Different solvent mixtures were examined for extraction of fumonisins from various naturally contaminated and spiked foods and foodstuffs: rough rice, retail rice, rice flour, white corn flour, corn meal, corn starch, corn flakes, tortilla/corn chips, white bean flour, white beans, mung beans, adzuki beans and infant cereals. Most of the naturally contaminated samples were analyzed using the extraction solvent mixtures methanol-acetonitrile-water (25:25:50) (solvent A) and methanol-water (75:25 or 80:20) (solvents B, BB); some were extracted with 0.1 M sodium hydrogen phosphate-acetonitrile (1:1, adjusted to pH 3.0 with o-phosphoric acid) (solvent C) and methanol-0.025 M borate buffer (3:1, adjusted to pH 9.2 with 1 N sodium hydroxide) (solvent D). A 1-ml SAX solid phase extraction column was used for the cleanup in all cases except for infant cereals, for which immunoaffinity chromatography was used; fumonisin concentrations were determined by liquid chromatography. Solvent A gave slightly better extraction of fumonisins from one of two samples of naturally contaminated rough rice than solvent B (fumonisin B1: 4080 ng/g versus 3150 ng/g; fumonisin B2:1100 ng/ g versus 922 ng/g) and much better extraction than solvent C (1210 ng/g fumonisin B1 and 315 ng/g fumonisin B2) or solvent D (372 ng/ g fumonisin B1 and 191 ng/g fumonisin B2). However, spike recoveries on a similar rice naturally contaminated at a lower level were only in the 43–53% range (solvent A). Recovery of fumonisins was very poor from spiked white rice flour but satisfactory from other rice foods. Solvent A similarly gave slightly better extraction of fumonisins from a sample of naturally contaminated white corn flour than solvent B (fumonisin B1 1260 ng/g versus 931 ng/g; fumonisin B2: 511 ng/g versus 447 ng/g ) and better extraction than solvents C and D. Solvent A was also a better solvent for extraction of fumonisins from naturally contaminated tortilla chips and infant cereals. Study of naturally contaminated corn starch was confounded by instability of fumonisins in this food. Recovery of fumonisins from spiked corn meal, tortilla chips, corn flakes, various types of beans and infant cereals with solvent A and/or solvent B (or BB) was satisfactory.  相似文献

Accelerated solvent extraction (ASE) is an alternative sample extraction procedure for ochratoxin A in roasted coffee. ASE results are comparable to that of the modified Koch method, but required less sample preparation time. Furthermore, ASE gave higher quantitative values than other methods reported for extraction of ochratoxin A. In the end less harmful water could be used for extraction.  相似文献

The extraction and purification methods used in the maleyl derivatization HPLC technique was evaluated with respect to the pH of the extraction mixture, the extraction solvent and the purification methods used in order to determine optimum conditions for quantification of fumonisins B1, B2, and B3 in corn cultures. The highest recovery of the three compounds was obtained by extraction at pH 3.5 with CH3OH–H20 (3∶1), whilst the subsequent solvent partitioning and reversedphase C18 Sep-pak purification have been shown to be very important in the quantification of the fumonisins in the corn cultures. The percentage recovery of the improved technique, utilizing a gradient HPLC solvent system for the simultaneous determination of the fumonisins, was 93.4% for FB1, 68.0% for FB2, and 82.6% for FB3. The study indicates that the polarity of the fumonisins and consequently their solubility during extraction as well as their behavior during the subsequent purification step play an important role in quantification of these mycotoxins in corn cultures.  相似文献

We analyzed 44 moldy corn samples for the B and C series of fumonisins by high-performance liquid chromatography. Of the 44 samples, 32 (73%) were contaminated with both the B and C series of fumonisins and 6 were contaminated with only the B series of fumonisins. The incidence of fumonisin C1 in moldy corn was 71%; the incidence was 11% for fumonisin C3 and 43% for fumonisin C4. Their mean levels ranged from 500 to 1,900 ng/g. This is the first report on the natural occurrence of the C series of fumonisins and fumonisin B4 in moldy corn.  相似文献

