The objectives of Experiment 1 were to determine a dose of eCG that would increase total luteal volume and plasma progesterone (P4) concentration on estrous cycle Day 7 in cows. The objectives of Experiment 2 were to determine the effects of treating embryo recipient lactating Holstein cows with eCG on pregnancy per embryo transfer (P/ET). In Experiment 1, lactating dairy cows at 63 ± 3 d postpartum (DIM) received no treatment (control, n = 10), or 600 (eCG6, n = 19), or 800 (eCG8, n = 19) IU of eCG 2 d after the start of the ovulation-synchronization protocol, Day -8 (Day -10 GnRH, Day -3 PGF, Day 0 GnRH). Blood was sampled on Days -10, -8, -3, 0, 7, and 14 for P4 concentration. Ovaries were examined by ultrasound on Days -10, -3, 0, and 7. In Experiment 2, lactating dairy cows were paired according to parity and previous insemination (0 or > 1 insemination) and assigned to receive 800 IU of eCG (eCG8, n = 152) 2 d after the start of the ovulation-synchronization protocol (Day -10 GnRH, Day -3 PGF, Day 0 GnRH) or to receive no treatment (control, n = 162). Blood was sampled on Days -10, -3, 0, 7, and 14 for determination of P4 concentration. Ovaries were examined by ultrasound on Days -10, -3, and 7, and cows with a CL > 20 mm in diameter on Day 7 received an embryo. In Experiment 1, P4 concentration on Day 7 was higher (P < 0.05) for eCG8 cows (2.3 ± 0.3 ng/mL) compared with control (1.2 ± 0.3 ng/mL) and eCG6 (1.1 ± 0.3 ng/mL) cows. In Experiment 2, eCG8 primiparous cows had more (P < 0.01) follicles > 10 mm on Day -3 compared with control primiparous cows (2.5 ± 0.9 vs 1.7 ± 0.5 mm), but multiparous control and eCG8 cows did not differ. A larger (P = 0.03) percentage of control cows received an embryo (87.5 vs 79.1%) compared with eCG8 cows. Among cows that received an embryo, total luteal volume on Day 7 was affected (P = 0.05) by treatment (eCG8 = 8.3 ± 0.4 cm3, control = 6.2 ± 0.4 cm3), but P4 concentration on Day 7 did not differ significantly between treatments. The percentage of cows pregnant 53 d after ET (overall, 24.2%) was not significantly different between control and eCG8 cows. In the current study, no differences in P/ET were observed between control and eCG8 cows and treatment with eCG increased the percentage of cows with asynchronous estrous cycle.  相似文献

Objectives were to determine progesterone concentration (P4) from days 4 to 28 relative to presumptive estrus necessary for maintenance of pregnancy in lactating Holstein cows. Cows were assigned to the low P4 (LowP4, n = 28) or control (n = 153) treatments. All cows were presynchronized with two injections of PGF (14 d apart) and an ovulation synchronization protocol (11 d later; GnRH on day −10, PGF on day −3; and GnRH on day 0 = presumptive estrus). Cows in the Low P4 treatment received 2 injections of prostaglandin F on days 4 and 5 (day 0 = presumptive estrus) and a new CIDR insert on day 5 that was replaced every 7 d until day 28. Blood was sampled on days −9, −2, 0, 4, 7,14, 21, and 28. Ovaries were examined with ultrasound on days −9, −3, and 7 and cows bearing a corpus luteum ≥20 mm on day 7 received an embryo. On days 0, 4 and 7 P4 did not differ (P ≥ 0.27) but control cows had greater (P < 0.01) P4 on days 14, 21, and 28. Progesterone concentration fold change from day 0 to 7 was not (P = 0.14) affected by treatment, but P4 concentration fold change from day 7 to 14 was (P < 0.01) greater for control cows compared with LowP4 cows. No LowP4 cows became pregnant after embryo transfer, whereas 35.7, 25.5, and 21.4% of control cows were pregnant on day 28, 42, and 63, respectively. Progesterone concentration fold changes from day 0 to 7 (P = 0.03) and from day 7 to 14 (P = 0.05) were associated with pregnancy outcomes on day 63. Among cows that were pregnant on day 63, the minimum P4 concentration fold changes from day 0 to 7 and from day 7 to 14 were 2.71 and 1.48, respectively. Interestingly, cows with P4 concentration <5 ng/mL on day 14 were (P = 0.01) and tended to be (P = 0.07) more likely to lose pregnancy from day 28 to 42 and from day 28 and 63, respectively. Faster rise in P4 concentration during the metestrus and early diestrus are associated with pregnancy establishment following embryo transfer, which suggests that early rise in P4 concentration has an indirect effect on embryo development through modulation of uterine environment and secretion of histotroph. Furthermore, the positive effects of early rise in P4 concentration appear to go beyond the phase of maternal recognition of pregnancy through adhesion and placentation stages.  相似文献

