构建小鼠heta-防御素2(murie beta-defensin 2,mBD2)的真核表达载体pcDNA-mBD2,并观察其在真核细胞中的表达.从小鼠肝组织中提取mRNA通过逆转录聚合酶链反应扩增得到mBD2基因,定向克隆至真核表达栽体pcDNA3.1(+)获得pcDNA-mBD2;经酶切和测序鉴定构建正确,应用脂质体法转染siha细胞,并通过蛋白免疫印迹法检测细胞内mBD2的表达.结果显示,构建了小鼠mBD2的真核表达载体,转染siha细胞培养并采用G418稳定筛选后,获得稳定转染细胞,收集并裂解转染细胞,蛋白免疫印迹法检测到转染细胞内mBD2的表达.成功构建了真核表达裁体pcDNA-mBD2,为研究mBD2在宫颈癌发生发展过程中的作用及作用机制奠定基础.  相似文献

目的:探讨β-榄香烯对卵巢癌细胞SKOV3增殖和凋亡的影响。方法:体外培养卵巢癌SKOV3细胞,将β-榄香烯(浓度梯度为25,50,100,150,200μg/m L),单独作用于卵巢癌SKOV3细胞,加药24 h、48 h后用噻唑蓝(MTT法)法检测细胞增殖情况;用流式细胞术检测加药24 h后对细胞凋亡的影响。结果:1MTT法结果显示β-榄香烯单独用药24 h、48 h后,与对照组相比,实验组对卵巢癌SKOV3细胞的抑制率均高于对照组(P<0.05),并且在一定程度上呈浓度和时间依赖性。2流式细胞术检测显示,β-榄香烯能够促进SKOV3细胞的凋亡。结论:β-榄香烯能够抑制卵巢癌细胞SKOV3的增殖,促进其凋亡。  相似文献

目的:探讨17β-雌二醇对谷氨酸诱导的SH-SY5Y细胞损伤的保护作用及可能机制。方法:选取SH-SY5Y细胞传代培养, 分为四组:(1)阴性对照组;(2)氧化损伤组:0.1 mmol·L-1谷氨酸作用24 h;(3)17β- 雌二醇低、高浓度组:加入(1.0× 10-4mmol·L-1和 1.0× 10-3 mmol·L-1)17β- 雌二醇作用24 h之后,加入谷氨酸作用24 h。采用MTT 比色法检测细胞存活率,流式细胞仪检测细胞活 性氧(ROS)水平,Hoechst-PI染色观查细胞凋亡,分光光度计检测上清液中Caspase-3 及Caspase-9 含量。结果:7β- 雌二醇能明显 抑制谷氨酸诱导的细胞活性的下降,减少谷氨酸所致SH-SY5Y 细胞内ROS 的生成,降低细胞凋亡率,减少凋亡因子的活性。结 论:17β-雌二醇对神经细胞损伤具有保护作用,这可能与其抗氧化作用有关。  相似文献

目的:研究β-榄香烯抗巨噬细胞源性泡沫细胞的形成及抑制巨噬细胞炎症因子分泌的作用。为探讨β-榄香烯抗动脉粥样硬化(AS)的作用提供依据。方法:采用氧化低密度脂蛋白(ox-LDL)诱导小鼠单核/巨噬细胞(RAW264.7)建立巨噬细胞源性泡沫细胞模型,采用油红O染色鉴定泡沫细胞形成。给予不同浓度(0.5,5,50μM)β-榄香烯干预后,ELISA方法检测巨噬细胞源性泡沫细胞内胆固醇含量和肿瘤坏死因子-α(TNF-α),白介素-6(IL-6)分泌量的变化。结果:β-榄香烯可降低巨噬细胞源性泡沫细胞内总胆固醇(P<0.05或P<0.01),胆固醇酯含量(P<0.01),减少炎症因子TNF-α,IL-6的分泌(P<0.05或P<0.01),并且呈现出一定的浓度依赖性。结论:β-榄香烯抑制巨噬细胞对ox-LDL的摄取,降低细胞内胆固醇的含量,抑制泡沫细胞的形成,同时改善巨噬细胞的炎症状态从而发挥抗动脉粥样硬化的作用。  相似文献

综合运用三维凝胶叠层法和发泡法制备了多孔β-磷酸三钙支架。将多孔支架在1.5倍模拟体液中浸泡14天,得到材料1;或者将其在氢氧化钠溶液中浸泡4天,再在1.5倍模拟体液中浸泡14天,得到材料2。测定了两种材料的物理性能,讨论了类骨磷灰石层对材料矿物组成及其显微结构等的影响。将两组材料分别与成骨前体细胞在体外复合培养,观察和测定了细胞的形态和增殖情况。结果表明复合材料的主要成分为β-磷酸三钙,表面具有结构不完整的含有碳酸磷灰石的类骨磷灰石,成骨细胞能在两组材料上正常粘附和增殖,而且材料2上的细胞粘附情况更好,说明多孔β-磷酸三钙与磷灰石的复合材料有望成为一种有应用前景的骨修复材料和骨组织工程支架材料。  相似文献

