S ummary : Japanese quail chicks died within a few days when fed diet containing raw navy beans, whereas a similar diet containing autoclaved beans supported normal growth. The diet containing raw beans was nonlethal to germfree quail chicks, but did not support such good growth as the heated bean diet. The effects of associating germfree quail on the raw bean diet with various combinations of 149 pure cultures of bacteria isolated from the intestines of ordinary quail have been studied. Strains of Escherichia coli were shown to be responsible for the lethal effects of the raw bean diet.  相似文献

Coenzyme Q10 (CoQ10) is a promising agent for neuroprotection in neurodegenerative diseases. We tested the effects of various doses of two formulations of CoQ10 in food and found that administration in the diet resulted in significant protection against loss of dopamine (DA), which was accompanied by a marked increase in plasma concentrations of CoQ10. We further investigated the neuroprotective effects of CoQ10, reduced CoQ10 (ubiquinol), and CoQ10 emulsions in the (MPTP) model of Parkinson's disease (PD). We found neuroprotection against MPTP induced loss of DA using both CoQ10, and reduced CoQ10, which produced the largest increases in plasma concentrations. Lastly, we administered CoQ10 in the diet to test its effects in a chronic MPTP model induced by administration of MPTP by Alzet pump for 1 month. We found neuroprotective effects against DA depletion, loss of tyrosine hydroxylase neurons and induction of alpha-synuclein inclusions in the substantia nigra pars compacta. The finding that CoQ10 is effective in a chronic dosing model of MPTP toxicity, is of particular interest, as this may be more relevant to PD. These results provide further evidence that administration of CoQ10 is a promising therapeutic strategy for the treatment of PD.  相似文献

The temperature and mass dependence of maximum consumption rate was measured for larval and early juvenile spotted seatrout Cynoscion nebulosus . Maximum consumption ( C MAX) estimates were obtained from feeding and gut evacuation experiments on larvae (3·8–19 mm standard length, L S) at three temperatures (24, 28 and 32° C), and maximum consumption experiments on juveniles at three temperatures (20, 26 and 32° C). Feeding levels were determined for larvae fed live prey ( Brachionus plicatilis and Artemia salina ) ad libitum . The midgut and total evacuation times were estimated for fish feeding continuously and discontinuously using alternate meals of tagged and untagged live prey. Temperature and fish size had significant effects on gut evacuation and consumption. The gut evacuation time increased with increasing fish size, and decreased with increasing temperatures. Mass‐specific midgut contents increased for small larvae <0·156 mg dry mass ( M D)( c . 4 mm L S), and decreased for larger larvae and juveniles. Maximum consumption was modelled by fitting a polynomial function to a reduced dataset of individuals feeding at high levels. The C MAX model predicted an initial increase in specific feeding rate from 70 to 155% M D day−1 for small larvae, before declining for larger larvae and juveniles.  相似文献

A rapid and simple method for enumerating total aerobic plate counts (APC) and coliforms in raw milk was developed and compared with conventional plating method. Following two-fold serial dilution of samples in a 96 well microtiter plate, double strength of two different modified media for APC or coliforms was added to each well. The final positive well (purple to yellow color) was determined and converted to dilution factors. The dilution factor of each sample was converted to Log10 DF (Dilution factors) and compared to actual microbial numbers Log10 CFU/mL. The results of 2-fold dilution method (Log10 DF) were strongly correlated to conventional plating method (Log10 CFU/mL) (P < 0.05). The correlation of the scatterplot of spread plating and 2-fold dilution method indicated a high level of agreement between two methods (R2= 0.921 for total counts and R2= 0.916 for forms from raw milk). This 2-fold dilution method is an easy, rapid, and economical method for enumeration of total microbial loads and coliforms in raw milk.  相似文献

Abstract The development, survivorship and reproduction of Bemisia tabaci B-biotype on three bean species were studied at 26±1°C in the laboratory. The developmental periods from egg to adult varied from 27.80 days on garden beans to 18.20 days on soybeans. The survivorship from egg to adult on soybeans, cowpeas and garden beans was 77.14, 70.14 and 64.28%, respectively. The average longevity of female adults ranged from 12.30 days on soybeans to 9.80 days on garden beans, and the oviposition of B. tabaci varied from 160.85 eggs on soybeans to 98.00 eggs on garden beans. Life table parameters were calculated as biological attributes for Bemisia tabaci populations fed on three bean species. The results indicated that the intrinsic rate of increase (rm), the finite rate of increase (λ) and net reproductive rate (R0) were high for populations fed on soybeans, with values of 0.1857, 1.2041 and 82.1576, respectively. The corresponding values were less for populations fed on garden beans, with values of 0.1097, 1.1159 and 31.2661, respectively. The parametric values for cowpeas were intermediate between soybeans and garden beans but no significant difference were observed for the rm values for soybeans and cowpeas. Experimental evidence in our investigation indicated that Bemisia tabaci is best adapted and shows the greatest preference for soybean of the three bean species tested in this study.  相似文献

