 As an extension of our earlier discoveries that ZnII-cyclen complex (1) (cyclen=1,4,7,10-tetraazacyclododecane) and ZnII-acridine-pendant cyclen complex ZnII-N-(9-acridin)ylmethyl-cyclen (3) are the first compounds to selectively recognize thymidine and uridine nucleosides in aqueous solution at physiological pH, the interaction of these and a relevant complex, bis(ZnII-cyclen) (7), has been investigated with a series of polynucleotides, single-stranded poly(U) and poly(G), and double-stranded poly(A)·poly(U), poly(dA)·poly(dT) and poly(dG)·poly(dC). These ZnII-cyclen complexes interact with the imide-containing nucleobases in the single-stranded poly(U), unperturbed by the presence of the anionic phosphodiester backbone. The affinity constant of 1 for each N(3)-deprotonated uracil base in poly(U) is determined to be log K= 5.1 by a kinetic measurement, which is almost the same as log K=5.2 for the interaction of 1 with uridine. Thus, they disrupt the A-U (or A-T) hydrogen bonds to unzip the duplex of poly(A)·poly(U) or poly(dA)·poly(dT), as demonstrated by lowering of the melting temperatures (T m) of poly(A)·poly(U) and poly(dA)·poly(dT) in 5 mM Tris-HCl buffer (pH 7.6, 10 mM NaCl) with increase in their concentrations. The order of the denaturing efficiency is well correlated with that of the 1 : 1 affinity constants for each complex with uracil or thymine;7>3>1. The comparison of circular dichroism (CD) spectra for poly(A)·poly(U), poly(A), and poly(U) in the presence of 3 has revealed a structural change from poly(A)·poly(U) to two single strands, poly(A) and poly(U), caused by 3 binding exclusively to uracils in poly(U). On the other hand, the acridine-pendant cyclen complex 3, which earlier was found to associate with guanine by the ZnII coordinating with guanine N(7), in addition to the π-π stacking, interacts with guanine in the double helix of poly(dG)·poly(dC) from outside and stabilized the double-stranded structure, as indicated by higher T m. 【目的】筛选高效降解木质素的菌株,并研究其以玉米秸秆为底物时木素降解酶活性。【方法】本研究以愈创木酚培养基和苯胺蓝培养基从吉林省不同经纬度的自然朽木及腐朽玉米秸秆土壤样品中分离、筛选得到高效降解木质素的菌株,并对其形态学鉴定,通过ITS序列分析构建系统发育树,分析菌株的分类地位。通过秸秆固体发酵过程产生的胞外木质素酶的活性分析,选出高效秸秆降解菌。【结果】筛选出1株高效降解秸秆的真菌,对其进行形态学特征和ITS序列分析,命名为白囊耙齿菌W2(Irpex lacteus W2)。该菌株在4–8 d内产生的锰过氧化物酶(Manganese peroxidase)呈上升趋势,并且在8 d达到峰值86.31 U/mL,与黄孢原毛平革菌(Phanerochaete chrysosporium)的最高酶活力45.86 U/mL相比,高出了88.20%(P0.01);该菌株的漆酶(Laccase)活力8 d时达到20.60 U/mL,比对照高40.76%(P0.05)。【结论】本研究分离到一株具有较强降解秸秆能力的真菌,初步鉴定为Irpex lacteus W2,具有较强的降解秸秆能力,其降解秸秆过程中产生较高的锰过氧化物酶与漆酶活力。  相似文献

Abstract

Confusion has existed over the validity of the taxon Urochloa multiculma (Andersson) Morrone & Zuloaga, which was originally described from Galapagos, and over the presence in the islands of the closely related U. fusca (Sw.) B.F. Hansen & Wunderlin. An examination of 14 characters in 182 specimens leads to the conclusions that U. multiculma is a valid taxon endemic to Galapagos, and that U. fusca is probably a recent human‐mediated introduction to the archipelago, which may now be hybridising with multiculma. Leaf pubescence and spikelet structure are used to construct an identification key.  相似文献

