The murine thymoma cell line EL-4 was used as an in vitro T-cell model to assess the immunomodulating effect of pure Ochratoxin A (OTA) and an Aspergillus ochraceus raw toxin preparation. Cytokine production (IL-2, IL-4, IL-5, IL-6) and cell viability of PMA-stimulated EL-4 cells were investigated in the presence of OTA. The cytotoxic effect of the raw toxin could be observed at lower concentrations than pure OTA. The IC50 values were 3 µg/ml and 11 µg/ml, respectively. Increasing concentrations of both OTA preparations caused an inhibition of cytokine production, but the inhibition effect of the raw toxin was stronger than of pure OTA. This is supposed to be the effect of further up to now not characterized substances in the raw toxin. Differences in the susceptibility of the mechanisms of production and regulation of each cytokine are indicated by the different concentrations for inhibition effects. Both toxin preparations showed also stimulating effects on some cytokines (IL-2 and IL-6) while others (IL-4 and IL-5) were depressed at these OTA concentrations. This indicates the immunomodulating properties of the toxin.  相似文献

Deoxynivalenol (DON) is a mycotoxin that reduces feed intake and animal performance, especially in swine. Arginine and glutamine play important roles in swine nutrition. The objective of this study was to determine the effects of dietary supplementation with arginine and glutamine on both the impairment induced by DON stress and immune relevant cytokines in growing pigs. A total of forty 60-d-old healthy growing pigs with a mean body weight of 16.28±1.54 kg were randomly divided into 5 groups, and assigned to 3 amino acid treatments fed 1.0% arginine (Arg), 1.0% glutamine (Gln) and 0.5% Arg+0.5% Gln, respectively, plus a toxin control and a non-toxin control. Pigs in the 3 amino acid treatments were fed the corresponding amino acids, and those in non-toxin control and toxin control were fed commercial diet with 1.64% Alanine as isonitrogenous control for 7 days. The toxin control and amino acid treatments were then challenged by feeding DON-contaminated diet with a final DON concentration of 6 mg/kg of diet for 21 days. No significant differences were observed between toxin control and the amino acid groups with regard to the average daily gain (ADG), although the values for average daily feed intake (ADFI) in the amino acid groups were significantly higher than that in toxin control (P<0.01). The relative liver weight in toxin control was significantly greater than those in non-toxin control, arginine and Arg+Glu groups (P<0.01), but there were no significant differences in other organs. With regard to serum biochemistry, the values of BUN, ALP, ALT and AST in the amino acid groups were lower than those in toxin control. IGF1, GH and SOD in the amino acid groups were significantly higher than those in toxin control (P<0.01). The IL-2 and TNFα values in the amino acid groups were similar to those in non-toxin control, and significantly lower than those in toxin control (P<0.01). These results showed the effects of dietary supplementation with arginine and glutamine on alleviating the impairment induced by DON stress and immune relevant cytokines in growing pigs.  相似文献

The present study which includes two feeding experiments was performed to investigate a possible performance enhancing effect of rare earth elements (REE) in piglets. This performance enhancing effect has been described in the Chinese literature for a long time, however, it was never tested under “western conditions”;. In the first feeding experiment 72 piglets at a mean BW of 7.3 kg were allotted to a control and to 4 REE groups at different levels of lanthanum chloride or an REE mixture containing mainly chlorides of lanthanum, cerium and praseodymium. The experimental period lasted 5 weeks. Positive effects of REE were found on body weight gain as well as on feed conversion ratio of the piglets. Compared to the control group, the daily weight gain was improved by 2 to 5% and feed conversion was better by up to 7%. These effects were, however, not significant. In the second feeding experiment, piglets (mean BW 17.3 kg) were fed for 8 weeks with a similar REE mixture. Significant positive effects of REE were found on both body weight gain and on feed conversion ratio by 19% and 10%, respectively. This is the first time that a performance enhancing effect of REE in pigs under western feeding conditions has been shown. Since the use of antibiotics as growth promoters in animal feed has been restricted in the European Union recently, rare earth elements might be of interest as new, safe and inexpensive alternative performance enhancers.  相似文献

