The major C14-labeled substance in sieve tube exudate of M. pyrifera is D-mannitol, comprising 3.6% (w/v). No sugars are detectable. Certain amino acids also possess some CWabeling and occur in significantly high concentrations in exudate. The exudate contains negligible ether-soluble lipid, but has a large amount of protein and a high concentration of K+. Neither protein nor lipid become labeled significantly in sieve tubes during short-term translocation experiments with C14. In general the chemical composition of the assimilate stream is comparable to that of vascular plants and does not, consequently, necessitate a different mechanism for translocation. 相似文献

The uptake of C14-labeled proteins (lysozyme, hemoglobin, lactoglobulin, and ovalbumin) from solution by tomato plants with sterile roots was studied. It was found that C14-compounds (proteins and/or protein-degradation products) were translocated to the foliage if the roots had undergone minor mechanical injury or if the plants were subjected to temporary wilting, i.e., physiological damage. C14-lysozyme was not transported to foliar tissue in healthy plants; C14-hemoglobin showed radioactivity in leaves of both healthy and injured plants, but there was evidence of a breakdown of the molecule; C14-ovalbumin gave a faint labeling of foliar tissues of some plants in which wilting or mechanical damage was below the threshold of detection. It is concluded, however, that translocation of proteins from roots in nutrient solution to tomato leaves does not occur in significant amounts in healthy plants in spite of the large uptake of proteins by root cortex, as found in earlier studies. 相似文献

Translocation patterns in the giant kelp, Macrocystis integrifolia Bory, were investigated in situ using 14C tracer; sources and sinks were identified. Export was first detected after 4 h of labeling; experiments were routinely 24 h continuous 14C application. Mature blades exported 14C to young blades on the same frond and on younger fronds, as well as to sporophylls and frond initials at the bases of the fronds. Blades <0.3 m from the apex imported and did not export; this distance did not change seasonally. In spring export from blades 0.3–1.25 m from the apex was exclusively upwards; older blades also exported downwards. In fall downward export began 0.5 m from the apex, and blades >2 m from the apex exported exclusively downwards. Carbon imported by frond initials, young fronds, and sporophylls in fall may partly be stored for growth in early spring. No translocation was seen in very young plants until one blade (secondary frond initial) bad been freed from the apical blade; this blade exported to the apical blade for a time, but imported when it began to develop into a frond. The second and third formed blades on the primary fronds (sporophylls also exported when <0.3 m from the apex, and later stopped. Frond initials and sporophylls on later-formed fronds did not export at all. The translocation pattern in M. integrifolia differs from that previously reported in M. pyrifera in seasonal change and in distances from the apex at which the changes take place. 相似文献

A quantitative accounting of the solids in the sieve tube sap from Macrocystis pyrifera (L.) e. Ag. was performed. The major organic compounds (mannitol, amino acids, and protein) and inorganic cations (K+, Na+, Mg2+ and Ca2+) were present near previously reported levels. The anions, until now unreported (except for iodide), included chloride as the major inorganic ion, bromide, phosphate, nitrate, and bicarbonate as the major inorganic carbon species. Sulfate and ammonium were below the detection limits of 1.5 ppm and 2 ppm, respectively. The elements B and As were also present although their speciation was not determined. 相似文献

Translocation of C14-labeled photosynthate was studied in nodulated pea and subterranean-clover plants which were grown either continuously without combined nitrogen or exposed to NaNO3 in the nutrient solution for five days prior to C14O2 assimilation. Supplying combined nitrogen decreased the proportion of photosynthate translocated to nodules with a corresponding increase in the proportion going to roots. Nodules on plants grown without combined nitrogen had a higher radioactivity than nodules on plants treated with sodium nitrate. 相似文献