Pregnancy per artificial insemination (AI) was evaluated in dairy cows (Bos taurus) subjected to synchronization and resynchronization for timed AI (TAI). Cows (n = 718) received prostaglandin F (PGF) on Days –38 and –24 (Days 39 and 53 postpartum), gonadotropin-releasing hormone (GnRH) on Day –10, PGF on Day –3, and GnRH and TAI on Day 0. Between Days –10 and –3, cows received a progesterone intravaginal insert (CIDR group) or no CIDR (Control group). Between Days 14 and 23, cows received a CIDR (Resynch CIDR group) or no CIDR (Resynch control group), GnRH on Day 23, with pregnancy diagnosis on Day 30. Cows in estrus (between Days 0 and 30) were re-inseminated at detected estrus (RIDE). Nonpregnant cows received PGF on Day 30 and GnRH and TAI on Day 33. Plasma progesterone was determined to be low or high on Days –24 and –10. Pregnancy rates were evaluated 30 and 55 d after AI. The CIDR insert included in the Presynch-Ovsynch protocol did not increase overall pregnancy per AI for first service (36.1% and 33.6% for CIDR; 34.1% and 28.8% for Control) but did decrease pregnancy loss (7.0% for CIDR and 15.6% for Control). The CIDR insert increased pregnancy per AI in cows with high progesterone at the time the CIDR insert was applied. Administration of a CIDR insert between Days 14 and 23 of the estrous cycle after first service did not increase overall pregnancy per AI to second service (24.7% and 22.7% for Resynch CIDR; 28.6% and 25.3% for Resynch control). For second service, RIDE cows had lower pregnancy rates in the Resynch CIDR group than in the Resynch control group. Cows with a CL (corpus luteum) at Day 30 had higher pregnancy rates in the Resynch CIDR group than those in the Resynch control group.  相似文献

Meticulous planning is required to minimize heat-stress conditions in barns. The objective of this study was to determine optimum barn characteristics for high-yielding dairy cows under Israeli (Mediterranean) summer ambient conditions, by using a new stress model that takes ambient temperature, relative humidity and wind velocity into account. During the summers of 2004 and 2005, three meteorological stations were alternately installed in 39 barns: two stations inside the barn at the prevailing downwind direction, and a third station outside the upwind end of the barn. Ambient temperature, relative humidity, wind speed and direction were measured and recorded every 10 min for 3 to 5 consecutive days at each barn in turn. The data were collected at different geographical and climatic conditions. Therefore, the data collected by an outside station were used as covariates. A heat-stress model was used to determine the threshold temperature (THRT) at which a cow begins to increase its respiratory rate; THRT was the response variable in the statistical model. The THRT model takes in account assumed values of a cow's physiological characteristics: daily milk yield of 45 kg, containing 3.5% fat, and 3 mm fur depth. The independent variables were: orientation, barn type, roof slope, roof ridge, marginal height, roof type (fixed or sliding) and barn width. Results showed that the optimal barn for high-yielding cows is the loose-housing type, oriented with its long axis perpendicular to the prevailing wind direction. Advantageous to the design would be an open ridge or pagoda with marginal height of over 4.7 m for north-south orientation and over 5 m for east-west orientation, roof slope over 11%, and barn width between 43 and 51 m for north-south orientation but lower than 42 m for east-west orientation. A sliding roof was also found to be an excellent solution when outside yards are banned by environmental regulations.  相似文献

Grass silage is typically fed to dairy cows in temperate regions. However, in vivo information on methane (CH4) emission from grass silage of varying quality is limited. We evaluated the effect of two rates of nitrogen (N) fertilisation of grassland (low fertilisation (LF), 65 kg of N/ha; and high fertilisation (HF), 150 kg of N/ha) and of three stages of maturity of grass at cutting: early maturity (EM; 28 days of regrowth), mid maturity (MM; 41 days of regrowth) and late maturity (LM; 62 days of regrowth) on CH4 production by lactating dairy cows. In a randomised block design, 54 lactating Holstein–Friesian dairy cows (168±11 days in milk; mean±standard error of mean) received grass silage (mainly ryegrass) and compound feed at 80 : 20 on dry matter basis. Cows were adapted to the diet for 12 days and CH4 production was measured in climate respiration chambers for 5 days. Dry matter intake (DMI; 14.9±0.56 kg/day) decreased with increasing N fertilisation and grass maturity. Production of fat- and protein-corrected milk (FPCM; 24.0±1.57 kg/day) decreased with advancing grass maturity but was not affected by N fertilisation. Apparent total-tract feed digestibility decreased with advancing grass maturity but was unaffected by N fertilisation except for an increase and decrease in N and fat digestibility with increasing N fertilisation, respectively. Total CH4 production per cow (347±13.6 g/day) decreased with increasing N fertilisation by 4% and grass maturity by 6%. The smaller CH4 production with advancing grass maturity was offset by a smaller FPCM and lower feed digestibility. As a result, with advancing grass maturity CH4 emission intensity increased per units of FPCM (15.0±1.00 g CH4/kg) by 31% and digestible organic matter intake (33.1±0.78 g CH4/kg) by 15%. In addition, emission intensity increased per units of DMI (23.5±0.43 g CH4/kg) by 7% and gross energy intake (7.0±0.14% CH4) by 9%, implying an increased loss of dietary energy with advancing grass maturity. Rate of N fertilisation had no effect on CH4 emissions per units of FPCM, DMI and gross energy intake. These results suggest that despite a lower absolute daily CH4 production with a higher N fertilisation rate, CH4 emission intensity remains unchanged. A significant reduction of CH4 emission intensity can be achieved by feeding dairy cows silage of grass harvested at an earlier stage of maturity.  相似文献