To evaluate the minimum effective dose of pure DON leading to measurable losses in weight gain and feed consumption a special feeding experiment was created to compare the effect of DON in natural contaminated wheat and — for the first time — a non-cereal diet (potato) spiked with pure DON. Examined parameters were weight gain, feed consumption and blood parameters.Three trials were conducted. In the first trial a concentration of 4000 µg DON/kg feed was used. In the second and third trial we used concentrations of 4000 and 6000 µg DON/kg feed. Severe effects on feed consumption and weight gain were found only in the second trial (naturally contaminated wheat ad lib.). By contrast, no differences in any parameter were found in the first (restricted feeding) and third trial (non-cereal diet spiked with pure DON ad lib.).  相似文献

Cyclopiazonic acid (CPA) was purified from cultures of Aspergillus flavus, and ca. 14 g of the toxin was collected for use in feeding studies. Chicken rations were artificially contaminated with purified CPA at concentrations of 10, 50, and 100 ppm (microgram/g) and fed ad libitum to eight groups of chickens for 7 weeks. Chickens receiving feed with 100 ppm of CPA had high mortality, decreased weight gain, and poor feed conversion when compared with birds receiving other doses. Postmortem examination showed that chickens fed the two greatest doses of CPA had proventricular lesions characterized by mucosal erosion and hyperemia (100 ppm) and by thick mucosa and dilated proventricular lumens (50 ppm). Birds given 100 ppm of CPA in feed also had numerous yellow foci in their livers and spleens. Microscopic examination of tissues of birds that received 100 ppm of CPA revealed ulcerative proventriculitis, mucosal necrosis in the gizzard, and hepatic and splenic necrosis and inflammation. Birds given 50 ppm of CPA had hyperplasia of the proventricular mucosal epithelium. Birds given 10 ppm of CPA and control birds had no significant treatment-related lesions.  相似文献

The effect of practically relevant mycotoxin concentrations on functions of immune cells was studied in in vitro experiments. Porcine mononuclear cells were exposed to a crudeAspergillus-ochraceus toxin containing OTA, a HPLC fraction identical with OTC derived from the crude toxin (RE2), as well as pure OTA and OTC in a concentration range from 0.46 to 3000 ng/ml. The influence of mycotoxin exposure on metabolic activity, mitogen induced proliferation, expression of the activation marker CD25 and the cell cycle of lymphocytes and on the formation of free oxygen radicals as well as the production of the cytokines IL-6 and TNF-α by monocytes was determined. Exposure to high concentrations of all mycotoxin preparations lead to non-specific suppression of the immune cell functions, which was related to cytotoxic effects. Low concentrations caused ambivalent reactions, especially on monocyte function. In general, the HPLC fraction RE2 had an up to 100-fold stronger effect than pure OTA. Ochratoxin-induced suppression of lymphocyte proliferation was not abrogated by phenylalanine or aspartame. The results indicate that immunomodulation can be caused by very low mycotoxin concentrations which are not related to clinical symptoms or loss of performance.  相似文献

The forty-two days long experiment was conducted on a total of 1000 Arbor Acres broilers, divided into two groups. Both groups of broilers were fed with a commercial feed mixture which consisted of standard feedstuffs and contained enough nutrients regarding the requirements. During the first three weeks of the trial, corn naturally contaminated with AFB1 0.0445 ppm per kg dry matter was used in the amount of 20% in the experimental group, while AFB1 free corn was given to birds in the control group. After the period of toxin administration until the end of the trial, broilers from both groups were normally fed another 5 weeks with uncontaminated feed. In the first phase of the trial, broilers in the control groups had an average daily gain (ADG) of 31 g, average daily feed intake (ADFI) of 45 g and a feed: gain ratio (FCR) of 1.42. In the same time, experimental broilers achieved an ADG of 25 g, ADFI of 35 g and a FCR of 1.39. During the whole period of the trial, control and experimental broilers achieved 49.12 g, 95.24 g and 1.94 and 39.71 g, 86.90 g and 2.19, respectively.  相似文献

