The aim of this study was to determine the survival of Campylobacter jejuni in chicken meat samples at frozen temperatures and given length of incubation and to determine the impact of aerobic bacteria on the survival of C. jejuni. The chicken meat samples were inoculated with C. jejuni NCTC 11351 suspensions and stored in bags at temperatures of -20°C and -70°C. The mean value of C. jejuni from meat samples decreased from 7.52 log10 CFU/g after 30 minutes of incubation at ambient temperature, to 3.87 log10 CFU/g on the eighth week of incubation at -20°C, and to 3.78 log10 CFU/g at incubation at -70°C after the same incubation period. Both freezing temperatures, -20°C and -70°C, decreased the number of campylobacters. The presence of aerobic mesophilic bacteria did not influence the survival of C. jejuni in chicken meet samples. Keeping poultry meat at freezing temperatures is important for the reduction of C. jejuni, which has a strong influence on the prevention of occurrence of campylobacteriosis in humans.  相似文献

The survival and development of cow eggs in the rabbit oviduct after storage at room temperature and after cooling and storage at 0-7-5 degrees C was examined. In PBS medium at room temperature 88% of Day-5 and 85% of Day-3 eggs showed normal development, but in TCM 199, 71% of Day-5 and only 49% of Day-3 eggs showed normal development. Duration of storage (1 1/2-2 hr or 6 1/2-7 1/2 hr) and cleavage stage before storage had no appreciable effect on development. Some retardation of development occurred in Day-3 eggs after 96 hr in the rabbit oviduct when compared to Day-5 eggs after 48 hr. Cooling of Day-5 and Day-6 eggs to 0-7-5 degrees C resulted in degeneration of a large proportion of eggs. Of the factors examined, storage medium (PBS or PBS+20%FCS), storage time (2 min, 24 hr) and storage temperature (0, 2, 5 or 7-5 degrees C) had little effect, but slower cooling rates tended to improve survival of eggs although the differences were not significant. More morulae (greater than 32 cells) than 8-to 24-celled eggs developed normally.  相似文献

A congenic pair ofCampylobacter jejuni has been previously developed in our laboratory that will (strain A74/C) and will not (strain A74/O) colonize 2-day-old chicks dosed with 105 colony forming units. Outer membrane protein (OMP) extracts of these organisms were prepared and studied by sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) and Western blot analyses. No consistent differences between the colonizer and noncolonizer were detected by SDS-PAGE. However, an antigen of 28 kD molecular weight was consistently found in the noncolonizer, but only at greatly reduced levels or not at all in the colonizer by Western blot analysis with rabbit anti-OMP serum. After affinity purification and cross absorption of serum against OMP from the colonizer, an antigen of 69 kD molecular weight was found unique to the colonizing strain. Exclusive association of the 69 kD antigen with the colonizing strain suggests that it may be a colonization factor.  相似文献

Freeze fracture electron microscopy studies were performed on samples of Anacystis nidulans quenched from different temperatures. Membrane lipid phase separations were observed to take place over the ranges 15–30°C, 5–25°C and –5–15°C for cultures grown at 38, 28 and 18°C, respectively. Differential scanning calorimetry heating curves showed endotherms which coincided with these temperature ranges. Variations of phase separation temperatures with growth temperature, and hysteresis effects in the calorimetric measurements, were related to changes in the fatty acid composition of membrane lipids.  相似文献

Egg parasitism of Trichogramma pretiosum strain RV when presented with eggs of Anticarsia gemmatalis and Pseudoplusia includens was investigated at 18, 20, 22, 25, 28, 30 and 32 °C. The number of eggs parasitized per day decreased for both hosts as a function of the age of parasitoids, reaching 80% of lifetime parasitism more rapidly as temperature increased; on the 4th day at 32 °C and on the 12th day at 18 °C. The lifetime number of parasitized P. includens eggs achieved by the parasitoid maintained at 20 °C (44.95 ± 3.94) differed from the results recorded at 32 °C (28.5 ± 1.33). Differently, the lifetime number of A. gemmatalis parasitized eggs did not differ among the temperatures. When T. pretiosum reached 100% of lifetime parasitism, each adult female had parasitized from 28.5 ± 1.33 to 44.95 ± 3.94 and from 29.58 ± 2.80 to 45.36 ± 4.50 P. includens and A. gemmatalis eggs, respectively. Also, the longevity of these adult T. pretiosum females, for which P. includens or A. gemmatalis eggs were offered, was inversely correlated with temperature. Not only were the survival curves of those adult T. pretiosum females of type I when they were presented with eggs of A. gemmatalis but also with eggs of P. includens, i.e., there was an increase in the mortality rate with time as the temperature increased. In conclusion, T. pretiosum strain RV parasitism was impacted by temperature when on both host eggs; however, the parasitoid still exhibited high survival and, more importantly, high number of parasitized A. gemmatalis and P. includens eggs even at the extremes tested temperatures of 18 and 32 °C. Those results indicate that T. pretiosum strain RV might be well adapted to this studied temperature range and, thus, be potentially suitable for use in biological control programs of P. includens and A. gemmatalis in different geographical areas that fits in this range. It is important to emphasize the results here presented are from laboratory studies and, therefore, field trials still need to be carried out in the future with this strain in order to support the full development of the technology intend to use this egg parasitoid in soybean fields worldwide.  相似文献