Neutral lipids, consisting primarily of triacylglycerols, were found to be a major form of carbon reserve in zoospores of the giant kelp Macrocystis pyrifera (L.) C. Ag. The fluorescent stain Nile Red revealed large lipid droplets in the posterior end of the cell, which comprised 20–41% of cellular carbon in newly released spores. Flow cytometric analyses of newly released spores stained with Nile Red revealed considerable variation in the neutral lipid content among spores that was independent of spore size. Lipid droplets were consumed during germination in spores maintained either under constant light or in continual darkness. The availability of light appeared to delay, but did not preclude, lipid use. The rate of lipid use during germination varied considerably among germlings with some cells consuming all of their lipid reserves within 5 h after release. In addition to zoospores, lipid droplets were observed in both male and female gametes. Numerous droplets were observed in eggs, while single lipid droplets were observed in sperm. Neutral lipid droplets were not observed in gametophytes or sporophytes except in developing gametes and spores. Large lipid reserves thus seem to be confined to the microscopic life history stages that presumably have relatively high energy demands. By serving as a supplemental fuel reserve, neutral lipids may be important in extending the effective range of zoospore dispersal. 相似文献

Radioactive bicarbonate was pulse fed to blades of Macrocystis pyrifera (L.) C. A. Ag. and the movement of the 11C-labelled photoassimilates was monitored in vivo using an externally mounted array of Geiger-Müller detectors. Results of experiments conducted in August 1982 and February 1983 showed kinetic transport profiles composed of short pulses of 11C (periods of two to three minutes and six to eight minutes) and a mass flow component travelling with a speed of 6–22 cm · h?1. The pulse-like movement of 11C-photoassimilates, revealed for the first time in a kelp, may be driven by an energy-assisted transport mechanism. Light microscopy revealed a putative symplastic transport pathway from the photo synthetic meristoderm to the medullary sieve cells in the M. pyrifera blade. Of particular importance were the connections between the inner cortical cells and thin-walled medullary sieve cells. Electron microscopy showed sieve plate pore diameters ranging between 35–60 nm in the cortex and ca. 40 nm in the end walls of the thin-walled sieve cells. 相似文献

Four-month-old carrot plants exposed to 14CO2 for 1 hr in light were harvested successively at 1, 5, 10, 26, and 78 hr after initial exposure. Half-mm thick transverse slices from 3 and 10 cm below the crown were frozen quickly, freeze-dried, and autoradiographed on film. Radioactivity was first localized in a ring surrounding the cambium. The radioactive region extended centrifugally along radii as discrete loci to half the phloem thickness in 10 hr, in a pattern similar to radial rows of callose-stained sieve elements. Median longitudinal sections stained for callose demonstrated the presence of anastomosing sieve tube strands between the more vertical sieve tubes of differing age. Radioactive materials did not move across the cambium for 5–10 hr. These data fit with the decreased growth, in earlier studies, of uniform phloem explants removed from increasing distances from the cambium and of the lesser growth than phloem of similar xylem explants. 相似文献

Sporophytes of Macrocystis pyrifera (L.) C. A. Agardh of various stages of growth were studied by light microscopy to determine the initiation and ontogeny of secretory cells and the accompanying duct system. Secretory cells are initiated by asymmetric, periclinal divisions of meristoderm cells; subsequent mitoses increase the number of secretory cells associated with each duct. Duct formation occurs by schizogeny of anticlinal cell walls adjacent to the site of secretory cell initiation. Differences in distribution and structure of the duct system occur in various parts of the sporophyte. The duct system does not have openings directly to the sporophyte surface. Histochemical techniques showed that the duct contents are mostly sulfated polysaccharides with perhaps some lipid. 相似文献

