Abstract: Nuclear magnetic resonance (NMR) spin relaxation techniques were used to measure the rate of water exchange across the chloroplast envelope membrane and to detect exchange of water-hydrogens between the bound and fluid states in Acer platanoides shade leaves. Results indicate that individual water molecules resided within a chloroplast for 320 ± 65 ms, on average, before exchanging through the envelope, and that exchangable bound hydrogens are at least six times more abundant in the chloroplast than in the nonchloroplast compartments.  相似文献

A leafminer of the Nearctic genus Coptodisca Walsingham (Lepidoptera: Heliozelidae), a species of potential economic interest, is reported for the first time from Europe, infesting the black (Juglans nigra L.) and the common walnut (Juglans regia L.). Mines were collected since September of 2010 in several sites of two Italian regions (Campania and Lazio). The species is rather similar to Coptodisca juglandella (Chambers), the only Coptodisca known to attack walnuts, but at present, an unambiguous identification cannot be provided because of the unsatisfactory characterization of this leafminer and congeneric species. Three generations were recorded per year and leafminers overwinter as mature larvae. The first adults emerged in May–June while mature larvae of the last generation started the overwintering in September. During the last generation of the year, infestation levels of leaves were 100% in all sampled localities. Several species of parasitoids were reared from infested mines, with specimens belonging to the genus Chrysocharis (Hymenoptera: Eulophidae) being the most frequent parasitoids.  相似文献

[15N]-depleted (NH4)2SO4 applied to the soil in 1985 resulted in residual labeling of about 16% of the storage nitrogen (N) pool of mature walnut ( Juglans regia L. cv. Serr) trees in 1987. Application of [15N]-depleted (NH4)2SO4 fertilizer to a different set of mature walnut trees in 1987 allowed monitoring of the kinetics and utilization of N from current year uptake in 1987 and resulted in >20% labeling of fruit N following completion of leaf expansion. Redistribution of storage N to the new growth predominated during the spring flush of growth although N derived from the soil during current-year uptake contributed increasingly during leaf expansion. Labeled N from current year uptake accumulated preferentially in the leaves as compared with reproductive organs during leaf expansion but subsequent to leaf expansion, fruit were more highly labeled with N derived from current-year uptake than leaves. Pistillate flower abortion was coincident with an apparent competition for N among developing vegetative and reproductive organs and preceded the period of significant N contribution from current-year uptake.  相似文献

We evaluated a new, two-dimensional (2-D) nuclear magnetic resonance (NMR) imaging technique as a method for measuring the distribution of chloroplasts in leaves. NMR images that showed the distribution of chloroplast water and of total water as a function of depth into Acer platanoides sun and shade leaves were compared with the distribution of chlorophyll in the same leaf types (as measured by fluorescence microscopy), with the cellular structure (by scanning electron microscopy), and with published information. Results showed that the volume fraction of chloroplast water was much larger in shade than in sun leaves, and that it averaged about one-third larger in the palisade than in the spongy parenchyma region of both leaf types. Chlorophyll fluorescence was more intense in shade than in sun leaves. In sun leaves, fluorescence was maximal in the palisade region near the junction with the spongy parenchyma, while in shade leaves, fluorescence was maximal in the upper part of the spongy layer. We concluded that 2-D NMR imaging reliably indicates the location of chloroplast water.  相似文献

通过对比核桃枝条和绿豆茎内δD值差异来分析核桃和绿豆水分来源和利用。结果表明,核桃-绿豆农林复合系统的根系在表层土壤(0—30cm)中交叉存在,生态位重叠。旱季中表层土壤含水量与δD值之间存在显著的负相关关系(R2=0.77,P=0.02),雨季相关关系不显著(R2=0.03,P=0.73)。δD值分析表明,旱季中核桃利用深层土壤(30—80cm)水分占总水分来源的51%以上,雨季中则主要利用浅层土壤水分,间作绿豆和单作绿豆主要利用表层土壤水分。雨季中表层土壤水分能同时满足核桃和绿豆生长需要,但复合系统中光能竞争导致间作绿豆光合速率显著地低于单作绿豆。旱季间作绿豆0—20cm土壤水分含量、凌晨叶片水势和光合速率明显高于单作绿豆,显示间作绿豆体内水分状况好于单作绿豆。线性模型分析结果显示间作绿豆体内约有1.58%—5.39%的水分来核桃夜晚水力提升,表明复合系统在旱季一定程度上缓冲季节性水分胁迫对农作物生长的影响。  相似文献