In all, 18 multiparous and 19 primiparous Holstein dairy cows were used in a completely randomized design with restrictions to evaluate the effects of feeding propylene glycol (PG) as a dry product, via two delivery methods, on production and blood parameters. PG treatments were administered from parturition through 21 days postpartum. Treatments were: (i) control, no PG; (ii) top dress, 162.5 g PG/day by top dressing onto the total mixed ration (TMR) and; (iii) mixing, 162.5 g PG/day as a part of the TMR by incorporating it into the TMR. PG used was a dry product which contained 65% pure PG and 35% silicon dioxide as the dry carrier. Coccygeal blood was sampled on 4, 7, 14 and 21 days in milk (±1.50 pooled s.d.). Supplementation of dry PG by top dressing onto, or incorporating into, the TMR had no effects on average dry matter intake, milk yield and composition, serum insulin, serum and plasma metabolites and milk ketones. Concentrations of urine ketones tended (P = 0.10) to be reduced by PG supplementation from 41.5 to 15.2 mg/dl. Supplementation of PG tended (P = 0.07) to decrease the incidence for subclinical ketosis from 39% to 24% and 13% for cows fed a TMR supplemented with no dry PG, with dry PG as a top dress and dry PG as a part of the TMR, respectively. It is concluded that supplementing PG as a dry product via incorporating into the TMR is as effective as when used as a top dress, based on the efficacies of both delivery methods to numerically reduce urine ketones concentrations and, therefore, the incidence for subclinical ketosis during the first 21 days of lactation. However, it should be noted that the number of cows used in the current study was minimal, and more cows are needed to confirm the efficacy of supplementing PG as a dry product on reducing the prevalence of subclinical ketosis in dairy cows during the first month of lactation.  相似文献

The aim of this study was to evaluate the effect of presynchronization with or without the detection of estrus on first service pregnancy per artificial insemination (P/AI) and on Ovsynch outcome in lactating dairy cows. A total of 511 cows were divided randomly but unevenly into 3 treatment groups at 44 to 50 days in milk (DIM). Ovsynch was started at the same time (69 to 75 DIM) in all three groups. Cows in the Ovsynch group (CON, N = 126) received no presynchronization before Ovsynch, and all cows were bred by timed AI (TAI). Cows in the presynchronization with estrus detection (PED) and the presynchronization with only TAI (PTAI) groups received two doses of prostaglandin F (PGF) 14 days apart, starting at 44 to 50 DIM. Ovsynch was initiated 11 days after the second PGF treatment. Cows in the PED group (N = 267) received AI if estrus was detected after either PGF injection. Cows that were not determined to be in estrus after PGF injection received Ovsynch and TAI. Cows in the PTAI group (N = 118) were not inseminated to estrus, with all cows receiving TAI after Ovsynch. The ovulatory response to the first GnRH injection administered as part of Ovsynch differed (P = 0.002) among treatment groups (83.1% in PTAI, 72.6% in PED, and 62.7% in CON). However, the ovulatory response to the second injection of GnRH during Ovsynch did not differ among treatment groups. Of the 267 PED cows, a total of 132 (49.4%) exhibited estrus and were inseminated. The P/AI at the 31-day pregnancy diagnosis was similar between the cows in the PED group with AI after estrus detection (37.9%; 50/132) and those bred with TAI (34.1%; 46/135). The P/AI in the CON group (46.8%; 59/126) was greater (P < 0.05) than that in the PED group (36.0%; 96/267). In addition, the P/AI in the CON group was greater (P = 0.04) than that in the PED cows receiving TAI (34.1%; 46/135) but less than that in the PED cows bred to estrus (37.9%; 50/132) (P = 0.16). At the 31-day pregnancy diagnosis, the cows in the PTAI group had greater P/AI (55.9%; 66/118) than both those in the PED group (P < 0.01; either estrus or TAI) and those in the CON group (P = 0.08). Thus, presynchronization with PGF (PTAI) increased the ovulatory response to Ovsynch and improved P/AI in dairy cows. Interestingly, the breeding of cows to estrus during presynchronization reduced fertility to the TAI and overall fertility, including cows bred to estrus and TAI. These results indicate that maximal fertility is obtained when all cows receive TAI after the presynchronization protocol.  相似文献

