Summary NAAG is one of the neuropeptides found in highest concentrations in the CNS. The presence of micromolar concentrations of NAAG in human CSF was demonstrated by using two different and complementary analytical approaches: 1) isocratic separation of endogenous NAAG by reverse-phase high performance liquid chromatography (HPLC) with dual wavelength detection and 2) derivatization of endogenous NAAG with acidic methanol and subsequent HPLC analysis of the derivative NAAG-trimethyl ester. The NAAG concentration was between 0.44µmol/l and 7.16µmol/l (mean of 2.19 ± 1.53µmol/l) in CSF samples from forty neuropsychiatric patients. Endogenous NAAG or [3H]NAAG added to CSF samples were not significantly degraded when the CSF was incubated at 37°C during one hour, suggesting that the peptide is a highly stable metabolite in the subarachnoid space. In addition, evidence is provided that NAAG does not present a concentration gradient along the lower subarachnoid space.  相似文献

Summary The heat resistance ofCitrobacter freundii NCTC 9750 between 45–65°C in media with various water activities has been determined.At a water activity of nearly 1.00, the Arrhenius plot of the death rate shows a sharp breakpoint at 56.5°C, suggesting the existence of at least two different thermal inactivation processes causing lethality of the bacterial cell. The activation energy below 56.5°C is 0.4186 MJ/mol (100 000 cal/mol), above 56.5°C it is 0.1863 MJ/mol (44 500 cal/mol). After addition of sucrose (1.8 mol/l) or NaCl (0.77 mol/l) to the heating medium, such a breakpoint is not observed. The activation energy for these processes are, for sucrose; 0.2097 MJ/mol, for NaCl; 0.3641 MJ/mol. However, at an NaCl concentration of 1.54 mol/l there is a breakpoint at 53.3°C.The influence of the sucrose concentration on the heat resistance can be described by the formula: ln kS=ln kOa [sucrose]. Such a simple correlation does not exist for the influence of NaCl or glycerol.The heat inactivation of whole cells ofC. freundii was also measured with a differential scanning calorimeter. One year old, individually tagged Lake Inari Arctic charr, Salvelinus alpinus, were reared at three constant temperatures, 10.3°C, 14.1°C and 18.1°C, over four weeks. Blood samples were collected from a group of unstressed fish after the cultivation period at the same time as another group of fish were subjected to acute handling stress treatment (2min netting in air and 40min (± 20min) recovery period in water). Plasma cortisol, calcium, sodium, potassium and chloride concentrations were measured on both groups. To study the effect of minor daily temperature fluctuations on the stress response of Arctic charr, two additional daily fluctuating temperature (14 ± 1°C, 18 ± 1°C) treatments were established. The samples were taken in the same manner as those in the constant temperature treatments. Growth was fastest at 10.3–14.1°C and clearly lower at 18.1°C. Pre-stress plasma cortisol levels were low but increased slightly with increasing temperature. After stressor treatment, the cortisol concentrations of Arctic charr were clearly higher in all temperature treatments but there were no significant differences in plasma cortisol concentrations among temperatures. Plasma calcium levels increased during the stress treatment but temperature did not modulate this effect. The plasma potassium concentrations declined at 14.1–18.1°C after acute stress but the response was not affected by temperature within this range. The concentrations of sodium and chloride were unaffected by acute stress. Temperatures of 10.3–18.1°C and fluctuating temperature treatments had no influence on any plasma ion concentrations. Arctic charr were able to maintain the plasma ion concentrations in fresh water at 10.3–18.1°C and after acute stress treatment. Results indicate that the optimum temperature for growth of Arctic charr has little to do with the plasma ion concentrations or the ability to maintain those concentrations after short-term stress. The plasma cortisol responses further indicate that the optimum temperature for growth of Arctic charr is not related to the suppressed ability to react to an acute handling stressor. Temperature fluctuations did not cause significant differences in cortisol levels when compared with constant temperatures.  相似文献