This study aimed to investigate the effect of supplemental camphor on the performance and immune functions of Japanese quail by feeding graded levels (0 (control), 250, 500, 750, 1000, 5000 or 10 000 ppm) of camphor during a 42-day feeding trial. In all, 280 1-day-old quail chicks were randomly assigned into 28 cages of 10 chicks each with separate feeders. The results clearly demonstrated that camphor did not have a significant effect on BW, BW gain, total experimental average daily feed intake, feed conversion ratio, internal organ relative weights and biochemical parameters such as uric acid, albumin, total protein and triglyceride; however, plasma cholesterol concentration was significantly different in a linear manner, in which 500 ppm of camphor resulted in a lower level of cholesterol. Alternatively, greater concentrations of glucose and high-density lipoprotein (HDL) were also found in 5000 and 1000 ppm of camphor groups, respectively. Cellular responses to the phytohaemagglutinin-P and 2,4-dinitro 1-chlorobenzene skin test were not influenced by dietary camphor. Humoral responses to secondary sheep red blood cells, avian influenza virus (AIV) and Newcastle disease virus (NDV) immunisations were positively influenced by camphor supplementation, in which greater secondary response to sheep erythrocytes belonged to 750 and 1000 ppm of camphor groups; whereas, diet supplementation with camphor had no significant effect on lymphoid organ weights and heterophil-to-lymphocyte ratio. The greatest AIV antibody titers were seen in groups, which received 1000 and 5000 ppm of camphor (P<0.05) and the values of NDV antibody titers increased with an increase in the camphor consumption. Furthermore, dietary inclusion of 500 ppm of camphor positively decreased coliform populations in the gastrointestinal tract (GIT). In addition, an increase in lactic acid bacteria was also observed in quails, which were fed on the diets containing 750 ppm camphor. Collectively, these data suggest that as a phytogenic feed additive, camphor may effectively act as a modulator of health status (by increasing glucose and HDL), GIT microbiota and immunological responses of the Japanese quail.  相似文献

BACKGROUND: Emodin, a widely available herbal remedy, was evaluated for potential effects on pregnancy outcome. METHODS: Emodin was administered in feed to timed-mated Sprague-Dawley (CD) rats (0, 425, 850, and 1700 ppm; gestational day [GD] 6-20), and Swiss Albino (CD-1) mice (0, 600, 2500 or 6000 ppm; GD 6-17). Ingested dose was 0, 31, 57, and approximately 80-144 mg emodin/kg/day (rats) and 0, 94, 391, and 1005 mg emodin/kg/day (mice). Timed-mated animals (23-25/group) were monitored for body weight, feed/water consumption, and clinical signs. At termination (rats: GD 20; mice: GD 17), confirmed pregnant dams (21-25/group) were evaluated for clinical signs: body, liver, kidney, and gravid uterine weights, uterine contents, and number of corpora lutea. Fetuses were weighed, sexed, and examined for external, visceral, and skeletal malformations/variations. RESULTS: There were no maternal deaths. In rats, maternal body weight, weight gain during treatment, and corrected weight gain exhibited a decreasing trend. Maternal body weight gain during treatment was significantly reduced at the high dose. In mice, maternal body weight and weight gain was decreased at the high dose. CONCLUSIONS: Prenatal mortality, live litter size, fetal sex ratio, and morphological development were unaffected in both rats and mice. At the high dose, rat average fetal body weight per litter was unaffected, but was significantly reduced in mice. The rat maternal lowest observed adverse effect level (LOAEL) was 1700 ppm; the no observed adverse effect level (NOAEL) was 850 ppm. The rat developmental toxicity NOAEL was > or =1700 ppm. A LOAEL was not established. In mice, the maternal toxicity LOAEL was 6000 ppm and the NOAEL was 2500 ppm. The developmental toxicity LOAEL was 6000 ppm (reduced fetal body weight) and the NOAEL was 2500 ppm.  相似文献

The effect of six different feeding times was tested on feed intake, growth performance, proximate body composition and nutrient retention in the rainbow trout Oncorhynchus mykiss . Using a non‐linear regression model, a significant rhythmic pattern over a 24 h period was observed for feed gain ratio and nutrient retention responses to feeding time. Specific growth rate and protein growth rate responses were also rhythmic but the trends were not significant. There was no clear effect of feeding time on feed intake and proximate body composition. The study suggested that feed intake, at least under the experimental conditions encountered, was synchronized to feeding time while some physiological rhythms involved in nutrient metabolism were probably synchronized to photoperiod.  相似文献

