Aims:  To compare bacterial populations and antimicrobial resistance patterns between clinical and sewage isolates from a regional hospital in northern Taiwan. The dissemination of antibiotic-resistant bacteria from hospital compartments to the hospital sewage treatment plant was examined.
Methods and Results:  A total of 1020 clinical isolates and 435 sewage isolates were collected between July and September 2005. The percentages of Gram-negative bacteria from the clinical and sewage isolates were 87·2% and 91·0%, respectively ( P  =   0·033). Escherichia coli were the leading bacterial isolates in both groups. Antimicrobial susceptibility testing showed a significant difference ( P  <   0·001) in resistance to ampicillin (85·6% vs 94·1%), ampicillin/sulbactam (31·7% vs 55·4%), cefazolin (29·2% vs 71·5%) and cefuroxime (20·7% vs 61·9%) between clinical and sewage coliform isolates, respectively.
Conclusions:  The sewage isolates had higher antimicrobial resistance rates than the clinical isolates from the same hospital.
Significance and Impact of the Study:  The low efficacy of the hospital sewage treatment may contribute to the dissemination of multidrug resistant bacteria from this hospital compartments to the environment. Practices which limit the disposal of antimicrobial agents into the wastewater system may be the possible measure to prevent the selection of multidrug-resistant bacteria from sewage treatment plants.  相似文献

Aims:  To design a cyclic voltammetry (CV) procedure to check the electrochemical activity of bacterial isolates that may explain the electrochemical properties of biofilms formed in compost.
Methods and Results:  Bacteria catalysing acetate oxidation in garden compost were able to form electrochemically active biofilms by transferring electrons to an electrode under chronoamperometry. They were recovered from the electrode surface and identification of the isolates using 16S rRNA sequencing showed that most of them were Gammaproteobacteria, mainly related to Enterobacter and Pseudomonas spp. A CV procedure was designed to check the electrochemical activity of both groups of isolates. Preliminary CVs suggested that the bacteria were not responsible for the catalysis of acetate oxidation. In contrast, both groups of isolates were found to catalyse the electrochemical reduction of oxygen under experimental conditions that favoured adsorption of the microbial cells on the electrode surface.
Conclusions:  Members of the genera Enterobacter and Pseudomonas were found to be able to catalyse the electrochemical reduction of oxygen.
Significance and Impact of the Study:  This study has shown the unexpected efficiency of Enterobacter and Pseudomonas spp. in catalysing the reduction of oxygen, suggesting a possible involvement of these species in biocorrosion, or possible application of these strains in designing bio-cathode for microbial fuel cells.  相似文献

对大亚湾海域表层水体的可培养细菌进行分离,测定了分离培养细菌的16S rRNA 基因V3 区序列,并采用该研究中的35 个序列与从GenBank 下载的46 个序列构建了系统树,对细菌进行了分子鉴定。在9 个站点的表层水体中共分离得到70株菌株,分属35 个种。其中α-变形菌纲(Alphaproteobacteria)和放线菌纲(Actinobacteria)为分离菌株最多的两个菌纲,分离率分别为40%和35.7%;而β-变形菌纲(Betaproteobacteria)仅分离到1 株(1.4%)。此外,还分离得到盖球菌属(Kytococcus)、间胞囊菌属(Intrasporangium)等非常见菌株。大亚湾海域细菌的Shannon-Wiener's 多样性指数为3.21。大亚湾海域表层水体可培养细菌丰富的种类多样性以及较高的α-变形菌分离率,说明了该海域具有良好的水质,同时也是潜在的微生物菌种资源库。  相似文献

Aims:  The aim of the present study was to reveal the microbial genetic diversity of epilithic biofilms using a DNA-based procedure.
Methods and Results:  A DNA extraction protocol was first selected to obtain PCR-amplifiable metagenomic DNA from a limestone biofilm. Extracted DNA was used to amplify either 16S rRNA genes or ITS regions from prokaryotic and eukaryotic genomes, respectively. Amplified DNAs were subsequently cloned, amplified by colony PCR and screened by restriction analysis [restriction analyses of amplified ribosomal DNA (ARDRA)] for DNA sequencing. Phylogenetic analysis using 16S rDNA sequences showed that predominating bacteria were Alphaproteobacteria belonging to the genera Sphingomonas , Erythrobacter , Porphyrobacter , Rhodopila and Jannashia ; Cyanobacteria and Actinobacteria were also identified. Analysis of ITS rDNA sequences revealed the presence of algae of the Chlorophyceae family and fungi related either to Rhinocladiella or to a melanized ascomycete. Statistical analysis showed that the specific richness evidenced was representative of the original sampled biofilm.
Conclusions:  The molecular methodology developed here constitutes a valuable tool to investigate the genetic diversity of microbial biofilms from building stone.
Significance and Impact of the Study:  The easy-to-run molecular method described here has practical importance to establish microbiological diagnosis and to define strategies for protection and restoration of stone surfaces.  相似文献

