In order to obtain information on the content and composition of the water-soluble polysaccharides from Ulva clathrata, an extraction at 60°C, in different media, was performed: water, EDTA and HCl (F-I), each followed by a sequential extraction in NaOH 0.1 M (F-II). The extracts were recovered and analyzed for total carbohydrates, proteins, rhamnose, uronic acids and sulfate content. Differences were obtained in the yield and composition in both fractions of the different media (F-I and F-II). Higher yields resulted in the first fraction on all media. HCl extraction was the best in both fractions (14.83 ± 1.5% and 5.96 ± 1.1%, F-I and F-II, respectively). In all cases, F-I was more sulfated ranging from 27.87% to 35.8% and higher in rhamnose content, whereas F-II had higher protein and slightly higher uronic acid content. FTIR spectra showed that soluble polysaccharides from the green seaweed U. clathrata are sulfated polysaccharides, similar to ulvan obtained from other Ulva species and confirmed by the 1 H-NMR spectrum, where the characteristic signal for the deoxy sugar (rhamnose) is present.  相似文献

Sulfated polysaccharides (SPS) were extracted from three species of seaweeds of Ulvacea (Ulva pertusa, Ulva conglobata and Entromorpha prolifera) for 4 hr at various temperatures and their physicochemical properties were studied using viscometric and equilibrium sedimentation measurements in order to determine the optimum extracting condition.

Sulfated polysaccharides extracted at various temperatures from the seaweed of U. pertusa had the same physicochemical properties, while the larger molecular components of SPS was not extracted from U. conglobata and E. prolifera, at the low temperature of 30~40°C. This was confirmed by analyses of their viscosity and molecular weight and by gel filtration chromatography, in which each SPS showed two or three peaks.

The larger molecular component of SPS could be extracted at the high temperature of 80~90°C in the thermostable form.  相似文献

1. The anostracan fairy shrimp Branchinecta gaini inhabits one of the most hostile environments on earth, living in pools and lakes in Antarctica. Between January 2002 and January 2003 temperatures in two pools where B. gaini are extremely abundant on Adelaide Island ranged from ?18.6 to ?15.7 °C in winter, to 19.4 to 17.1 °C in summer, whilst air temperatures ranged from ?34 to 6.3 °C. 2. Branchinecta gaini survives winter as cysts, but endures large summer temperature fluctuations as adults. Cysts froze between ?24.4 and ?25.7 °C. In experiments adults survived 0–10 °C with no mortality for 1 week, 25 °C for nearly 48 h with 50% mortality, and at 32 °C complete mortality occurred in <1 h. 3. Oxygen consumption (M?O2) in B. gaini approximately doubled for every 10 °C temperature rise (Q10 = 2.04) up to 20 °C where it reached a peak. Females had, on average 19% higher M?O2 than males. Females also had greater metabolic scopes, (maximum–minimum M?O2 across temperatures was ×3.6 for females, ×3.1 for males). 4. Ventilation frequency increased linearly with temperature, and did not decline at 25 °C, indicating animals were ‘trying’ progressively harder to supply oxygen to tissues, and oxygen deficiency was the probable cause of death. Females had a higher ventilation frequency than males (8.6–17.1% higher) and they also exhibited greater scope to raise ventilation frequency (×2.4 for females versus ×1.5 for males). 5. Great metabolic flexibility allows B. gaini to exploit extreme, highly fluctuating environments, and larger ventilatory and respiratory scopes allow females to survive higher temperatures than males. Because of this flexibility their prospects for coping with physical environmental change are high.  相似文献