The CO2 production of individual larvae of Apis mellifera carnica, which were incubated within their cells at a natural air humidity of 60–80%, was determined by an open-flow gas analyzer in relation to larval age and ambient temperature. In larvae incubated at 34 °C the amount of CO2 produced appeared to fall only moderately from 3.89±1.57 µl mg–1 h–1 in 0.5-day-old larvae to 2.98±0.57 µl mg–1 h–1 in 3.5-day-old larvae. The decline was steeper up to an age of 5.5 days (0.95±1.15 µl mg–1 h–1). Our measurements show that the respiration and energy turnover of larvae younger than about 80 h is considerably lower (up to 35%) than expected from extrapolations of data determined in older larvae. The temperature dependency of CO2 production was determined in 3.5-day-old larvae, which were incubated at temperatures varying from 18 to 38 °C in steps of 4 °C. The larvae generated 0.48±0.03 µl mg–1 h–1 CO2 at 18 °C, and 3.97±0.50 µl mg–1 h–1 CO2 at 38 °C. The temperature-dependent respiration rate was fitted to a logistic curve. We found that the inflection point of this curve (32.5 °C) is below the normal brood nest temperature (33–36 °C). The average Q10 was 3.13, which is higher than in freshly emerged resting honeybees but similar to adult bees. This strong temperature dependency enables the bees to speed up brood development by achieving high temperatures. To determine the contribution of food ingestion (trophic pathway) to PCB contamination of zooplankton in the river Meuse (Belgium), we used 14C-labelled algae (Dictyosphaerium ehrenbergianum) to measure ingestion and assimilation rates in the rotifer species Brachionus calyciflorus. When the concentration of algae in the culture medium varied from 20 103 to 200 103 algal cells ml–1 (0.12 to 1.18 mg Cl–1), the Brachionus calyciflorus ingestion rate varied from 0.25 ± 0.12 to 1.52 ± 0.43 ng C ind–1 h–1 at 15 °C and from 0.74 ± 0.17 to 5.93 ± 0.61 ng C ind–1 h–1 at 20 °C. The assimilation efficiency (ratio of the assimilation rate to the ingestion rate) measured in a culture medium containing 200 103 algal cells ml–1 was 55.7 ± 5.8%. Since the PCB concentration measured in the phytoplankton of the river Meuse is about 3 µg PCBs g–1 D.W., the estimated PCB contamination of zooplankton ascribable to the trophic pathway ranges from 0.22 ± 0.17 to 1.31 ± 0.77 µg PCBs g–1 D.W. at 15 °C and from 0.64 ± 0.34 to 5.10 ± 2.10 µg PCBs g–1 D.W. at 20°C. The lower figure based on measurements effected at 20 °C is comparable to the actual level measured in zooplankton samples collected in the river Meuse (0.69 ± 0.20 µg PCBs g–1 D.W.). The applicability of the formula used in our estimate was checked in a 48-hour in vitro experiment in which the rotifers were fed contaminated algae. The PCB accumulation measured in the rotifers was found to coincide with the calculated PCB contamination. Additional experiments were carried out to determine the contribution of the direct pathway to PCB contamination of zooplankton living in the river Meuse (0.02 µg PCBs l–1 of water; average dissolved organic matter: 3 mg C 1–1). The PCB concentration in zooplankton resulting from direct uptake of PCBs from the water was estimated at 0.19 ± 0.05 µg PCBs g–1 D.W. These results show that in zooplankton living in polluted ecosystems, PCBs are likely to accumulate via the trophic pathway to concentrations up to 30 times higher than by direct contamination. Furthermore, our estimates of PCB contamination via the trophic pathway coincide quite well with actual concentrations measured in situ.  相似文献

Summary The effects of microwave radiation in reducing decalcification time were evaluated by measuring rates of calcium removal from samples of rat and cat teeth in 0.1 mol l–1 EDTA. In some cases, 3% glutaraldehyde was added to the decalcifying solution. Test specimens were placed in a microwave oven at 39±2°C for repeated periods of 1–2h. Control specimens were placed in a coventional oven at 39°C for the same times or held at room temperature. The calcium removed during each treatment was measured using an atomic absorption spectrophotometer. There was no consistent difference between the results obtained with microwave radiation as compared with heating in a conventional oven, although in both cases decalcification was slightly faster than at room temperature. These results are attributed to thermal effects. No evidence for non-thermal effects of micro-radiation was found.  相似文献

Field observations on temperature and pH of a small pond showed that a amphipod population of Hyalella azteca was exposed to variable seasonal pH between 5.10–5.85, and water temperatures between 2–21 °C. Laboratory experiments were designed to simulate seasonal temperatures and field pHs of a small pond habitat. Laboratory bioassay experiments were conducted to determine the survival of Hyalella azteca at pHs 4, 5, 6 and 7, and varying temperatures of 5°, 10°, 15°, 20° and 25 °C.The LT100 at pH 4 and 25 °C was 5.7 ± 0.47 days, compared to 47.3 ± 2.49 days at 5 °C. An Analysis of Variance (ANOVA) showed temperature was a significant (p > 0.0001) source of variation in the acute lethality of pH to H. azteca. A Duncans Multiple Range Test (DMRT) further showed that in laboratory experiments at pH 4, there was a significant difference ( = 0.01) between the LT100s at 5°, 10°, 15° and 20 °C, but not between temperatures 20° and 25 °C.  相似文献

