The large-particle fraction from the cytoplasm of chick embryo fibroblasts infected with Semliki Forest virus was found to catalyze the incorporation of the 5'-triphosphates of guanosine, adenine, cytidine, and uridine into an acid-insoluble alkali-labile product. The conditions affecting the preparation and assay of this enzyme were investigated. The ribonucleic acid (RNA) polymerase was not present in uninfected cells, and it appeared in infected cells at the time of rapid viral RNA synthesis. The polymerase was found to catalyze the synthesis of a species of RNA which was resistant to ribonuclease and which exhibited the sedimentation properties, buoyant density, and thermal transition temperature of the double-stranded RNA found in vivo in chick cells infected with Semliki forest virus. Attempts to demonstrate that the reaction product of this enzyme also included single-stranded viral RNA were not successful. Although other interpretations are possible, these results give some support to the suggestion that more than one enzyme may be involved in the replication of viral RNA.  相似文献

An immune ribonucleic acid (iRNA) preparation was made using phenol extracts of spleens of mice previously immunized with Salmonella tennessee flagella. An enzyme, also prepared from the spleens of these mice, induced the incorporation of 3H-UTP into the acid-insoluble fraction in a cell-free system in the presence of this RNA. The enzyme activity could be demonstrated from the spleens of immunized mice but not from normal ones, and this activity was also inhibited by two derivatives of rifamycin. Treatment with ribonuclease or heating at alkaline pH resulted in a loss of activity in added RNA. The 3H-uridine-labeled product was found resistant to ribonuclease treatment but became sensitive when the product was subjected to heat treatment. However, actinomycin D, mitomycin C or bleomycin A2 did not inactivate the enzyme activity. These results suggest that this enzyme induces the incorporation of UTP into the acid-insoluble fraction using iRNA as a template and the product may be a newly synthesized RNA which forms a hybrid with iRNA. This enzyme activity may play a role in the antibody formation process, and may account for the in vivo replication of iRNA by this enzyme, viz., probably an RNA-dependent RNA replicase.  相似文献

"L-cell virion," obtained from chronically infected 929 L-cell lines, was purified on 70% sucrose pads and its ribonucleic acid (RNA) was extracted and analyzed on sucrose gradients. Two RNA components were isolated from the virion, a rapidly sedimenting species (82S) and a more slowly sedimenting species (6S). When the fast-moving component was heated to 45 C, it yielded a homogeneous peak which now sedimented at approximately 72S. Upon heating to 70 C, the peak dissociates into a smaller homogeneous RNA species with a sedimentation coefficient of approximately 33S. Ribonuclease treatment and base composition analysis revealed that the fast-moving species is a single-stranded molecule. Base ratio analysis also revealed that the small (6S) RNA component is similar in its base composition to L-cell ribosomal RNA and distinctly different from the base composition of the large (72S) RNA component. Actinomycin D inhibits production of the L-cell virion by the L-cell line.  相似文献

A virus-induced ribonucleic acid (RNA) polymerase activity was found in L cells infected with type 3 reovirus. Most of the enzyme is associated with the "large particle" fraction of the infected cells. The enzyme first appeared at 3 to 5 hr after infection and increased in amount until 7 to 9 hr. All four ribonucleoside triphosphates are incorporated in vitro into an acid-insoluble form by the enzyme. The major part of the product formed in vitro is a double-stranded RNA indistinguishable from viral RNA by electrophoresis on polyacrylamide gel. Approximately 40% of the product is a single-stranded RNA of relatively small molecular weight. More than 95% of the nucleotides incorporated into double-stranded RNA by the enzyme are bound in internal 3'-5'-phosphodiester linkages extending back from both 3'- and 5'-termini of the RNA strands.  相似文献