【背景】城市垃圾渗滤液是一种成分复杂的有机废水,含氮量高,如果未经处理直接排放到环境中会造成严重的环境污染。【目的】筛选可以耐受垃圾渗滤液中高浓度氨氮并高效去除污水中氮素的异养硝化好氧反硝化菌株,为解决垃圾渗滤液的氮素污染提供功能菌株。【方法】从垃圾渗滤液中筛选分离能耐受高氨氮浓度的菌株,通过测定各菌株的脱氮能力,筛选到一株脱氮能力最强的菌株,命名为U1,通过测定16S rRNA基因序列和生理生化特性确定该菌株为铜绿假单胞菌。进一步研究了菌株U1在不同初始氨氮浓度、碳源、转速、初始pH、碳氮比等单因素变量下的脱氮能力,并结合L9(34)正交试验研究了菌株U1的最佳脱氮条件。【结果】分离出一株铜绿假单胞菌并命名为U1。该菌株的最优脱氮条件为:初始氨氮浓度为1 000 mg/L,红糖和柠檬酸三钠的混合碳源,pH 6.0,C/N为10,转速为130 r/min,菌株U1的最大总氮去除率为64.37%,最大氨氮去除率为76.73%。对于总氮和氨氮含量分别是2 345 mg/L和1 473.8 mg/L的垃圾渗滤液,菌株U1最大总氮去除率为27.86%...  相似文献

Aim. Pulmonary complications are known to occur in over half the patients exposed to sulphur mustard. Many studies have focused on the clinical complications, often ignoring the pathogenesis of sulphur mustard. Also, the reasons for the variable severity of lung injuries caused by sulphur mustard are unclear. Hence, the current study was performed to evaluate the correlation between superoxide dismutase (SOD) and catalase (CAT) activity and pulmonary function in patients exposed to sulphur mustard. Methods. Our study was a comparative cross-sectional survey. Two hundred and fifty incident survivors were selected from the Sardasht population who were exposed to sulphur mustard in 1987. A control group from non-exposed civilians was also selected. We used a pulmonary function test, and SOD and CAT activity was measured in these groups. Results. The mean SOD activity in the healthy control group (70.5±10.8 U ml?1) was higher than in the moderate-to-severe group (67.0±6.1 U ml?1) (p <0.001, one-tail ANOVA, least significant difference (LSD) post hoc). The mean activity in the mild group (72.5±6.9 U ml?1) was no higher than in the healthy control group (70.5±10.8 U ml?1) (p=0.095 one-tail ANOVA, LSD post hoc). The mean CAT activity in the healthy control group (4.9±1.5 U ml?1) was lower than in the moderate-to-severe group (8.0±1.8 U ml?1) (p <0.001, one-tail ANOVA, LSD post hoc) and in the mild group (7.5±1.5 U ml?1) (p=0.012 one-tail ANOVA, LSD post hoc). Conclusion. According to our findings, it is reasonable to hypothesize that re-establishment of the activation–inactivation or oxidant–antioxidant balance in favour of the activation and antioxidant balances would be useful as a therapeutic strategy to suppress pathological mechanisms underlying lung injuries.  相似文献

Abstract

The main features to distinguish the seven native Utricularia species occurring in central Europe are found in flower shape, but being rarely flowering identification is often doubtful and uncertain. A recent morphometric work highlighted that there are no univocal reliable extra-floral morphological features allowing a safe identification at species level. Therefore, DNA barcoding approach is attempted here. Molecular analyses were performed to search for DNA barcodes using nuclear ITS (rDNA), plastid (cpDNA) trnL-trnF IGS and rps16 intron sequences. Generally, the barcoding approach failed to discriminate Utricularia species, although it could be of some help in the U. minor aggregate. With few exceptions, U. bremii shows peculiar DNA regions different from U. minor for both plastid markers investigated. However, interesting hypotheses could be derived from the obtained networks, including hybridization events to explain the rise of mostly sterile species, such as U. stygia. This species clusters with the other species of the U. intermedia aggregate in plastid phylogenetic graphs, while it is closely related to species of the U. minor aggregate in ITS phylogenetic graphs. Additionally to U. stygia, U. ochroleuca also shows some incongruences in the different markers, at least for some accessions, pointing to the possible occurrence of hybrids.  相似文献

ABSTRACT

Deninger’s bears (Ursus deningeri) have been studied less frequently than Ursus spelaeus s.l. Our objective is to present, for the first time, an analysis of the skull shape of U. deningeri.

