The longitudinal development of spontaneous motility in chick embryos was studied by Kovach's method (Kovach 1970) from the 10th day of incubation up to hatching, in completely intact eggs. From the 10th to 12th day of incubation, very low amplitude movements of a burst character predominated in spontaneous motility. From the 13th day, both low and high amplitude movements could be distinguished. From the 18th day, high amplitude movements alternating with intervals of motor inactivity preponderated. This discontinuous motility, which was most pronounced on the 20th day of incubation, changed to periodic strong hatching movements. Reduction of spontaneous motility after the 17th day of incubation was not confirmed. Strychnine already activated spontaneous motility in 11-day embryos, but typical convulsions did not appear until the 15th incubation day. With picrotoxin, motility was likewise stimulated in 11-day embryos and paroxysmal activation did not occur until the 15th incubation day. In older embryos, convulsions were gradually succeeded by a continuous increase in spontaneous motility. The effect of picrotoxin had a much longer latent period than the effect of strychnine.  相似文献

The effects of the acute and chronic administration of chlordiazepoxide on spontaneous motility and on the reactivity of the generator of embryonic motility were studied in chick embryos from the 4th to the 19th day of incubation. 1. The acute administration of chlordiazepoxide (5 mg/kg e.w.) significantly depressed spontaneous motility from the 13th day of incubation. 2. The chronic administration of chlordiazepoxide (12.2 mg/kg e.w./24 h) from the 4th to the 8th, 12th and 16th day of incubation enhanced the reduction of the spontaneous motility of 17-day-old embryos. 3. The chronic administration of chlordiazepoxide significantly modified the activity of both activators (strychnine, metrazol, bicuculline, picrotoxin) and inhibitors (GABA, chlordiazepoxide) of the spontaneous motility of chick embryos.  相似文献

From the 4th to the 16th day of incubation, GABA was administered continuously to chick embryos in a mean dose of 9.04 +/- 0.98 mg/kg e.w./24 h. On the 17th day of incubation, spontaneous motility was evaluated from the frequency of spontaneous movements as resting motility and motility after the acute administration of GABA (100 mg/kg e.w.), bicuculline (1 mg/kg e.w.) and oxazepam (10 mg/kg e.w.). 1) The chronic administration of GABA reduced the spontaneous motor activity of the experimental embryos to 38.4-47.8% of the control value. To obtain this effect it was sufficient to administer GABA between the 4th and the 8th day of incubation. 2) The inhibitory effect of the acute administration of GABA in the experimental embryos was only half its effect in the controls. Conversely, the relative size of bicuculline activation of motility was distinctly greater in the experimental embryos, which were also significantly more sensitive to oxazepam. The results show that GABA has a dual effect during embryogenesis--a) an early effect between the 4th and 8th day of incubation causing a chronic debt in the development of spontaneous motor activity, and b) an inhibitory effect as a central transmitter, which begins to be manifested in embryonic spontaneous motility of chick embryos on about the 15th day of incubation.  相似文献

The effects of acute and chronic application of ketamine on the resting spontaneous motility, its development and reactivity was studied in chick embryos of white Leghorns. 1. Acute application of ketamine (Narcamon) in a dose of 12.5 mg/kg e.w. partially depressed spontaneous motility as early as in 11-day old chick embryos. From day 15 of incubation ketamine very effectively blocked spontaneous motility. 2. Ketamine was fully ineffective in spinal preparations (decapitation on day 2 of incubation) of 11- and 13-day-old embryos. It was not until day 15 evoked that it depressed motility as in normal embryos. 3. Chronic continuous supply of ketamine (average dose 6.34 +/- 0.72 mg/kg e.w./24 h) from day 4 of incubation till day 8, 12, or 16 of incubation reduced the developmental decrease of spontaneous motility by 23.1-6.0% as compared to the controls. This effect was already observed after the first 4 days of chronic application of ketamine. 4. Chronic application of ketamine significantly diminished the strychnine activation and GABA-mediated depression of spontaneous motility. The depressive effect of the acute application of ketamine itself was hardly affected. The results have shown that ketamine interferes with the development of the endogenous rhythm of intrinsic activity and with the development of reactivity of the generator of embryonic spontaneous motility.  相似文献