Lying behaviour is important for the welfare of the cow. Therefore, reliable electronic devices may improve the management of the cows, and the devices can be used as a tool in research. However, accelerometer-based devices measure acceleration, and an algorithm is therefore necessary for the calculation of lying behaviour. Thus, validation of such devices is imperative prior to use. The objective of this study was to validate the use of the AfiTagII device for measurements of the lying time and frequency of lying bouts of Danish Holstein (DH) and Danish Jersey (DJ) cows in a loose-house system on two different bedding materials. The validation included correlations and linear regression analyses of data collected by the AfiTagII compared with data collected both by direct observations and recordings from a previously validated device (IceQube). In total, 40 cows were observed directly with primiparous and multiparous DJ and DH cows, equally represented. Furthermore, 21 cows were monitored with both AfiTagII and IceQube devices, and data from both devices were collected simultaneously. The devices were attached to the hind leg of the cow. The estimates of the lying time from the AfiTagII device was highly correlated with the recordings from direct observations (r=0.98), and there was a linear relation between these with an intercept equal to 0 and a slope close to 1. The estimates of the lying time from the AfiTagII device was also highly correlated with the IceQube recordings (r=0.94). However, the intercept deviated from 0. The frequency of lying bouts recorded by the AfiTAgII compared to direct observations showed a positive predictive value of 0.96 for lactating cows on the slatted floor and of 0.85 for the dry cows on the deep bedding. The correlations between frequency of lying bouts recorded with the two devices were high (r=0.94), but the intercept deviated from 0. In conclusion, the AfiTagII has a high accuracy for the measurements of lying behaviour in both DH and DJ cows kept on different bedding materials.  相似文献

As a result of the increase in herd size and the intensification of production, the complexity of reproductive management has been growing in dairy herds. The aim of our study was to examine the associations of management practices and reproductive performance in Holstein cows on large commercial dairy farms. Management practices applied to cows were surveyed between 22 May and 6 November 2015 in 34 large Holstein-Friesian dairy herds in Hungary. Individual data of 23 784 cows that calved between 1 January 2014 and 31 December 2014 in the surveyed herds were gathered. Associations between the management practices and the reproductive parameters were analyzed by mixed effects models. Regarding heat abatement we found that ventilation with sprinklers was associated with the shortest breeding interval (P<0.01), the shortest calving to conception interval (P<0.01), and the highest odds of being pregnant by 200 days in milk (P<0.01). Solely ventilation showed similar (P>0.05) results to lack of heat stress protection. It was also revealed, that lack of a well-established voluntary waiting period (VWP) or a VWP shorter than 50 days was associated with less days to first service (P<0.01), shorter breeding interval (P<0.01) and calving to conception interval (P<0.05), as well as higher odds of carrying a calf by 200 days in milk (P<0.01) compared with those using a VWP of at least 50 days. Those farms that applied transrectal ultrasonography were more likely to use ventilation with sprinklers (P<0.05), hormonal synchronization (P<0.01) and to perform early pregnancy diagnosis followed by pregnancy recheck (P<0.05). The application of transrectal ultrasonography with one of the aforementioned practices was associated with reduced days to first service (P<0.05), shorter breeding interval (P<0.05) and higher odds of pregnancy at 200 days in milk (P<0.05). Our study highlights the management practices most closely related to improved reproductive performance, which are, therefore, suggested to be applied on dairy farms, considering the local circumstances of the individual farms.  相似文献

Involuntary soil intake by cows on pasture can be a potential route of entry for pollutants into the food chain. Therefore, it appears necessary to know and quantify factors affecting soil intake in order to ensure the food safety in outside rearing systems. Thus, soil intake was determined in two Latin square trials with 24 and 12 lactating dairy cows. In Trial 1, the effect of pasture allowance (20 v. 35 kg dry matter (DM) above ground level/cow daily) was studied for two sward types (pure perennial ryegrass v. mixed perennial ryegrass–white clover) in spring. In Trial 2, the effect of pasture allowance (40 v. 65 kg DM above ground level/cow daily) was studied at two supplementation levels (0 or 8 kg DM of a maize silage-based supplement) in autumn. Soil intake was determined by the method based on acid-insoluble ash used as an internal marker. The daily dry soil intake ranged, between treatments, from 0.17 to 0.83 kg per cow in Trial 1 and from 0.15 to 0.85 kg per cow in Trial 2, reaching up to 1.3 kg during some periods. In both trials, soil intake increased with decreasing pasture allowance, by 0.46 and 0.15 kg in Trials 1 and 2, respectively. In Trial 1, this pasture allowance effect was greater on mixed swards than on pure ryegrass swards (0.66 v. 0.26 kg reduction of daily soil intake between medium and low pasture allowance, respectively). In Trial 2, the pasture allowance effect was similar at both supplementation levels. In Trial 2, supplemented cows ate much less soil than unsupplemented cows (0.20 v. 0.75 kg/day, respectively). Differences in soil intake between trials and treatments can be related to grazing conditions, particularly pre-grazing and post-grazing sward height, determining at least in part the time spent grazing close to the ground. A post-grazing sward height lower than 50 mm can be considered as a critical threshold. Finally, a dietary supplement and a low grazing pressure, that is, high pasture allowance increasing post-grazing sward height, would efficiently limit the risk for high level of soil intake, especially when grazing conditions are difficult. Pre-grazing and post-grazing sward heights, as well as faecal crude ash concentration appear to be simple and practical tools for evaluating the risk for critical soil intake in grazing dairy cows.  相似文献