The vitamin B12-producing capacity of micro-organisms isolated from baboon faeces and gastric contents was measured using Lactobacillus leichmannii . The animals were fed either a diet deficient in or supplemented with vitamin B12 (controls). Samples of gastric and small intestinal contents, obtained at laparotomy from two young vitamin B12-deprived baboons, contained varying quantities of vitamin B12. Many of the organisms isolated from these aspirates produced vitamin B12 in vitro . The highest levels of vitamin B12 were produced by anaerobic organisms. Gastric juice samples from vitamin B12 deprived and control baboons contained similar types of organisms with like vitamin B12-producing capacity. The vitamin B12 content, pH and total bacterial counts of gastric juice samples aspirated after a 6 h fast from vitamin B12 deprived baboons were not significantly different from those of the control animals. The pH values of gastric juice samples aspirated 18 h after feeding, however, were significantly lower than those of 6 h fasting samples in both groups. The mean vitamin B12 levels in the total volumes of gastric juice aspirated after each fasting period were similar. The possible involvement of the gastrointestinal flora in the vitamin B12 status of the baboon is discussed.  相似文献

Microbiological characteristics of anevato: a traditional greek cheese   总被引:1,自引:0,他引:1  
Nine batches of Anevato, raw goat milk cheese, were examined throughout a 60 day storage time at three different periods within the lactation season of the goat. High mean log counts per gram of cheese for aerobic bacteria (7·92–9·56), lactic acid bacteria (7·78–9·32), Gram-negative organisms 5·64–9·67), psychrotrophs (7·90–11·79) and proteolytic bacteria (7·57–9·36) were found. Enterobacteriaceae, coliforms and yeasts were considerably lower. Enterobacteriaceae and coliforms in the curd of cheese made in May were lower by approximately 3·0 log10 cfu g−1 than counts in curd made in January, and were lower by about 2·5 log10 cfu g−1 than those in cheese made in March. This coincided with lower pH and higher counts of lactic acid bacteria in cheese made in March and May. Yeast populations were affected by the season and were higher in May than March and/or January. Lactococci dominated in the cheese until 15 days, but lactobacilli became predominant after 30 days. Lactococcus lactis was the most abundant species of lactic acid bacteria found in Anevato cheese. Results suggest the need for improving milk quality and/or using heat-treated milk to produce Anevato cheese; the use of L. lactis as a starter would possibly eliminate or suppress the growth of undesirable organisms.  相似文献

The metamorphosis of Solea senegalensis was studied in larvae reared at 20° C and fed four different feeding regimes. A, Artemia (4 nauplii ml−1); B, Artemia (2 nauplii ml−1); C, mixed diet (2 nauplii ml−1 and 3 mg ml−1 microencapsulated diet); and D, microencapsulated diet (3·7 mg ml−1). Rotifers were also supplied in all cases during the first days of feeding. These feeding regimes supported different growth rates during the pre-metamorphosis period (regime A, G=0·376 day−1; regime B, G=0·253 day−1; regime C, G=0·254 day−1; regime D, G=0·162 day−1). Larvae started metamorphosis 9 days after hatching (DAH) when fed the regime A, 13 DAH with regime B, 11 DAH with regime C and 15 DAH with regime D. A minimum 5·6–5·9 mm LT was required under all feeding regimes to initiate the metamorphosis. Eye translocation was completed when the larvae reached 8·6–8·7 mm LT (regimes A, B and C), but only 7·3 mm LT with regime D. 4·4–6·2 days were required to complete eye migration under the regimes A, B and C, and 18·3 days under the regime D. This transformation is concomitant with changes in body reserves, and with the pattern of some digestive enzymes.  相似文献