Development of the effectivensess of the systemic administration of oxotremorin (200 micrograms/kg egg weight) on spontaneous motility in chick embryos was studied from the 11th to the 19th day of incubation. 1) Oxotremorin activated spontaneous motility from the 15th day of incubation; subsequently, its effect increased with the embryo's age. 2) Glycine (100 mg/kg e.w.) blocked the activating effect of oxotremorin, again in a direct correlation to the embryo's age. 3) The spontaneous motility of spinal embryos was activated significantly less, thereby demonstrating the importance of the supraspinal component of the effect of oxotremorin. 4) The results are evaluated primarily as evidence of the role of the cholinergic mechanism (Everett 1964) in embryonal development of supraspinal control of the spontaneous activity of spinal motoneurones.  相似文献

The effect of imipramine on the spontaneous motility and development of chick embryos was studied from the 4th to the 19th day of incubation. On acute administration (a single dose of 12.5 of 25 mg/kg egg weight), imipramine already induced significant depression of spontaneous motility in 11-day embryos--an effect which increased significantly after the 15th day of incubation. The similar effect of imipramine in spinal embryos testifies to its direct action on the spinal cord and draws attention to certain details of the role of supraspinal structures of the CNS in the acute effect of imipramine. The chronic administration of imipramine showed that it had an almost 100% lethal effect from 4th to the 7th day of incubation. Between the 8th and the 10th day it caused longlasting depression of spontaneous motility. When it was administered between the 11th and 16th day of incubation, no significant effect on the development of spontaneous motor activity was found in chick embryos.  相似文献

The motility of isolated uterine horns as well as the generation of PGE and PGF like material by the uterus from estrus and spayed rats, treated or untreated with 17-beta estradicl, were studied. Following 40 minutes of mounting the spontaneous motility of uteri from estrus rats had a lower magnitude than that from spayed ones. The amount of PGF-like material was similar in both groups whereas the first one liberated less PGE-like substance. In spayed animals treated with 1 μg of 17-beta estradiol the decay of spontaneous contractile force was higher than that observed in untreated rats, and similar to that displayed by uteri from estrus. Less PGE-like material was liberated in comparison with spayed animals and a tendency to produce higher quantity of PGF-like compounds was observed, although the level was not significantly different. With 50 μg of 17-beta estradiol the spontaneous reduction of contractile activity was higher than in spayed animals and than in those treated with 1 μg. The amount of PGF-like material liberated was higher than in spayed rats and less PGE-like substance was generated comparing with spayed and 1 μg-treated animals. These findings show that estradiol decreases the release of PGE-like compound. It would also appear that this may have some relationship with the levels of spontaneous contractile activity of the isolated rat uterus.  相似文献

The participation of central monoaminergic systems in the regulation of spontaneous motility in developing chick embryos was tested by systemic administration of several drugs, which affect the different sites of central aminergic systems. Amphetamine (2 mg.kg-1 egg weight) evoked an age-dependent depression of spontaneous motility, which first occurred in a significant fashion on day 13 of incubation. Two thirds of this depression depended upon supraspinal influences. The effect of p-chlorophenylalanine (100 mg.kg-1 e.w.) consisted from day 15 of incubation in a short-lasting depression followed by partial recovery of resting motility. alpha-methyldopa (100 mg.kg-1 e.w.) depressed the spontaneous motility from day 13 of incubation, without any signs of recovery within the first hour after drug administration. The most pronounced depressive effect was evoked by melatonin (25 mg.kg-1 e.w.) even in 11-day-old embryos. Along with the increased depth of depression the recovery of motility declined until full cessation of motility was achieved in 17-day-old embryos. The results were interpreted as constituting further evidence for the involvement of central aminergic systems in the development of supraspinal control over spontaneous motor activity generated by the CNS.  相似文献

