The maximum carboxylation capacity of Rubisco, Vc,max, is an important photosynthetic parameter that is key to accurate estimation of carbon assimilation. The gold‐standard technique for determining Vc,max is to derive Vc,max from the initial slope of an ACi curve (the response of photosynthesis, A, to intercellular CO2 concentration, Ci). Accurate estimates of Vc,max derived from an alternative and rapid “one‐point” measurement of photosynthesis could greatly accelerate data collection and model parameterization. We evaluated the practical application of the one‐point method in six species measured under standard conditions (saturating irradiance and 400 μmol CO2 mol?1) and under conditions that would increase the likelihood for successful estimation of Vc,max: (a) ensuring Rubisco‐limited A by measuring at 300 μmol CO2 mol?1 and (b) allowing time for acclimation to saturating irradiance prior to measurement. The one‐point method significantly underestimated Vc,max in four of the six species, providing estimates 21%–32% below fitted values. We identified ribulose‐1,5‐bisphosphate‐limited A, light acclimation, and the use of an assumed respiration rate as factors that limited the effective use of the one‐point method to accurately estimate Vc,max. We conclude that the one‐point method requires a species‐specific understanding of its application, is often unsuccessful, and must be used with caution.  相似文献

The environmental benefits of fuel cells and electrolyzers have become increasingly recognized in recent years. Fuel cells and electrolyzers that can operate at intermediate temperatures (300–450 °C) require, in principle, neither the precious metal catalysts that are typically used in polymer‐electrolyte‐membrane systems nor the costly heat‐resistant alloys used in balance‐of‐plant components of high‐temperature solid oxide electrochemical cells. These devices require an electrolyte with high ionic conductivity, typically more than 0.01 S cm?1, and high chemical stability. To date, however, high ionic conductivities have been found in chemically unstable materials such as CsH2PO4, In‐doped SnP2O7, BaH2, and LaH3?2xOx. Here, fast and stable proton conduction in 60‐at% Sc‐doped barium zirconate polycrystal, with a total conductivity of 0.01 S cm?1 at 396 °C for 200 h is demonstrated. Heavy doping of Sc in barium zirconate simultaneously enhances the proton concentration, bulk proton diffusivity, specific grain boundary conductivity, and grain growth. An accelerated stability test under a highly concentrated and humidified CO2 stream using in situ X‐ray diffraction shows that the perovskite phase is stable over 240 h at 400 °C under 0.98 atm of CO2. These results show great promises as an electrolyte in solid‐state electrochemical devices operated at intermediate temperatures.  相似文献

The present study investigated the effect of different culture conditions on the vegetative growth of a new species, Haematococcus alpinus (strain LCR‐CC‐261f) using airlift photobioreactors. The influence of culture medium, aeration rates, CO2 concentration in air‐gas mixture, temperature, light intensities, and wavelengths were investigated to achieve sustainable high cell density cultures. Growth parameters were determined by fitting the data to a form of the logistic equation that included a lag phase. The shear‐sensitive vegetative cells favored lower aeration rates in the photobioreactors. MLA medium increased to 40 mM nitrate produced high density cultures. Temperatures between 12°C and 18°C, 3% (v/v) CO2 concentration and a narrow photon flux density ranging between 37 and 48 μmol photons · m?2 · s?1 were best suited for growth. The wavelength of the light source also impacted growth and a high cell density of 9.6 × 105 cells · mL?1 was achieved using a mixture of red and blue compared to warm white, red, or blue LEDs.  相似文献

The unabated rise in atmospheric [CO2] is associated with increased air temperature. Yet, few CO2‐enrichment studies have considered pre‐industrial [CO2] or warming. Consequently, we quantified the interactive effects of growth [CO2] and temperature on photosynthesis of faster‐growing Eucalyptus saligna and slower‐growing E. sideroxylon. Well‐watered and ‐fertilized tree seedlings were grown in a glasshouse at three atmospheric [CO2] (290, 400, and 650 µL L?1), and ambient (26/18 °C, day/night) and high (ambient + 4 °C) air temperature. Despite differences in growth rate, both eucalypts responded similarly to [CO2] and temperature treatments with few interactive effects. Light‐saturated photosynthesis (Asat) and light‐ and [CO2]‐saturated photosynthesis (Amax) increased by ~50% and ~10%, respectively, with each step‐increase in growth [CO2], underpinned by a corresponding 6–11% up‐regulation of maximal electron transport rate (Jmax). Maximal carboxylation rate (Vcmax) was not affected by growth [CO2]. Thermal photosynthetic acclimation occurred such that Asat and Amax were similar in ambient‐ and high‐temperature‐grown plants. At high temperature, the thermal optimum of Asat increased by 2–7 °C across [CO2] treatments. These results are the first to suggest that photosynthesis of well‐watered and ‐fertilized eucalypt seedlings will remain strongly responsive to increasing atmospheric [CO2] in a future, warmer climate.  相似文献

