Sink-limited conditions, defined as treatment with continuousillumination, cause a reduction in the rate of photosyntheticfixation of CO2 in single-rooted leaves of soybean (Glycinemax. Merr. cv. Turunoko). We suggested previously that thisreduction is due to a deactivation of ribulose-1,5-bisphosphatecarboxylase (RuBPcase, E.C. 4.1.1.39 [EC] ) that is caused by a decreasein the level of Pi in the leaves [Sawada et al. (1989) PlantCell Physiol. 30: 691, Sawada et al. (1990) Plant Cell Physiol.31: 697]. In the present study, the mechanism of regulationof RuBPcase activity by Pi was examined. The activity of RuBPcasein the sink-limited leaves, exposed for 6 or 7 d to continuousillumination to alter the source/sink balance, was enhancedwith increasing concentrations of Pi, in a CO2-free preincubationmedium in the presence of 5 mM MgCl2 The maximum value [6.3µmole CO2 (mg Chl)–1 min–1] was obtained atapproximately 5 mM Pi after a 5 min incubation, being 3 timesof the activity without the preincubation. The activity of acrude preparation of RuBPcase that had been deactivated by removalof CO2 and Mg2+ ions by the gel filtration was 5.2–9.3nmole CO2 (mg protein)–1 min–1 and was also enhancedby Pi plus Mg2+ ions. The maximum value [147–151 nmoleCO2 (mg protein)–1 min–1] was attained at 5 mM Piafter a 5 min incubation. The cycle of activation and inactivationof deactivated crude RuBPcase was perfectly reversible by additionof Pi to the enzyme and removal of Pi from the enzyme. Levelsof free Pi and of esterified phosphate in the sink-limited leaveswere 69% and 31% of the total phosphate, respectively. By contrast,in the control leaves, these values were 87% and 13%, respectively.These results support our previously stated hypothesis and indicatean important role for free Pi in the regulation of RuBPcaseactivity, in particular in sink-limited plants. Photosynthesis is known to occur in rice panicles, but littlehas been reported about the photosynthetic or biochemical characteristicsof such panicles. The estimated gross amount of photo-syntheticallyassimilated CO2 in a panicle is 30% of that in a flag leaf.This result and the good light-intercepting characteristicsof the panicle in the canopy suggest that photosynthesis inthe panicle may contribute significantly to grain filling. Therice panicle is composed of spikelets and of rachis-branchesincluding rachis which have estimated gross rates of photosynthesisduring the 30-day period after anthesis of 130 to 180 and 50to 100 µmol CO2.(mg Chl)–1.h–1, respectively.The corresponding rate for the flag leaf is 180 to 230 µmolCO2.(mg Chl).h. On the basis of Chl, spikeletshave a high photosynthetic capability which is similar to thatof the flag leaf. The activities of ribulose-l,5-bisphosphate carboxylase (RuBPCase),phosphoenolpyruvate carboxylase (PEPCase), and pyruvate.Pi dikinase(PPDK) in spikelets were 129, 220, and 87 µmol.(mg Chl).h,respectively. The activities of PEPCase and PPDK in spikeletswere considerably higher than those in the flag leaf or rachis-branches.Oxygen-insensitive photosynthesis was found only in spikelets.The Km of NaHCO3 for photosynthesis by slices of spikelets inan aqueous solution (0.6 mM) was considerably lower than thatfor slices of flag leaf (4.2 mM). All these results indicatethat spikelets have different photosynthetic characteristicsfrom those of the flag leaf and rachis-branches. In the present study, we investigatedwhether, in intact soybean plants, sink-limited conditions wouldalso cause a reduction in the rate of photosynthesis and whethersuch a reduction might be due to the deactivation of RuBPcasevia the same regulatory mechanism as that suggested from previousstudies with single-rooted leaves. Continuous removal of flowerbuds from intact plants brought a large decrease in ratio ofthe dry weight of sink organs (stem, roots, pods) to sourceorgan (leaves) as a result of the absence of pod formation.Pods are likely to function as the major sink at the reproductivestage. Upon continuous removal of flower buds, the treated (sink-limited)plants showed a large decrease in the rate of photosyntheticfixation of CO2 as compared to control plants. RuBPcase in theleaves of treated plants was continuously inactivated with thedecrease in photosynthetic activity. However, the inactivatedenzyme was totally reactivated upon incubation in the presenceof 10 mM NaHCO3 and 5 mM MgCl2. The levels of sucrose and ribulose-1,5-bisphosphatein leaves of the treated plants