Aim:  Fresh kava beverages have a limited shelf life under refrigerated conditions. The objective of this study was to isolate and identify bacteria in aqueous extracts of kava rhizome.
Methods and Results:  The internal part of kava rhizome was used to minimize soil contamination. Three kava extracts were prepared, serially diluted and plated on nutrient agar. Isolated colonies were identified by sequencing polymerase chain reaction amplicons targeting the eubacterial 16S rDNA and the tuf gene of Staphylococcus . Seventy-five bacterial isolates belonged to 16 genera. Bacillus , Cellulomonas , Enterococcus , Pectobacterium and Staphylococcus were identified in all kava extracts.
Conclusions:  Kava rhizome contains large amounts of starch and fibre, which justify the presence of polysaccharide-degrading bacteria in the extracts. Bacillus cereus group and Staphylococcus species may produce toxins and cause foodborne illness.
Significance and Impact of the Study:  The results of this study provide fundamental information that may be used to enhance the microbial quality and safety of kava beverages.  相似文献

Aims:  To understand the diversity, taxonomy and antagonistic potential of rice-associated bacteria, and to discover new bacteria for biocontrol of rice foliar pathogens.
Methods and Results:  Amplified ribosomal DNA restriction analysis (ARDRA), BOX-PCR and 16S rRNA gene sequence analysis were used to identify the diversity of 203 rice-associated antagonistic bacteria. Eleven potential biocontrol bacteria were used to test their biological control of rice blast in a natural field experiment. Eleven different genera were encountered in five divisions, including Bacilli , Alphaproteobacteria , Betaproteobacteria , Gammaproteobacteria and Deinococci . The most prominent genus in all microenvironments was Bacillus (68·5%). The efficacy of rice leaf blast biocontrol was 64·35% for strain 1Pe2, 57·86% for strain 2R37 and 56·44% for strain 1Re14.
Conclusions:  Biocontrol data from the field experiments demonstrated no positive correlation between antagonism, physiological characteristics and biocontrol efficacy. There was significant diversity among the rice-associated bacteria isolated from different microenvironments. The most prominent genus of all microenvironments was Bacillus . Brevibacillus brevis strain 1Pe2 and Deinococcus aquaticus strain 1Re14 have good potential for field application and commercial use.
Significance and Impact of the Study:  This is the first attempt to study the diversity and identification of rice-associated antagonistic bacteria from different microenvironments, and endophytic bacteria Deinococcus aquaticus strain 1Re14, Acidovorax sp. isolate 3Re21 and Brevibacillus brevis strain 1Pe2 are first reported as rice-associated bacteria.  相似文献

Aims:  To evaluate the performance of the VITEK2 Bacillus identification card (BCL) for the identification of aerobic endospore-forming bacteria, using fresh isolates and reference strains.
Methods and Results:  One hundred and nine industrial, environmental and clinical isolates were tested using the BCL card. The card contained 46 substrates for measuring carbon source utilization, enzymatic activities, inhibition by 6·5% NaCl and resistance to the antibiotics kanamycin, oleandomycin and polymyxin B. Identifications were made after 14 h incubation, using a database allowing identification of 42 species of the genera Aneurinibacillus , Bacillus , Brevibacillus , Geobacillus , Paenibacillus and Virgibacillus . The reference identities of all isolates were authenticated by phenotypic methods, with 16S rRNA gene sequencing used to resolve discrepancies.
Conclusions:  One hundred and one strains (93%) were identified correctly to species level, seven strains (6%) were incorrectly identified, and one strain (1%) remained unidentified.
Significance and Impact of the Study:  The VITEK2 BCL card provides a major advance in the reliable identification of Bacillus species and members of related genera.  相似文献

A 16-month pilot study in two similar 1200 m water mains was conducted to determine the effects of granular activated carbon (GAC) filtration on drinking water quality in a distribution system. The results demonstrated that despite the higher initial disinfectant residue, the increase in bacteria in the conventionally treated and postozonated water was higher than in the water additionally treated with GAC filtration and u.v.-disinfection. Accordingly, a significant decline in assimilable organic carbon in the postozonated water was observed throughout the main, whereas in the GAC-filtered water this decline was shifted to the GAC filters. In the GAC-filtered water the conversion of ammonia to nitrite and nitrite to nitrate was more intense than in postozonated water. The findings confirm that GAC filtration increases the biological stability of drinking water even when treating cold humic waters in which biodegradation is generally limited by phosphorus. However, it appears that biological treatment favours the slow kinetics of nitrifying bacteria, thus allowing nitrification to occur even under cold water conditions in a distribution system.  相似文献

