实验在50例麻醉、麻痹和切断双侧迷走神经的家兔上进行。用双极银丝电极引导膈神经放电,经放大、幅度积分而后记录在 X-Y 记录仪上。脊髓蛛网膜下腔注射乙酰胆碱(ACh)50μg 后,膈神经吸气放电幅度增加,平均增加30.6±11.6%.ACh 的这一效应可被阿托品阻断,但不能被六甲双铵、酚妥拉明和心得安阻断。但单独注射上述任何一种受体阻断剂均不能改变膈神经的放电活动。上述结果提示,脊髓蛛网膜下腔注射 ACh 可兴奋膈运动神经元,而且 ACh 的这一兴奋效应是由 Μ 受体中介的,但在正常生理情况下,胆碱能递质系统对膈神经元似无紧张性兴奋作用。因此,ACh 递质可能在脊髓水平对高级中枢下传的呼吸驱动信息的整合中起兴奋性调制作用。  相似文献

Abstract— In the lobster nerve the fixation of CO, at various levels of pCO2 was studied by the incorporation of [l-14C]pyruvate. Incorporation of 14C was solely dependent on CO2 fixation since the C-1 was decarboxylated in the formation of acetyl-CoA. Paired-nerve studies with [2-14C]pyruvate afforded a study of pyruvate metabolism in the lobster nerve. [I14C]Pyruvate was incorporated to nearly the same extent at all levels of pCO2 including zero pCO2, a finding that suggested metabolic recycling of CO2. The magnitude of the metabolic recycling of C-1 of pyruvate or pyruvate dismutation was estimated to be nearly 20 per cent of total CO2 fixation. Re-evaluation of the relative contributions of the CO2 fixation. and acetyl-CoA pathways on the basis of more extensive data gave a ratio of 2:3.
The pCO2 affected synthesis of ACh and the level of citrate. With increasing pCO2, the specific radioactivity of ACh decreased much more than the content of ACh. The decrease in the specific radioactivity of ACh but not that of citrate further suggested metabolic compartmentation. The implication of these findings is discussed.
Alanine functioned as a metabolic sink for the incorporated pyruvate. Pyruvate levels were estimated to be approximately 0.1 nmol/mg of protein.  相似文献

Abstract— Acetylcholine is synthesized and stored in the nerve endings from which the liberation of the nerve transmittor is regulated by the nerve activity. The aim of the present investigation was to measure the in vivo turnover of acetylcholine in this subcellular acetylcholine pool. This has been carried out by injecting labelled choline intravenously and then by measuring at different time intervals the ratio between labelled choline and acetylcholine in the fractions obtained after subcellular fractionation. It was found that the ratio radioactive choline to radioactive acetylcholine was the same (2:1) in whole brain and in the nerve ending fraction 2 to 20 min after injection. Since it was assumed that the same ratio is true also for the endogenous compounds the choline pool in the nerve terminals was considered to make up 13 nmoles/g brain. The results also indicate that plasma choline is rapidly equilibrated with the nerve terminals and transformed to acetylcholine at a rate of about 5 nmoles/g brain/min.  相似文献

Abstract— Acetylcholine, its precursor (choline), and the enzymes of its biosynthesis and degradation (choline acetyltransferase and acetylcholinesterase, respectively) have been studied and quantified in extracts of several regions of the nervous system of the lobster and in single, isolated axons of identified efferent excitatory, efferent inhibitory and afferent sensory neurons. The choline acetyltransferase is a soluble enzyme similar to that from other species. The predominant acetylcholine-hydrolysing enzyme is largely membrane-bound and has been characterized as a specific acetylcholinesterase. A single peak of acetylcholinesterase activity can be detected upon velocity sedimentation analysis of Triton X-100-treated extracts of all regions of the nervous system. Choline acetyltransferase distribution parallels that of sensory neural elements, and its specific activity shows nearly a 500-fold difference from the richest to the poorest neural source. Acetylcholinesterase levels span only a 23-fold range, and activity is found in all neural regions, including those free of known sensory components. A radiochemical microassay for choline and acetylcholine in the range of 20–2000 pmol is described in detail. All 3 types of axons contain comparable levels of choline ( ca. 2 pmol/μg protein), but acetylcholine is asymmetrically distributed. Efferent axons contain no detectable acetylcholine, while sensory axons from abdominal muscle receptor organs have an average of 1·9 pmol/μg protein. Choline acetyltransferase is similarly distributed; sensory axons show at least 500-fold greater activity than efferent axons. Acetylcholinesterase is nearly uniformly distributed among the three types of fibres. These results are discussed in terms of a general view of transmitter accumulation in single neurons.  相似文献