The objective of this study was to describe the genetic and phenotypic relationship between milk urea nitrogen (MUN) and reproductive traits in Iranian Holstein dairy cows. Test-day MUN data obtained from 57 301 dairy cows on 20 large dairy herds in Iran between January 2005 and June 2009. Genetic parameters for MUN and reproductive traits were estimated with a five-trait model using ASREML program. Random regression test-day models were used to estimate heritabilities separately for MUN from first, second and third lactations. Regression curves were modeled using Legendre polynomials of order 3. Herd-year-season along with age at calving was included as fixed effects in all models for reproductive traits. Heritabilities for MUN and reproductive traits were estimated separately for first lactation, second lactation and third lactation. The estimated heritabilities for MUN varied from 0.18 to 0.22. The heritability estimate was low for reproductive traits, which ranged from 0.02 to 0.06 for different traits and across parities. Except for days open, phenotypic and genetic correlations of MUN with reproductive performance traits were close to zero. Genetic correlations between MUN and days open were 0.23, 0.35 and 0.45 in first, second and third lactation, respectively. However, the phenotypic correlation between MUN at different parities was moderate (0.28 to 0.35), but the genetic correlation between MUN at different parities was high and ranged from 0.84 to 0.97. This study shows a limited application of MUN for use in selection programs to improve reproductive performance.  相似文献

Milk fatty acid (FA) profile has been previously used as a predictor of enteric CH4output in dairy cows fed diets supplemented with plant oils, which can potentially impact ruminal fermentation. The objective of this study was to investigate the relationships between milk FA and enteric CH4 emissions in lactating dairy cows fed different types of forages in the context of commonly fed diets. A total of 81 observations from three separate 3×3 Latin square design (32-day periods) experiments including a total of 27 lactating cows (96±27 days in milk; mean±SD) were used. Dietary forages were included at 60% of ration dry matter and were as follows: (1) 100% corn silage, (2) 100% alfalfa silage, (3) 100% barley silage, (4) 100% timothy silage, (5) 50 : 50 mix of corn and alfalfa silages, (6) 50 : 50 mix of barley and corn silages and (7) 50 : 50 mix of timothy and alfalfa silages. Enteric CH4output was measured using respiration chambers during 3 consecutive days. Milk was sampled during the last 7 days of each period and analyzed for components and FA profile. Test variables included dry matter intake (DMI; kg/day), NDF (%), ether extract (%), milk yield (kg/day), milk components (%) and individual milk FA (% of total FA). Candidate multivariate models were obtained using the Least Absolute Shrinkage and Selection Operator and Least-Angle Regression methods based on the Schwarz Bayesian Criterion. Data were then fitted into a random regression using the MIXED procedure including the random effects of cow, period and study. A positive correlation was observed between CH4 and DMI (r=0.59,P<0.001), whereas negative associations were observed between CH4 and cis9-17:1 (r=−0.58, P<0.001), and trans8, cis13-18:2 (r=−0.51,P<0.001). Three different candidate models were selected and the best fit candidate model predicted CH4 with a coefficient of determination of 0.84 after correction for cow, period and study effects and was: CH4 (g/day)=319.7−57.4×15:0−13.8×cis9-17:1−39.5×trans10-18:1−59.9×cis11-18:1−253.1×trans8, cis12-18:2−642.7×trans8, cis13-18:2−195.7×trans11, cis15-18:2+16.5×DMI. Overall and linear prediction biases of all models were not significant (P>0.19). The objective of this experiment was to quantify the effects of unroasted or roasted ground-shelled corn (GSC), when fed with alfalfa ensiled in bag, bunker, or O2-limiting tower silos on ruminal digestion and microbial CP synthesis in lactating dairy cows. The roasted corn was heat-treated in a propane-fired roasting system. Alfalfa was harvested as second cutting from fields with regrowth of the same maturity. A portion of each field was allotted to each silo. The diets with 3 × 2 factorial arrangement of treatments were fed to six multiparous rumen-cannulated Holstein cows in a cyclic change-over design with five 21-day periods. Experimental diets were comparable and averaged (on dry matter (DM) basis): 410 g/kg alfalfa silage (AS), 150 g/kg corn silage, 350 g/kg GSC, 50 g/kg soybean meal, 40 g/kg roasted soybeans, 177 g/kg CP, 264 g/kg NDF and 250 g/kg starch. Nutrient flow was quantified by the omasal sampling technique with use of three markers (Co, Yb and indigestible NDF). Continuous infusion of 10% atom excess (15NH4)2SO4 was used to label microbial CP. None of the interactions between storage structure of dietary AS and corn type were significant. DM intake was not different among dietary treatments, averaging 24.5 kg/day across diets. Means of ADF digested in the rumen for cows fed diets with AS from bag, bunker and O2-limiting tower silo were 2.1, 1.7 and 2.1 kg/day, respectively, and was lower in cows fed AS from the bunker silo. This response may partly be a reflection of the higher intake of ADF by cows fed AS ensiled in the O2-limiting tower silo compared with the bunker. There was a slightly greater supply of fermentable substrates for cows fed diets with roasted compared with unroasted GSC. The small increases in yield of milk protein and lactose observed in the previous production trial in cows fed diets containing roasted corn may have occurred because of greater supply of fermentable substrates.  相似文献

