Isozyme phenotypes are described for 45 structural loci and 1 modifier locus in bishop pine (Pinus muricata D. Don,) and segregation data are presented for a subset of 31 polymorphic loci from 19 enzyme systems. All polymorphic loci had alleles that segregated within single-locus Mendelian expectations, although one pair of alleles at each of three loci showed significant segregation distortion. The consistency of resolution and segregation at many loci in bishop pine makes electrophoretic analyses feasible for many purposes in this species.  相似文献

Genetic control of GOT, GDH, DIA, MDH, SOD, FDH, ADH, ACP, and LAP enzymes was studied in the seed megagametophytes of cembra pine (Pinus cembra L.) from the natural population of the Ukrainian Carpa-thian mountains. Efficient electrophoretic separation was obtained for 21 loci products. The analysis of allele segregation in heterozygous trees confirms monogenic inheritance of the revealed variants.  相似文献

The comparative analysis of the genetic variation in the plantations of Pinus pallasiana D. Don in Krivbass and the Crimean populations has been conducted using the electrophoretic division of isoenzymes of 10 geneenzymous systems, encoded by 23 loci. In the artificial plantations of industrial ecotopes of Krivbass the level of genetic variation has been higher than in natural stands of Pinus pallasiana D. Don. within the range of one microslope in the Mountainous Crimea. The Nei' s genetic distance (Dn) between two regions varied within 0.016 to 0.026.  相似文献

Electrophoretic spectra of GOT, GDH, DIA, MDH, SOD, FDH, ADH, ACP, IDH enzymes in the megagametophytes of seeds of 69 mountain pine (Pinus mugo Turra) trees from natural populations of the Ukrainian Carpathian mountains have been described. 19 loci products had efficient electrophoretic separation. The analysis of alleles segregation of the heterozygous trees on the whole confirms monogenic inheritance of the discovered variants.  相似文献

On the basis of electrophoretic analysis of 9 enzymous systems encoded by 20 gene loci the level of intra- and inter-population variation of two relict populations of Pinus sylvestris L. in the Ukrainian Carpathians and two ones in Rastochiye was studied. The less allele representation and the lower level of heterozygosity are typical for the Carpathian populations. Fst and Gst, parameters of populations subdivision, were not high--0.020 and 0.022 correspondingly and the coefficient DN was 0.008 in average. The results of the cluster analysis showed that only the populations of Rastochiye were united in one group indicating their genetic affinity.  相似文献

In order to analyze the genetic structure, subdivision and differentiation within and between two small isolated populations of the Crimea relict endemic, Pinus stankewiczii (Sukacz.) Fomin, electrophoretic analysis of the isozyme variation at nine enzymatic systems was carried out using 183 oldest trees. It was demonstrated that in populations of P. stankewiczii, 80% of the genes were in polymorphic state. Each tree was heterozygous at 19.1% loci, and at 21.6% loci in artificial 50-year-old plantation. The genetic structure of two populations was less differentiated (DN = 0.006), compared to their individual localities (DN = 0.008-0.009). Within-population subdivision of the diffusely dispersed populations was higher (FST-GST = 1.8-2.0%) than that of the populations themselves (0.8%).  相似文献

Allozyme variation has been and continues to be a major source of information on the level of genetic variation among plant species. Deciphering the molecular basis of electrophoretic variation is essential for understanding the forces affecting the protein level variation. In this study, the relationship between allozyme heterozygosity and nucleotide diversity in plants is investigated among and within species. Allozyme and nucleotide diversity in 27 plant species was reviewed. At the multilocus level, the two methods are congruent: a clear correlation between the two measures of genetic diversity among plant species was observed, strengthening the view that effective population size is the major determinant of genome-wide diversity. Nucleotide diversity at six allozyme coding genes (6pgdB, aco, gdh, gotC, mdhA, and mdhB) in conifer Pinus sylvestris was investigated jointly with electrophoretic data. Single non-synonymous charge-changing mutations were found together with electrophoretic alleles that consequently were mutationally unique. Synonymous site nucleotide diversity (point estimate of θ W—0.009 per bp) and silent site divergence from Pinus pinaster at allozyme coding loci were at comparable levels with other loci in the species. Linkage disequilibrium was extensive compared to earlier estimates from P. sylvestris and other trees, spanning several kilobases. Allozyme coding genes had an excess of closely related haplotypes whose frequency has recently increased possibly as a result of partial selective sweeps or balancing selection, but complex demographic effects cannot be excluded.  相似文献

