Understanding spatio‐temporal variability in recruitment is vital to studies of kelp population dynamics. Research on settlement and post‐settlement processes has suggested that arrival of kelp zoospores to suitable substrate is important in limiting kelp recruitment, yet the role of planktonic processes in kelp population dynamics has not been studied due to difficulties in sampling and identifying zoospores. I developed a method to estimate kelp zoospore abundance from in situ plankton samples and used it to study various processes regulating the availability of giant kelp (Macrocystis pyrifera) zoospores for settlement. My studies focused on (1) identifying temporal scales over which zoospore abundance is most variable, (2) describing physical and biological processes that regulate this variability, and (3) determining the relationship between zoospore abundance and settlement. I found that short‐term variability in zoospore abundance (<24 hrs) was not due to changes in supply but rather dispersion, caused by oscillating hydrodynamic forces (e.g. waves). Long‐term variability in zoospore abundance, however, was best explained by the size and density of reproductive adult plants, with zoospore abundance being most variable at the scale of days to months. Changes in adult reproductive condition caused rapid changes in zoospore abundance suggesting that the supply of kelp zoospores is sensitive to environmental regulation of adult physiology. Thus, unlike with marine animals, these results indicate that variability in kelp propagule supply, over scales most likely to affect subsequent settlement and recruitment, is more tightly coupled to demographic and reproductive mechanisms than to physical transport processes.  相似文献

Elevated irradiance has a profound effect on the successful dispersal and establishment of kelp zoospores, affecting their physiology and viability. The research to date, however, has been on zoospores localized near the benthos, with little attention on the importance of vertical transportation and subsequent exposure to increased irradiance. Therefore, we wanted to investigate the effects of exposure to high irradiance on the reproductive planktonic life‐history stages of kelps Macrocystis pyrifera (L.) C. Agardh and Pterygophora californica Rupr. Zoospores of both species were exposed to different irradiances (75, 275, 575, 1,025 μmol photons · m?2 · s?1) over varying durations (1, 2, 4, 8, 12 h) and subsequently monitored for settlement competency, gametophyte development, and reproductive viability. Settlement success for M. pyrifera was uniform throughout all irradiance × time treatments, while settlement for P. californica decreased with increasing exposure time but not irradiance, although settlement was generally reduced at the highest irradiance level. Following zoospore settlement, germ tube development was visible in the gametophytes of both species within 1 week, although a significant decline of germ tube density in P. californica was observed with increasing irradiance. Similarly, a decrease in germ tube development with increasing exposure was observed across all irradiance levels for M. pyrifera, but irradiance itself was not significant. Further development into embryonic sporophytes was remarkably similar to gametophyte development, suggesting that the effect of exposure of kelp zoospores to high irradiance on subsequent sporophyte production is mediated through gametophyte development as well as zoospore survival.  相似文献

Successful kelp recruitment is important for kelp population persistence and associated kelp forest communities. The proximity of settled kelp zoospores is a known requirement for successful kelp recruitment and proximity can be increased as zoospores aggregate. Substrate rugosity can also be an important factor affecting macroalgal settlement and recruitment in wave‐swept areas, and may affect kelp recruitment by aggregating zoospores. In this study, kelp zoospores were cultured at different levels of small‐scale aggregation and kelp recruitment was quantified. Sporophyte production significantly increased as zoospores became more aggregated indicating that processes that aggregate kelp zoospores have the potential to enhance kelp recruitment. A 13‐month field experiment demonstrated differential kelp recruitment onto settlement plates that mimicked surface rugosities of two common rock types within Stillwater Cove, Carmel Bay in central California (Carmelo Formation sandstone and Santa Lucia granodiorite). Significantly more kelp recruited to molds mimicking granodiorite over the yearlong study (granodiorite = 2.7 recruits ± SE 0.50, sandstone = 1.2 recruits ± SE 0.51). There was a significant difference in recruitment between seasons and this variability was due to the fact that spring had the highest average number of kelp recruits per mold. However, the interaction between substrate and season was not significant. This study emphasizes the importance of kelp zoospore aggregation on kelp recruitment and demonstrates that small‐scale rugosity affects kelp recruitment.  相似文献