Bear crania and mandibles were digitised with a Microscribe or CT-scanned and the surface models subsequently landmarked. The landmarks were chosen based on a compromise between functional morphology and sample size.

Results show that U. deningeri and U. spelaeus mandibles display very similar morphologies and allometric trajectories, both to each other and to Ailuropoda melanoleuca. It is inferred that masticatory adaptations to a herbivorous diet were already present in the Middle Pleistocene. U. deningeri displays a cranial morphology that is similar to that of U. spelaeus when comparing all species, but U. deningeri has a relatively narrower and dorsoventrally lower zygomatic arch than U. spelaeus, although the masticatory signal is less strong in the skull.

We observe intraspecific differences between different populations of U. deningeri, which could parallel the genetic diversity found in U. spelaeus. The intraspecific differences found within U. deningeri may be temporal and/or geographical in nature and could be related to the evolution of the Late Pleistocene cave bear, but this hypothesis remains to be tested.  相似文献

Abstract

In this study, the effects of Aspergillus niger in coculture with the basidiomycetes, Trametes versicolor, T. maxima, and Ganoderma spp., were studied to assess H2O2 production and laccase (Lac), Lignin Peroxidase (LiP), and manganese peroxidase (MnP) activities. The results indicated that maximum discoloration was of 97%, in the T. maxima and A. niger coculture, where the concentration of H2O2 was 5?mg/L and 6.3?mg/L in cultures without and with dye, respectively. These concentrations of H2O2 were 1.6- and 1.8-fold higher than monocultures of T. maxima (3.37?mg/L) and A. niger (3.87?mg/L), respectively. In the same coculture, the LiP and MnP enzyme activities also increased 12-fold, (from 0.08?U/mg to 0.99?U/mg), and 67-fold, (from 0.11?U/mg to 7.4?U/mg), respectively. The Lac activity increased 1.7-fold (from 13.46?U/mg to 24?U/mg). Further, a Box–Behnken experimental design indicated a 1.8-fold increase of MnP activity (from 7.4?U/mg to 13.3?U/mg). In addition, dye discoloration regression model obtained from the Box–Behnken experimental design showed a positively correlation with H2O2, (R2?=?0.58) and a negatively correlation with Lac activity (R2 = –0.7).  相似文献

Uncaria rhynchophylla (Gouteng) is a famous traditional Chinese medicine used for psychiatric and hypotensive purposes in China. In this study, the ethyl acetate (EtOAc) part of U. rhynchophylla was revealed with protein tyrosine phosphatase 1B (PTP1B) inhibitory activity. Subsequent investigation on the EtOAc part yielded one new triterpenoid, 3β-formyloxy-6β,19α-dihydroxyurs-12-en-28-oic acid (1) and four known ones, 3β,6β,19α-trihydroxyurs-12-en-28-oic acid (2), 2-oxopomolic acid (3), 3β,19α-dihydroxy-6-oxo-olean-12-en-28-oic acid (4) and sumaresinolic acid (5). The structure of compound 1 was determined by extensive HRESIMS, IR, 1D and 2D NMR spectroscopic analyses. Two ursane-type triterpenoids (2 and 3) showed selective inhibition on PTP1B with IC50 values of 48.2 and 178.7 μM. The enzyme kinetic study suggested that compounds 2 and 3 were mix-type inhibitors on PTP1B with Ki values of 15.6 and 132.5 μM. This investigation manifests the antidiabetic potency of U. rhynchophylla with triterpenoids as the active constituents.  相似文献

ABSTRACT

The morphology of both crowns and tooth-roots reflects dietary specialisation in mammalian carnivores. In this article, we analyse the tooth-root morphology of maxillary teeth from CT scans of living bears (Ursus arctos, Ursus americanus, Ursus maritimus, Ursus thibetanus, Melursus ursinus, Helarctos malayanus, Tremarctos ornatus and Ailuropoda melanoleuca) in order to make inferences about the diet and feeding behaviour of the extinct cave bear (Ursus spelaeus sensu lato). Specifically, we investigate two major mitochondrial clades of extinct cave bears recognized by previous authors: Ursus ingressus and Ursus spelaeus (U. spelaeus spelaeus, U. spelaeus ladinicus, U. spelaeus eremus). Our results indicate a close association between tooth-root surface area and feeding behaviour in all living bear species. Tooth-root surface area values of cave bears suggest that they relied more on vegetative matter than living brown bears (Ursus arctos) but subtle differences between these species/subspecies could also indicate different feeding strategies among the members of cave bear complex.  相似文献