In vitro cultures of Nephrolepis exaltata and Cordyline fruticosa were stored at 5°, 9° or 13°C, at a low irradiance (3–5 mol m–2 s–1) or in darkness. Prior to storage the cultures were subjected to 18°, 21°, 24° or 27°C and 15, 30 or 45 mol m–2 s–1 in a factorial combination.The optimal storage conditions for Nephrolepis were 9°C in complete darkness. These cultures were still transferable to a peat/perlite mixture at the end of the experimental period of 36 months.The optimal storage conditions for Cordyline were 13°C and a low light level (±3–5 mol m-2 s-1). When the pre-storage conditions were normal growth room conditions (24°C and 30 mol m-2 s-1), in vitro cultures could be stored for 18 months. With the most favourable pre-storage treatment (18°C and 15 mol m-2 s-1) some cultures still had green shoots after 36 months of storage, but did not survive transfer to peat/perlite.Pre-conditioning before storage was most favourable for Nephrolepis, and not that important, but still favourable, for Cordyline. There was an interaction between pre-storage temperature and pre-storage irradiance. Summary The present study was performed with the aim to determine the kinetics and the caracteristics of cellular uptake of L-3,4-dihydroxyphenylalanine (L-DOPA) in rat renal proximal tubules. Incubation of renal tubules at 4°C in the presence of increasing concentrations of L-DOPA results in a linear and concentration-dependent accumulation of the substrate. In experiments carried out at 37°C, the accumulation of L-DOPA in renal tubules was found to be greater than that occurring at 4°C and showed a trend for saturation. The saturable component of L-DOPA uptake was derived from the total amount of L-DOPA accumulated in renal tubules at 37°C subtracted with the values obtained in experiments conducted at 4°C. The Vmax and Km values for the saturable component of L-DOPA uptake in renal tubules were, respectively, 241 ± 32 fmol µg protein–1min–1 and 567 ± 63 µM. Cyanine 863 (5 and 10 µM) was found to decrease the tubular uptake of L-DOPA, whereas probenecid (50 µM) did not change the rate of uptake of L-DOPA into renal tubules. The Vmax and Km values for the saturable component of L-DOPA uptake in renal tubules incubated in the presence of 10 µM cyanine 863 were, respectively, 97 ± 11 fmol µg protein–1min–1 and 160 ± 22 µM. It is suggested that the anionic L-DOPA may behave as an amphoteric substance, both hydroxyl groups in the aromatic ring determining the binding of the molecule to the organic cation transporter.  相似文献

The life cycle of Laminaria abyssalis (Laminariales,Phaeophyta) in culture   总被引:2,自引:2,他引:0  
Laminaria abyssalis occurs in deep water in tropical latitudes of the Brazilian coast (19° 23 S, 38° 28 W to 22° 54 S, 42° 13 09 W). Its life cycle has been completed in the laboratory in seven months using different conditions of light and temperature. The gametophytic stage required for growth the low photon flux density of 1.2 ± 0.3 µmol m–2 s–1 and 18 °C, while the juvenile and adult sporophytes needed 15 µmol m–2 s–1 and 18 °C. The sporophytes became fertile at 23 °C. Our results showed that light and temperature are the main factors regulating the growth and life history of this species under the culture conditions tested.  相似文献

Sugar cane bagasse hemicellulosic fraction was hydrolysed by treatment with 70 mg of sulphuric acid per gram of dry mass at 125 °C for 2 h. The hydrolysate was used as the substrate to grow Candida langeronii RLJ Y-019 at 42 °C; initial pH 6.0; stirring at 700 rev/min and aeration at 1.0 and 2.0 v/v/min. The utilization of D-xylose, L-arabinose, and acetic acid were delayed due to the presence of D-glucose, but after D-glucose depletion the other carbon sources were utilized. The kinetic parameters calculated for both cultivations at 1.0 and 2.0 v/v/min included: maximum specific growth rate (max) of 0.29 ± 0.01 h–1 and 0.43 ± 0.016 h–1, yields (Y x/s) of 0.36 ± 0.012 and 0.40 ± 0.012 gx/gs and productivity (Q x) of 0.81 ± 0.016 and 0.97 ± 0.012 gx/l/h, respectively, and compared favourably with published results obtained with Candida utilis and Geotrichum candidum. Candida langeronii appeared superior to C. utilis for biomass production from hemicellulose hydrolysate, in that it utilized L-arabinose and was capable of growth at higher temperatures. The biomass contained 48.2, 1.4, 5.8 and 23.4% of total protein, DNA, RNA and carbohydrate, respectively and contained essential amino acids for animal feed.  相似文献