Wetting of the upper leaf surface of Juglans regia L. and of model surfaces colonized by epiphytic micro-organisms was investigated by measuring contact angles of aqueous solutions buffered at different pH values. During June to October 1995, contact angles of aqueous solutions on the leaf surface of J. regia decreased by angles ranging from 12° (low pH values) to 25° at high pH values. At the end of this vegetation period, wetting was strongly dependent on pH showing significantly lower contact angles with alkaline solutions (pH 9·0) than with acidic solutions (pH 3·0). Contact angle titration measured angles on the leaf surface as a function of the pH of buffered aqueous solutions, covering a pH range from 3·0 to 11·0. Titration curves revealed inflection points around 7·5, indicating the existence of ionizable carboxylic groups at the interface of the phylloplane. Altered leaf-surface wetting properties observed on the intact leaf surface could be simulated in model experiments by measuring contact angles on artificial surfaces colonized by Pseudomonas fluorescens and by epiphytic micro-organisms isolated from the phylloplane of J. regia . Strong evidence is provided that interfacial carboxylic groups derive from epiphytic micro-organisms present on the phylloplane. Results suggest that the age-dependent increase in, and pH dependence of, wetting as leaves mature are related to the presence of epiphytic micro-organisms on the phylloplane. Ecological consequences of increased leaf-surface wetting, concerning the structure of the leaf surface as a microhabitat for epiphytic micro-organisms, are discussed.  相似文献

The model RATP (radiation absorption, transpiration and photosynthesis) is presented. The model was designed to simulate the spatial distribution of radiation and leaf-gas exchanges within vegetation canopies as a function of canopy structure, canopy microclimate within the canopy and physical and physiological leaf properties. The model uses a three-dimensional (3D) representation of the canopy (i.e. an array of 3D cells, each characterized by a leaf area density). Radiation transfer is computed by a turbid medium analogy, transpiration by the leaf energy budget approach, and photosynthesis by the Farquhar model, each applied for sunlit and shaded leaves at the individual 3D cell-scale. The model typically operates at a 20–30 min time step. The RATP model was applied to an isolated, 20-year-old walnut tree grown in the field. The spatial distribution of wind speed, stomatal response to environmental variables, and light acclimation of leaf photosynthetic properties were taken into account. Model outputs were compared with data acquired in the field. The model was shown to simulate satisfactorily the intracrown distribution of radiation regime, transpiration and photosynthetic rates, at shoot or branch scales.  相似文献

Diel variation in specific hydraulic conductivity (ks) was recorded in petioles of two savanna tree species, Schefflera macrocarpa and Caryocar brasiliense, from central Brazil. These two species have compound leaves with long petioles (10–30 cm). In both species, petiole ks decreased sharply with increasing transpiration rates and declining leaf water potentials (ψL) during the morning. Petiole ks increased during the afternoon while the plants were still transpiring and the water in the non‐embolized vessels was still under tension. Dye experiments confirmed that in both species diel variation in ks was associated with embolism formation and repair. When transpiration was prevented in individual leaves, their petiole ks and water potential remained close to their maximum values during the day. When minimum daily ψL on selected branches was experimentally lowered by 0.2–0.6 MPa, the rate of ks recovery during the afternoon was slower in comparison with control branches. Several field manipulations were performed to identify potential mechanisms involved in the refilling of embolized petiole vessels. Removal of the cortex or longitudinal incisions in the cortex prevented afternoon recovery of ks and refilling of embolized vessels. When distilled water was added to petiole surfaces that had been abraded to partially remove the cuticle, ks increased sharply during the morning and early afternoon. Evidence of starch to sugar conversion in the starch sheath cells surrounding the vascular bundles of the petioles was observed during periods of rapid transpiration when the abundance of starch granules in the starch sheath cells surrounding the vascular bundles decreased. Consistent with this, petiole sugar content was highest in the early afternoon. The most parsimonious explanation of the field observations and the experimental results was that an increase in osmotically active solutes in cells outside the vascular bundles at around midday leads to water uptake by these cells. However, the concurrent increase in tissue volume is partially constrained by the cortex, resulting in a transient pressure imbalance that may drive radial water movement in the direction of the embolized vessels, thereby refilling them and restoring water flow. This study thus presents evidence that embolism formation and repair are two distinct phenomena controlled by different variables. The degree of embolism is a function of tension, and the rate of refilling a function of internal pressure imbalances.  相似文献

目的探讨传统开腹手术与腹腔镜手术治疗子宫肌瘤的临床疗效。方法选择2013年2月~2015年11月在我院接受治疗的子宫肌瘤患者140例为研究对象,按患者自愿原则分为对照组和观察组各70例,对照组采用传统开腹手术治疗,观察组采用腹腔镜手术治疗,并对两组患者手术时间、术中出血量、术后感染情况和住院时间进行比较。结果观察组在手术时间、术中出血量、术后感染率及住院时间等方面均优于对照组,差异具有显著统计学意义(P0.05)。结论腹腔镜下子宫肌瘤切除术与传统开腹手术比较,具有出血量少、住院时间短、恢复快、并发症少等优点,值得临床推广应用。  相似文献