Based on electrophoretic analysis of 21 isozyme loci controlling 10 enzyme systems, the intra- and inter-population variation was studied in two peat-bog and three dry-meadow populations of the dwarf mountain pine Pinus mugo Turra from the highlands of the Ukrainian Carpathians. In the studied samples (a total of 164 trees), on average 62% of the studied genes were polymorphic; the mean heterozygosity was 21.3%. The dry-meadow populations differed from the peat-bog populations by allele and genotype diversity and by heterozygosity although the indices characterizing population heterogeneity (Fst and Gst) were small (0.027 and 0.032, respectively). Nei's genetic distances between the populations ranged of 0.011 to 0.032 with the mean of 0.018.  相似文献

用酸性聚丙烯酰胺凝胶(APAGE)技术分析了华北地区5个油松种群在醇溶蛋白水平上的遗传多样性。95份材料分离出12条带,其中11条具有多态性,得到23种不同的带型。种群多态位点比例在16.67%~66.67%之间。Shannon信息指数和Nei’s指数的统计结果表明:关帝山油松种群的遗传多样性高于其它种群;华北地区油松种内遗传多样性为0.248 9;种群内的遗传多样性为0.150 7,占总变异的60.55%;种群间的遗传多样性为0.098 2,占总变异的 39.45%。  相似文献

Using electrophoretic analysis of 22 isozyme loci controlling ten enzyme systems, we studied intrapopulation and interpopulation variation of Scots pine Pinus sylvestris L. in the main forest regions of Ukraine. In 15 of the populations examined, 76.5% of genes were polymorphic, and an average plant was shown to be heterozygous at 23.4% of the genes. The lowest and highest values of major polymorphism parameters were characteristic of respectively the relic populations of Ukrainian Carpathians and the populations from the steppe and forest-steppe zones. Nei's genetic distances between the populations varied from 0.006 to 0.031 (on average 0.016). Cluster analysis failed to show clear trends in the population distribution relative to their geographical position.  相似文献

Summary An Expectation-Maximization (EM)-algorithm procedure is presented that extends Cheliak et al. (1983) method of maximum-likelihood estimation of mating system parameters of mixed mating system models. The extension permits the estimation of the rate of self-fertilization (s) and allele frequencies (Pi) at loci in outcrossing pollen, at marker loci having recessive null alleles. The algorithm makes use of maternal and filial genotypic arrays obtained by the electrophoretic analysis of cohorts of progeny. The genotypes of maternal plants must be known. Explicit equations are given for cases when the genotype of the maternal gamete inherited by a seed can (gymnosperms) or cannot (angiosperms) be determined. The procedure can accommodate any number of codominant alleles, but only one recessive null allele at each locus. An example, using actual data from Pinus banksiana, is presented to illustrate the application of this EM algorithm to the estimation of mating system parameters using marker loci having both codominant and recessive alleles.Issued as AECL-8745  相似文献

用RAPD技术分析了分布于中国东北的3个红松(Pinus koraiensis Seib.et Zucc.)天然群体的遗传多样性及群体间的遗传分化。38个随机引物共检测到241个可重复的位点,其中多态位点139个,占总位点的57.68%。Shannon信息指数和Nei指数的统计结果都表明,红松种内的遗传变异主要存在于群体内,凉水群体的遗传多样性水平高于黑河、虎林群体。群体内遗传相似度为0.927,群体间为0.845。红松现阶段对偏低的遗传多样性水平与第四纪冰期所遭受的严重打击和人类近期的干扰有较大关系。  相似文献

A method is presented for estimating relative mutation rates or relative effective population sizes, under the hypothesis of adaptively neutral allelic variation. This method was applied to seven surveys of electrophoretic variation. It was observed that electrophoretic mutation rates so obtained follow the gamma distribution and, in Drosophila, are positively correlated with the molecular weights of the enzymes subunits. The variance in mutation rate is larger under the step-wise model of electrophoretic mutation than under the infinite-alleles model. Rates for the most variable loci may exceed rates for less variable loci by a factor of 500. For completely invariant loci, this factor may be as high as 4 X 10(4), an observation suggesting that these loci are subject to purifying selection. In contrast to mutation rates, effective population sizes may vary at the most by a factor of ten. These results support the hypothesis that differences in the amount of electrophoretic variability among polymorphic loci may reflect differences in the rate by which electrophoretically detectable variation is generated in population.  相似文献