Motilin and motilin receptors have been found in most regions of the brain, including the amygdala, one of the most important parts of the limbic system. Our previous study found that administration of motilin in the hippocampus stimulates gastric motility. We now explore the effect of motilin in the amygdala on gastric motility. In conscious rats, gastric motility was recorded after microinjection of motilin, motilin receptor antagonist (GM-109) or a mixture of the two into the basomedial amygdala nucleus (BMA). In anesthetized rats the changes of spontaneous discharges of gastric distention sensitive neurons (GDSN) in the BMA were recorded after intracerebroventricular (i.c.v.) microinjection of motilin or GM-109. In conscious rats the amplitude of gastric contractions increased dose-dependently after microinjection of motilin in the BMA, and decreased after microinjection of GM-109. The excitatory or inhibitory effects induced by motilin or GM-109 alone, were weakened by microinjection of a mixture solution of both. The spontaneous discharge frequency of gastric distention excitatory neuron (GDEN) was mainly inhibited by i.c.v. microinjection of motilin but excited by GM-109. In contrast, the spontaneous discharge frequency of gastric distention inhibitory neuron (GDIN) was mainly excited by motilin, but inhibited by GM-109. Our findings suggest that motilin may regulate gastric motility by modulating neural pathways in the BMA.  相似文献

In the investigations of the rate of oxygen uptake by sperm cells in frozen bull semen it was found that exogenous glutathione (GSH) in a 5 mM concentration failed to stimulate mitochondrial oxidative processes in the sperm cells. At the same time it was observed that the respiration of sperm cells changed in relation to the season of the year when ejaculates had been collected and preserved. The highest values were noted in winter and the lowest ones in summer. Moreover, in the observations on the motility of spermatozoa a difference was observed in the reaction of sperm cells to exogenous GSH, and this difference depended on the season. A significant (p less than or equal to 0.05) rise was disclosed in the number of sperm cells with maintained advancing motility in the presence of GSH in the medium as compared to controls without GSH, but this rise was found only in semen obtained and preserved in winter.  相似文献

The consequences of systemic administration of aminergic transmitters (n-adrenaline 16 microgram/kg egg weight; serotonin 2.5 and 5 mg/kg e.w.; dopamine 2.5 and 5 mg/kg e.w.) for the spontaneous motility and heart rate of 11- to 19-day chick embryos were studied intack eggs. The following results were characteristic for all three transmitters: a) when administered to 11- and 13-day embryos their effect was non-significant; the first signs of activity did not appear until the 15th day of incubation. The effect on 17- and 19-day embryos was stronger. b) After the 15th day of incubation, all these transmitters had a predominantly inhibitory effect on spontaneous motility; in 17- and 19-day embryos this acquired a periodic character. c) The changes in spontaneous motility did not correlate significantly in any way with the relatively small heart rate changes. It is concluded from the results that aminergic mechanisms begin to participate in regulation of the spontaneous motility of chick embryos from the 15th day of incubation, and not before.  相似文献

Development of the effect of glycine and gamma-aminobutyric acid [GABA] on spontaneous motility was studied in 11- to 19-day-old chick embryos under normal conditions and after acute and chronic decapitation. Chronic decapitation was performed on the 2nd day of incubation. Glycine (100 mg/kg egg weight) and GABA (103 mg/kg egg weight) (applied onto the shell membrane) demonstrably inhibited spontaneous motility only from the 15th day of incubation, the inhibitory effect increasing with the embryo's age. When administered together in half doses, glycine and GABA completely inhibited spontaneous motility for the first time in 19-day-old embryos. Neither amino acid influenced depression of motility immediately after decapitation, but 24 and 48 hours after, in 17- and 19-day-old embryos, they had a paradoxical effect, i.e. they transiently activated motor activity and even caused motor paroxysms. After chronic decapitation, both glycine and GABA again had a mild, protracted inhibitory effect. A comparison of spontaneous motility in normal and chronically decapitated embryos showed that the role of supraspinal factors in spinal motor output increases significantly with development of the chick embryo from the 15th day of incubation and that inhibition of these supraspinal factors plays the decisive role in the effect of glycine and GABA.  相似文献

