Background

Food contaminated with fungi and their toxins is a problem that threatens many developing countries. Kingdom of Saudi Arabia depends on the exported grain and legume seeds.

Materials and methods

The study involved examination of 160 samples of rice and maize seeds collected from different locations in the Kingdom of Saudi Arabia. Heterogeneity in the 18s rRNA gene of toxigenic Alternaria sp. and Fusarium sp. was unraveled. The seeds were disinfected and cultured on Potato Dextrose Agar (PDA), Yeast Extract Sucrose (YES) media and incubated at 25?°C/7?days. The isolated fungi were subjected to 18s rRNA gene sequencing. Five toxins were extracted from maize and rice grains infected with isolated fungi.

Results

The isolated fungi were identified based on morphological and spores characters as Fusarium sp. and Alternaria sp. Molecular identification based on18s rDNA barcode' was performed due to its high degree of inter specific variability, conserved primer sites and multi-copy nature in the genome. Fusarium sp. produced the highest detected (2070?μg/kg) fumonisin especially in cereal production season 2011. The collected grain from Dammam recorded the highest percentage (5485.2?g/kg) of toxins.

Conclusion

This work highlights that 50% of samples were found contaminated with toxins in various concentrations which impose a threat for public health and necessitate rapid identification methods for toxigenic fungi such as 18s rDNA sequencing.  相似文献

Objective

This study identified the biological role of miR-4728 in Burkitt lymphoma (BL) process.

Methods

Ramos cells were used to analyze MicroRNA-4728 (miR-4728) biological functions. MiR-4728 expression was investigated in 14 randomly chosen tumor tissues and 12 noncancerous tissues by qRT-PCR. Cyquant assay was used to monitor cell proliferation. Colony formation assay was performed to study the effectiveness of miR-4728 on the proliferation of cells. The effects of miR-4728 on MAPK signaling pathway were detected by luciferase reporter assay. The significance of differences between groups were evaluated by SPSS.

Results

In this study, MiRNA-4728 was observed to down-regulated in BL tissues compared to the noncancerous tissues. Additionally, miR-4728 inhibited Ramos cell proliferation. Moreover, miR-4728 overexpression also decreased the MAPK signaling activity.

Conclusion

Our results suggested that miR-4728 serves as a suppressor and antagonist of oncogenic MAPK in Burkitt lymphoma. The appropriate regulation of miR-4728 might be vital to improve BL treatment.  相似文献

Objective

To compare and analyze three therapies on patients with primary central nervous system lymphoma (PCNSL), aiming to provide evidences for future treatment and prognosis.

Methods

Clinical data of 26 cases of PCNSL with normal immune system confirmed by postoperative pathology were retrospectively analyzed. Among them there were six cases with operation only, nine cases with operation and radiotherapy, and 11 cases with operation, radiotherapy and chemotherapy, and their survival rate was compared as well.

Results

The survival time of patients with operation only, operation combined with radiotherapy and operation combined with radiotherapy and chemotherapy was 6–11?months, 15–24?months and 24–51?months, respectively. And their median survival time was only nine months, 21?months and 38?months, respectively.

Conclusions

Operation combined with radiotherapy and chemotherapy can dramatically extend PCNSL patients’ survival time, therefore, it can be regarded as the first-line therapy.  相似文献

Background

Oral squamous cell carcinoma (OSCC) oncogenic mechanisms still remain elusive. Herein, we proposed to understand the biological role of a newly discovered OSCC miRNA.

Methods

MiR-1297 related to OSCC was selected for this study. TaqMan qRT- PCR assay was used to profile miRNA and gene expression in 16 tumors with matched adjacent non-tumor tissues. CCK-8 assay and colony formation assay were used to demonstrate cell proliferation. Protein level was determined by western blotting. SPSS was used for statistical analyses.

Results

MiR-1297 is repressed and PTEN activated in OSCC. Moreover, the miR-1297 overexpressing cell lines displayed a decrease in cell growth rate. And, suppression of miR-1297 reversed the cell growth rate. In addition, PTEN silencing display the similar pattern as miRNA-1297 overexpression to enhance OSCC cell growth.