Fertility in dairy cows has been declining for the past three decades. Genetic selection for increased milk production has been associated with changes in key metabolic hormones (growth hormone, insulin, IGF and leptin) that regulate metabolism by homoeostasis and homeorhesis. These metabolic hormones, particularly insulin, provide signals to the reproductive system so that regulation of ovarian function is coordinated with changes in metabolic status. Studies have shown, for example, that increasing circulating insulin concentrations during the early post partum period can advance the resumption of oestrous cycles by enhancing follicular growth. However, high concentrations of insulin can be detrimental to the developmental competence of oocytes, which is also influenced by the supply of fatty acids at the systemic level and at the ovarian level. Insulin status is also associated with the incidence and characteristics of abnormal ovarian cycles. These changes can occur without significant variation in circulating gonadotrophin concentrations. This suggests that additional factors, such as peripheral metabolites, metabolic hormones and locally produced growth factors, may have a modulating role. Recent evidence has demonstrated that ovarian responses to metabolic signals and nutrient profile vary according to the stage of the reproductive cycle. Improved understanding of this multifactorial process enables nutrition to be matched to genotype and milk production, with a positive impact on pregnancy rate.  相似文献

The objective of this study was to evaluate the effect of supplemented condensed tannins (CT) from the bark of the Black Wattle tree (Acacia mearnsii) on production variables and N use efficiency in high yielding dairy cows. A feeding trial with 96 lactating German Holstein cows was conducted for a total of 169 days, divided into four periods. The animals were allotted to two groups (control (CON) and experimental (EXP) group) according to milk yield in previous lactation, days in milk (98), number of lactations and BW. The trial started and finished with a period (period 1 and 4) where both groups received the same ration (total-mixed ration based on grass and maize silage, ensiled sugar beet pulp, lucerne hay, mineral premix and concentrate, calculated for 37 kg energy-corrected milk). In between, the ration of EXP cows was supplemented with 1% (CT1, period 2) and 3% of dry matter (DM) (CT3, period 3) of a commercial A. mearnsii extract (containing 0.203 g CT/g DM) which was mixed into the concentrate. In period 3, samples of urine and faeces were collected from 10 cows of each group and analyzed to estimate N excretion. Except for a tendency for a reduced milk urea concentration with CT1, there was no difference between groups in period 2 (CON v. CT1; P>0.05). The CT3 significantly reduced (P<0.05) milk protein yield, the apparent N efficiency (kg milk N/k feed N) and milk urea concentration; but total milk yield and energy-corrected milk yield were not affected by treatment. Furthermore, as estimated from 10 cows per group and using urinary K as a marker to estimate the daily amount of urine voided, CT3 caused a minor shift of N compounds from urine to faeces, as urea-N in urine was reduced, whereas the N concentration in faeces increased. As an improvement in productivity was not achieved and N use efficiency was decreased by adding the CT product it can be concluded that under current circumstances the use in high yielding dairy cows is not advantageous.  相似文献