Genetic and biochemical studies have suggested that signal transduction by the Wnt pathway is very different to the familiar protein-kinase cascade of the Ras pathway. Among the more intriguing findings is that beta-catenin, a component of intercellular adhesive junctions, is a central component in the Wnt signalling pathway. Recent studies suggest ways in which beta-catenin might serve as a focal point for regulation of adhesion, gene expression and cell proliferation.  相似文献

β-1,6-glucanases degrade the polysaccharide β-1,6-glucan, a cell wall component in some filamentous fungi. A single copy of a β-1,6-glucanase gene, designated gcnA, was identified in each of the grass endophytic fungi Neotyphodium lolii and Epichloë festucae. Phylogenetic analysis indicates that the GcnA protein is a member of glycosyl hydrolase family 5, and is closely related to fungal β-1,6-glucanases implicated in mycoparasitism. The E. festucae gcnA gene was expressed in mycelium grown in culture and in both vegetative and reproductive tissues of perennial ryegrass. A gcnA replacement mutant had reduced β-1,6-glucanase activity when grown in media containing pustulan as the major carbon source. β-1,6-glucanase activity was restored in the replacement mutant by introducing multiple copies of the gcnA gene. Growth of ΔgcnA and gcnA-overexpressing strains in vegetative grass tissues was indistinguishable from wild type strains.  相似文献

目的:肝癌的放射治疗可导致放射性肝损伤(RILD)、甚至肝纤维化及肝硬化等并发症的发生,因此寻找较佳的血清标记物对放射性肝纤维化的无创诊断及监测具有重要意义。本文通过建立放射性肝纤维化大鼠模型,检测血清转化生长因子β1(TGF-β1)的动态表达,从而探讨其与放射性肝纤维化严重程度的相关性及其作为血清标记物的诊断价值。方法:雄性SD大鼠40只,随机分为模型组(30只)和对照组(10只)。除对照组外,模型组大鼠右半肝均接受单次6MV X线25Gy照射,于照射2月、4月、6月后,随机抽取10只,酶联免疫吸附法(ELISA)检测血清TGF-β1的表达,同时将大鼠肝组织进行HE染色,观察肝组织病理变化及大鼠肝纤维化程度,将后者与血清TGF-β1值进行相关性分析。结果:在照射第2月、4月、6月后,模型组大鼠血清TGF-β1值(分别为551.03±69.00 ng/L、645.31±109.29 ng/L、737.89±118.11 ng/L)逐渐升高,均明显高于对照组(451.71±51.12 ng/L,P<0.05)。通过相关性分析表明,大鼠血清TGF-β1值与肝纤维化程度正相关(r=0.82,P<0.01)。结论:在放射性肝纤维化发生发展中,TGF-β1随着肝纤维化严重程度增加,其表达亦升高。本研究为放射性肝纤维化严重程度的监测提供了一种新的无创、操作简单的手段,为后续TGF-β1作为血清标记物应用于放射性肝纤维化的临床研究奠定了理论基础。  相似文献

The positive inotropic effect of isoproterenol was quantified in the presence of several beta-adrenergic blocking agents in ventricular strips of carp heart. Isoproterenol had a concentration-dependent positive inotropic effect. The effect was markedly inhibited by propranolol and carteolol, but was extremely insensitive to atenolol. Practolol totally failed to alter the effect. These results indicated that the positive inotropic effect of isoproterenol may be mediated by mostly beta-2 adrenergic receptors in ventricular strips of carp heart.  相似文献

The 11beta- and 18-hydroxylase activities of a highly purified cytochrome P-450 from bovine adrenocortical mitochondria have been examined in detail with a view to determining whether the two activities are shown by a single protein or by two distinct proteins. The purified enzyme shows a single N-terminal residue (glutamic acid) and its amino acid composition is reported. Both enzyme activities decay considerably during storage at 4 degrees C for 11 days and the rates of decay are similar for the two activities. Metyrapone inhibits both activities competitively (Ki = 1.50 and 1.43 micrometer for 11beta- and 18-hydroxylase, respectively). Carbon monoxide inhibits both activities and the ratio CO:O2 for 50% inhibition is similar for the two activities (K = 1.69 and 1.53). A variety of nonspecific inhibitors produce approximately the same inhibition of both activities. Finally, antiserum produced by rabbits to the purified enzyme on double diffusion in agarose gels gives a single band with the purified enzyme. Increasing concentrations of antiserum added to the assay system produce increasing and proportionate inhibition of both activities. The evidence strongly supports earlier suggestions that the two hydroxylase activities occur in a single protein.  相似文献