Stichococcus, a genus of green algae, distributes in ice-free areas throughout Antarctica. To understand adaptive strategies of Stichococcus to permanently cold environments, the physiological responses to temperature of two psychrotolerants, S. bacillaris NJ-10 and S. minutus NJ-17, isolated from rock surfaces in Antarctica were compared with that of one temperate S. bacillaris FACHB753. Two Antarctic Stichococcus strains grew at temperature from 4 to 25°C, while the temperate strain could grow above 30°C but could not survive at 4°C. The photosynthetic activity of FACHB753 at lower than 10°C was less than that of Antarctic algae. Nitrate reductase in NJ-10 and NJ-17 had its optimal temperature at 20°C, in comparison, the maximal activity of nitrate reductase in FACHB753 was found at 25°C. When cultured at 4–15°C a large portion of unsaturated fatty acids in the two Antarctic species was detected and the regulation of the degree of unsaturation of fatty acids by temperature was observed only above 15°C, though the content of the major unsaturated fatty acid αC18:3 in FACHB753 decreased with the temperatures elevated from 10 to 25°C. Elevated nitrate reductase activity and photosynthetic rates at low temperatures together with the high proportion of unsaturated fatty acids contribute to the ability of the Antarctic Stichococcus to thrive.  相似文献

The macroalga Ulva ohnoi constitutes a considerable fraction of green tides in coastal areas of Japan, but little is known about the physiological characteristics of this species. To investigate the environmental factors that promote the formation of green tides, we tested the responses of U. ohnoi and another common Japanese species, Ulva pertusa, to various levels of irradiance at different water temperatures. Because the two species are morphologically similar, we identified them using the PCR‐restriction fragment length polymorphism method. Under laboratory conditions, we evaluated the photosynthetic, dark respiration, and relative growth rate at a range of water temperatures (5 to 35°C) and photosynthetically active radiation (0 to 1000 μmol photons m?2 s?1). The maximum gross photosynthetic rate of U. ohnoi was larger than that of U. pertusa. The dark respiration rates revealed no significant differences among the species and temperature conditions. At 500 μmol photons m?2 s?1, the relative growth rate of U. ohnoi was larger than that of U. pertusa in higher temperature and the difference was the largest at 20°C. The estimated compensation irradiance and estimated saturation irradiance of U. ohnoi and U. pertusa ranged from 0.709 to 5.510 and 40.530 to 58.674 μmol photons m?2 s?1, which were lower than those in other intertidal green macroalgae, from 6 to 11 and 50 to 82 μmol photons m?2 s?1, respectively. Thus, U. ohnoi which exists as free‐floating near the water surface and accumulating inside the green tide can survive extensively in the water column of the intertidal zone, furthermore, the species can maintain rapid growth in this situation. Therefore, as a result of this study, it is suggested that the ecological success of U. ohnoi in shallow waters such as the tidal flats, estuarine, and coasts of the inner bay in comparison with U. pertusa.  相似文献

The minimum and maximum temperatures for germination of uredospores of Puccinia striiformis, P. recondite, P. coronata and P.gramnis isolates from south-central Chile were 0°C and 26°C, 0°C and 32°C, 8°C and 30°C, and 4°C and 34°C, respectively, whereas the shortest latent period was 8.5 days for P. struformis at 20°C, 5 days for P. recondite at 26°C, 5.5 days for P. coronata at temperatures, reaching the minimum and maximum threshold temperatures at 0.006°C and 23°C for P. strüformis, 0.31°C for P. recondita, 2.87°C and 30°C for P. coronata, and 1.72°C and 32°C for P. graminis. rspectively. The cardinal temperatures give no explanation as to the observed sequential appearance of these rusts during the growing season. Other phenomena like the systemic mycelial growth of P.striiformis might be involved here. At temperatures between 19°C and 22°C, the average daily increase of the area of sporulation of P.striiformis in wheat leaves varied between 9.05 and 22.48 mm2/infection site. This variation was due to substrate (plant) history and environmental factors.  相似文献

To investigate the ecological effect of macroalgae on de-eutrophication and depuration of mariculture seawater, the variation of dissolved inorganic nitrogen (DIN) and phosphate (DIP), the amount of Vibrio anguillarum, and total heterotrophic bacteria in Ulva clathrata culture, as well as on the algal surface, were investigated by artificially adding nutrients and V. anguillarum strain 65 from February to April 2006. The results indicated that U. clathrata not only had strong DIN and DIP removal capacities, but also showed a significant inhibitory effect on V. anguillarum, although not reducing the total heterotrophic bacteria. Vibrio anguillarum 65 dropped from 5∼8 × 107 cfu mL−1 to 10 cfu mL−1 (clone-forming units per mL) in 10 g L−1 of fresh U. clathrata culture within 2 days; i.e., almost all of the Vibrios were efficiently eradicated from the algal culture system. Our results also showed that the inhibitory effect of U. clathrata on V. anguillarum strain 65 was both DIN- and DIP-dependent. Addition of DIN and DIP could enhance the inhibitory effects of the algae on the Vibrio, but did not reduce the total heterotrophic bacteria. Further studies showed that the culture suspension in which U. clathrata was pre-cultured for 24 h also had an inhibitory effect on V. anguillarum strain 65. Some unknown chemical substances, either released from U. clathrata or produced by the alga associated microorganisms, inhibited the proliferation of V. anguillarum 65.  相似文献

