Abstract

Evidence is presented showing that the chromosomes of diploid and triploid races of Atrichum undulatum are significantly shorter than those of haploid plants. Relative DNA contents of the three cytotypes have been estimated and they differ significantly from an expected 1:2:3 ratio in haploid, diploid and triploid races.  相似文献

Bryophytes, the shade loving plants, have tremendous medicinal properties. The aqueous and alcoholic extracts of Atrichum undulatum (Hedw.) P. Beauv. were analysed for antimicrobial properties against the fungi Aspergillus fumigatus and Fusarium oxysporum and the bacteria Escherichia coli, Bacillus mycoides, Proteus mirabilis, Staphylococcus aureus and Salmonella typhi. The study is an attempt to investigate the medicinal properties of Atrichum undulatum (Hedw.) P. Beauv. using disc-diffusion method. No inhibition was observed against A. fumigatus and P. mirabilis. For bacteria S. typhi and E. coli (20 and 15 mm), aqueous and alcoholic extracts of Atrichum showed significant inhibition. However, alcoholic extract was found remarkably effective against bacteria rather than aqueous extract.  相似文献

A system for plant regeneration from protoplasts of the moss, Atrichum undulatum (Hedw.) P. Beauv. in vitro, is first reported. Viable protoplasts were isolated at about 9 × 105 protoplasts g−1 fresh weight from 10 to 18 days protonemata. For regeneration of protoplasts, viable protoplasts were cultured in liquid–solid medium containing surface liquid medium MS (0.4 M mannitol) and subnatant solid medium Benecke (0.3 M mannitol) at 20 °C under a 16-h photoperiod white light after 12 h preculture in darkness at 20 °C. The great majority of protoplasts follow a regenerative sequence: formation of asymmetric cells in 2–3 days; division of the asymmetric cells to 2–3 cells in 4–5 days, and further develop to produce a new chloronemal filament in 15 days. Juvenile gametophyte can be visible in 20 days. The plating ratio of cell cluster regenerated from protoplasts reaches up to 45%. Transient expression experiments indicate the electroporation uptake of DNA is possible.  相似文献

The extraction and partial purification of phytochrome from light-grownAtrichum undulatum P. Beauv., a chlorophyllous moss, is described. Polyethyleneimine and salt fractionation followed by hydroxyapatite and Affi-gel-blue chromatography were used to separate phytochrome from chlorophyll, and to purify the pigment. All steps were performed in the presence of Triton X-100 which improved the yield by a factor of about three. The protein has a molecular weight some-what larger than that ofAvena phytochrome (124 kDa), as determined by sodium dodecyl sulfate-polyacrylamide gel electrophoresis and immunoblot analysis. 通过对采自河北雾灵山海拔1500m的仙鹤藓(Atrichum undulatum)的孢子萌发以及原丝体发育的观察,发现仙鹤藓孢子无休眠现象,孢子接种3天左右萌发:其原丝体发育分为绿丝体和轴丝体两个阶段。扩大培养实验结果表明。仙鹤藓茎叶体在添加2%葡萄糖的MS培养基上,置于25℃/20℃、14小时光照/10小时黑暗、36μmol·m^-2·s^-1条件下培养.产生新生茎叶体最多,且茎叶体长势最好,可以获得大量无菌材料。仙鹤藓愈伤组织诱导实验显示,形成愈伤组织的最佳培养基为添加2%葡萄糖和1.0mg·L^-16-BA的MS培养基。  相似文献

Rates of leaf initiation, emergence, and growth have been measuredduring the period between the production of one and of ten matureleaves on the primary shoot of Agropyron repens. There is aprogressive decline in the growth rate of successively formedleaves so that at the time of formation of the next leaf primordiumeach primordium is smaller than its predecessor at a comparablestage of development. There is also a trend towards a diauxicpattern of growth with a lag phase apparently coinciding withthe transition from apical to intercalary growth of the youngleaf. Up to the six-leaf stage the rate of leaf formation exceedsthe rate of emergence and leaf primordia accumulate on the shootapex. Thereafter the rate of emergence exceeds the rate of formation.These changes in rates of leaf formation and growth are interpretedin terms of competition for assimilates between expanding leavesand leaf primordia, and between the primary and axillary shootapices.  相似文献