Umbelopsis gibberispora is described as a new species in the genus Umbelopsis, Umbelopsidaceae, Mucorales. The species differs from others in this genus by ellipsoidal sporangiospores with unilaterally thickened walls. Phylogenetic analyses based on nuclear large subunit ribosomal DNA (nLSU rDNA) partial sequences suggest that U. gibberispora, U. swartii, and U. westeae form a clade together with the strains of Umbelopsis ramanniana. The ex-type strain of Micromucor ramannianus var. angulisporus is found to be very close to Umbelopsis vinacea, whereas other isolates identified under the former name in the sense of Linnemann fall in the U. ramanniana subclade. For these isolates, a new species, Umbelopsis angularis, is introduced. Phylogenetic relationships among Umbelopsis species are discussed related to their attributes of the sporangial wall and mature spore shapes. Fungal specimens of discomycetes in the family Hyaloscyphaceae were collected from across Japan and identified. In this report, glassy members of the tribe Hyaloscypheae, subfamily Hyaloscyphoideae are described and illustrated. The 7 species involved include two new species (Urceolella brunneola andU. pseudopani), and five species new to Japan (Hyalopeziza millepunctata, H. pygmaea, Mollisina uncinata, Urceolella carestiana andU. crispula). Background: The research of G protein-coupled receptors (GPCRs) is a promising strategy for drug discovery. In cancer therapy, there is a need to discover novel agents that can inhibit proliferation and induce apoptosis in cancer cells. JTC-801 is a novel GPCR antagonist with the function of reversing pain and anxiety symptoms. This study aims to investigate the antitumor effects of JTC-801 on human osteosarcoma cells (U2OS) and elucidate the underlying mechanism.

Materials and methods: The Cell Counting Kit-8 assay was used to detect the viability of U2OS cells treated with JTC-801 in vitro. The cell apoptosis was evaluated using a flow cytometry assay with Annexin V-FITC/PI double staining. The inhibitory effect of JTC-801 on invasion and migration of U2OS cells were determined by the Transwell assays. Western blot assay was performed to measure the levels of proteins related to cell apoptosis and its mechanism.

Results: The JTC-801 significantly decreased the viability of U2OS cells (p?p?p?Conclusions: JTC-801 may exert osteosarcoma cell growth inhibition by promoting cell apoptosis, through PI3K/AKT signaling pathway participation.  相似文献

Objective

To solve the bottleneck of plasmid instability during microbial fermentation of l-DOPA with recombinant Escherichia coli expressing heterologous tyrosine phenol lyase.

Results

The tyrosine phenol lyase from Fusobacterium nucleatum was constitutively expressed in E. coli and a fed-batch fermentation process with temperature down-shift cultivation was performed. Efficient strategies including replacing the original ampicillin resistance gene, as well as inserting cer site that is active for resolving plasmid multimers were applied. As a result, the plasmid stability was increased. The co-use of cer site on plasmid and kanamycin in culture medium resulted in proportion of plasmid containing cells maintained at 100% after fermentation for 35 h. The specific activity of tyrosine phenol lyase reached 1493 U/g dcw, while the volumetric activity increased from 2943 to 14,408 U/L for l-DOPA biosynthesis.

Conclusions

The established strategies for plasmid stability is not only promoted the applicability of the recombinant cells for l-DOPA production, but also provides important guidance for industrial fermentation with improved microbial productivity.

Abstract

The induction of leukemic cell differentiation is a hopeful therapeutic modality. We studied the effects of monochloramine (NH2Cl) on erythroleukemic K562 cell differentiation, and compared the effects observed with those of U0126 and staurosporine, which are known inducers of erythroid and megakaryocytic differentiation, respectively. CD235 (glycophorin) expression, a marker of erythroid differentiation, was significantly increased by NH2Cl and U0126, along with an increase in cd235 mRNA levels. Other erythroid markers such as γ-globin and CD71 (transferrin receptor) were also increased by NH2Cl and U0126. In contrast, CD61 (integrin β3) and CD42b (GP1bα) expression, markers of megakaryocytic differentiation, was increased by staurosporine, but did not change significantly by NH2Cl and U0126. NH2Cl retarded cell proliferation without a marked loss of viability. When ERK phosphorylation (T202/Y204) and CD235 expression were compared using various chemicals, a strong negative correlation was observed (r = ?0.76). Paradoxically, NH2Cl and staurosporine, but not U0126, induced large cells with multiple or lobulated nuclei, which was characteristic to megakaryocytes. NH2Cl increased the mRNA levels of gata1 and scl, decreased that of gata2, and did not change those of pu.1 and klf1. The changes observed in mRNA expression were different from those of U0126 or staurosporine. These results suggest that NH2Cl induces the bidirectional differentiation of K562. Oxidative stress may be effective in inducing leukemic cell differentiation.  相似文献