Cabbage, green beans, or seeds from sunflowers grown either on municipal sewage sludge-amended soil or soil alone were fed to male and female Coturnix quail, as 50% of a complete diet, for 5 weeks. Specific activities of liver glutathione S-transferase (GST) were similar in all quail fed the latter two plant diets and also similar to quail fed a nonplant, control diet. Sludge-grown cabbage-treated quail exhibited liver GST activities significantly higher (P less than 0.05) than levels of liver GST in birds fed the other plants, with a further twofold activity increase in quail fed the soil-grown cabbage. This response seems to be correlated with the levels of glucosinolates present in the cabbage, i.e., 3040 and 9253 ppm (dry basis) in the sludge-grown and soil-grown cabbage, respectively.  相似文献

When whole plants were exposed to 14CO2, almost the same amount of radioactivity was taken up initially by each leaf regardless of its position on the stem and of the presence of beans at that node. Thus, although developing beans are a powerful sink for assimilated carbon, they do not increase the CO2 uptake by adjoining leaves.
The distribution of labelled assimilates 6 hours after feeding 14CO2 to a single leaf for 1 hour varied with both the position of the treated leaf and the stage of development of the plant. Before any flowers were set most of the radioactivity from all expanded leaves moved downwards to the roots and the stem below the treated leaf (lower stem). Later, during pod-fill, the upper leaves maintained this supply to the roots and lower stem, whilst most of the carbon translocated from the lower and mid-stem leaves went to the beans. However, we found no exclusive relationship between a leaf and the supply to beans developing on the same node.
The amount of radioactivity moving out of a source leaf at a fruiting node increased over successive samplings up to 48 h; the pattern of distribution of the 14CO2 however remained virtually unchanged.  相似文献

The economy of Ca utilization is under the control of vitamin D3, particularly its active metabolite 1,25-dihydroxy cholecalciferol [1,25(OH)2D3]. In sufficient Ca absorption leads to tibial dyschondroplasia resulting in not attaining optimum body weight. Our earlier studies [T.P. Prema, N. Raghuramulu, Phytochemistry 37 (1994) 167] have shown that the Cestrum diurnum (CD) leaves contain vitamin D3 metabolites. It was felt whether incorporation of CD as a source of 1,25(OH)2D3 could improve the Ca absorption in broilers. Four groups of 60 birds each were fed with either normal diet or normal diet + 0.25% CD or normal diet without vitamin D3 or normal diet without vitamin D3 + 0.25% CD leaf powder for 45 days. In subsample of six birds it was observed that incorporation of CD leaves in the feed had the maximal effect on all the parameters studied. The results indicate that the intestinal Ca transport as represented by Serosa/Mucosa (S/M) ratio was found to be significantly (p < 0.01) higher in broilers fed diet with CD leaf powder and the 1 hydroxylase activity in kidney is significantly (p < 0.001) higher in negative controls. On the other hand the supplementation of CD leaves enhanced the serum Ca, body weight, tibia weight, density and strength resulting in the disappearance of tibial dyschondroplasia. No lesions of toxicity were observed in any of the soft tissue examined. The results suggest that the incorporation of CD leaf powder in poultry feed could be beneficial to the poultry.  相似文献

The protective effects of dietary Ca2+ supplementation against Cd accumulation in rainbow trout Oncorhynchus mykiss fed with Cd-contaminated food were evaluated in relation to chronic changes in intestinal absorption rates. The changes were measured ' in vitro '. The control diet contained c. 20 mg Ca2+ g−1 food and 0·25 μg Cd g−1 food; the experimental diets were supplemented with CaCO3 and Cd(NO3)2·4H2O to levels of 50 mg Ca2+ g−1 food and 300 μg Cd g−1 food, alone and in combination. The Ca2+ and Cd absorption rates were measured using radiotracers (45Ca, 109Cd) at total Ca2+ and Cd concentrations of 3·0 and 0·12 mmol l−1, respectively in the intestinal saline. Chronically elevated dietary Cd caused a significant increase in Cd absorption rate by up to 10-fold at 30 days in the mid-intestine. The high Ca2+ diet prevented this up-regulation of Cd transport rate. Conversely, intestinal Ca2+ absorption was significantly increased by two- to five-fold by the Ca2+-supplemented diet at 30 days in both the mid- and posterior intestine, and this effect was eliminated when Cd was simultaneously elevated in the diet. Ca2+ and Cd probably interact at common pathways and transport mechanisms in the intestine, though independent pathways may also exist.  相似文献