Conclusions

MiRNA and gene expression changes are common event in OSCC. Our results suggest miR-1297 may drive tumor progression through PTEN. And, miR-1297 could be a promising candidate for future investigation.  相似文献

Aim

To test the fitness-gain curve model proposes that cosexual plants adjust their sex ratios and resource allocation depending on their size. In this study, the monoecious species Sagittaria potamogetifolia was used as a model to determine the effects of plant size and density on gender modification and reproductive allocation.

Methods and materials

Various traits, including flower number and plant biomass, were measured under four different artificially constructed population density treatments. More male flowers were produced than female flowers per individual at high densities, while the opposite trend was observed at low densities. This trend was particularly evident in the highest density treatment.

Results

A trade-off was discovered between male–female sex allocations in the highest density treatment (40 individuals m?2). The allometric growth of reproductive organs compared with plant size was detected, as evidenced by the reproductive structures’ biomass and flower numbers. However, in the highest density treatment, size was weakly negatively correlated with femaleness.

Conclusion

Thus, S. potamogetifolia has a reproductive strategy that easily adjusts to different reproductive environmental densities.  相似文献

Purpose

To investigate the effect of Fructus schisandrae syrup on bronchial asthma mice model.

Methods

Sixty Kunming mice were randomly divided into normal control group, bronchial asthma model group, low-, middle-, and high-dose Fructus schisandrae syrup groups. Bronchial asthma was induced by injection of ovalbumin combined smoking. Two hours after the last administration, the change of lung function were observed, the contents of NO, IL-6 in serum were detected, the morphological changes of lung and bronchial were also observed, so as to explore the effect of Fructus schisandrae syrup on bronchial asthma mice.

Results

Compared with model group, the Schisandrae Fructus syrup groups can significantly increase the tidal volume of mice and decrease the respiratory frequency and the degree of bronchial stenosis (P?<?0.01); The Schisandrae Fructus syrup groups can decrease the levels of NO and IL-6 in serum and improve the pathological changes of lung and bronchus in different degrees.

Conclusion

Fructus schisandrae syrup can significantly improve the biochemical indexes and pathological status of mice with bronchial asthma.  相似文献

Purpose

Explore the effect of Baihe Dihuang powder on chronic stress depression rat models.

Methods

Chronic stress depression rat models were established with different stimuli for 21?days. At the same time, the drug was administered for 21 consecutive days. The animals were weighed once a week after the start of the formal experiment. On the second day after the end of drug administration, conduct sugar water consumption test and open-filed box experiment, and conduct behavioral observation; At the end of behavioral testing, blood was taken from the eyeball and plasma was separated to measure MDA level and erythrocyte SOD activity; Take brain for homogenate, then measure the contents of 5-HT, NE and DA in brain tissue homogenate; Take the thymus and spleen, stained with 10% formalin fixation, embedding and HE staining, then use microscope to observe the histopathological changes.

Results

Chronic stress depression rats model replicated successfully. Each group of given drugs could increase the weight, the consumption of sugar water, and improve the behavioral score, increase erythrocytes SOD activity and decrease MDA level of plasma, increase the content of 5-HT, NE and DA of brain homogenate, and improve the pathological changes of thymus and spleen of chronic stress depression model animals.

Conclusion

Chronic stress depression rat model replicates successfully. Baihe Dihuang powder can interfere chronic stress depression rats model through different action pathways.  相似文献

Objective

To investigate the correlation between the level of glucose in serum and the development of acute pancreatitis (AP).

Methods

Data of 153 AP cases were collected, in which there were 130 patients with mild AP (MAP), 4 with moderate-severe AP (MSAP) and 19 with severe AP (SAP). At the time of admission, following indexes of patients were recorded: glucose, APACHE II score, TNF-α and C-reaction protein (CRP).

Results

At the time of admission, the levels of glucose in serum and APACHE II scores in the MSAP and SAP groups were significantly higher than those in the MAP group, but after treatment, the level of glucose in serum was recovered in 95.8% of the patients in the MAP group, while this digit in the SAP group remained to be 68.4%; in the SAP group, the levels of TNF-α and CRP in patients with sustained hypertension were significantly higher than those with non-persistent hypertension; in terms of the length of stay in hospital, the SAP group was shorter than that in the non-treatment group, and the difference had statistical significance (p?<?0.05). Moreover, we found that the level of glucose in serum was positively correlated with the APACHE II scores, TNF-α and CRP.