Understanding of the extent of acclimation of light‐saturated net photosynthesis (An) to temperature (T), and associated underlying mechanisms, remains limited. This is a key knowledge gap given the importance of thermal acclimation for plant functioning, both under current and future higher temperatures, limiting the accuracy and realism of Earth system model (ESM) predictions. Given this, we analysed and modelled T‐dependent changes in photosynthetic capacity in 10 wet‐forest tree species: six from temperate forests and four from tropical forests. Temperate and tropical species were each acclimated to three daytime growth temperatures (Tgrowth): temperate – 15, 20 and 25 °C; tropical – 25, 30 and 35 °C. CO2 response curves of An were used to model maximal rates of RuBP (ribulose‐1,5‐bisphosphate) carboxylation (Vcmax) and electron transport (Jmax) at each treatment's respective Tgrowth and at a common measurement T (25 °C). SDS‐PAGE gels were used to determine abundance of the CO2‐fixing enzyme, Rubisco. Leaf chlorophyll, nitrogen (N) and mass per unit leaf area (LMA) were also determined. For all species and Tgrowth, An at current atmospheric CO2 partial pressure was Rubisco‐limited. Across all species, LMA decreased with increasing Tgrowth. Similarly, area‐based rates of Vcmax at a measurement T of 25 °C (Vcmax25) linearly declined with increasing Tgrowth, linked to a concomitant decline in total leaf protein per unit leaf area and Rubisco as a percentage of leaf N. The decline in Rubisco constrained Vcmax and An for leaves developed at higher Tgrowth and resulted in poor predictions of photosynthesis by currently widely used models that do not account for Tgrowth‐mediated changes in Rubisco abundance that underpin the thermal acclimation response of photosynthesis in wet‐forest tree species. A new model is proposed that accounts for the effect of Tgrowth‐mediated declines in Vcmax25 on An, complementing current photosynthetic thermal acclimation models that do not account for T sensitivity of Vcmax25.  相似文献

Sphingobium sp. strain SYK‐6 is able to use a phenylcoumaran‐type biaryl, dehydrodiconiferyl alcohol (DCA), as a sole source of carbon and energy. In SYK‐6 cells, the alcohol group of the B‐ring side chain of DCA was first oxidized to the carboxyl group, and then the alcohol group of the A‐ring side chain was oxidized to generate 5‐(2‐carboxyvinyl)‐2‐(4‐hydroxy‐3‐methoxyphenyl)‐7‐methoxy‐2,3‐dihydrobenzofuran‐3‐carboxylate (DCA‐CC). We identified phcF, phcG and phcH, which conferred the ability to convert DCA‐CC into 3‐(4‐hydroxy‐3‐(4‐hydroxy‐3‐methoxystyryl)‐5‐methoxyphenyl)acrylate (DCA‐S) in a host strain. These genes exhibited no significant sequence similarity with known enzyme genes, whereas phcF and phcG, which contain a DUF3237 domain of unknown function, showed 32% amino acid sequence identity with each other. The DCA‐CC conversion activities were markedly decreased by disruption of phcF and phcG, indicating that phcF and phcG play dominant roles in the conversion of DCA‐CC. Purified PhcF and PhcG catalysed the decarboxylation of the A‐ring side chain of DCA‐CC, producing DCA‐S, and showed enantiospecificity towards (+)‐ and (–)‐DCA‐CC respectively. PhcF and PhcG formed homotrimers, and their Km for DCA‐CC were determined to be 84 μM and 103 μM, and Vmax were 307 μmol?min?1?mg?1 and 137 μmol?min?1?mg?1 respectively. In conclusion, PhcF and PhcG are enantiospecific decarboxylases involved in phenylcoumaran catabolism.  相似文献