Abstract.  1. Colonisation of host trees by an endophytic herbivore, the spruce beetle, Dendroctonus rufipennis , is accompanied by invasion of its galleries by a number of fungal species. Four of these associated species were identified as Leptographium abietinum , Aspergillus fumigatus , Aspergillus nomius , and Trichoderma harzianum .
2.  Trichoderma and Aspergillus significantly reduced spruce beetle survival and reproduction in controlled assays.
3. A previously undescribed behaviour was observed, in which spruce beetle adults exuded oral secretions, especially within fungus-pervaded galleries.
4. These oral secretions inhibited the growth of fungi except A. nomius , and disrupted the morphology of the latter. Administration of these secretions indicated a dose-dependent inhibitory effect.
5. Oral secretions cultured on microbiological media yielded substantial bacterial growth.
6. Filter-sterilised secretions failed to inhibit fungal growth, evidence that the bacteria are responsible for the antifungal activity.
7. Nine bacterial isolates belonging to the Actinobacteria, Firmicutes, Gammaproteobacteria, and Betaproteobacteria taxa were obtained from the secretions.
8. Bacterial isolates showed species-specific inhibitory activity against the four fungi antagonistic to spruce beetle. The bacterium with the strongest fungal inhibition activity was the actinomycete Micrococcus luteus .
9. The production of bark beetle secretions containing bacteria that inhibit fungal growth is a novel finding. This suggests an additional level of complexity to ecological associations among bark beetles, conifers, and microorganisms, and an important adaptation for colonising subcortical tissue.  相似文献

The dominant genera of microorganisms isolated from interstitial waters and GAC particles were Achromobacter, Acinetobacter, Aeromonas, Alcaligenes, Bacillus, Chromobacterium, Corynebacterium, Micrococcus, Microcyclus, Paracoccus, and Pseudomonas. Coliform bacteria were found in small numbers in the effluents from some of the GAC columns in the later months of the study. Oxidation of influent waters with ozone and maintenance of aerobic conditions on the GAC columns failed to appreciably enhance the microbial growth on GAC.  相似文献

Aims:  To identify lactic acid bacteria (LAB) present in Moroccan dairy products to establish and preserve their microbial species diversity.
Methods and Results:  Thirty-seven samples were collected from different farms. A total of 146 LAB were isolated and subjected to (GTG)5-PCR analysis. Comparison of the profiles with data available at the Moroccan Coordinated Collections of Micro-organisms allowed identification of 85 isolates. The remaining 61 were subjected to SDS-PAGE analysis of whole cell proteins. Comparison of the profiles with data available at the Belgian Coordinated Collections of Micro-organisms allowed identification of 43 isolates. Several of the remaining 18 isolates exhibited identical protein electrophoretic fingerprints. Therefore, eight representatives of them were subjected to partial pheS gene sequencing which allowed identification of all remaining isolates. In raw milk, six genera were found while in 'lben', three were found. This is the first report of Leuconostoc kimchii in dairy products.
Conclusions:  LAB diversity was established using a stepwise polyphasic identification approach. It used the expertise of both research bodies involved in this study and proved to be cost-effective for the identification of all isolates.
Significance and Impact of the Study:  To establish LAB diversity in Moroccan dairy products which could be a source of strains with specific properties.  相似文献

Aims:  The microbial diversity of bioaerosols released during operational activities at composting plants is poorly understood. Identification of bacteria and fungi present in such aerosols is the prerequisite for the definition of microbial indicators that could be used in dispersal and exposure studies.
Methods and Results:  A culture-independent analysis of composting bioaerosols collected at five different industrial open sites during the turning of composting piles in fermentation was performed by building 16S rDNA and 18S rDNA libraries. More than 800 sequences were analysed. Although differences in the phylotypes distribution were observed from one composting site to another, similarities in the structure of microbial diversity were remarkable. The same phyla dominated in the five bioaerosols: Ascomycota among fungi, Firmicutes and Actinobacteria among bacteria. For each phylum, some dominant phylotypes were common to at least four bioaerosols. These common phylotypes belonged to Thermomyces , Aspergillus , Penicillium , Geobacillus, Planifilum , Thermoactinomyces , Saccharopolyspora , Thermobifida and Saccharomonospora .
Conclusions:  The microbial signature of aerosols produced during the thermophilic phase of composting was determined. The similarities observed may be explained by the selection of thermophilic and sporulating species.
Significance and Impact of the Study:  Several bacteria and fungi identified in this study may represent potential indicators of composting bioaerosols in air.  相似文献