The dairy farming systems of Western Europe are based on a simple feeding system composed of grazed and preserved grass, maize silage and concentrates in variable proportions. There is, nevertheless, a great diversity of feeding strategies between dairy farms. Over 5 years, we studied the direct and delayed effects of four feeding strategies on the lactation and reproduction performances of Holstein and Normande dairy cows. The four feeding strategies (denoted Hh, Hl, Lh and Ll) correspond to two total mixed rations applied in winter from calving to turnout (maize silage with 30% concentrate or grass silage with 15% concentrate), which were subsequently crossed with two levels of concentrate supplementation at grazing to 210 days. Each year, 72 dairy cows managed in grouped winter calving were assigned to the four strategies. Finally, the results of 325 lactations and 295 inseminated cows were analysed. The four strategies resulted in considerable variation in nutrient intake and, in particular, in differences in concentrates consumed, with values of 1407, 1026, 773 and 392 kg dry matter per cow for strategies Hh, Lh, Hl and Ll, respectively. Total milk production (7567, 7015, 6720 and 6238 kg per cow for treatments Hh, Lh, Hl and Ll, respectively), milk fat content (39.0, 37.1, 40.3 and 38.5 g/kg, respectively), milk protein content (33.0, 31.8, 33.1 and 31.6 g/kg, respectively), and the character of the lactation and body condition curves were all highly sensitive to the strategies applied. While no significant interaction was detected on total lactation yield, the Holstein cows reacted more dramatically to each dietary change at each period, compared with the Normande cows. Winter feeding did not affect the production of milk at pasture whereas, at pasture, the milk from the cows of the H groups in winter was higher in milk fat and protein content. Reproduction performance was unaffected by feeding strategy. The Holstein cows, well fed and producing the most milk (Hh and Hb), had the lowest rate of success at first artificial inseminations (21.5%). The dual-purpose Normande cows had a pregnancy rate 10 points higher than Holstein cows. This comparison of strongly contrasting feeding strategies confirms the immediate reactivity of dairy cows (in terms of milk performance and body condition) to variations of nutritive intake throughout lactation, with a weak carryover effect from feeding levels early in lactation. In contrast, reproduction performance was less sensitive to variation in nutrient supply.  相似文献

On the basis of the potential benefits to human health there is an increased interest in producing milk containing lower-saturated fatty acid (SFA) and higher unsaturated fatty acid (FA) concentrations, including cis-9 18:1 and cis-9, trans-11-conjugated linoleic acid (CLA). Twenty-four multiparous Holstein cows were used in two experiments according to a completely randomized block design, with 21-day periods to examine the effects of incremental replacement of prilled palm fat (PALM) with sunflower oil (SFO) in high-concentrate diets containing 30 g/kg dry matter (DM) of supplemental fat (Experiment 1) or increases in the forage-to-concentrate (F : C) ratio from 39 : 61 to 48 : 52 of diets containing 30 g/kg DM of SFO (Experiment 2) on milk production, digestibility and milk FA composition. Replacing PALM with SFO had no effect on DM intake, but tended to increase organic matter digestibility, yields of milk, protein and lactose, and decreased linearly milk fat content. Substituting SFO for PALM decreased linearly milk fat 8:0 to 16:0 and cis-9 16:1, and increased linearly 18:0, cis-9 18:1, trans-18:1 (Δ4 to 16), 18:2 and CLA concentrations. Increases in the F : C ratio of diets containing SFO had no effect on intake, yields of milk, milk protein or milk lactose, lowered milk protein content in a quadratic manner, and increased linearly NDF digestion and milk fat secretion. Replacing concentrates with forages in diets containing SFO increased milk fat 4:0 to 10:0 concentrations in a linear or quadratic manner, decreased linearly cis-9 16:1, trans-6 to -10 18:1, 18:2n-6, trans-7, cis-9 CLA, trans-9, cis-11 CLA and trans-10, cis-12 CLA, without altering milk fat 14:0 to 16:0, trans-11 18:1, cis-9, trans-11 CLA or 18:3n-3 concentrations. In conclusion, replacing prilled palm fat on with SFO in high-concentrate diets had no adverse effects on intake or milk production, other than decreasing milk fat content, but lowered milk fat medium-chain SFA and increased trans FA and polyunsaturated FA concentrations. Increases in the proportion of forage in diets containing SFO increased milk fat synthesis, elevated short-chain SFA and lowered trans FA concentrations, without altering milk polyunsaturated FA content. Changes in fat yield on high-concentrate diets containing SFO varied between experiments and individual animals, with decreases in milk fat secretion being associated with increases in milk fat trans-10 18:1, trans-10, cis-12 CLA and trans-9, cis-11 CLA concentrations.  相似文献