Ecklonia radiata is the main foundation species in Australian temperate reefs, yet little has been published on its reproduction and how this may change across its depth range (1–50+ m). In this study, we examined differences in sporophyte morphology and zoospore production during a reproductive season and across four depths (7, 15, 25, and 40 m). Additionally, we examined differences in germination rate, survival, and morphological traits of gametophytes obtained from these four depths, cultured under the same light and temperature conditions. Multivariate morphology of sporophytes differed significantly between deep (~40 m) and shallow sites (7 and 15 m), but individual morphological traits were not significantly different across depths. Total spore production was similar across depths but the peak of zoospore release was observed in February at 15 m of depth (6,154 zoospores · mm?2 of tissue) and the minimum observed in January at 7, 25, and 40 m (1,141, 987, and 214 zoospores · mm?2 of tissue, respectively). The source depth of zoospores did not have an influence in the germination rate or the survival of gametophytes, and only gametophytes sourced from 40 m sites presented significantly less surface area and number of branches. Overall, these results indicate that E. radiata’s reproductive performance does not change across its depth range and that kelp beds reproducing in deeper areas may contribute to the replenishment of their shallow counterparts. We propose that deep kelps may constitute a mechanism of resilience against climate change and anthropogenic disturbances.  相似文献

Recent research suggests that fungal zoospores have important roles in ecosystems. Ultrastructural characteristics of zoospores and sequences of ribosomal RNA genes are increasingly being used in taxonomic and phylogenetic studies with zoosporic fungi. However, our current knowledge of the physiology and ecology of zoospores lags far behind our knowledge of phylogenetic relationships. Some aspects of the ecology of fungal zoospores are discussed in this review. Fungal zoospores have two mechanisms for active short range dispersal: flagellar and amoeboid movement. Flagellar movement is more common in open water and amoeboid movement more common on surfaces. All phases of the life cycle can be passively transported by currents in water for long range dispersal. As far as is known the zoospore never has a cell wall but can have a cell coat. This may cause zoospores to be more susceptible to damage by osmotic and mechanical forces than other phases in the life cycle. The environmental cues for release of zoospores are not understood. Zoospores can often respond to both light and chemical gradients. The behavior of zoospores may influence the structure of the colony formation. Little is known about the modes of nutrition of zoospores. Some zoospores may be osmotrophic but there is no evidence for holozoic nutrition.  相似文献

Some species of macroalgae continue to live for extended periods of time after detachment and may even maintain reproductive structures, yet very little is known about this process. Here, we describe the presence of sporophylls (with sporogenous tissues) on floating kelp rafts of Macrocystis spp. along the coast of Chile. Surveys were conducted at nine sites (18–50° S) during austral summer 2002, and floating kelp rafts were seen and collected at seven of these nine sites (between 22 and 50° S). Fifteen (26.8%) of the 56 samples had sporophylls, indicating maintenance of sporophylls after detachment. Some of the kelp sporophytes with reproductive blades showed signs of having been afloat for long periods (indicated by the large size of attached stalked barnacles). Additionally, experiments showed that floating kelps released viable zoospores. To understand the reproductive dynamics of floating kelps, we compared these results with information from attached populations of Macrocystis spp. at nearby coastal sites. In general, attached kelp had higher proportions of sporophylls than floating rafts, suggesting that detachment may negatively affect reproductive status. Nevertheless, floating kelps remained functionally reproductive, suggesting that zoospores may be dispersed via floating rafts. Published reports on other macroalgae indicate that some species (Lessoniaceae, Fucaceae, and Sargassaceae) are fertile and probably release zoospores or zygotes while floating or drifting in ocean currents. Because dispersal distances achieved by spores of most macroalgae are relatively short, release of spores from floating algae may be an alternative mechanism of long‐distance dispersal.  相似文献

One of the most significant processes in the life history of an alga is the colonization of a new substratum. In the present study, we evaluate whether different organic compounds, such as agar, gelatine, chicken albumin, glycerine and polylysine, promote zoospore recruitment and germination in a periphytic, fresh-water green microalga of the genus Pseudulvella(Chlorophyta). Given the low adhesion capacity of its zoospores a series of experiments were conducted in order to find a substance and its optimal concentration that increases zoospore recruitment and allows us to follow the processes of settlement, attachment and germination of zoospores. Polylysine significantly increased the number of zoospores attached with no significant effect on the germination rate. The minimum effective concentration of polylysine for improving zoospore settlement was 0.1%. %  相似文献