目的:观察不同浓度乙酰胆碱(ACh,10-10~10-5mol/L)对大鼠体外抗体生成的影响,并初步探讨其作用机制,从细胞水平了解乙酰胆碱与免疫功能之间的关系。方法:用体外抗体生成的检测方法,用绵羊红细胞(SRBC)刺激大鼠肠系膜淋巴结B细胞转化成抗体形成细胞(AFC),然后检测其抗体生成量。结果:①10-10~10-7mol/LACh能显著抑制体外抗体生成,其中10-8和10-7mol/LACh的作用较强,而10-6和10-5mol/LACh无明显的抑制作用;②M型胆碱能受体激动剂毛果芸香碱(10-8和10-7mol/L)能明显减弱体外抗体生成,而N型受体激动剂烟碱(10-8和10-7mol/L)没有显著的减弱作用,M型受体拮抗剂阿托品(10-7和10-6mol/L)可完全阻断ACh抑制体外抗体生成的作用;③ACh分别在B细胞用SRBC刺激后3~48h中的6个不同时间与淋巴细胞作用,其抗体生成仍然是减少的。结论:ACh可非浓度依赖性地抑制大鼠的体外抗体生成;此作用可能由B细胞上的M型胆碱能受体介导;且ACh可能主要影响B细胞转化的后期过程。  相似文献

The effect of oxotremorine (1 mg kg-1 i.p.) on the steady state concentration of acetylcholine (ACh) and choline (Ch) and the transformation of radioactive choline ([3H]Ch) was studied in different brain regions of the mouse following death by microwave irradiation of the head. Oxotremorine significantly increased the concentration of endogenous ACh in the cortex and hippocampus and of endogenous Ch in the cortex. Pretreatment with atropine (5 mg kg-1 i.p.) prevented the increase in ACh. The biosynthesis of radioactive ACh ([3H]ACh) was decreased in all brain regions. Atropine (5 mg kg-1) pretreatment counteracted this effect of oxotremorine (1 mg kg-1), while methylatropine (5 mg kg-1) had no effect except in the striatum. A calculation of the apparent turnover rate of ACh showed that oxotremorine (1 mg kg-1) decreased the turnover in the cortex, hippocampus, midbrain. and striatum.  相似文献

The diazotrophic cyanobacteria Trichodesmium spp. contribute approximately half of the known marine dinitrogen (N2) fixation. Rapidly changing environmental factors such as the rising atmospheric partial pressure of carbon dioxide (pCO2) and shallower mixed layers (higher light intensities) are likely to affect N2‐fixation rates in the future ocean. Several studies have documented that N2 fixation in laboratory cultures of T. erythraeum increased when pCO2 was doubled from present‐day atmospheric concentrations (~380 ppm) to projected future levels (~750 ppm). We examined the interactive effects of light and pCO2 on two strains of T. erythraeum Ehrenb. (GBRTRLI101 and IMS101) in laboratory semicontinuous cultures. Elevated pCO2 stimulated gross N2‐fixation rates in cultures growing at 38 μmol quanta · m?2 · s?1 (GBRTRLI101 and IMS101) and 100 μmol quanta · m?2 · s?1 (IMS101), but this effect was reduced in both strains growing at 220 μmol quanta · m?2 · s?1. Conversely, CO2‐fixation rates increased significantly (P < 0.05) in response to high pCO2 under mid‐ and high irradiances only. These data imply that the stimulatory effect of elevated pCO2 on CO2 fixation and N2 fixation by T. erythraeum is correlated with light. The ratio of gross:net N2 fixation was also correlated with light and trichome length in IMS101. Our study suggests that elevated pCO2 may have a strong positive effect on Trichodesmium gross N2 fixation in intermediate and bottom layers of the euphotic zone, but perhaps not in light‐saturated surface layers. Climate change models must consider the interactive effects of multiple environmental variables on phytoplankton and the biogeochemical cycles they mediate.  相似文献