increased significantly. C4 model plants composed of single-rooted Amarantus cruentusL. leaves were developed to study source-sink relationships.The photosynthetic activity of CO4 fixation in the rooted leavescultured under the control condition (10-h light/14-h darkness,730 µmol photons m–2s–1) remained constantand high. When the leaves kept for 8 or 9 d under the controlcondition were exposed to continuous light (CL) for 4 d, theactivity decreased steeply. The starch content increased significantlyduring the first 2 d in CL whereas the sucrose content increasedalmost linearly during the 4 d in CL. The close relationshipbetween the decrease in photosynthetic activity and the increasein sucrose indicates that the activity was subjected to feedbackinhibition when the end product was forced to accumulate inthe leaves. In the treated leaves, the initial activity of ribulose-1,5-bisphoshatecarboxylase decreased significantly. A large increase in ribulose-1,5-bisphosphateand decrease in 3-phosphoglycerate occurred following the deactivationin RuBPcase, which was fully restored by the preincubation withCO2 and Mg2+ and/or P1 and Mg2+. The activity of phosphoenolpyruvatecarboxylase also decreased but not as rapidly as RuBPcase. Thealanine and pyruvate levels decreased markedly but the oxaloacetate,glutamine and asparagine levels significantly increased whilephosphoenolpyruvate and glutamate were kept at almost constantlevels. The malate level first increased slightly and then decreasedsignificantly. The mechanism of coordinated decrease in carboxylationreactions between phtosynthetic carbon reduction cycle and C4metabolism in response to change in the source-sink balanceis discussed. The capacity for C4 photosynthesis in Panicum milioides, a specieshaving reduced levels of photorespiration, was investigatedby examining the activity of certain key enzymes of the C4 pathwayand by pulse-chase experiments with 14CO2. The ATP$P1 dependentactivity of pyruvate,P1 dikinase in the species was extremelylow (0.14–0.18 µmol mg chlorophyll–1 min–1).Low activity of the enzyme was also found in Panicum decipiensand Panicum hians (related species with reduced photorespiration)and in Panicum laxum (a C3 species). The antibody to pyruvate,P1dikinase caused about 70% inhibition of the ATP$P1 dependentactivity of the enzyme in P. milioides. The activity of NAD-malicenzyme and NADP-malic enzyme in P. milioides was equally low(approximately 0.1–0.2 µmol mg chlorophyll–1min–1) and similar to the activity in P. decipiens, P.hians and P. laxum. Photosynthetic pulse-chase experiments underatmospheric conditions showed a typical C3-like pattern of carbonassimilation including the labelling of glycine and serine asexpected during photorespiration. During the pulse with 14CO2only about 1% of the labelled products appeared in malate and2–3% in aspartate. During a chase in atmospheric levelsof CO2 for up to 6 min there was a slight increase in labellingin the C4 acids. The amount of label in carbon 4 of aspartatedid not change during the chase, indicating little or no turnoverof the C4 acid via decarboxylation. The results indicate thatunder atmospheric conditions P. milioides assimilates carbondirectly through the C3 pathway. Bot. 38: 1937–1948. Associations between photosynthesis, the activation state ofRuBP carboxylase and the contents of photosynthetic intermediateswere compared in soybean and wheat leaves before and after exposureto photoinhibitory treatments in the presence of atmosphericO2. Exposing attached leaves to a supra-saturating irradiance(3 800 µmol quanta m– 2 s–1) for 2 h in CO2-freeair decreased carboxylation efficiency and the light-saturatedphotosynthetic rate in air by approximately 50%. Exposure tothe photoinhibitory treatment for periods in excess of 2 h didnot cause a further decrease of photosynthesis in soybean. Althoughphotosynthesis was reduced, the initial and total (fully-activated)activities of ribulose 1,5-bisphosphate carboxylase (RuBPCase)in leaf extracts were unaltered in each species by the photoinhibitorytreatment. This was true for leaves sampled under both air andat a rate-limiting intercellular CO2 partial pressure (Ci) of75 µPa Pa–1. The contents of ribulose l,5-bisphosphate(RuBP) and 3-phosphoglyceric acid (3-PGA) were reduced by thephotoinhibitory treatment in soybean leaves sampled in air andat a rate-limiting Ci, although the RuBP/3-PGA ratio was unaffected.The relative reduction of RuBP content in soybean leaves