Diseases transmitted by hematophagous (blood-feeding) insects are responsible for millions of human deaths worldwide. In hematophagous insects, the blood meal is important for regulating egg maturation. Although a high concentration of iron is toxic for most organisms, hematophagous insects seem unaffected by the iron load in a blood meal. One means by which hematophagous insects handle this iron load is, perhaps, by the expression of iron-binding proteins, specifically the iron storage protein ferritin. In vertebrates, ferritin is an oligomer composed of two types of subunits called heavy and light chains, and is part of the constitutive antioxidant response. Previously, we found that the insect midgut, a main site of iron load, is also a primary site of ferritin expression and that, in the yellow fever mosquito, Aedes aegypti, the expression of the ferritin heavy-chain homologue (HCH) is induced following blood feeding. We now show that the expression of the Aedes ferritin light-chain homologue (LCH) is also induced with blood-feeding, and that the genes of the LCH and HCH are tightly clustered. mRNA levels for both LCH- and HCH-genes increase with iron, H2O2 and hemin treatment, and the temporal expression of the genes is very similar. These results confirm that ferritin could serve as the cytotoxic protector in mosquitoes against the oxidative challenge of the bloodmeal. Finally, although the Aedes LCH has no iron responsive element (IRE) at its 5'-untranslated region (UTR), the 5'-UTR contains several introns that are alternatively spliced, and this alternative splicing event is different from any ferritin message seen to date.  相似文献

Aedes aegypti feeding on chickens infected with Plasmodium gallinaceum take less blood and lay fewer eggs than those feeding on uninfected hosts. Both activities show an inverse correlation with the degree of parasitemia. Mosquitoes feeding on infected chickens ingest blood in amounts directly proportional to the length of time spent on the hosts, whereas there is no relationship between host contact and blood meal size for mosquitoes feeding on uninfected hosts. Feeding and probing choice experiments demonstrate that infected chickens are less attractive to Aedes aegypti than uninfected chickens.  相似文献

A bioassay of mosquito iridescent virus (MIV) of Aedes taeniorhynchus was developed using cell cultures of Aedes aegypti. The dilution end point technique was based on the occurrence of cytopathic effects which were optimum at 31°C. Peleg's A. aegypti cell line was more sensitive and reliable than Singh's A. aegypti cell line for infectivity titration of the “R” and “T” strains of MIV. The highest tissue culture infectivity dose 50s (TCID50) were elicited by virion:cell ratios of approximately 10. TCID50 titers were significantly reduced by virus neutralization with either homologous or heterologous antiserum to either RMIV or TMIV. The virus propagated in either cell line was not infectious to A. taeniorhynchus larvae, or to the respective cells from which the virus was produced. An electron microscopical study was conducted on the pathology of the mosquito iridescent virus (MIV) of Aedes taeniorhynchus in monolayer cultures of Aedes aegypti cells. The sequence of events in the pathology, from the initiation of attachment through maturation and release, is presented.MIV attaches to cells and is taken up by the process of viropexis (phagocytosis) within 15 min after inoculation. Intact virions are released into the cytoplasm at 30–60 min by disruption of the phagocytic vesicles. Discrete foci of replication (viroplasm) develop in the cytoplasm within 1 day after infection. Progeny virus is assembled in the viroplasm within 2 days after infection and later appears at the cell surface, where it acquires an envelope from the plasma membrane upon budding from the cell. Virus does not accumulate to form aggregates in the cytoplasm; instead, it buds from the cell after assembly.  相似文献

The phenotype of a ouabain-resistant Aedes albopictus cell line has been partially characterized. Treatment of ouabain-sensitive cells with 0.005-1.0 mM ouabain resulted in an 80% reduction in the uptake of 86rubidium (86Rb+), an ion with an affinity for the K+ pump binding site; ouabain-resistant cells showed only a 40% reduction with 1.0 mM ouabain. When ouabain-sensitive cells were incubated in the presence of ouabain (0.1 mM) for one and one-half to three hours, the molar ratio of intracellular Na+/K+ rose from 0.2 to 4.2. In ouabain-resistant cells, a similar treatment had very little effect. Based on [3H] ouabain-binding studies, ouabain-resistant cells were estimated to have 60% fewer binding sites per cell than ouabain-sensitive cells. The spontaneous mutation rate from ouabain sensitivity to ouabain resistance was calculated to be 1-6 x 10(-8) mutations/cell/generation, a value similar to that reported for mammalian cells at the analogous locus.  相似文献