Conclusion

Glucose level in serum can be used as one of the indicators for evaluating the severity and development of AP in clinical practice.  相似文献

Objective

Observe anti-inflammatory effect and the effect on acute pharyngitis rats model induced by ammonia water of compound Lobelia oral liquid, providing experimental basis for its clinical use.

Methods

Use egg white establish foot swelling rats model and use carboxymethyl cellulose establish white blood cell migration rats model. Then observe the anti-inflammatory effect of compound Lobelia oral liquid. Use 15% ammonia spray at pharyngeal establish acute pharyngitis rats model, Visual observation and conduct grading of pharyngeal tissue stimulation in rats, measure the levels of TNF-α and IL-6 in serum. Pharyngeal tissue was taken to observe the morphological changes.

Result

All dose groups of compound Lobelia oral liquid can reduce the rate of foot swelling of rats at all time points (P?<?0.01 or P?<?0.05), and significantly reduce the number of white blood cells of rats (P?<?0.01); And improve the local hyperemia degree, reduce secretion, reduce local swelling of pharyngeal tissue, reduce the serum TNF-α and IL-6 levels of acute pharyngitis rats with different degrees (P?<?0.01 or P?<?0.05).

Conclusion

Compound Lobelia oral liquid has a good anti-inflammatory effect on foot swelling and white blood cell migration rats model, as well as significant improvement effect on acute pharyngitis rats model.  相似文献

Objective

To explore the expression level of FGF5 in the peripheral blood of primary hypertension patients and its clinical significance.

Methods

The 34 patients with primary hypertension treated in this hospital from June 2012 to June 2014 were selected as the observation group, while the 25 patients at this hospital who had physical exam with heathy results were selected as control group. Venous blood was drawn early in the morning after an overnight fast. FGF5, mRNA and protein level changes in the peripheral blood cells and peripheral blood serum were analyzed by real-time fluorescence based quantitative PCR (RT-PCR) and enzyme-linked immunosorbent assay (ELISA). FGF5 gene SNP (rs16998073) were amplified by PCR and inserted into T vector, and its genetic variation were analyzed by sequencing. The relationship of FGF5 protein levels and genetic variation with diastolic/systolic blood pressure was also analyzed.

Results

Comparing with the control group, the observation group’s FGF5 mRNA and protein levels significantly increased in the peripheral blood cells and peripheral blood. The difference was statistically significant (P?<?.05). Correlation analysis showed that FGF5 protein level and systolic/diastolic blood pressure were positively correlated (P?<?.05). T/A genetic variation of FGF5 gene SNP (rs16998073) and diastolic/systolic blood pressure were positively correlated (P?<?.05).

Conclusion

The FGF5 mRNA and protein expression levels of the patients with primary hypertension were abnormal and had genetic variation, which were associated with blood pressure of the patients with primary hypertension.  相似文献

Introduction

Thrombotic and inflammatory mechanisms are involved in the pathophysiology of acute coronary syndrome (ACS). The aim of the study was the evaluation of inflammation (white blood cells count/WBC, C-reactive protein/CRP, interleukin-6/IL-6) and platelet (platelet count/PLT, mean platelet volume/MPV, large platelet/LPLT, beta-thromboglobulin/β-TG) biomarkers in the groups of ACS patients depending on the severity of signs and symptoms and compared to controls without coronary artery disease.

Materials and methods

The study group included 93 patients categorized into 3 subgroups depending on the severity of signs and symptoms of ACS. PLT, MPV, LPLT, and WBC were determined on hematological analyzer, IL-6 and β-TG were measured using the ELISA method.

Results

In the whole group of ACS patients WBC, CRP, IL-6, MPV, and β-TG were significantly higher as compared to controls. Analyzing the inflammation and platelet biomarkers depending on the severity of signs and symptoms in comparison to controls, statistically significant differences for above-mentioned parameters were also found. There were no significant differences between the advancement of coronary artery changes and inflammation as well as platelet parameters, except for CRP concentrations. The AUCs for all inflammation parameters tested were similar, however the highest AUCs showed WBC and CRP. Among platelet parameters the highest AUC revealed β-TG.