The effect of elevated UV‐B radiation on CO2 exchange of a natural flark fen was studied in open‐field conditions during 2003–2005. The experimental site was located in Sodankylä in northern Finland (67°22′N, 26°38′E, 179 m a.s.l.). Altogether 30 study plots, each 120 cm × 120 cm in size, were randomly distributed between three treatments (n=10): ambient control, UV‐A control and UV‐B treatment. The UV‐B‐treated plots were exposed to elevated UV‐B radiation level for three growing seasons. The instantaneous net ecosystem CO2 exchange (NEE) and dark respiration (RTOT) were measured during the growing season using a closed chamber method. The wintertime CO2 emissions were estimated using a gradient technique by analyzing the CO2 concentration in the snow pack. In addition to the instantaneous CO2 exchange, the seasonal CO2 balances during the growing seasons were modeled using environmental data measured at the site. In general, the instantaneous NEE at light saturation was slightly higher in the UV‐B treatment compared with the ambient control, but the gross photosynthesis was unaffected by the exposure. The RTOT was significantly lower under elevated UV‐B in the third study year. The modeled seasonal (June–September) CO2 balance varied between the years depending on the ground water level and temperature conditions. During the driest year, the seasonal CO2 balance was negative (net release of CO2) in the ambient control and the UV‐B treatment was CO2 neutral. During the third year, the seasonal CO2 uptake was 43±36 g CO2‐C m−2 in the ambient control and 79±45 g CO2‐C m−2 in the UV‐B treatment. The results suggest that the long‐term exposure to high UV‐B radiation levels may slightly increase the CO2 accumulation to fens resulting from a decrease in microbial activity in peat. However, it is unlikely that the predicted development of the level of UV‐B radiation would significantly affect the CO2 balance of fen ecosystems in future.  相似文献

Bypass of the penicillin‐binding proteins by an l ,d ‐transpeptidase (Ldtfm) confers cross‐resistance to β‐lactam and glycopeptide antibiotics in mutants of Enterococcus faecium selected in vitro. Ldtfm is produced by the parental strain D344S although it insignificantly contributes to peptidoglycan cross‐linking as pentapeptide stems cannot be used as acyl donors by this enzyme. Here we show that production of the tetrapeptide substrate of Ldtfm is controlled by a two‐component regulatory system (DdcRS) and a metallo‐d ,d ‐carboxypeptidase (DdcY). The locus was silent in D344S and its activation was due to amino acid substitutions in DdcS or DdcR that led to production of DdcY and hydrolysis of the C‐terminal d ‐Ala residue of the cytoplasmic peptidoglycan precursor UDP‐MurNAc‐pentapeptide. The T161A and T161M substitutions affected a position of DdcS known to be essential for the phosphatase activity of related sensor kinases. Complete elimination of UDP‐MurNAc‐pentapeptide, which was required specifically for resistance to glycopeptides, involved substitutions in DdcY that increased the catalytic efficiency of the enzyme (E127K) and affected its interaction with the cell envelope (I14N). The ddc locus displays striking similarities with portions of the van vancomycin resistance gene clusters, suggesting possible routes of emergence of cross‐resistance to glycopeptides and β‐lactams in natural conditions.  相似文献

Although there is now a considerable literature on the inhibition of leaf respiration (CO2 evolution) by light, little is known about the effect of other environmental conditions on day respiratory metabolism. In particular, CO2 and O2 mole fractions are assumed to cause changes in the tricarboxylic acid pathway (TCAP) but the amplitude and even the direction of such changes are still a matter of debate. Here, we took advantage of isotopic techniques, new simple equations and instant freeze sampling to follow respiratory metabolism in illuminated cocklebur leaves (Xanthium strumarium L.) under different CO2/O2 conditions. Gas exchange coupled to online isotopic analysis showed that CO2 evolved by leaves in the light came from ‘old’ carbon skeletons and there was a slight decrease in 13C natural abundance when [CO2] increased. This suggested the involvement of enzymatic steps fractionating more strongly against 13C and thus increasingly limiting for the metabolic respiratory flux as [CO2] increased. Isotopic labelling with 13C2‐2,4‐citrate lead to 13C‐enriched Glu and 2‐oxoglutarate (2OG), clearly demonstrating poor metabolism of citrate by the TCAP. There was a clear relationship between the ribulose‐1,5‐bisphosphate oxygenation‐to‐carboxylation ratio (vo/vc) and the 13C commitment to 2OG, demonstrating that 2OG and Glu synthesis via the TCAP is positively influenced by photorespiration.  相似文献