Quantitative counts of regenerative sieve tubes and vessels were made in a large number of samples of mature internode #5 of C. blumei, with concomitant study of the fine details of vascular regeneration and the occurrence of the normally developing phloem anastomoses. Such anastomoses were found in many of the plants, but their average number in the small regenerating area was low (viz., 0.9 ± 0.2). With the phloem anastomoses excluded from the counts, the time course of regeneration was clear cut—no strands completed their regeneration around the wound until three days after wounding. More regenerative sieve tubes completed their differentiation under all conditions than did regenerative vessels. The number of sieve tubes and vessels regenerated by four days was closely related to the number of preexisting bundles of that type of vascular cell that had been severed by the transverse wound. The ratio of bundles severed by the wound in the phloem to those in the xylem was 2.14, and the ratio of the regenerative sieve tubes to the regenerative vessels was 2.24. For both tracheary and sieve tube cells the initial regeneration was strongly polar (mostly above the wound), as expected from earlier IAA transport data. The path of tracheary regeneration was obviously related to that of the sieve tubes on the other side of the cambium. 相似文献

1. Starch, containing 7.05 atom per cent C13 excess has been produced in the mesophyll cells of bean leaves, starting with C13O2 containing 7.26 atom per cent C13 excess. Approximately 67 per cent of the C13 taken up by the leaves was determined in the starch fraction. 2. The soluble carbohydrate, containing 6.72 atom per cent C13 excess, accounts for approximately 23 per cent of the C13 taken up by the leaves. The normal supply of growth substances to a young soybean plant was altered by removing the plant's apical meristem and replacing this meristem with an aqueous solution of either indole-3-acetic acid (IAA), gibberellic acid (GA), or water. The length of each experiment was 1 hr. In the middle of it, 14CO2 was administered to one of the primary leaves of the plant, and at the end distribution of 14C in various parts of the plant was determined. It was found that an addition of growth substances stimulated translocation in three different ways. Both IAA and GA increased the total amounts of sucrose-14C translocated, increased the rate of its translocation, and affected the distribution pattern of translocated sucrose throughout the plant. Experiments using IAA-14C have shown that the action of IAA is on the longitudinal translocation in the stem and not on the transfer of photosynthate from the mesophyll to the conducting tissues of the leaf. 相似文献

通过细胞学观察,在普通小麦栽培品种“丰抗13”和“京红1号”的杂交后代中,发现有多价体出现,这就表明有染色体易位发生。为进一步弄清究竟是哪条染色体发生了易位,我们采用单体测交方法,观察鉴定所有各单体系F_1的花粉母细胞第一次减数分裂中期Ⅰ(以下简称PMCs中Ⅰ)染色体构型。从鉴定结果发现,凡2n=42的F_1 PMCs中Ⅰ出现19~Ⅱ 1~Ⅳ,而2n=41的F_1PMCs中Ⅰ的染色体构型不同,单体与易位有关的两个单体系4B和1D F_1 PMCs中的Ⅰ构型中有部分呈现为19个二价体加1个三价体,即19~Ⅱ 1~Ⅲ,没有单价体,而其余各单体系F_1 PMCs中Ⅰ构型则表现为18个二价体,1个四价体和1个单价体,即18~Ⅱ 1~Ⅰ 1~Ⅳ。因此,可以肯定“丰抗13”存在1个染色体易位,其有关染色体就是4B和1D。 相似文献

Allium cernuum, Vicia faba, and Tradescantia paludosa were treated by root immersion in maleic hydrazide (1 mM/liter) labeled with C14 (C14-MH) for 1 hour to determine the location within the cell to which MH moves during various periods of time after treatment. Root tips were fixed 24 hours, 48 hours, 72 hours, and 3 weeks after treatment. Autoradiographs of root tips squashed 24 to 72 hours after fixation showed that C14-MH was distributed throughout the nuclei and was particularly concentrated in the nucleoli. The nucleolar localization of the chemical was transitory, fixations made 3 weeks after treatment showing well labeled nuclei many of which completely lacked label in the nucleoli. The chromosomes seen in mitotic divisions of all three species had the same amount of label in euchromatic as heterochromatic areas. Since the chemical was not accumulated preferentially in heterochromatic areas, it seems likely that the reported specificity of MH for the breakage of heterochromatin can not be due to preferential heterochromatic incorporation. 相似文献