Natural occurrence of fumonisins B1 (FB1) and B2 (FB2), a promoter for hepato-carcinogenesis, was investigated in corn and corn — based products sampled in Japan, Nepal, and China by high — performance liquid chromatographic method. From the 9 imported corn kernel and 6 gluten feed samples, FB1 was detected in 8 corn (0.6 ~ 4.1μg/g) and all gluten feed (0.3 ~ 2.4μg/g) samples, while FB2 was found in the same corn (0.3 ~ 10μg/g) and 3 gluten feed (0.8 ~ 8.5μg/g) samples. ELISA analysis also revealed the contamination of aflatoxin B1 in 2 corn and all gluten feed samples along with fumonisins. Of 17 corn grit samples, 14 and 5 samples were contaminated with fumonisin B1 and B2, with maximum levels of 2.6 and 2.8μg/g, respectively. As for corn-based foodstuffs marketed in Japan, no significant contamination of fumonisins was observed. Among 24 corn kernel samples in Nepal, 12 and 7 samples were positive for FB1 and FB2, and averaged to 0.6 and 1.6μg/g, respectively. One sample showed the highest fumonisin contents as 4.6 and 5.5μg/g, respectively. In corn samples harvested at Shanghai and Beijing, China, FB1 and FB2 were detected in various concentrations. Mycological survey has also revealed the presence of a fumonisin — producing fungus in a crop field of Japan. These findings have for the first time demonstrated high levels of contamination of fumonisins in corn and corn — based products in Asian countries. Natural co — occurrence of fumonisins and aflatoxin B1 was also detected in raw materials for mixed feed.  相似文献

The fumonisin mycotoxins, which are worldwide contaminants of corn, inhibit de novo sphingolipid biosynthesis leading to elevation in the ratio of the sphingoid bases, sphinganine and sphingosine, in the serum of animals exposed to fumonisins. A new HPLC method for the determination of the ratio of these bases in serum has been developed involving lipid extraction, clean-up on a silica minicolumn and alkaline hydrolysis prior to precolumn o-phthaldialdehyde derivatisation and HPLC separation and quantification by fluorescence detection. Based on serum from both normal and fumonisin-exposed vervet monkeys, the method was shown to be reproducible (R.S.D.<10%).  相似文献

Fumonisins are mycotoxins produced by several species of Fusaria. They are found on corn and in corn-based products, can cause fatal illnesses in some animals and are suspected human esophageal carcinogens. Fumonisins are believed to cause toxicity by blocking ceramide synthase, a key enzyme in sphingolipid biochemistry which converts sphinganine (or sphingosine) and fatty acyl CoA to ceramide. Relatively fewfungal species have been evaluated for their ability to produce fumonisins. Fewer have been studied to determine if they produce ceramide synthase inhibitors, whether fumonisin-like structures or not, therefore potentially having toxicity similar to fumonisins. We analyzed corn cultures of 49 isolates representing 32 diversespecies of fungi for their ability to produce fumonisins. We also evaluated the culture extracts for ceramide synthase activity. Only cultures prepared with species reported previously to produce fumonisins – Fusarium moniliforme and F. proliferatum – tested positive for fumonisins. Extracts of these cultures inhibited ceramide synthase, as expected. None of the other fungal isolates we examined produced fumonisins or other compounds capable of inhibiting ceramide synthase. Although the fungi we selected for these studies represent only a few ofthe thousands of species that exist, they share the commonality that they are frequently associated with cereal grasses, including corn, either as pathogens or as asymptomatic endophytes. Thus,these results should be encouraging to those attempting to find ways to genetically manipulate fumonisin-producing fungi, tomake corn more resistant, or to develop biocontrol measures because it appears that only a relatively few fungal contaminants of corn can produce fumonisins. Optimal conditions for the extraction and analysis of furanocoumarins from fruits of Archangelica officinalis Hoffm. have been determined. The following extraction methods were used: exhaustive extraction in a Soxhlet apparatus, ultrasonication at 25 and 60 degrees C, microwave-assisted solvent extraction in open and closed systems, and accelerated solvent extraction (ASE). In most cases the yields of furanocoumarins were highest using the ASE method. The effects of extracting solvent, temperature and time of extraction using this method were investigated. The highest yield of furanocoumarins by ASE was obtained with methanol at 100-130 degrees C for 10 min. The extraction yields of furanocoumarins from plant material by ultrasonication at 60 degrees C and microwave-assisted solvent extraction in an open system were comparable to the extraction yields obtained in the time- and solvent-consuming exhaustive process involving the Soxhlet apparatus.  相似文献