To determine the effects of elevated CO2 concentration ([CO2]) on the temperature‐dependent photosynthetic properties, we measured gas exchange and chlorophyll fluorescence at various leaf temperatures (15, 20, 25, 30, 35 and 40°C) in 1‐year‐old seedlings of the Japanese white birch (Betula platyphylla var. japonica), grown in a phytotron under natural daylight at two [CO2] levels (ambient: 400 µmol mol?1 and elevated: 800 µmol mol?1) and limited N availability (90 mg N plant?1). Plants grown under elevated [CO2] exhibited photosynthetic downregulation, indicated by a decrease in the carboxylation capacity of Rubisco. At temperatures above 30°C, the net photosynthetic rates of elevated‐CO2‐grown plants exceeded those grown under ambient [CO2] when compared at their growth [CO2]. Electron transport rates were significantly lower in elevated‐CO2‐grown plants than ambient‐CO2‐grown ones at temperatures below 25°C. However, no significant difference was observed in the fraction of excess light energy [(1 ? qP)× Fv′/Fm′] between CO2 treatments across the temperature range. The quantum yield of regulated non‐photochemical energy loss was significantly higher in elevated‐CO2‐grown plants than ambient, when compared at their respective growth [CO2] below 25°C. These results suggest that elevated‐CO2‐induced downregulation might not exacerbate the temperature‐dependent susceptibility to photoinhibition, because reduced energy consumption by electron transport was compensated for by increased thermal energy dissipation at low temperatures.  相似文献

2D vanadium carbide MXene containing surface functional groups (denoted as V2CTx , where Tx are surface functional groups) is synthesized and studied as anode material for Na‐ion batteries. V2CTx anode exhibits reversible charge storage with good cycling stability and high rate capability through electrochemical test. The charge storage mechanism of V2CTx material during Na+ intercalation/deintercalation and the redox reaction of vanadium are studied using a combination of synchrotron based X‐ray diffraction, hard X‐ray absorption near edge spectroscopy (XANES), and soft X‐ray absorption spectroscopy (sXAS). Experimental evidence of a major contribution of redox reaction of vanadium to the charge storage and the reversible capacity of V2CTx during sodiation/desodiation process are provided through V K ‐edge XANES and V L 2,3‐edge sXAS results. A correlation between the CO32? content and the Na+ intercalation/deintercalation states in the V2CTx electrode observed from C and O K ‐edge in sXAS results implies that some additional charge storage reactions may take place between the Na+‐intercalated V2CTx and the carbonate‐based nonaqueous electrolyte. The results of this study provide valuable information for the further studies on V2CTx as anode material for Na‐ion batteries and capacitors.  相似文献

Recombination between repeated DNA elements in the genomes of Mycoplasma species appears to lie at the basis of antigenic variation of several essential surface proteins. It is therefore imperative that the DNA recombinatorial pathways in mycoplasmas be unravelled. Here, we describe the proteins encoded by the Mycoplasma genitalium MG352 and Mycoplasma pneumoniae MPN528a genes (RecUMge and RecUMpn respectively), which share sequence similarity with RecU Holliday junction (HJ) resolvases. RecUMge was found to: (i) bind HJ substrates and large double‐stranded DNA molecules and (ii) cleave HJ substrates at the sequence 5′‐G/TC↓C/TTA/GG‐3′ in the presence of Mn2+. Interestingly, RecUMpn(from M. pneumoniae subtype 2 strains) did not possess obvious DNA binding or cleavage activities, which was found to be caused by the presence of a glutamic acid residue at position 67 of the protein, which is not conserved in RecUMge. Additionally, RecUMpn appears not to be expressed by subtype 1 M. pneumoniae strains, as these possess a TAA translation termination codon at position 181–183 of MPN528a. We conclude that RecUMge is a HJ resolvase that may play a central role in recombination in M. genitalium.  相似文献