Summary A 25-l scale protocol is devised for the optimal secretion and recovery of fungal cellulase. Using a selected higher yieldingTrichoderma viride SMC strain, a protocol consisted of: a) an optimized production medium rich in microcrystalline cellulose (MCC), fortified with 1% (w/v) ammonium sulphate, 0.5% (w/v) soybean flour, 0.1% (v/v) Tween-80 and other trace nutrients; b) optimized physical parameters of production, such as an inoculum containing a homogeneous suspension of 6×107 conidia per 1,28±1°C, pH 4.0±0.5, 300±20 rpm, 11000±1000 l/h aeration, and 170–220 h duration; c) optimal recovery through a filter press (450 l/h rate of filtration) followed by precipitation with 2.5–3.0 volumes of acetone (15°C and basket centrifugation (27°C, 1700 rpm)); and d) vacuum drying (35°C, 4–6 h). This afforded 70% recovery of cellulase in the form of white fluffy powder containing 20000±2000 carboxy methyl cellulase and 1000±50 units filter paperase per g activities, with raw material cost of US$ 8–10 per million carboxy methyl cellulase units. During storage for 18 months at 4°C, ambient temperature and 37°C, the cellulase preparation was found to retain 100, 75 and 60% of its initial activity, respectively.  相似文献

The effect of thermal acclimation for 15 days on the gametogenesis of Poecilobdella viridis was studied. Leeches conditioned at laboratory temperature (25.5°–27.5°C), when warm acclimated (32.0° ± 0.5° C), displayed a significant (p < 0.05) increase in the percentage of oocytes having a diameter of 12.5 µ and 15.0 µ and in the percentage of fully mature sperm clusters over the controls, whereas cold acclimation (10.0° ± 0.5°C) did not induce significant (p > 0.05) changes in the oocytes and sperm clusters. Thus it is concluded that in P. viridis temperature is of paramount importance in initiating gametogenesis.  相似文献

Previously we have shown that human red blood cells (RBCs) undergo a sudden change from blocking to passing through a 1.3±0.2-µm micropipette when applying an aspiration pressure of 2.3 kPa at a critical transition temperature (Tc=36.4±0.3 °C). Low-shear viscosity measurements suggested that changes in the molecular properties of hemoglobin might be responsible for this effect. To evaluate structural changes in hemoglobin at the critical temperature, we have used circular dichroism (CD) spectroscopy. The thermal denaturation curves of human hemoglobin A (HbA) and hemoglobin S (HbS) upon heating between 25 and 60 °C were non-linear and showed accelerated denaturation between 35 and 39 °C with a midpoint at 37.2±0.6 °C. The transition was reversible below 39 °C and independent of solution pH (pH 6.8–7.8). It was also independent of the oxygenation state of hemoglobin, since a sample that was extensively deoxygenated with N2 showed a similar transition by CD. These findings suggest that a structural change in hemoglobin may enable the cellular passage phenomenon as well as the temperature-dependent decrease in viscosity of RBC solutions.  相似文献

Summary In order to analyse hydroxyproline (HYP) in urine, a high-performance liquid chromatographic method was modified. The primary amino groups were blocked with o-phthalaldehyde, and then the secondary amino groups were derivatized with 4-dimethylaminoazobenzene-4-sulphonyl chloride. In addition, the dabsylated samples were treated with ethyl acetate to obtain a simple elution profile in high-performance liquid chromatography. The dabsyl-HYP and -proline were eluted at 4.7 min and 8.0 min, respectively. The chromatographic analysis was completed within 10 min, including the time needed for reequilibration of the column. Using the present method, the concentration of HYP in urine was determined to 260 ± 6µmol/l.  相似文献

The aim of this study was to investigate if voluntary activation and force variability during maximal voluntary contraction (MVC) depends more on muscle (local) or body (core) temperature. Ten volunteers performed a 2-min MVC of the knee extensors under the control (CON) conditions (ambient temperature (21 °C), relative humidity (30%), and air velocity (∼0.1 m/s)) as well as after heating (HT) and cooling (CL) of the lower body. During water manipulation procedure lower body was immersed up to the waist in a water bath at ∼44 °C for 45 min for HT experiment, and ∼15 °C for 30 min for CL experiment. Peak torque, torque variability, muscle voluntary activation and half-relaxation time were assessed during the exercise. HT increased muscle (2.8±0.2 °C) and rectal (1.9±0.1 °C) temperatures while CL lowered muscle (2.2±0.2 °C) temperature, but did not affect rectal temperature. During 2-min MVC, peak torque decreased (P<0.05; SP>90%) and to a lower level in HT compared to CON and CL experiments (52.6±2.3% versus 69.0±2.3% and 65.6±1.9% MVC, respectively, P<0.05; SP>90%). Torque variability increased significantly during exercise and was significantly larger in HT and lower in CL compared to CON experiment. Voluntary activation of exercising muscle was more depressed in HT (i.e. greater central fatigue) and the smallest effect was found in CL compared to CON. In conclusion increased core and muscle temperature impairs voluntary activation and increases force variability of the exercising muscles while a local muscle cooling decrease force variability but has a small effect on central fatigue.  相似文献