Eight feeding trials using 708 cross-bred pigs were conducted to evaluate the addition of virginiamycin and a commercially-available lactobacillus fermention product (probiotic) to starter, grower and finisher diets based on maize and soya bean meal. Virginiamycin was added at a level of 11 mg/kg of diet, and lactobacillus probiotic at 1 g/kg in starter diets and 500 mg/kg in grower and finisher diets. In the combined analysis of four starter trials, pigs fed on diets containing virginiamycin tended to eat more and grow faster than pigs fed on the control diet, but feed efficiency was not different. Furthermore, daily gain, daily feed intake and feed efficiency were not improved by the addition of lactobacillus probiotic to starter diets. In the combined analysis of a starter—grower—finisher and a grower—finisher trial, virginiamycin supplementation did not alter overall pig performance compared with control. Pigs fed on a positive control diet containing chlortetracycline, penicillin and sulfamethazine had a nonsignificantly greater daily gain and feed intake.Daily gain for the lactobacillus probiotic-fed pigs was lower (P < 0.05) than that of the negative controls, but daily feed intake and feed efficiency were not significantly different. In three growing—finishing trials in which virginiamycin and lactobacillus probiotic were compared in a 2 × 2 factorial arrangement of treatments, daily gain and feed intakes were greater (P < 0.01) for pigs given virginiamycin. The lactobacillus probiotic did not improve daily gain, feed intake or feed efficiency. Overall, no interaction between virginiamycin and lactobacillus probiotic occurred.  相似文献

A total of 240 pigs, 74 days old, half boars and half females, were included in a trial designed to assess the effect of the stress caused by changes in the pattern of food administration on the concentration of acute phase proteins (APP) and productive performance parameters. Half of the animals (pigs fed ad libitum, AL group) had free access to feed, while the rest were fed following a disorderly pattern (DIS group), in which animals had alternating periods of free access to feed and periods of no feeding, when food was removed from the feeder. The periods of free access to feed (two daily periods of 2-h duration) were randomly assigned, and varied from day to day. Total feed supplied per day was identical in both groups, and exceeded the minimal amount required for animals of these ages. Pen feed intake, individual body weights and the main positive pig APP pig major acute phase protein (Pig-MAP), haptoglobin, serum amyloid A (SAA), C-reactive protein (CRP), and the negative APP apolipoprotein A-I (ApoA-I) and transtherytin were determined every 2 weeks during the period 76 to 116 days of age. Animals fed ad libitum had better average daily gain (ADG) than DIS animals in the whole experimental period (P < 0.01) but the differences in ADG were only produced in the two first experimental sub-periods (60 to 74 and 74 to 116 days of age), suggesting that the stress diminished when the animals get used to the DIS feeding. Interestingly differences in ADG between DIS and AL pigs were due to males, whereas no differences were observed between females. The same differences observed for ADG were found for APP. DIS males had higher Pig-MAP concentration than AL males at 74 and 116 days of age, lower ApoA-I concentration at 74 days of age and higher haptoglobin and CRP concentration at 116 days of age (P < 0.05). The results obtained in this trial show an inverse relationship between weight gain and APP levels, and suggest that APP may be biomarkers for the evaluation of distress and welfare in pigs.  相似文献

The effect of rumen microbes on six mycotoxins (aflatoxin B1, ochratoxin A, zearalenone, T-2 toxin, diacetoxyscirpenol, and deoxynivalenol ) considered to be health risks for domestic animals was investigated. The mycotoxins were incubated with intact rumen fluid or fractions of rumen protozoa and bacteria from sheep and cattle in the presence or absence of milled feed. Rumen fluid had no effect on aflatoxin B1 and deoxynivalenol . The remaining four mycotoxins were all metabolized, and protozoa were more active than bacteria. Metabolism of ochratoxin A, zearalenone, and diacetoxyscirpenol was moderately or slightly inhibited by addition of milled feed in vitro. The capacity of rumen fluid to degrade ochratoxin A decreased after feeding, but this activity was gradually restored by the next feeding time. Ochratoxin A was cleaved to ochratoxin alpha and phenylalanine; zearalenone was reduced to alpha-zearalenol and to a lesser degree to beta-zearalenol; diacetoxyscirpenol and T-2 toxin were deacetylated to monoacetoxyscirpenol and HT-2 toxin, respectively. Feeding of 5 ppm (5 mg/kg) of ochratoxin A to sheep revealed 14 ppb (14 ng/ml) of ochratoxin A and ochratoxin alpha in rumen fluid after 1 h, but neither was detected in the blood. Whether such conversions in the rumen fluid may be considered as a first line of defense against toxic compounds present in the diet is briefly discussed.  相似文献