Abstract— It is shown that transketolase activities in red blood cells and whole brain of normal and thiamine-deficient rats correlate well with heart frequencies.
The effect of thiamine depletion on the levels of acetylcoenzyme A (acetyl-CoA) and acetylcholine (ACh), and on the activities of pyruvate dehydrogenase, choline acetyl-transferase and acetylcholine esterase was studied in whole brains of thiamine-deficient, thiamine-supplemented ad libitum and pair-fed rats. The concentrations of acetyl-CoA and ACh decreased in thiamine-deficient brains by 42 and 35 per cent, respectively.
Total pyruvate dehydrogenase activity did not change during vitamin B1 deficiency. The 'resolved' enzyme, reconstituted with thiamine diphosphate, had an association constant of 5.4 × 10−6 m . Choline acetyltransferase and acetylcholine esterase activities remained unchanged in thiamine deficiency.
Possible mechanisms which could explain the reduced Ach levels in vitamin B1 deficiency are discussed.  相似文献

The time course of the incorporation of choline from plasma into a high and a low molecular weight fraction from mouse brain synaptosomes was studied. The fractions were obtained from lysed synaptosomes by gel filtration on Sephadex G-25. An extremely rapid incorporation of radioactivity into acetylcholine was found in both fractions and in the time interval 0.25-9 min after the intravenous administration of labelled choline, higher specific radioactivities of acetylcholine were found in the high molecular weight fraction than in the low molecular weight fraction. However, the specific radioactivity of choline in the high molecular weight fraction was much lower than that of acetylcholine. It was found that barbiturate anaesthesia caused a marked decrease in the labelling of acetylcholine in the high molecular weight fraction while the incorporation into the low molecular weight fraction was affected to a much smaller extent. Acetylcholine of the high molecular weight fraction showed properties similar to those of vesicle-bound acetylcholine. The recoveries of labelled and endogenous acetylcholine and choline from the brain homogenates were calculated in different steps of the fractionation procedure. In the fraction containing lysed synaptosomes the recovery of radioactive acetylcholine was lower than that of endogenous acetylcholine. This may indicate the presence of two types of bound acetylcholine in the synaptosomes. Different models for the intraneuronal synthesis of acetylcholine are discussed and it is proposed that a site of acetylcholine synthesis in vivo may be closely associated with some constituent of the high molecular weight fraction and directly coupled with the storage of the transmitter.  相似文献

本实验应用微量离子电泳技术,研究了乙酰胆碱对明或暗适应状态青蛙(Rana esculenta)松果体无色感或光敏神经元(Achromatic or Lumninance neuron)自发放电的影响.在所检测过的124个神经元中,乙酶胆碱导致其自发放电频率显著上升的大约占82%,另外有2%的神经元对乙酰胆碱的反应为抑制性,其余的16%无反应.乙酰胆碱引起的这种兴奋性反应具有剂量相关性,可逆性和重复性.  相似文献