Using two PGF treatments 14 days apart as a way to enhance estrus detection rate following the 2nd treatment is a reproductive management tool that continues to be used on large dairy farms. In one study, in cows with a functional CL and a dominant follicle, treatment with cloprostenol vs. dinoprost resulted in greater peripheral estradiol concentrations. The objective of the present study was to determine if cloprostenol could enhance pregnancy rates of cows in a large dairy herd using a PGF program for 1st artificial insemination (AI). Lactating dairy cows (n = 4549) were randomly assigned to receive two treatments of either 500 μg cloprostenol or 25 mg dinoprost 14 days apart, with the 2nd treatment on the 1st day of the voluntary waiting period (57 DIM). Cows detected in estrus within 5 days after the 2nd treatment were inseminated. There was no effect of treatment on day of estrus detection, with 78% of cows inseminated on Days 3 or 4 following treatment. Cloprostenol increased (P < 0.01) estrus detection rates in 1st parity cows compared to dinoprost, 42.4 vs. 34.0%. In cows inseminated on Days 3 or 4 after treatment, cloprostenol increased (P = 0.05) conception rates compared to dinoprost, 38.3 vs. 34.4%. When treatments and parities were combined, conception rates increased (P < 0.02) with interval after treatment (27.0, 36.4, and 44.5% for Days 1 or 2, Days 3 or 4, and Day 5, respectively). Cloprostenol increased (P = 0.02) overall pregnancy rate compared to dinoprost, 14.4 vs. 12.2%. In summary, cloprostenol increased fertility in 1st parity cows inseminated on Days 3 or 4 following treatment and subsequently enhanced pregnancy rates of 1st parity lactating dairy cows compared to dinoprost. Fertility appeared greater in cows expected to have had a young antral ovarian follicle at treatment.  相似文献

Cobalamin (CBL), the biologically active form of vitamin B12, and its analogs, are produced by bacteria only if cobalt supply is adequate. The analogs differ generally by the nucleotide moiety of the molecule. In CBL, 5,6-dimethylbenzimidazole (5,6-DMB) is the base in the nucleotide moiety. The present study aimed to determine if a supplement of 5,6-DMB could increase utilization of dietary cobalt for synthesis of CBL and change ruminal fermentation, nutrient digestibility, omasal flow of nutrients and ruminal protozoa counts. Eight ruminally cannulated multiparous Holstein cows (mean±standard deviation=238±21 days in milk and 736±47 kg of BW) were used in a crossover design. Cows were randomly assigned to a daily supplement of a gelatin capsule containing 1.5 g of 5,6-DMB via the rumen cannula or no supplement. Each period lasted 29 days and consisted of 21 days for treatment adaptation and 8 days for data and samples collection. Five corrinoids, CBL and four cobamides were detected in the total mixed ration and the omasal digesta from both treatments. The dietary supplement of 5,6-DMB increased (P=0.02) apparent ruminal synthesis of CBL from 14.6 to 19.6 (s.e.m. 0.8) mg/day but had no effect (P>0.1) on apparent ruminal synthesis of the four analogs. The supplement of 5,6-DMB had no effect (P>0.1) on milk production and composition, or on protozoal count, ruminal pH and concentrations of volatile fatty acids and ammonia nitrogen in rumen content. The supplement had also no effect (P>0.1) on intake, omasal flow and apparent ruminal digestibility of dry matter, organic matter, NDF, ADF and nitrogenous fractions. Plasma concentration of CBL was not affected by treatments (P=0.98). Providing a preformed part of the CBL molecule, that is, 5,6-DMB, increased by 34% the apparent ruminal synthesis of CBL by ruminal bacteria but had no effect on ruminal fermentation or protozoa count and it was not sufficient to increase plasma concentrations of the vitamin. Even though the efficiency of cobalt utilization for apparent synthesis of CBL was increased from 2.0% to 2.7% by the 5,6-DMB supplement, this improved efficiency was still very low. Further research is needed to identify the factors affecting efficiency of utilization of cobalt for synthesis of CBL by the bacterial populations in rumen.  相似文献

Twenty-four lactating Holstein dairy cows (12 first lactation and 12 multiparous; day in milk = 11 ± 5 days) were allotted to a randomised complete block design in a 2 × 3 factorial with four replicates per treatment to evaluate the effects of two methods of alfalfa feeding (dry and reconstituted to achieve a theoretical dry matter (DM) content of 350 g/kg) and three geometric mean (GM) particle sizes of alfalfa (9.13, 4.51 and 1.20 mm) on performance of dairy cows for a period of 28 days. Diets were offered for ad libitum intake as total mixed rations (TMR). The GM particle size, its standard deviation, and the values of physical effectiveness factor of alfalfa and TMR decreased as alfalfa particle size decreased. Reduction of particle size and reconstitution of alfalfa increased the bulk density and the functional specific gravity of alfalfa and rations. Reduction of particle size decreased insoluble dry matter, water-holding capacity, and hydration rate of alfalfa. As particle size decreased, the amount of physically effective NDF in the ration (g/kg) decreased but the daily intake of physically effective NDF (kg/day) increased. Reduction of particle size and reconstitution increased dry matter intake (DMI) and ruminal passage rate, but reduced NDF and ash digestibilities, ruminal pH, N-NH3, milk fat, total chewing activity, rumination and eating time, total and ruminal mean retention time, and time delay of marker. Increased functional specific gravity, from reduced forage particle size and the reconstitution of alfalfa, was the most important factor influencing DMI, milk composition, and chewing activity.  相似文献