Ribonucleic acid (RNA)-dependent RNA polymerase activity was demonstrated in the microsomal and ribosomal fraction from the spleen cells of immunized mice. The enzyme activity was solubilized by Triton X-100 from the fraction and partially purified by Biogel A 1.5 M column chromatography. The RNA-dependent RNA polymerase activity was eluted in a single peak from the column. High activity was demonstrated with an RNA preparation (iRNA) as template made from the spleens of immunized mice but very low activity was found with an nRNA preparation made from the spleens of normal mice. Incorporation of 3H-UTP markedly decreased in the presence of RNase but not in the presence of DNase. DNA preparations made from the spleens of immunized mice were inactive as template for this enzyme. The iRNA preparation was fractionated by sucrose density gradient centrifugation. A fraction corresponding to 12–13 S was most active as a template. It was followed by a fraction corresponding to 6–7 S. Sucrose gradient analysis of the 3H-UTP-labeled product was attempted. The effect of the ribonucleic acid (RNA) control (RC) gene on the biosynthesis of viral RNA has been examined in an RC(str) and an RC(rel) host infected with R17 RNA bacteriophage under conditions in which host RNA and protein synthesis were inhibited by the addition of rifampicin. Methionine and isoleucine starvation depressed viral RNA biosynthesis in an RC(str) host but not in an RC(rel) host. However, histidine starvation had little effect on viral RNA and protein synthesis in both RC(str) and RC(rel) cells, although it had a marked effect on host protein and RNA synthesis in an RC(str) host. Chloramphenicol relieved the effect of amino acid starvation on viral RNA synthesis in an RC(str) host. It is concluded that stringent control of viral RNA biosynthesis does not require the continued biosynthesis of the RC gene product (RNA or protein) and that a preformed RC gene product can regulate the biosynthesis of the exogenous RNA. It is suggested that the amino acid dependence of viral RNA biosynthesis is due to its obligatory coupling with the translation of the viral coat protein which lacks histidine. It may be inferred that the amino acid requirement of bacterial RNA is due to its coupling with the translation of a host-specific protein (other than the RC gene product) which requires a full complement of amino acids. Since chloramphenicol is known to permit ribosome movement in the absence of protein synthesis, it is suggested that ribosome movement along the nascent RNA chain is a sufficient condition for the continuation of RNA synthesis.  相似文献

Oncornavirus 60 to 70S ribonucleic acids (RNA), such as those from avian myeloblastosis virus, Schmidt-Ruppin virus, or mouse sarcoma-mouse leukemia viruses, isolated by conventional techniques, contain 4S transferlike RNA molecules that are released upon dissociation of the 60 to 70S RNA with heat. The 4S RNA represents 2.5 to 3.0% of the RNA in the 65S aggregate or 4 to 5 molecules per molecule of 35S RNA formed.  相似文献

The nature and properties of the 20S ribonucleic acid which accumulates only during the sporulation of Saccharomyces cerevisiae were examined. The 20S ribonucleic acid (RNA) has a base composition considerably different from ribosomal RNA species and is virtually unmethylated. The 20S RNA did, however, exhibit approximately 70% homology with 18S RNA by RNA-deoxyribonucleic acid filter hybridization competitions. The 20S RNA showed a hybridization saturation plateau level 30 to 40% higher than 18S, consistent with measurements of the size difference in polyacrylamide gels. Pulse-chase experiments in the presence and absence of cycloheximide indicate that the 20S RNA has a presumptive relationship to the 20S ribosomal RNA precursor normally observed only in short pulse-labeling in vegetative cells.  相似文献

The synthesis of ribonucleic acid (RNA) and of protein in Escherichia coli during glucose-lactose diauxie lag have been examined. The rate of RNA synthesis is about 7%, of the corresponding rate during exponential growth and the rate of protein synthesis 10 to 15%. Inhibition of RNA synthesis occurs to the same extent in both rel and rel(+) strains. The RNA which accumulates during 20 min in diauxie lag is composed of about 50% ribosomal and transfer RNA species and about 50% of a fraction which resembles messenger RNA (mRNA) in its heterogeneous sedimentation properties. Decay of the heterogeneous fraction occurs in the presence of glucose and actinomycin D with a half-life of 3 min, the same as that of pulse-labeled mRNA; however, during the diauxie lag, the half-life of this RNA is about 25 min. Accumulation of the heterogeneous RNA is further increased when protein synthesis is blocked by chloramphenicol. The data suggest that the disproportionate accumulation of mRNA during diauxie lag and energy source shift-down may be attributed at least in part to increased stability of mRNA, but do not rule out a preferential synthesis of mRNA.  相似文献

In an MS2 phage ribonucleic acid (RNA)-directed in vitro protein-synthesizing system, the coat protein cistron and the adjacent RNA polymerase cistron are translated non-continuously. The ribosomes which have completed the synthesis of coat protein dissociate from the MS2 RNA and do not read through the intercistronic gap. Purified poliovirus single- and double-stranded ribonucleic acids (RNA) were examined by electron microscopy. The length of both molecules was found to be 2.37 mum. The uncorrected sedimentation coefficient for single-stranded RNA is 33S, as compared to 27S for the RNA of tobacco mosaic virus. It is calculated from these results that the molecular weight of the sodium form of poliovirus is 2.6 x 10(6) daltons.  相似文献