A spontaneous rhythmic motility of the isolated intestine in snail, Chryptomphalus hortensis, has been registered at 30 degrees C with the aid of an electronic transductor and an appropriate amplification. These slow regular movements are affected by ionic composition changes in the suspension medium. Na+, K+ and Ca++ ions prove to be important in this motility, and the addition of Ba++ markedly stimulates it. ACh produces hypermotility from 1.8 X 10(-11) g/ml. Its effect decreases in the presence of atropine and increases in that of pyridostigmine. The intestine is sensitive to 5-HT from 10(-10) g/ml, which stimulates its activity. The effect of histamine is weak. Low concentrations of adrenaline tend to increase the amplitude, whereas concentrations from 10(-5) g/ml onward produce a cease of motility in relaxation.  相似文献

The continuous administration of d-tubocurarine (6.5 +/- 0.4 mg/kg e.w./24 h) to chick embryos from the 4th to the 12th day of incubation had a positive effect on defects produced in the development of spontaneous motility either by decentralization of the spinal cord or by chemical phenobarbital depression, or by a combination of both experimental factors. The sarafotoxins (SRTX) and endothelins (ET) were shown to influence the motility of the isolated rat uterus by inducing an increase in the rate and in the maximum tension of the spontaneous rhythmic contractions and a suppression of the relaxation phase of these contractions. Ovariectomized rats, 24 weeks post-operation, show no spontaneous motility of their uteri and the SRTX/ET peptides induce only a slight tonic increase in the uterine tension. Treatment with 17 beta estradiol restores spontaneous motility and sensitivity to the SRTX/ET peptides in all three contraction modes. It is concluded that the influence of the SRTXs and ETs on uterine motility depends on the hormonal status of the animal.  相似文献

Pregnant women's blood serum (the whole as well as 50- and 100-diluted) did not alter parameters of spontaneous motility, i.e. it did not affect the uterus myocytes' spontaneous motility, whereas the whole or 50-diluted serum raised considerable the rat myometrium's beta-adrenoreaction. The data obtained suggest that the beta-adrenoreaction of the rat uterus' myocytes increases after a prolonged contact with pregnant women's blood serum which may be due to the beta-adrenoreceptor synthesis activation and to an increase in their concentration because of diffusion from the blood serum. This suggests existence of humoral mechanisms of the myometrium beta-adrenoreaction regulation, the mechanisms creating the optimum uterus' contractile activity.  相似文献

The mechanisms by which dopamine (DA) influences gastrointestinal (GI) tract motility are incompletely understood and complicated by tissue- and species-specific differences in dopaminergic function. To improve the understanding of DA action on GI motility, we used an organ tissue bath system to characterize motor function of distal colonic smooth muscle segments from wild-type and DA transporter knockout (DAT -/-) mice. In wild-type mice, combined blockade of D(1) and D(2) receptors resulted in significant increases in tone (62 +/- 9%), amplitude of spontaneous phasic contractions (167 +/- 24%), and electric field stimulation (EFS)-induced (40 +/- 8%) contractions, suggesting that endogenous DA is inhibitory to mouse distal colonic motility. The amplitudes of spontaneous phasic and EFS-induced contractions were lower in DAT -/- mice relative to wild-type mice. These differences were eliminated by combined D(1) and D(2) receptor blockade, indicating that the inhibitory effects of DA on distal colonic motility are potentiated in DAT -/- mice. Motility index was decreased but spontaneous phasic contraction frequency was enhanced in DAT -/- mice relative to wild-type mice. The fact that spontaneous phasic and EFS-induced contractile activity were altered by the lack of the DA transporter suggests an important role for endogenous DA in modulating motility of mouse distal colon.  相似文献

Background

Curcuma has long been used as an anti-inflammatory agent in inflammatory bowel disease. Since gastrointestinal motility is impaired in inflammatory states, the aim of this work was to evaluate if Curcuma Longa had any effect on intestinal motility.