Macroalgal rafts frequently occur floating in coastal waters of temperate regions of the world’s oceans. These rafts are considered important dispersal vehicles for associated organisms with direct development. However, environmental factors may limit the floating potential of kelp and thereby the dispersal of associated organisms. To examine the effect of water temperature and grazing on growth, reproductive output, and survival of floating Macrocystis spp., experiments were conducted in outdoor tanks during austral summer 2006/2007 at three sites along the Chilean Pacific coast (20° S, 30° S, 40° S). At each site, Macrocystis spp. was maintained individually at three different water temperatures (ambient, ambient − 4°C, ambient + 4°C) and in the presence or absence of the amphipod grazer Peramphithoe femorata for 14 d. High water temperatures (>20°C) provoked rapid degradation of Macrocystis spp. rafts. At moderate temperatures (15°C–20°C), algal survival depended on the presence of associated grazers. In the absence of grazers, algal rafts gained in biomass while grazing caused considerable losses of algal biomass. Algal survival was the highest under cooler conditions (<15°C), where raft degradation was slow and grazer-induced biomass losses were compensated by continuing algal growth. Our results indicate that floating kelp rafts can survive for long time periods at the sea surface, but survival depends on the interaction between temperature and grazing. We suggest that these processes limiting the survival of kelp rafts in warmer temperatures may act as a dispersal barrier for kelp and its associated passengers.  相似文献

Significantly higher numbers of zoospores of the fouling, green alga Enteromorpha adhered to silicone elastomers cured by dibutyltin dilaurate (DBTDL) than to identical polymers cured by dibutyltin diacetate (DBTDA). Enhanced zoospore adhesion was shown to be due to the presence of DBTDL and the effect was concentration‐dependent. Further experiments revealed that enhanced zoospore adhesion also occurred to glass coverslips coated with lauric acid (C12) and other saturated fatty acids. The possibility that fatty acids may act as chemical cues (chemoattractants), guiding motile zoospores to the substratum for settlement in the natural environment is discussed. Settlement of other fouling organisms to DBTDL‐cured silicone elastomers is currently being investigated.  相似文献

N -acylhomoserine lactone (AHL) quorum-sensing molecules modulate the swimming behaviour of zoospores of the macroalga Ulva to facilitate the location of bacterial biofilms. Here we show that the intertidal surfaces colonized by Ulva are dominated by Alphaproteobacteria , particularly the Rhodobacteraceae family, and the Bacteroidetes family Flavobacteriaceae , and that this diverse assemblage both produces and degrades AHLs. N -acylhomoserine lactones could also be extracted from the surfaces of pebbles recovered from intertidal rock-pools. Bacteria representative of this assemblage were isolated and tested for the production and degradation of AHLs, and for their ability to modulate zoospore settlement at different biofilm densities. Of particular interest was a Shewanella sp. This strain produced three major AHLs (OC4, OC10 and OC12) in the late exponential phase, but the longer-chain AHLs were rapidly degraded in the stationary phase. Degradation occurred via both lactonase and amidase activity. A close relationship was found between AHL synthesis and Ulva zoospore settlement. The Shewanella isolate also interfered with AHL production by a Sulfitobacter isolate and its ability to enhance zoospore settlement in a polymicrobial biofilm. This influence on the attachment of Ulva zoospores suggests that AHL-degrading strains can affect bacterial community behaviour by interfering with quorum sensing between neighbouring bacteria. More importantly, these interactions may exert wider ecological effects across different kingdoms.  相似文献

We investigated the toxicity of Karenia mikimotoi toward three model grazers, the cladoceran Moina mongolica, the copepod Pseudodiaptomus annandalei, and the crustacean Artemia salina, and explored its chemical response upon zooplankton grazing. An induction experiment, where K. mikimotoi was exposed to grazers or waterborne cues from the mixed cultures revealed that K. mikimotoi might be toxic or nutritionally inadequate toward the three grazers. In general, direct exposure to the three grazers induced the production of hemolytic toxins and the synthesis of eicosapentaenoic acid (EPA). Both EPA and the hemolytic toxins from K. mikimotoi decreased the survival rate of the three grazers. In addition, the survival rates of M. mongolica, P. annandalei, and A. salina in the presence of induced K. mikimotoi that had previously been exposed to a certain grazer were lower than their counterparts caused by fresh K. mikimotoi, suggesting that exposure to some grazers might increase the toxicity of K. mikimotoi. The chemical response and associated increased resistance to further grazing suggested that K. mikimotoi could produce deterrents to protect against grazing by zooplankton and that the substances responsible might be hemolytic toxins and EPA.  相似文献