Rising global CO2 is changing the carbonate chemistry of seawater, which is expected to influence the way phytoplankton acquire inorganic carbon. All phytoplankton rely on ribulose‐bisphosphate carboxylase oxygenase (RUBISCO) for assimilation of inorganic carbon in photosynthesis, but this enzyme is inefficient at present day CO2 levels. Many algae have developed a range of energy demanding mechanisms, referred to as carbon concentrating mechanisms (CCMs), which increase the efficiency of carbon acquisition. We investigated CCM activity in three southern hemisphere strains of the coccolithophorid Emiliania huxleyi W. W. Hay & H. P. Mohler. Both calcifying and non‐calcifying strains showed strong CCM activity, with HCO3? as a preferred source of photosynthetic carbon in the non‐calcifying strain, but a higher preference for CO2 in the calcifying strains. All three strains were characterized by the presence of pyrenoids, external carbonic anhydrase (CA) and high affinity for CO2 in photosynthesis, indicative of active CCMs. We postulate that under higher CO2 levels cocco‐lithophorids will be able to down‐regulate their CCMs, and re‐direct some of the metabolic energy to processes such as calcification. Due to the expected rise in CO2 levels, photosynthesis in calcifying strains is expected to benefit most, due to their use of CO2 for carbon uptake. The non‐calcifying strain, on the other hand, will experience only a 10% increase in HCO3?, thus making it less responsive to changes in carbonate chemistry of water.  相似文献

Aims: Algae are favourable as a biofuel source because of the potential high oil content and fast generation of biomass. However, one of the challenges for this technology is achieving high oil content while maintaining exponential or high growth of the organism. Introducing a two‐stage reactor to optimize both growth and oil content of the algae could be a solution to this hurdle. The aim of this study was to determine the reactor design parameters of the first‐stage reactor, which would optimize growth of two algal strains, Oocystis sp. and Amphora sp. Methods and Results: Growth kinetics were monitored by in vivo fluorescence and correlated to dry mass for both cultures under several environmental conditions during exponential growth. Temperatures of 25 and 30°C and light intensities of 150 and 80 μmol m?2 s?1 provided the most robust growth for Oocystis sp. and Amphora sp., respectively. Both strains showed optimized growth at a light : dark cycle of 16 : 08. At these conditions, the doubling rate for Oocystis sp. was 0·333 d?1 and for Amphora sp. was 0·179 d?1. Conclusions: For both cultures, growth rate was more dependent on light : dark cycle and temperature than light intensity. Both strains grew slower in this work than data reported in the literature, however agitation and air/CO2 sparging were not incorporated in the system under study. The highest doubling rate for Amphora sp. was observed near the maximum tolerable temperature, and it is suggested to grow this strain at 30°C for a consistent high growth rate. Significance and Impact of Study: Optimized growth conditions were determined for two lipid producing strains identified in the Aquatic Species Program summary report. An optimized, first‐stage growth reactor operating at these conditions would thus offer the maximum productivity for an algal biomass feed stream into a lipid‐optimized second‐stage reactor.  相似文献