NaClO(4) was employed in a technique for the rapid extraction of reovirus ribonucleic acid (RNA). The extracted RNA, which was purified in a Cs(2)SO(4) equilibrium density gradient, had a buoyant density of 1.61 g/cm(3) and a sedimentation coefficient of 15S in a 7 to 20% sucrose gradient. It was 90% resistant to ribonuclease in a solution of high ionic strength (0.1 m NaCl). The sensitivity of reovirus RNA to ribonuclease increased with decreasing ionic strength. The thermal denaturation transition of the RNA began at 78 C and was complete at 85 C. The T(m) of the transition was 81 C in 0.01 m tris(hydroxymethyl)aminomethane buffer (pH 7.2) containing 0.001 m ethylenediaminetetraacetate. Thermal denaturation of reovirus RNA resulted in the formation of three ribonuclease-sensitive fractions. Denaturation at 25 C in the presence of dimethyl sulfoxide resulted in the formation of two ribonuclease-sensitive fractions.  相似文献

Daunomycin and ethidium bromide, two deoxyribonucleic acid-intercalating drugs, inhibit ribonucleic acid (RNA) and protein synthesis in Saccharomyces cerevisiae. Both agents rapidly curtail uptake of radioactive adenine, whereas the kinetics of radioactive leucine uptake after drug addition are consistent with translation of a pool of exponentially decaying messenger RNA. Messenger RNA half-life determinations from these experiments gave identical results over a range of drug concentrations; this value is 21 +/- 4 min at 30 C. In a temperature-sensitive mutant in which RNA synthesis is curtailed at the nonpermissive temperature, a similar half-life for messenger RNA decay is found both in the absence and in the presence of either drug. This indicates that at the concentrations used in this study, neither daunomycin nor ethidium bromide has an appreciable direct effect on translation and do not increase the lability of messenger RNA.  相似文献

A 26 base long oligodeoxyribonucleotide complementary to a common RNA sequence of potato spindle tuber viroid (PSTV) and chrysantemum stunt viroid (CSV) was synthesized. The 3'-end biotinylated one was used for the detection of PSTV and CSV RNA immobilized on nitrocellulose filters by nucleic acid hybridization. Visualization of hybridization results was performed by two ways, either by streptavidin-alkaline phosphatase conjugate or streptavidine and biotinylated alkaline phosphatase. It was possible to detect 0.65 ng of purified CSV and PSTV RNA. The suggested system of viroid diseases detection can be used by agricultural and horticultural enterprises.  相似文献

Foot-and-mouth disease virus (FMDV)-specific ribonucleic acid (RNA) was analyzed by electrophoresis on 0.5% agarose gels. Four classes of RNA were resolved as a function of mobility in agarose: two classes of slowly migrating multistranded RNA, the infectious viral RNA with intermediate mobility, and a minor fast-moving class of lower-molecular-weight single-stranded RNA. The major RNA species were infectious viral RNA and the slowest migrating class of multistranded RNA. The latter RNA was polydisperse when analyzed by sucrose gradient centrifugation, it was partially ribonuclease resistant, and it was the predominant RNA species labeled during the initial period of (3)H-uridine triphosphate incorporation in the cell-free system. Heat treatment studies indicated that part of the slowest-moving RNA was degraded at 60 C and almost complete degradation was detected at 100 C. It was concluded that this RNA is the replicative intermediate in viral RNA synthesis. The second class of multistranded RNA contained both a ribonuclease-resistant RNA and a second RNA peak which was detected only after heat treatment at temperatures above 75 C. Fractions of FMDV-specific RNA isolated by sucrose gradient centrifugation were analyzed by agarose-gel electrophoresis. Infectious viral RNA was detected only in the 37S zone and was the major species of RNA in this part of the gradient. The ribonuclease-resistant RNA (the 20S zone) contained about equal amounts of multistranded RNA (both classes) and the low-molecular-weight single-stranded RNA. All sucrose gradient fractions between 20 and 40S were found to contain the replicative intermediate, although the major portion was detected in the 20 to 25S region.  相似文献

Pyronin, when used in the methyl green-pyronin stain, is useful in localizing ribonucleic acid (RNA). That it has rarely been used alone is perhaps a result of the observation (Kurnick 1955) that pyronin stains deoxyrobonucleic acid (DNA) of animal tissue when not competitively inhibited by methyl green. The tests described in this note indicate that pyronin alone can be used to demonstrate RNA in fixed plant tissues.  相似文献

A cytoplasmic structure containing the viral ribonucleic acid (RNA) polymerase has been isolated by sucrose density centrifugation from cells infected with Sindbis virus. Uninfected cells did not contain any such structure. Preliminary experiments indicated that the structure may be associated with membranes. This structure incorporated (3)H-guanosine triphosphate in vitro in the absence of added template. The RNA synthesized in vitro by the enzyme consisted of single-stranded 40S RNA, the ribonuclease-resistant replicative form, and possibly the replicative intermediate form of viral RNA. The products formed in vitro by the enzyme are identical in sedimentation rates to those formed in the infected cells in vivo.  相似文献