Methods

The biological activity of Curcuma extract was evaluated against Carbachol induced contraction in isolated mice intestine. Acute and chronic colitis were induced in Balb/c mice by Dextran Sulphate Sodium administration (5% and 2.5% respectively) and either Curcuma extract (200 mg/kg/day) or placebo was thereafter administered for 7 and 21 days respectively. Spontaneous contractions and the response to Carbachol and Atropine of ileum and colon were studied after colitis induction and Curcuma administration.

Results

Curcuma extract reduced the spontaneous contractions in the ileum and colon; the maximal response to Carbachol was inhibited in a non-competitive and reversible manner. Similar results were obtained in ileum and colon from Curcuma fed mice. DSS administration decreased the motility, mainly in the colon and Curcuma almost restored both the spontaneous contractions and the response to Carbachol after 14 days assumption, compared to standard diet, but a prolonged assumption of Curcuma decreased the spontaneous and Carbachol-induced contractions.

Conclusions

Curcuma extract has a direct and indirect myorelaxant effect on mouse ileum and colon, independent of the anti-inflammatory effect. The indirect effect is reversible and non-competitive with the cholinergic agent. These results suggest the use of curcuma extract as a spasmolytic agent.  相似文献

Antibodies binding to sea urchin flagellar outer-doublet tubulin have been isolated from rabbit sera by tubulin-affinity chromatography employing electrophoretically purified tubulin as the immobilized substrate. This procedure provides "induced" antitubulin antibody from immune sera and "spontaneous" antitubulin antibody from preimmune sera. These antitubulins were characterized in terms of their specificity, ability to bind to sea urchin axonemes, and effects on the motility of reactivated spermatozoa. Induced antitubulin antibody specifically reduced the bend angle and symmetry of the movement of demembranated reactivated spermatozoa without affecting the beat frequency. At identical concentrations, spontaneous antitubulin had no effect on motility. Affinity-purified induced antitubulins from three other rabbits all gave specific bend-angle inhibition, whereas their corresponding spontaneous antitubulins had no effect on the flagellar movement. The effects of antitubulin on microtubule sliding were examined by observing the sliding disintegration of elastase-digested axonemes induced by MgATP2+-. Affinity-purified induced antitubulin antibody, in quantities sufficient to completely paralyze reactivated flagella, did not inhibit microtubule sliding. The amplitude-inhibiting effect of induced antitubulin on reactivated spermatozoa may be caused by action on a mechanism responsible for controlling flagellar bending rather than by interference with the active sliding process. This is the first report of an antitubulin antibody having an inhibitory activity on microtubule-associated movement.  相似文献

More than two consecutive miscarriages in less than 20 weeks of gestation is defined as recurrent spontaneous miscarriage. Various causes such as uterine anatomical anomalies, genetic factors, and infectious and endocrine disorders have been reported for RPL. However, approximately 50% of the causes are unknown, which can be due to male factors. Several studies have been done on semen parameters to determine the unknown causes and risk factors for miscarriages, however, only studying common semen parameters have not been sufficient. In this study, the relationship between sperm DNA fragmentation, the amount of free radicals, and total antioxidant capacity (TAC) in semen have been considered as a risk factor for spontaneous miscarriage. Semen samples were collected from 42 men whose partners had a history of spontaneous miscarriage and 42 fertile men as the control group. Volume, pH, viscosity, concentration, and motility of semen, as well as sperm morphology were measured. Sperm DNA fragmentation was analyzed by the sperm chromatin structure assay (SCSA) and TUNEL methods, the amount of sperm free radicals was measured by the luminescence method and the total amount of semen antioxidant was measured using the TAC kit. The results have shown that sperm motility in the experimental group was significantly less than the control group (P?=?0.001). The percentage of sperm DNA fragmentation and the amount of free radicals in the experimental group were significantly higher than the control group (P?<?0.001). The total amount of antioxidant was lower in the experimental group compared to the control. Spouses of men with lower sperm motility and higher DNA fragmentation had a higher chance of spontaneous miscarriage when compared to the control group. The results of this study support the hypothesis that sperm DNA fragmentation is a major contributor to spontaneous miscarriage.The relationship between SDF, ROS and TAC with RPL.  相似文献