Rapid expansion of microsatellite sequences in pines   总被引:9,自引:0,他引:9  
Microsatellite persistence time and evolutionary change was studied among five species of pines, which included a pair of closely related species (Pinus sylvestris and Pinus resinosa) in the subgenus Pinus, their relative Pinus radiata, and another closely related species pair (Pinus strobus and Pinus lambertiana) in the subgenus Strobus. The effective population sizes of these species are known to have ranged from the very small bottlenecks of P. resinosa to vast populations of P. sylvestris. This background allowed us to place the microsatellite evolution in a well-defined phylogenetic setting. Of 30 loci originating from P. strobus and P. radiata, we were able to consistently amplify 4 in most of the these pine species. These priming sites had been conserved for over 100 Myr. The four microsatellites were sequenced in the five species. Flanking sequences were compared to establish that the loci were orthologous. All microsatellites had persisted in these species, despite very different population sizes. We found a recent microsatellite duplication: a closely related pair of loci in P. strobus, where the other four species had just one locus. On two independent occasions, the repeat area of this same microsatellite (locus RPS 105a/b) had grown from a very low repeat number to 15 or 17 in the last 10-25 Myr. Other parts of the same compound microsatellite had remained virtually unchanged. Locus PR 4.6 is known to be polymorphic in both P. radiata and P. sylvestris, but the polymorphism in the two species is due to different motifs. The very large pine genomes are highly repetitive, and microsatellite loci also occur as gene families.  相似文献

BACKGROUND AND AIMS: Studying the genome structure of pines has been hindered by their large genomes and uniform karyotypes. Consequently our understanding of the genome organization and evolutionary changes in different groups of pines is extremely limited. However, techniques are now available that can surmount these difficulties. The purpose of this study was to exploit some of these techniques to characterize the genome differentiation between the two subgenera of Pinus: Pinus and Strobus. METHODS: Double-probe fluorescence in-situ hybridization (FISH) was used to localize the 5S and 18S rDNA loci on chromosomes of five species from the subgenus Strobus: P. bungeana, P. koraiensis, P. armandii, P. wallichiana and P. strobus. * KEY RESULTS: The rDNA FISH pattern varied considerably among the five species, with P. bungeana being the most distinct. By comparing the results obtained with those of previous rDNA FISH studies of members of the subgenus Pinus, several general features of rDNA loci distribution in the genus Pinus can be discerned: (a) species of subgenus Strobus generally have more rDNA loci than species of subgenus Pinus, correlating with their larger genomes in the subgenus Strobus; (b) there is a clear differentiation in 5S and 18S rDNA loci linkage patterns between the two subgenera; (c) variations in the rDNA FISH pattern correlate with phylogenetic relationships among species within the subgenus; (d) P. bungeana has fewer 18S rDNA sites than other pines investigated to date, but they give intense signals, and may reflect the primary distribution of the 18S-25S rDNA loci in the genus. CONCLUSIONS: The stable differentiation in rDNA FISH pattern between the subgenera suggests that chromosomal rearrangements played a role in the splitting of the two subgenera, and transpositional events rather than major structural changes are likely responsible for the variable rDNA distribution patterns among species of the same subgenus with conserved karyotypes.  相似文献

Electrophoretic patterns of blood proteins and enzymes have been analysed among 152 Arvicanthis, 75 Mastomys huberti and 13 Mastomys erytholeucus captured in 19 localities in Senegal. Individual variability (genetic polymorphism) was observed at four loci, while other loci appeared monomorphic. Differences in electrophoretic migration of some proteins were observed among populations originating from distant geographic areas. The overall trait in Arvicanthis is that individual variability is rather poorly developed while populations diverge first by the extent of polymorphism and the loci concerned and also by different electrophoretic behaviour of some proteins.  相似文献

The alterations on the electrophoretic patterns of albumin and globulin fractions in both embryo and endosperms of Pinus pinea seeds during artificial ageing have been investigated. The results show that the electrophoretic patterns of aged seeds are drastically different from those of seeds with high germination capacity. These alterations were observed even when the quantitative analysis did not show any significant changes.  相似文献