Granular activated carbon (GAC) beds may be used for removal of dissolved organic matter during the treatment of drinking water. However, they might also change the microbiological quality of the water entering the distribution system either by changing the predominant bacteria or the bacterial density of the treated water. A 3-year pilot plant study of water treatment using GAC beds was conducted at the Baxter Water Treatment Plant in Philadelphia. During the study, bacteria were isolated from the raw water and from the effluents of the GAC treatment units. At the end of the study, bacteria were also isolated from the GAC units and from sand beds operated in parallel with the GAC units. Bacterial genera in the GAC effluents and in the GAC units themselves were similar to those found in the raw water and in the sand beds. Prechlorination and (or) preozonation of the water before GAC treatment had no noticeable effect on the bacterial genera found as compared with GAC unit having no predisinfection. The bacterial genera found in this study were similar to those found in seven other studies of GAC water treatment that used a variety of treatment schemes and a variety of heterotrophic plate count techniques to evaluate bacterial populations. From these several studies it appears that GAC treatment does not change the nature of the bacterial populations associated with drinking water.  相似文献

1. Planktonic ciliates were examined during a spring–summer period (November 1998–April 1999) in the ultraoligotrophic Lake Moreno Oeste (41°5' S and 71°33' W, 758 m a.s.l), which belongs to the Nahuel Huapi System (Patagonia, Argentina). The lake is deep ( Z max=90 m) and warm monomictic.
2. Sampling was performed at a mid-lake station, where vertical profiles of temperature and light were measured in situ , and samples for bacteria and ciliates enumeration were taken throughout the water column.
3. The peritrich Ophrydium naumanni , a freshwater pelagic ciliate with endosymbiotic Chlorella , was the dominant ciliate in the lake.
4.  Ophrydium naumanni and autotrophic picoplankton exhibit a clear coincidence in their vertical distribution ( P  < 0.05), preferring levels at or near the 1% of surface photosynthetically active radiation (PAR) irradiance. Both may have the same light requirements, or the coincidence may reflect a trophic relationship.
5. Dependence on light and prey by O. naumanni were studied using field experiments, in which we analysed ciliate grazing on bacteria, and in laboratory experiments, in which we compared particle uptake under dark and light conditions.
6.  Ophrydium naumanni was able to ingest particles [latex microspheres and fluorescently labelled bacteria (FLB)] in field and laboratory experiment, indicating that it has the potential to affect bacteria population of Lake Moreno Oeste.
7. Ciliate particle ingestion was observed to be dependent on light availability because under dark conditions, the ingestion was lowered ( P  < 0.05).  相似文献

Aims:  To identify the types of micro-organisms involved in the formation of biofilms on dairy ultrafiltration and reverse osmosis membranes and investigate factors affecting the attachment of those isolates.
Methods and Results:  Micro-organisms isolated from industrial membranes following standard cleaning were identified using the API culture identification system. Thirteen different isolates representing eight genera were isolated and their ability to attach to surfaces was compared using a microtitre plate assay. Three Klebsiella strains attached best, while mixed strains of Pseudomonas and Klebsiella attached better than individual strains. Whey enhanced the attachment of the isolates. The micro-organisms were characterized according to cell surface hydrophobicity using the microbial adhesion to hydrocarbon (MATH) test, and cell surface charge by measuring the zeta potential. These cell surface characteristics did not show a clear relationship with the attachment of our strains.
Conclusions:  A variety of different micro-organisms is associated with dairy ultrafiltration and reverse osmosis membranes after cleaning, suggesting several possible sources of contamination. The cleaning of these membranes may be inadequate. The attachment of the different isolates is highly variable and enhanced in the presence of whey.
Significance and Impact of the Study:  Knowledge of persistent microflora colonizing dairy membrane systems will help develop strategies to mitigate biofilm development in this environment, improving hygiene in membrane processing plants.  相似文献

