolated from infected quarters were from 3 species: C. kefyr, C. catenulata and C. lambica, which were also among the yeasts species recovered from normal milk. Among the three species, only one i.e. Candida kefyr is able to grow above 40 ° C and from there can be considered as potentially pathogenic, even if bacterial association is necessary to cause mastitis.  相似文献

In the framework of initiatives, aiming at a better definition of biodiversity in microorganisms and of its role in ecosystem function, the biology of yeasts is revisited with particular emphasis on their ecology and classification. The limited number of different species isolated from, as well as the exceedingly low yeast cell counts in, most natural environments are discussed and possible explanations presented. The impact of the use of molecular taxonomic techniques of classification on the extent of biodiversity within yeasts is also debated, as well as the possible repercussions of more aggressive pre-isolation treatments of samples.  相似文献

Aims:  Isolation and identification of yeasts converting xylose to ethanol.
Methods and Results:  A total of 374 yeasts were isolated from a variety of rotten fruits and barks of trees. Out of these, 27 yeast strains were able to assimilate xylose and produce 0·12–0·38 g of ethanol per gram of xylose. Based on phylogenetic analysis of D1/D2 domain sequence of LSU (Large Subunit) rRNA gene and phenotypic characteristics the ethanol-producing strains were identified as member(s) of the genera Pichia, Candida , Kluyveromyces, Issatchenkia, Zygosacchraomyces , Clavispora, Debaryomyces , Metschnikowia , Rhodotorula and Cryptococcus.
Conclusion:  Yeast strains producing ethanol from xylose have been isolated from a variety of rotten fruits and barks of trees and identified.
Significance and Impact of the Study:  Environmental isolates of yeasts which could convert xylose to ethanol could form the basis for bio-fuel production and proper utilization of xylan rich agricultural and forest wastes.  相似文献

Fifty individuals of the endemic Alpine salamander, Salamandra atra, representing 13 populations throughout the range of the two currently recognized subspecies, atra and aurorae, were examined for sequence variation in a large portion (1050 bp) of the mitochondrial cytochrome b gene. We revealed a large number of mitochondrial DNA (mtDNA) haplotypes (10). Interpopulation sequence divergence was very low, ranging from 0 to 3.1%. The relationships among haplotypes were poorly resolved. The divergence time estimate between several mtDNA haplotypes suggested a pre-Pleistocene differentiation approximately 3 million years ago. Moreover, the impact of the Pleistocene glaciations on the phylogeographical patterns appears to have been secondary, although a somewhat reduced genetic variability was found in populations living in areas that were directly affected by the glaciation.  相似文献

To elucidate the phylogenetic relationships of Alpine voles comprising the Microtus multiplex complex and related species the mitochondrial control region ( CR ) was employed as a genetic marker. Forty specimens were analysed representing the taxa Microtus liechtensteini , Microtus multiplex , and Microtus bavaricus (samples from 11 geographic regions) as well as specimens of Microtus subterraneus , Microtus tatricus , Microtus arvalis , Microtus agrestis , Microtus oeconomus , and Chionomys nivalis. The haplotypes from Tuscany and the Swiss canton of Valais can be ascribed to M. multiplex , whereas the haplotypes isolated from the geographic samples from Croatia, Slovenia, Carinthia, Styria, East Tyrol, and South Tyrol represent M. liechtensteini . The molecular data indicate that the distribution range of M. liechtensteini extends further to the north (Upper Styria) than has been assumed earlier. The haplotypes of M. bavaricus , together with the populations from North Tyrol, form a cluster clearly separated from M. liechtensteini . This result suggests that the North Tyrolean samples belong to M. bavaricus , a species thought to be extinct. Microtus tatricus splits off at the basis of the multiplex complex. Our sequence data do not support a close relationship between M. bavaricus and M. tatricus . The phylogenetic relationships deduced from the sequence data favour a hypothesis assuming three glacial refugia, in which M. multiplex , M. liechtensteini and M. bavaricus , respectively, survived the last one or two Alpine glaciations.  相似文献