Summary The genetic diversity of a world collection of foxtail millet strains (Setaria italica) and some samples of wild populations (Setaria viridis) was studied by means of electrophoresis on five enzymes (10 loci) Est, Acph, Got, Mdh, Pgd. In spite of an overall limited polymorphism, the diversity appeared to be clearly regionalized. The wild populations collected in France and China introduced new genetic variability to the cultivated forms. However, the interregional diversity within both species was greater than the between species (S. viridis/S. italica) diversity.  相似文献

A green cell suspension culture was established from callus tissue obtained from a moss,Atrichum undulatum. This suspension culture was subcultured every ten days in Murashige and Skoog's liquid medium (MS) containing 10?6 M 2, 4-D and 4% glucose. The suspended cells grew vigorously with the passage of subculture, maintaining small clusters made up of a few cells. In this suspension culture, round spore-like cells released from the cell clusters were found. The yield of protoplasts from the suspension culture was significantly higher than that of the conventional method using moss protonemata developed directly from spores.  相似文献

Chlorophyll-fluorescence parameters of three mosses, Rhytidiadelphusloreus (Hedw.) Warnst.,Anomodon viticulosus (Hedw.) Hook &Tayl. and Grimmia pulvinata (Hedw.) Summary A study on a series of genetic markers was run on five hybrids of foxtail millet, Setaria italica, and on one interspecific hybrid S. viridisxS. italica (S. viridis is the wild relative of S. italica). Seven enzymatic systems were investigated using starch gel electrophoresis (esterase, alcohol dehydrogenase, glutamate oxaloacetate transaminase, acid phosphatase, malate dehydrogenase, 6-phosphogluconate dehydrogenase, cathodic peroxidase). This genetic analysis of the 6 F2 has allowed us to define 12 polymorphic loci: Est-1, -2 and -3, Adh-1, Got-1 and -2, Acph-1, Mdh-1 and -2, Pgd-1 and -2, and Pox-1. All of them behaved like dimers, except Est-1 and Est-2 which showed monomeric structures. Two other markers were examined: waxy endosperm, which appeared to be controlled by one locus, and anthocyanic pigmentation of the collar, for which at least two loci are responsible. Studies of linkage carried out on three F2 showed two linkage groups: Mdh-1, Pox-1, Wx, Est-3, and a locus for collar colour, and Est-2, and one or two other loci of colouring.  相似文献

Histochemically, thesewalls have cellulose, pectins, proteins, phospholipids, andRNA as components. Ultrastructurally, the walls contain fragmentsof the plasmalemma and what appear to be aggregations of endoplasmicreticulum and vesicles. Background and Aims

Evolutionary transitions between separate and combined sexes have frequently occurred across various plant lineages. In mosses, which are haploid-dominant, evolutionary transitions from separate to combined sexes are often associated with genome doubling. Polyploidy and hermaphroditism have strong effects on the inbreeding depression of a population, and are subsequently predicted to affect the mating system.

Methods

We tested the association between ploidy (haploid, diploid or triploid gametophytes) and mating system in 21 populations of Atrichum undulatum sensu lato, where sex ratios vary widely. For each population, we measured the sex ratio, estimated selfing rates using allozyme markers and determined the level of ploidy through flow cytometry.

Key Results

Hermaphrodites in A. undulatum were either diploid or triploid. However, many diploid populations were strictly separate-sexed, suggesting that hermaphroditism is not a necessary result of genome doubling. Levels of selfing were strongly supported as being greater than zero in one population with strictly separate-sexed individuals, and one-third of populations with hermaphrodites.

Conclusions

Although hermaphrodites are associated with triploidy, hermaphroditism is not a necessary outcome of genome duplication. Hermaphroditism, but not genome duplication alone, increased estimated selfing rates, probably due to the occurrence of selfing within a gametophyte. Thus, genome duplication can influence the mating system and the associated evolution and maintenance of reproductive traits.  相似文献