Contamination of corn with the fungus Fusarium moniliforme and its secondary metabolites, the fumonisins, has been associated with several human and animal diseases. This paper summarizes present knowledge and presents new data on the levels of fumonisins present in foods and feeds associated with these diseases as well as in commercial corn and corn-based products. The doses of fumonisins to which humans and animals consuming these products would be exposed are compared with those doses known to produce LEM in horses and hepatocarcinogenesis in rats. It is concluded that the known naturally occurring levels of fumonisins present a potential threat to human and animal health and realistic tolerance levels need to be set.  相似文献

为建立一种加速溶剂萃取-毛细管区带电泳测定女贞子中齐墩果酸和熊果酸的新方法.考察了萃取温度、萃取时间和萃取次数对目标物萃取效率的影响,并考察了硼砂浓度,β-环糊精浓度、pH及甲醇浓度对目标物分离的影响.结果表明:(1)萃取温度和萃取次数影响目标物的萃取效率,而萃取时间的影响很小;萃取压力影响萃取过程的重现性.(2)优化的萃取条件为:萃取压力6.9 MPa,萃取温度100℃,萃取时间5 min和萃取次数2次.(3)优化的缓冲体系为:40 mmol/L硼砂,1 mmol/Lβ-环糊精,pH 9.5及6%甲醇.(4)齐墩果酸和熊果酸分别在10~200、10~160 mg/L范围内线性良好,相关系数均大于0.996,检测限分别为3.2 mg/L和3.0 mg/L,加标回收率为93%~97%.对比了加速溶剂萃取、索氏提取以及超声提取的提取效率.(5)不同提取方法比较结果表明,加速溶剂萃取的提取效率与索氏提取法接近,但高于超声提取;加速溶剂萃取法的主要优点是消耗提取溶剂量少,提取时间短,样品用量小.  相似文献

Fusarium verticillioides (Gibberella fujikuroi mating population A) is a producer of fumonisins and one of the main contaminants of corn grains. In Brazil, some studies analyzing strains isolated from corn have demonstrated high levels of fumonisins, whereas the levels for strains isolated from sorghum have been found to be low. In the present study, we investigated the genetic diversity of 22 F. verticillioides strains isolated from corn and 21 strains isolated from sorghum cultivated in the State of S?o Paulo, Brazil. Differences in the genetic profile were observed between the strains isolated from the two substrates using single primer amplification reaction by polymerase chain reaction (SPAR-PCR). Fumonisins levels were higher in strains isolated from corn than in those isolated from sorghum. The MAT-1 and MAT-2 alleles were identified by PCR, and the isolates were subsequently crossed with Fusarium thapsinum (G. fujikuroi mating population F) reference strains because this species is morphologically similar to F. verticillioides and produces low levels of fumonisins. The SPAR haplotypes of some strains isolated from sorghum were similar to the F. thapsinum reference strain haplotypes, but there was no fertile mating between the strains isolated from the two substrates and the F. thapsinum references strains. The MAT-1:MAT-2 proportion was 5:17 and 14:7 for isolates from corn and sorghum, respectively.  相似文献

Using the seed- plate technique, we have isolated a strain ofF. proliferatum from rye grains that produces 3 fumonisins, fumonisin B1 (FB1), FB2 and FB3 on inoculated rice and corn. Inoculated corn and rice were extracted with an aqueous methanol solution and fumonisin concentrations estimated using high performance liquid chromatography. Production of all 3 fumonisins (FB1, FB2 and FB3) was much higher on rice than corn; ranging from 3816, 1068 and 985 ppm to 1643, 350 and 162 ppm respectively. We conclude that all natural substrates whereF. proliferatum is present as a component of the mycoflora should be monitored for the presence of fumonisins.  相似文献