The purification and characterization of psychro‐thermoalkalistable protease from psychrotrophic Pseudomonas putida isolate is being reported for the first time. A ~53 kDa protease was purified 21.4‐folds with 57.2% recovery by ultrafiltration and hydrophobic interaction chromatography. Kinetic analyses revealed the Km and Vmax to be 1.169 mg mL?1 and 0.833 mg mL?1 min?1, respectively. The kcat value of 3.05 × 102 s?1 indicated high affinity and catalytic efficiency toward casein. The protease was most active at pH 9.5 and 40°C, with 100% stability in pH and temperature range of 6.0–11.0 and 10–40°C, respectively. Presence of Zn2+ increased the thermostability of protease (at 70°C) by 433%. Ethylene diamine tetra acetic acid (EDTA) and 1,10‐phenanthroline were inhibitory, whereas phenyl methyl sulfonyl fluoride (PMSF), p‐chloro mercuric benzoate (PCMB), and β‐mercaptoethanol were ineffective, revealing the enzyme to be a metalloprotease. Zinc, calcium, iron, nickel, and copper at 1 mM increased the enzyme activity (102–134%). Complete reversion of enzyme inhibition (caused by Ethylene diamine tetra acetic acid [EDTA]) by Zn2+ affirmed this enzyme as zinc‐dependent metalloprotease. At 0.1% concentration, Triton X‐100 and Tween 80 slightly increased, while SDS and H2O2 reduced the protease activity. In the presence of 0.1% commercial detergents, the enzyme was fairly stable (54–81%). In the presence of organic solvent, the protease was remarkably stable exhibiting 72–191% activities. In contrast, savinase exhibited good stability in the presence of hydrophilic solvents, while chymotrypsin showed elevated activities with benzene, toluene, and xylene only. Circular dichroism analysis revealed the protease as a β‐rich protein, having large fraction (~40%) of β‐sheets. Presence of different environmental conditions altered the β‐content, which accordingly affected the protease activity. © 2012 American Institute of Chemical Engineers Biotechnol. 3‐Hydroxypropionic acid (3‐HP) is an important platform chemical that can be used to synthesize a range of chemical compounds. A previous study demonstrated that recombinant Escherichia coli stains can produce 3‐HP from glycerol in the presence of vitamin B12 (coenzyme B12), when overexpressed with a coenzyme B12‐dependent glycerol dehydratase (DhaB) and an aldehyde dehydrogenase. The present study examined the production of 3‐HP in recombinant Klebsiella pneumoniae strains, which naturally synthesizes vitamin B12 and does not require supplementation of the expensive vitamin. The NAD+‐dependent gamma‐glutamyl‐gamma‐aminobutyraldehyde dehydrogenase (PuuC) of K. pneumoniae alone or with its DhaB was overexpressed homologously, and two major oxidoreductases, DhaT and YqhD, were disrupted. Without vitamin B12 addition, the recombinant K. pneumoniae ΔdhaTΔyqhD overexpressing PuuC could produce ~3.8 g/L 3‐HP in 12 h of flask culture. However, this was possible only under the appropriate aeration conditions; 1,3‐propanediol (1,3‐PDO) (instead of 3‐HP) was mainly produced when aeration was insufficient, whereas a very small amount of both 3‐HP and 1,3‐PDO were produced when aeration was too high. The production of a small amount of 3‐HP under improper aeration conditions was attributed to either slow NAD+ regeneration (under low aeration) or reduced vitamin B12 synthesis (under high aeration). In a glycerol fed‐batch bioreactor experiment under a constant DO of 5%, the strain, K. pneumoniae ΔdhaTΔyqhD, overexpressing both PuuC and DhaB could produce >28 g/L 3‐HP in 48 h with a yield of >40% on glycerol. Only small amount of 3‐HP was produced when cultivation was carried out at a constant aeration of 1 vvm or constant 10% DO. These results show that K. pneumoniae is potentially useful for the production of 3‐HP in an economical culture medium that does not require vitamin B12. The results also suggest that the aeration conditions should be optimized carefully for the efficient production of 3‐HP while using this strain. Biotechnol. The Arctic is particularly sensitive to climate change, but the independent effects of increasing atmospheric CO2 concentration (pCO2) and temperature on high‐latitude forests are poorly understood. Here, we present a new, annually resolved record of stable carbon isotope (δ13C) data determined from Larix cajanderi tree cores collected from far northeastern Siberia in order to investigate the physiological response of these trees to regional warming. The tree‐ring record, which extends from 1912 through 1961 (50 years), targets early twentieth‐century warming (ETCW), a natural warming event in the 1920s to 1940s that was limited to Northern hemisphere high latitudes. Our data show that net carbon isotope fractionation (Δ13C), decreased by 1.7‰ across the ETCW, which is consistent with increased water stress in response to climate warming and dryer soils. To investigate whether this signal is present across the northern boreal forest, we compiled published carbon isotope data from 14 high‐latitude sites within Europe, Asia, and North America. The resulting dataset covered the entire twentieth century and spanned both natural ETCW and anthropogenic Late Twentieth‐Century Warming (~0.7 °C per decade). After correcting for a ~1‰ increase in Δ13C in response to twentieth century pCO2 rise, a significant negative relationship (r = ?0.53, P < 0.0001) between the average, annual Δ13C values and regional annual temperature anomalies is observed, suggesting a strong control of temperature on the Δ13C value of trees growing at high latitudes. We calculate a 17% increase in intrinsic water‐use efficiency within these forests across the twentieth century, of which approximately half is attributed to a decrease in stomatal conductance in order to conserve water in response to drying conditions, with the other half being attributed to increasing pCO2. We conclude that annual tree‐ring records from northern high‐latitude forests record the effects of climate warming and pCO2 rise across the twentieth century.  相似文献