atrate-limiting C1 was similar to the corresponding reductionof carboxylation efficiency. For wheat,the relative reductionof RuBP content at rate-limiting Ci (–19%) caused by thephotoinhibitory treatment was considerably less than the correspondingdecrease of carboxylation efficiency (–49%).The RuBP/3-PGAratio of wheat was also increased significantly by the photoinhibitorytreatment The significance of these observations to the regulationof CO2-limited photosynthesis in leaves experiencing photoinhibitionunder atmospheric oxygen is discussed. Consideration is alsogiven to the previous contention that contemporary measurementsof initial activity in crude extracts may provide a spuriousindication of the amount of the enzyme-CO2-Mg2 + form of RuBPcarboxylase present in the leaf. The relationship between chlorophyll a (Chl a) and primary productivity(PP) in the uppermost water layer and the water column-based(0–15 m) integral values of those variables were examinedusing measurements taken in Lake Kinneret (Israel) from 1990to 2003. In 81% of all Chl a profiles examined, the distributionwas fairly uniform within the entire 0–15 m water column,and 12.3% of instances showed a prominent subsurface maximum,when the lake phytoplankton was dominated by the dinoflagellatePeridinium gatunense. Chl a can be reliably estimated by remotesensing techniques in the productive and turbid water of LakeKinneret, since Chl a concentration at surface layers can beextrapolated to the entire water column. Light vertical attenuationcoefficient average for wavelengths from 400 to 700 nm, Kd,ranged from 0.203 to 1.954 m–1 and showed high degreeof temporal variation. The maximal rate of photosynthetic efficiency,PBopt [average 3.16 (±1.50)], ranged from 0.25 to 8.85mg C m–3 h–1 mg Chl a–1. Using measured dataof Chl a, PBopt, and light as an input, a simple depth-integratedPP model allowed plausible simulation of PP. However, a lackof correlation between photosynthetic activity and temperature(or other variable with remotely sensed potential) renders theuse of models that require input of photosynthetic efficiencyto calculate integrated PP of little value in the case of productiveand turbid Lake Kinneret.  相似文献

To study the effect of chronically elevated CO2 on the excitability and function of neurons, we exposed mice to 7.5–8% CO2 for 2 wk (starting at 2 days of age) and examined the properties of freshly dissociated hippocampal neurons. Neurons from control mice (CON) and from mice exposed to chronically elevated CO2 had similar resting membrane potentials and input resistances. CO2-exposed neurons, however, had a lower rheobase and a higher Na+ current density (580 ± 73 pA/pF; n = 27 neurons studied) than did CON neurons (280 ± 51 pA/pF, n = 34; P < 0.01). In addition, the conductance-voltage curve was shifted in a more negative direction in CO2-exposed than in CON neurons (midpoint of the curve was –46 ± 3 mV for CO2 exposed and –34 ± 3 mV for CON, P < 0.01), while the steady-state inactivation curve was shifted in a more positive direction in CO2-exposed than in CON neurons (midpoint of the curve was –59 ± 2 mV for CO2 exposed and –68 ± 3 mV for CON, P < 0.01). The time constant for deactivation at –100 mV was much smaller in CO2-exposed than in CON neurons (0.8 ± 0.1 ms for CO2 exposed and 1.9 ± 0.3 ms for CON, P < 0.01). Immunoblotting for Na+ channel proteins (subtypes I, II, and III) was performed on the hippocampus. Our data indicate that Na+ channel subtype I, rather than subtype II or III, was significantly increased (43%, n = 4; P < 0.05) in the hippocampi of CO2-exposed mice. We conclude that in mice exposed to elevated CO2, 1) increased neuronal excitability is due to alterations in Na+ current and Na+ channel characteristics, and 2) the upregulation of Na+ channel subtype I contributes, at least in part, to the increase in Na+ current density. sodium ion channels; oxygen deprivation  相似文献