Conclusion

Markers of inflammation and platelet activation may be associated to myocardial ischemia and myocardial injury. WBC, CRP and IL-6 as inflammation parameters and MPV and β-TG as platelet biomarkers may be useful indicators of the presence of coronary artery disease.  相似文献

Background

The current study was designed to investigate the effect of crocetin on the proliferation inhibition of colon cancer cells and the underlying mechanism.

Methods

MTT assay showed inhibition of proliferation of colon cancer cells in a dose based manner by crocetin treatment. At 30 µM concentration of crocetin proliferation rate of colon cancer cells was reduced to 14% after 24 h. Flow cytometry and fluorescence microscopy revealed induction of apoptosis in colon cancer cells on treatment with crocetin. The tube formation was suppressed significantly in the cultures of HUVEC treated with 30 µM concentration of crocetin compared to the control cultures.

Results

The results from transwell assay revealed a significant reduction in the population of DU-145 cells passing through filters of transwell on treatment with crocetin compared to the control cells. Treatment of the DU-145 cells with crocetin caused a significant reduction in the expression levels of NF-κB, VEGF and MMP-9. The results from RT-PCR analysis revealed a significant reduction in the expression of genes involved in inflammation including, HMGB1, IL-6 and IL-8 on treatment of DU-145 cells with crocetin. However, the expression of NAG-1 gene was increased by crocetin treatment in DU-145 cells significantly compared to the control cells.

Conclusion

Crocetin inhibits growth of colon cancer cells and prevents tube formation through induction of apoptosis. Therefore, crocetin can be used efficiently for the treatment of colon cancer.  相似文献

Purpose

To obtain protein hydrolysates from fresh water green algae Scenedesmus obliquus by three different enzymes and evaluate its antioxidant and antiviral activity.

Methods

Enzymatic hydrolysates of green algae Scenedesmus obliquus protein were prepared by treatment with: 1.2% solution of pepsin, trypsin or papain. Protein was extracted from S. obliquus by three different extraction methods. Protein extracts and hydrolysates were assessed from stained gels following SDS–PAGE of samples. Antioxidant activity of protein hydrolysates was investigated.

Results

S. obliquus cells and protein extracts were rich in Arg, Lys, Asp, Ala, and His. Protein hydrolyzed by papain (Sd1pa) and protein hydrolyzed by trypsin (Sd2Try) induced highest antioxidant activity based on 1,1-diphenyl-2-picryl-hydrazyl (DPPH) radical-scavenging (41.41% and 40.62%) respectively, and on 2,2′-azinobis 3-ethyl-benzothiazoline-6-sulphonate (ABTS) radical (87.03% and 45.12%) respectively, at 150?µg/ml. The inhibitory effect and mode of action of protein hydrolysates were evaluated against Coxsackie B3 virus (CVB3). Protein hydrolyzed by papain (Sd2pa) and protein hydrolyzed by pepsin (Sd1pep) at 100?µg/ml exhibited antiviral activity (66.2% and 57.6%, respectively), against (CVB3) from all protein hydrolysates.

Conclusion

S. obliquus protein hydrolysates have a potential as antioxidative neutraceutical ingredients and a potential therapeutic agent against CVB3.  相似文献

The phosphatidylinositol 3-kinases (PIK3s) are lipid kinases. Mutation in the exon 9 and exon 20 determined as a predictive factor in anti-HER-2 therapy. In some countries, such as Singapore, China, and Peru, PIK3CA exon 9 E545A was reported to produce the highest rate of mutation. In this research, we developed and optimized PIK3CA exon 9 E545A detection methods with intercalating dye SYBR Green I based on the Tm Shift approach by using prepared recombinant plasmid pGEMT-easy PIK3CA exon 9 and PIK3CA exon 9 E545A. Recombinant plasmid was used due to the limited number of samples.

Methods

Recombinant plasmid was prepared based on manufactured procedures, and this process was then followed by Tm prediction with Poland software, Tm Shift SYBR Green I development, and its characterization (reproducibility, repeatability, sensitivity, qPCR efficiency, and qPCR amplification), respectively.