Effects of growth light intensity on the temperature dependence of CO2 assimilation rate were studied in tobacco (Nicotiana tabacum) because growth light intensity alters nitrogen allocation between photosynthetic components. Leaf nitrogen, ribulose 1·5‐bisphosphate carboxylase/oxygenase (Rubisco) and cytochrome f (cyt f) contents increased with increasing growth light intensity, but the cyt f/Rubisco ratio was unaltered. Mesophyll conductance to CO2 diffusion (gm) measured with carbon isotope discrimination increased with growth light intensity but not with measuring light intensity. The responses of CO2 assimilation rate to chloroplast CO2 concentration (Cc) at different light intensities and temperatures were used to estimate the maximum carboxylation rate of Rubisco (Vcmax) and the chloroplast electron transport rate (J). Maximum electron transport rates were linearly related to cyt f content at any given temperature (e.g. 115 and 179 µmol electrons mol?1 cyt f s?1 at 25 and 40 °C, respectively). The chloroplast CO2 concentration (Ctrans) at which the transition from RuBP carboxylation to RuBP regeneration limitation occurred increased with leaf temperature and was independent of growth light intensity, consistent with the constant ratio of cyt f/Rubisco. In tobacco, CO2 assimilation rate at 380 µmol mol?1 CO2 concentration and high light was limited by RuBP carboxylation above 32 °C and by RuBP regeneration below 32 °C.  相似文献

The cosmopolitan, bloom‐forming diatom, Skeletonema costatum, is a prominent primary producer in coastal oceans, fixing CO2 with ribulose 1,5‐bisphosphate carboxylase/oxygenase (RubisCO) that is phylogenetically distinct from terrestrial plant RubisCO. RubisCOs are subdivided into groups based on sequence similarity of their large subunits (IA–ID, II, and III). ID is present in several major oceanic primary producers, including diatoms such as S. costatum, coccolithophores, and some dinoflagellates, and differs substantially in amino acid sequence from the well‐studied IB enzymes present in most cyanobacteria and in green algae and plants. Despite this sequence divergence, and differences in isotopic discrimination apparent in other RubisCO enzymes, stable carbon isotope compositions of diatoms and other marine phytoplankton are generally interpreted assuming enzymatic isotopic discrimination similar to spinach RubisCO (IB). To interpret phytoplankton δ13C values, S. costatum RubisCO was characterized via sequence analysis, and measurement of its KCO2 and Vmax, and degree of isotopic discrimination. The sequence of this enzyme placed it among other diatom ID RubisCOs. Michaelis‐Menten parameters were similar to other ID enzymes (KCO2 = 48.9 ± 2.8 μm ; Vmax = 165.1 ± 6.3 nmol min?1 mg?1). However, isotopic discrimination (ε = [12k/13k ? 1] × 1000) was low (18.5‰; 17.0–19.9, 95% CI) when compared to IA and IB RubisCOs (22–29‰), though not as low as ID from coccolithophore, Emiliania huxleyi (11.1‰). Variability in εvalues among RubisCOs from primary producers is likely reflected in δ13C values of oceanic biomass. Currently, δ13C variability is ascribed to physical or chemical factors (e.g. illumination, nutrient availability) and physiological responses to these factors (e.g. carbon‐concentrating mechanisms). Estimating the importance of these factors from δ13C measurements requires an accurate εvalue, and a mass‐balance model using the εvalue for S. costatum RubisCO is presented. Clearly, appropriate εvalues must be included in interpreting δ13C values of environmental samples.  相似文献

Na‐ion technology is increasingly studied as a low‐cost solution for grid storage applications. Many positive electrode materials have been reported, mainly among layered oxides and polyanionic compounds. The vanadium oxy/flurophosphate solid solution Na3V2(PO4)2F3‐y O2y (0 ≤ y ≤ 1), in particular, has proven the ability to deliver ≈500 Wh kg‐1, operating on the V3+/V4+ (y = 0) or V4+/V5+ redox couples (y = 1). This paper reports here on a significant increase in specific energy by enabling sodium insertion into Na3V2(PO4)2FO2 to reach Na4V2(PO4)2FO2 upon discharge. This occurs at ≈1.6 V and increases the theoretical specific energy to 600 Wh kg?1, rivaling that of several Li‐ion battery cathodes. This improvement is achieved by the judicious modification of the composition either as O for F substitution, or Al for V substitution, both of which disrupt Na‐ion ordering and thereby enable insertion of the 4th Na. This paper furthermore shows from operando X‐Ray Diffraction (XRD) that this energy is obtained in the cycling range Na4V2(PO4)2FO2–NaV2(PO4)2FO2 with a very small overall volume change of 1.7%, which is one of the smallest volume changes for Na‐ion cathodes and which is a crucial requisite for stable long‐term cycling.  相似文献