Among six strains of Bacillus thuringiensis and five other species of Bacillus, only two strains of B. thuringiensis, strains HD-1 and BA-068, were toxic to Aedes aegypti larvae within 24 hr. The LC50s were 5.6 × 104 and 2.4 × 105 spores/ml for strains HD-1 and BA-068, respectively. The toxic factor(s) was heat sensitive and γ ray resistant and preliminary evidences indicated that it was associated with the crystalline body of B. thuringiensis.  相似文献

Six essential oils: asteraceae oil, rutaceae oil, mentha piperta oil, carvacryl oil, citronella oil, and eucalyptus oil were tested for evaluation of their repellent effects against Aedes albopictus mosquitoes under laboratory conditions. Only citronella oil and eucalyptus oil were tested with human beings. There was considerable protection for mice. Carvacryl oil (7%) provided 100% protection for mice after 7 h. Eucalyptus oil (15%) gave protection to humans for least 3 h; the protection time was prolonged to 5 h after adding 5% vanillin. The mixture could be developed into a practical product after the field evaluation.  相似文献

Cultured mosquito cells were found to produce Sindbis virus nearly as efficiently as BHK-21 cells at 28 C. In virtually all of the cells observed in the electron microscope, virus morphogenesis was found to occur within complex vesicular structures which developed after viral infection. Viral nucleocapsids were first seen in these vesicles and appeared to be enveloped within these structures. The process of envelopment within these inclusions differed in some respects from the process previously described for the envelopment of nucleocapsids at the plasma membrane of vertebrae cells. Free nucleocapsids were only rarely seen in the cytoplasm of infected mosquito cells, and budding of virus from the cell surface was detected so infrequently that this process of virus production could not account for the amount of virus produced by the infected cells. The vast majority of extracellular virus was produced by the fusion of the virus-containing vesicles with the plasma membrane releasing mature virions and membrane nucleocapsid complexes in various stages of development.  相似文献

Immediate (24 hours exposure) and long-term (until the emergence of the adults) effects of different doses of a primary powder of Bacillus thuringiensis subsp. israelensis (B.t.i.) against first and second instar Aedes aegypti larvae were monitored. The long-term effect was dose-dependent and was materialized by a prolongation of the preimaginal development and continuous cumulative mortality until the emergence of the adults. Mortality values were higher during the fourth larval instar and the pupal stage. Some of the larvae reaching the fourth instar were smaller in size and remained in this state a 2-4 times longer period than in the control and finally died as larvae or very small pupae. The long-term effect was more intense as the treatment was applied earlier during the larval development. The correlation of the immediate lethal effect with the late effect allows the evaluation of the total impact of a larvicidal treatment.  相似文献

We describe the use of DNA reassociation kinetics to determine the total genome size and complexity together with the individual complexity and copy number of the single copy, middle repetitive and highly repeated DNA fractions of cell line and larval DNA from the mosquito, Aedes aegypti. The genome of Ae. aegypti is both large and complex, being one third the size of the human genome, and exhibits a short period interspersed repeat pattern. The implications of patterns of sequence arrangement and genome complexities for experiments aimed at isolating specific classes of DNA sequences, such as mobile genetic elements, are discussed.  相似文献

Palp-antenna is a homeotic mutant of Aedes aegypti that modifies the apex of the maxillary palps of both sexes into a variable number of antenna-like segments. The fine structure of the antenna, including sexual dimorphism, is apparent in the mutant palps. Palp-antenna is a sex-linked recessive. a linkage distance of 27.9 +/- 0.45 crossover units from sex is estimated from F2 and backcross data. Penetrance is complete; expressively is variable. The fitness of the mutant compares favorably with that of the wild type. The mapping of the ppa locus lengthens the known chromosome 1 linkage map of Aedes aegypti.  相似文献

The mosquito, Aedes aegypti, is the primary, worldwide arthropod vector for the yellow fever and dengue viruses. As it is also one of the most tractable mosquito species for laboratory studies, it has been and remains one of the most intensively studied arthropod species. This has resulted in the development of detailed genetic and physical maps for Ae. aegypti and considerable insight into its genome organization. The research community is well-advanced in developing important molecular tools that will facilitate a whole genome sequencing effort. This includes generation of BAC clone end sequences, physical mapping of selected BAC clones and generation of EST sequences. Whole genome sequence information for Ae. aegypti will provide important insight into mosquito chromosome evolution and allow for the identification of genes and gene function. These functions may be common to all mosquitoes or perhaps unique to individual species, possibly specific to host-seeking and blood-feeding behaviors, as well as the innate immune response to pathogens encountered during blood-feeding. This information will be invaluable to the global effort to develop novel strategies for preventing arthropod-borne disease transmission.  相似文献