One hundred and seventy yeast strains belonging to 14 genera and 29 species were isolated from 112 water samples of the river Danube in the area of Bratislava. The samples were collected through the year from April to March.Saccharomyces cerevisiae, Candida maltosa, Aureobasidium pullulans, Cystofilobasidium capitatum, Rhodotorula glutinis, Geotrichum candidum, and Candida krusei were the most frequent. The basidiomycetous yeasts and yeast-like organisms with oxidative metabolism were present in approximately equal numbers to those with fermentative metabolism. Saccharomyces cerevisiae was the dominant yeast and was isolated from 50% of all samples examined and represented approximately one quarter of the yeast community.Yeast densities ranged from 100 to 21,100 CFU per litre. The highest population density was observed in October. Cryptococcus albidus, Saccharomyces cerevisiae, Rhodotorula glutinis, and Aureobasidium pullulans formed the main part of the yeast population in this month.  相似文献

AIMS: The present study was conducted to screen for psychrophilic yeasts that are able to degrade pectin compounds at low temperature, and to examine the cold-active pectinolytic enzymes produced by the isolated psychrophilic yeasts. METHODS AND RESULTS: Psychrophilic yeasts, which grow on pectin as a sole carbon source, pectinolytic-psychrophilic yeast (PPY) strains PPY-3, 4, 5 and 6, were isolated from soil from Abashiri (Hokkaido, Japan). The sequences of 28S rDNA D1/D2 of strains PPY-3 and 4 indicated a taxonomic affiliation to Cryptococcus cylindricus and Mrakia frigida, respectively, strains PPY-5 and 6 belonged to Cystofilobasidium capitatum. The isolated strains were able to grow on pectin at below 5 degrees C, and showed the activities of several cold-active pectinolytic enzymes. CONCLUSION: The findings of this study indicate the possibility that the isolated strains produce novel pectinolytic enzymes that are able to degrade pectin compounds at low temperature. Significance and Impact of the Study: It is possible that the cold-active pectinolytic enzymes from the isolated strains can be applied to the food industry, e.g. the clarification of fruit juice below 5 degrees C.  相似文献

A key aspect for understanding the biological and biochemical environment of subglacial waters, on Earth or other planets and moons in the Solar system, is the analysis of material embedded in or underneath icy layers on the surface. In particular the Antarctic lakes (most prominently Lake Vostok) but also the icy crust of Jupiter’s moon Europa or the polar caps of Mars require such investigation. One possible technique to penetrate thick ice layers with small and reliable probes is by melting, which does not require the heavy, complex and expensive equipment of a drilling rig. While melting probes have successfully been used for terrestrial applications e.g. in Antarctic ice, their performance in vacuum is different and theory needs confirmation by tests. Thus, a vacuum chamber has been used to perform a series of melting tests in cold (liquid nitrogen cooled) water ice samples. The feasibility of the method was demonstrated and the energy demand for a space mission could be estimated. Due to the high energy demand in case of extraterrestrial application (e.g. Europa or polar caps of Mars), only heating with radioactive isotopes seems feasible for reaching greater depths. The necessary power is driven by the desired penetration velocity (approximately linearly) and the dimensions of the probe (proportional to the cross section). In comparison to traditional drilling techniques the application of a melting probe for exploration of Antarctic lakes offers the advantage that biological contamination is minimized, since the Probe can be sterilized and the melting channel freezes immediately after the probe’s passage, inhibiting exchange with the surface layers and the atmosphere. In order to understand the physical and chemical nature of the ice layers, as well as for analysing the underlying water body, a melting probe needs to be equipped with a suite of scientific instruments that are capable of e.g. determining the chemical and isotopic composition of the embedded or dissolved materials.  相似文献

Several yeast cultures belonging to five non-Saccharomyces species associated with wine-making were evaluated for their oenological properties. Results showed that Candida stellata and Torulaspora delbrueckii could positively affect the taste and flavour of alcoholic beverages. Apiculate yeasts exhibited large amounts of negative byproducts, particularly ethyl acetate. Nevertheless, Kloeckera apiculata showed a significantly negative correlation between either acetic acid and ethyl acetate formation and ethanol production. Selected non-Saccharomyces yeast cultures could be applied profitably in wine-making for optimization of wine bouquet using new fermentation technologies.  相似文献