This study was conducted to determine the chromium content of different feedstuffs, their estimated requirement, and effect of dietary Cr supplementation on nutrient intake, nutrient utilization, growth performance, and mineral balance in buffalo calves during summer season. Levels of Cr was higher in cultivated fodder, moderate in cakes and cereal grains, while straw, grasses, and non-conventional feeds were poor in Cr content. To test the effect of Cr supplementation in buffalo calves, 0, 0.5, 1.0, and 1.5 ppm of inorganic Cr were fed to 24 buffalo calves. Buffalo calves were randomly assigned to four treatments (n?=?6) and raised for 120 days. A metabolic trial for a period of 7 days was conducted after 3 months of dietary treatments. Blood samples were collected at fortnight interval for plasma mineral estimation. The results suggested that dietary Cr supplementation in summer did not have any affects (P?>?0.05) on feed consumption, growth performance, nitrogen balance, and physiological variables. However, dietary Cr supplementation had significant effect (P?<?0.05) on balance and plasma Cr (ppb) levels without affecting (P?>?0.05) balance and plasma levels of other trace minerals. The estimated Cr requirement of buffalo calves during summer season was calculated to be 0.044 mg/kg body mass and 10.37 ppm per day. In conclusion, dietary Cr supplementation has regardless effect on feed consumption, mass gain, and nutrient utilization in buffalo calves reared under heat stress conditions. However, supplementation of Cr had positive effect on its balance and plasma concentration without interacting with other trace minerals.  相似文献

Earlier studies have shown that larvae of the green lacewing predator Chrysoperla carnea are negatively affected when preying on lepidopteran larvae that had been fed with transgenic maize expressing the cry1Ab gene from Bacillus thuringiensis. To test whether the observed effects were directly caused by the Cry1Ab toxin, we have developed a bioassay which allows us to feed high concentrations of the toxin directly to the predator. The results of these feeding studies show no direct toxic effect of Cry1Ab on C. carnea larvae. The amount of toxin ingested by first instar C. carnea in the present study was found to be a factor 10,000 higher than the concentration ingested when feeding on Bt-reared lepidopteran larvae, a treatment that was previously shown to have a negative impact on the predator. In addition, feeding first instar C. carnea with the Cry1Ab toxin did not affect the utilisation of subsequently provided prey. Furthermore, the quality of the prey provided to first instars did not affect the sensitivity of second and third instar C. carnea to the Bt-toxin. The presented results strongly suggest that C. carnea larvae are not sensitive to Cry1Ab and that earlier reported negative effects of Bt-maize were prey-quality mediated rather than direct toxic effects. These results, together with the fact that lepidopteran larvae are not regarded as an important prey for C. carnea in the field, led us to conclude that transgenic maize expressing Cry1Ab poses a negligible risk for this predator.  相似文献

The aim of this study was to investigate protein requirements for the maintenance and growth of blue-breasted quail (Excalfactoria chinensis) from 7 to 21 days of age. A total of 180 quails, 7 days old, were randomly assigned to 36 cages and for 2 weeks were fed diets with a metabolisable energy concentration of 12.13 MJ/kg and a dietary CP concentration of 125, 150, 175, 200, 225 or 250 g/kg. The average BW per cage and the feed intake per cage were recorded daily. The results showed that quails fed 125 g/kg CP could not maintain their BW and had negative feed efficiency. There were linear and quadratic relationships between CP level and response criteria, including BW, weight gain, feed intake, feed efficiency, final body nitrogen mass and body nitrogen accretion (P<0.05). The dietary CP requirements, as calculated using a one-slope quadratic broken-line model, were 211 and 202 g/kg according to weight gain and feed efficiency, respectively. The regression equations, on the basis of metabolic BW, of daily weight gain on daily protein intake according to the model were Y=0.137-2.128(0.113-X) if X<0.113 and Y=0.137 if X>or=0.113 (R2=0.96, P<0.001), which meant that the protein requirement for maintenance was 0.049 times the metabolic BW and that to gain 1 g weight quails needed to ingest an extra 0.47 g protein after the maintenance requirement was satisfied. The regression equations, on the basis of metabolic BW, of daily body nitrogen accretion on daily protein intake according to the model were Y=5.667-76.700(0.119-X) if X<0.119 and Y=5.667 if X>or=0.119 (R2=0.95, P<0.001), which meant that quails had to receive an amount of protein equal to their metabolic BW multiplied by 0.045 to satisfy the requirement for maintenance and then ingest an extra 13 g protein to accrete 1 g body nitrogen.