Result

A method for PIK3CA E545A detection based on TM shift SYBR Green I has been successfully developed. The melting temperature for PIK3CA exon 9 was 78.1 ± 0.1 °C, while that for PIK3CA exon E545A was 80.20 °C. The Tm of mutant was the same as that predicted using Polland Software. The reproducibility of the methods was high, with the coefficient values for inter and intra assays were below 10% with a high sensitivity at 1%, while R2 0.99 and PCR efficiency was 97.75%.

Conclusion

The results presented here demonstrate that the PIK3CA exon 9 E545A detection method has a good sensitivity and efficacy assay, which proves that the method has a high diagnostic accuracy in breast cancer.  相似文献

Purpose

To explore how the use of a laser needle-knife affects vertebroarterial morphology and protein expression of PI-3K, AKT and VEGF in the carotid artery of a rabbit model of cervical spondylotic arteriopathy (CSA), and to determine its primary treatment mechanism.

Methods

The CSA rabbit model consisted of 30 rabbits with CSA that were randomly divided into a model group (n?=?10), an acupuncture group (n?=?10) and a laser needle-knife group (n?=?10) plus a further 10 wild type rabbits placed into a normal group. Rabbits in the acupuncture group were treated by needling the Fengchi (GB 20) and Jiaji (EX-B2) acupoints, while those in the laser needle-knife group were treated with a laser needle-knife on the Jiaji (EX-B2) near the spinous process of the fifth cervical vertebra, the rabbit in both groups immobilized during treatment. Rabbits in the other two groups received no treatment, but were immobilized for a similar duration. The morphology of the right vertebral artery and the distribution of pore size before and after treatment were compared using BET and SEM, and the protein expression of PI-3K, AKT and VEGF in the carotid artery of the four groups measured using Western blot analysis.

Results

The pore diameter and specific surface area of the right vertebral artery increased after treatment, as did the number of micropores. Compared with the normal group, the protein expression of PI-3K, AKT and VEGF in the carotid artery of the model group was significantly lower (P?<?0.05), while that of the laser needle-knife group was significantly higher (P?<?0.05 and P?<?0.01). In addition, rabbits from the acupuncture and laser needle-knife groups demonstrated significantly higher levels of protein expression of PI-3K and VEGF in the carotid artery compared with the model group (P?<?0.05 and P?<?0.01).

Conclusions

By promoting micrangium hyperplasia within the vertebral artery of rabbits with CSA, treatment with a laser needle-knife modified the protein expression of PI-3K, AKT and VEGF, suggesting that laser needle-knife therapy possibly treats CSA though these signaling pathways.  相似文献

Objective

Anti-inflammatory activity of rhein in animal models with potential mechanism of actions.

Methods

Rhein was isolated from Cassia fistula L. flowers collected in Chennai, Tamil Nadu, India. Its anti-inflammatory activity was then investigated in Wistar rats and mice using carrageenan-induced hind paw oedema, croton oil-induced ear oedema, cotton pellet-induced granuloma and acetic acid-induced vascular permeability models.

Results

Administration of rhein (10, 20, 40 mg/kg) significantly (p < 0.05) inhibited carrageenan-induced paw oedema in rats and croton oil-induced ear oedema in mice in dose-dependent manners. Continual administration of rhein to rats using implanted cotton pellets significantly (p < 0.05) reduced granuloma formation (20 mg/kg: 17.24%; 40 mg/kg: 36.12%) compared to control group animals. Administration of rhein increased the activities of catalase (CAT), superoxide dismutase (SOD) and glutathione peroxidase (GSH-px) and decreased the levels of nitrite, interleukin-6 (IL-6), interleukin-1β (IL-1β), tumor necrosis factor-α (TNF-α), malondialdehyde (MDA) and vascular endothelial growth factor (VEGF) compared to control animals. Western blotting results revealed that rhein diminished carrageenan-induced cyclooxygenase (COX)-2 and inducible nitric oxide synthase (iNOS) and increased heme oxygenase (HO)-1, nuclear factor erythroid 2–related factor 2 (Nrf2), peroxisome proliferator-activated receptor gamma (PPAR)-γ and heat shock protein (HSP)-72 expression after 6 h in the paw oedema model.