Single, seed-grown plants of ryegrass (Lolium perenne L. cv.Melle) were grown for 49 d from the early seedling stage ingrowth cabinets at a day/night temperature of 20/15 C, witha 12 h photoperiod, and a CO2 concentration of either 340 or680µI 1–1 CO2. Following complete acclimation tothe environmental regimes, leaf and whole plant CO2 effluxesand influxes were measured using infra-red gas analysis techniques.Elevated CO2 increased rates of photosynthesis of young, fullyexpanded leaves by 35–46% and of whole plants by morethan 50%. For both leaves and whole plants acclimation to 680µI–1 CO2 reduced rates of photosynthesis in bothCO2 regimes, compared with plants acclimated to 340µll–1. There was no significant effect of CO2 regime onrespiration rates of either leaves or whole plants, althoughleaves developed in elevated CO2 exhibited generally lower ratesthan those developed in 340µI I–1 CO2. Initially the seedling plants in elevated CO2 grew faster thantheir counterparts in 340µI I–1 CO2, but this effectquickly petered out and final plant weights differed by onlyc. 10%. Since the total area of expanded and unexpanded laminaewas unaffected by CO2 regime, specific leaf area was persistently13–40% lower in elevated CO2 while, similarly, root/shootratio was also reduced throughout the experiment. Elevated CO2reduced tissue nitrogen contents of expanded leaves, but hadno effect on the nitrogen contents of unexpanded leaves, sheathsor roots. The lack of a pronounced effect of elevated CO2 on plant growthwas primarily due to the fact that CO2 concentration did notinfluence tiller (branch) numbers. In the absence of an effecton tiller numbers, any possible weight increment was restrictedto the c. 2.5 leaves of each tiller. The reason for the lackof an effect on tillering is not known. The causes of interspecific differences in the µ-l relationshipare examined in the context of a mechanistic model which relatesµ to irradiance in terms of six factors:, kc photosyntheticquotient (PQ), Chl a:C, respiration and excretion. The effectof cell size on the light saturated growth rate is also considered.It is shown that photosynthetic efficiency and PQ exhibit remarkablylittle interspecific variability, and average 0.024 ±0.005 µg C(µg Chl a)–1 h–1 (µEm–2 s–1)–1 and 1.5 ± 0.2 mol 02 molC–1 (when NO3 is the nitrogen source) respectively.Two useful relationships were derived: (i) between growth efficiency,g and Chl a:C at µ. = 0; (ii) between the compensationintensity, Ic and the Chl a-specific maintenance respirationrate. Both relationships were independent of temperature anddaylength. Species best adapted to growth at low light werefound to exhibit high Chl a:C ratios and low maintenance respirationrates. As a group, diatoms were consistently the best adaptedfor growth at low irradiance. Chiorophytes, haptophytes, chrysophytesand cryptophytes were intermediate in their performance at lowirradiance. Dinoflagellates exhibited extreme diversity, withspecies spanning the spectrum from very good performance atlow irradiance to very poor. A new µmax-cell carbon relationshipis given based on growth rates normalized to 15°C. Evidenceis presented to show that noise in this relationship can besignificantly reduced by using only carbon-specific growth ratesand using only data for species grown at the same daylength.  相似文献

Permeability coefficients (PS values) for CO2 of the plasmamembrane (PM) of the unicellular green algae Eremosphaera viridis,Dunaliella parva, and Dunaliella acidophila, and of mesophyllprotoplasts isolated from Valerianella locusta were determinedfrom 14CO2 uptake experiments using the rapid separation ofcells by the silicone oil layer centrifugation technique. Theexperimental PS values were compared with calculated numbersobtained by interpolation of Collander plots, which are basedon lipid solubility and molecular size, for D. parva cells,mesophyll protoplasts isolated from Spinacia oleracea, mesophyllcells and guard cells of Valerianella, and guard cell protoplastsisolated from Vicia faba. The conductivity of algal plasma membranes for CO2 varies between0.1 and 9 ? 10–6 m s–1, whereas for the plasmalemmaof cells and protoplasts isolated from leaves of higher plantsvalues between 0.3 and 11 ? 10–6 m s–1 were measured.By assuming that these measurements are representative for plantsand algae in general, it is concluded that the CO2 conductivityof algal PM is of the same order of magnitude as that of thehigher plant cell PM. Ps values of plasma membranes for CO2are lower than those for SO2, but are in the same order of magnitudeas those measured for H2O. On the basis of these results itis concluded that theoretical values of about 3000 ? 