Haplogloia andersonii (Farlow) Levring is an anti-tropical species that occurs on cold and warm-temperate Pacific coasts of both Americas. In its habit it resembles the subantarctic species Chordaria linearis (Hooker et Harvey) Cotton. Culture studies show that the species differ in morphology and ecophysiology of their microscopic gametophytes and in gamete behavior. Details of sporophyte anatomy are presented that also allow the distinction of field plants. In South America, H. andersonii occurs only on the Pacific coast, from central Perú (14°S) to southern Chile (50°S). Chordaria linearis occurs on the Pacific coast from Chiloé Island (43°S) to Cape Horn (56°S). In the shared area the species may co-occur. On the Atlantic coast, C. linearis was newly collected at a locality in northern Patagonia (41°S). In addition, C. linearis occurs in Antarctica. Haplogloia moniliformis Richer, recently described from Macquarie Island, is probably synonymous with Chordaria linearis.  相似文献

Pseudonitzschia seriata, isolated from sea ice in the Canadian Arctic, was grown in silicon-limited batch and semi-continuous culture to determine the effects of temperature on its nutrient utilization. Resource-saturated growth rate (μmax) increased significantly with temperature from -1.5 to 6°C with a Q10 of 1.63. The efficiency of the algae in using exogenous silicic acid for growth had no significant monotonic relationship with temperature but was significantly (P<0.05) greater in cultures at >0°C than in those at lower temperatures. Silicic acid uptake kinetics did not differ between high and low temperatures. Silicon cell quotas were significantly higher at temperatures <0°C, indicating greater silicon demand at low temperatures.P. seriata should not suffer silicon limitation in its natural ice habitat based on the observed kinetics, but its behaviour provides some support for the suggestion that temperatures <0°C are associated with diminished efficiency of nutrient utilization by cold ocean microalgae.  相似文献

The germination of seeds of three species of forage grasses, Lolium perenne, Festuca pratensis and Dactylis glomerata, was studied after storage for 3–5 years under five different storage conditions: in aluminium foil packets at —25°C, 0°C and laboratory temperature (c. 18°C), and in manilla paper packets at 0°C and laboratory temperature. With Lolium perenne and Festuca pratensis high germination values at 3 and 7 days were obtained from seed stored at — 25 °C and 0°C in foil packets (5% moisture), but at laboratory temperatures, seed from foil packets gave lower germination values than those from manilla paper packets. At all three temperatures Dactylis glomerata germination after 7 and 14 days was higher in seed stored in foil than in manilla packages. With all three species stored in manilla packets, germination was higher after laboratory than cold storage.  相似文献

Callophyllis variegata is a red alga that has been exported to Japan as an edible seaweed over the past few years. Available data strongly suggest that after a few years of exploitation of the C. variegata stands in southern Chile, a decline of its abundance seems to be occurring. However, there is not sufficient knowledge available to sustain harvesting strategies or to develop cultivation techniques. This study describes the C. variegata landings in the south of Chile and also provides some basic data on the biology of this species. Experiments related to the cultivation of early developmental stages, frond cultivation under controlled conditions and the regeneration capacity of the holdfast of this red alga are also presented. Spore production occurs mainly in autumn and winter and survival of carpospores decreased as the temperature increased from 8 to 12°C. Survival of tetraspores increased significantly, from 50 to 60% to over 80%, when temperature was raised from 8 to 12°C. Carpospore survival was also significantly affected by the photon flux density. This effect was mainly at 8°C, whereas at 33 µmol m?2 s?1 the survival of the spores is always low. The cultivation of apical portions of C. variegata under laboratory conditions showed that lower temperatures (8°C) significantly increased growth. Salinity and photon flux density did not have an effect on specific growth rate. Laboratory experiments demonstrated that holdfasts can regenerate fronds and that these fronds can be excised and cultivated, and are far more tolerant to environmental factors than the apical portions.  相似文献