Conclusion

The anti-inflammatory mechanisms of rhein might be related to decrease in the levels of MDA, iNOS and COX-2 and the stimulation of HO-1, PPAR-γ and Nrf2 expression via increases in the activities of CAT, SOD and GSH-px through the suppression of nitrite, TNF-α, IL-6 and IL-1β.  相似文献

Objective

Study the effect of the 3:7 ratio of Astragalus total saponins and Curcumin on the model of diabetic nephropathy rats, and explore its mechanisms.

Methods

Diabetic nephropathy rats model was established by high-fat and high-sugar feed feeding combined with streptozotocin (STZ) injection in sublingual vein. Measured fasting blood glucose of rats on the 10, 20 and 30th day, and measured urine protein content in urine of rats on 30th days. Two hours after the last administration, measured glycated serum protein (GSP), insulin antibody (IA), triglyceride (TG), total cholesterol (TC), low density lipoprotein (LDL), high density lipoprotein (HDL), malondialdehyde (MDA), insulin, superoxide dismutase (SOD), glutathione (GSH), urea nitrogen (BUN), creatinine (Cr) in the serum and calculated the renal index of rat. Take the viscera of pancreas and kidney, and HE staining, so as to observe pathological changes.

Result

Astragalus total saponins and Curcumin 3:7 compatibility each dose group can significantly reduce the diabetic nephropathy rats blood glucose of 30th days, significantly reduce the level of GSP, IA, TG, TC, LDL (P?<?0.01), and reduce MDA levels with different degrees (P?<?0.01 or P?<?0.05), and significantly increase the level of insulin (P?<?0.01), increase the level of HDL, SOD and GSH with different degrees (P?<?0.01 or P?<?0.05 or P?>?0.05); Astragalus total saponins and Curcumin 3:7 compatibility each dose group also can decrease renal index, UN, and Cr levels with different degrees and improve the pathological changes of pancreatic tissue and kidney tissue in diabetic nephropathy rats with different degrees (P?<?0.01 or P?<?0.05 or P?>?0.05).

Conclusion

The 3:7 ratio of Astragalus total saponins and Curcumin can achieve the treatment and protection effects on diabetic nephropathy rats by improve the glycometableolism, insulin resistance, lipid metableolism, oxidative stress levels, and pathological changes.  相似文献

Objective

To study the putative effects of Advanced Oxidation Protein Products (AOPPs) and Advanced Glycation End Products (AGEs) in the development and progression of cardiovascular disease (CVD).

Methodology

AGEs, AOPPs, e-NOS, lipid profile, circulating stress and inflammatory biomarkers were evaluated among fifty cardiovascular patients and fifty controls. Independent student’s t-test was done for statistical analysis.

Results

The malondialdehyde mean level in CVD patients (5.45?nmol/ml) was significantly higher than control (1.36?nmol/ml) (p value?=?0.018). Nitric oxide in CVD patients (55.72?ng/ml) was remarkably increased as compared to normal subjects (19.19?ng/ml). A significant change in the mean serum level of AGEs in CVD patients (2.74?ng/ml) and normal individuals (0.85?ng/ml) was recorded (p value?=?0.000). The AOPPs also showed significant increased levels in CVD group (132.07?ng/ml) in comparison with normal subjects (83.05?ng/ml) (p value?=?0.011). The mean eNOS serum level in CVD group (15.50?U/L) was higher than control group (11.28?U/L) (p value?=?0.004). Cardiovascular disease patients, in comparison with healthy controls, showed increased level of total cholesterol (5.48?mmol/L vs 4.45?mmol/L), triglycerides (2.59?mmol/L vs 1.24?mmol/L), and low density lipoprotein (2.47?mmol/L vs 2.31?mmol/L) along with decrease in high density lipoprotein (1.39?mmol/L vs 1.74?mmol/L). The mean MMP-11 serum levels in CVD group (98.69?ng/ml) was almost double of control group (45.60?ng/ml) (p value?=?0.017). The mean serum level of TNF-α and IL1-α were 32.16?pg/ml and 6.64?pg/ml in CVD patient. The significant decreasing trend of SOD (p value?=?0.041), CAT (p value?=?0.018), GSH (p value?=?0.036) and GRx (p value?=?0.029) but increasing drift of GPx (0.023) level was observed in CVD patients.