10–6m s–1 believed to be representative for higher plant cells(Nobel, 1983) and which are frequently used for computer-basedmodels of photosynthesis, lack experimental confirmation andrepresent considerable overestimations. However, with severalsystems, including higher plant cells, the conductance of thePM for CO2 was significantly higher in light than in darkness.This suggests that in light, additional mechanisms for CO2 uptakesuch as facilitated diffusion or active uptake may operate inparallel with diffusional uptake. Species-specific differences in the assimilation of atmosphericCO2 depends upon differences in the capacities for the biochemicalreactions that regulate the gas-exchange process. Quantifyingthese differences for more than a few species, however, hasproven difficult. Therefore, to understand better how speciesdiffer in their capacity for CO2 assimilation, a widely usedmodel, capable of partitioning limitations to the activity ofribulose-1,5-bisphosphate carboxylase-oxygenase, to the rateof ribulose 1,5-bisphosphate regeneration via electron transport,and to the rate of triose phosphate utilization was used toanalyse 164 previously published A/Ci, curves for 109 C3 plantspecies. Based on this analysis, the maximum rate of carboxylation,Vcmax, ranged from 6µmol m–2 s–1 for the coniferousspecies Picea abies to 194µmol m–2 s–1 forthe agricultural species Beta vulgaris, and averaged 64µmolm–2 s–1 across all species. The maximum rate ofelectron transport, Jmax, ranged from 17µmol m–2s–1 again for Picea abies to 372µmol m–2 s–1for the desert annual Malvastrum rotundifolium, and averaged134µmol m–2 s–1 across all species. A strongpositive correlation between Vcmax and Jmax indicated that theassimilation of CO2 was regulated in a co-ordinated manner bythese two component processes. Of the A/Ci curves analysed,23 showed either an insensitivity or reversed-sensitivity toincreasing CO2 concentration, indicating that CO2 assimilationwas limited by the utilization of triose phosphates. The rateof triose phosphate utilization ranged from 4·9 µmolm–2 s–1 for the tropical perennial Tabebuia roseato 20·1 µmol m–2 s–1 for the weedyannual Xanthium strumarium, and averaged 10·1 µmolm–2 s–1 across all species. Despite what at first glance would appear to be a wide rangeof estimates for the biochemical capacities that regulate CO2assimilation, separating these species-specific results intothose of broad plant categories revealed that Vcmax and Jmaxwere in general higher for herbaceous annuals than they werefor woody perennials. For annuals, Vcmax and Jmax averaged 75and 154 µmol m–2 s–1, while for perennialsthese same two parameters averaged only 44 and 97 µmolm2 s–1, respectively. Although these differencesbetween groups may be coincidental, such an observation pointsto differences between annuals and perennials in either theavailability or allocation of resources to the gas-exchangeprocess. The influence of salinity in the growing media on ribulose-1,5-bisphosphate (RuBP) carboxylase and on CO2 fixation by intactsugar beet (Beta vulgaris) leaves was investigated. RuBP carboxylase activity was mostly stimulated in young leavesafter exposure of plants for 1 week to 180 mM NaCl in the nutrientsolution. This stimulation was more effective at the higherNaHCO2 concentrations in the reaction medium. Salinity also enhanced CO2 fixation in intact leaves mostlyat rate-limiting light intensities. A 60 per cent stimulationin CO2 fixation rate was obtained by salinity under 450 µEm–2 s–1. At quantum flux densities of 150 µEm–2 s–1 (400–700 nm) this stimulation was280 per cent. Under high light intensities no stimulation bysalinity was found. In contrast, water stress achieved by directleaf desiccation or by polyethylene glycol inhibited enzymeactivity up to fourfold at –1.2 MPa. Planktonic primary production in the German Wadden Sea   总被引:8,自引:0,他引:8  
By combining weekly data of irradiance, attenuation and chlorophylla concentrations with photosynthesis (P) versus light intensity(E) curve characteristics, the annual cycle of planktonic primaryproduction in the estuarine part of the Northfrisian WaddenSea was computed for a 2 year period. Daily water column particulategross production ranged from 5 to 2200 mg C m–2 day–1and showed a seasonal pattern similar to chlorophyll a. Budgetcalculation yielded annual gross particulate primary productionsof 124 and 176 g C m–2 year–1 in 1995 and 1996,respectively. Annual amounts of phytoplankton respiration, calculatedaccording to a two-compartment model of Langdon [in Li,W.K.W.and Maestrini,S.Y. (eds), Measurement of Primary Productionfrom the Molecular to the Global Scale. International Councilfor the Exploration of the Sea, Copenhagen, 1993, pp. 20–36],and dissolved production in 1996, were both in the range of24–39 