All intertidal gastropods for which cold tolerance strategies have been assessed have been shown to be freeze tolerant. Thus, freeze tolerance is considered an adaptation to the intertidal environment. We investigated the cold tolerance strategies of three species of subtropical and temperate snails (Gastropoda: Littorinidae) to determine whether this group is phylogenetically constrained to freeze tolerance. We exposed dry acclimated and wet rehydrated snails to low temperatures to determine temperature of crystallisation (Tc), lower lethal temperature and LT50 and to examine the relationship between ice formation and mortality. Tc was lowest in dry Afrolittorina knysnaensis (–13.6±0.4 °C), followed by dry Echinolittorina natalensis (–10.9±0.2 °C) and wet A. knysnaensis (–10.2±0.2 °C). The Tc of both A. knysnaensis and E. natalensis increased with rehydration, whereas Tc of dry and wet Afrolittorina africana did not differ (–9.6±0.2 and –9.0±0.2 °C respectively). Wet snails of all species exhibited no or low survival of inoculative freezing, whereas dry individuals of A. knysnaensis could survive subzero temperatures above –8 °C when freezing was inoculated with ice. In the absence of external ice, Afrolittorina knysnaensis employs a freeze-avoidance strategy of cold tolerance, the first time this has been reported for an intertidal snail, indicating that there is no family-level phylogenetic constraint to freeze tolerance. Echinolittorina natalensis and A. africana both showed pre-freeze mortality and survival of some internal ice formation, but were not cold hardy in any strict sense.  相似文献

Summary Literature data suggest that water accumulation by the human fetus is driven by osmotic gradients of small solutes. However, the existence of such gradients has not been supported by prior measurements. Attempts to estimate the size of the gradient necessary to drive net water movement have been seriously hampered by the lack of permeability data for the syncytiotrophoblast membranes. Stopped-flow light scattering techniques were employed to measure the osmotic water permeability (P f )of microvillous (MVM) and basal membrane (BM) vesicles isolated from human term placenta. At 37°C, the P f was determined to be 1.9±0.06 × 10+–3 cm/sec for MVM and 3.1±0.20 × 10+–3 cm/sec for BM (mean ±SD, n = 6). At 23°C, P f was reduced to 0.7±0.04 × 10+–3 cm/sec in MVM and 1.6±0.05 × 10+–3 cm/sec in BM. These P f values are comparable to those observed in membranes where water has been shown to permeate via a lipid diffusive mechanism. Arrhenius plots of P f over the range 20–40°C were linear, with activation energies of 13.6 ± 0.6 kcal/mol for MVM and 12.9±1.0 kcal/mol for BM. Water permeation was not affected by mercurial sulfhydryl agents and glucose transport inhibitors. These data clearly suggest that water movement across human syncytiotrophoblast membranes occurs by a lipid diffusion pathway. As noted in several other epithelial tissues, the basal membrane has a higher water permeability than the microvillous membrane. It is speculated that water accumulation by the human fetus could be driven by a solute gradient small enough to be within the error of osmolarity measurements.We thank the staff of the labor and delivery ward at University of San Francisco Medical Center for help in obtaining placental tissue. A series of comparative culture experiments were conducted in order to determine responses of Laminaria longicruris male and female gametophytes and juvenile sporophytes to several temperatures (5, 10, 15, 20 °C), light levels (10, 35, 75 µmol m–2 s–1) and media nitrogen concentrations (0, 20, 100 µM ammonium-nitrogen). Responses were measured as numbers of male and female gametophytes producing gametangia and number of sporophytes produced following fertilization. Both male and female gametogenesis was reduced at 5 and 20 °C versus 10 and 15 °C. At 20 °C gametogenesis inhibition was greater with higher levels of ammonium-nitrogen concentration (100 µM). Sporophyte production was more sensitive to light, temperature and nitrogen concentration than gametogenesis. Production of sporophytes was inhibited completely at 20 °C. At lower temperatures, increasingly higher nutrient concentrations produced greater inhibition of production of sporophytes.  相似文献
