The occurrence of mountain hare mitochondrial DNA in wild brown hares   总被引:4,自引:0,他引:4  
If interspecific hybrids are fertile and backcross to either parental species, transmission of mitochondrial DNA over the species barrier can occur. To investigate if such transmission has occurred between the brown hare Lepus europeus Pall and the mountain hare L. timidus L. in Scandinavia, an analysis of genetic variation in mitochondrial DNA from 36 hares, collected from 15 localities, was performed. Sequence divergence of mtDNA between species was estimated at 8 ± 1% (SD). Intraspecific mtDNA sequence divergence varied between 0.09 and 0.38% in brown hares and 0.10 and 1.44% in mountain hares. In six out of 18 brown hares examined, two different haplotypes of mountain hare origin were detected, demonstrating a transmission of mtDNA haplotypes from mountain hares to brown hares. The results indicate that interspecific hybridization between the two species occurs in wild populations.  相似文献

Sequence variation of the 5′ end of the mitochondrial DNA control region (600 bp) was examined in the population samples of Eurasian wigeon Anas penelope from Anadyr’ and Primorye. A total of 11 different mtDNA haplotypes were identified, with one of these belonging to American wigeon Anas americana. The presence of the mtDNA haplotype from the species closely relative to A. penelope in the Anadyr’ sample can be considered as the genetic evidence in favor of interspecific hybridization. This suggestion is in the good agreement with ornithological data. Genetic differentiation of the Primorye and Anadyr’ populations was low (Φst = 0.096). The phylogeographic structure was not pronounced.  相似文献

Dung counts are frequently employed to infer abundance of antelope species in African forests, but the accuracy of dung identification has rarely been tested. We used non-invasive genetic methods to test the accuracy of both field identification and morphometrics for identifying dung samples collected in the Udzungwa Mountains, Tanzania. Species identity was established by sequencing part of the mitochondrial control region from faecal DNA. Field identification was found to be correct in only 58–76% of cases depending on the observer. Discriminant analysis of dung pellet length correctly classified 80% of samples but a larger reference sample size is needed before using this method to classify dung of unknown origin. The results of this study illustrate the potential inaccuracy of dung counts as a monitoring tool for sympatric forest antelope species when the probability of correct identification is unknown. We recommend molecular testing of species identity during forest antelope surveys before conclusions are drawn on the basis of other identification methods.  相似文献

The carnivore community of the altiplano ecosystem of the high Andes, including the Andean mountain cat (Leopardus jacobita) and pampas cat (Leopardus colocolo), is one of the least studied in the world. We determined the origin of 186 carnivore samples (184 faeces and two skulls) collected above 3000 m above sea level in northern Chile, including 33 from the Andean mountain cat and 75 from the pampas cat using diagnostic molecular genetic sequence variation. We determined for the first time food habits, habitat and physiographic associations, and general patterns of molecular genetic variation of the Andean mountain cat and the pampas cat in Chile. Both species had narrow dietary niches dominated by small rodents and there was a wide overlap in diet composition (0.82), suggesting low levels of prey partitioning between species. The mountain viscacha (Lagidium viscacia) made up a large proportion of the biomass of the diet of both species, especially for the Andean mountain cat (93.9% vs. 74.8% for the pampas cat), underscoring the importance of further research and conservation focus on this vanishing prey species. Although the probability of finding Andean mountain cat scats increased with altitude and slope, there was substantial geographical overlap in distribution between species, revealing that the pampas cat distribution includes high-altitude grassland habitats. The Andean mountain cat had relatively low levels of mitochondrial DNA (mtDNA) genetic variation (two mtDNA haplotypes) compared with the pampas cat (17 mtDNA haplotypes), suggestive of a distinct evolutionary history and relatively smaller historic populations. These insights will facilitate and provide tools and hypotheses for much-needed research and conservation efforts on these species and this ecosystem.  相似文献