g C m–2 year–1. Annual total net productionwas thus very similar to particulate gross production (127 and177 g C m–2 year–1 in 1995 and 1996, respectively).Phytoplankton growth was low or even negative in winter. Inspring and summer, production/biomass (Pr/B) ratios varied from0.2 up to 1.7. Phytoplankton growth during the growth seasonalways surpassed average flushing time in the area, thus underliningthe potential of local phytoplankton bloom development in thispart of the Wadden Sea. The chlorophyll-specific maximum photosyntheticrate (PBmax) ranged from 0.8 to 9.9 mg C mg–1 Chl h–1and was strongly correlated with water temperature (r2 = 0.67).By contrast, there was no clear seasonal cycle in B, which rangedfrom 0.007 to 0.039 mg C mg–1 Chl h–1 (µmolphotons m–2 s–1)–1. Its variability was muchless than PBmax and independent of temperature. The magnitudeand part of the variability of PBmax and B are presumably causedby changes in species composition, as evidenced from the rangeof these parameters found among 10 predominant diatom speciesisolated from the Wadden Sea. Mass spectrometry has been used to investigate the transportof CO2 in the freshwater diatom Navicula pelliculosa. The timecourseof CO2 formation in the dark after addition of 100 mmol m–3dissolved inorganic carbon (DIC) to cell suspensions showedthat no external carbonic anhydrase (CA) was present in thesecells. Upon illumination, cells pre-incubated at pH 75 with100 mmol m–3 DIC, removed almost all free CO2 from themedium at an initial rate of 285 µmol CO2 mg–1Chl h–1. Equilibrium between HCO3 and CO2 in themedium occurred rapidly upon addition of bovine CA, showingthat CO2 depletion resulted from a selective uptake of CO2 ratherthan an uptake of all inorganic carbon species. However, photosyntheticO2 evolution rate remained constant after CO2 had been depletedfrom the medium indicating that photosynthesis is sustainedprimarily by active HCO3 uptake. Treatment of cells with2-iodoacetamide (83 mol m–3) completely inhibited CO2fixation but had little effect on CO2 transport since initialrates of CO2 depletion were about 81% that of untreated cells.Transfer of iodoacetamide-treated cells to the dark caused arapid increase in the CO2 concentration in the medium largelydue to the efflux of the unfixed intracellular DIC pool whichwas found to be about 194 times the concentration of that inthe external medium. These results indicate that Navicula pelliculosaactively takes up molecular CO2 against a concentration gradientby a process distinct from HCO3 transport. Spinach plants (Spinacia oleracea L.) were grown hydroponicallywith different concentrations of nitrate nitrogen, ranging from0.5 to 12 mM, in a glasshouse under full sunlight. Using anopen gas exchange system, the rate of CO2 assimilation, A, wasdetermined as a function of intercellular partial pressure ofCO2, Pi, with a constant amount of absorbed light per unit Chl.When expressed on a leaf area basis, A measured at high irradianceand at pi=500 µbar, was proportional to the in vitro rateof uncoupled whole-chain electron transport as well as to Chlcontent. There was a curvilinear relationship between the mesophyllconductance (the slope of the A : Pi curve near the CO2 compensationpoint) and the in vitro RuBP carboxylase activity. The curvaturedid not appear to be due to enzyme inactivation in vivo in leaveswith high nitrogen contents. The curvature suggested the presenceof a CO2 transfer resistance between the intercellular spacesand the site of carboxylation of 2.2 m2 s bar mol–1 CO2,which is similar to that previously observed in wheat. Thisimplied that, while nitrogen deficiency increased the ratioof in vitro activity of electron transport to that of RuBP carboxylase,the two activities remained balanced in vivo. Irradiance response curves were determined by both net CO2 andO2 exchange. The two methods gave reasonable agreement at lightsaturation. Agrostis capillaris L.5, Festuca vivipara L. and Poaalpina L.were grown in outdoor open-top chambers at either ambient (340 3µmol mol–1) or elevated (6804µmol mol–1)concentrations of atmospheric carbon dioxide (CO2) for periodsfrom 79–189 d. Photosynthetic capacity of source leaves of plants grown atboth ambient and elevated CO2 concentrations was measured atsaturating light and 5% CO2. Dark respiration of leaves wasmeasured using a liquid phase oxygen electrode with the buffersolution in equilibrium with air (21% O2, 0.034% CO2). Photo-syntheticcapacity