Until recently it had been widely accepted that birds are energetically adapted to the latitude they inhabit, having an increased basal metabolic rate (BMR) at higher latitudes. Latterly, this general view has been questioned and the influence of phenotypic flexibility, due to factors such as habitat, life‐history or acclimatization has received increased attention. In particular, focus has been directed towards comparing species from arid and mesic habitats, but less attention has been given to species which breed in cold climates. We chose to study northern wheatears Oenanthe oenanthe from two populations at different latitudes (southern Norway, Iceland), but with similar life‐histories and habitat requirements throughout the year, in a common‐garden experiment. In order to assess true latitudinal trends in metabolic rate, we estimated the nocturnal resting metabolic rate (RMR) of northern wheatears from southern Norway and Iceland at different temperatures from 0° to 30°C. We found that Norwegian birds had overall lower metabolic rates than Icelandic birds, which were also slightly larger. This difference was not observed at 0°C, which might indicate that Icelandic birds might rely on better feather insulation reducing metabolic costs at very low temperatures. At temperatures above 10°C birds of both populations had constant metabolic values, indicating that their thermoneutral range almost completely covered the temperatures experienced during the breeding period. This study shows that the northern wheatear, which is one of only a few insectivorous long‐distance migratory songbirds occurring at such high latitudes, has evolved metabolic adaptations to life at cold temperatures which are endogenously determined.  相似文献

The incubation period and percentage hatching of eggs of pigmented and unpigmented Biomphalaria glabrata at constant temperatures were investigated in the range 14 °C to 34 °C. In order to determine the influence of extreme temperatures on adult snails, specimens of the same species were exposed to 0 °C and 40 °C for selected time periods. The results indicate that sustained temperatures below 16 °C and above 32 °C are detrimental to the development and hatching of B. glabrata embryos. The optimum temperatures for incubation period and hatching differ from each other. As far as temperature is concerned, this foreign snail species should be capable of successfully colonizing the warmer parts of southern Africa.  相似文献

The absence of the larval Anisakis simplex s. l. in samples from central Chile but heavy infections in fish from southern localities suggest the existence of two stocks of Strangomera bentincki, one closely associated with the central coast of Chile (landing port San Antonio and Talcahuano, c. 33° 30′ S–36° 40′ S) and the other associated with southern Chile (landing port Puerto Montt, c. 41° 30′ S). Results confirm the usefulness of metazoan parasites, like A. simplex s. l. as biological markers.  相似文献

The effect of genotype, the origin of genotype, and germination temperature on Scots pine pollen grain size, hydration rate, germinability, and tube growth was studied in vitro. The mean sizes of dry and germinated pollen grains varied among pollen genotypes in different ways, thus the hydration rate varied among genotypes. Pollen from Scots pine that originates in northern Finland hydrated more than pollen from a population in southern Finland. Germination temperature had no effect on the hydration rate. Germinability and tube growth rate of northern genotypes were higher at 20 °C than at 15 °C. Differences among southern genotypes were not significant. At 15 °C, the germinability and pollen tube growth rate of northern genotypes were lower than southern genotypes. At 20 °C, the differences were not significant. It appears that germination and growth of pollen from northern populations are enhanced at higher temperatures whereas pollen from southern populations is unaffected.  相似文献