Abstract: Rhabditis nematodes fed a diet of Escherichia coli defecate viable undigested bacteria. These bacteria retain phenotypic characteristics, including those encoded on plasmids. Nematodes can survive a 2-min surface sterilization with 2% chlorine bleach; internalized bacteria also survive this treatment and are released in the nematode wastes. Bacteria alone or on the surface of dead nematodes are unable to survive incubation with this solution. There were 3.2 × 105 viable bacteria per nematode, indicating that sufficient bacteria were present for gene transfer. Transconjugants ( lac nal R str R cm R) were recovered in the nematode fecal material following a protocol where nematodes were initially fed a plasmidless lac nal R str S cm S E. coli and then, after surface sterilization, a lac + nal S E. coli plasmid donor containing the conjugative R100JA ( str R cm R) plasmid. The presence of plasmids in the transconjugants was confirmed by gel electrophoresis. The occurrence of conjugation in the gut was confirmed by dissection of individual surface-sterilized nematodes and isolation of transconjugants.  相似文献

Abstract A method is proposed that allows the enrichment and most probable number estimation of H2/CO2-utilizing acetogenic bacteria. It is based on the difference in acetate production for serial dilutions incubated under either a test H2/CO2 (4:1), or a control N2/CO2 (4:1) headspace atmosphere. A nutritionally non-selective medium was used, containing bromoethane-sulfonic acid as inhibitor of methanogenic archaea and 10% pre-incubated clarified rumen fluid. Acetogenic bacteria were enumerated in rumen and hindgut contents of animals and in human feces. They ranged from below 102 to above 108 per gram wet weight gut content and their population levels were the highest in the absence of methanogenesis. The method described therein should prove useful to better understand the diversity and ecological importance of dominant gut acetogens.  相似文献

Previously, we have reported that rye significantly increased both viscosity and Clostridium perfringens proliferation when compared with corn in an in vitro digestive model. Two independent trials were conducted to evaluate the effect of rye as a source of energy on bacterial translocation, intestinal viscosity, gut microbiota composition, and bone mineralization, when compared with corn in turkey poults. In each experiment, day-of-hatch, turkey poults were randomly assigned to either a corn or a rye diet (n = 0 /group). At 10 d of age, in both experiments, 12 birds/group were given an oral gavage dose of fluorescein isothiocyanate dextran (FITC-d). After 2.5 h of oral gavage, blood and liver samples were collected to evaluate the passage of FITC-d and bacterial translocation (BT) respectively. Duodenum, ileum and cecum gut sections were collected to evaluate intestinal viscosity and to enumerate gut microbiota. Tibias were collected for observation of bone parameters. Broilers fed with a rye diet showed increased (p<0.05) intestinal viscosity, BT, and serum FITC-d. Bacterial enumeration revealed that turkey poults fed with rye had increased the number of total lactic acid bacteria (LAB) in all three sections of the gastrointestinal tract evaluated when compared to turkey poults fed with corn. Turkey poults fed with rye also had significantly higher coliforms in duodenum and ileum but not in the ceca, whereas the total number of anaerobes increased only in duodenum. A significant reduction in bone strength and bone mineralization was observed in turkey poults fed with rye when compared with corn fed turkey poults. In conclusion, rye evoked mucosal damage in turkey poults that increased intestinal viscosity, increased leakage through the intestinal tract, and altered the microbiota composition and bone mineralization. Studies to evaluate dietary inclusion of selected Direct-Fed Microbial (DFM) candidates that produce exogenous enzymes in rye fed turkey poults are currently being evaluated.  相似文献

The postprandial excretion pattern of ammonia in dependence of feeding regime (fasting, 1 ×/day, 2 ×/day, 4 ×/day and continuous feeding), was determined in rainbow trout ( Oncorhynchus mykiss ), using continuous flow analysis. In fed fish ammonia peaked c. 7 h after the first meal with no differences in pattern between treatments. Fasting fish did not show a pattern. Overall production rates (NH4 and NO2+ NO3) ranged from 0.29–0.31 g kg−1 BW/d in fed fish and were around 0.07 g kg−1 BW/d in fasting fish. Additionally determined total N (Kjeldahl) showed much higher values in fed fish (0.78–1.05 g kg−1 BW/d) but only slightly higher values in fasting fish (0.11 g kg−1 BW/d). Budgets of nitrogen (N) and energy (E) showed low recoveries ( c. 50% and between 50% and 70%, respectively). When correcting ammonia excretion (NH4 and NO2+ NO3) using literature data on urea excretion of O. mykiss and assuming that total N partly stemmed from uneaten but undetected feed, both N and E budgets reached a recovery of around 100% in all four fed groups. Implications of this approach are discussed in the light of incomplete budgets as determined in earlier studies.  相似文献