of P. alpina was reduced by growth at 680 µmolmol–1 CO2 by 105 d, and that of F. vivipara was reducedat 65 d and 189 d after CO2 enrichment began, suggesting down-regulationor acclimation. Dark respiration of successive leaf blades ofall three species was unaltered by growth at 680 relative to340 µmol mol–1 CO2. In F. vivipara, leaf respirationrate was markedly lower at 189 d than at either 0 d or 65 d,irrespective of growth CO2 concentration. There was a significantlylower total non-structural carbohydrate (TNC) concentrationin the leaf blades and leaf sheaths of A. capillaris grown at680µmol mol–1 CO2. TNC of roots of A. capillariswas unaltered by CO2 treatment. TNC concentration was increasedin both leaves and sheaths of P. alpina and F. vivipara after105 d and 65 d growth, respectively. A 4-fold increase in thewater-soluble fraction (fructan) in P. alpina and in all carbohydratefractions in F. vivipara accounted for the increased TNC content. In F. vivipara the relationship between leaf photosyn-theticcapacity and leaf carbohydrate concentration was such that therewas a strong positive correlation between photosynthetic capacityand total leaf N concentration (expressed on a per unit structuraldry weight basis), and total nitrogen concentration of successivemature leaves reduced with time. Multiple regression of leafphotosynthetic capacity upon leaf nitrogen and carbohydrateconcentrations further confirmed that leaf photosynthetic capacitywas mainly determined by leaf N concentration. In P. alpina,leaf photosynthetic capacity was mainly determined by leaf CHOconcentration. Thus there is evidence for down-regulation ofphotosynthetic capacity in P. alpina resulting from increasedcarbohydrate accumulation in source leaves. Leaf dark respiration and total N concentration were positivelycorrelated in P. alpina and F. vivipara. Leaf dark respirationand soluble carbohydrate concentration of source leaves werepositively correlated in A. capillaris. Changes in source leafphotosynthetic capacity and carbohydrate concentration of plantsgrown at ambient or elevated CO2 are discussed in relation toplant growth, nutrient relations and availability of sinks forcarbon. The relationships between CO2 concentrating mechanisms, photosyntheticefficiency and inorganic carbon supply have been investigatedfor the aquatic macrophyte Littorella uniflora. Plants wereobtained from Esthwaite Water or a local reservoir, with thelatter plants transplanted into a range of sediment types toalter CO2 supply around the roots. Free CO2 in sediment-interstitial-waterranged from 1–01 mol m–3 (Esthwaite), 0.79 mol m–3(peat), 0.32 mol m–3 (silt) and 0–17 mol m–3(sand), with plants maintained under PAR of 40 µmol m–2s–1. A comparison of gross morphology of plants maintained underthese conditions showed that the peat-grown plants with highsediment CO2 had larger leaf fresh weight (0–69 g) andtotal surface area (223 cm2 g–1 fr. wt. including lacunalsurface area) than the sand-grown plants (0.21 g and 196 cm2g–1 fr. wt. respectively). Root fresh weights were similarfor all treatments. In contrast, leaf internal CO2 concentration[CO2], was highest in the sand-grown plants (2–69 molm–3, corresponding to 6.5% CO2 in air) and lowest inthe Esthwaite plants (1–08 mol m–3). Expressionof CAM in transplants was also greatest in the low CO2 regime,with H+ (measured as dawn-dusk titratable acidity) of 50µmolg fr. wt., similar to Esthwaite plants in natural sediment.Assuming typical CAM stoichiometry, decarboxylation of malatecould account largely for the measured [CO2]1 and would makea major contribution to daytime CO2 fixation in vivo. A range of leaf sections (0–2, 1–0, 5–0 and17–0 mm) was used to evaluate diffusion limitation andto select a suitable size for comparative studies of photosyntheticO2 evolution. The longer leaf sections (17.0 mm), which weresealed and included the leaf tip, were diffusion-limited witha linear response to incremental addition of CO2 and 1–0mol m–3 exogenous CO2 was required to saturate photosynthesis.Shorter leaf sections were less diffusion-limited, with thegreatest photosynthetic capacity (36 µmol O2 g–1 fr. wt. h–1) obtainedfrom the 1.0 mm size and were not infiltrated by the incubatingmedium. Comparative studies with 1.0 mm sections from plants grown inthe different sediment types revealed that the photosyntheticcapacity of the sand-grown plants was greatest (45 µmolO2 g–1 fr. wt. h–1) with a K0.5 of 80 mmol m–3.In terms of light