Abstract— The effect of zuclomiphene, a hypocholesterolemic agent, on developing rat CNS cholesterol biosynthesis was examined. Sterol content and composition was studied in relation to age in four regions of the CNS, cerebrum, brain stem, spinal cord and cerebellum. Sterol content of all four regions was slightly lower in drug-treated animals than in controls. Brain stem and spinal cord were more susceptible to the effects of zuclomiphene than were cerebrum and cerebellum. Drug treatment resulted in the accumulation of desmosterol and zymosterol (5 x -cholesta-8,24-dien-3β-ol) in all CNS regions. After 15 days of drug treatment, desmosterol constituted more than 50% of the total sterol in the four examined regions. Six to 9% of the total sterol was zymosterol.
Examination by electron microscopy indicated only minimal morphological changes. Occasionally, neuronal membranous cytoplasmic inclusion bodies were evident.  相似文献

Abstract— Acetylcholine and choline were identified and their concentrations measured, by means of gas chromatography/mass spectrometry, in extracts obtained from nerve fibers of the hindmost stellar nerve of the squid Sepioteuthis sepioidea. These compounds were quantitated in samples of stellar nerve devoid of giant fiber, intact giant nerve fiber, extruded axoplasm, and axoplasm-free giant nerve fiber sheaths. In 11 samples of stellar nerve devoid of giant fiber, weighing an average of 20.8 ± 2.3 mg ( s.e.m. ), 756 ± 91 pmol ACh and 8.65 ± 0.62 nmol of choline were found. The total ACh content of the largest fibre in this group (10 μ m in diameter), for a 5 cm length of nerve, is in the order of 0.16 pmol. The average wet weights of a single giant nerve fiber (270-420 μ m in diameter) and its separate components ( s.e.m .; in mg; number of fibers in parentheses) were: intact fiber, 4.58 ± 0.19 (25); extruded axoplasm, 3.38 ± 0.13 (20); sheaths, 1.21 ± 0.11 (16). The average ACh content per unit weight of sample was about 2-3 times higher in the sheaths (5-13 pmol-mg−1) than in the axoplasm (2-4 pmol mg−1), whereas the ACh concentrations estimated per unit volume of cellular water were about 40 times higher in the Schwann cell (107-222 μ m ) than in the axon (2-5 μ m ). These experimental findings establish the presence of ACh in the giant nerve fiber of S. sepioidea. They also indicate the Schwann cells themselves as the main source for the release of ACh, responsible for their long-lasting hyperpolarizations following the conduction of nerve impulse trains by the axon.  相似文献

Abstract— Pre-treatment of rat brain slices with organic mercurials prevents the increased acetylcholine release induced by tityustoxin. This inhibition is reversed by dithiothreitol. N-Ethylmaleimide blocks the tityustoxin effect irreversibly. Simultaneous incubation with mercurials and toxin reveals a competition of both ligands for a membrane sulfhydryl group apparently required for tityustoxin activity.  相似文献

Abstract— Slices of electric organ of Torpedo marmorata were chopped and incubated in a saline-urea-sucrose medium. This preparation of minced tissue exhibited a relative enrichment in ACh and nerve endings, which was attributed to a loss of electroplaque cytoplasm. Electron microscopic controls showed nerve endings of normal morphology, some of them forming 'chaplets' separated from electro-plaques. Miniature endplate potentials were recorded on sealed fragments also present in this preparation. ACh levels remained unchanged during incubation periods as long as 19 h. The time course of the incorporation of [1-14C]acetate of [2-14C]pyruvate into ACh pools was studied. These incorporations were similarly affected by the choline added to the medium. In the presence of increasing choline concentrations (up to 10-4 m ), the incorporation of [14C]acetate or [14C]pyruvate into ACh increased. They both diminished when choline was added above 10-4M. The ACh content of the tissue was not affected by added choline. From the constancy of ACh levels in the presence of various choline concentrations and from the steady state of our preparation, we can conclude that the release of transmitter varied in parallel to the incorporation rate of the precursor of the acetyl moiety of ACh. This fact was also found using the efflux of [14C]acetate as an evaluation of ACh release. The values of release calculated by this method were in good agreement with those determined from the incorporations of acetate and pyruvate into ACh. It is suggested that the primary action of choline is on its high affinity carrier system. This triggers a secondary action on the ACh release mechanisms.  相似文献