Little is known about genetic variation in the 6–8 subspecies of Colobus angolensis, currently distinguished by pelage differences. We present a comparative genetic analysis of one of these subspecies, C. a. palliatus, in Kenya and Tanzania that assesses evolutionary relationships and patterns of mitochondrial genetic diversity in 103 individuals across its geographic range. Fecal samples from approximately 156 individuals were collected in four localities: (1) Diani Forest, Kenya; (2) Shimoni, Kenya; (3) Udzungwa Mountains National Park, Eastern Arc Mountains, Tanzania; and (4) Mount Rungwe, Southern Highlands, Tanzania. These samples represent at least six groups, with 5–15 samples from each. Comparative sequence analysis of a 1,795 base pair mtDNA fragment revealed 19 unique haplotypes in four populations. Phylogenetic analyses suggest that sampled Kenyan haplotypes are paraphyletic, with one Kenyan haplotype basal to all other sampled haplotypes. Analysis of molecular variance (AMOVA) suggests high levels of genetic variation among populations (ΦST 0.72, P<0.001). Genetic data are concordant with a subspecies level differentiation between C. a. palliatus populations in Kenya and those in Central and southern Tanzania, as earlier suggested based on pelage differences. This study highlights the evolutionary distinctiveness of Kenyan populations of C. a. palliatus relative to Tanzanian populations. Although C. a. palliatus habitat in Tanzania is currently better protected than in Kenya, our results suggest Kenyan and Tanzanian populations should be considered distinct units, and the protection of C. a. palliatus habitat in Kenya, as well as habitat connectivity between Kenyan populations, should be prioritized for conservation and management. 该研究采用非损伤性DNA基因分型技术,对可可西里地区10个藏羚羊(Pantholops hodgsonii)个体的mtDNA非编码区部分片段(444~446bp)进行了序列分析,结果显示A、T%含量(61.8%)明显高于G、C%含量(38.2%),共发现10种单倍型,包括48个多态位点,其中转换位点44个、颠换位点1个、插入位点1个、缺失位点2个。单倍型间平均遗传距离为0.031,单倍型多态性(h)为1.000,核苷酸多态性(π)为2.96%。说明藏羚羊线粒体控制区存在着丰富变异,最后从藏羚羊的生态习性及地理分布两方面对这一结果进行了分析探讨。   相似文献

Heliocopris dilloni is the largest dung-beetle found in the Tsavo National Park, Kenya, which during the biannual rainy seasons comprises the most important agency of elephant dung removal. The species is shown to exhibit considerable morphological variation. The mode of construction of the brood chamber is described together with details of development from egg to pupa. It is suggested that the depth to which these beetles burrow is related to avoiding the extreme climate near the surface and to predator avoidance.  相似文献

Night counts of hares and other animals were made on a disused airstrip in the Queen Elizabeth National Park, Uganda and on the road between Magadi and Nairobi in Kenya. Lepus crawshayi is the only species of hare which occurs in the Queen Elizabeth Park but L. capensis is also found in Kenya. No regular seasonal fluctuations were found in the number of hares recorded due probably to the continuous breeding of these equatorial species. The number of waterbuck seen in Uganda on moonlit nights was significantly greater than that counted on dark nights. This is not thought to be due to better viewing conditions under a full moon since the phenomenon was not found with other species. The number of hares recorded varied inversely with the number of other mammals present but bore no relationship with the number of birds. It is concluded that night counts can be useful in revealing population trends but not for assessing total numbers or for comparing densities in different areas.  相似文献

We examined sequence variation in the mitochondrial cytochrome b and NADH dehydrogenase subunit 5 genes (2,360 bp total) for 26 lions from eleven locations throughout sub-Saharan Africa. Six distinct haplotypes were observed in the combined sequences, forming two clades: the eastern and the western savannas. The Uganda-Western Kenya haplotype grouped at a basal position with the eastern clade of lions from Tsavo south to the Transvaal and Natal regions. The phylogenetic position of the haplotype from Sabi Sands in the southern part of Kruger National Park remained poorly resolved. The haplotypes found in Namibia and Botswana formed the western clade. The modest genetic variation documented here argues against taxonomic distinctions among living African lions.  相似文献