Conclusion

This study provides strong evidence that CVD patients presented with elevated oxidative stress, enhanced inflammation and lipid profile in their serum. Objective

To investigate the levels of blood fat, C-reactive protein (CRP) and hemorheological indicators in the elder patients with coronary heart disease (CHD), so as to provide evidence for prospective study and treatment of elder CHD.

Methods

We collected the clinical data of 127 elder CHD patients who admitted to this hospital between July 2016 and December 2017 to detect the levels of blood fat, CRP and hemorheological indicators.

Results

In elder CHD patients, levels of the total cholesterol (TC), triglyceride (TG) and low density lipoprotein cholesterin (LDL-C) were significantly higher than the normal reference, and comparison with the control group also showed significant increases (p?<?0.01); average levels of the high-density lipoprotein cholesterin (HDL-C), phospholipid (PL), lipoprotein a [LP (a)] and free fatty acid were in the range of normal reference. Abnormal levels of TC, TG, LDL-C and HDL-C were identified in 59.06%, 58.27%, 51.18% and 18.11% of the elder CHD patients, most of which were concomitant with obesity or hypertension, and levels of these indicators were significantly higher than those in the control group with statistically significant differences (p?<?0.01). Comparisons of the age, gender distribution, hypotension, exercise and sleep showed that differences had no statistical significance (p?>?0.05). In comparison with the control group, the levels of CRP, the whole blood viscosities at high and low shears, plasma viscosity, hematocrit value, aggregation index and rigidity index of red blood cells (RBC) were all higher than those in the control group, and the differences had statistical significance (p?<?0.01). However, the erythrocyte sedimentation rate (ESR), deformity index of RBC, blood flow rates in the bilateral middle cerebral arteries (MCA), anterior cerebral arteries (ACA), terminal internal carotid artery (TICA), posterior cerebral arteries (PCA), vertebral arteries (VA) and basilar artery (BA) were significantly lower than those in the control group, and the differences had statistical significance (p?<?0.05 or 0.01).

Conclusion

In elder CHD patients, anomaly is mainly seen in levels of TC, TG and LDL-C with concentrated, adhesive and aggregating blood.  相似文献

Objective

Explore the possible protective effect of Sargentodoxa cuneata total phenolic acids on cerebral ischemia reperfusion injury rats.

Methods

Focal cerebral ischemia reperfusion rats model were established by linear thrombus. Nimodipine group, Naoluotong group, the high, middle and low dose of Sargentodoxa cuneata total phenolic acids groups were given related drugs via intragastric administration before operation for seven days, once a day. At the same time sham operation group, and ischemia reperfusion group were given the same volume of physiological saline. One hour after the last administration, establish focal cerebral ischemia- reperfusion model in rats by thread method, and the thread was taken out after 2?h ischemia to achieve cerebral ischemia reperfusion injury in rats. After reperfusion for 24?h, the rats were given neurologic deficit score. The brain tissue was taken to measure the levels of IL-6, IL-1β, TNF-α, Bcl-2, Bax, Casp-3 and ICAM-1; HE staining observed histopathological changes in the hippocampus and cortical areas of the brain; Immunohistochemistry was used to observe the expression of NGF and NF-KBp65.

Result

Focal cerebral ischemia reperfusion rats model was copyed successed. Compared with model group, each dose group of Sargentodoxa cuneata total phenolic acids could decreased the neurologic deficit score (P?<?0.05 or P?<?0.01), decreased the levels of IL-6, IL-1β, ICAM-1, TNF-α, Bax and Caspase-3 in brain tissue (P?<?0.05 or P?<?0.01), increased the levels of IL-10, Bcl-2, NGF in brain tissue (P?<?0.05 or P?<?0.01), decreased the express of NF-KBp65 in brain (P?<?0.05 or P?<?0.01).

Conclusion

Sargentodoxa cuneata total phenolic acids can improve focal cerebral ischemia reperfusion injury rats tissue inflammation, apoptosis pathway, increase nutrition factor to protect the neurons, reduce the apoptosis of nerve cells, activate brain cells self-protect, improve the histopathological changes in the hippocampus and cortical areas of the brain, reduce cerebral ischemia reperfusion injury.  相似文献