The kelp Laminaria hyperborea is host for a rich fauna of mobile invertebrates. Dispersal patterns of these animals may be crucial for their availability to visual predators like fish, which are known to search for food in these productive habitats. Diurnal, horizontal and vertical dispersal within and out of the kelp forest were studied by analysing colonization of artificial substrata placed among kelps. The species composition of the fauna was different on three parts of the kelp: lamina, stipes (with epiphytes) and holdfast. The artificial substrata were colonized mainly by species associated with kelp. More species and individuals colonized the artificial substrata at night than during the day. The most abundant faunal groups on the kelps and the artificial substrata were amphipods and gastropods. Both groups dispersed at a significantly higher rate at night than during the day. Rapid horizontal dispersal out of the kelp forest was found. The artificial substrata were also colonized quickly by kelp fauna at all vertical levels inside and above the kelp forest. However, species associated with the kelp holdfast tended to disperse close to the bottom, while stipe-associated fauna moved through all parts of the kelp forest and even above the canopy layer. A high dispersal rate appears common amongst the mobile species living on kelp and seems to be advantageous, despite the risks involved in emigrating from the habitat and being exposed to predators. Higher activity at night may reduce availability to predators.  相似文献

Zoospores are major dispersal and infective propagules of pythiaceous species. Built upon a recently developed ‘wet‐plate’ method, the objectives of this study were to develop a better understanding about zoospore production biology. Four broth media and five incubation temperatures were evaluated with 12 isolates of Phytophthora nicotianae and 17 other pythiaceous species in this study. The ‘wet‐plate’ method worked the best for heterothallic species, especially those isolates that do not produce chlamydospores. These species included Phytophthora citrophthora, P. nicotianae, Phytophthora palmivora and Phytophthora tropicalis. They readily produced 105–106 zoospores/ml. Overall, most species and isolates produced more zoospores with 20% clarified V8 broth than the other three media: rye, lima bean and carrot. The optimal temperature for nutrient‐deprived culture without free‐flowing water to produce sporangia typically is 5°C cooler than that for vegetative growth. Fresh and revived cultures are more prolific than those that had been subcultured multiple times. These findings will assist oomycete researchers, adding quality, productivity and efficiency to their future zoospore‐based studies.  相似文献

Climate change is expected to favour infectious diseases across ecosystems worldwide. In freshwater and marine environments, parasites play a crucial role in controlling plankton population dynamics. Infection of phytoplankton populations will cause a transfer of carbon and nutrients into parasites, which may change the type of food available for higher trophic levels. Some phytoplankton species are inedible to zooplankton, and the termination of their population by parasites may liberate otherwise unavailable carbon and nutrients. Phytoplankton spring blooms often consist of large diatoms inedible for zooplankton, but the zoospores of their fungal parasites may serve as a food source for this higher trophic level. Here, we investigated the impact of warming on the fungal infection of a natural phytoplankton spring bloom and followed the response of a zooplankton community. Experiments were performed in ca. 1000 L indoor mesocosms exposed to a controlled seasonal temperature cycle and a warm (+4 °C) treatment in the period from March to June 2014. The spring bloom was dominated by the diatom Synedra. At the peak of infection over 40% of the Synedra population was infected by a fungal parasite (i.e. a chytrid) in both treatments. Warming did not affect the onset of the Synedra bloom, but accelerated its termination. Peak population density of Synedra tended to be lower in the warm treatments. Furthermore, Synedra carbon: phosphorus stoichiometry increased during the bloom, particularly in the control treatments. This indicates enhanced phosphorus limitation in the control treatments, which may have constrained chytrid development. Timing of the rotifer Keratella advanced in the warm treatments and closely followed chytrid infections. The chytrids' zoospores may thus have served as an alternative food source to Keratella. Our study thus emphasizes the importance of incorporating not only nutrient limitation and grazing, but also parasitism in understanding the response of plankton communities towards global warming.  相似文献

Neutral lipids, consisting primarily of triacylglycerols, were found to be a major form of carbon reserve in zoospores of the giant kelp Macrocystis pyrifera (L.) C. Ag. The fluorescent stain Nile Red revealed large lipid droplets in the posterior end of the cell, which comprised 20–41% of cellular carbon in newly released spores. Flow cytometric analyses of newly released spores stained with Nile Red revealed considerable variation in the neutral lipid content among spores that was independent of spore size. Lipid droplets were consumed during germination in spores maintained either under constant light or in continual darkness. The availability of light appeared to delay, but did not preclude, lipid use. The rate of lipid use during germination varied considerably among germlings with some cells consuming all of their lipid reserves within 5 h after release. In addition to zoospores, lipid droplets were observed in both male and female gametes. Numerous droplets were observed in eggs, while single lipid droplets were observed in sperm. Neutral lipid droplets were not observed in gametophytes or sporophytes except in developing gametes and spores. Large lipid reserves thus seem to be confined to the microscopic life history stages that presumably have relatively high energy demands. By serving as a supplemental fuel reserve, neutral lipids may be important in extending the effective range of zoospore dispersal.  相似文献