Growth of Enterobacteriaceae, coliforms and faecal coliforms in Manchego cheese during the first 24 h after manufacture was retarded by treatment of milk with 0.1 g 1-1 H2O2 compared to growth in control cheese made from untreated milk. Moreover, the decrease in their numbers during cheese ripening was accelerated by the H2O2 treatment of milk. In contrast, KNO3 or KNO2 addition to milk enhanced the growth of these micro-organisms during cheese manufacture and favoured their survival during ripening.  相似文献

Abstract: When a fistulated cow was fed an all forage diet, ruminal pH remained more or less constant (6.7 to 6.9). The ruminal pH of a concentrate-fed cow decreased dramatically in the period soon after feeding, and the pH was as low as 5.45. Mixed ruminal bacteria from the forage-fed cow converted CO2 and H2 to methane, but the ruminal fluid from the concentrate-fed cow did not produce methane. When the pH of the ruminal fluid from the concentrate-fed cow was adjusted to pH 7.0, methane was eventually detected, and the absolute rate constant of methane production was as high as the one observed with ruminal fluid from the forage fed cow (0.32 h−1). Based on the zero-time intercepts of methane production, it appeared that the concentrate-fed cow had fewer methanogens than the forage-fed cow. When the mixed ruminal bacteria were incubated in a basal medium containing 100 mM acetate, methanogenesis was pH-dependent, and no methane was detected at pH values less than 6.0. Because the removal of acetic acid completely reversed the inhibition of methanogenesis, it appeared that volatile fatty acids were causing the pH-dependent inhibition. Based on these results, concentrate diets that lower ruminal pH may provide a practical means of decreasing ruminal methane production.  相似文献

Abstract: The Egyptian fruit bat Rousettus aegyptiacus develops severe vitamin B12 deficiency when fed a diet of fresh peeled fruit and water. In a group of bats fed this diet, B12 concentrations in the serum and brain were low, and neurological impairment, evidenced by deficient or absent hindlimb groping or grasping ability and climbing difficulties, was manifest. Control bats fed the identical diet supplemented with B12 showed no such changes. Fatty acid analysis of whole brain homogenates showed a higher level of 20:4 in the deficient group. Phosphatidylcholine showed a marginally higher percentage of 18:3. The total percentage of branched chain fatty acids of phos-phatidylethanolamine was four times higher in deficient brains, comprising 2% of the total. Sphingomyelin showed a slightly higher percentage of 15:0, and a significantly lower percentage of long chain fatty acids C-24 and longer ( p < 0.01). The compositions of nonhydroxy fatty acids in cerebroside were unchanged. Examination of phospholipids showed that 8.9 ± 0.4% of total phosphorus was present as sphingomyelin in deficient bats, compared with 11.9 ± 1.2% in control animals (p < 0.05). There were no statistically significant differences in the concentrations of total brain lipid, protein, phospholipid, glycosphingolipid, cholesterol and plasmalogen between B12-deficient and control bats.  相似文献

Abstract Concentrations of oxalate-degrading anaerobes in ruminal contents of sheep were determined from counts of colonies producing clear zones on a calcium oxalate medium (D agar with 7 mM CaCl2). Viable counts of oxalate degraders from a 55-kg sheep fed a diet containing 32% halogeton (4.6% oxalate) averaged 2.6 × 106/ g (dry weight). When the halogeton concentration in the diet was reduced to 16%, counts of oxalate degraders decreased nearly 300-fold. Oxalate-degrading isolates from this sheep were similar to OxB, the type strain of Oxalobacter formigenes . When a 45-kg sheep was fed diets containing 2.2, 1.5, and 0.8% oxalate, viable counts of oxalate degraders (enumerated on D agar with 14 mM CaCl2 and 20% filter-sterilized ruminal fluid) represented 0.85, 0.52, and 0.06% of the total viable population, respectively; total viable counts were essentially unchanges by these concentrations of dietary oxalate. Similar percentages of oxalate degraders were also observed when a 23-kg sheep was fed diets containing 1.5 or 0.8% oxalate. This report presents the first direct measurements of the concentrations of oxalate-degrading bacteria in the rumen and supports the concept that the availability of oxalate in the diet influences the proportion of oxalate-degrading bacteria in the rumen  相似文献