This study investigated composition and selectivity in diet for waterbuck in the Pendjari National Park in north-western Benin, through the use of micrographic analysis of faecal samples. Three plant species ( Panicum anabaptistum, Echinochloa stagnina and Andropogon gayanus ) were regularly consumed all year round. Meanwhile, three other species (i.e., Hyparrhenia involucrata, Acroceras amplectens and Oryza barthii ) are mostly found in its diet during the beginning of the rainy season. During the dry season, long life grasses (>40%) and tree forage (about 35%) were the most dominant life form in the diet. On the contrary at the beginning of the rainy season, annual species (> 50%) were dominant. In conclusion, the waterbuck has a grazer regime when plant species are abundant and a mixed diet during the dry season. Waterbuck's food niche breath, defined by Hespenheide [ Ecology and Evolution of communities . Harvard Univ. Press, 1975], was lower than 1, implying this antelope does not eat all food categories in a proportional way. Shannon diversity index showed that the diet was more diversified during the rainy season and less diversified at the end of the dry season. Based on [ Ecology , 64 (1983), 1297] diet selectivity index, waterbuck exerted a positive selection on the major graminaceous species.  相似文献

The occurrence of coprophilous fungi on samples of antelope, buffalo, zebra and hippopotamus dung collected from the Marula Estates in Kenya was recorded. A hundred and seventy three fungal isolates representative of 40 genera and 59 species were identified. Among these species, 23 were Ascomycetes (39%), 30 Deuteromycetes (50.8%), 5 Zygomycetes (8.5%) and 1 Basidiomycete (1.7%). Ascobolus immersus, Coprotus niveus, Iodophanus carneus, Lasiobolus lasioboloides, Podospora anserina, P. australis and Sporormiella minima were the dominant species occurring in the dung of these animals. Notes on infrequent or interesting Ascomycetes include Kernia nitida, Saccobolus versicolor, Sordaria fimicola and others. The interdependence of the vegetation of this area with the fungi occurring on the dung of these animals is discussed. Land use change and human population growth are accelerating the fragmentation and insularization of wildlife habitats worldwide. The conservation and management of wildlife in the resultant ‘island’ ecosystems in the context of global warming is challenging due to the isolation and reduced size of the ecosystems and hence the scale over which ecosystem processes can operate. We analyzed trends in numbers of nine large herbivores in Kenya’s Lake Nakuru National Park to understand how rainfall and temperature variability, surrounding land use changes, and boundary fencing affected wildlife population dynamics inside the park during 1970–2011. Buffalo, zebra and Thomson’s gazelle numbers increased persistently. Grant’s gazelle and impala increased initially then gradually declined. Waterbuck and warthog numbers progressively declined to levels that potentially threatened their local population persistence. The total biomass of ungulates tripled from 1970 to 2011, with buffalo replacing waterbuck as the predominant species in biomass. Increased competition from buffalo and zebra, heightened predation and illicit human harvests probably all contributed to the declines by waterbuck and warthog. Density-dependent limitation of population growth within the park confines was evident for buffalo, impala, eland, giraffe, Grant’s and Thomson’s gazelles. Fluctuations in the lake level related to varying rainfall affected changes in animal abundance through expansion of the lake area and flooding of grasslands bordering the lake. Unusually, the most stressful conditions were associated with high water levels following high rainfall. There was also evidence of carry-over effects from prior habitat conditions affecting all species. The relatively stable populations of all species except warthog and waterbuck demonstrate the remarkable capacity of this small, insularized park to retain viable populations of most of the large herbivores, without much management intervention.  相似文献