Mechanisms involved in the pathogenesis of the AIDS-dementia complex are still unclear. The dichotomy between a small number of HIV-infected cells in the brain and their marked dysfunction could be related to a cellular amplification and/or activation of cerebral viral load by several cytokines. This link between cytokines and viral load could play a role in the generation of the clinical dementia syndrome. We have studied cerebral levels of transforming growth factor-beta1 and interferon-alpha, both in the mild and severe AIDS-dementia complex and also compared these cytokines with HIV RNA load in patients with different degrees of dementia. Our data indicate that production of different cytokines characterized the expression of clinical dementia. In the mild AIDS-dementia complex, there was a significant inverse correlation between interferon-alpha and transforming growth factor-beta1 (r = - 0.743; p < 0.001), and HIV-RNA was present in inverse proportion to transforming growth factor beta1 (r = - 0.751; p < 0.001). In patients with severe AIDS-dementia, transforming growth factor-beta1 was undetectable, while interferon-alpha level were higher than in mild AIDS dementia and correlated positively to cerebral HIV-RNA. No significant difference was evident between these cytokines in the serum of ADC patients and in the control samples. Our study suggests that a relationship is possible between productive HIV infection in the cerebral nervous system and a heterogenous and different expression of the immune response via a complex interaction of cytokines with a differential modulation of the dementia phenotype.  相似文献

Experimental allergic encephalomyelitis (EAE) is an autoimmune disease of the central nervous system mediated by T cells bearing TCR of restricted heterogeneity. Thus, in the murine PL strain, V beta-8.2 is used by 80% of the encephalitogenic T cells. This observation has led to the successful prevention and reversal of EAE by the in vivo use of mAb directed to these restricted gene products. In SJL mice, the V beta-17a gene product has been shown to be used by approximately 50% of encephalitogenic T cells subsequent to immunization with a myelin basic protein (MBP)-derived peptide. However, the other V beta genes used by encephalitogenic T cells in SJL EAE have remained uncharacterized. We now report, for the first time, the beta-chain-encoding DNA sequence of two encephalitogenic, MBP-reactive, SJL-derived T cell clones. These clones which are specific for H-2s and the carboxyl-terminus (amino acid 92-103) of MBP, use TCR encoded by V beta-4. In addition, we demonstrate that the transfer of EAE by a heterogenous SJL-derived encephalitogenic T cell line can be prevented using an anti-V beta-4 antibody in vivo. V beta-4 usage has been previously described in a H-2u/MBP amino-terminus-reactive encephalitogenic T cell. The present findings may thus further support the "V region-disease" hypothesis.  相似文献

The specificity of the mouse class I-specific antibody COB6-3 was examined in detail. It was found to react with the mouse class I molecules H-2Db, Kd, and Qa-2, and with human HLA-A, –B, –C antigens. The specificity pattern of COB6-3, despite its different origin, was similar to that of the monomorphic HLA class I-specific antibody W6/32. Cross-inhibition studies show that on human cells the antigenic determinants recognized by the two antibodies are situated close together and may be identical. Graminan-type fructans are temporarily stored in wheat (Triticum aestivum) stems. Two phases can be distinguished: a phase of fructan biosynthesis (green stems) followed by a breakdown phase (stems turning yellow). So far, no plant fructan exohydrolase enzymes have been cloned from a monocotyledonous species. Here, we report on the cloning, purification, and characterization of two fructan 1-exohydrolase cDNAs (1-FEH w1 and w2) from winter wheat stems. Similar to dicot plant 1-FEHs, they are derived from a special group within the cell wall-type invertases characterized by their low isoelectric points. The corresponding isoenzymes were purified to electrophoretic homogeneity, and their mass spectra were determined by quadrupole-time-of-flight mass spectrometry. Characterization of the purified enzymes revealed that inulin-type fructans [beta-(2,1)] are much better substrates than levan-type fructans [beta-(2,6)]. Although both enzymes are highly identical (98% identity), they showed different substrate specificity toward branched wheat stem fructans. Although 1-FEH activities were found to be considerably higher during the fructan breakdown phase, it was possible to purify substantial amounts of 1-FEH w2 from young, fructan biosynthesizing wheat stems, suggesting that this isoenzyme might play a role as a beta-(2,1)-trimmer throughout the period of active graminan biosynthesis. In this way, the species and developmental stage-specific complex fructan patterns found in monocots might be determined by the relative proportions and specificities of both fructan biosynthetic and breakdown enzymes.  相似文献

There are a lack of biomarkers which can be used to predict clinical outcomes for multiple sclerosis (MS) patients receiving interferon beta (IFN-β).