response, saturation of photosynthesis intissue slices occurred at 850–1000 µmol m–2s–1 although light compensation points (6–11 µmolm–2s–1) and chlorophyll a: b ratios (1.3) were low.While CO2 and PAR responses were obtained using varying numbersof sections with a constant fresh weight, the relationshipsbetween photosynthetic capacity and CO2 supply or PAR were maintainedwhen the data were expressed on a chlorophyll basis. It is concludedthat under low PAR, CO2 concentrating mechanisms interact inintact plants to maintain saturating CO2 levels within leaflacunae, although the responses of the various components ofCO2 supply to PAR require further investigation. To study possible changes in the transport metabolites betweenchloroplasts and cytoplasm during CAM induction of Mesembryanthemumcrystallinum, we compared substrate specificity of P11 translocator(s)in isolated chloroplasts from the C3 and CAM-induced plants.The [14C]glu-cose 6-phosphate (G6P) transport activity was significantonly in the chloroplasts of CAM-mode plants and not detectablein those of C3-mode, while a similar high rate of [32P]Pi uptakewas observed with both types of chloroplasts. Kinetic analysisof G6P uptake in the CAM chloroplasts showed a high Vmax [10.6µmol (mg Chl)–1 h–1] and a comparatively lowKm value (0.41 mM); the latter was similar to Ki values of Pi,3-phosphoglycerate and phospho-enolpyruvate, 0.30, 0.34 and0.47 mM, respectively. On the other hand, [32P]Pi uptake inthe CAM chloroplasts was inhibited competitively by G6P witha Ki value (8.4 mM) 20-fold higher than the Km value for G6Puptake, while that in C3 chloroplasts was not inhibited at all.These results suggest that a new G6P/Pi, counterexchange mechanismis induced in the chloroplast envelope of CAM-induced M. crystallinumin addition to the ordinary type of P, translocator, that cannottransport G6P, already present in the C3-type chloroplasts. Trends in several photosynthetic parameters and their responseto changed growth light were followed for 15 d in leaves ofyoung birch saplings using a rapid-response gas exchange measuringequipment. These in vivo measurements were compared to biochemicalassays that were made from the same leaves after the gas exchangestudies. The measurements were made on leaves that were selectedprior to the study and were at that time of similar age. Forthe first 7 d the photosynthetic parameters were followed fromthe growth conditions of moderate light (200 µmol m–2s–1; referred to as controls later in the text). On day7 some of the saplings were transferred to grow either underhigh (450 µmol m–2 s–1; referred to as highlight plants) or low (75 µmol m–2 s–1; referredto as low light plants) light and the capability of the preselectedleaves for acclimation was followed for 6 d. For comparison,at the end of the experiment the measurements were made on bothcontrols and on young leaves that had developed under high andlow light. Generally the in vivo measured rate of CO2 uptake (gross photosynthesis)both at 310 ppm CO2 and 2000 ppm CO2 corresponded very wellto the biochemically determined CO2 fixation capacity in vitroafter rapid extraction (measured as the initial and total activityof Rubisco, respectively). However, if the flux of CO2 intothe chloroplasts was limited by the closure of the stomata,as was the case of the high light plants, then the in vitromeasured Rubisco activity was greater than the in vivo measuredCO2 uptake. Vmax, calculated from the mesophyll conductanceat 1% O2, exceeded the initial activity of Rubisco (assayedat saturating RuBP and CO2) constantly by 60%. The catalyticactivity of Rubisco in birch leaves was overall very low, evenwhen calculated from the total activity of Rubisco (Kcat 0.63–1.18 s–1), when compared to herbaceous C3 species. Signs of light acclimation were not observed in most of thephotosynthetic parameters and in chloroplast structure whenmature birch leaves were subjected to changes in growth lightfor 6 d. However, the change of the growth light either to highor low light caused day-to-day fluctuations in most of the measuredphotosynthetic parameters and in the case of the high lightplants signs of photoinhibition and photodestruction were alsoobserved (decrease in the amount of chlorophyll and increasein chlorophyll a/b ratio). As a result of these fluctuationsthese plants achieved a new and lower steady-state conditionbetween the light and dark reactions, as judged from the molarratio of RuBP to Rubisco binding site.