Patterns of mitochondrial DNA (mtDNA) variation were examined in 133 mole-rats constituting all four chromosomal species (2n = 52, 2n = 54, 2n = 58, and 2n = 60) of the Spalax ehrenbergi superspecies in Israel, as well as the peripheral isolates of 2n = 60. In the main range of the complex, a total of 28 mtDNA haplotypes were found in 64 mole-rats, with most haplotypes being unique to either a single chromosomal species or population. mtDNA divergence increased from low to high diploid number in a north-to-south direction in Israel. Overall levels of mtDNA diversity were unexpectedly the highest in the 2n = 60, the youngest species of the complex. The mtDNA haplotypes can be separated into two major groups, 2n = 52-54 and 2n = 58-60, and a phylogenetic analysis for each group revealed evidence of a few haplotypes not sorted by diploid number. The overall patterns of mtDNA divergence seen within and among the four chromosomal species are consistent with the parapatric mode of speciation as suggested from previous studies of allozyme and DNA hybridization. In a separate data set the patterns of mtDNA variation were examined across the main geographic range and across peripheral semi-isolates and isolates of the 2n = 60 chromosomal species. Fifteen haplotypes were found in 69 mole-rats. High levels of mtDNA diversity characterized the main range, semi-isolated, and even some desert isolated populations. The peripheral isolates contain much mtDNA diversity, including novel haplotypes.   相似文献

Bryde’s whales (Balaenoptera brydei) differ from other typical baleen whale species because they are restricted to tropical and warm temperate waters in major oceans, and frequent trans-equatorial movement has been suggested for the species. We tested this hypothesis by analyzing genetic variation at 17 microsatellite loci (N = 508) and 299 bp of mitochondrial DNA (mtDNA) control region sequences (N = 472) in individuals obtained from the western North Pacific, South Pacific, and eastern Indian Ocean. Combined use of microsatellite and mtDNA markers allowed us to distinguish between contemporary gene flow and ancestral polymorphism and to describe sex-specific philopatry. A high level of genetic diversity was found within the samples. Both nuclear and mtDNA markers displayed similar population structure, indicating a lack of sex-specific philopatry. Spatial structuring was detected using both frequency-based population parameters and individual-based Bayesian approaches. Whales in the samples from different oceanic regions came from genetically distinct populations with evidence of limited gene flow. We observed low mtDNA sequence divergence among populations and a lack of concordance between geographic and phylogenetic position of mtDNA haplotypes, suggesting recent separation of populations rather than frequent trans-equatorial and inter-oceanic movement. We conclude that current gene flow between Bryde’s whale populations is low and that effective management actions should treat them as separate entities to ensure continued existence of the species.  相似文献

Channa marulius (Hamilton, 1822) is a commercially important freshwater fish and a potential candidate species for aquaculture. The present study evaluated partial Cytochrome b gene sequence of mtDNA for determining the genetic variation in wild populations of C. marulius. Genomic DNA extracted from C. marulius samples (n = 23) belonging to 3 distant rivers; Mahanadi, Teesta and Yamuna was analyzed. Sequencing of 307 bp Cytochrome b mtDNA fragment revealed the presence of 5 haplotypes with haplotype diversity value of 0.763 and nucleotide diversity value of 0.0128. Single population specific haplotype was observed in Mahanadi and Yamuna samples and 3 haplotypes in Teesta samples. The analysis of data demonstrated the suitability of partial Cytochrome b sequence in determining the genetic diversity in C. marulius population.  相似文献

Patterns of genetic structure in eusocial naked mole-rat populations were quantified within and among geographically distant populations using multilocus DNA fingerprinting and mitochondrial DNA (mtDNA) sequence analysis. Individuals within colonies were genetically almost monomorphic, sharing the same mtDNA control region haplotype and having coefficients of band sharing estimated from DNA fingerprints ranging from 0.93 to 0.99. Family analysis of a hybrid captive colony of naked mole-rats with increased levels of genetic variability using multilocus DNA fingerprinting gave results consistent with Mendelian inheritance, and has revealed for the first time that multiple paternity can occur. In a survey of wild colonies from Ethiopia, Somalia and locations in northern and southern Kenya, we have examined mtDNA control region sequence variation in 42 individuals from 15 colonies, and together with multilocus DNA fingerprinting and mtDNA cytochrome- b sequence analysis in selected individuals have shown that these populations show considerable genetic divergence. Most of the variance in sequence divergence was found to be between geographical locations (Φct= 0.68) and there was a significant correlation between sequence divergence and geographical separation of haplotypes. Six colonies from Mtito Andei in southern Kenya shared the same control region haplotype, suggesting a recent common maternal ancestor. In contrast, out of four colonies at Lerata in north Kenya, three haplotypes were identified, and phylogenetic analysis suggests that this area may be a zone where two distinct lineages are in close proximity. Genetic distances were maximal between Ethiopian and southern Kenyan populations at 5.8% for cytochrome- b , and are approaching interspecific values seen between other Bathyergids.  相似文献