Analysis of plant material is an important task in chemotaxonomical investigations, in search of plants with pharmacological activity or in standardisation of plant drugs. The choice of optimal conditions for the analysis of plant material and effect of extraction method on the yield of furanocoumarins from Pastinaca sativa fruits were examined. The following extraction methods were used in experiments: exhaustive extraction in Soxhlet apparatus, ultrasonification (USAE) at 25 and 60 degrees C, microwave-assisted solvent extraction in open and closed system (MASE) and accelerated solvent extraction (ASE). In Entre Ríos, Argentina, corn is one of the most important cereal grains produced, being an important income for the regional economy. The aim of this work was to assess aflatoxins, zearalenone, deoxynivalenol (DON) and fumonisins (FB) in corn harvest in 2003 and 2004 in the most contaminated departments found in previous studies in selected sampling places. At the harvest time, when the trucks arrived to store plants, samples of corn were taken from seven different positions of the trucks and from five in the trailer. Composite samples were randomised reduced to 10 kg. The samples were analysed by immunological tests, by thin layer chromatography (TLC), high performance liquid chromatography (HPLC) and/or gas liquid chromatography-electron capture detector (GLC-ECD). In 2003 average contamination was 3.19 u.g/kg for aflatoxins, 118.5 μg/kg for deoxynivalenol, 230.8 μg/kg for zearalenone and 10200 μg/kg of total fumonisins (HPLC and ELISA quantification showed a linear correlation (r2 =0.9618), but RIDASCREEN®FAST values were 1.7 higher than HPLC values); in 2004 deoxynivalenol and zearalenone were not detected and an average of 2.0 μg/kg for aflatoxins and 4700 μg/kg for total fumonisins was found.This province, with the earliest harvested corn in the country each summer, tends to display different contaminations from the rest of the provinces, probably due to climate characteristics, particularly hotter weather.  相似文献

A survey was conducted to determine the levels of fumonisins B1 and B2 in corn and corn-based products available in Colombia for human and animal consumption. A total of 120 samples were analyzed by acetonitrile-water extraction, cleanup with a strong-anion-exchange column, and liquid chromatography with o-phthaldialdehyde-2-mercaptoethanol derivatization and fluorescence detection. The samples of corn and corn-based products for animal intake were taken at different feed manufacturing plants, whereas the samples used for human foods where purchased from local retail stores. The number of positive samples for fumonisin B1 was 20.0% higher in corn and corn-based products for animal intake (75.0%) than in corn and corn-based products for human consumption (55.0%). The levels of fumonisin B1 were also higher in corn and corn-based products for animal intake (mean = 694 μg/kg; range = 32–2964 μg/kg), than in corn and corn-based products for human intake (mean = 218 μg/kg; range = 24–2170 μg/ kg). The incidence and levels of fumonisin B2 were lower than those for fumonisin B1. Corn and corn-based products for animal consumption had an incidence of fumonisin B2 of 58.3%, with a mean value of 283 μg/kg, and a range of 44–987 μg/kg. The incidence of fumonisin B2 in corn-based products for human intake was 35.0%, with a mean value of 118 μg/kg and a range of 21–833 μg/kg. The highest incidence and levels of fumonisins were found in samples of hominy feed, with concentrations ranging from 86 to 2964 μg/kg fumonisin B1 and 57 to 987 μg/kg fumonisin B2.  相似文献

Fusarium moniliforme is the predominant fusarium species in the grain mycoflora of corn grown in Northern Caucasus, accounting for 95% of fusarium isolates. Eighty-five Fusarium moniliforme strains were grown on grain substrate and checked for the presence of fumonisins (B1 + B2 + B3) by indirect solid-phase enzyme immunoassay (EIA). All strains were capable of producing fumonisins (0.95 to 32,000 mg/kg). Strains sampled in the Krasnodar krai produced the highest fumonisin levels (averaging 5490 mg/kg).  相似文献

Fumonisins are secondary metabolites of the fungus Fusarium moniliforme, a common mycotoxin in corn, which are known to cause cancer in a number of experimental animals and have been linked to human esophageal cancer in China and South Africa. A high-performance liquid chromatographic (HPLC) method is currently the most widely used method for the quantitative determination of fumonisins. This method utilizes precolumn derivatization with o-phthaldialdehyde, isocratic elution, and fluorescence detection. In this study, the HPLC method was chosen as the reference method to evaluate the reproducibility and accuracy of FILIA (flow-injection liposome immunoanalysis) for the detection of the fumonisin B1 (FmB1). Studies indicate that a recovery of 86-90% could be obtained when commercial yellow cornmeal spiked with FmB1 was extracted in 75% methanol, which correlated favorably (correlation coefficient, r(2)=0.945) with the result of 80-92% obtained using the flow-injection liposome immunoanalysis (FILIA) system. The data suggest that the FILIA method is comparable to HPLC for the detection of fumonisins in corn, animal feeds, and human foods. Important features of FILIA as compared to HPLC are, most importantly, lower detection limit (ca. 25 x lower), and also less complex and faster sample preparation and therefore increased analytical throughput. In addition, 24 human corn-based foods and 6 animal feeds were examined for the presence of FmB1 using HPLC and FILIA.  相似文献