Aims:  To investigate the factors affecting benzene biodegradation and microbial community composition in a contaminated aquifer.
Methods and Results:  We identified the microbial community in groundwater samples from a benzene-contaminated aquifer situated below a petrochemical plant. Eleven out of twelve groundwater samples with in situ dissolved oxygen concentrations between 0 and 2·57 mg l−1 showed benzene degradation in aerobic microcosm experiments, whereas no degradation in anaerobic microcosms was observed. The lack of aerobic degradation in the remaining microcosm could be attributed to a pH of 12·1. Three groundwaters, examined by 16S rRNA gene clone libraries, with low in situ oxygen concentrations and high benzene levels, each had a different dominant aerobic (or denitrifying) population, either Pseudomonas , Polaromonas or Acidovorax species. These groundwaters also had syntrophic organisms, and aceticlastic methanogens were detected in two samples. The alkaline groundwater was dominated by organisms closely related to Hydrogenophaga .
Conclusions:  Results show that pH 12·1 is inimical to benzene biodegradation, and that oxygen concentrations below 0·03 mg l−1 can support aerobic benzene-degrading communities.
Significance and Impact of the Study:  These findings will help to guide the treatment of contaminated groundwaters, and raise questions about the extent to which aerobes and anaerobes may interact to effect benzene degradation.  相似文献

Aims:  To analyse the arsenic-resistant bacterial communities of two agricultural soils of Bangladesh, to isolate arsenic-resistant bacteria, to study their potential role in arsenic transformation and to investigate the genetic determinants for arsenic resistance among the isolates.
Methods and Results:  Enrichment cultures were performed in a minimal medium in the presence of As(III) and As(V) to isolate resistant bacteria. Twenty-one arsenic-resistant bacteria belonging to different genera of Gram-positive and Gram-negative bacteria were isolated. The isolates, with the exception of Oceanimonas doudoroffii Dhal Rw, reduced 2 mmol l−1 As(V) completely to As(III) in aerobic conditions. Putative gene fragments for arsenite efflux pumps were amplified in isolates from Dhal soil and a putative arsenate reductase gene fragment was amplified from a Bacillus sp. from Rice soil.
Conclusions:  Phylogenetically diverse arsenic-resistant bacteria present in agricultural soils of Bangladesh are capable of reducing arsenate to arsenite under aerobic conditions apparently for detoxification purpose.
Significance and Impact of the Study:  This study provides results on identification, levels of arsenic resistance and reduction of arsenate by the bacterial isolates which could play an important role in arsenic cycling in the two arsenic-contaminated soils in Bangladesh.  相似文献

Aims:  This study aimed to determine the microbial diversity in the starter of Fen Liquor.
Methods and Results:  The plate method was used to enumerate the micro-organisms; meanwhile, the 16S rDNA of bacteria and the internal transcribed spacer of fungi were used to determine microbial diversity. Several genera were accordingly identified. Among the bacteria, Lactobacillales and Actinomycetales were detected only on the surface of the starter, whereas Bacillales was dominant within the starter. Among the fungi, Saccharomycopsis and Issatchenkia were the main genera in surface and interior starter, respectively; in addition, Thermomyces was found in interior starter, while other species of fungi were detected on the surface.
Conclusions:  The culture-dependent and polymerase chain reaction-based methods revealed the significant microbial diversity in different locations in the starter of Fen Liquor.
Significance and Impact of the Study:  This study is the first to identify the bacterial and fungal communities associated with the starter of Fen Liquor using both traditional and molecular methods; it is also the first to compare the microbial diversity on the surface of starter with that in the interior. The results enrich our knowledge on liquor-related micro-organisms, and can be used to promote the development of the traditional fermentation technology.  相似文献

Aims:  To assess the impact of Bacillus amyloliquefaciens and Microbacterium oleovorans on bacterial and fungal groups associated to the roots of field-grown maize.
Methods and Results:  Identification and count of bacterial and fungal culturable populations associated to the roots of maize seedlings, changes in culturable community structure according to the richness and diversity indexes concept and shifts in microbial activity through analysis of cellulolytic, ammonification and nitrification potentials were determined, in relation to kernel treatment with biological control agents. Following the treatment of maize kernels with B. amyloliquefaciens at 107 CFU ml−1, an increase in bacterial diversity was observed at the rhizoplane of resultant seedlings. Bacterial richness was significantly increased at the root inner tissues of seedlings treated with Mic. oleovorans . Fusarium , Aspergillus , Penicillium and Trichoderma were the main fungal genera isolated and there population sizes were unequally affected by the addition of biocontrol agents.
Conclusions:  Numbers and types of isolated bacteria and fungi changed in response to the addition of biocontrol agents, while microbial activity remained unchanged with respect to control.
Significance and Impact of the Study:  This study provides an insight of the effects of proven biocontrol agents on micro-organisms naturally associated to the target crop.  相似文献