We studied the feeding ecology of the bongo (Tragelaphus eurycerus) in the Dzanga National Park in the rainforest of the Central African Republic to understand better the trade-off between food selectivity, ranging behaviour and social organization of a large, forest-dwelling, social antelope. Food plants and vegetation types were registered along a 311-km route travelled by bongos. Food availability was determined by identifying and counting the plants in 19 randomly chosen forest plots. Bongos showed pronounced selectivity for 26 out of 100 woody forest species. They predominantly consumed younger leaves, which suggests that high protein and low fibre content influence plant choice. In addition to leaves, bongos also ate fruits of two and flowers of one species. Furthermore, the diet was supplemented by grasses and herbs consumed on large natural licks. Such licks were regularly visited by the bongos. According to Jarman's ecological classification of antelopes, selective browsers are relatively small and live alone or in pairs to avoid competition over food. The bongo's large size and gregariousness should not allow it to survive in the rainforest as a pure selective browser. Our data suggest that the bongo relies on the opportunity to graze in bulk which it finds on the natural licks. We hypothesize that such licks either limit the distribution of bongos in other rainforest areas or allow larger group sizes than in areas without licks. Patterns of social grouping in ungulates of Tsavo National Park, Kenya   总被引:1,自引:0,他引:1  
The paper presents data on group size frequencies and mean group sizes in 11 species of ungulates and, for seven of them, more detailed information on the composition of groups in Tsavo East National Park, Kenya. In several species, mean group size was higher in a part of the study area with more open vegetation. No significant seasonal variations in mean group size were found, except in waterbuck which concentrated markedly in one location during the dry season. Minor local or seasonal variations in the proportions and/or sizes of different group types were evident in most species. In general, grouping patterns were consistent with the type of social organization found in other areas for the species concerned. Frequencies and types of interspecific associations, and the possible factors underlying their formation, are also discussed.  相似文献

This paper reports the use of molecular methods to characterize the coprophilous fungal communities (CFC) that inhabit the dung of four species of mammalian herbivores at two sites, Sevilleta National Wildlife Refuge (SNWR) in New Mexico and Wind Cave National Park (WCNP) in South Dakota. Results reveal that CFC from domesticated cattle (Bos taurus) at SNWR, and bison (Bison bison) and black-tailed prairie dogs (Cynomys ludovicianus) at WCNP were diverse but dominated primarily by members within eight taxonomic orders, including the rarely cultured and anaerobic order Neocallimastigales. In addition, 7.7% (138 of 1,788) of the sequences obtained from all dung samples were at least 97% similar to root-associated fungal (RAF) sequences previously described from blue grama (Bouteloua gracilis), a common forage grass found throughout North America and growing at both study sites. In contrast, 95.8% (295 of 308) of the sequences and four of the total seven operational taxonomic units obtained from pronghorn antelope (Antilocapra americana) dung belonged to the Pleosporalean order. We hypothesize that some herbivore vectors disperse non-systemic (non-clavicipitaceous) fungal endophytes. These dispersal events, it is argued, are most likely to occur via herbivores that occasionally forage and masticate root tissue, especially in arid regions where aboveground vegetation is sparse. The results of this study suggest that some (possibly many) members of the RAF community can expand their ecological role to include colonizing dung.  相似文献