The marcoalga Ulva pertusa was cultured under (20 ± 2)°C, (20 ± 4)°C, (20 ± 6)°C, (20 ± 8)°C and (20 ± 10)°C circadian rhythms of fluctuating temperature conditions, and constant temperature of 20°C was used as the control. The growth rate of macroalga at (20 ± 2)°C, (20 ± 4)°C and (20 ± 6)°C were significantly higher than that at constant temperature of 20°C, while growth rate at (20 ± 8)°C and (20 ± 10)°C were significantly lower than that at constant temperature of 20°C. The growth rate of macroalga was a quadratic function of the thermal amplitude. Such a growth model can be described by G = β 0 + β 1(TA) + β 2(TA)2, where G represents the relative growth rate, TA is thermal amplitude in degree Celsius, β 0 is the intercept on the G axis, and β 1 and β 2 are the regression coefficients. The optimal thermal amplitude for the growth of thallus at mean temperature of 20°C was estimated to be ± 3.69°C. Analysis of biochemical composition at the final stages of thaulls growth revealed that diel fluctuating temperature caused various influences (P < 0.05). The content of chlorophyll, protein and total solute carbohydrate at (20 ± 2)°C and (20 ± 4)°C were slightly higher than those at constant temperature of 20°C, however no statistically significant differences were found among them (P > 0.05). While osmolytes (total solute carbohydrate and free proline) at (20 ± 10)°C were significantly higher than that at 20°C (P < 0.05). Therefore, more chlorophyll and carbohydrate production might account for the enhancement in the growth of macroalga at the diel fluctuating temperatures in the present study. Handling editor: S. M. Accommodating weanling horses in loose housing (sleeping hall with deep-litter bed and paddock) environments in winter at northern latitudes exposes the nonhuman animals to low ambient temperatures. We determined the heat loss of nine weanling horses in a cold environment by infrared thermography to assess their thermoregulatory capacity. The rate of heat loss was 73.5 to 98.7 W/m2 from the neck and 69.9 to 94.3 W/m2 from the trunk. The heat loss was higher at -16 °C than at 0 °C and -9 °C (p ≤ .01), indicating that the lower critical temperature may have been between -9 °C and -16 °C. Surprisingly, the heat loss from the trunk was less at -23 °C than -16 °C (p < .05). The frost on the surface of the hair coat at -23 °C probably disturbed the thermographic examination. Thus, thermography is not necessarily suitable for determining heat loss at very cold temperatures. Our results emphasize the importance of taking the housing temperature into account in planning the feeding of cold-housed weanling horses in northern latitudes.  相似文献

Antarctic wetlands are characterized by the presence of liquid water during short austral summer. Filamentous cyanobacteria are often dominant there and are exposed to severe conditions, of which the changes in the desiccation–rehydration and freeze–thaw cycles are two of the most stressful. Vigor, after freezing and desiccation, was laboratory tested in cyanobacterial and algal strains from wetland habitats collected in maritime and continental Antarctica. Whereas minor sub-zero temperatures (−4°C), demonstrating summer diurnal freeze–thaws did not cause significant damage on either cyanobacteria or algae, low sub-zero temperatures (−40, −100, −196°C), demonstrating annual winter freeze, caused little harm to cyanobacteria, but was fatal for more than 50% of the population of algae. Freezing and desiccation tolerance of these strains was compared using multiregression methods: cyanobacteria from continental Antarctica were significantly more tolerant to low sub-zero temperatures than similar strains from maritime Antarctica (P = 0.026; F = 3.66); and cyanobacteria from seepages habitat were less tolerant to freezing and desiccation than cyanobacteria from other wetlands (P = 0.002; F = 5.69).  相似文献

The psychrotolerant yeast Mrakia frigida 2E00797 isolated from sea sediment in Antarctica was found to be able to produce killer toxin against the pathogenic yeast (Metschnikowia bicuspidata WCY) in crab. When the psychrotolerant yeast was grown in the medium with pH 4.5 and 3.0% (wt/vol) NaCl and at 15°C, it could produce the highest amount of killer toxin against the pathogenic yeast M. bicuspidata WCY. The crude killer toxin activity against the pathogenic yeast M. bicuspidata WCY was the highest when it grew at 15°C in the assay medium with 3.0% (wt/vol) NaCl and pH 4.5. At temperatures higher than 25°C, the killing activity produced by M. frigida 2E00797 was completely lost and after the crude killer toxin was pre-incubated at temperatures higher than 40°C for 4 h, the killing activity was also completely lost. The killer toxin produced by M. frigida 2E00797 could kill only M. bicuspidata WCY, Candida tropicalis and Candida albicans among all the fungal species and bacterial species tested in this study.  相似文献