大别山五针松( Pinus dabeshanensis C. Y. Cheng et Y. W. Law)为松科( Pinaceae)松属( Pinus Linn.)植物,自然种群数量极少,目前仅发现在安徽省岳西县大王沟海拔900~1300 m阴坡和半阴坡有相对集中的分布,种群规模计200余株,且多为成年个体,林下幼苗极少,自然更新困难,为中国特有珍稀树种之一[1]。大别山五针松常在每年3月份至4月份形成花芽,5月中下旬花粉成熟并散发,花粉具气囊,此时雌球花张开接受传粉,球果翌年9月成熟[2]。据作者近年的调查,大别山五针松种子败育率较高,且种子质量较低,这也是该种类濒危的重要原因之一。  相似文献

The survival of a human strain of Campylobacter jejuni in raw, pasteurized and ultra-heat-treated goat's milk stored at 5°, 10°, 15° and 20°C was studied. No viable units were detected in raw milk after 24 h at 20°C and 48 h at 15°C. None were detected in pasteurized milk after 48 h at 20°C. In all other samples, there was a decline in viable units in the first 24 h but very little decline in the next 24 h period. The organism survived best at 5° and 10° C.  相似文献

Flagella are essential for motility and have been implicated to be one of the pathogenic determinants. The flagellum ofCampylobacter jejuni is a polymeric structure of a 62-kd protein. Using a high-affinity flagellin antibody to screen a lambda gt 11 phage genomic expression library ofC. jejuni strain TGH9011 (Serotype LIO36), a recombinant phage clone lambda gt 11RK that expresses theC. jejuni flagellin protein was isolated. The recombinant lambda gt 11 RK produced a 56-kd protein upon induction with isopropylthiogalactoside, which reacted specifically with anti-flagellin antibody. The flagellin gene was sequenced, and comparative analysis of the nucleotide and amino acid sequence identified a region of the flagellin that shows hypervariability among differentCampylobacter species and strains.  相似文献

Initial psychrotroph counts determined by a Most Probable Number technique were correlated with shelf-lives of pasteurized milk determined at a number of storage temperatures. The initial psychrotroph count was also correlated with a bacterial count carried out on milk agar containing crystal violet penicillin and nisin after previous incubation of the milk at 15°C for 25 h.
Pre-incubation counts carried out at a variety of temperatures and on a variety of media were examined for their relation to shelf-life. Shelf-lives at four pre-set temperatures (2, 6, 10 and 14°C) could best be predicted by pre-incubation of pasteurized milk at 15°C before inoculation on milk agar.
An equation which allows prediction of shelf-life of pasteurized milk at any storage temperature is described.  相似文献

Survival of Vibrio cholerae El Tor serotype Inaba was examined in pasteurized milk, freshwater fish, raw beef and raw chicken at a variety of temperatures. Both food type and incubation temperature affected survival. At the lowest temperatures, V. cholerae remained viable in meats for up to 90 d at—5°C and 300 d at —25°C. In milk, however, it was not detectable after 34 d at —5°C and 150 d at —25°C. At 7°C it survived 32 d, on average, in milk and only 18–20 d in the other foods. At room temperatures survival periods were shorter, never exceeding 10 d, and it was not detected after 2 d incubation at 35°C in chicken and fish.  相似文献

Summary The effect of temperature on the nuclear envelope structure and the transport of total RNA and ribosomal subunits from nucleus to cytoplasm was examined inTetrahymena cells propagated at two different temperatures. Freeze-etch electron microscopy of cells grown at 23 and 18°C detects the emergence of smooth areas on the fracture faces of the nuclear membranes upon lowering the temperature below 15 and 12°C, respectively. Coincident with these freeze-etch changes, a discontinuous decrease is observed in the nucleocytoplasmic RNA-transport; this is probably not due to a cease in RNA-synthesis. Below the thermotropic discontinuity observed in the transport of total RNA in 18°-cells the nucleocytoplasmic transport of the small and large ribosomal subunits is equally retarded. Recent temperature studies on the endoplasmic reticulum membranes ofTetrahymena suggest that the freeze-etch changes in the nuclear membranes are induced by a thermotropic clustering of the membrane lipids. We conclude that this lipid clustering induces the permanent protein constituents in the nuclear envelope pore complexes to change from a relatively open into a relatively closed state thus causing the observed decrease in RNA-transport.  相似文献

Components of outer membrane preparations, heated saline extracts, and phenolwater lipopolysaccharide extracts obtained from strains ofCampylobacter jejuni representing seven passive hemagglutination serotypes (Penner serotypes 1–4, 13, 16, and 50) were electrophoresed in sodium dodecyl sulfate-polyacrylamide gels. Tests of gel eluates demonstrated that lipopolysaccharide antigens are involved in serotypingC. jejuni by passive hemagglutination and that other cell surface components have no activity. This finding was confirmed by hemagglutination inhibition. In the typing ofC. jejuni by passive hemagglutination, each serotype is probably defined by the presence of one or more specific lipopolysaccharides. These findings may lead to a clarification of the serotyping nomenclature for those systems that depend on passive hemagglutination. It is recommended that a single internationally agreed numbering system be adopted for lipopolysaccharides derived fromC. jejuni.  相似文献