The processes limiting the population recovery of the kelp Saccharina latissima after recent large‐scale loss from the south coast of Norway are poorly understood. Previous investigations do, however, suggest that the impacts of biotic interactions (epibiosis and competition) and increased water turbidity are important. We investigated the depth‐related patterns of growth, epibiosis, and mortality in two sample populations of kelp, from the south and the southwest coast of Norway. The investigations were performed over a period of seven months, in a crossed translocational study, where kelps were mounted on rigs at six depths (1, 3, 6, 9, 15, and 24 m). In a second experiment, the amounts of light blocked by different epibiont layers growing on the kelp frond were investigated. While growth decreased with depth in spring and summer, the kelp grew faster at 15 m than at shallower depths in fall. Survival was low both in shallow water and below 15 m depth. Epibionts covered the kelp growing at depths from 1 to 9 m, and the laboratory study showed that the coverage may have deprived the individuals of as much as 90% of the available light. Although the depth‐related results we present apply—in the strictest sense—only to kelp translocated on rigs, we argue that the relative patterns are relevant for natural populations. Growth and survival of S. latissima is likely to be reduced by heavy loads of epibionts, while depths where epibionts are sparse may be close to the lower limit of the kelps depth distribution along the south coast of Norway. This suggests that a vertical squeeze, or narrowing of the distribution range of kelp forests may be occurring in Norway.  相似文献

The green alga Enteromorpha is the most important macroalga that fouls ships, submarines and underwater structures. Major factors in its success in colonising new substrata are the production of enormous numbers of swimming spores and their ability to locate surfaces on which to settle. Factors facilitating the settlement and adhesion of asexual zoospores are examined in this article. Settlement and adhesion may be regulated by topographical, biological, chemical and physico-chemical cues, all of which are modified by the presence of microbial biofilm. The level of gregarious zoospore settlement is related to spore density and may be mediated by a number of external cues including fatty acids and 'detritus'.  相似文献

Bacteria utilize quorum sensing to regulate the expression of cell density-dependant phenotypes such as biofilm formation and virulence. Zoospores of the marine alga Ulva intestinalis exploit the acyl-homoserine lactone (AHL) quorum sensing system to identify bacterial biofilms for preferential settlement. Here, we demonstrate that AHLs act as strong chemoattractants for Ulva zoospores. Chemoattraction does not involve a chemotactic orientation towards the AHL source. Instead, it occurs through a chemokinesis in which zoospore swimming speed is rapidly decreased in the presence of AHLs. The chemoresponse to AHLs was dependant on the nature of the acyl side chain, with N-(3-oxododecanoyl)-homoserine lactone (30-C12-HSL) being the most effective signal molecule. Mean zoospore swimming speed decreased more rapidly over wild-type biofilms of the marine bacteria Vibrio anguillarum relative to biofilms of the vanM mutant, in which AHL synthesis is disrupted. These data implicate a role for AHL-mediated chemokinesis in the location and preferential settlement of Ulva zoospores on marine bacterial assemblages. Exposure to AHLs did not inhibit the negative phototaxis of Ulva zoospores, indicating that chemoattraction to bacterial biofilms does not preclude the response to a light stimulus in substrate location.  相似文献

Atlantic menhaden Brevoortia tyrannus develop characteristic skin ulcers in response to infection by the oomycete Aphanomyces invadans. To investigate pathogenicity, we conducted a dose response study. Juvenile menhaden were inoculated subcutaneously with 0, 1, 5, 10, 100, and 500 secondary zoospores per fish and monitored for 37 d post-injection (p.i.). Survival rates declined with increasing zoospore dose, with significantly different survivorship curves for the different doses. Moribund and dead fish exhibited characteristic ulcerous lesions at the injection site starting at 13 d p.i. None of the sham-injected control fish (0 zoospore treatment) died. The LD50 (lethal dose killing 50% of exposed menhaden) for inoculated fish was estimated at 9.7 zoospores; however, some fish receiving an estimated single zoospore developed infections that resulted in death. Menhaden were also challenged by aqueous exposure and confirmed that A. invadans was highly pathogenic by this more environmentally realistic route. Fish that were acclimated to culture conditions for 30 d, and presumably free of skin damage, then aqueously exposed to 100 zoospores ml(-1), exhibited 14% lesion prevalence with 11% mortality. Net-handled fish that were similarly infected had a significantly higher lesion prevalence (64%) and mortality (64%). Control fish developed no lesions and did not die. Scanning electron microscopy of fish skin indicated that zoospores adhered to intact epidermis, germinated and penetrated the epithelium with a germ tube. Our results indicate that A. invadans is a primary pathogen of menhaden and is able to cause disease at very low zoospore concentrations.  相似文献