Abstract

Aim: The main purpose of this work was to develop new protocols for high yield purification of secretory phospholipase A2 (PLA2) and to investigate its biophysical properties.

Materials and methods: We have used a Pichia pastoris expression system for PLA2 expression and two-stage chromatography for its purification. The biophysical properties of PLA2 were investigated by circular dichroism.

Results: A scalable method for high yield purification of recombinant Streptomyces violaceruber PLA2 was developed. The PLA2 from S. violaceruber was expressed in the methylotrophic yeast P. pastoris. Functional active phospholipase A2 with specific activity 73?U/mg was purified with a concentration of at least 3?mg/mL. The role of different divalent ions in PLA2 thermostability were evaluated. Ca2+ and Ba2+ ions significantly increased thermostability of the enzyme.  相似文献

【背景】醇脱氢酶AdhS能催化不对称还原反应制备(R)-2-氯-1-苯乙醇,但由于自身再生辅酶NADH的能力不足,需要辅酶再生酶协助其再生NADH。谷氨酸脱氢酶能以谷氨酸为底物,再生辅酶NAD(P)H,具有辅酶再生酶的潜力。【目的】克隆表达谷氨酸脱氢酶基因gdhA,构建谷氨酸脱氢酶GdhA与醇脱氢酶AdhS的大肠杆菌共表达体系,提高AdhS制备(R)-2-氯-1-苯乙醇的转化效率。【方法】从枯草芽孢杆菌(Bacillus subtilis) 168中克隆基因gdhA,并在大肠杆菌(Escherichia coli) BL21(DE3)中表达,分析辅酶再生活力;再与醇脱氢酶AdhS共表达,优化表达条件;分析不同辅酶再生方案对制备(R)-2-氯-1-苯乙醇的转化效率的影响。【结果】谷氨酸脱氢酶GdhA再生NADH的比活力为694 U/g。经GdhA与AdhS的共表达及表达条件优化后,制备(R)-2-氯-1-苯乙醇的转化效率达465 U/L。经比较,GdhA协助再生辅酶NADH,可使AdhS制备(R)-2-氯-1-苯乙醇的转化效率提高到约3倍。【结论】谷氨酸脱氢酶GdhA为NADH高效再生酶,与醇脱氢酶AdhS共表达可显著提高AdhS制备(R)-2-氯-1-苯乙醇的转化效率。  相似文献

Abstract

Enzymatic activity of five lysosomal hydrolases: acid p‐nitrophenyl phosphatase (EC 3.1.3.2), acid β‐glycerophosphatase (EC 3.1.3.2), arylsulphatase (EC 3.1.6.1), β‐galactosidase (EC 3.2.1.23) and β‐N‐acetylhexosaminidase (EC 3.2.1.30) was studied in the supernatants of homogenates of hearts of unirradiated mice, serving as controls, and a group of U.V.‐irradiated mice.

In the control group, determinations made at 6‐hr intervals showed rhythmic diurnal changes in activities of three acid hydrolases. These changes were statistically significant in the case of acid p‐nitrophenyl phosphatase, acid β‐glycerophosphatase, and β‐N‐acetylhexosaminidase. The effect of U.V.‐irradiation was manifested mainly by depression of enzyme activities of the acid hydrolases during the first few hours after exposure. Depression of activities of arylsulphatase and β‐N‐acetylhexosaminidase by U.V. was statistically significant. Presumably, the fall in enzyme activities of the acid hydrolases was due to chemical mediators formed in the skin under the influence of U.V.‐irradiation and adrenal corticoids secreted into the blood.  相似文献