Objective: To evaluate difficulty chewing, consequent food choice and impacts using both open and closed questions within a semi‐structured interview format. Design: A cross‐sectional study using a semi‐structured interview (SSI) with open and closed questions Setting: Independently living people in South‐West London interviewed in their own homes Subjects: A group of 54, edentate people (mean age 82, R= 60‐93 years) were studied. Results: Most subjects (69%) expressed difficulty eating at least one type of food, half were unwilling to eat the foods they found difficult, others were conscious of cooking longer or cutting smaller in order to manage these foods, although no one accepted that they overcooked food. There were dramatic differences in the number of subjects expressing difficulty to chew various foods when responding to open and closed questions. Roast beef proved a good discriminator in this group of predominantly English subjects. The importance of the questioning process was also shown by the discrepancy of responses between variously prepared apples and the inability of any subjects to bite an unpeeled apple. Conclusions: For sensitive appraisal of food choice it is obviously essential to assess relevant foods, for the culture of the group being studied. Open questions provide a way of exploring which are the appropriate foods and food preparation as criteria to assess perceived chewing ability and treatment benefit. The semi‐structured interview method is particularly valuable for the study of older people. Closed questions provide different information which is also necessary to secure recall by older people.  相似文献

The short-chain oxidoreductase (SCOR) family of enzymes includes over 6000 members, extending from bacteria and archaea to humans. Nucleic acid sequence analysis reveals that significant numbers of these genes are remarkably free of stopcodons in reading frames other than the coding frame, including those on the antisense strand. The genes from this subset also use almost entirely the GC-rich half of the 64 codons. Analysis of a million hypothetical genes having random nucleotide composition shows that the percentage of SCOR genes having multiple open reading frames exceeds random by a factor of as much as 1 x 10(6). Nevertheless, screening the content of the SWISS-PROT TrEMBL database reveals that 15% of all genes contain multiple open reading frames. The SCOR genes having multiple open reading frames and a GC-rich coding bias exhibit a similar GC bias in the nucleotide triple composition of their DNA. This bias is not correlated with the GC content of the species in which the SCOR genes are found. One possible explanation for the conservation of multiple open reading frames and extreme bias in nucleic acid composition in the family of Rossman folds is that the primordial member of this family was encoded early using only very stable GC-rich DNA and that evolution proceeded with extremely limited introduction of any codons having two or more adenine or thymine nucleotides. These and other data suggest that the SCOR family of enzymes may even have diverged from a common ancestor before most of the AT-rich half of the genetic code was fully defined.  相似文献

This work represents an attempt to assess the relative contribution of the factors limiting productivity ofSpirulina platensis in open raceways throughout the year. Temperature of the culture during daylight exerted the predominant effect on productivity and elevating the temperature resulted in a significant rise in productivity even in summer. Photoinhibition had a decisive role in summer in determining productivity ofSpirulina in open raceways in that growth almost ceased after mid-day. Contamination by other microorganisms, particularlyS. minor andChlorella sp. was estimated to reduce the net biomass yield by at least 15 to 20%, but measures to curtail the establishment of these species in the raceway have been devised. The effect of harvest time during the day on the yield of dry mass was examined: no conclusion could be drawn except in mid-summer, when evening harvesting resulted in a significant increase in the output rate of dry weight over morning harvesting.It was estimated that in a subtropical climate with little cloudiness, it should be readily possible to obtain an annual output rate of dry mass of ca. 60 to 70 t ha–1. Such output, however, which would reduce very substantially the cost of production to-date, is possible only if the optimal temperature forSpirulina can be maintained throughout daylight, photoinhibition essentially controlled, harvesting in summer performed in the evening, and night-loss of dry mass as well as the extent of contamination by other cyanobacteria or microalgae can be kept low. The pronounced daily fluctuations in the output rate at peak productivity in summer suggest that when environmental limitations of growth are minimal, other limitations become dominant. These should be identified to facilitate an even greater increase in the productivity ofSpirulina in outdoor cultures.  相似文献

Aspects of the population dynamics and feeding activity of piscivorous birds in the small (c. 5 ha) intermittently open Riet River estuary, on the south-eastern coastline of South Africa, were investigated monthly from August 2005 to July 2006. A total of 188 birds of 13 species were recorded, of which six were wading piscivores, four aerial divers and three were pursuit swimmers. The Reed Cormorant (Phalacrocorax africanus) was the numerically dominant species, with a mean of 8.25 (SD ± 7.90) individuals per count. Mean numbers of the Pied Kingfisher (Ceryle rudis) and Giant Kingfisher (Megaceryle maximus) were 3.42 (SD ± 1.20) and 1.17 (SD ± 0.60) individuals per count, respectively. The remaining 10 species revealed mean values <0.5 individuals per count. Breaching events were associated with a change in feeding groups from waders to pursuit feeders, and a decrease in total bird numbers, most likely due to loss of potential littoral zone foraging habitat for waders resulting from reduced water levels. The highest bird numbers were recorded in winter reflecting the migration of large numbers of Reed Cormorant into the system. Monthly food consumption by all piscivorous birds showed large temporal variability, ranging from 26.35 to 140.58 kg per month.  相似文献

1. Acute air exposure of an air-breathing fish Channa argus immediately induced hypercapnic acidemia while total CO2 content of blood remained unchanged. Upon reimmersion, paco2 and pHa quickly restored to pre-exposure levels followed by gradual rise of [HCO-3]. 2. Artificial air ventilation of air-exposed fish restored acid-base status and greatly depressed voluntary air ventilatory movements.
